Cyanobacteria show high metabolic plasticity by re‐allocating macromolecular resources in response to variations in both environmental inorganic carbon (Ci) and light. We grew cultures of the picoplanktonic cyanobacterium Synechococcus elongatus Nägeli across a 50‐fold range of growth irradiance at either a dissolved [Ci] <0.1 mM, sufficient to induce strongly the carbon‐concentrating mechanism (CCM) or a dissolved [Ci] of ~4 mM, sufficient to strongly induce the CCM to basal constitutive activity. There was no detectable growth cost of acclimation to low Ci across the entire range of irradiance and growth was nearly light saturated at 50 l mol photons·m?2·s?1. Cells acclimated to low Ci significantly re‐allocated macromolecular resources to support their CCM, while maintaining near homeostatis of metabolic flux per unit photosynthetic complex. Changing growth irradiance also drove re‐organization of the photosynthetic machinery to balance excitation flux and metabolic demands, but flux per complex varied widely across the range of tolerable growth irradiances. Across the range of growth irradiance, low Ci cells had significantly less phycocyanin than high Ci cells, which corresponded to a lower PSII absorbance capacity. Furthermore, low Ci cells maintained more PSI per cell?1 than high Ci cells under high growth irradiance. Low Ci cells could therefore maintain more of their PSII reaction centers open at high growth irradiance than could high Ci cells, which experienced a significant PSII closure. Thus, acclimation to growth under high available Ci actually constrained acclimation to high light by restricting electron transport downstream from PSII in S. elongatus.  相似文献

Synechococcus elongatus strain PCC7942 cells were grown in high or low environmental concentrations of inorganic C (high-Ci, low-Ci) and subjected to a light shift from 50 µmol m–2 s–1 to 500 µmol m–2 s–1. We quantified photosynthetic reductant (O2 evolution) and molar cellular contents of phycobilisomes, PSII, PSI, and ribulose-1,5-bisphosphate carboxylase oxygenase (Rubisco) through the light shift. Upon the increase in light, small initial relative decreases in phycobilisomes per cell resulted from near cessation of phycobilisome synthesis and their dilution into daughter cells. Thus, allocation of reductant to phycobilisome synthesis dropped fivefold from pre- to post-light shift. The decrease in phycobilisome synthesis liberated enough material and reductant to allow a doubling of Rubisco and up to a sixfold increase in PSII complexes per cell. Low-Ci cells had smaller initial phycobilisome pools and upon increased light; their reallocation of reductant from phycobilisome synthesis may have limited the rate and extent of light acclimation, compared to high-Ci cells. Acclimation to increased light involved large reallocations of C, N, and reductant among different components of the photosynthetic apparatus, but total allocation to the apparatus was fairly stable at ca. 50% of cellular N, and drew 25–50% of reductant from photosynthesis.  相似文献

Recently, several studies reported that the optimum temperature for the initial slope [IS(Ci)] of the light-saturated photosynthetic rate (A) versus intercellular CO2 concentration (Ci) curve changed, depending on the growth temperature. However, few studies compare IS(Ci) with ribulose 1,5-bisphosphate carboxylase/oxygenase (Rubisco) properties. Here, we assessed Rubisco activation state and in vitro Rubisco kinetics, the main determinants of IS(Ci), in spinach leaves grown at 30/25 [high temperature (HT)] and 15/10 degrees C [low temperature (LT)]. We measured Rubisco activation state and A at a CO2 concentration of 360 microL L(-1) (A360) at various temperatures. In both HT and LT leaves, the Rubisco activation state decreased with increasing temperatures above the optimum temperatures for A360, while the activation state remained high at lower temperatures. To compare Rubisco characteristics, temperature dependences of the maximum rate of ribulose 1,5-bisphosphate (RuBP) carboxylation (Vcmax), specificity factor (Sc/o) and thermal stability were examined. We also examined Vcmax, and thermal stability in the leaves that were transferred from HT to LT conditions and were subsequently kept under LT conditions for 2 weeks (HL). Rubisco purified from HT, LT and HL leaves are called HT, LT and HL Rubisco, respectively. Thermal stabilities of LT and HL Rubisco were similar and lower than that of HT Rubisco. Both Vcmax and Sc/o in LT Rubisco were higher than those of HT Rubisco at low temperatures, while these were lower at high temperatures. Vcmax in HL Rubisco were similar to those of LT Rubisco at low temperatures, and to those of HT Rubisco at high temperatures. The predicted photosynthetic rates, taking account of the Rubisco kinetics and the Rubisco activation state, agreed well with A360 in both HT and LT leaves. This study suggests that photosynthetic performance is largely determined by the Rubisco kinetics at low temperature and by Rubisco Kinetics and the Rubisco activation state at high temperature.  相似文献

Inorganic carbon acquisition in red tide dinoflagellates   总被引:3,自引:0,他引:3  
Carbon acquisition was investigated in three marine bloom-forming dinollagellates-Prorocentrum minimum, Heterocapsa triquetra and Ceratium lineatum. In vivo activities of extracellular and intracellular carbonic anhydrase (CA), photosynthetic O2 evolution, CO2 and HCO3- uptake rates were measured by membrane inlet mass spectrometry (MIMS) in cells acclimated to low pH (8.0) and high pH (8.5 or 9.1). A second approach used short-term 14C-disequilibrium incubations to estimate the carbon source utilized by the cells. All three species showed negligible extracellular CA (eCA) activity in cells acclimated to low pH and only slightly higher activity when acclimated to high pH. Intracellular CA (iCA) activity was present in all three species, but it increased only in P. minimum with increasing pH. Half-saturation concentrations (K1/2) for photosynthetic O2 evolution were low compared to ribulose 1,5-bisphosphate carboxylase/oxygenase (Rubisco) kinetics. Moreover, apparent affinities for inorganic carbon (Ci) increased with increasing pH in the acclimation, indicating the operation of an efficient CO2 concentration mechanism (CCM) in these dinoflagellates. Rates of CO2 uptake were comparably low and could not support the observed rates of photosynthesis. Consequently, rates of HCO3- uptake were high in the investigated species, contributing more than 80% of the photosynthetic carbon fixation. The affinity for HCO3- and maximum uptake rates increased under higher pH. The strong preference for HCO3- was also confirmed by the 14C-disequilibrium technique. Modes of carbon acquisition were consistent with the 13C-fractionation pattern observed and indicated a strong species-specific difference in leakage. These results suggest that photosynthesis in marine dinoflagellates is not limited by Ci even at high pH, which may occur during red tides in coastal waters.  相似文献

Acclimation to rapidly fluctuating light, simulating shallow aquatic habitats, is altered depending on inorganic carbon (Ci) availability. Under steady light of 50 μmol photons·m?2·s?1, the growth rate of Synechococcus elongatus PCC7942 was similar in cells grown in high Ci (4 mM) and low Ci (0.02 mM), with induced carbon concentrating mechanisms compensating for low Ci. Growth under fluctuating light of a 1‐s period averaging 50 μmol photons·m?2·s?1 caused a drop in growth rate of 28%±6% in high Ci cells and 38%±8% in low Ci cells. In high Ci cells under fluctuating light, the PSI/PSII ratio increased, the PSII absorption cross‐section decreased, and the PSII turnover rate increased in a pattern similar to high‐light acclimation. In low Ci cells under fluctuating light, the PSI/PSII ratio decreased, the PSII absorption cross‐section decreased, and the PSII turnover remained slow. Electron transport rate was similar in high and low Ci cells but in both was lower under fluctuating than under steady light. After acclimation to a 1‐s period fluctuating light, electron transport rate decreased under steady or long‐period fluctuating light. We hypothesize that high Ci cells acclimated to exploit the bright phases of the fluctuating light, whereas low Ci cells enlarged their PSII pool to integrate the fluctuating light and dampen the variation of the electron flux into a rate‐restricted Ci pool. Light response curves measured under steady light, widely used to predict photosynthetic rates, do not properly predict photosynthetic rates achieved under fluctuating light, and exploitation of fluctuating light is altered by Ci status.  相似文献

雾凉季研究了西双版纳热带雨林4种植物幼苗对生长光环境的适应,其中两个树种幼苗喜光(团花和滇南插柚紫),两个树种幼苗耐荫(滇南红厚壳和玉蕊)发现弱光环境中生长的4种植物比叶重、光合能力、光饱和点、光补偿点暗呼吸速度、叶绿素a/b比较低,叶绿素含量较高。玉蕊和滇南红厚壳幼苗的光合能力和呼吸速度 于团花树和滇南插柚紫。团花树和滇南插机紫的比叶重和光合作用的可塑性大于玉蕊和滇南红厚壳。高光强下生长的团花树和滇南插机紫增加叶氮分配给羧化酶的比较。减少分配给叶绿素的比例。滇南红厚壳和玉蕊适应弱光环境的能力略强于团花树和滇南插机紫,但适应强光的能力较差。研究结果支持树种的生理生态特性决定了其演替状况和生境选择的假说,单位干重叶的光合能力和呼吸速率并未表现出利于光适应的可塑性,表明4种植物生理适应能力较差,形态学上的适应在4种热带雨林树种幼苗光适应方面起到了重要的作用,叶氮分配也是它们光适应的策略之一。  相似文献

于分蘖、拔节和抽穗3个时期在空气CO2浓度(380μmol·mol-1)下测定稻田中稗草叶片的净光合速率(Pn),发现在开放式CO2浓度增高(FACE)条件下生长的稗草叶片后2个时期的Pn显著低于普通空气中生长的对照,比对照下降约20%,说明FACE条件下稗草叶片光合作用对高CO2浓度发生了明显的适应.同时,叶片的气孔导度(Gs)和胞间CO2浓度(Ci)的下降更为明显.与对照相比,叶片可溶性蛋白含量明显降低,拔节期只有对照的62.4%;高CO2浓度下生长的稗草叶片Rubisco含量也降低,分蘖期和拔节期分别为对照的87%和84%,但其差异未达到显著水平.可以认为,长期生长在高CO2浓度下的C4植物稗草叶片光合作用的适应是叶片气孔部分关闭和可溶性蛋白含量下降的结果.  相似文献

Comprehensive studies on the processes involved in photosynthetic acclimation after a sudden change in light regime are scarce, particularly for trees. We tested (i) the ability of photosynthetic acclimation in the foliage of walnut trees growing outdoors after low‐to‐high and high‐to‐low light transfers made early or late in the vegetation cycle, and (ii) the relative importance of changes in total leaf nitrogen versus changes in the partitioning of leaf nitrogen between the different photosynthetic functions during a 2 month period after transfer. Changes in maximum carboxylation rate, light‐saturated electron transport rate, respiration rate, total leaf nitrogen, ribulose 1·5‐bisphosphate carboxylase/oxygenase (Rubisco) and total chlorophylls were surveyed before and after the change in light regime. Respiration rate acclimated fully within 1 week of transfer, and full acclimation was observed 1 month after transfer for the amount of Rubisco. In contrast, total nitrogen and photosynthetic capacity acclimated only partially during the 2 month period. Changes in photosynthetic capacity were driven by changes in both total leaf nitrogen and leaf nitrogen partitioning. The extent of acclimation also depended strongly on leaf age at the time of the change in light regime.  相似文献

Net phytoplankton (> 20 μm) comprised 51 ± 9% of the total chlorophyll (Chl) in a Skeletonema costatum– dominated spring bloom in Delaware Bay. The net phytoplankton had low C:N and high protein: carbohydrate ratios, indicating that their growth was nutrient-replete. Their photosynthetic responses were characterized by low specific absorption, low light-limited and light-saturated rates of photosynthesis, and high quantum yields, indicative of acclimation to low irradiance and internal self-shading. High fucoxanthin: Chi ratios also indicated low light acclimation, but high photoprotective xanthophyll: Chi ratios suggested a high capacity for photoprotective energy dissipation. Ribulose-1,5-bisphosphate carboxylase/oxygenase (Rubisco) could be activated and deactivated in response to changes in irradiance and was fully activated at the surface of the water column and fully deactivated in aphotic deep water. Maximum Rubisco activity was correlated with Rubisco content and bulk protein content of the phytoplankton and with light-saturated rates of photosynthesis measured in short (< 20-min) incubations. Long (60-min) incubations caused a decrease in the light-saturated rate of photosynthesis, possibly because of feedback limitation. While feedback limitation is unlikely to occur in the water column, it should be considered when estimating productivity in well-mixed waters from fixed light-depth incubations.  相似文献

Hizikia fusiformis thalli experience dynamic incident light conditions during the period of growth. The present study was designed to examine how changing photon irradiance affects the photosynthesis both in the short and long terms by culturing H. fusiformis under three different light levels: 35 μmol photons m-2 s-1 (low light, LL), 85 μmol photons m-2 s-1 (intermediate light, IL), and 165 μmol photons m-2 s-1 (high light, HL). A similar relative growth rate was observed between IL- and HL-grown algae, but the growth rate was significantly reduced in LL-grown algae. The photosynthetic rates (P n) measured at their respective growth light levels were found to be lowest in the thalli grown at LL and highest at HL. However, LL-grown algae exhibited much higher P n in comparison with IL- and the HL-grown thalli at the same measuring photosynthetic photon flux density, indicating the photosynthetic acclimation to low growth light in H. fusiformis. The photosynthesis–light curves showed that LL-grown algae had a highest light-saturating maximum P n (P max) in comparison with IL- or HL-grown algae when the photosynthetic rates were expressed on the biomass basis. However, P max was highest in HL-grown algae compared to IL- or LL-grown algae when the rates were normalized to chlorophyll a. The photosynthesis–inorganic carbon (Ci) response curves were also significantly affected by the growth light conditions. The highest value of apparent photosynthetic conductance occurred in LL-grown algae while the lowest value in HL-grown algae. Additionally, the activity of external carbonic anhydrase (CA) tended to increase while the total CA activity inclined to decrease in H. fusiformis thalli when the growth light level altered from 35 to 165 μmol photons per square meter per second. The external CA inhibitors showed a higher inhibition in HL-grown algae compared with LL-grown algae. It was proposed that photosynthetic acclimation to low light condition in H. fusiformis was achieved through an increase in the number of reaction centers and increased capacities of electron transport and of Ci transport within cells. The ability of photosynthetic acclimation to low light confers H. fusiformis thalli to overcome the environmental low light condition as a result of the attenuation of seawater or self-shading through enhancing its photosynthetic performance and carbon assimilation necessary for growth.  相似文献

The potential role of foliar carbon export features in the acclimation of photosynthetic capacity to differences and changes in light environment was evaluated. These features included apoplastic vs. symplastic phloem loading, density of loading veins, plasmodesmatal frequency in intermediary cells, and the ratio of loading cells to sieve elements. In initial studies, three apoplastic loaders (spinach, pea, Arabidopsis thaliana) exhibited a completely flexible photosynthetic response to changing light conditions, while two symplastic loaders (pumpkin, Verbascum phoeniceum), although able to adjust to different long-term growth conditions, were more limited in their response when transferred from low (LL) to high (HL) light. This suggested that constraints imposed by the completely physical pathway of sugar export might act as a bottleneck in the export of carbon from LL-acclimated leaves of symplastic loaders. While both symplastic loaders exhibited variable loading vein densities (low in LL and high in HL), none of the three apoplastic loaders initially characterized exhibited such differences. However, an additional apoplastic species (tomato) exhibited similar differences in vein density during continuous growth in different light environments. Furthermore, in contrast to the other apoplastic loaders, photosynthetic acclimation in tomato was not complete following a transfer from LL to HL. This suggests that loading vein density and loading cells per sieve element, and thus apparent loading surface capacity, play a major role in the potential for photosynthetic acclimation to changes in light environment. Photosynthetic acclimation and vein density acclimation were also characterized in the slow-growing, sclerophytic evergreen Monstera deliciosa. This evergreen possessed a lower vein density during growth in LL compared to HL and exhibited a more severely limited potential for photosynthetic acclimation to increases in light environment than the rapidly-growing, mesophytic annuals.  相似文献

Rice (Oryza sativa L.) has been used to study the long-term responses of photosynthesis to high irradiance focusing on the composition of the photosynthetic apparatus and leaf morphology. Typical sun/shade differences in chloroplast composition are seen in the fifth leaf following growth in high irradiance compared with low irradiance (1000 and 200 micromol m(-2) s(-1), respectively): higher light-saturated rates of photosynthesis (P(max)), higher amounts of Rubisco protein, and a lower chlorophyll a:b ratio. In addition, leaves were thicker under high light compared with low light. However, responses appear more complex when leaf developmental stage is considered. Using a system of transferring plants from low to high light in the laboratory responses that occur before and after full leaf extension have been studied. Acclimation of photosynthesis is limited by leaf age: the transfer to high light, post-leaf extension, is characterized by alterations in chlorophyll a:b but not in Rubisco protein, which may be limited by leaf morphology. Microarray analysis of gene expression was carried out on plants that were transferred to high light post-leaf extension. A down-regulation of light-harvesting genes was seen. No change in the expression level of Rubisco genes was observed. Up-regulation of genes involved in photoprotection was observed. It was also shown that high-light leaf morphology is established prior to formation of the zone of cellular elongation and division. The endogenous and environmental factors which establish the characteristics of high light acclimation may be important for attaining high rates of assimilation in leaves and crop canopies, and the fifth leaf in rice provides a convenient model system for the determination of the mechanisms involved.  相似文献

Net photosynthetic rates (Pns) in leaves were compared between rice plants grown in ambient air control and free-air CO2 enrichment (FACE, about 200 micromol mol(-1) above ambient) treatment rings. When measured at the same CO2 concentration, the Pn of FACE leaves decreased significantly, indicating that photosynthetic acclimation to high CO2 occurs. Although stomatal conductance (Gs) in FACE leaves was markedly decreased, intercellular CO2 concentrations (Ci) were almost the same in FACE and ambient leaves, indicating that the photosynthetic acclimation is not caused by the decreased Gs. Furthermore, carboxylation efficiency and maximal Pn, both light and CO2-saturated Pn, were decreased in FACE leaves, as shown by the Pn-Ci curves. In addition, the soluble protein, Rubisco (ribulose-1,5-bisphosphate caboxylase/oxygenase), and its activase contents as well as the sucrose-phosphate synthase activity decreased significantly, while some soluble sugar, inorganic phosphate, chlorophyll and light-harvesting complex II (LHC II) contents increased in FACE leaves. It appears that the photosynthetic acclimation in rice leaves is related to both ribulose-1,5-bisphosphate (RuBP) carboxylation limitation and RuBP regeneration limitation.  相似文献

Photosynthesis and growth to maturity of antisense ribulose-1,5-bisphosphate carboxylase/oxygenase (Rubisco) activase Arabidopsis thaliana with reduced concentrations of activase relative to wild-type (Wt) plants were measured under low (200 mumol m-2 s-1) and high (600 mumol m-2 s-1) photosynthetic photon flux density growing conditions. Both growth and photosynthesis were significantly reduced in an Arabidopsis clone (R100) with 30 to 40% Wt activase, an effect that was more pronounced in high light. The aboveground biomass of the antisense clone R100 reached 80% of Wt under low light and 65% of Wt under high light. Decreased growth in the antisense plants was attributed to reduced relative rates of growth and leaf area expansion early in development; all plants attained similar values of relative rates of growth and leaf elongation by 21 d after planting. Reductions in photosynthesis were attributed to decreased Rubisco activation in the antisense plants. Rubisco constituted about 40% of total soluble protein in both Wt and clone R100 under both light regimes. Activase content was 5% and 1.4% of total soluble protein in Wt and clone R100, respectively, and also was unaffected by growth irradiance. The stoichiometry of Rubisco to activase was estimated at 20 Rubisco active sites per activase tetramer in Wt Arabidopsis and 60 to 80 in the transgenic clone R100. We conclude that Wt Arabidopsis does not contain Rubisco activase in great excess of the amount required for optimal growth.  相似文献

Acclimation of rice photosynthesis to irradiance under field conditions   总被引:8,自引:0,他引:8  
Acclimation to irradiance was measured in terms of light-saturated photosynthetic carbon assimilation rates (P(max)), Rubisco, and pigment content in mature field-grown rice (Oryza sativa) plants in tropical conditions. Measurements were made at different positions within the canopy alongside irradiance and daylight spectra. These data were compared with a second experiment in which acclimation to irradiance was assessed in uppermost leaves within whole-plant shading regimes (10% low light [LL], 40% medium light [ML], and 100% high light [HL] of full natural sunlight). Two varieties, japonica (tropical; new plant type [NPT]) and indica (IR72) were compared. Values for Rubisco amount, chlorophyll a/b, and P(max) all declined from the top to the base of the canopy. In the artificial shading experiment, acclimation of P(max) (measured at 350 microL L(-1) CO(2)) occurred between LL and ML for IR72 with no difference observed between ML and HL. The Rubisco amount increased between ML and HL in IR72. A different pattern was seen for NPT with higher P(max) (measured at 350 microL L(-1) CO(2)) at LL than IR72 and some acclimation of this parameter between ML and HL. Rubisco levels were higher in NPT than IR72 contrasting with P(max). Comparison of data from both experiments suggests a leaf aging effect between the uppermost two leaf positions, which was not a result of irradiance acclimation. Results are discussed in terms of: (a) acclimation of photosynthesis and radiation use efficiency at high irradiance in rice, and (b) factors controlling photosynthetic rates of leaves within the canopy.  相似文献

The temperature response of C(3) and C(4) photosynthesis   总被引:1,自引:0,他引:1  
We review the current understanding of the temperature responses of C(3) and C(4) photosynthesis across thermal ranges that do not harm the photosynthetic apparatus. In C(3) species, photosynthesis is classically considered to be limited by the capacities of ribulose 1.5-bisphosphate carboxylase/oxygenase (Rubisco), ribulose bisphosphate (RuBP) regeneration or P(i) regeneration. Using both theoretical and empirical evidence, we describe the temperature response of instantaneous net CO(2) assimilation rate (A) in terms of these limitations, and evaluate possible limitations on A at elevated temperatures arising from heat-induced lability of Rubisco activase. In C(3) plants, Rubisco capacity is the predominant limitation on A across a wide range of temperatures at low CO(2) (<300 microbar), while at elevated CO(2), the limitation shifts to P(i) regeneration capacity at suboptimal temperatures, and either electron transport capacity or Rubisco activase capacity at supraoptimal temperatures. In C(4) plants, Rubisco capacity limits A below 20 degrees C in chilling-tolerant species, but the control over A at elevated temperature remains uncertain. Acclimation of C(3) photosynthesis to suboptimal growth temperature is commonly associated with a disproportional enhancement of the P(i) regeneration capacity. Above the thermal optimum, acclimation of A to increasing growth temperature is associated with increased electron transport capacity and/or greater heat stability of Rubisco activase. In many C(4) species from warm habitats, acclimation to cooler growth conditions increases levels of Rubisco and C(4) cycle enzymes which then enhance A below the thermal optimum. By contrast, few C(4) species adapted to cooler habitats increase Rubisco content during acclimation to reduced growth temperature; as a result, A changes little at suboptimal temperatures. Global change is likely to cause a widespread shift in patterns of photosynthetic limitation in higher plants. Limitations in electron transport and Rubisco activase capacity should be more common in the warmer, high CO(2) conditions expected by the end of the century.  相似文献

Disruption of the ccmM gene in the cyanobacterium Synechocystis sp. PCC 6803 causes a deficiency of carboxysomes and impairs growth in ambient CO2. The effect of this gene defect on cellular metabolism was investigated using electron microscopy, biochemical and fluorescence analysis. Mutant cells were devoid of the characteristic dense polyhedral bodies called carboxysomes. The photosynthetic oxygen evolution was considerably lower in mutant cells compared to wild type, while Rubisco activity in cell extracts was similar. During photosynthetic CO2-dependent oxygen evolution, Rubisco Vmax dropped from 142 micromol mg-1 chlorophyll h-1 (WT) to 77 micromol mg-1 chlorophyll h-1 in the mutant cells, and the Km for Ci (inorganic carbon) increased from 0.5 mM (WT) to 40 mM. The fluorescent indicator, acridine yellow, was used for non-invasive measurements of cytoplasmic pH changes in whole cells induced by addition of Ci, making use of the decrease in fluorescence yield that accompanies cytoplasmic acidification. The experimental results indicate that control of the cytoplasmic pH is linked to the internal carbon pool (Ci). Both wild-type and ccmM-deficient cells showed a linear response of acridine yellow fluorescence quenching and, thus, of internal acidification, with respect to externally added inorganic carbon. However, the fluorescence analysis of mutant (carboxysome-free) cells indicated slower kinetics of Ci accumulation.  相似文献
