蓖麻毒素是免疫毒素的重要组成之一。本文采用硫酸胺盐析法,从蓖麻籽匀浆液中直接制备粗毒。然后,在酸化Sepharose 4B柱上用半乳糖溶液梯度洗脱。由此得到分子量为65000道尔顿的电泳纯蓖麻毒素。其小鼠LD50为0.53μg,M olt-4细胞毒性试验IC为3.8×10-10M与以前报道用改良Olsnes法所得结果相近。但操作简便、得率提高近5倍。  相似文献

【背景】猪链球菌4型(Streptococcus suis serotype 4,SS4)分离率日益升高,对养殖业和公共卫生安全造成严重危害,目前尚无有效的SS4疫苗。【目的】筛选致病力强、抗原性好、遗传性状稳定的SS4疫苗菌种。【方法】以7株SS4分离株(代号为A1—A7)为受试菌株,通过累积法测定菌株半数致死量(LD50),ELISA测定免疫小鼠血清中IgG效价,攻毒保护试验测定免疫保护率,并采集小鼠脏器观察病理组织学变化。再连续传代培养受试菌株,分别对第10、20、30代菌株进行致病性和抗原性试验。【结果】A1—A7菌株对小鼠的LD50分别为2.19×108、1.76×108、1.83×108、1.01×108、4.05×108、1.19×108和9.03×107 CFU。二免7 d后,A1、A2、A3、A4、A6、A7免疫组IgG效价分别为1:1 600、1:1 600、1:3 200、1:6 400、1:3 200和1:6 400,免疫保护率分别为30%、30%、50%、70%、60%和80%,而且A4、A7免疫组小鼠组织病变较其余4组轻微。体外传至30代后,A4菌株的LD50上升至3.81×108 CFU,IgG效价下降至1:1 600,免疫保护率下降至40%,而A7菌株的LD50上升至2.49×108 CFU,IgG效价和免疫保护率稳定保持为1:6 400和80%,而且A7免疫组小鼠的组织病变较A4免疫组轻微。【结论】A7菌株(原始编号为HBgu18-4)具有强致病力和良好的抗原性,而且遗传性状均一、稳定,可作为SS4制苗候选菌株。  相似文献

采用酸处理Sepharose 6B亲和层析和SephacrylS-200凝胶过滤,首次从中药北寄生中分得一种有毒凝集素,称为北寄生凝集素(Viscum coloratum lectin,VCL),是一种高毒性的植物毒蛋白,对小鼠静脉给药的LD50为4.4ug/kg,其粗提物也具有很强的毒性,对小鼠腹腔给药的LD50为0.2g/kg,分子量52 000,等电点10.5。  相似文献

采用一种改进的方法,简便快速地从丝瓜(Luffa cylindrica)籽中得到丝瓜蛋白α和β.它们在SDS-PAGE上均呈一区带,其分子量分别为28000和29000.等电聚焦测定等电点均为10.它们对无细胞体系蛋白质合成都有强烈的抑制活性,其ID50分别为10μg/L和50μg/L,是目前发现的单链核糖体失活蛋白中活性最高的.  相似文献

The effects of tridemorph and vinclozolin were studied on different types of microorganisms, urea hydrolysis and nitrification in soil and in culture. The fungicides adversely affected the population of bacteria, fungi and actinomycetes as a function of time of incubation. Urea hydrolysis both in culture and soil were also inhibited by the fungicides, and tridemorph was more detrimental. In soil, 45μg/g of tridemorph inhibited 50 % of ammonification of urea, ID50 for nitrite production was 750μg/g. In urea-hydrolyzing cultures, 80, 75 and 77 mg/L of tridemorph were the ID50 values for urea hydrolysis byMicrococcus sp.,Proteus sp. andP. vulgaris respectively.  相似文献

为研究蝎毒素BmkTXKβ对家兔心房肌细胞瞬间外向钾电流(Ito)的影响,采用全细胞膜片钳技术记录应用BmkTXKβ前后的Ito电流. 结果显示BmkTXKβ(1 μmol/L)使心房肌细胞Ito(刺激电压为+50 mV)从(13.63±0.87)pA/pF减少到(7.98±0.78)pA/pF,抑制率为41.4% (n=16, P<0.001).冲洗后, Ito部分恢复至(11.18±0.82)pA/pF (n=6, P<0.01,与给药后比较).在0.01~100 μmol/L范围内BmkTXKβ呈浓度依赖性地抑制Ito,IC50的均值为0.95 μmol/L (n=10, P<0.01),但无频率依赖性(n=6,P>0.05). 1 μmol/L的BmkTXKβ可使Ito通道的失活动力学曲线明显左移,V1/2分别为(-23.6±2.7) mV和(-35.3±3.6) mV(n=8,P<0.05),曲线斜率基本不变.同时可使Ito通道的恢复过程明显减慢,恢复曲线右移,τ值从(51.2±8.5) ms延长至(93.5±13.4) ms(n=9,P<0.01),但不影响其激活过程.据此推断BmkTXKβ对家兔心房肌细胞Ito具有显著的抑制作用,主要作用于失活过程,延长该通道的恢复时间.  相似文献

采用盆栽试验方法,探讨了香樟(Cinnamomum camphora)凋落叶不同土壤添加水平(0 g/盆为CK、25 g/盆、50 g/盆,100 g/盆)对受体作物小白菜(Brassica chinensis)、莴笋(Lactuca sativa)幼苗生长和光合特性的影响。结果显示:(1)香樟凋落叶分解对两种作物的地径、株高、生物量、叶片数和叶面积均有明显的抑制作用,且抑制效应随凋落叶添加量的增加而增强,但随着分解时间的延长其抑制作用逐渐减弱甚至表现为促进作用。(2)香樟凋落叶分解对两种作物的光合色素含量均有明显的抑制效应,并随凋落叶添加量的增加抑制作用增强,且持续时间延长。(3)经凋落叶处理的两种作物叶片净光合速率(Pn)、气孔导度(Gs)、蒸腾速率(Tr)、水分利用率(WUE)总体上均低于CK,而胞间CO2浓度(Ci)在各凋落叶处理下均高于CK。(4)随着土壤中凋落叶量的增加,两种作物在光饱和以及CO2饱和状态下的最大净光合速率(Pn max)、表观量子效率(AQY)、羧化速率(CE)、光呼吸速率(Rp)和暗呼吸速率(Rd)均不断下降,而光补偿点(LCP)、光饱和点(LSP)、CO2饱和点(CSP)、CO2补偿点(CCP)因受体作物的不同,表现出不同的变化趋势。研究表明,土壤中香樟凋落叶分解释放的化感物质,能通过降低受体作物的光合色素合成和光合能力,限制其营养生长,最终影响生物量积累;相对于莴笋,小白菜对香樟凋落叶分解产生的化感胁迫效应具有更强的耐受性,可能更适宜在香樟林间种植。  相似文献

选取桂林喀斯特石山生境中常见的50种植物为研究对象,分别测定叶片单位面积最大净光合速率(Aarea)、单位质量最大净光合速率(Amass)、气孔导度(Gs)、水分利用效率(WUE)、胞间/环境CO2浓度比值(Ci/Ca)和蒸腾速率(Tr)等光合特性指标,探讨不同物种光合特性的差异以及光合特性之间的内在联系,以此探究不同植物适应喀斯特石山生境所表现出的光合生理特性。结果表明,50种植物叶片Aarea,Amass,Gs,WUE,Ci/Ca和Tr的平均值分别为8.35 μmol m-2 s-1,110.98 nmol g-1 s-1,0.10 mol m-2 s-1,94.84 μmol/mol,0.57和2.37 mmol m-2 s-1;方差分析表明,不同物种之间在Aarea,Amass,Gs,WUE,Ci/CaTr之间存在显著差异。Pearson相关性分析表明,表征50种常见植物叶片光合特性的6个指标相关性除Ci/CaAareaAmass,WUE与Amass不一致外,其他指标两两之间相关性均表现为一致性,其中GsCi/Ca呈极显著的正相关。主成分分析表明,在6个光合特性指标中,GsCi/Ca可作为反映喀斯特石山植物适应生境的重要光合指标,主要表征对水分条件的敏感程度以及耐旱性强弱,同时反映了植物叶片光合速率大小,用于衡量植物对喀斯特生境的生理生态适应性。基于GsCi/Ca进行聚类分析表明,50种植物划分为3类:即中等Gs较高Ci/Ca型,较低Gs较高Ci/Ca型和较低Gs,Ci/Ca型。本研究表明,喀斯特生境植物在生理生态方面所表现出的适应策略主要为对资源利用方式及抵御外界不利环境的适应策略,这为后续选择物种加速植被恢复演替进程提供了参考。  相似文献

It has now been over twenty years since a novel herpesviral genome was identified in Kaposi's sarcoma biopsies. Since then, the cumulative research effort by molecular biologists, virologists, clinicians, and epidemiologists alike has led to the extensive characterization of this tumor virus, Kaposi's sarcoma-associated herpesvirus(KSHV; also known as human herpesvirus 8(HHV-8)), and its associated diseases. Here we review the current knowledge of KSHV biology and pathogenesis, with a particular emphasis on new and exciting advances in the field of epigenetics. We also discuss the development and practicality of various cell culture and animal model systems to study KSHV replication and pathogenesis.  相似文献

Comprises species occurring mostly in subtidal habitats in tropical, subtropical and warm-temperate areas of the world. An analysis of the type species, V. spiralis (Sonder) Lamouroux ex J. Agardh, a species from Australia, establishes basic characters for distinguishing species in the genus. These characters are (1) branching patterns of thalli, (2) flat blades that may be spiralled on their axis, (3) width of the blade, (4) primary or secondary derivation of sterile and fertile branchlets and (5) position of sterile and fertile branchlets on the thalli. Application of the latter two characters provides an important basic method for separation of species into three major groups. Osmundaria , a genus known only in southern Australia, was studied in relation to Vidalia , and its separation from the Vidalia assemblage is not accepted. Species of Vidalia therefore are transferred to the older genus name, Osmundaria. Two new species, Osmundaria papenfussii and Osmundaria oliveae are described from Natal. Confusion in the usage of the epithet, Vidalia fimbriala Brown ex Turner has been clarified, and Vidalia gregaria Falkenberg, described as an epiphyte on Osmundaria pro/ifera Lamouroux, is revealed to be young branches of the host, Osmundaria prolifera.  相似文献

Fifteen chromosome counts of six Artemisia taxa and one species of each of the genera Brachanthemum, Hippolytia, Kaschgaria, Lepidolopsis and Turaniphytum are reported from Kazakhstan. Three of them are new reports, two are not consistent with previous counts and the remainder are confirmations of very scarce (one to four) earlier records. All the populations studied have the same basic chromosome number, x = 9, with ploidy levels ranging from 2x to 6x. Some correlations between ploidy level, morphological characters and distribution are noted.  相似文献

采用原位分子杂交方法检测HCV RNA及HBV X基因;采用免疫组织化学方法研究HCV核心抗原,非结构区C33c抗原及HBxAg在肝细胞肝癌中的定位及分布.结果表明(1)HCV RNA、HBV X基因在肝细胞肝癌组织检出率分别为40%(55/136)和82%(112/136).HCV RNA定位于癌细胞的胞浆内,阳性细胞呈散在、灶状及弥漫分布三种形式;HBV X基因在肝癌细胞中的分布呈胞浆型、核型及核浆型,阳性细胞也呈上述三种分布形式;(2)HCV C33c抗原、核心抗原在肝细胞肝癌中的阳性率为81%(133/164)及86%(141/164).C33c抗原定位于癌细胞及肝细胞的胞浆内;核心抗原既定位于癌细胞核中,又可定位于胞浆中.C33c抗原阳性细胞以灶状分布为主;而核心抗原阳性细  相似文献

For a plant selection model with frequency-independent viabilities, fertilities and selfing rates, it is shown that apart from global fixation, for certain parameter combinations a protected polymorphism and facultative fixation (either allele may become fixed according to initial frequencies) may both occur. Facultative fixation requires different selling rates for the dominant and recessive type. Protection of the polymorphism requires resource allocation for male and female function. In this connection the problem of purely genetically caused population extinction is discussed.
For general frequency dependence and regular segregation, the chances for establishment of a completely recessive gene are compared to those of a completely dominant gene. It is proven that the process of establishment of the recessive gene, despite a fitness advantage, may be considerably endangered by drift effects if random mating prevails. The recessive gene may reach the same effectivity in establishment as a dominant gene, only if the recessive homozygote mates exclusively with its own type during the period of establishment.  相似文献
