目的观察蓝莓花色苷(blueberryanthocyanin,BBA)预处理对实验性急性心肌梗死大鼠心肌梗死面积,心肌肌钙蛋白-T(cTn-T)表达,Bax、Bcl-2mRNA表达的影响,探讨其干预心肌梗死的机制。方法40只Wistar大鼠随机分为假手术组,心肌梗死模型组,BBA低、中、高剂量组,药物干预4周,末次给药30min后结扎左冠状动脉前降支建立心梗动物模型。24h后,TTC检测心肌梗死面积;Westernblotting方法检测心肌细胞cTn-T蛋白表达;realtimePCR方法检测Bcl-2mRNA、BaxmRNA表达。结果模型组和假手术组相比,模型组心肌梗死面积显著升高(P〈0.01),心肌细胞cTn.T蛋白表达下降(P〈0.05),Bcl-2mRNA表达下降(P〈0.05),BaxmRNA表达显著升高(P〈0.01),Bcl-2/Bax比值显著降低(P〈0.01)。BBA干预给药组和模型组相比,中剂量组心肌梗死面积低于模型组(P〈0.05),低剂量组心肌细胞cTn-T蛋白表达升高(P〈0.05),中剂量组Bcl-2mRNA表达升高(P〈0.05),低、中剂量组BaxmRNA表达下降(P〈0.05),中剂量组Bcl-2/Bax比值升高(P〈0.05)。结论蓝莓花色苷对心肌梗死后心肌细胞具有明确的保护作用,其机制可能与减少心肌梗死面积,上调心肌细胞eTn-T蛋白的表达,上调Bcl-2mRNA表达、下调BaxmRNA表达,抑制心肌梗死后心肌细胞凋亡有关。  相似文献

目的:探讨间歇有氧运动对心肌梗死(MI)大鼠肾脏CD40表达的影响,揭示运动改善MI肾脏功能的可能机制。方法:36只雄性SD大鼠,随机分为假手术组(Sham)、心肌梗死组(MI)、心梗+间歇运动组(ME),每组12只。MI组采用心脏左冠状动脉前降支(LAD)结扎法,建立MI模型。Sham组大鼠实施假手术,ME组大鼠在MI手术后1周进行8周跑台运动。运动开始速度为10 m/min运动10 min后,速度逐渐增至25 m/min×7 min,再以15 m/min×3 min运动,之后依次交替进行。每天60 min×1次,每周5 d,共8周。训练结束后次日,各组大鼠评定心功能,腹主动脉取血及获取肾脏组织后,测定肾脏胶原容积百分比(CVF)、CD40、hs-CRP、TNF-α、IL-6、p-NF-κBp65、BUN和sCr等指标变化。结果:与Sham组比较,MI组大鼠LVEDP升高,LVSP和±dp/dt max显著降低,肾脏CVF升高;MI后可见肾脏肾小管细胞胞浆中CD40阳性染色,CD40蛋白和mRNA表达增多,血清及肾脏hs-CRP、TNF-α和IL-6表达升高,同时肾脏p-NF-κBp65蛋白表达增多,血清BUN和sCr表达增高。与MI组比较,ME组大鼠LVEDP降低,LVSP和±dp/dt max升高,肾脏CVF降低;肾脏CD40蛋白和mRNA表达减少,血清及肾脏hs-CRP、TNF-α和IL-6表达降低;同时肾脏p-NF-κBp65蛋白表达降低,血清BUN和sCr表达减少。结论:间歇有氧运动可显著降低心梗大鼠肾脏CD40表达,抑制NF-κB通路,减少血清及肾脏炎症因子表达,改善心梗大鼠肾脏功能。  相似文献

目的应用免疫组化技术和电生理技术记录心肌细胞/胶原复合体对心梗大鼠梗死周边区的有效不应期(ERP)及缝隙连接蛋白43(Cx43)改变,探讨梗死周边区的电偶联网络变化。方法将成年SD大鼠随机分组:假手术组、模型组、移植组。后2组制作心肌梗死动物模型,假手术组仅开胸,不结扎冠状动脉,移植组移植心肌细胞与胶原材料复合组织。结果①左室ERP变化:与假手术组相比,心梗组梗死周边区ERP显著延长(P〈0.01);移植组梗死周边区ERP延长,但较心梗阻ERP缩短,差异无显著性(P〉0.01)。②Cx43免疫组化结果:移植组Cx43阳性蛋白表达高于心梗组。结论移植的心肌细胞/胶原复合体移植可改善大鼠心肌梗死周边区缝隙连接电偶联网络,进而调控心肌细胞/胶原复合体与宿主心肌同步收缩。  相似文献

The temporal and spatial expression of transforming growth factor (TGF)-beta(1) and connective tissue growth factor (CTGF) was assessed in the left ventricle of a myocardial infarction (MI) model of injury with and without angiotensin-converting enzyme (ACE) inhibition. Coronary artery ligated rats were killed 1, 3, 7, 28, and 180 days after MI. TGF-beta(1), CTGF, and procollagen alpha1(I) mRNA were localized by in situ hybridization, and TGF-beta(1) and CTGF protein levels by immunohistochemistry. Collagen protein was measured using picrosirius red staining. In a separate group, rats were treated for 6 months with an ACE inhibitor. There were temporal and regional differences in the expression of TGF-beta(1), CTGF, and collagen after MI. Procollagen alpha1(I) mRNA expression increased in the border zone and scar peaking 1 week after MI, whereas collagen protein increased in all areas of the heart over the 180 days. Expression of TGF-beta(1) mRNA and protein showed major increases in the border zone and scar peaking 1 week after MI. The major increases in CTGF mRNA and protein occurred in the viable myocardium at 180 days after MI. Long-term ACE inhibition reduced left ventricular mass and decreased fibrosis in the viable myocardium, but had no effect on cardiac TGF-beta(1) or CTGF. TGF-beta(1) is involved in the initial, acute phase of inflammation and repair after MI, whereas CTGF is involved in the ongoing fibrosis of the heart. The antifibrotic benefits of captopril are not mediated through a reduction in CTGF.  相似文献

Our prior study had shown that resveratrol was a potent cardioprotective agent in rats with myocardial infarction (MI). In this study, we further evaluated the mechanism of cardioprotection of resveratrol by proteomic analysis. After permanent ligation of the left anterior descending artery under isoflurane anesthesia, surviving rats were randomly allocated to three groups and treated with resveratrol at 1 mg/kg/day (MI/R group), or vehicles (sham group and MI group) once daily for four weeks. In proteomic analysis, the MI group showed decreased expression of adenylate kinase 1 (AK1) and mitochondrial NADP?-dependent isocitrate dehydrogenase (IDPm) after MI compared with the sham group. These variations were reversed by resveratrol in the MI/R group. Validation with Western blot and immunohistochemical analyses showed similar trends in protein expression profiling. Our studies suggest that the beneficial effects of resveratrol on ventricular modeling may be due to increased expression of AK1 and IDPm, which have been known to increase myocardial energetic efficiency and reduce reactive oxygen species-mediated damage, respectively.  相似文献

The myocardial infarct causes prolonged activation of the renin-angiotensin system and profoundly influences cardiac performance and renal excretory capabilities. The aim of the present study was to determine whether the myocardial infarct is also associated with an altered expression of AT1a receptors (AT1aR) mRNA in the heart and the kidney. To this end male Sprague-Dawley rats were subjected either to the left coronary artery ligation or to the sham surgery. Four weeks after the surgery the animals were sacrificed. In 11 infarcted and 10 sham-operated rats expression of AT1aR mRNA in the walls of the left and right ventricle of the heart, and in the renal cortex and renal medulla was determined by semiquantitative PCR method. In another group of 10 infarcted and 14 sham-operated rats the diameter of cardiomyocytes in the left and right cardiac ventricle was determined. The size of the infarct in the rats used for mRNA determination and for morphometric measurements was equal to 29.4 +/- 1.8% and to 31.0 +/- 1.2 % of the left ventricular wall, respectively. Expression of AT1aR mRNA was significantly greater in the left (P< 0.01) and right ventricle (P<0.03) of the heart in the infarcted than in the sham operated rats. AT1aR mRNA expression was also significantly greater (P<0.02) in the renal medulla of the infarcted rats than in the renal medulla of the sham operated rats whereas no significant difference was found in the renal cortex. The myocardial infarct was associated with a significant increase of diameter of cardiomyocytes of the left ventricle of the heart (P< 0.0001), however there was no significant correlation between changes in AT1aR mRNA expression and diameter of cardiomyocytes. The results provide evidence that the myocardial infarct results in significant and prolonged upregulation of AT1a receptors mRNA expression in the heart and in the medullary region of the kidney.  相似文献

We tested whether a decrease in the ratio of interleukin-10 (IL-10) to tumor necrosis factor-alpha (TNF-alpha) correlates with the decrease in cardiac function in heart failure. It has been suggested that TNF-alpha plays a role in the progression of heart failure, and the effect of TNF-alpha in many tissues is modulated by IL-10. Any relation of these two cytokines to heart failure has never been examined. Cardiac function was assessed by echocardiographic and hemodynamic techniques in coronary artery-ligated rats at 1, 4, 8, and 16 wk after myocardial infarction (MI). Membrane-bound and soluble fractions of TNF-alpha and IL-10 proteins, the ratio of TNF-alpha to IL-10, and TNF-alpha and IL-10 mRNA levels were analyzed. Losartan was used to modify cardiac function in rats 4 wk after MI to further validate the relation between the IL-10-to-TNF-alpha ratio and cardiac function. Cardiac function deteriorated with time in all coronary artery-ligated groups, with severe failure at 16 wk after MI. Membrane-bound and soluble TNF-alpha protein fractions were increased 1 and 4 wk after MI, whereas TNF-alpha mRNA was increased 4 and 8 wk after MI. Membrane-bound IL-10 protein and mRNA levels were decreased 4, 8, and 16 wk after MI. The decrease in the IL-10-to-TNF-alpha protein ratio in all coronary artery-ligated groups correlated with the depressed cardiac function. Losartan improved cardiac function, membrane-bound and soluble TNF-alpha and IL-10 protein levels, the ratio of IL-10 to TNF-alpha, and IL-10 mRNA. This study suggests that a decrease in IL-10 and IL-10-to-TNF-alpha ratio correlates with depressed cardiac function.  相似文献

目的:研究上调血红素氧合酶1(HO-1)的表达对糖尿病心肌梗死大鼠心功能的影响及其机制。方法:将60只成年Wistar雄性大鼠随机分为5组(n=12),分别为假手术组、糖尿病组、模型组、诱导剂组、抑制剂组。心梗造模后次日开始给药,1次/周,持续6周,术后28周,应用心脏超声、经颈动脉左心室内插管术等方法观察HO-1诱导剂钴原卟啉(CoPP)及HO活性抑制剂锡中卟啉(SnMP)干预后对心室重构及心功能各项指标的远期影响;测定血糖(GLU)、总胆红素(TB)、C反应蛋白(CRP)、血清肌酐(cr)、转氨酶(ALT)等指标;采用ELISA测定白介素-6(IL-6)、肿瘤坏死因子(TNE)、一氧化氮(NO)、前列环素(PG12)、脂联素、超敏CRP(HsCRP)等水平。结果:应用CoPP上调HO-1水平,能够改善糖尿病心梗大鼠左心室压力最大变化速率、左室射血分数、左室短轴缩短率,缩小左室舒张末期内径,升高血清胆红素、一氧化氮和前列环素水平,提高心肌组织磷酸化的内皮型一氧化氮合酶(peNOS)、磷酸化的活化蛋白激酶(pAkt)、磷酸化的腺苷活化的蛋白激酶(pAMPK)的表达,降低血清TNF-α、hs-CRP水平。使用SnMP后,能够阻断CoPP的上述作用。结论:上调HO-1通过peNOS、pAMPK途径能够长期地改善血管内皮功能,抑制炎症反应,提升血清胆红素等,有效抑制心室重塑,改善梗死后糖尿病大鼠远期的心功能。  相似文献

目的:通过观察心肌肥大大鼠加速纤维肉瘤/丝裂素活化蛋白激酶激酶/胞外信号调节蛋白激酶(Raf/MEK/ERK)通路关键因子的基因和蛋白表达及蛋白磷酸化修饰水平上的变化,了解Raf/MEK/ERK通路在心肌肥大调控中的作用。方法: 20只SD大鼠随机分为假手术组和模型组,通过主动脉弓缩窄(TAC)法建立心肌肥大模型,12周后颌下静脉取血分离血清,检测氨基末端脑钠肽前体(NT-proBNP)含量,之后进行超声心动图测定和麻醉下的血流动力学测定,收集心肌标本,观察心肌组织的病理学改变,检测心肌组织Raf/MEK/ERK通路的关键因子基因、蛋白表达水平及蛋白磷酸化水平的变化。结果:与假手术组比较,TAC模型组大鼠超声心动图的左室舒张末期室间隔厚度(IVSd)、左室收缩末期室间隔厚度(IVSs)、左室后壁舒张末期厚度(LVPWd)、左室后壁收缩末期厚度(LVPWs)显著增厚(P<0.05,P<0.01),左室收缩末期内径(LVIDs)显著减小(P<0.01),左心室质量(LV Mass)、左心系数LW(LV Mass/Weight)比值显著增加(P<0.05,P<0.01);大鼠心率(HR)、左心室最大收缩速率(+dp/dtmax)、左心室最大舒张速率(-dp/dtmax) 均显著降低(P<0.01),血清中NT-pro BNP含量显著增加(P< 0.01);心肌细胞排列杂乱,心肌细胞肥大、胞质明显增多,炎症细胞浸润,出现大量胶原纤维沉积,大面积心肌细胞呈现蓝色;大鼠心肌组织中c-Raf在Ser259和Ser338上的磷酸化蛋白phospho-c-Raf (Ser259)和phospho-c-Raf (Ser338) 表达水平显著升高(P<0.01),其下游MEK1/2、ERK1/2的磷酸化蛋白phospho-MEK1/2(Ser217/Ser221)和phospho-ERK1/2 (Thr202/Tyr204)表达水平也显著增高(P<0.01)。结论: Raf/MEK/ERK通路在心肌肥大中的调控作用,可能通过激活关键因子c-Raf、MEK1、MEK2、ERK1和ERK2特异性位点的磷酸化实现的。  相似文献

Angiotensin II (AngII) is the biologically active peptide of the renin-angiotensin system (RAS). Tissue- based, local RAS has been identified in the prostate, testis, epididymis and coagulating glands. Experimental and clinical studies have consistently shown that myocardial infarction (MI) is associated with activation of the systemic RAS with increased concentration of angiotensin peptides in the blood and changes in expression of angiotensin receptors (AT). Changes in angiotensin receptors in the renal and cardiovascular system after MI are well recognized, but the effects of MI influence on changes in other tissue like the prostate gland are unknown. In the present study, we investigated the effect of myocardial infarction on angiotensin receptor protein and mRNA expression in the rat prostate gland. MI model was established in Wistar rats by ligating the left coronary artery (modified Selye method). The levels of AT1a-b and AT2 receptor mRNAs and proteins were measured in the rat prostate. Our study demonstrates tissue-specific changes in AT1a-b and AT2 receptor expression after myocardial infarction. The results show that MI has a strong influence on the expression of angiotensin receptor type AT1 in the prostate at the protein and mRNA level.  相似文献

The impact of angiotensin II receptor blockers (ARBs) on electrical remodelling after myocardial infarction (MI) remains unclear. The purpose of the present study was to evaluate the effect of valsartan on incidence of ventricular arrhythmia induced by programmed electrical stimulation (PES) and potential link to changes of myocardial connexins (Cx) 43 expression and distribution in MI rats. Fifty-nine rats were randomly divided into three groups: Sham (n = 20), MI (n = 20) and MI + Val (20 mg/kg/day per gavage, n = 19). After eight weeks, the incidence of PES-induced ventricular tachycardia (VT) and fibrillation (VF) was compared among groups. mRNA and protein expressions of Cx43, angiotensin II type 1 receptor (AT1R) in the LV border zone (BZ) and non-infarct zone (NIZ) were determined by real-time PCR and Western blot, respectively. Connexins 43 protein and collagen distribution were examined by immunohistochemistry in BZ and NIZ sections from MI hearts. Valsartan effectively improved the cardiac function, reduced the prolonged QTc (163.7 ± 3.7 msec. versus 177.8 ± 4.5 msec., P < 0.05) after MI and the incidence of VT or VF evoked by PES (21.1% versus 55%, P < 0.05). Angiotensin II type 1 receptor expression was significantly increased in BZ and NIZ sections after MI, which was down-regulated by valsartan. The mRNA and protein expressions of Cx43 in BZ were significantly reduced after MI and up-regulated by valsartan. Increased collagen deposition and reduced Cx43 expression in BZ after MI could be partly attenuated by Valsartan. Valsartan reduced the incidence of PES-induced ventricular arrhythmia, this effect was possibly through modulating the myocardial AT1R and Cx43 expression.  相似文献

目的:观察ghrelin对心肌梗死(MI)大鼠心肌重塑和心脏功能的影响,并探讨其可能的机制。方法:应用冠状动脉结扎术创建大鼠MI模型,并设立假手术组作为对照;造模成功后每天2次注射ghrelin(100μg/kg),持续4周,以此作为MI-ghrelin组,并以每天注射生理盐水的MI大鼠作为MI-生理盐水组。检测和比较各组大鼠左心室重塑和血流动力学的改变情况;非梗死心肌中白介素(IL)-1β、肿瘤坏死因子-α(TNF-α)、基质金属蛋白酶(MMP)-2、MMP-9 mRNA和蛋白的表达;梗死边界心肌细胞的凋亡情况。结果:Ghrelin可使心肌梗死后的MI大鼠降低的缩短分数(FS)、左室内压最大变化率均显著下降(dP/dtmax)、疤痕厚度明显升高,增加左室舒张末压(LVEDP)、左室收缩末内径(LVESD)、左室舒张末期内径(LVEDD)、梗死边界心肌细胞的凋亡指数显著降低。此外,ghrelin可抑制心肌梗死后的MI大鼠非梗死心肌中白介素(IL)-1β、肿瘤坏死因子-α(TNF-α)、质金属蛋白酶(MMP)-2和MMP-9的mRNA和蛋白的表达。结论:Ghrelin可缓解MI后大鼠LV功能紊乱及心室重塑,这可能与其抑制炎症反应及基质金属蛋白酶的表达有关。  相似文献

The expression of the chemorepellent Sema3a is inversely related to sympathetic innervation. We investigated whether overexpression of Sema3a in the myocardial infarction (MI) border zone could attenuate sympathetic hyper‐innervation and decrease the vulnerability to malignant ventricular tachyarrhythmia (VT) in rats. Survived MI rats were randomized to phosphate buffered saline (PBS, n = 12); mock lentivirus (MLV, n = 13) and lentivirus‐mediated overexpression of Sema3a (SLV, n = 13) groups. Sham‐operated rats served as control group (CON, n = 20). Cardiac function and electrophysiological study (PES) were performed at 1 week later. Blood and tissue samples were collected for histological analysis, epinephrine (EPI), growth‐associated factor 43 (GAP43) and tyrosine hydroxylase (TH) measurements. QTc intervals were significantly shorter in SLV group than in PBS and MLV groups (168.6 ± 7.8 vs. 178.1 ± 9.5 and 180.9 ± 8.2 ms, all P < 0.01). Inducibility of VT by PES was significantly lower in the SLV group [30.8% (4/13)] than in PBS [66.7% (8/12)] and MLV [61.5% (8/13)] groups (P < 0.05). mRNA and protein expressions of Sema3a were significantly higher and the protein expression of GAP43 and TH was significantly lower at 7 days after transduction in SLV group compared with PBS, MLV and CON groups. Myocardial EPI in the border zone was also significantly lower in SLV group than in PBS and MLV group (8.73 ± 1.30 vs. 11.94 ± 1.71 and 12.24 ± 1.54 μg/g protein, P < 0.001). Overexpression of Sema3a in MI border zone could reduce the inducibility of ventricular arrhythmias by reducing sympathetic hyper‐reinnervation after infarction.  相似文献

目的建立大鼠心肌纤维化(myocardial fibrosis,MF)模型,探讨其病变规律,为临床防治MF研究提供实验动物模型。方法 100只雄性Wistar大鼠随机分为模型组(92只)和伪手术组(8只),模型组进行心脏冠状动脉结扎(coronary artery ligation,CAL),手术后第7、14、21、28、35、42、49、56天分别处死;留取心脏标本,HE染色和Masson染色观察心肌组织基本结构,定量测定心脏组织羟脯氨酸含量、心肌胶原和转化生长因子β1(transfor-ming growth factor,TGF-β1)的表达。另设立伪手术组作为对照。结果与伪手术组组相比,模型组大鼠手术7 d后心肌组织炎性反应即已严重,心肌细胞断裂,心肌胶原含量显著升高(P〈0.01),羟脯氨酸含量升高(P〈0.05),TGF-β1表达显著增高并持续保持在较高水平(P〈0.01),纤维化反应在第42天达到高峰,其后有好转趋势。结论 CAL法能成功建立可靠的心肌纤维化动物模型,其机制可能与上调TGF-β1表达有关。  相似文献

The present study aimed to investigate the protective effects of rAAV9-CyclinA2 combined with fibrin glue (FG) in vivo in rats after myocardial infarction (MI). Ninety male Sprague–Dawley rats were randomized into 6 groups (15 in each group): sham, MI, rAAV9-green fluorescent protein (GFP)?+?MI, rAAV9-CyclinA2?+?MI, FG?+?MI, and rAAV9-CyclinA2?+?FG?+?MI. Packed virus (5?×?1011vg/ml) in 150 µl of normal saline or FG was injected into the infarcted myocardium at five locations in rAAV9-GFP?+?MI, rAAV9-CyclinA2?+?MI, and rAAV9-CyclinA2?+?FG?+?MI groups. The sham, MI, and FG?+?MI groups were injected with an equal volume of normal saline or FG at the same sites. Five weeks after injection, echocardiography was performed to evaluate the left ventricular function. The expressions of CyclinA2, proliferating cell nuclear antigen (PCNA), and phospho-histone-H3 (H3P), vascular density, and infarct area were assessed by Western blot, immunohistochemistry, immunofluorescence, and Masson staining. As a result, the combination of rAAV9-CyclinA2 and FG increased ejection fraction and fractional shortening compared with FG or rAAV9-CyclinA2 alone. The expression level of CyclinA2 was significantly higher in the rAAV9-CyclinA2?+?FG?+?MI group compared with the rAAV9-CyclinA2?+?MI and FG?+?MI groups (70.1?±?1.86% vs. 14.74?±?2.02%, P?<?0.01; or vs. 50.13?±?3.80%; P?<?0.01). A higher expression level of PCNA and H3P was found in the rAAV9-CyclinA2?+?FG?+?MI group compared with other groups. Comparing with other experiment groups, collagen deposition and the infarct size significantly decreased in rAAV9-CyclinA2?+?Fibrin?+?MI group. The vascular density was much higher in the rAAV9-CyclinA2?+?FG?+?MI group compared with the rAAV9-CyclinA2?+?MI group. We concluded that fibrin glue combined with rAAV9-CyclinA2 was found to be effective in cardiac remodeling and improving myocardial protection.  相似文献

One of the proposed mechanisms for the myocardial protective effects of heat shock (HS) treatment has been a reduction in the inflammatory response. The objective of the present study was to evaluate the impact of HS treatment in an established model of polymorphonuclear cell (PMN) migration following myocardial infarction (MI). Isolated purified PMNs (10 x 10(6) cells) labeled with (51)Cr were injected into Lewis rats following a left thoracotomy and ligation of the left anterior descending coronary artery causing MI. Two experimental groups of animals were created: MI group (n = 11) and HS+MI group (n = 7). HS treatment consisted of an elevation in core temperature to 42 degrees C for 15 min 24 h prior to MI. An additional group of control animals underwent sham thoracotomy (n = 5). All animals were euthanized at 24 h after MI, and gamma counts were obtained to estimate PMN migration. Myocardial injury was confirmed in all experimental animals (histology and echocardiography). The serum troponin I and infarct size (triphenyltetrazolium chloride) were similar in both groups. Labeled PMN migration was significantly higher in HS+MI animals (14.3 x 10(4) +/- 3.7 x 10(4) PMN) compared with MI group (9.5 x 10(4) +/- 3.6 x 10(4); P = 0.01), suggesting increased PMN migration as a result of HS treatment. HS treatment did not affect PMN migration to positive skin control sites (LPS). ICAM-1 myocardial expression was not significantly increased in HS+MI compared with MI group. In summary, HS treatment results in increased PMN migration into myocardium following MI independent of ICAM-1. These findings suggest that the proposed cardioprotective effect of HS may not be entirely due to a downregulation of myocardial inflammation as previously proposed.  相似文献

This study evaluated the effects of angiotensin-converting enzyme (ACE) inhibition after myocardial infarction (MI) on cardiac remodeling and gene expression and localization of components (ligands, receptors, and binding proteins) of the cardiac insulin-like growth factor (IGF) system. After ligation of the coronary artery, rats were randomized to vehicle or ACE inhibitor (Captopril, 50 mg/kg/day) for 4 weeks. Blood pressure, cardiac remodeling, and components of the IGF system were localized in the heart using in situ hybridization (ISH) and immunohistochemistry (IHC). The average infarct size was 42%. There were regional differences in the expression of the IGF system after MI, with increased IGF-I mRNA abundance in the border (24-fold) and infarct (12-fold) and increased IGF-binding protein (IGFBP)-3 mRNA in all areas of the failing left ventricle (threefold). Captopril reduced blood pressure, attenuated cardiac remodeling, and caused a threefold increase in IGF-I receptor mRNA and protein in infarct, border zone, and viable myocardium (p<0.01). Captopril had no effect on IGF-I mRNA but further increased IGFBP-3 mRNA and protein in the border zone, (p<0.05). The changes in the cardiac IGF system following MI are highly localized, persist for at least 4 weeks, and can be modulated by ACE inhibition. These data suggest that the benefits of ACE inhibitors in attenuation of cardiac remodeling may be mediated in part through increased expression of the IGF-I receptor sensitizing the myocardium to the positive effects of endogenous IGF-I.  相似文献
