真菌提取物JN219抗人巨细胞病毒机制的初步研究 总被引:1,自引:0,他引:1
目的探讨真菌提取物JN219的抗病毒机制。方法(1)样品对病毒入侵宿主细胞的抑制作用:将样品(JN219 50μg/ml、柱洗脱组分5 g/L)分别与人巨细胞病毒(HCMV半数组织感染量TCID50为105/ml)等容量混合,不同时间(混合后立刻、混合后37℃作用1 h)以不同稀释度、100μl/孔接种至多孔板人胚肺细胞(HEL)上;(2)样品对病毒复制的抑制作用:将HCMV(100个TCID50)100μl/孔接种于HEL上1 h后,加不同稀释度的样品;(3)灭活病毒对样品抗病毒作用的抑制作用:将紫外线灭活的HCMV与JN219、柱洗脱组分等容量混合,37℃作用1 h,加等容量HCMV(TCID50为105/ml)混合37℃作用1 h,以不同稀释度、100μl/孔接种于96孔板内的HEL上。以上各实验组同时设细胞对照组、病毒对照组、空白对照组、相应稀释度的样品对照组,37℃、5%CO2培养,每日观察细胞CPE,当病毒对照病变90%以上,终止培养,中性红染色,在540 nm读取吸光度A值,用Reed-Muench法计算细胞半数有效浓度(IC50)和病毒TCID50。通过比较组间差异,探讨在哪一复制周期样品对病毒产生抑制作用。结果(1)样品对病毒入侵宿主细胞的抑制作用:JN219、柱洗脱组分与HCMV等容量混合后立刻接种细胞,测得病毒TICD50为104.8/ml,与病毒对照组比较(TCID50为105/ml)差异无显著性;与病毒作用1 h后接种细胞,JN219、柱洗脱组分IC50分别为0.168μg/ml、0.185μg/ml,第一孔JN219 TCID50为101.6/ml,柱洗脱组分标本未测得病毒,可使HCMV得到抑制;(2)样品对病毒复制的抑制作用:先将病毒接种细胞1 h,然后加JN219、柱洗脱组分,IC50分别为3.29μg/ml、13.1 g/L,样品不能完全抑制人巨细胞病毒病变的发生;(3)灭活病毒对样品抗病毒作用的抑制作用:紫外线灭活的病毒与样品作用1 h,再加等容量病毒后接种细胞,样品JN219、柱洗脱组分与病毒混合物TCID50分别为103.5、103.75,样品抗病毒位点已被紫外线灭活病毒外膜蛋白占据,抗病毒效果降低。结论JN219抗人巨细胞病毒作用机制主要是阻断病毒入侵宿主细胞,作用位点主要为人巨细胞病毒外膜蛋白。 相似文献

本文分别采用环己烷、正丁醇、乙酸乙酯、丙酮、甲醇5种极性不同的溶剂分别提取青蒿内生真菌Alternaria sp.(PQH12)的菌丝体。采用MTT法分别测定不同极性溶剂提取物对3种肿瘤细胞株(HL-60、MCF-7和COLO205)的生长抑制率,采用DPPH自由基清除法和超氧阴离子O-·2清除法评价了青蒿内生真菌Alternaria sp.(PQH12)不同极性溶剂提取物的抗氧化活性。结果显示,正丁醇和乙酸乙酯提取物均表现出对肿瘤细胞生长的抑制作用;浓度为20μg/mL时,正丁醇提取物对HL-60细胞的生长抑制率为80.56%,乙酸乙酯提物对MCF-7和COLO205细胞的生长抑制率分别为54.47%和83.09%;在两种抗氧化活性评价体系中,甲醇提取物的抗氧化活性均明显优于其他溶剂的提取物,其IC50值分别为0.20 mg/mL(DPPH)和0.19 mg/mL(O-·2)。该结果表明,青蒿内生真菌Alternaria sp.(PQH12)具有较好的细胞毒和抗氧化潜力,具有一定的开发利用价值,可以作为进一步实验研究的对象。 相似文献

从南海沉积物中分离得到23株海洋真菌,通过ITS测序进行鉴定。以新月弯孢霉(Curvularia lunata)、柱枝双胞霉(Cylindrocladium scoparium)、链格孢(Alternaria alternata)、胶孢炭疽菌(Colletotrichum gloeosporioides)为受试植物病原真菌,以神经胶质瘤细胞(SF-268)、乳腺癌细胞(MCF-7)、大细胞肺癌细胞(NCI-H460)和肝癌细胞(HepG-2)为受试肿瘤细胞,分别采用生长速率法和SRB法对这些菌株的发酵液粗提物进行抗植物病原真菌和细胞毒活性测试,发现11个菌株的粗提物在浓度为50 mg/mL时,对至少1种受试植物病原真菌的抑制率在50%以上,有9个菌株的粗提物在浓度为100μg/mL时,对至少1种肿瘤细胞株的抑制率在80%以上,其中菌株Eupenicillium sp.FS100、Penicillium sp.FS105、Dichotomomyces cejpii FS110、Acaromyces ingoldii FS121对植物病原真菌和(或)肿瘤细胞具有明显的抑制活性。 相似文献

玉米(Zea mays L.)种子在萌发期间热激处理(42℃)时蛋白质合成率低于对照(25℃);高活力种子胚热激蛋白的合成率最高。在42℃热激处理时玉米种胚子合成的热激蛋白的分子量分别为73、65、62、54、18kD等5种。高活力种胚合成的热激蛋白在最上高于低活力种子,高活力种胚合成的特异性热激蛋白56kD可以作为衡量种子活力的指标。双向电泳表明高低活力种子间热激蛋白的合成有更多质上的差异。 相似文献

选用240种常用中草药,研究其蒸馏水和甲醇浸提液原液及其10倍、100倍和1 000倍稀释液对小列当种子发芽的影响.结果表明:小列当种子在蒸馏水浸提液刺激下发芽率超过70%的中草药有灯心草、降香和莲须,在甲醇浸提液刺激下发芽率超过70%的中草药有百部、桑椹、谷芽和萆薢;蒸馏水浸提液对小列当种子发芽抑制率超过50%的中草药有菊花、棕榈、苦杏仁和黄柏,甲醇浸提液对小列当种子发芽抑制率超过70%的有续断、地黄、肉豆蔻、地肤子、黄连、木贼和胡黄连.大部分中草药浸提液在10倍稀释液条件下对小列当种子发芽的刺激或抑制作用最明显.研究发现,一些参试中草药蒸馏水和甲醇浸提液能够刺激或抑制小列当种子萌发,并且其对种子萌发的影响程度随浸提液浓度的变化而异,不同中草药最佳处理浓度不同,而且用甲醇浸提液可筛选出更多种具有化感作用的中草药. 相似文献

植物的根际促生菌能使植物增产,其中菌群产生的挥发性成分起重要作用。3-甲基-1-丁醇(3-methyl-1-butanol),即异戊醇,作为一种常见的微生物挥发性有机成分,但3-甲基-1-丁醇对高等植物种子萌发和生长的影响尚鲜见报道。本文研究了3种浓度(1、10和100 mg·L-1)的3-甲基-1-丁醇对小麦和玉米种子萌发的影响。结果表明,1和10 mg·L-1的3-甲基-1-丁醇处理均可提高玉米和小麦种子的活力指数;在低浓度的3-甲基-1-丁醇处理下,玉米和小麦种子可溶性糖和可溶性蛋白质含量减少,而幼芽SOD活性和叶绿素含量比对照组显著增加。本文研究表明,3-甲基-1-丁醇在适宜的浓度下可以促进玉米和小麦生长。 相似文献

The effects of ultradry storage on the starch mobilization in maize (Zea mays L.) seed after aging were investigated. The results indicated that there were no significant differences in the content of ATP,starch, and soluble sugar, as well as the activity of amylase, between ultradried seeds and seeds stored at -20 ℃ during germination. These results were consistent with the higher level of vigor of the ultradried seed. Sieve tube introduction of a fluorescence dye (carboxyl fluoresceindiacetate) and laser confocal microscopy were used to study the development of plasmodesmata in the ultradried seeds. The results indicated that plasmodesmata developed well in ultradried seeds. Fluorescence analysis also showed that the fluorescence intensity in the radicle of ultradried seeds was stronger than that in seeds with a higher moisture content. This suggests that ultradry treatment has no adverse effects on the seeds. After seed imbibition, cell orgaelles could be resumed. It is concluded that ultradry seed storage is beneficial for maintaining seed vigor and that starchy mobilization proceeds regularly during germination. 相似文献

Biological Control of Stenocarpella maydis in Maize Seed with Antagonistic Streptomyces sp. Isolates
The effectiveness of two Streptomyces sp. isolates, isolated from maize rhizosphere soil and designated as DAUFPE 11470 and DAUFPE 14632, was evaluated in vitro and under greenhouse conditions for control of Stenocarpella maydis in maize seeds. Stenocarpella maydis incidence was detected in all subsamples of disease‐free maize seeds by in vitro survey test, and ranged from 10.8% to 65.2%. In a filter paper test with surface‐disinfected seeds inoculated with S. maydis, Streptomyces sp. isolates DAUFPE 11470 and DAUFPE 14632 significantly reduced (P ≤ 0.05) the pathogen incidence by 93.2% and 92.3%, respectively. Seed germination in the same treatments was increased by 30.0% and 28.2%, respectively. Treatments of non‐disinfected seeds with the isolates DAUFPE 11470 and DAUFPE 14632, under greenhouse conditions reduced disease incidence in the seedlings by 87.3% and 85.6%, respectively. The reductions in disease incidence in surface‐disinfected seeds were 85.0% and 83.0% for the same isolates. Seedling emergence significantly (P ≤ 0.05) increased in disinfected and non‐disinfected seeds inoculated with the Streptomyces sp. isolates. The results indicate the potential of using Streptomyces sp. isolates as an additional tool to control Stenocarpella ear rot by significantly reducing the incidence of S. maydis in maize seeds and seedlings. 相似文献

研究了不同光照和温度条件对大百合种子萌发的影响。结果表明:光照对大百合种子的萌发有明显促进作用,可提高种子萌发率。种子萌发率以24 h光照20℃恒温最佳。避光条件下温度对大百合种子萌发有影响,其种子萌发最适温度为20℃,11 d开始萌动,3周左右萌发完全,随着温度的升高或降低其种子萌发率下降。萌发前经不同预处理的大百合种子的萌发率不同。同一果实内中部种子萌发最好,上部种子次之,下部种子最差,上、中、下三部分种子萌发率与其千粒重呈正相关。 相似文献
