This study was undertaken to evaluate the combined effects of sucrose feeding and exercise training on serum insulin, triglycerides, as well as cholesterol and its distribution into lipoproteins of female Wistar rats. The animals were fed ad libitum either laboratory chow alone, or chow and a 32% aqueous sucrose solution. Half of each dietary group was submitted to an exercise-training program. Both sucrose feeding and exercise training elicited greater energy intake. Sucrose feeding produced a marked elevation in triglyceridemia that was prevented by exercise training. Insulin levels paralleled those of triglycerides. The sucrose-fed animals had higher total cholesterol levels than the animals given chow. Although exercise training did not affect total cholesterol in the chow-fed animals, it partly prevented the sucrose-induced elevation in total cholesterol. Cholesterol in the lipoproteins of lower densities was increased significantly with sucrose feeding, and exercise training totally prevented this augmentation. The amount of cholesterol carried by high-density lipoprotein (HDL) was not affected by exercise training in the chow-fed animals. In contrast, sucrose feeding increased HDL-cholesterol in sedentary animals, whereas exercise training partly prevented this increase. The HDL/total cholesterol ratio was similar in all groups. Changes in insulin concentration underline the importance of this hormone in the regulation of blood lipid levels.  相似文献

Groups S and IN were fed diets containing 5% of sucrose or inulin, respectively, for the whole experimental period of 40 days. Group IN/S was fed IN and S diets, whereas group S/IN was fed S and IN diets, in the first and the second 20-day period, respectively. Groups IN(up) and IN(down) were fed diets in which the content of inulin increased from 1-5% and decreased from 5-1%, every 8 days, respectively. The common effects of inulin on caecal fermentation, i.e. enlargement of tissue, acidification of digesta, a decrease in activities of potentially harmful bacterial enzymes (beta-glucuronidase and beta-glucosidase), and an increase in the total volatile fatty acids concentration and pool, were especially observed in the IN, S/IN and IN(up) groups. The results suggested that the intensity of caecal fermentation is increased when inulin is present at a relatively high dietary level and that these changes are easily reversible after inulin withdrawal from feed.  相似文献

This study describes the effect of substituting dietary linoleic acid (18:2 n-6) with alpha-linolenic acid (18:3 n-3) on sucrose-induced insulin resistance (IR). Wistar NIN male weanling rats were fed casein based diet containing 22 energy percent (en%) fat with approximately 6, 9 and 7 en% saturated fatty acids (SFA), monounsaturated fatty acids (MUFA) and polyunsaturated fatty acids (PUFA) respectively for 3 months. IR was induced by replacing starch (ST) with sucrose (SU). Blends of groundnut, palmolein, and linseed oil in different proportions furnished the following levels of 18:3 n-3 (g/100 g diet) and 18:2 n-6/18:3 n-3 ratios respectively: ST-220 (0.014, 220), SU-220 (0.014, 220), SU-50 (0.06, 50), SU-10 (0.27, 10) and SU-2 (1.1, 2). The results showed IR in the sucrose fed group (SU-220) as evidenced by increase in fasting plasma insulin and area under the curve (AUC) of insulin in response to oral glucose load. In SU-220, the increase in adipocyte plasma membrane cholesterol/phospholipid ratio was associated with a decrease in fluidity, insulin stimulated glucose transport, antilipolytic effect of insulin and increase in basal and norepinephrine stimulated lipolysis in adipocytes. In SU-50, sucrose induced alterations in adipocyte lipolysis and antilipolysis were normalized. However, in SU-2, partial corrections in plasma insulin, AUC of insulin and adipocyte insulin stimulated glucose transport were observed. Further, plasma triglycerides and cholesterol decreased in SU-2. In diaphragm phospholipids, the observed dose dependent increase in long chain (LC) n-3 PUFA was associated with a decrease in LC-n-6 PUFA but insulin stimulated glucose transport increased only in SU-2. Thus, this study shows that the substitution of one-third of dietary 18:2 n-6 with 18:3 n-3 (SU-2) results in lowered blood lipid levels and increases peripheral insulin sensitivity, possibly due to the resulting high LCn-3 PUFA levels in target tissues of insulin action. These findings suggest a role for 18:3 n-3 in the prevention of insulin resistant states. The current recommendation to increase 18:3 n-3 intake for reducing cardiovascular risk may also be beneficial for preventing IR in humans.  相似文献

The incidence of the dietary source of energy on lipid transport and accumulation was investigated over a full nycthemeral cycle in adapted rats fed ad libitum. Starch, sucrose and lard were compared. Lipoprotein composition of the plasma, liver and plasma lipids and insulinemia were analyzed every 3 hours over 24 hours. The pattern of VLDL concentration was dependent on the nature of the energetic substrate. Feeding starch resulted in a remarkable stability of lipoproteins, liver and plasma lipids, despite clearcut diurnal variations in plasma non esterified fatty acids, insulinemia and liver glycogen. In sucrose-fed rats VLDL rose to a sharp maximum in the post prandial period (9-12:00) and were totally cleared by 18:00. In fat-fed rats, HDL were elevated during the night, suggesting a possible stimulation of their synthesis by dietary fat in the intestine. LDL were constantly elevated with peak values at 21:00 while VLDL were very low, even at night, despite elevated levels of non-esterified fatty acids. It is concluded that, in animals adapted to a high fat-diet, a high level of circulating non esterified fatty acids is not sufficient to promote the synthesis of VLDL. The main regulating factor appears to be the intensity of hepatic lipogenesis which is stimulated by sucrose and inhibited by lard. No correlation was found between variations in plasma VLDL and insulinemia.  相似文献

The effect of various dietary sugars on the uptake of 1 mM leucine and 1 mM lysine by intestinal cells isolated from stock-fed and sucrose-fed rats was determined. Leucine uptake was activated by 10 mM fructose and inhibited by 10 mM glucose or 20 mM sucrose on both diets. The major dietary effect noted was a significant increase in the inhibition of leucine by glucose in the sucrose-fed rats. The uptake of lysine was minimally affected by the sugars irrespective of the diet fed. These results demonstrate an important dichotomy in the properties of glucose and fructose transport in the intestine and suggest that dietary fructose may increase the transport of certain amino acids.  相似文献

We have previously reported that normal Wistar rats fed an isocaloric, sucrose-rich (63%) diet (SRD) developed glucose intolerance and elevated triglyceride levels in plasma (P) as well as in heart (H) and liver (L) tissue. This metabolic state was accompanied by hyperinsulinism both in vivo and in vitro, suggesting that a state of insulin resistance has developed. In order to gather information on the role of hyperinsulinemia and glucose intolerance in the development of the above lipid metabolism abnormalities, diazoxide, a known insulin release blocking agent was administered (120 mg/kg/day) together with the diet (SRD + DZX) for 22 days. Control groups fed a standard chow (STD) or the STD plus diazoxide (STD + DZX) were included in the study. Under the present experimental design, DZX was able to prevent the development of hyperinsulinism, glucose intolerance and elevated levels of triacylglycerol in plasma, heart and liver present in animals fed on a sucrose rich diet. Our results suggest that mechanisms involved in the development of this nutritionally induced syndrome may include an interaction of hyperinsulinemia, with a direct effect of sucrose on several steps of lipid metabolism.  相似文献

Using an EDTA extraction procedure, bones from pregnant Wistar rats were analyzed for their content of collagen and non-collagenous components (sialoprotein, proteoglycan and carbohydrate). The bone matrix size was found to be smaller in pregnant rats than in normal rats (19.5% vs 17.5% of the dry weight bone). The EDTA extractability of the bone protein from pregnant rats was higher than that from controls (2.6% vs 1.9% dry weight bone). EDTA extracts from pregnant rats contained higher amounts of soluble collagen (1.6% vs 0.5% of dry weight tissue) and lower amounts of non-collagenous components (1.65% vs 2.23% for hexoses, 2.38% vs 3.95% for sialic acid and 1.24% vs 1.73% for uronic acid). In bone matrix, collagen content was lower in the pregnant rats (9.45% vs 10.6%). Similarly, the amounts of non-collagenous components were slightly decreased in the bone matrix from the pregnant rats. The respective values were: 0.91% vs 0.93% for hexoses, 0.45% vs 0.52% for sialic acid and 0.39% vs 0.50% for uronic acid. These results suggest that in pregnancy collagen and non-collagenous protein content in bone is decreased while the total mineral content is increased.  相似文献

This experiment examined skeletal effects of moderate zinc (Zn) supplementation of a low-calcium diet. Male weanling rats were fed experimental diets for about 4 wk. One diet was adequate (control), whereas two others were calcium-deficient, but otherwise adequate. One of the low-calcium (Ca) diets was supplemented with Zn. Dimensions, weight, mineral content, and mechanical properties of femurs were measured. Ca deficiency reduced bone mineral content and strength markedly. Adding Zn to the low-Ca diet further reduced bone strength and elasticity, compared with the unsupplemented low-Ca diet. When the Ca intake is low, possible benefits of Zn supplements should be weighed against risk of deterioration of mechanical properties of bone.  相似文献

During the last two decades Zinc (Zn) as a micronutrient is being used indiscriminately in agricultural and husbandry practices and also in baby foods and multivitamin supplements with a view that Zn is non-toxic and promotes linear growth and body weight in the consumers. The long-term effect of increasing Zn load in the body has not been worked out so far. In this study, three groups of rats were fed on a semi-synthetic diet containing 20 mg (control, group-I), 40 mg (group-II) and 80 mg Zn /kg (group-III) diet respectively for 6 months. The results revealed that the gain in body weight increased in rats in Zn-concentration dependent manner. The urine examined on weekly basis showed glucosuria in group-II on week 10 and in group-III on week 8 and thereafter. The arterial blood pressure was significantly higher in group-II and III than their control counter parts on monthly basis. Histochemical examination of skin revealed an increase in the number of adipocytes filled with triglycerides making a subcutaneous fatty tissue thicker in group-II and group-III than that of control group. The blood profile after 180 days of dietary treatment, displayed a significant rise in glucose, total lipids, cholesterol, triglycerides, LDL-cholesterol, VLDL-cholesterol, insulin, cortisol and aldosterone whereas HDL-cholesterol, T3, T4 and TSH showed a reduction in their levels in the blood serum. The tissue metal status showed an increase of Zn, Cu and Mg in the serum, a rise in Zn in liver, hair and abdominal muscles and fall in Cu and Mg concentrations in liver, hair and abdominal muscles. This data suggest that Zn in excess in diet when fed for longer periods of time induces metabolic syndrome-X.  相似文献

Human and bovine bone matrices were extracted with salt solutions of different composition and the extracts tested for stimulation of incorporation of radioactivity from [3H]glucosamine and [35S]SO4 into the hyaluronic acid and chondroitin sulfate of the cell pellet, the cell surface and the medium fractions of human synovial cells in culture. Stimulatory activity was extracted with a solution of 0.3 M EDTA in 2.5 M NaCl from bovine but not human bone. Subsequent extraction of the residues with 4 M guanidinium hydrochloride yielded activity from both matrices. A major stimulation of incorporation of radioactivity was observed in the cell surface fractions. Human synovial cells constitute a more sensitive assay system for the stimulatory activity than rabbit synovial cells.  相似文献

Two 90 L anaerobic baffled reactors were used to study the granulation of sludge and the effect of the organic loading rate and NaHCO3/COD ratios on reactor performance. Furthermore, it was determined whether an anaerobic baffled reactor would promote phase separation and if additive of bentonite or granular active carbon was capable of enhancing granule formation. In order to minimize feed variations, and have a totally biodegradable substrate, a synthetic sucrose substrate was used. Granulation was achieved in both reactors within 75 days. However, the granules from the granular active carbon amended reactor appeared earlier and were larger and more compact. The reactors were maintained at a hydraulic retention time of 20 h during performance study stage. The results showed that when organic loading rate were changed from 2.15 to 6.29 kg COD m(-3)day(-1), chemical oxygen demand (COD) removal was not decreased (91-93%), but a slight increase in effluent COD was observed. It was found that the COD removals were generally good (87-92%) and had not obviously change with the decreasing NaHCO3/COD ratios. From the bacterial distribution and the concentration of volatile fatty acids in four compartments, it was concluded that a separation of phases occurred within the anaerobic baffled reactors.  相似文献

The effect of feeding garlic oil to white albino rats maintained on high sucrose and alcohol diets was studied. It is proposed that the mechanism of the hypolipidemic effect of the oil involves the active principle, diallyl disulphide, inactivating enzymes and substrates containing thiol groups in an exchange reaction; increased hydrolysis of triacylglycerols as increased lipase activity is induced by the oil; and the reduction in the biosynthesis of triacylglycerols as NADPH is made unavailable for the process by the metabolism of the oil.  相似文献

The study objective was to determine if male and female rats fed a diet rich in fish oil had femurs and vertebrae that were stronger and more resistant to fracture than rats not fed omega-3 long chain polyunsaturated fatty acids. Weanling rats were randomized to a control or a fish oil diet for 5 weeks. Feeding fish oil to males had no effect on biomechanical strength properties of femurs and vertebrae as measured by three point bending and compression, respectively. In contrast, females fed fish oil had reduced length growth and a lower vertebral peak load. These effects may have been partly mediated by a lower food intake but were not associated with differences in serum IGF-I, estradiol or urinary calcium. The effect of consuming a fish oil diet into later adulthood should be investigated to determine if femur strength is also affected among females.  相似文献

Confluent cultures of rat muscle fibroblastic cells respond by increased glycosaminoglycan (GAG) synthesis when cultured in medium containing a solubilized bone matrix fraction (SBM) at a concentration of 100 micrograms/ml. The metabolism of the GAG associated with the cell pellet, the cell surface and the tissue culture medium fractions was studied, in the presence and absence of SBM, by measuring the incorporation of radioactivity from [3H]glucosamine and [35S]SO4 into the isolated GAG. Net synthesis of hyaluronic acid and of chondroitin sulfate in the medium fraction increased more rapidly in cultures containing SBM compared to controls, and the accumulation of labelled GAG in the medium of the treated cultures was approximately linear with respect to the length of incubation. The addition of SBM also resulted in increased incorporation of 3H and of 35S into the GAG of the cell surface and cell pellet fractions. In these fractions, stimulation of incorporation of radioactivity occurred in two waves: an early, relatively minor increase and a later relatively major increase. The relatively major stimulation of radioactivity into the GAG of the cell surface fraction occurred between 24 and 48 h and was independent of any apparent effect of serum.  相似文献
