Burbot Lota lota sampled from lakes Mjosa and Losna in southeastern Norway between 2005 and 2008 were found to be infected with Mycobacterium salmoniphilum at a culture-positive prevalence of 18.6 and 3.3%, respectively. The condition factor (CF) of mycobacteria-affected fish sampled from Mj?sa in 2008 was lower than the average CF of total sampled fish the same year. Externally visible pathological changes included skin ulceration, petechiae, exopthalmia and cataract. Internally, the infections were associated with capsulated, centrally necrotic granulomas, containing large numbers of acid-fast bacilli, found mainly in the mesenteries, spleen, heart and swim bladder. Mycobacterial isolates recovered on Middlebrook 7H10 agar were confirmed as M. salmoniphilum by phenotypical investigation and by partial sequencing of the 16S rRNA, rpoB and Hsp65genes as well as the internal transcribed spacer (ITS1) locus. This study adds burbot to the list of fish species susceptible to piscine mycobacteriosis and describes M. salmoniphilum infection in a non-salmonid fish for the first time.  相似文献

Records of burbot Lota lota (L.) captures from the early nineteenth century to the present have been gathered and arranged watershed by watershed in chronological order. Most are from eastward-flowing river systems from Durham southwards to the Great Ouse, but a few records from westward-flowing systems are considered. In many areas the records imply a decline of burbot numbers and distribution during the present century, though burbot may never have been more than locally abundant. Local over-fishing, pollution and habitat changes are considered the most likely causes of decline. Conservation measures seem desirable.  相似文献

The parasite fauna of the burbot (Lota lota) within its natural range is reviewed. The sent paper summarizes the data on parasites of the burbot from water bodies of Eurasia and North America, based on published monographs, reviewed journals, scientific reports, conference contributions, and PhD theses. The checklist includes all protozoan and metazoan parasites of the burbot. A total of 242 parasite species/taxa were recorded in the burbot (Ki-netoplastomonada--4, Parasitomonada--3, Coccidiomorpha--1. Microsporidea--3, Myxosporidia--35, Pleurostomata--1. Cyrtostomata--3, Peritricha--20. Protozoa incertae sedis--1. Monogenea--8, Cestoda--23, Digenea--50. Nematoda--36, Acanthocephala--28, Hirudinea--11. Bivalvia--5, Crustacea--10). Most parasites belong to digenean trematodes. Most of these species (183 species/taxa) were recorded on Eurasian and only 92--in North America fishes. Several parasite species recorded from the burbot are discussed in relation to host specificity and their geographical distribution.  相似文献

We developed 21 polymorphic dinucleotide microsatellite loci, (CA)n and (CT)n, for the Holarctic freshwater fish, Lota lota, using an enriched genomic library protocol. The species has an interesting life history because winter‐spawning adults migrate over long distances to form spawning aggregations, a behaviour which should maintain genetic homogeneity across large spatial scales. Availability of the reported microsatellites will facilitate the investigation of population genetic structure with regard to postglacial colonization history and conservation strategies. The primers were screened on 30 individuals from a natural population (Lake Constance, southern Germany), revealing three to 24 alleles per locus with expected heterozygosities ranging from 0.48 to 0.93.  相似文献

A rearing experiment was conducted to test whether temperature protocols that differed from a simulation of natural conditions might induce maturation after isothermal grow‐out in burbot Lota lota. Lota lota were acclimated to two different temperature regimes: low temperature (LT), close to natural temperature at 4·0° C and elevated, high temperature (HT) at 8·5° C over 40 and 27 days respectively, with all fish then wintered for 47 days. Every second fish was treated with a gonadotropin‐releasing hormone analogue. Maturational competence of oocytes was assessed with a germinal vesicle breakdown assay using a novel staining strategy. In both treatments, puberty and maturational progress occurred, characterised by an elevated gonado‐somatic index and advanced gonadal stages (histological analysis). Progress of gonadal maturation was reflected by elevated plasma concentrations of testosterone and 11‐ketosterone in males and 17β‐oestradiol in females. Ovulation was not observed. Sperm could be activated equally across treatments. In general, LT was more effective than HT treatment, indicated by advanced gonadal stages, higher numbers of oocytes undergoing germinal vesicle breakdown in vitro and elevated sex steroid levels. Hormone treatment could improve effectiveness at HT. In conclusion, less drastic temperature regimes as previously reported in combination with hormone treatments seem sufficient to induce maturation in L. lota after isothermal grow‐out.  相似文献

The burbot (Lota lota Linnaeus, 1758) is the only freshwater species from the cod family. Various taxonomic hypotheses were tested against molecular data by sequencing the mitochondrial cytochrome b locus of 120 burbot from 41 populations together with the related species Molva molva (ling) and Brosme brosme (tusk), which represented the other Lotinae genera. Within the genus Lota two distinct phylogroups were observed: one in North America south of the Great Slave Lakes (Lota lota maculosa) and one in Eurasia and the remainder of the Nearctic region (Lota lota lota). The burbot lineage separated 10 Myr BP from the other Lotinae, while the genetic variation within burbot appeared to be approximately 1 Myr old. However, fossil evidence suggested that burbot already existed in the Early Pliocene in Europe, from were it probably colonized North America in the Early Pleistocene. While Nearctic burbot survived climatic oscillations and diverged in several refugia, the Eurasian form became extinct or was reduced to a very small population. In the Late Pleistocene the species recolonized the Palearctic region to establish its present distribution range.  相似文献

Sperm motility and composition of the seminal fluid in Lota lota were investigated. Fives after motility initiation, 88.2 ± 12.4% of the spermatozoa were motile, their mean average path swimming velocity was 61.6 ± 16.3 μm s?1 and their principal swimming type the linear motion (77.4 ± 20.9%). In distilled water the rate of motile spermatozoa decreased to 0% in 40s. In 25–50 mosmol kg?1 electrolyte (NaCl) or non-electrolyte (glucose, sucrose) solutions, motility was prolonged for 10s and these solutions can therefore increase the efficiency of artificial fertilization when used for sperm motility activation. When semen was diluted in electrolyte or non-electrolyte solutions with osmolalities higher than 50 mosmol kg?1, sperm motility rates and swimming velocities decreased, and at osmolalities of 400 mosmol kg?1 motility was completely suppressed. In the seminal fluid with an osmolality of 290.08 ± 45.22 mosmol kg?1, sodium levels of 139.86 ± 23.79 mmol × 1?1, potassium levels of 11.59 ± 2.45 mmol × 1?1 and calcium levels of 0.20 ± 0.08 mmol × 1?1, sperm motility was inhibited. Under in vitro conditions, artificial saline solutions resembling the seminal plasma composition and 400 mosmol kg?1 NaCl or glucose solutions were useful as motility inhibiting solutions for predilution of semen. Sperm motility was not affected by pH 7.5–9.0, but at pH 6 the motility rate and the swimming velocity were reduced; seminal fluid pH was 8.47 ± 0.02. Therefore buffering of the artificial saline solutions can provide more stabile conditions for semen during storage and activation. Temperature optimum of semen was between 2 and 5°C. At higher temperatures semen became spontaneously motile. Therefore, controlled temperature conditions are an important factor for handling of semen. The qualitative, organical composition of seminal fluid was similar as in other fresh water teleosts.  相似文献

The cryopreservation of spermatozoa of a teleost fish, the burbot, Lota lota (Gadidae) was investigated. Cryopreserved semen had the highest motility rate (46.6+/-8.0%, fresh semen control 86.5+/-8.2%) and fertility (78.1+/-2.7% embryo survival in hatching stage, fresh semen control 82.2+/-2.9%) when 10% methanol, 1.5% glucose and 7% hen egg yolk were used as cryoprotectants. Freezing was performed in 0.5-ml straws in the vapour of liquid nitrogen at 1cm above the level of liquid nitrogen and thawing in water at 25 degrees C for 20s. For optimal fertilization cryopreserved semen was first mixed with the eggs and then 25 or 50 mmol/L NaCl solution (pH 8.5) was added at a ratio of 1:24 (semen:saline solution). Under these conditions fertilization ratios in the range of fresh semen control were obtained at minimal sperm to egg ratios of 1.7 x 10(6):1. Fertilization with cryopreserved semen had no influence on the embryonic development, as the ratio of embryos which stopped development and the ratio of embryonic malformations were similar to fresh semen.  相似文献

A novel sampling scheme, using a combination of electrofishing, visual exploration by scuba divers, two types of fyke nets and longlines, was tested in four reservoirs (including their inlets and outlets) to monitor a population of burbot Lota lota. This was supplemented by fry trawling and vertical hydro‐acoustics, to detect L. lota larvae in two deep reservoirs that have had a long‐term stocking programme. The majority of the L. lota detected were juveniles, captured by electrofishing in the littoral zones of the reservoirs and in running waters. Older individuals were rarely captured with longlines or fyke nets in deeper zones or structured habitats within the reservoirs. A combination of multiple sampling methods provided an assessment of the whole population. Population establishment could not be demonstrated as the age structure of the sampled fish corresponded with that of the stocked fish. Low post‐stocking survival, migratory behaviour, interactions with other species and warmer water temperatures are considered the potential drivers for unsuccessful establishment of L. lota populations in these reservoirs.  相似文献

An experimental approach was used to analyze preferences of the burbot (Lota lota) with regard to water depth, substrate type and flow velocity. In total, 30 burbots used in the experiments were caught in the middle reaches of the River Elbe by electro‐fishing. Immediately after capture, they were placed in a 180 L transportation box with aerated freshwater and taken to the research aquarium of the Zoological Museum Hamburg where they were divided into two groups according to total length: group I, 10.0–16.5 cm; group II, 20.0–30.0 cm. Prior to the experiments the fish were adapted to laboratory conditions for four weeks at 20°C in four tanks, each containing 720 L water. For each experiment, three L. lota were transferred to an additional 1350 L experimental tank. After a one‐day adaptation phase each experiment ran for three continuous day and night phases. Burbot preferences were tested for (i) water depth, (ii) substrate size, (iii) water depth and substrate size combined, and (iv) water depth, substrate and flow velocity in combination. Diurnal and nocturnal activities were recorded and analyzed throughout all experiments. Burbots of both length groups showed a significant preference for deeper water depths and large cobble substrate. Substrate type was found to be the dominant factor determining habitat preferences of L. lota when investigated in combination with water depth. In experiments combined with flow velocity, substrate and water depth were the most important factors determining burbot habitat preferences, and flow velocity of minor importance. In general, burbots were most active at night, while daytime activity was much lower. However, in all experimental approaches in the smaller length group the burbots displayed much higher day‐ and night‐time activity than did the individuals in the larger length group.  相似文献

Three different species of the genus Chloromyxum Mingazzini were found in burbot (Lota lota L.) collected in south-west Bohemia (Czechoslovakia). Comparison with existing records revealed that one species could be identified as C. pseudomucronatum Kashkovskiy, 1966. Found in the urinary bladder, it had subspherical spores with fine surface ridges and polysporic plasmodia. The two other myxosporea were established as new species: C. lenorae n. sp. was found in the kidney, renal corpuscles, renal tubules and interstitium, and had ellipsoid spores with surface ridges barely perceptible in the light microscope but clearly revealed by transmission electron microscopy. In the polysporic plasmodia, spores developed in pansporoblasts. C. reticulatum n. sp. was found in the gall bladder. It had polysporic plasmodia and spherical spores (average size 8.1 m in diameter) with a unique surface structures: elevated crests marking off irregular fields which appeared as a reticulum. In five of the fish infected with C. lenorae, bloodstream myxosporean stages of an extrasporogonic cycle were found. Further research is needed to determine whether they are stages of Sphaerospora cristata Shulman, 1962, a species also found in two of the burbot examined, or stages belonging to the Chloromyxum life cycle.  相似文献

The burbot, Lota lota, is the only freshwater species of the codfish family and has a Holarctic distribution. Pleistocene glaciations caused significant geographical differentiation in the past, but its life history characterized by winter spawning migrations over large distances is likely to homogenize populations by contemporary gene flow. We investigated the population genetic structure of 541 burbots from Lake Constance and adjacent Rhine and Danube tributaries in Europe using the entire mitochondrial DNA (mtDNA) control region and 11 microsatellites. Microsatellites revealed considerable population divergence (F(ST) = 0.26) and evidenced recent bottlenecks in two Central European rivers. In accordance to previous evidence two main phylogeographic lineages (Atlantic and Danubian) were found co-occurring at similar frequencies in Lake Constance, where they currently undergo random mating as indicated by microsatellites. The Danubian lineage contributed only a small proportion to the lake's mtDNA diversity, and probably expanded within the lake shortly after its formation approximately 10,000-15,000 BP. The larger Atlantic haplotype diversity suggested a population expansion older than the lake itself. Levels of admixture at microsatellite loci were less obvious due to their high variability, and coalescence methods were used to estimate past admixture proportions. Our results reinforce a model of a two-step colonization of Europe by burbot from an ancestral Danubian refuge, and confirm the persistence of a secondary Atlantic refuge, as proposed to exist for other freshwater fish. We conclude that the present-day burbot population in Lake Constance bears the genetic signature of both contemporary gene flow and historical separation events.  相似文献

Climatic oscillations during the Pleistocene epoch had a dramatic impact on the distribution of biota in the northern hemisphere. In order to trace glacial refugia and postglacial colonization routes on a global scale, we studied mitochondrial DNA sequence variation in a freshwater fish (burbot, Lota lota; Teleostei, Gadidae) with a circumpolar distribution. The subdivision of burbot in the subspecies Lota lota lota (Eurasia and Alaska) and Lota lota maculosa (North America, south of the Great Slave Lake) was reflected in two distinct mitochondrial lineages (average genetic distance is 2.08%). The lota form was characterized by 30 closely related haplotypes and a large part of its range (from Central Europe to Beringia) was characterized by two widespread ancestral haplotypes, implying that transcontinental exchange/migration was possible for cold-adapted freshwater taxa in recent evolutionary time. However, the derived mitochondrial variants observed in peripheral populations point to a recent separation from the core group and postglacial recolonization from distinct refugia. Beringia served as refuge from where L. l. lota dispersed southward into North America after the last glacial maximum. Genetic variation in the maculosa form consisted of three mitochondrial clades, which were linked to at least three southern refugia in North America. Two mitochondrial clades east of the Continental Divide (Mississippian and Missourian clades) had a distinct geographical distribution in the southern refuge zones but intergraded in the previously glaciated area. The aim of this study was to investigate the energy metabolism of the burbot (Lota lota, n=38) before, during, and after spawning, which represents the greatest annual metabolic demand for the species. A decrease in body mass, relative weight of the livers, and glycogen concentration of the livers was observed toward the end of spawning. The prespawning period was characterized by high rates of liver glycogenolysis and lipid mobilization. Also, plasma triiodothyronine and sex steroid levels were high before reproduction. During spawning, liver lipolysis was reduced and muscle glycogenolysis stimulated. The levels of triiodothyronine and sex steroids decreased. After reproduction, liver glycogenolysis was suppressed and the rate of gluconeogenesis increased. Thyroid hormone levels were elevated after spawning. Leptin protein and a ghrelin-immunoreactive peptide were detected in burbot plasma. Their concentrations were relatively low before and during reproduction but increased after spawning. The functions of leptin and the ghrelin-immunoreactive peptide in the physiology of the burbot are not consistent with the models of their function in mammals.  相似文献

The diet, prey selection, and growth of larval and juvenile Oneida Lake burbot, Lota lota (L.), was investigated from the time of first hatch in mid-April until their disappearance from the pelagic zone at the beginning of June. Mean length increased exponentially from 3–20 mm on 18 April to 15–00 mm on 29 May. The number and size of prey in burbot stomachs, and the prey size most selected by burbot increased as they grew. Prey width, not prey length, limited the size of prey ingested. The smallest burbot ate and selected highly for the largest prey they could ingest, but older burbot did not. As burbot grew, prey selection was highest first for the rotifer Asplanchna , followed by copepod nauplii, then cyclopoid copepods. Daphnia and calanoid copepods and cyclopoid copepods were selected to similar degrees by 29 May. For a given size, however, cyciopoid copepods were selected more strongly than any other prey type from 18 to 29 May. There were no significant differences in prey selection among offshore, nearshore, and onshore habitats, nor among depths in the offshore habitat.  相似文献
