The hind limb muscles of the spring hare (Pedetes capensis) were found to be relatively heavier than the hind limb muscles of the cane rat (Thryonomys gregorianus). The distribution of succine dehydrogenase activity was investigated in four of these muscles (m. gluteus superficialis, m.semimembranosus, m. biceps femoris and m.rectus femoris) from both animals. It was found that the spring hare had a higher proportion of low-activity fibres in all four muscles than the cane rat. All muscle fibre types were also smaller in diameter in the spring hare than the cane rat. These results are discussed in relation to the different locomotry habits of the two animals.  相似文献

Summary Intrafusal muscle fibres of the slow soleus (Sol) and fast vastus lateralis (VL) muscles of the rat were studied histochemically. Serial transverse sections were incubated for the localization of succinate dehydrogenase (SDH), alpha glycerophosphate dehydrogenase (GPD) and adenosine triphosphatase (ATPase). The latter was examined further after preincubation in acidic solution held at either low or room temperature (RT). The bag2 intrafusal fibres in both muscles displayed high regular and acid stable ATPase, but low SDH and GPD activities. Bag1 intrafusal fibres showed low to moderate regular ATPase, a regional heterogeneity after RT acid preincubation (low activity in juxtaequatorial and high in polar zones), moderate SDH, but low GPD reactions. In both muscles the chain fibres usually exhibited high ATPase for both regular and cold acid preincubated reactions, but usually low activity after RT acid preincubation; they had high SDH but variable GPD activities. In Sol muscle, however, approximately 25% of spindles contained chain fibres that showed high acid-stable ATPase reaction after both cold and RT acid preincubation. In contrast, chain fibres in some VL spindles had a characteristically low ATPase reaction even after cold acid preincubation. This study, therefore, has delineated the existence of an inherent heterogeneity among chain fibres (with respect to their histochemical reactions) in muscle spindles located within slow and fast muscles and also between those found within populations of either Sol or VL muscle spindles.  相似文献

The fibres of superficial and deep abductor muscles of the pectoral fins of the stripped weakfish, Cynoscion guatucupa have been studied using histochemical techniques: succinic dehydrogenase (SDH) for mitochondria, periodic acid–Schiff (PAS) for glycogen, myosin‐adenosintriphosphatase (mATPase) to identify different fibre types based on the contraction speed and modified ATPase to identify capillaries. The fibre diameters were measured, and the capillaries of the main fibre types – red, pink and white— were counted. The two muscles showed both macroscopically and microscopically two well‐differentiated zones with predominant white fibres. The area of insertion of muscles into the fin rays had red, pink and white fibres. The origin zone of the muscle into the bone was composed by white fibres only. Both zones of white muscle evidenced a mosaic of small, medium and large polygonal white fibres. Red, pink and white muscles showed a wide histochemical diversity of fibre subtypes. The area per peripheral capillary increased from the red to the white muscles. Due to the predominance of white fibres, the pectoral fins of C. guatucupa were mainly involved in rapid movements to stop/discontinue and stabilize the body during swimming.  相似文献

The hind limb muscles of the Egyptian desert rodent Gerbillus pyramidum and the albino rat were studied macroscopically and microscopically, using ordinary and histochemical techniques. The results revealed a relatively long hind limb and a relatively long tendon of the gastrocnemius group in G. pyramidum as compared with the albino rat. The dark fibres (red) were dominant in the muscles of the former animal and its spindles similarly contained more dense intrafusal fibres than the spindles of the albino rat. The correlation between these findings and the frequency and power of movement of the hind limbs in G. pyramidum was discussed.  相似文献

The present results give evidence by using cytochemical markers of motor end-plates (ChE, AchR) and ultrastructural techniques that the Levator ani (LA) muscle is also present in adult females: it is composed of differentiated and innervated fibres. A significant difference, both on the number of fibres per muscle (n) and on their average cross-sectional area (acsa), was observed between sexes:-male: n = 5300 +/- 687, acsa: 522 +/- 68.6 microns 2;-female: n = 565 +/- 246.9, acsa: 68 +/- 8.6 microns 2. These results suggest that testosterone could control, at least partially, the number and the diameter of muscle fibres during development.  相似文献

Summary Activity levels of succinate dehydrogenase (SDH) were determined kinetically by means of comparative microphotometric measurements in situ. Activities were correlated with fibre types classified histochemically according to Brooke and Kaiser (1970). Analyses of tibialis anterior muscles in the mouse, rat, guinea pig, rabbit, cat and the human showed pronounced variations in the activity profiles of type I, type IIA and IIB fibres of these muscles. Large scattering of enzyme activity existed in the three fibre populations. Overlaps of varying extent were found for the SDH profiles between the different muscles. Type I fibres reveal species diffeences in aerobic oxidative capacity. Whereas the majority of the IIB fibres in rabbit muscle tended to be low in SDH activity, the main fraction of this fibre population was characterized by high activities in mouse muscle. Similarly, the IIA fibre populations revealed opposite properties in mouse and rabbit muscles. These extremes as well as intermediate activity patterns indicate that no general scheme exists according to which the histochemically assessable myosin ATPase is correlated with the aerobic oxidative capacity of muscle fibres in various mammalian muscles.  相似文献

Summary The dependence of adenosine-triphosphatase (ATPase) and succinic dehydrogenase (SDH) histochemical reactions on the pH of the preincubation medium was studied in serial cross sections of 1- to 6-month-old rat extensor digitorum longus (EDL) and soleus (SOL) muscles.The use of a wide spectrum of pH values confirmed the previous results showing that: (1) according to their ATPase and SDH reactions 3 types of extrafusal muscle fibres, i.e., fast-twitch glycolytic (FG), fast-twitch oxidative-glycolytic (FOG) and slow-twitch oxidative (SO) and 3 types of intrafusal muscle fibres, i.e. typical and intermediate nuclear bag fibres and nuclear chain fibres were observed; (2) only acid preincubation (pH 4.35) is necessary to demonstrate the reversal of the ATPase reaction; while (3) alkali preincubation (pH 10.4) does not provide any new important information as compared with ATPase without preincubation. Furthermore, it was shown that: (4) fast-twitch muscle fibres exhibited high ATPase activity on preincubations at pH 4.9 to 10.4, slow-twitch fibres had very high ATPase activity on preincubation at pH 4.3 and 4.5; (5) after preincubation at pH 4.5 two types of FOG fibres were observed, differing in their ATPase activity; (6) in both muscles there were fibres with intermediate ATPase activity both after acid and/or alkali preincubations; (7) the intrafusal muscle fibres exhibited some specific characteristics when compared with extrafusal fibres.In contrast to the ATPase reactions, SDH activity was decreased equally, in both extra- and intrafusal fibres, with increasing acidity and alkality of the preincubation medium.  相似文献

A histochemical study has been carried out on the eye muscles of the carp. On the base of the ATP-ase and SDH activity and with regard to the localization and diameter of muscle fibres six types of muscle fibres situated in the defined zones can be distinguished. The type 1 and 2 fibres display a moderately high ATP-ase activity at pH 9.4 and rather low activity after alkaline and acid preincubation. Type 1 shows a high SDH activity in contrast to type 2 with a low SDH activity. The other four types of fibres have the high ATP-ase activity at pH 9.4. Type 3 contains fibres with a moderately high ATP-ase activity after alkaline preincubation with a rather low activity after acid preincubation and with a low SDH activity. The fibres of type 4 characterized by the high ATP-ase activity after alkaline and acid preincubation and by the high SDH activity. The fibres of type 5 display high ATP-ase activity after alkaline and acid preincubation and the low SDH activity. They are situated in the white and intermediate fibre zones. The fibres of type 6 are comparable to the fibres of type 5, however they differ diameter and localization, i.e. they are situated in small diameter fibre zone. Using the electron microscope four types of fibres (A, B, C, and D) are found. They vary in the localization of T system, the organization of Z-line and in M-line appearance. In the type B of muscle fibres two different localizations of T system have been discerned.  相似文献

Summary Morphological, histochemical and ultrastructural characteristics of intrafusal fibre types were studied in rat muscle spindles. The existence of three intrafusal fibre types, namely the typical bag, the intermediate bag and the chain fibres was confirmed. Intrafusal fibres differ in diameter, length and number of nuclei in the equatorial zone. Histochemically, typical bag fibres exhibit both alkali-and acid-stable ATPase activity and low SDH activity. Intermediate bag fibres possess low alkali-stable ATPase activity; after acid-preincubation, however, they have low activity only in the juxtaequatorial region, whereas in the polar zones they exhibit high acid-stable ATPase activity. The SDH activity varies from moderate to high. The chain fibres exhibit high alkali-stable and low acid-stable ATPase and high SDH activity in the extensor digitorum longus muscle, whereas in the soleus muscle the acid-stable ATPase activity varies from a low one to a high one, either among individual chain fibres in one spindle, and/or repeatedly along the fibre length.Since there are regional differences in morphological characteristics and in staining properties of intrafusal fibres, a reliable identification of intrafusal fibre types can only be achieved by an analysis of serial sections.  相似文献

The histochemical activities of succinic dehydrogenase (SDH), myofibrillar Adenosine triphosphatase (ATPase) and alpha glycerophosphate dehydrogenase were studied in serial sections of rat vastus lateralis (red) (RVL), gastrocnemius and diaphragm muscles. Three main fibre-types were distinguished. The "Type I" fibres of RVL and gastrocnemius muscles fell into two distinct groups: one category--"Type IA" showed very low ATPase activity. The second category of "Type IB" fibres displayed moderate ATPase reaction. The "Type IA" fibres were divisible into two sub-groups when tested for SDH reaction. "Type IA1" fibres possessed a homogenous distribution of diformazan granules throughout the fibre: "Type IA2" fibres displayed characteristic "moth-eaten" pattern of diformazan localization. The diaphragm muscle did not show either "Type IB" or "Type IA2" varieties. The great majority of TypeI fibres were sub-type IA1 in the three fast muscles studied. It is also demonstrated here that an inherent heterogeneity exists between Type I filores of diaphragm and leg muscles in regard to alpha-GPD localization. This histochemical data emphasizes the fact that subdivision of TypeI striated muscle fibres of mammalian animals into two sub-types is only approximate and that a further subcategorization is possible.  相似文献

The measurements showed a large scatter of the two enzyme activities in these two myosin-based fibre types. In rabbit TA, IIA and IIB fibres have similar GP-OX activities, whereas generally IIA fibres have higher SDH activities than IIB fibres. An inverse distribution of the two enzymes exists in mouse muscle. Generally, IIA fibres of mouse TA display low SDH and IIB fibres high SDH activities. The mean activity of GP-OX is slightly higher in IIA than in IIB fibres of mouse TA. Since measurements of both enzymes were taken in the same fibres, the ratio of their activities in each fibre could be evaluated. The SDH/ GP-OX activity ratios vary significantly between the two fibre populations both in rabbit and in mouse. The ratio is high in IIA and low in IIB fibres of rabbit TA, whereas it is low in IIA and high in IIB fibres of mouse TA.  相似文献

The aim of this study was to evaluate the effect of lead and excess zinc on the adaptation of mitochondria from skeletal muscles to physical effort. Rats were intoxicated once a week for 12 weeks by subcutaneous injection of the solution containing 2 mg Zn+2 and/or 3 mg Pb2+ per kg of body weight. During the last 6 weeks, 6 times weekly, rats performed endurance training which involved swimming 15 minutes daily with additional load of 5% of the body weight. The activities of isocitrate (ICD), malate (SDH), succinate (MDH) dehydrogenases, cytochrome oxidase (COX) and protein content (PM) were determined in the mitochondrial fractions obtained from the soleus muscle (ST fibres), and from the superficial (FTb fibres) and deep (FTa fibres) parts of the gastrocnemius muscle. In the control group (C), which was injected with saline, higher activities of ICD and MDH were obtained in FTa and FTb fibres than in the ST fibres. SDH and COX had higher activities in FTa and ST compared to FTb fibres. Zinc treatment (Zn) caused diminution of ICD, SDH and COX activities in ST fibres. Lead intoxication (Pb) resulted in a decrease of MDH activity in all fibre types, and in a decrease of SDH activity in ST fibres. Simultaneous action of zinc and lead produced an increase in ICD activity and diminution of COX activity in FTb fibres. It also resulted in an increase of SDH and decrease of COX activity in ST fibres. These results suggest that the ST fibres are more susceptible to disturbances of adaptation to physical exercise caused by zinc and lead. There are no signs of uniform antagonism between zinc and lead action in the processes under investigation.  相似文献

The pulmonary veins of albino Wistar rats were studied by means of light and electron microscopy. The media of larger veins consists of cardiac muscle fibers which extend until the vessels attain about 100 μ in diameter. This coat consists of external longitudinal fibers and internal circular fibers. The vasa vasorum are well developed and the capillaries show pseudofenestrations. The numerous adrenergic and cholinergic nerve endings do not form typical motor end-plates as seen in skeletal muscles. The ultrastructure of these media muscle fibers is similar to that of rat hearts. The smooth muscle layer of larger pulmonary veins is not continuous as it is in smaller veins where it forms cushions. Comparisons of albino rats and other rodents reveal striking differences. Action potential shape and propagation velocity (0.5–1.2 m/s) along the myocardial coat of the pulmonary vein were similar to those observed in the left atrium and so was their sensitivity to locally applied acetylcholine. The physiological direction of propagation in rat pulmonary veins is toward the lung. This finding lends support to the hypothesis of a rhythmic, valve-like action of the striated musculature of the pulmonary venous wall during the systole and a possible role in the capacitance of the pulmonary circulation.  相似文献

Prone hip extension has been used as a self-perturbation task to test the stability of the lumbopelvic region. However, the relationship between recruitment patterns in the hip and trunk muscles and lumbopelvic kinematics remains unknown. The present study aimed to examine if the balance of hip and trunk muscle activities are related to pelvic motion and low back muscle activity during prone hip extension. Sixteen healthy participants performed prone hip extension from 30° of hip flexion to 10° of hip extension. Surface electromyography (of the gluteus maximus, semitendinosus, rectus femoris, tensor fasciae latae, multifidus, and erector spinae) and pelvic kinematic measurements were collected. Results showed that increased activity of the hip flexor (tensor fasciae latae) relative to that of hip extensors (gluteus maximus and semitendinosus) was significantly associated with increased anterior pelvic tilt during hip extension (r=0.52). Increased anterior pelvic tilt was also significantly related to the delayed onset timing of the contralateral and ipsilateral multifidus (r=0.57, r=0.53) and contralateral erector spinae (r=0.63). Additionally, the decrease of the gluteus maximus activity relative to the semitendinosus was significantly related to increased muscle activity of the ipsilateral erector spinae (r=-0.57). These results indicate that imbalance between the agonist and antagonist hip muscles and delayed trunk muscle onset would increase motion in the lumbopelvic region.  相似文献

Pathological movement patterns like crouch gait are characterized by abnormal kinematics and muscle activations that alter how muscles support the body weight during walking. Individual muscles are often the target of interventions to improve crouch gait, yet the roles of individual muscles during crouch gait remain unknown. The goal of this study was to examine how muscles contribute to mass center accelerations and joint angular accelerations during single-limb stance in crouch gait, and compare these contributions to unimpaired gait. Subject-specific dynamic simulations were created for ten children who walked in a mild crouch gait and had no previous surgeries. The simulations were analyzed to determine the acceleration of the mass center and angular accelerations of the hip, knee, and ankle generated by individual muscles. The results of this analysis indicate that children walking in crouch gait have less passive skeletal support of body weight and utilize substantially higher muscle forces to walk than unimpaired individuals. Crouch gait relies on the same muscles as unimpaired gait to accelerate the mass center upward, including the soleus, vasti, gastrocnemius, gluteus medius, rectus femoris, and gluteus maximus. However, during crouch gait, these muscles are active throughout single-limb stance, in contrast to the modulation of muscle forces seen during single-limb stance in an unimpaired gait. Subjects walking in crouch gait rely more on proximal muscles, including the gluteus medius and hamstrings, to accelerate the mass center forward during single-limb stance than subjects with an unimpaired gait.  相似文献

The histochemical pattern of red, pink and white muscle of fish living in fresh, brackish, and salt water is reported. The muscle fibres were stained routinely during the year for lactate dehydrogenase (LDH), menadione α-glycerophosphate dehydrogenase (Mα—GPDH), succinic dehydrogenase (SDH), myosin adenosine triphosphatase (myosin ATPase), phosphorylase, lipids and glycogen. The pink and red muscles contain more glycogen and lipids and have a higher SDH activity, which is in accord with their aerobic metabolism and function in sustained swimming activity. The acid labile myosin ATPase activity characteristic of fast twitch fibres is present in the white fibres of most species, however in the white muscle of Gobius paganellus the enzyme activity is stable to both acid and alkali and, in addition, there is a scattered distribution of different fibre types in red and, especially, pink muscle. A study of seasonal variation patterns of myosin ATPase in white muscle of mugilidae over a period of two years has demonstrated, in late summer, the appearance of new small diameter fibres, with a high acid stable enzyme activity, that develop into the large diameter acid labile fibres.  相似文献

Moment arm lengths of three hip extensor muscles, the gluteus maximus, the hamstrings and the adductor magnus, were determined at hip flexion angles from 0 degrees to 90 degrees by combining data from ten autopsy specimens and from twenty patients, the latter examined by computed tomography. A straight-line muscle model for muscle force was used for the hamstrings and adductor magnus, and for the gluteus maximus a two-segment straight-line muscle force model was used. With the joint in its anatomical position the moment arm of the gluteus maximus to the bilateral motion axis averaged 79 mm, for the hamstrings 61 mm and for the adductor magnus 15 mm. The moment arm of gluteus maximus decreased with increasing hip flexion angle. The hamstrings showed an increase in moment arm length up to an average of 35 degrees hip flexion and then a decrease with increasing hip flexion angle. The corresponding figures for the adductor magnus moment arm showed an increase up to 75 degrees and then a decrease. Statistical analysis revealed significant differences in moment arm length between men and women.  相似文献
