Concurrent training is a strategy employed in both general fitness and sports conditioning. The purpose of this study was to compare the responses of VO2 in different combinations of strength exercise with aerobic interval exercise. Eight men (23.6 ± 4.2 years, 178 ± 6.3 cm, 77 ± 7.9 kg, 7.67 ± 1.95% body fat) completed 3 combinations of strength training (ST) and aerobic training (AT) in a randomized order with a 7-day recovery period: AT before ST exercises, AT between 2 blocks of ST exercises, and AT after ST exercises. The ST comprised 4 exercises performed in 3 sets of 10 reps and 2 exercises, abdominal crunch and lumbar extension, performed in 3 sets of 30 and 20 reps, respectively. The AT consisted of a 20-minute interval cycling. There were no significant differences in the values of absolute or relative VO2, in the heart rate (HR) and in the respiratory exchange ratio (RER) when the 3 sessions (during + postexercise measurements) were compared (values are mean ± SD). Analyzing only ST in each session, differences were detected in the RER values (F = 4.714; p < 0.05; η2 = 0.308) between AT before ST and AT in the middle of ST (1.01 ± 0.97 vs. 1.11 ± 0.07, respectively). In all sequences, there was a significant increase (p < 0.05) in the values of relative and absolute VO2 and HR, and a significant decrease in RER values (p < 0.05) from the first to the second part of the ST session. The values of absolute or relative VO2, HR, and RER did not vary significantly among the 3 sessions as compared with the AT after ST. These data support the hypothesis that ST and AT, when performed in sequence in the same session, do not seem to affect the overall oxygen consumption during the exercise session. Therefore, training sessions may incorporate both modalities without apparent impact on aerobic exercise.  相似文献

The fundamental pulmonary O(2) uptake (.VO(2)) response to moderate, constant-load exercise can be characterized as (d.VO(2)/dt)(tau)+Delta.VO(2) (t)=Delta.VO(2SS) where Delta.VO(2SS) is the steady-state response, and tau is the time constant, with the .VO(2) kinetics reflecting intramuscular O(2) uptake (.QO(2)) kinetics, to within 10%. The role of phosphocreatine (PCr) turnover in .QO(2) control can be explored using (31)P-MR spectroscopy, simultaneously with .VO(2). Although tau.VO(2) and tauPCr vary widely among subjects (approx. 20-65 s), they are not significantly different from each other, either at the on- or off-transient. A caveat to interpreting the "well-fit" exponential is that numerous units of similar Delta.VO(2SS) but with a wide tau distribution can also yield a .VO(2) response with an apparent single tau. This tau is, significantly, inversely correlated with lactate threshold and .VO(2max)(but is poorly predictive; a frail stamen, therefore), consistent with tau not characterizing a compartment with uniform kinetics. At higher intensities, the fundamental kinetics become supplemented with a slowly-developing phase, setting .VO(2)on a trajectory towards maximum .VO(2). This slow component is also demonstrable in Delta[PCr]: the decreased efficiency thereby reflecting a predominantly high phosphate-cost of force production rather than a high O(2)-cost of phosphate production. We also propose that the O(2)-deficit for the slow-component is more likely to reflect shifting Delta.VO(2SS) rather than a single one with a single tau.  相似文献

To assess the rate-limiting factor of oxygen uptake ( ) kinetics at the onset of exercise, six healthy male sedentary subjects performed repeated one-legged constant-load cycle exercise. The one-legged constant-load exercise test consisted of two 5-min periods of pedalling at an exercise intensity of 50 W, with a 5-min rest between periods (these exercise periods, i.e. first and second exercises, were performed by the same leg). The exercise was then repeated using the other leg. In addition, two-legged incremental exercise was investigated to establish whether kinetics were affected by slower heart rate kinetics. The incremental exercise test consisted of two-legged pedalling first with the cycle unloaded as a warm-up for 5 min followed by 50-W exercise for 5 min. The exercise intensity was then increased to 100 W for 5 min. During exercise, gas exchange parameters were determined by the breath-by-breath method and cardiac output ( ) was determined continuously by an impedance technique with a computer-based automated system. To determine the kinetics of heart rate (HR), and , a best fit procedure was employed using least-squares criteria with a time delay, except during the initial increase. During the one-legged constant-load exercise test, kinetics were significantly accelerated by repeated exercise using the same leg. On the other hand, when the exercise was changed to the other leg, kinetics were significantly slower, although kinetics continued to be faster. During the incremental exercise test, although the HR response was slower at the transition from 50-W to 100-W exercise than at the transition from warm-up to 50-W exercise, there were no significant changes in kinetics. These findings suggest that kinetics may be affected by metabolic conditions in the muscle, but not by blood flow ( and/or HR) kinetics.  相似文献

Four incremental protocols of knee extension exercise of different stage durations were compared to study the effect of the protocol upon power output at the last stage (P peak). Previous studies of knee extension have found very different power outputs with similar ergometers and these large differences have been interpreted as being the result of the fatigue due to the durations of the protocols. The knee extension device used in previous studies was modified to avoid the action of the knee and hip flexors: the subjects pushing on a lever instead of pulling a rod. In the present study five subjects performed four incremental knee extension exercises which differed with regard to stage duration (60, 90, 180 or 360 s) on this ergometer. The P peak, peak oxygen uptake (O2peak) and peak heart rate (HRpeak) were measured at the end of each of these four incremental protocols. In eight subjects, the reliability of the protocols with the two shortest increments (60 and 90-s stages) was verified by measuring P peak at 60 s and 90 s (P peak60, P peak90) twice. The knee ergometer proposed in the present paper was easy to use without any special training and should improve the measurement of P peak. The P peak60 [49.4 (SD 5.6) W] was higher than at 180 s [P peak180, 43.6 (SD 5.8) W, P < 0.05] and at 360 s [P peak360, 43.4 (SD 5.3) W, P < 0.05]. All the other differences in P peak, O2 peak and HRpeak were not significant. All correlations between P peak60, P peak90, P peak180 and P peak360 were significant, except those between P peak360 and P peak90 or P peak180. The effect of the stage duration on power output and oxygen uptake at the end of the knee extension exercises was not great. Consequently, the large differences in power output and oxygen uptake observed in previous studies cannot be explained by the protocol only. The significant difference between P peak 60 and P peak90 was of the order of 10% in agreement with findings in the literature using cycle ergometry. The reliability of P peak60 and P peak90 was high and the use of these protocols can be recommended if further studies show that the measurement of P peak, is useful in the evaluation of local endurance. 1. The hypoglycaemic compound diphenyleneiodonium causes rapid and extensive swelling of rat liver mitochondria suspended in 150mm-NH(4)Cl, and in 150mm-KCl in the presence of 2,4-dinitrophenol and valinomycin. This indicates that diphenyleneiodonium catalyses a compulsory exchange of OH(-) for Cl(-) across the mitochondrial inner membrane. Br(-) and SCN(-) were the only other anions found whose exchange for OH(-) is catalysed by diphenyleneiodonium. 2. Diphenyleneiodonium inhibited state 3 respiration of mitochondria and slightly stimulated state 4 respiration with succinate or glutamate as substrate in a standard Cl(-)-containing medium. 3. Diphenyleneiodonium did not inhibit state 3 respiration significantly in two Cl(-)-free media (based on glycerol 2-phosphate or sucrose) but caused some stimulation of state 4. 4. In Cl(-)-containing medium diphenyleneiodonium only slightly inhibited the 2,4-dinitrophenol-stimulated adenosine triphosphatase and it had little effect in the absence of Cl(-). 5. The inhibition of respiration in the presence of Cl(-) is dependent on the Cl(-)-OH(-) exchange. 2,4-Dichlorodiphenyleneiodonium is ten times as active as diphenyleneiodonium both in causing swelling of mitochondria suspended in 150mm-NH(4)Cl and in inhibiting state 3 respiration in Cl(-)-containing medium. Indirect evidence suggests that the Cl(-)-OH(-) exchange impairs the rate of uptake of substrate anions. 6. It is proposed that stimulation of state 4 respiration in the absence of Cl(-) depends, at least in part, on an electrogenic uptake of diphenyleneiodonium cations. 7. Tripropyl-lead acetate, methylmercuric iodide and nine substituted diphenyleneiodonium derivatives also catalyse Cl(-)-OH(-) exchange across the mitochondrial membrane. 8. Diphenyleneiodonium is compared with the trialkyltin compounds, which are also known to mediate Cl(-)-OH(-) exchange and which have in addition strong oligomycin-like effects on respiration. It is concluded that diphenyleneiodonium is specific for catalysing anion-OH(-) exchange and will be a useful reagent for investigating membrane-dependent systems.  相似文献

This study investigated the oxygen consumption of the putative oxygen conformer marbled swamp eel Synbranchus marmoratus during progressive hypoxia. Earlier studies have not reached an agreement on whether S. marmoratus is a conformer or a regulator. Reactive oxygen species (ROS) are generated by a variety of sources from the environment (e.g., photo-oxidations and emissions) and normal cellular functions (e.g., mitochondrial metabolism and neutrophil activation). ROS include free radicals (e.g., superoxide and hydroxyl radicals), nonradical oxygen species (e.g., hydrogen peroxide and peroxynitrite) and reactive lipids and carbohydrates (e. g., ketoaldehydes, hydroxynonenal). Oxidative damage to DNA can occur by many routes including the oxidative modification of the nucleotide bases, sugars, or by forming crosslinks. Such modifications can lead to mutations, pathologies, cellular aging and death. Oxidation of proteins appears to play a causative role in many chronic diseases of aging including cataractogenesis, rheumatoid arthritis, and various neurodegenerative diseases including Alzheimer's Disease (AD). Our goal is to elucidate the mechanism(s) by which oxidative modification results in the disease. These studies have shown that (a) cells from old individuals are more susceptible to oxidative damage than cells from young donors; (b) oxidative protein modification is not random; (c) some of the damage can be prevented by antioxidants, but there is an age-dependent difference; and (d) an age-related impairment of recognition and destruction of modified proteins exists. It is believed that mechanistic insight into oxidative damage will allow prevention or intervention such that these insults are not inevitable. Our studies are also designed to identify the proteins which are most susceptible to ROS damage and to use these as potential biomarkers for the early diagnosis of diseases such as AD. For example, separation of proteins from cells or tissues on one- and two-dimensional gels followed by staining for both total protein and specifically oxidized residues (e.g., nitrotyrosine) may allow identification of biomarkers for AD.  相似文献

Decreases in dissolved organic carbon (DOC) and dissolved oxygen (DO) with increasing depth below the groundwater table are often considered as evidence for aerobic respiration; however, they may reflect mixing of infiltrating water and groundwater. We found that groundwater DOC concentration was on average 0.3 mg C l?1 higher and DO concentration 1.5 mg O2 l?1 lower at recharge sites replenished with stormwater than at reference sites fed by direct infiltration of rain water from the land surface. Groundwater DOC increased and DO decreased with increasing vadose zone thickness (VZT) at both recharge and reference sites. There was no significant interaction between the effects of stormwater infiltration and VZT. Vertical changes in DOC and DO below the groundwater table at recharge sites could account for by simple mixing of infiltrating stormwater and groundwater. Moreover, aquifer sediment respiration (SR) was not significantly higher at recharge sites than at reference sites. However, slow filtration column experiments showed that SR increased significantly with an increasing supply of easily biodegradable DOC. We conclude that the observed reduction in DOC below the groundwater table at recharge sites was essentially due to water mixing rather than biological uptake because of the low biodegradability of the DOC and the short transit time of stormwater in the upper layers of groundwater. Our results highlight the need to distinguish between the effect of hydrological and biological processes on DOC and DO patterns below the groundwater before conclusions are made on the efficiency of groundwater in degrading surface-derived DOC.  相似文献

We have studied metabolic responses of six male volunteers who were exposed to hypoxic nitrogen–oxygen and argon–oxygen respiratory mixtures under hermetic chamber conditions for a long time. We measured the values of 44 biochemical parameters of venous blood, which reflected the state of different parts of metabolism, as well as the state of organs and tissues. Under the conditions of argon–oxygen respiratory mixture with the level of oxygen of 12.8% within the first five days and 12.0% from six to ten days of the experiment, the activation of processes of anaerobic glycolysis and lipolysis with the development of metabolic acidosis was observed. Against this background, biochemical signs of unfavorable changes in the myocardium were recorded. When we used the nitrogen–oxygen respiratory mixture with the level of oxygen of 13.0% during the first five days of the experiment and 12.1% from the sixth to the tenth day, these changes were expressed much more significantly: the activation of lipolysis and a decrease in the level of serum iron were observed and the signs of damage of mitochondria, depression of the kidney function, and development of hypodynamia were recorded. Complete stabilization of metabolic processes was reached only after seven to eight days of the recovery period for both mixtures. These findings lead to the conclusion that it is physiologically more preferable to use oxygen–argon respiratory mixtures at small immersion depths.  相似文献

It has recently been shown that, in subthylakoid particles prepared using detergent, there is inhibition of oxygen production reactions in photosynthesis by thermodynamic feedback from oxygen build-up, with 50% inhibition at 230 kPa partial pressure of oxygen. This article presents a comprehensive analysis of laboratory data on the effects of high oxygen partial pressures on photosynthesis, and on photo-lithotrophic and chemo-organotrophic growth, of oxygen-producing organisms. The article also contains an analysis of the extent to which high oxygen concentrations occur at the site of photosystem II (PSII) activity under natural conditions today and in the past. The conclusion is that the oxygen concentrations found in nature are very unlikely to reach that needed to cause 50% inhibition of the photosynthetic oxygen production reaction in subthylakoid particles, but that it is just possible that a small part of the inhibition of photosynthesis and of photo-lithotrophic growth by oxygen can be attributed to inhibition of oxygen production by PSII.  相似文献

The present study was conducted to validate the applicability of Cooper''s 12-minute run test (CRT) for predicting VO2max in male university students of Kolkata, India, to bypass the exhaustive and complicated protocol of direct estimation of VO2max. Eighty-eight sedentary male university students recruited by simple random sampling from the University of Calcutta, Kolkata, were randomly assigned to the study group (N = 58) and the confirmatory group (N = 30). VO2max of each participant was determined by the direct procedure and the indirect CRT method. The mean value of predicted VO2max (PVO2max) (42.8±4.0 ml · kg−1 · min−1 with a range of 33.7–50.9) showed a significant difference with VO2max (39.8±4.0 ml · kg−1 · min−1 with a range of 33.5–47.7) in the study group. Limits of agreement between PVO2max and VO2max were large enough (0.10 to 5.94 ml · kg−1 · min−1) with poor confidence intervals indicating inapplicability of the current protocol of CRT in the studied population. The prediction norm [Y = 21.01X – 11.04 (SEE = 0.193 ml · kg−1 · min−1)] was computed from the significant correlation (r = 0.93, P < 0.001) between distance covered in CRT and VO2max. Application of this norm in the confirmatory group revealed an insignificant difference between PVO2max and VO2max. The modified equation is recommended for application of CRT as a valid method to evaluate the cardiorespiratory fitness in terms of VO2max in sedentary male Indian youth.  相似文献

UV-VIS absorbance, inductively coupled plasma-optical emission spectroscopy (ICP-OES), and particle beam/hollow cathode-optical emission spectroscopy (PB/HC-OES) are presented as techniques for determining Cr3+ loading into transferrin (Tf), with and without Fe3+. The methods are compared based on loading percentages (i.e. 100% loading would be equal to 2 M(n+): 1 Tf) determined for Cr3+ loading into apo-transferrin. Spectral interferences and overlapping LMCT bands cause inaccurate chromium (qualitative) and iron (qualitative and quantitative) results for the UV-VIS absorbance method. The ICP-OES and PB/HC-OES methods are in good agreement providing evidence that the PB/HC-OES method is a valid technique for investigating metal-protein complexes. Maximum Cr3+ loading into apo-transferrin over a 24 h period was determined to be 26.8 3.5% by the ICP-OES method and 25.3 2.2% by the PB/HC-OES method. Loading percentages were increased to 49.7 1.9% (ICP-OES) and 55.7 3.2% (PB/HC-OES) when the metal-transferrin solution was allowed to incubate for up to 10 days. Under non-excess carbonate conditions the Cr3+ loading is elevated over a 24 h incubation time, but under physiological conditions the loading is inhibited. Equal loading of Fe3+ and Cr3+ into apo-transferrin was achieved when chromium was at a level more than 5 times in excess of iron. Inhibition of Cr3+ loading was only observed when an excess of Fe3+ was available to bind into apo-transferrin. The ability for Cr3+ to displace Fe3+ from holo-transferrin was observed as small amounts of Cr3+ were loaded into the once occupied metal binding site.  相似文献

MEGF10 functions as a receptor for the uptake of amyloid-β   总被引:1,自引:0,他引:1  
MEGF10 is predominantly expressed in the brain and known to function as a phagocytic receptor. Here, we provide evidence that MEGF10 is involved in the uptake of amyloid-β peptide (Aβ42) in the brain. Overexpression of MEGF10 dramatically increased Aβ42 uptake in Hela cells. Knockdown of endogenous MEGF10 expression significantly decreased Aβ42 uptake in N2A neuroblastoma cells. MEGF10-mediated Aβ uptake is mostly dependent on lipid raft endocytosis pathway. Furthermore, site-directed mutagenesis revealed that the conserved cytoplasmic NPxY and YxxØ motifs are crucial for MEGF10-mediated uptake of Aβ42 peptide. Thus, the identification of the MEGF10 as a functional receptor that mediates the uptake of amyloid-β peptide will help elucidate the molecular mechanisms of amlyoid-β clearance in Alzheimer’s disease.

Summary The kinetics of Cu uptake in nutritionally starved cells of the diazotrophic cyanobacteriumNostoc calcicola Bréb. have been compared with those in cells recovering from starvation. Unstarved cyanobacterial cells assimilated 97.0 nmol Cu mg–1 protein within 1 h when incubated in medium containing 40 M Cu. Uptake was markedly inhibited in carbon-starved cells and, to a lesser extent, in cells starved of nitrogen or sulphur. The intracellular concentrations of protein and photopigments were markedly lower in cells starved of carbon, nitrogen, sulphur or phosphorus, whilst that of carbohydrate was lower in cells starved of carbon, sulphur or phosphorus, but almost doubled in cells starved of nitrogen. The ability to assimilate Cu was partially restored in cells after 72 h of recovery from phosphorus or sulphur deprivation, but showed little improvement during recovery from carbon or nitrogen starvation. A possible role of phosphorus in regulating Cu transport and accumulation is discussed.  相似文献

Antioxidant enzymes in erythrocytes such as superoxide dismutase (SOD), catalase (CAT), glutathione peroxidase (GSH-Px) and antioxidant sex hormone oestradiol in serum and malondialdehyde (MDA) production as a marker of lipid peroxidation in erythrocytes were investigated in male and female underwater rugby (UWR) players. Results showed that except for GSH-Px activity in female players, all antioxidant enzymes increased significantly (p < 0.05) after an UWR game. It was interesting to note that while the concentration of oestradiol in female players did not change, it increased significantly (p < 0.05) in male players. Lipid peroxidation (LPO) levels in female players as well as oestradiol concentration did not change significantly (p > 0.05) whereas LPO levels in male players increased significantly (p < 0.05) compared to pre-exercise values. In conclusion, the results showed that underwater rugby can stimulate over-production of ROS and antioxidant systems and affect oestradiol levels in male players. Because of increased LPO levels observed in male players, complex antioxidant supplementation including co-factors of antioxidant enzymes such as Cu, Zn, Fe, Se and antioxidant vitamins such as vitamin C and E may be recommended to players before the UWR game.  相似文献

Using a photoacoustic technique it has been possible to observe fast oxygen evolution and uptake transients at a high time resolution (approx. 0.2 s), when a dark-adapted leaf is reilluminated. There is initially a rapid pulse of oxygen evolution, correlated with the initial fluorescence rise (total duration under the experimental conditions used about 1–2 s), corresponding presumably to the photoreduction of the plastoquinone pool. This phenomenon may be utilized to calibrate the oxygen-evolution photoacoustic signal.
