蛋白质组学的基础研究之一是蛋白质鉴定.规模化的蛋白质鉴定通常采用"鸟枪法",即选择一些酶切肽段(母离子)碎裂生成二级谱图,通过二级谱图及其母离子质量鉴定肽段,再推断对应的蛋白质.在鉴定过程中,母离子质量是一个关键参数.母离子是否是肽段的单同位素峰决定了正确肽段是否能进入候选,母离子的质量精度决定了候选肽段的数目.本文从判断单同位素峰和系统误差校准这两个角度研究了母离子的准确检测技术.判断单同位素峰的技术在蛋白质上已有研究,包括电荷判断、单同位素峰判断和重叠同位素峰判断.可以借鉴蛋白质水平的技术研究母离子的单同位素峰判断方法.同时母离子的系统误差校准也有较为成熟的方法.这两个角度的研究有助于提高规模化蛋白质的鉴定率.  相似文献

In the hemipteroid insects of the suborder Sternorrhyncha, B chromosomes are relatively common in comparison with other suborders of Hemiptera. However, the occurrence of supernumerary chromosomes is restricted, in most cases, to several genera or closely related species. At least in some species of Psylloidea with the XY sex determination system, a mitotically stable B chromosome integrated into an achiasmatic segregation system with the X, and became fixed as a Y chromosome. In some Aphidoidea with a multiple X system of sex determination, B chromosomes appear to be in fact non-functional X chromosomes. Supernumerary chromosomes thus probably play an important role in the evolution of sex determination systems in Sternorrhyncha.  相似文献

In Drosophila melanogaster, the mechanism of sex determination is substantially different in the germ line and in the soma. In the germ line, the process is not completely cell-autonomous, but requires some signals from the soma. Only some of the genes involved in somatic sex determination are also needed for germ cell development. Recent genetic studies have identified loci required for germ-line sex determination.  相似文献

The somatic integration definition of life is familiar from the debate on the determination of death, with some bioethicists arguing that it supports brain death while others argue that some brain‐dead bodies exhibit sufficient somatic integration for biological life. I argue that on either interpretation, the somatic integration definition of life implies that neither the preimplantation embryo nor the postimplantation embryo meet the somatic integration threshold condition for organismal human life. The earliest point at which a somatic integration determination of life could be made would be the beginning of the fetal stage, 9 weeks postfertilization. Bioethical implications are considered, specifically with respect to the moral status of the postimplantation embryo in embryo research and abortion.  相似文献

为了分析IVIG抗补体活性测定出现负值与绵羊红细胞质量的关系。采用CH50试验测定IVIG ACA时,在被检样品和其他试验条件相同的情况下,比较了被污染的和未污染的绵羊红细胞对IVIG抗补体活性测定的影响。结果显示,使用被污染的绵羊红细胞(作为试验材料时所得的)检测10份样品ACA(%)均为负值;使用未污染的绵羊红细胞未出现负值。提示被污染的绵羊红细胞干扰了抗补体活性测定,从而引起CH50试验结果出现偏差。  相似文献

通过回顾近年来以多种植物为材料进行的性染色体观察,性别决定基因及调控方式的研究,对植物性别决定的机制进行了初步探讨,从而可以看出不同植物具有不同的性别决定机制:对于有性染色体的植物而言,目前已经从Y染色体上分离和鉴定了许多与雄性发育紧密相关的基因;部分性别决定基因和调控序列已利用构建减法文库,诱导突变体等方法从一些植物中获得。此外,还有研究表明,DNA脱甲基化,以及某些激素(如赤霉素、乙烯、Ace)都对植物的性别决定有重要作用。  相似文献

The precipitate of zinc phosphate helps to remove from crude leaf extracts some but not all the interfering material. In some species of self-fertile pulmonate snails, two sexual morphs co-occur in natural populations: regular individuals and aphallic individuals that cannot transmit sperm to other snails. Purely aphallic populations therefore reproduce obligatorily by selfing. Understanding the evolution of aphally and selfing in these snails requires a precise knowledge of phally determination. In this paper, we investigate the genetic and environmental determination of aphally in Bulinus truncatus by a survey of the family (offspring) aphally ratio of 233 individuals originating from seven natural populations and a study of the reaction norm of the family aphally ratio to temperature using 60 individuals from 10 selfed lineages of one population. Our results indicate a high genetic variability for the determination of aphally between populations and within some populations, associated with a high level of genetic determination. Our second experiment indicates a significant temperature and lineage effect though no interaction between these two effects. We discuss our results in the framework of threshold models developed for dimorphic traits with polygenic inheritance. We propose that the sexual morph of an individual at a given temperature is determined by a temperature threshold value depending on both the individual genotype and probabilistic processes.  相似文献

Sex is prevalent in nature and sex determination is one of the most fundamental biological processes, while the way of initiating female and male development exhibits remarkable diversity and variability across vertebrates. The knowledge on why and how sex determination mechanisms evolve unusual plasticity remains limited. Here, we summarize sex determination systems, master sex-determining genes and gene-regulatory networks among vertebrates. Recent research advancements on sex determination system transition are also introduced and discussed in some non-model animals with multiple sex determination mechanisms. This review will provide insights into the origin, transition and evolutionary adaption of different sex determination strategies in vertebrates, as well as clues for future perspectives in this field.  相似文献

The development of primordia as leaves, petals, or as organsintermediate between leaves and petals can be regulated by photoperiodin Impatiens. In intermediate organs only some parts of theorgan differentiated as petal, and then only in some cell layers.Allometric measurements of primordium shape suggested that intermediateorgans may begin development as petals, and that their intermediatecharacter at maturity resulted from a switch of some parts ofthe organs from petal to leaf development when the primordiawere between 0.5 and 1 mm long. In reverted apices made to re-flower,primordia were not completely determined as leaves until theywere about 750 µm long. Determination typically occurredfirst at the tips and last at the bases of these primordia.The determination of primordia as leaves or petals in Impatiensis discussed in relation to primordium determination in otherspecies. It is suggested that the lack of commitment to flowermay result in relatively late primordium determination in Impatiens. Interference of low concentrations of Hepes and other buffers commonly used in protein determination was studied. The data show that some of these buffers interfere to differing degrees with protein determination according to the Lowry method. A study of the structure-interference relationship suggests that the group ethanolamine is involved in this interference. No interference was observed when protein was measured using bicinchonic acid at the same concentration as the Lowry reagent.  相似文献

Although gonadogenesis has been extensively studied in vertebrates with genetic sex determination, investigations at the molecular level in nontraditional model organisms with temperature-dependent sex determination are relatively new areas of research. Results show that while the key players of the molecular network underlying gonad development appear to be retained, their functions range from conserved to novel roles. In this review, we summarize experiments investigating candidate molecular players underlying temperature-dependent sex determination. We discuss some of the problems encountered unraveling this network, pose potential solutions, and suggest rewarding future directions of research.  相似文献

Early mammalian development is regulative - it is flexible and responsive to experimental intervention. This flexibility could be explained if embryogenesis were originally completely unbiased and disordered; order and determination of cells only arising later. Alternatively, regulative behaviour could be consistent with the embryo having some order or bias from the very beginning, with inflexibility and cell determination increasing steadily over time. Recent evidence supports the second view and indicates that the sequence and the orientations of cell divisions help to build the first asymmetries.  相似文献

A hypothesis on the evolutionary origin of the genetic pathway of sex determination in the nematode Caenorhabditis elegans is presented here. It is suggested that the pathway arose in steps, driven by frequency-dependent selection for the minority sex at each step, and involving the sequential acquisition of dominant negative, neomorphic genetic switches, each one reversing the action of the previous one. A central implication is that the genetic pathway evolved in reverse order from the final step in the hierarchy up to the first. The possible applicability of the model to the other well-characterized sex determination pathway, that of Drosophila melanogaster, and to sex determination in mammals, is discussed, along with some potential implications for pathway evolution in general. Finally, the specific molecular and population genetic questions that the model raises are described and some tests are proposed.  相似文献

The degree of genetic determination of 25 quantitative dermatoglyphic characteristics has been studied on family: twin material: 45 pairs of MZ and 75 single-sex DZ twins; and 53 single-sex "parent-child" pairs. Approximating formulae were used to estimate main components of phenotypic variance due to additive interaction of genetic factors, to non-linear effects (intralocus dominance) and to the effect of total-familiar and random environmental factors. All the finger dermatoglyphic characteristics studied had a high degree of genetic determination (G greater than 0,80), and for most of them the contribution into the large variance of intralocus dominance effects was comparable with that of additive gene interaction, included in the determination of these characters. There are some palm dermatoglyphic characteristics ("ad" distance "cd" comb counting, "bad", "adt" and "cda" angles), which degree of genetic determination is low (G less than 0,35). At least ten quantitative finger and palm dermatogliphic characteristics with a high degree of genetic determination can be used for special studies in frames of multidimentional genetical analysis, including determination of twin zygosity type. Earlier described "indices" (using twin data) of relative role of genetic and environment factors in the determination of populational variability of quantitative characters are considered. None of them is shown to be a reliable estimate of the coefficient of genetic character determination. The use of these indices in practical studies can result in wrong conclusions on the degree and the character of genetic determination of quantitative characters.  相似文献

Screening plant extracts for antifungal activity is increasing due to demand for new antifungal agents, but the testing methods present many challenges. Standard broth microdilution methods for antifungal susceptibility testing of available antifungal agents are available now, but these methods are optimised for single compounds instead of crude plant extracts. In this study we evaluated the standard NCCLS method as well as a modification which uses spectrophotometric determination of the end-points with a plate reader. We also evaluated another standard method, the EUCAST method, which is a similar microdilution assay to the NCCLS method, but uses a larger inoculum size and a higher glucose concentration in the medium as well as spectrophotometric end-point determination. The results showed that all three methods had some drawbacks for testing plant extracts and thus we modified the NCCLS broth microdilution method by including a colorimetric indicator-resazurin for end-point determination. This modified method showed good reproducibility and clear-cut end-point, plus the end-point determination needed no instruments. It enabled us to evaluate the activity of a selection of extracts from six Combretaceous plants against three Candida spp. and thus provided pharmacological evidence for some traditional uses of these plants while assisting the identification of the active ingredients.  相似文献

An improved enzymatic method for the determination of pyrroloquinoline quinone, a novel prosthetic group of some important oxidoreductases, has been developed with cytoplasmic membrane of Escherichia coli K-12, in which D-glucose dehydrogenase (EC 1.1.99.17) was completely resolved to apo-enzyme by EDTA treatment. Incubation of the EDTA-treated membrane with exogenous pyrroloquinoline quinone in the presence of magnesium ions gave a quantitative determination of pyrroloquinoline quinone by assaying the restored D-glucose dehydrogenase activity. This novel enzymatic method was confirmed to be highly reproducible up to 10 ng of pyrroloquinoline quinone and could be applied to a routine assay of pyrroloquinoline quinone.  相似文献
