Modified d-glucose and d-mannose analogs are potentially clinically useful metabolic inhibitors. Biological evaluation of 2-deoxy-2-halo analogs has been impaired by limited availability and lack of efficient methods for their preparation. We have developed practical synthetic approaches to 2-deoxy-2-fluoro-, 2-chloro-2-deoxy-, 2-bromo-2-deoxy-, and 2-deoxy-2-iodo derivatives of d-glucose and d-mannose that exploit electrophilic addition reactions to a commercially available 3,4,6-tri-O-acetyl-d-glucal.  相似文献

Treatment of L-fucose with an excess of pyridine-sulphur trioxide gave an equilibrium mixture of mono-, di-,and tri-sulphates. L-Fucose was sulphated under optimal conditions for monosulphate formation, and the monoester fraction was isolated by chromatography on DEAE-cellulose. The isomeric L-fucose 2-, 3-, and 4-sulphates (1-3) were separated on a DEAE-cellulose column by elution with borate buffer. The structures of 1-3 were established by electrophoresis, colour tests, periodate oxidation, and, for the 2-isomer, by comparison with a specimen of 1 that had been definitively synthesised via methyl 3,4-O-isopropylidene-α-L-fucopyranoside (6) and methyl α-L-fucopyranoside 2-(barium sulphate) (5). The latter was rapidly hydrolysed in hot, dilute acetic acid to 1 and methyl α-L-fucopyranoside (4).  相似文献

Reaction of trifluoro(fluoroxy)methane at ca. −80° with 3,4,6-tri-O-acetyl-D-galactal affords trifluoromethyl 3,4,6-tri-O-acetyl-2-deoxy-2-fluoro-α-D-galactopyranoside (2, 39%), 3,4,6-tri-O-acetyl-2-deoxy-2-fluoro-α-D-galactopyranosyl fluoride (3, 37%), trifluoromethyl 3,4,6-tri-O-acetyl-2-deoxy-2-fluoro-α-D-talopyranoside (4, 3%), and 3,4,6-tri-O-acetyl-2-deoxy-2-fluor-α-D-talopyranosyl fluoride (5, 2%). The structures of compounds 25 have been established by n.m.r. spectroscopy. Acid hydrolysis of 2 or 3 allords 2-deoxy-2-fluoro-D-galactose.  相似文献

l-Alanylglycyl-l-alanylglycyl-l-alanylglycyl-l-serylglycine and its pentachlorophenyl ester methanesulphonate have been synthesized as monomers for the preparation of silk fibroin model polypeptide. The former octapeptide was polymerized with diphenylphosphorylazide (DPPA) and triethylamine in DMSO or in HMPA—pyridine, and the latter octapeptide pentachlorophenylester was polymerized by adding triethylamine in DMSO to give poly(l-alanylglycyl-l-alanylglycyl-l-alanylglycyl-l-serylglycine). This sequential polypeptide gave a similar i.r. pattern to the crystalline part of Bombyx mori silk fibroin, which indicated antiparallel β-conformation. Dialysis of the solution of this polymer in 60%, aqueous LiBr against water gave mainly the polymer of α-form. O.r.d. measurements suggest that this polypeptide exists as a random structure in dichloroacetic acid on in 60% aqueous LiBr.  相似文献

Filiferin B is identical to timosaponin A-III, which had previously been shown to be 3-O-[2-O-(β-d-glucopyranosyl)-β-d-galactopyranosyl]sarsasapogenin. A larger-scale isolation of filiferin B from the seeds of Yucca filifera led to the isolation of filiferin A, now shown to be 3-O-[2-O-β-d-xylopyranosyl)-β-d-galactopyranosyl]-sarsasapogenin. The presence of the xylose residue was established by way of hydrolysis. 8-Methoxycarbonyloctyl 2-O-(β-d-glucopyranosyl)-β-d-galactopyranoside was synthesized to serve as a model for interpretation of the 13C-n.m.r. spectrum of filiferin B. The information thus gained, together with the 13C-n.m.r. spectra of other, simple model-compounds, permitted assignment of the structure for filiferin A. 8-Methoxycarbonyloctyl 2-O-(α-d-glucopyranosyl)-β-d-galactopyranoside was also synthesized.  相似文献

The cotyledons, leaves and inflorescences of the radish, Raphanus sativus, have been examined for their content of hydroxylated cinnamic acid malate esters. 2-O-(p-coumaroyl)-l-malate, 2-O-caffeoyl-l-malate and 2-O-feruloyl-l-malate were found to be quantitatively predominating compounds in the fraction of carboxylic acids isolated from radish leaves and inflorescences. Identifications were based on PC, TLC, HVE, GC, HPLC, UV and NMR spectroscopy. The l- or (2S)-configuration of malic acid released by hydrolysis from the esters was determined by using l-malate dehydrogenase. A combined column chromatography technique applied prior to HVE, HPLC and GC was useful for separating the malate esters from most other low MW plant constituents. The significance of the present investigation is briefly discussed in relation to the metabolism of phenolic constituents of glucosinolate-containing plants, and the effects of these compounds in relation to insect feeding behaviour.  相似文献

Attempted cyclization of 2,3,4-tri-O-methyl-5-seleno-L-arabinose dimethyl acetal in acidic solution gave the corresponding diselenide. Intramolecular attack by the selenobenzyl group at C-5 of 5-O-p-tolylsulfonyl-L-arabinose dibenzyl diseleno-acetal resulted in the formation of benzyl 1,5-diseleno-L-arabinopyranoside. Similarly, 2,3,5-tri-O-methyl-4-O-p-tolylsulfonyl-D-xylose dibenzyl diselenoacetal gave benzyl 2,3,5-tri-O-methyl-1,4-diseleno-L-arabinofuranoside, and 2,3,4-tri-O-acetyl-5-O-p-tolylsulfonyl-D-xylose (or ribose) dibenzyl diselenoacetal gave benzyl 2,3,4-tri-O-acetyl-1,5-diseleno-D-xylo- (or ribo-)pyranoside. The glycosylic benzylseleno group was removed from the pyranoside with mercuric acetate, but attempted deacetylation of the product led to decomposition and not to the expected 5-seleno-D-xylopyranose.  相似文献
