Growth and proteinase production by Micrococcus sp. INIA 528 in a batch-operated laboratory fermentor were investigated, with trypticase soy broth as the basal medium for studies on optimum temperature, pH and medium composition. Maximum growth was recorded at 34°C and pH 715, whereas optimum temperature and pH for proteinase production were 31°C and pH 6.25. Maximum rate of enzyme production occurred during the late log and early stationary phases of growth. Addition of 5.0 g 1-1 yeast extract, 1.0 g 1-1 glucose, 1.0 g 1-1 MgSO4 or 1.0 g 1-1 K2HPO4 to basal medium resulted in a lower enzyme yield, but supplementation of basal medium with 2.5 g 1-1 (NH4)2SO4 increased enzyme production by 45%. A high initial biomass added to fresh broth supplemented with 2.5 g 1-1 (NH4)2SO4 only increased enzyme activity by 19%, compared to the maximum enzyme activity achieved with the standard inoculum.  相似文献

Bavaricin MN, a bacteriocin produced by Lactobacillus bavaricus MN, reached titres of 2000 AU ml-1 in APT broth maintained at pH 6.0, 30°C in a batch fermenter. Levels of bavaricin MN at pH 5.5 and 6.5 were lower despite comparable levels of producer cells. The addition of 3.0 g l-1 beef extract to APT broth resulted in increases in both the growth rate of the culture and the production of bavaracin MN. The titre of bavaricin MN in batch fermenters controlled at pH 6.0 in APT broth plus 3 g l-1 beef extract reached 3200 AU ml-1 at 30°C. This level was reduced to 800 AU ml-1 by 76 h. Glucose-limited continuous culture of Lact. bavaricus MN under the same conditions resulted in an increase in the titre of bavaricin MN to 6400 AU ml-1. This level was maintained, independent of growth rate, for 345 h. Growth rates of 0.205, 0.118, 0.169 and 0.058 h-1 were examined.  相似文献

A 24 h screen to detect viable salmonellas in faeces was developed by studying growth dynamics of salmonellas and competing flora in combinations of enrichment media and artificially-inoculated pig faeces. Muller-Kauffmann tetrathionate (MK) broth, incubated overnight at 42°C, maintained the lowest ratio of salmonella: competing flora and identified all inoculated samples. A 4 h postenrichment in M broth plus novobiocin reduced the number of false-positive results in subsequent ELISAs. Adjusting the negative cut-off values and incubation time of the chromogenic substrate from that recommended in the ELISA instructions reduced the rate of false-positive results further and allowed the detection of 103 salmonellas per ml in the presence of up to 107 ml−1 aerobic-competing cells. Suspension of faeces diluted 1 in 2 and 1 in 5, rather than 1 in 10 in MK broth did not necessitate further adjustments to the ELISA baseline values. The proposed screen protocol is an overnight incubation of faeces suspended 1 in 10 in MK broth, a 1 in 100 subculture into M broth plus 10 μg ml−1 novobiocin (MbN) for 4 h, steam inactivation of MbN cultures and testing by ELISA, and can detect three salmonella cells per g faeces.  相似文献

Abstract The effects of O2 tension, temperature, salt concentration and organic matter concentration on the growth and nitrifying activity of Nitrosomonas N3 isolated from Tay Estuary sediments have been investigated. Chemostat-grown cultures were able to grow and nitrify at dissolved O2 concentrations as low as 0.1 mg O2· 1−1 (cell population densities were 15% of those obtained in fully aerated cultures). This bacterium was sensitive to reduced temperatures as chemostat-grown cultures washed out at growth temperatures below 15°C, at dilution rates > 0.025 · h−1. Batch-grown cultures of Nitrosomonas N3 were used to study the effects of NaCl and complex organic matter concentration on nitrifying activity. Maximum rates of NH+4 oxidation were recorded at NaCl concentrations of 1% w/v, whilst tryptone soya broth (TSB), nutrient broth (NB), yeast extract broth (YEB) and peptone were inhibitory at concentrations > 10 mg · 1−1.  相似文献

By reducing the concentration of nitrogen (from 5.0 to 2.5 mmol 1-1), batch cultures of Xanthomonas campestris induced the enzyme UDP-glucose dehydrogenase and stimulated the Entner-Doudoroff pathway enzyme glucose-6-P dehydrogenase. The surplus energy generation was directed to xanthan biosynthesis resulting in a 10% polysaccharide increase. The nitrogen restriction led to a higher consumption of nitrogen (93%) whereas glucose consumption did not surpass 75% utilization. Low concentrations of both magnesium and sulphur exerted a negative effect on xanthan formation. Both restrictions reduced the phosphomannose isomerase enzyme activity by 10-fold turning the mannose transference presumably into the rate-limiting step for xanthan biosynthesis. Conversely, the rate of synthesis of glucuronic acid residues did not affect the rate of xanthan biosynthesis. Polysaccharide synthesis in magnesium and sulphur cultures was negatively affected in comparison with cell formation as the cell volumetric production rate increased from 0.037 to 0.091 g 1-1 h-1 and the xanthan volumetric production rate dropped from 0.133 g 1-1 h-1 to the minimum obtained at 0.083 g 1-1 h-1. The efficiency of the carbon substrate conversion was also greatly changed.  相似文献

The effect of MRS broth on the stability of hydrogen peroxide (H2O2) has been studied. Known concentrations (1–100 μg ml−1) of H2O2 were prepared in distilled water, phosphate buffer (pH 7·0) and MRS broth (pH 6·2 and 3·9). H2O2 was very stable in aqueous and buffer solutions but it was rapidly degraded in MRS broth (pH 3·9). The presence of H2O2 in MRS broth (pH 6·2) could not be detected.  相似文献

Fifty-one essential oils extracted from plants of known origin were tested for their antimicrobial activity against three bacteria, Pseudomonas aeruginosa , Staphylococcus aureus , Escherichia coli and four yeasts, Torulopsis utilis , Schizosaccharomyces pombe , Candida albicans and Saccharomyces cerevisiae using the drop diffusion method. All showed antimicrobial activity against at least one of the micro-organisms. Following this preliminary screening, 13 essential oils showing antimicrobial activity against at least five of the micro-organisms were tested in the range 50 μg ml−1 to 500 μg ml−1 using broth micro dilution techniques with dimethylsulphoxide (DMSO) as a dispersing solvent. The concentration of most of the oils required for total inhibition of growth was >500 μg ml−1. Further studies on the antimicrobial action of cinnamon oil in the range 10–150 μg ml−1 showed that 50-fold higher activity was found when no dispersing solvent was used.  相似文献

Abstract: Changes of the main hydrodynamic and oxygen transfer parameters during Aspergillus niger cultivation in an external-loop air-lift bioreactor of 200 dm3 operating capacity were investigated. The final average concentrations of biomass and citric acid obtained in batch fermentations were about 17 g 1-1 and 90 g 1-1, respectively. Significant influence of the increasing biomass concentration on the rheological properties of the broth and operating parameters was found. Enterobacter agglomerans isolated from a soil sample collected in the vicinity of a sugar house was grown on low-grade maple sap: When cultivated in shake flask, Ent. agglomerans produced exopolysaccharide and α-ketoglutarate. No polysaccharide and no α-ketoglutarate were produced during growth at dissolved oxygen concentrations lower than 1% saturation in a 3.5 1 bioreactor, but succinate, lactate and formate were accumulated. In order to increase polysaccharide production, various agitation systems were adapted to 7 1 bioreactors with working volumes of 41. Highest polysaccharide yield (Yp/s= 0.70) and biomass yield (Yx/s= 0.17) were obtained with bioreactors agitated with four-and six-bladed Rushton turbines. Accumulation of mixed organic acids during polysaccharide production indicated that both aerobic and anaerobic conditions prevailed inside the bioreactors. Maple sap concentration higher than 60 g 1−1 (as sucrose equivalent) decreased both growth and exopolysaccharide production.  相似文献

Synthetic chlorochromate derivatives of pyridine and quinoline were active in vitro against type cultures of Escherichia coli (ATCC 128), Staphylococcus aureus (ATCC 14775), Pseudomonas aeruginosa (ATCC 10145) and Bacillus subtilis (NCTC 8236). The minimum inhibitory concentrations (MIC) were 125–250 μg ml−1 and 250–500 μg ml−1 for pyridinium chlorochromate and quinolinium chlorochromate, respectively. An established derivative of quinoline (Perfloxacin) had an MIC of 125–250 μg ml−1. The extinction time for 105 cfu in broth was 90 min for pyridinium chlorochromate and 120 min for quinolinium chlorochromate, except for B. subtilis which survived up to about 180 min and 360 min. A combination of the two compounds produced an antagonistic effect. The 50% lethal dose (LD50 toxicity) in mice was estimated at 76 μg g−1 and 33 μg g−1 body weight for the quinolinium and pyridinium chlorochromates. The compounds also exhibited some potential for suppressing a simulated staphylococcal infection in mice at the dosage levels of ca 22 μg g−1 for pyridinium chlorochromate and 45 μg g−1 for quinolinium chlorochromate.  相似文献

Field experiments on the use of recommended and reduced application rates of aphicides for control of summer aphid infestations on winter wheat have been done at five sites each year from 1990 to 1992. The experiment compared sprays of pirimicarb at recommended rate (140g a.i. ha-1) and reduced rate (25 g a.i. ha-1) and of alphacypermethrin at recommended rate (15 g a.i. ha-1) and one third recommended rate (5 g a.i. ha-1), each applied either at flag leaf emergence or at flowering. Aphid infestations were allowed to develop naturally and were measured at critical growth stages. In 1990, overwintered aphid infestations, together with established populations of parasitoids, were detected at four of the five sites when the first assessment was made in April. Parasitoids provided sufficient aphid control to prevent yield loss at two sites, without pesticide applications. At three sites both aphicides gave similar levels of control and yield response at both recommended and reduced application rates. In 1991 and 1992 no overwintered aphids were detected in the crops and aphid infestations developed much later. Aphicide applications gave significant yield responses at four sites in 1992. Recommended rates gave better control of aphids than reduced rates. There was no significant difference in the yield response obtained with different rates of aphicide.  相似文献

This study describes a broad host transformation protocol that enables the uptake of plasmid DNA into 10 different species of Bifidobacterium , some of which have never been transformed before. The vector pNC7 (4·9 kb) was used to optimize the electroporation protocol. Transformation efficiencies ranged from 3·6×10−1 to 1·2×105 transformations per μg DNA. The impact of growth medium composition and electric field strength on transformation efficiency were independently optimized. Electrocompetent cells were grown in Iwata medium broth enriched with ActilightRP 16%, harvested during the early exponential growth phase, and pulsed at 12·5 kV cm−1, 100 Ω and 25 μF.  相似文献

The antimicrobial activities of bovine lactoferrin (bLF), its pepsin hydrolysate (bLFH) and the active peptide lactoferricin® B (LFcinB) against four clinical isolates of enterohaemorrhagic Escherichia coli O157:H7 were studied. The MICs against these isolates were 3 mg ml−1 for bLF, 0·1–0·2 mg ml−1 for bLFH and 8–10 μg ml−1 for LFcinB in 1% Bactopeptone broth. LFcinB killed these bacteria within 3 h at concentrations above 10 μg ml−1. Transmission electron microscopy findings suggested that LFcinB acts on the bacterial surface and affects cytoplasmic contents. LFcinB was shown to influence the levels of verotoxins in the culture supernatant fluid of an E. coli 0157:H7 strain. These results demonstrate that E. coli O157:H7 strains are susceptible to the antimicrobial effects of bLF and its peptides.  相似文献

The growth of a strain of Rhizobium trifolii and of R. meliloti was studied in broth and peat cultures to determine the relative toxicity of Na+ and Cl-. The following salts were added in a range of concentrations: Na2HPO4 as a source of Na+, CaCl2.2H2O as a source of Cl-, and NaCl. Disodium hydrogen orthophosphate affected the growth rate of both strains in broth culture but not in peat culture. Unexpectedly, calcium chloride was more toxic than NaCl in broth and peat culture. The toxicity of NaCl can be ascribed to the Cl-. Rhizobium meliloti strains grew on 3·5% NaCl after adaptation during a long period. Rhizobia for soya bean and cowpea grew at 0·5% NaCl and those for clover and pea, at 1·0% NaCl.  相似文献

Abstract Growth of Pyrococcus furiosus was studied in batch cultures with cellobiose, maltose and pyruvate as limiting substrates. Fermentation mass balances of all conversions were complete. These data suggested that the pathway for conversion of the disaccharides is essentially the same. The molar growth yields on maltose and cellobiose were about equal (±50 g cell dry weight (cdw) per mol disaccharide). The molar growth yield on pyruvate was ±6.3 g cdw mol−1. Growth yields were influenced by the hydrogen partial pressure. When P. furiosus was co-cultured with a methanogen a yield of 56 g cdw mol−1 disaccharide and 8.6 g cdw mol−1 pyruvate was obtained. When the data were interpreted according to the proposed pyroglycolytic pathway, the calculated YATP values were different for the various pH2 conditions, suggesting that an additional energy conserving site may be present in the pathway.  相似文献

Abstract Attempts to study bacterial resistance to metal ions in broth media can be complicated by precipitation reactions. Precipitation from Luria-Bertani (LB) broth occurred at Cd2+ concentrations higher than 1.6 mM and was complete within about 10 min. The precipitate obtained after addition of 3 mM Cd2+ was analysed by elemental analysis. 1H and 31P nuclear magnetic resonance (NMR) spectroscopy. It consisted of cadmium phosphates together with organic material containing valine- and glutamate-rich polypeptides. Precipitation significantly reduced the levels of the essential trace elements Fe and Zn in the growth medium.  相似文献

A reappraisal of oxygen uptake by Sarotherodon mossambicus was undertaken using a continuous flow respirometer. Measurements were obtained over the temperature range 16°C–37°C for fish weighing between 10 g and 150 g. The metabolism of low-molecular-weight thiols was investigated in seeds of Pisum sativum L. cv. Kleine Rheinländerin during imbibition in water for 14 h. The amount of oxidized glutathione (GSSG) decreased from 319 nmol (g dry weight)−1 in dry seeds to 38 nmol (g dry weight)−1 within the first 14 h of imbibition. The decrease may have been due to the reduction of GSSG to reduced glutathione (GSH), catalyzed by the enzyme glutathione reductase (GR; EC 1.6.4.2). The enzyme activity was high in dry seeds [25 nkat (g dry weight)−1] and decreased to 20 nkat (g dry weight)−1 within 14 h of imbibition. The activity of glucose-6-phosphate dehydrogenase (EC 1.1.1.49) decreased from 100 nkat (g dry weight)−1 in dry seeds to 67 nkat (g dry weight)−1 after 14 h of imbibition. Within 14 h the amount of γ-glutamyl-cysteine (γ-GC) decreased from 135 to 38 nmol (g dry weight)−1, whereas the cysteine content rose from 81 nmol (g dry weight)−1 in dry seeds to a maximum of 170 nmol (g dry weight)−1 after 12 h of imbibition, which may be due to the degradation of γ-GC into cysteine.  相似文献

A fungal strain (S33-2), able to grow on cooked starch and produce a substantially high level of kojic acid, was isolated from morning glory flower ( Bixa orellana ). The fungus was characterized and identified as Aspergillus flavus. The effect of different types of starch (sago, potato and corn starch) on growth of strain S33-2 and kojic acid production was examined using shake flasks. It was found that strain S33-2 grew well on all types of starch investigated. However, kojic acid production was highest when corn starch was used, with the maximum kojic acid obtained being comparable to fermentation using glucose. The highest kojic acid production (19·2 g l−1) was obtained when 75 g l−1 corn starch was used. This gave a yield, based on starch consumed, and an overall productivity of 0·256 g g−1 and 0·04 g l−1 h−1, respectively.  相似文献

Nitrogen fixation activity was determined for Lotus tenuis. Medicago lupulina and Trifolium pratense . The three species grew in clones in grassland in an area reclaimed from brackish water in the 1940s. The N2[C2H2]-fixation was measured in soil cores throughout 1974 and 1975. From cores taken in dense and uniform stands of the species, the yearly N2[C2H2]-fixation at maximum cover was estimated. L. tenuis fixed about 4 g N m−2 yr−1 (area with max. cover 130%), i.e. 30–56% of its requirement. Both M. lupulina and T. pratense fixed about 7 g N m−2 yr−1 (maximum cover 37% and 80%) i.e. 67% of their N-requirement. Average N2[C2H2]-fixation for the whole area was 0.4 g N m−2 yr−1, considerably less than the N-addition through rainfall.  相似文献
