Interspecific host discrimination and within-host competition between Encarsia formosa Gahan and Encarsia pergandiella (Howard), two endoparasitoids of whiteflies, were studied under laboratory conditions. Interspecific host discrimination was studied at two time intervals (0 h and 72 h after the first species had oviposited). Parasitized and unparasitized Trialeurodes vaporariorum (Westwood) hosts were accepted for oviposition at the same rate by the two parasitoid species. Host type did not affect the handling time of the two parasitoids. The outcome of within-host competition was investigated after females of the two species parasitized the hosts at various time intervals. In four treatments, E. pergandiella was allowed to oviposit 0, 24, 48 and 72 h after E. formosa while in the other two, E. formosa was allowed to oviposit 0 and 72 h after E. pergandiella. In four of these treatments: E. formosa following E. pergandiella at 0 and 72 h, and E. pergandiella following E. formosa at 0 and 24 h, E. pergandiella prevailed. In the host discrimination experiment (72 h interval), 20% of E. pergandiella eggs were killed by E. formosa females. Interspecific ovicide was also observed in the within-host competition experiment, in which 6% of 72-h-old E. pergandiella eggs were killed by E. formosa females.  相似文献

The parasitoid Encarsia formosa Gahan (Hymenoptera: Aphelinidae) has been used successfully for the control of Trialeurodes vaporariorum (Westwood) (Homoptera: Aleyrodidae). The development of UV-blocking plastic films has added a new component to future integrated pest management systems by disrupting insect pest infestation when UV light is excluded. Because both T. vaporariorum and E. formosa are reported to have similar spectral efficiency, there was a need to identify the impact of UV-blocking films on the dispersal behavior of both the pest and the natural enemy. In field studies, using choice-chamber experiments, E. formosa showed some preference to disperse into compartments where less UV light was blocked. However, further studies indicated that the effect was primarily attributable to the different light diffusion properties of the films tested. Thus, unlike its whitefly host, when the UV-absorbing properties of the films were similar, but the light diffusion properties differed, E. formosa adults preferred to disperse into compartments clad with films that had high light diffusion properties. When the plastic films differed most in their UV-absorbing capacity and had no light-diffusion capability, the initial dispersal of E. formosa between treatments was similar, although a small preference toward the environment with UV light was observed over time. When parasitoid dispersal was measured 3 h after release, more parasitoids were found on plants, suggesting that the parasitoids would search plants for whitefly hosts, even in a UV-blocked light environment. The potential for the integration of UV-blocking films with E. formosa in an advanced whitefly management system is discussed.  相似文献

The eggs of most oviparous animals are provisioned with a class of protein called vitellogenin (Vg) which is stored as the major component of yolk. Until recently, deduced amino acid sequences were available only from vertebrate and nematode Vgs, which proved to be homologous. The sequences of several insect Vgs are now known, but early attempts at pairwise alignments with vertebrate and nematode Vgs have been problematic, leading to conflicting conclusions about how closely insect Vgs are related to the others. In this paper we demonstrate that insect Vg sequences can be confidently aligned with one another along their entire lengths and with multiple vertebrate and nematode Vg sequences along most of their spans. Although divergence is high, conservation among insect, vertebrate, and nematode Vg sequences is widespread with a preponderance of glycine, proline, and cysteine residues among strictly conserved amino acids, establishing conclusively that Vgs from the three phyla are homologous. Areas of least-certain alignment are primarily in and around insect and vertebrate polyserine domains which are not homologous. Phylogenetic reconstructions of Vgs based on sequence identities indicate that the insect lineage is the most diverged and that the mammalian serum protein, apolipoprotein B-100, arose from a Vg ancestor after the nematode/vertebrate divergence. Vitellogenin (Vg) is a precursor of the major yolk protein, an essential nutrient for the embryonic development of oviparous animals including insects. Here, the gene(CceVg [Corcyra cephalonica Vg] ) encoding the Vg (CceVg of moth, C. cephalonica, was cloned and sequenced. The gene sequence was 6,721‐bp long and contained 5five introns and six exons that together formed a 5,382‐bp open reading frame. The deduced protein (CceVg) consisted of 1,793 amino acid residues, including a 16‐amino‐acid signal peptide. The putative molecular weight of the primary Vg protein was 202.46 kDa. The CceVg contained all conserved domains and motifs that were commonly found in most insect Vgs except the presence of a polyserine tract at the C‐terminal region, which had not been reported in other lepidopteran Vgs. The expression pattern showed thatCceVg was first transcribed at a very low level in the early larval stage but disappeared in later stage larva. In female, theCceVg mRNA was detected in early pupal stage and throughout adult stage. Interestingly, theCceVg mRNA was detected only in mated males at low levels, not in the virgin ones. Injection ofCceVg double‐stranded RNA into early‐emergent females caused severely abnormal ovaries.  相似文献

We previously reported a cDNA for vitellogenin (Vg) from the cockroach, Leucophaea maderae (Lm). In the present study, we identified another cDNA encoding a second Vg (Vg2) having stretches of amino acid sequences different from the first one, Vg1, reported earlier. The complete nucleotide sequence of Vg2 consisted of 5,915 bp, which encoded a primary protein of 1,911 residues including a 16-residue putative signal peptide. The regions different in both Vg precursors (Pro-Vg1 and pro-Vg2) were four in number, and two, relatively longer, existed at the carboxy terminal. The presence of two Vg-related cDNAs was confirmed by sequencing of RT-PCR products generated using primers designed based on the common sequences flanking the regions different in amino acid sequences. Both forms were transcribed since they could be amplified on mRNA from fat bodies of different individual females. Southern blot analysis of digested genomic DNA revealed the existence of two Vg-related genes in L. maderae indicating that each Vg cDNA originated from a separate gene. Also, the immunoblot analysis using antibodies generated against peptides unique to both Vg1 and Vg2 probed the same antigen in the same individual, suggesting LmVg to be a product coded by two different Vg precursors. Both Vg primary products showed 96% similarity at an amino acid level. Compared to other insect Vgs, Vg2 showed a slightly higher (1-2%) similarity than Vg1. We previously reported, based on amino-terminal sequence analysis, that L. maderae pro-Vg was cleaved into four subunit polypeptides (112-, 100-, 92-, and 55-kD), which were deposited in the egg as four respective vitellin (Vn) polypeptides. We show now based on immunoblot analysis that the 112-kD polypeptide is further cleaved, near the C-terminus, to an 87-kD polypeptide before it is secreted into the hemolymph. Both the L. maderae Vgs were compared with each other and with other insect Vgs and the processing pattern is discussed.  相似文献

Vitellogenins (Vgs) are precursors of the major egg storage protein, vitellin (Vn), in many oviparous animals. Insects Vgs are large molecules (∼200-kD) synthesized in the fat body in a process that involves substantial structural modifications (e.g., glycosylation, lipidation, phosphorylation, and proteolytic cleavage, etc.) of the nascent protein prior to its secretion and transport to the ovaries. However, the extent to which Vgs are processed in the fat body varies greatly among different insect groups. We provide evidence by cloning and peptide mapping of four Vg molecules from two cockroach species (Periplaneta americana and Leucophaea maderae) that, in hemimetabolous insects, the pro-Vg is cleaved into several polypeptides (ranging from 50-to 180-kD), unlike the holometabolans where the Vg precursor is cleaved into two polypeptides (one large and one small). An exception is the Vg of Apocrita (higher Hymenoptera) where the Vg gene product remains uncleaved. The yolk proteins (YPs) of higher Diptera (such as Drosophila) form a different family of proteins and are also not cleaved. So far, Vgs have been sequenced from 25 insect species; 9 of them belong to Hemimetabola and 16 to Holometabola. Alignment of the coding sequences revealed that some features, like the GL/ICG motif, cysteine residues, and a DGXR motif upstream of the GLI/CG motif, were highly conserved near the carboxy terminal of all insect Vgs. Moreover, a consensus RXXR cleavage sequence motif exists at the N-terminus of all sequences outside the Apocrita except for Lymantria dispar where it exists at the C-terminus. Phylogenetic analysis using 31 Vg sequences from 25 insect species reflects, in general, the current phylogenies of insects, suggesting that Vgs are still phylogenetically bound, although a divergence exists among them.  相似文献

The genus Encarsia F?rster includes parasitoid species that are effective natural enemies of whitefly and armoured scale insect agricultural pests. Within this genus, several species groups have been recognized on the basis of morphological similarity, although their monophyly appears uncertain. It is often difficult to separate morphologically similar species, and there is evidence that some species could in fact be complexes of cryptic species. Their correct identification is fundamental for biological control purposes. Recently, due to unreliability of morphological characters, molecular techniques have been investigated to identify markers that differentiate closely related species. In this study, DNA variation in an approximately 900 bp segment of the mitochondrial cytochrome oxidase subunit I (COI) gene was examined by both sequencing and PCR-RFLP. Two pairs of species that are difficult to distinguish morphologically were analysed: Encarsia formosa Gahan and Encarsialuteola Howard, belonging to the luteola group, and two populations of Encarsiasophia (Girault & Dodd) from Pakistan and Spain, belonging to the strenua group, recently characterized as cryptic species. High sequence divergence and species-specific restriction patterns clearly differentiate both species pairs. Parsimony analysis of the nucleotide sequences was also performed, including Encarsiahispida De Santis (luteola group) and Encarsia protransvena Viggiani (strenua group). Two monophyletic clades supporting the two groups of species considered were resolved. The results of this study support the use of the COI gene as a useful marker in separating species of Encarsia, for which morphological differences are subtle. Moreover, the COI gene appears potentially useful for understanding phylogenetic relationships in this genus.  相似文献

The American cockroach, Periplaneta americana has two vitellins (Vn1 and Vn2) and corresponding vitellogenins (Vg1 and Vg2). Vns/Vgs were separated on the SDS-PAGE as three major polypeptide bands [170, 100 (multisubunits), and 50 kD] and a minor polypeptide band (150 kD) both in the egg (mature terminal oocyte) extract and in the female hemolymph. We previously cloned one Vg (Vg1) cDNA and showed that the 170-kD polypeptide originated from the C-terminus of the Vg1. In the present study, we cloned the other Vg (Vg2) cDNA. It is 5,826 bp long encoding 1,876 amino acid residues (including 16 residues for putative signal peptide) in a single ORF. The deduced amino acid sequences of both Vgs (Vg1 and Vg2) of P. americana showed 30% identity. The GL/ICG motif is followed by eight cysteine residues at conserved locations near the C-terminal and the DGXR motif starts 18 residues upstream of the GL/ICG motif. The chemically determined N-terminal amino acid sequences of the 150-kD and of the 50-kD polypeptides matched exactly with each other and with the deduced N-terminal amino acid sequence of the Vg2 cDNA. The pattern of processing in P. americana Vns/Vgs is discussed.  相似文献

The ovipositional patterns of the heteronomous hyperparasitoid Encarsia pergandiella Howard (Hymenoptera: Aphelinidae) in the presence of its primary host Bemisia argentifolii Bellows & Perring (Hemiptera: Aleyrodidae), and in the presence or absence of conspecific and heterospecific secondary hosts (Encarsia formosa Gahan andEretmocerus mundus Mercet; Hymenoptera: Aphelinidae) were examined to assess host species preferences. Host preferences by heteronomous hyperparasitoids may affect the relative abundance of co-occurring parasitoid species and may influence host population suppression by the parasitoid community. Four combinations of hosts were tested: (1) B. argentifolii, E. mundus, and E. formosa, (2) B. argentifolii, E. formosa, and E. pergandiella, (3) B. argentifolii, E. mundus, and E. pergandiella, and, (4) B. argentifolii, E. mundus, E. formosa, and E. pergandiella. Arrays of hosts (24) were constructed in Petri dishes using leaf disks, each bearing one host. Thirty arrays of each host combination were exposed to single females for 6 h. All hosts were dissected to determine number of eggs per host. Encarsia pergandiella parasitized E. formosa hosts as frequently as E. mundus hosts. However, E. pergandiella parasitized either of these heterospecific hosts more frequently than conspecific hosts in treatments including two secondary host species. When a third parasitoid species was included in host arrays, E. pergandiella parasitized conspecific hosts as frequently as heterospecific hosts. Developmental stage of the hosts did not significantly influence host species selection by E. pergandiella. Our results indicate that host selection and oviposition by heteronomous hyperparasitoids like E. pergandiella, vary with the composition of hosts available for parasitization, and suggest a preference for heterospecific over conspecific secondary hosts.  相似文献

We cloned and sequenced the cDNA of vitellogenin (Vg) from the cicada Graptopsaltria nigrofuscata (Homoptera). The deduced amino acid sequence of 1987 residues (including 16 residues for a putative signal peptide) was obtained. The pro-Vg was cleaved into two subunits between residues 379 and 380 following a consensus RXXR cleavage site sequence, secreted as S-Vg (apparent molecular weight 43 kDa) and L-Vg (200 kDa), sequestered, and stored in the egg as two vitellins (Vns), S-Vn and L-Vn, with similar respective molecular weights. There was a single long serine-rich stretch closely following the cleavage site. The entire amino acid sequences of the Vgs from the eight insects so far reported could be aligned confidently. The presence of subdomains I-V (areas of relatively high amino acid conservation) and of 10 cysteines at conserved locations at the C-terminus, noted previously among insect Vgs, were confirmed. Antisera raised against G. nigrofuscata S- and L-Vn cross-reacted with the S- and L-Vg/Vn, respectively, of all three other cicada species examined. Another major egg protein (170 kDa) unrelated to Vg/Vn, was also detected in all species examined.  相似文献

Only a few biochemical and molecular studies on yolk proteins (vitellins) have been carried out in mollusks, mainly in bivalves, while information on prosobranch vitellogenesis is still limited. In this study, we cloned a full-length cDNA encoding vitellogenin (Vg) in the Pacific abalone Haliotis discus hannai. The complete Vg cDNA consists of 7753 nucleotides with a long open reading frame encoding 2391 amino acid residues. The deduced primary structure contains the N-terminal amino acid sequences of the 95 kDa and 150 kDa subunits of vitellin of the abalone and shows similarities to Vgs of other mollusk, fish, nematode and coral species. In common with bivalve Vgs, the abalone Vg gene was expressed only in the ovary. In situ hybridization analysis further localized Vg mRNA to the follicle cells in the ovary. We conclude that the follicle cells are the site of Vg synthesis in H. discus hannai.  相似文献

Abstract.  Insects, particularly phloem-feeding Sternorrhyncha, are known to produce sugars in their honeydew (excreta) that are not found in their host plants. Of these, Bemisia tabaci , the sweet potato whitefly, is the only insect known to produce trehalulose [α- d -glucose (1,1) d -fructose] as a major component of its honeydew. The present study aims to determine whether trehalulose is comparable to sucrose as a nutrient source for three whitefly parasitoids ( Encarsia formosa , Encarsia pergandiella and Eretmocerus eremicus ). In addition, the study also examines trehalulose feeding effects on longevity for a parasitoid of muscoid Diptera, Nasonia vitripennis . Parasitoids are provided diets of either sucrose or trehalulose in varying concentrations (from 0.1% to 70%) or a water control. Sucrose and trehalulose are not significantly different in affecting survival when compared at the same concentration. This was true for all Bemisia parasitoids and N. vitripennis. Certain specific diets are significantly different in pairwise combination tests. There is a significant effect of species, diet type and the interaction of these two factors on the longevity of the three different Bemisia parasitoid species; however, within species, there is no significant increase in longevity observed for either carbohydrate diet. This result contrasts with expectations for the effects of host-modified carbohydrates on longevity. The implications are that, although carbohydrate feeding is essential for these parasitoids, these host-provided sources of carbohydrates are equally capable of extending longevity.  相似文献

A cDNA encoding vitellogenin (Vg) of the cockroach, Leucophaea maderae was cloned and sequenced. The deduced amino acid sequence consisting of 1913 residues (including 15 residues for a putative signal peptide) was obtained. Amino-terminal sequence analysis demonstrated that the pro-Vg was cleaved into four polypeptide 'subunits' following the three consensus RXXR cleavage site sequences, which were secreted as four Vg polypeptides (apparent molecular weights = 112-, 100-, 92- and 55-kD), sequestered, and deposited in the egg as four respective vitellin (Vn) polypeptides. There was, however, an additional 90-kD Vn polypeptide existed in the egg. We show that this polypeptide is a processed product from 92-kD Vn polypeptide. Northern blot analysis of poly (A)+ RNA reveals that mRNA coding for Vg is present only in the female fat body cells but neither in the ovary nor in the male fat body cells. The deduced amino acid sequence contained a serine-rich stretch at the C-terminal region. This stretch occurred also in Vgs of Periplaneta americana (Vg1 and Vg2) and Blattella germanica. The Vg of L. maderae had 26% and 31% homology with those of P. americana (Vg1 and Vg2) and B. germanica, respectively. Phylogenetic analysis (neighbour-joining) was made using four cockroach Vgs and the tree was compared with other molecular and conventional phylogenetic trees.  相似文献

Using the electroretinogram (ERG) technique the spectral efficiency of the compound eye of the glasshouse whitefly Trialeurodes vaporariorum (Westwood) (Homoptera: Aleyrodidae) and its main parasitoid Encarsia formosa (Gahan) (Hymenoptera: Aphelinidae) was measured at selected wavelengths between 340 nm and 670 nm. The form of the ERG for both T. vaporariorum and E. formosa was found to be monophasic in nature. For both male and female T. vaporariorum and female E. formosa a primary peak of efficiency occurred in the blue-green-yellow region, peak 520 nm and a secondary peak in the ultraviolet (UV) region. The compound eye of female E. formosa gave a significantly greater response in the UV region than either the dorsal or ventral regions of the compound eye of T. vaporariorum relative to the responses in the blue-green-yellow region. T. vaporariorum has divided compound eyes with distinct dorsal and ventral regions. In this study it was found that the percentage response in the UV, of the dorsal region of the eye, is significantly greater than that of the ventral region of the eye relative to the percentage response in the blue-green-yellow region and there is a significant shift in the blue-green yellow peak towards the right of the spectrum.  相似文献

The cDNA for vitellogenin (Vg) of the parasitoid wasp Pimpla nipponica (Hymenoptera: Apocrita) was cloned and sequenced.1 The deduced amino acid sequence with 1807 residues was obtained. The N-terminal 20 amino acids chemically determined for vitellin (Vn) agreed completely with the deduced 20 amino acids that follow the 16 amino acid residues for putative signal peptide. The cDNA clone for the Vg of the turnip sawfly Athalia rosae (Hymenoptera: Symphyta), previously obtained and partially sequenced, was also completely sequenced and the amino acid sequence deduced. Amino acid sequences were compared between these two species and also with known Vg sequences from other insects. Common to all these insects is the presence of two long regions with relatively well-conserved amino acid sequences, one near the N-terminal extending 267–282 residues (including two cysteines at conserved locations), and the other starting at position 450 to 655 and extending 279–283 residues, and of a region at the C-terminal extending some 200 residues (about 250 in Aedes aegypti due to the presence of a serine-rich stretch) with 10 cysteines at conserved locations. Abstract.  1. Studies of inter-specific competition in parasitoids have largely focused on the outcome of within-host competition and the behavioural mechanisms by which female parasitoids prevent competition. Another, less well studied, possibility is oviposition preceded by 'heterospecific ovicide', the destruction of the other species' egg. Heterospecific ovicide essentially eliminates within-host competition.
2. This study investigated the mechanisms and outcome of within-host competition in Encarsia formosa and Encarsia luteola , solitary endoparasitoids of whitefly pests. These species are known to commit ovicide of conspecific eggs.
3. Competition experiments indicated that the offspring of second-ovipositing females had an apparent advantage in competition, regardless of whether the second female was E. formosa or E. luteola .
4. Observations of ovipositor movement through the cuticle of host whitefly nymphs showed that both species often committed heterospecific ovicide and then oviposited or host-fed. Multiparasitism and heterospecific host discrimination were less common and absent respectively.
5. Heterospecific ovicide appears to explain the second-female advantage in competition between these species. Second-female advantage is contrary to the paradigmatic view of multiparasitism, where the first-ovipositing female has an advantage in competition or one of the species consistently prevails in competition.  相似文献

This study assessed the quality of three commercially available natural enemies used for pest management in greenhouses: the whitefly parasitoid Encarsia formosa Gahan (Hymenoptera: Aphelinidae), the aphid parasitoid Aphidius colemani Viereck (Hymenoptera: Braconidae), and the aphid predatory midge Aphidoletes aphidimlyza (Rondani) (Diptera: Cecidomyiidae). Shipment packaging was consistent for all natural enemies. However, there was high variability in delivery punctuality, product cost, and product information provided by each of the six selected companies. Product quantity, percentage of emergence upon arrival, percentage of total emergence, percentage of females, and percentage of flying insects were assessed using International Organization for Biological Control (IOBC) recommended procedures. The parameters with greatest variability between companies were percentage of emergence upon arrival (0.9-10.5%) and percentage of flying insects (35.4-85.0%) for E. formnosa; product quantity (623.3-833.8 aphid mummies), percentage of emergence upon arrival (6.1-41.2%) and percentage of females (51.1-54.8%) for A. colemani; and percentage of emergence upon arrival (0.0-7.7%) and percentage of females (54.6-76.2%) for A. aphlidimyza. Results are discussed in terms of the value to consumers and compared with IOBC standards.  相似文献

The flight behaviour of Encarsia formosa Gahan (Hymenoptera: Aphelinidae), a parasitoid of Trialeurodes vaporariorum (Westwood) (Homoptera: Aleyrodidae) has been studied in response to the following natural components: bean plant (Phaseolus vulgaris L.) infested with T. vaporariorum (PHC, plant host complex), host damaged bean plant from which whiteflies were removed (HDP, host damaged plant), T. vaporariorum third instar larvae (H, host), uninfested bean plant (P, plant) and T. vaporariorum empty pupal cases (EPC, empty pupal cases). A combination of chemical and visual stimuli is necessary in host location from a distance by E. formosa and seems to deeply affect the dispersal of this parasitoid. The presence of host (PHC, H) or of its parts (EPC) led to a substantial increase of oriented flights suggesting the occurrence of host derived chemicals. At a closer range, a combination of visual and chemical stimuli deriving from all the components of the plant host complex elicited the transition flying-landing and the following landing on the source.  相似文献
