The authors propose a complex mathematical method for analysis of the microcirculatory link of the vascular bed of muscles and muscular organs. On the basis of the data of the amount and diameters of the vessels the method permits calculating the following parameters according to the proposed pattern of the table-report and formulas: 1. the square surface of the cross-section of the vascular bed; 2. the square surface of the exchange or contact with the tissue surface of the vascular bed; 3. the density of the vascular network in percentage; 4. the blood volume in the vascular bed in volumetric units and average data; 5. the blood volume in one vessel on the average; 6. the volume of the tissue fed by one vessel; 7. the volume of blood per a unit of the surface of the vascular wall.  相似文献

The method is suggested for the quantitative characteristics of dynamic vascular tone. It allows the determination of a degree of vascular vasoconstriction as a definite value--an index. This index is determined as real area of vascular cross-sections to their maximum possible area ratio with the inner elastic membrane fully straightened. The index determined in such a way permits to assess quantitatively the degree of vascular vasoconstriction, which is independent of the vascular diameter.  相似文献

The rodent retina is perhaps the most accessible mammalian system in which to investigate neurovascular interplay within the central nervous system (CNS). It is increasingly being recognized that several neurodegenerative diseases such as Alzheimer’s, multiple sclerosis, and amyotrophic lateral sclerosis present elements of vascular compromise. In addition, the most prominent causes of blindness in pediatric and working age populations (retinopathy of prematurity and diabetic retinopathy, respectively) are characterized by vascular degeneration and failure of physiological vascular regrowth. The aim of this technical paper is to provide a detailed protocol to study CNS vascular regeneration in the retina. The method can be employed to elucidate molecular mechanisms that lead to failure of vascular growth after ischemic injury. In addition, potential therapeutic modalities to accelerate and restore healthy vascular plexuses can be explored. Findings obtained using the described approach may provide therapeutic avenues for ischemic retinopathies such as that of diabetes or prematurity and possibly benefit other vascular disorders of the CNS.  相似文献

The low-viscosity bolus method was used to determine the longitudinal distributions of vascular resistance and intravascular pressure with respect to cumulative vascular volume from the lobar artery to the lobar vein in isolated dog lung lobes near functional residual capacity under zone 3 conditions. We found that the resistance distribution had two modes, a larger one upstream and a smaller one downstream from a local minimum. Over the range of vascular pressures studied the total vascular resistance decreased and the vascular volume increased with increasing vascular pressure. However, the shape of the normalized resistance distribution was independent of vascular pressure. Comparisons of the resistance distributions with the distributions of arterial, capillary, and venous volumes suggest that the modes represent regions of relatively high resistance proximal and distal to the capillary bed. These results are consistent with the concept that within the lobar vascular bed the highest resistance per unit blood volume is in the smallest arteries and veins, as suggested by morphometric data from other sources.  相似文献

Effect of some preparatory methods to make ready earthworm tissues for electron microscopical investigation has been studied in order to reveal structural components of the vascular wall. The method for an immediate fixation of the earthworm tissues in liquid nitrogen with a subsequent additional fixation in cooled 2.5% glutar aldehyde, applied in combination with OsO4-thiocarbohydroside-OsO4 method is optimal. This method of samples preparation gives a reliable information concerning existence of an internal--endothelial--lining in the earthworm vessels. A conclusion is made on a qualitatively new level of cell differentiation of the internal vascular lining in the earthworm, which is called phase of blast transformation of endotheliocytes.  相似文献

The structure of the microcirculatory bed in the pleural villi was studied by the method of impregnation with silver nitrate, after V.V. Kuprianov. In human and animal mediastinal pleura, the microcirculatory ways are regulated within polymerously repeated complexes--moduli. A characteristic feature of the pleural villi blood supply is the presence of capillary glomeruli with a vascular peduncle consisting of an arteriole and a venule. Four types of structure are revealed in the vascular peduncle consisting of an arteriole and a venule. Four types of structure are revealed in the vascular peduncle of the pleural villi. They were studied morphometrically. Correlation in the development of the villi microcirculatory bed and that of the vascular peduncle was stated. Participation of villous vessels in the regulation of fluid balance in the pleural cavity was discussed.  相似文献

Diagnosis of retinal vascular diseases depends on ophthalmoscopic findings that most often occur after severe visual loss (as in vein occlusions) or chronic changes that are irreversible (as in diabetic retinopathy). Despite recent advances, diagnostic imaging currently reveals very little about the vascular function and local oxygen delivery. One potentially useful measure of vascular function is measurement of hemoglobin oxygen content. In this paper, we demonstrate a novel method of accurately, rapidly and easily measuring oxygen saturation within retinal vessels using in vivo imaging spectroscopy. This method uses a commercially available fundus camera coupled to two-dimensional diffracting optics that scatter the incident light onto a focal plane array in a calibrated pattern. Computed tomographic algorithms are used to reconstruct the diffracted spectral patterns into wavelength components of the original image. In this paper the spectral components of oxy- and deoxyhemoglobin are analyzed from the vessels within the image. Up to 76 spectral measurements can be made in only a few milliseconds and used to quantify the oxygen saturation within the retinal vessels over a 10-15 degree field. The method described here can acquire 10-fold more spectral data in much less time than conventional oximetry systems (while utilizing the commonly accepted fundus camera platform). Application of this method to animal models of retinal vascular disease and clinical subjects will provide useful and novel information about retinal vascular disease and physiology.  相似文献

For the injury of the lower leg associated with both bone and soft-tissue defect, the combined free flap and the Ilizarov distraction method were described as a useful treatment modality. During the procedure of distraction, however, revisions were frequently needed to change the pin position or to change the flap configuration. In case of flap ischemia, distraction should be delayed or abandoned. Then, a vascularized bone transfer might be necessary. To avoid these complications and achieve safe distraction, the configuration of the flap with its vascular pedicle should be carefully planned in terms of the future bony lengthening procedures and the concomitant soft-tissue changes of the lower leg. According to the response of local tissue to the distraction process, the lower limb can be divided into four compartments (active mobile, passive mobile, receptive, and restrictive). The configuration of the transferred free flap with its vascular pedicle can be classified into five types. To minimize the undue forces to the vascular pedicle and reduce the possibility of vascular compromise, the transferred free flap should have the configuration that its vascular pedicle lies in the territory of the mobile compartment. In performing free-tissue transfer combined with the Ilizarov method in the lower extremity, the configuration of the flap with its vascular pedicle should be carefully planned, and the characteristics of lower leg tissue should be kept in mind during the distraction.  相似文献

Long noncoding RNAs (lncRNAs) play important roles in human diseases including vascular disease. Given the large number of lncRNAs, however, whether the majority of them are associated with vascular disease remains unknown. For this purpose, here we present a genomic location based bioinformatics method to predict the lncRNAs associated with vascular disease. We applied the presented method to globally screen the human lncRNAs potentially involved in vascular disease. As a result, we predicted 3043 putative vascular disease associated lncRNAs. To test the accuracy of the method, we selected 10 lncRNAs predicted to be implicated in proliferation and migration of vascular smooth muscle cells (VSMCs) for further experimental validation. The results confirmed that eight of the 10 lncRNAs (80%) are validated. This result suggests that the presented method has a reliable prediction performance. Finally, the presented bioinformatics method and the predicted vascular disease associated lncRNAs together may provide helps for not only better understanding of the roles of lncRNAs in vascular disease but also the identification of novel molecules for the diagnosis and therapy of vascular disease.  相似文献

本文采用质外体染料示踪法研究了葡萄果实不同发育期根域限制和非根域限制(对照)处理果实维管束水分运输变化。结果表明,在葡萄果实发育的第一次快速生长期,根域限制和对照处理的果实周缘维管束被染色数量最多,染料溶液在葡萄果实中的运输速率也最高,分别为1.72和1.63cm·h-1,根域限制处理果实中周缘维管束和中央维管束中染料溶液的运输速度低于对照处理,但胚珠维管束中染料运输速度要高于对照,为1.32cm·h-1;进入生长停滞期后,根域限制和对照处理的葡萄果实中周缘维管束被染色数目降低,染料溶液在中央维管束中的运输速度分别降低为0.72和0.70cm·h-1;在果实的第二次快速生长期,根域限制和对照处理果实周缘维管束染色数目仍然低于果实发育的第一次快速生长期,根域限制果实周缘维管束染色范围和染料运输速率都高于对照处理。  相似文献

Previous studies showed that 20 Gy whole-body gamma irradiation results in a decreased response of the abdominal aorta to the stable thromboxane A2 (TXA2) mimic, U46619. The present study evaluated the effect of WR2721 on this radiation-induced decrease in vascular responsiveness. Rats receiving WR2721 (200 mg/kg, i.p.) 20 min before irradiation showed no depression in vascular reactivity to U46619 compared to control. The abolition of the radiation-induced decrease in vascular responsiveness was not caused by a direct vasoconstrictor action of WR2721 or its metabolites. The vascular response of rat abdominal aortic rings to KCl was unchanged after in vivo exposure to ionizing radiation. WR2721 did not alter the vascular response to KCl. These studies confirm that exposure to whole-body ionizing radiation decreased abdominal aortic vascular responsiveness to U46619. This depressed vascular reactivity can be abolished by pretreatment with the radioprotectant, WR2721. These observations may provide a rapid initial screening method for evaluating the in vivo efficacy of radioprotectant drugs.  相似文献

1H magnetic resonance microtomography imaging was applied to study vascular systems in developing bean (Phaseolus limensis L.) seeds. Using the gradient echo method, we recorded 2D tomographic sections in the sagittal and axial planes of the fruits sampled from a vegetating plant on days 10, 17, 24, and 31 after fertilization. Any vascular connection between the tissues of maternal plant (bean pod and seed coat) and the embryo were undetectable. The embryo has an autonomous branched network of procambial strands in the cotyledons, converging to the embryonic axis. The bean pods are covered with a network of vascular bundles; large vascular strands run along the dorsal and ventral sutures. The seed coat vascular bundles are formed in the process of seed ripening and are represented by a developed vascular system multiply branching in the middle part of the ground parenchyma at the stage of physiological maturity. They are connected with the source of assimilates via the lateral pod veins and a large vascular bundle, entering the seed below the hilum via the placenta. Assimilates enter the external part of the seed coat, which contains no vascular bundles, via the funiculus vascular bundles and hilum tissue.  相似文献

The microsphere method has been used extensively to measure regional blood flow in large laboratory animals. A fundamental premise of the method is that microspheres do not alter regional flow or vascular tone. Whereas this assumption is accepted in large animals, it may not be valid in the pulmonary circulation of smaller animals. Three studies were performed to determine the hemodynamic effects of microspheres on the rat pulmonary circulation. Increasing numbers of 15-microm-diameter microspheres were injected into a fully dilated, isolated-lung preparation. Vascular resistance increased 0.8% for every 100,000 microspheres injected. Microspheres were also injected into an isolated-lung preparation in which vascular tone was increased with hypoxia. Microspheres did not induce vasodilatation, as reported in other vascular beds. Fluorescent microspheres were injected via tail veins into awake rats, and the spatial locations of the microspheres were determined. Regional distributions remained highly correlated when microspheres of one color were injected after microspheres of another color. This indicates that the initial injection did not alter regional perfusion. We conclude that, when used in appropriate numbers, 15-microm-diameter microspheres do not alter regional flow or vascular tone in the rat pulmonary circulation.  相似文献

A new method to salvage severed composite-tissue segments (denuded hindleg) by vascular implantation associated with subcutaneous burying was successfully undertaken in rabbits. The implantation of vascular bundle can preserve more viability of the fresh segment than burying alone and serves as the vascular pedicle for later transplantation. A devitalized segment can also be vascularized by this method, although no survival or regeneration of preexisting tissues could be found.  相似文献

An iterative method of calculating propagation parameters at harmonics of heart rate for a uniform vascular segment from a combination of four arterial waveform measurements is presented. Measurements of blood pressure, vascular diameter, and blood flow-rate may be combined arbitrarily provided only that at least one measurement of pressure and one of flow-rate be included; the requirement of four measurements implies at least two measurement sites along the vessel. The analysis is thus a generalization of those associated with previous methods of determining propagation parameters, allowing for instance relaxation of the requirement of equal spacing in the three-point method. Results are presented for the propagation of an impulse along a rubber tube when the measurements are pressure at two sites, flow-rate and diameter.  相似文献

We have used confocal microangiography to examine and describe the vascular anatomy of the developing zebrafish, Danio rerio. This method and the profound optical clarity of zebrafish embryos make it possible to view the entire developing vasculature with unprecedented resolution. A staged series of three-dimensional images of the vascular system were collected beginning shortly after the onset of circulation at 1 day postfertilization through early- to midlarval stages at approximately 7 days postfertilization. Blood vessels in every region of the animal were imaged at each stage, and detailed "wiring patterns" were derived describing the interconnections between every major vessel. We present an overview of these data here in this paper and in an accompanying Web site "The interactive atlas of zebrafish vascular anatomy" online at (http://eclipse.nichd.nih.gov/nichd/lmg/redirect.html). We find a highly dynamic but also highly stereotypic pattern of vascular connections, with different sets of primitive embryonic vessels severing connections and rewiring in new configurations according to a reproducible plan. We also find that despite variation in the details of the vascular anatomy, the basic vascular plan of the developing zebrafish shows strong similarity to that of other vertebrates. This atlas will provide an invaluable foundation for future genetic and experimental studies of vascular development in the zebrafish.  相似文献

The viability of axial pattern skin flaps in pigs was assessed by the use of intravenous fluorescein, intradermal injection of 133Xe in saline, intravenous 51Cr tagged red cells, and angiography. The results were correlated with flap survival at 4 days postoperatively. Intravenous fluorescein provided the most accurate method for prediction of viable tissue at the time of operation. There was no evidence of vascular perfusion in the distal portions of these flaps. These axial pattern flaps differed in their viability from similar flaps in humans, and anastomoses between discrete vascular territories were infrequent in pigskin.  相似文献

A combined method for clearing soft tissues, staining cartilage and bone, and injecting the vascular system of small mammals was developed using Mus musculus (house mouse). Mammalian muscle tissue remains milky or even opaque after "clearing" by previous techniques due to the relatively high content of intramuscular fat. A method employing chloroform-ethanol successfully renders soft tissues of mammalian specimens translucent without damaging or bleeding color from the latex injected in the circulatory system. Resulting specimens yield an excellent view of the skeletal system and the injected vascular system without obstruction by opaque tissues or disruption by physical removal of connective tissue.  相似文献

A method is presented for the introduction and removal of cyroprotective agents from kidneys, utilizing the principle of gradual addition and elution during continuous perfusion. The method differs from those described previously in that it utilizes a standard perfusion machine (Water MOX 100) and a motorized syringe pump; both of which are widely available in laboratories involved in clinical and experimental organ preservation. In addition, a simple method for measuring vascular resistance is described which relies on a comparison of flow rates through the perfused organs to that through a fixed resistance. The utility of this approach is illustrated by studies of organ function and vascular resistance.  相似文献
