Nitrogen Dynamics in the Steeply Stratified,Temperate Lake Verevi,Estonia   总被引:2,自引:0,他引:2  
The dynamics of different nitrogen compounds and nitrification in diverse habitats of a stratified Lake Verevi (Estonia) was investigated in 2000–2001. Also planktonic N2-fixation (N2fix) was measured in August of the observed years. The nitrogen that accumulated in the hypolimnion was trapped in the non-mixed layer during most of the vegetation period causing a concentration of an order of magnitude higher than in the epilimnion. The ammonium level remained low in the epilimnion (maximum 577 mgN m−3, average 115 mgN m−3) in spite of high concentrations in the hypolimnion (maximum 12223 mgN m−3, average 4807 mgN m−3). The concentrations of NO2 and NO3 remained on a low level both in the epilimnion (average 0.94 and 9.09 mgN m−3, respectively) and hypolimnion (average 0.47 and 5.05 mgN m−3, respectively). N2fix and nitrification ranged from 0.30 to 2.80 mgN m−3 day−1 and 6.0 to 107 mgN m−3 day−1, respectively; the most intensive processes occurred in 07.08.00 at depths of 2 and 5 m, accordingly. The role of N2fix in the total nitrogen budget of Lake Verevi (in August 2000 and 2001) was negligible while episodically in the nitrogen-depleted epilimnion the N2fix could substantially contribute to the pool of mineral nitrogen. Nitrification was unable to influence nitrogen dynamics in the epilimnion while some temporary coupling with ammonium dynamics in the hypolimnion was documented.  相似文献

The vertical and temporal distribution of metazooplankton in the small hypertrophic, strongly stratified, temperate Lake Verevi (Estonia) was studied during 1998–2001. The zooplankton of Lake Verevi is characteristic of hypertrophic lakes, with a small number of dominant species, rotifers being the main ones, and juveniles prevailing among copepods. In 1999–2001, the average abundance of metazooplankton in the lake was 1570 × 103 ind m−3; in the epilimnion 2320 × 103 ind m−3, in the metalimnion 2178 × 103 ind m−3, and in the hypolimnion 237 × 103 ind m−3. The average biomass of metazooplankton was 1.75 g m−3; in the epi-, meta- and hypolimnion, accordingly, 2.16, 2.85 and 0.26 g m−3. The highest abundances – 19,136 × 103 ind m−3 and 12,008 × 103 ind m−3 – were registered in the lower half of the metalimnion in 24 May and 5 June 2001, respectively. Rotifer Keratella cochlearis f. typica (Gosse, 1851) was the dominating species in abundance. In biomass, Asplanchna priodonta Gosse, 1850, among the rotifers, and Eudiaptomus graciloides (Lilljeborg, 1888), among the copepods, dominated. According to the data from 2000–2001, the abundance and biomass of both copepods and rotifers were highest in spring. Zooplankton was scarce in the hypolimnion, and no peaks were observed there. During the summers of 1998 and 1999, when thermal stratification was particularly strong, zooplankton was the most abundant in the upper half of the metalimnion, and a distinct peak of biomass occurred in the second fourth of the metalimnion. Probably, the main factors affecting the vertical distribution of zooplankton in L. Verevi are fish, Chaoborus larvae, and chemocline, while food, like phytoplankton, composition and abundance may affect more the seasonal development of zooplankton.  相似文献

An overview on studies of macrozoobenthos in the small, hard-water, stratified and hypertrophic Lake Verevi (South-Eastern Estonia) is given. The list of macroinvertebrates comprises at least 105 taxa. In the open water habitats, the biomass and abundance of macrozoobenthos (except the phantom midge Chaoborus flavicans) was rather constant beginning from the epilimnion up to the upper hypolimnion (depth 2–4 m), but very low in the lower hypolimnion (depth 6 m), which was inhabited mainly by Chaoborus. Comparison with long-term reference data from other Estonian lakes, belonging to similar limnological types, indicated that the total biomass and abundance (without Chaoborus) in the profundal of Verevi were very low.  相似文献

Primary Production of Phytoplankton in a Strongly Stratified Temperate Lake   总被引:7,自引:7,他引:0  
Lake Verevi (12.6 ha, maximum depth 11.0 m, mean depth 3.6 m) is a strongly eutrophic and stratified lake. Planktothrix agardhii is the most characteristic phytoplankton species in summer and autumn, while photosynthesizing sulphur bacteria can occur massively in the metalimnion. Primary production (PP) and chlorophyll a concentration (Chl a) were seasonally studied in 1991, 1993, 2000, and 2001. Vertical distribution of PP was rather complex, having usually two peaks, one at or near the surface (0–1 m), and another deeper (at 3–7 m) in the metalimnion. The values of dark fixation of CO2 in the metalimnion were in most cases higher than those in the upper water layer. Considering the average daily PP 896 mg C m−2 and yearly PP 162 mg C m−2, Secchi depth 2.34 m, and epilimnetic concentrations of chlorophyll a (19.6 mg m−3), total nitrogen and total phosphorus (TP, 52 mg m−3) in 2000, L. Verevi is a eutrophic lake of a ‘good’ status. Considering the total amounts of nutrients stored in the hypolimnion, the average potential concentrations in the whole water column could achieve 1885 mg m−3 of TN and 170 mg m−3 of TP reflecting hypertrophic conditions and a ‚bad’ status. Improvement of the epilimnetic water quality from the 1990s to the 2000s may have resulted from incomplete spring mixing and might not reflect the real improvement. A decreased nutrient concentration in the epilimnion has supported the establishment of a ‘clear epilimnion state’ allowing light to penetrate into the nutrient-rich metalimnion and sustaining a high production of cyanobacteria and phototrophic sulphur bacteria.  相似文献

The surface level of Lake Kinneret is regulated to remain between –209 m and –213 m msl. During the stratified period, soluble reactive phosphorus (SRP), ammonium (NH inf4 sup+ ) and dissolved sulphide (H2S) accumulate in the hypolimnion. The concentration of these solutes, which are direct and indirect products of the decomposition of organic matter, increase considerably in summers with lower lake levels. A numerical model describing depth-averaged hypolimnion and epilimnion current velocities for high and low lake levels was adapted for Lake Kinneret. Simulated hypolimnetic currents were shown to be stronger for low lake levels as a result of the fact that low lake levels are characterized by a thinner hypolimnion while the thickness of the epilimnion remains unchanged. We suggest that the stronger hypolimnetic currents have the following consequences: 1. turbulence is induced, 2. the enhanced turbulence results in higher resuspension, 3. because SO4= is available to bacteria on resuspended particles, mineralization rates are enhanced, and 4. focusing of fine sediments and associated organic matter to the pelagic zone is enhanced.  相似文献

We analysed the long-term dynamics (1980–2007) of hypolimnetic and epilimnetic bacterial abundances and organic carbon concentrations, both dissolved (DOC) and particulate (POC), in the deep holo-oligomictic Lake Maggiore, included in the Southern Alpine Lakes Long-Term Ecological Research (LTER) site. During the 28 years of investigation, bacterial abundance and POC concentrations did not decrease with declining phosphorus concentrations, while DOC concentrations showed a pronounced decrease in the epi- and hypolimnion. We used the annual mean total lake heat content and total annual precipitation as climate-related variables, and in-lake total phosphorus as a proxy for trophic state. The model (forward stepwise regression, FSR) showed that reduced anthropogenic pressure was more significant than climate change in driving the trend in DOC concentrations. Bacterial dynamics in the hypolimnion mirrored the fluctuations observed in the epilimnion, but average cell abundance was three times lower. The FSR model indicates that bacterial number variability was dependent on POC in the epilimnion and DOC in the hypolimnion. In the hypolimnion, cell biovolumes for rod and coccal morphotypes were significantly larger than in the epilimnion.  相似文献

The effects of viral lysis and heterotrophic nanoflagellate (HNF) grazing on bacterial mortality were estimated in a eutrophic lake (Lake Plußsee in northern Germany) which was separated by a steep temperature and oxygen gradient into a warm and oxic epilimnion and a cold and anoxic hypolimnion. Two transmission electron microscopy-based methods (whole-cell examination and thin sections) were used to determine the frequency of visibly infected cells, and a model was used to estimate bacterial mortality due to viral lysis. Examination of thin sections also showed that between 20.2 and 29.2% (average, 26.1%) of the bacterial cells were empty (ghosts) and thus could not contribute to viral production. The most important finding was that the mechanism for regulating bacterial production shifted with depth from grazing control in the epilimnion to control due to viral lysis in the hypolimnion. We estimated that in the epilimnion viral lysis accounted on average for 8.4 to 41.8% of the summed mortality (calculated by determining the sum of the mortalities due to lysis and grazing), compared to 51.3 to 91.0% of the summed mortality in the metalimninon and 88.5 to 94.2% of the summed mortality in the hypolimnion. Estimates of summed mortality values indicated that bacterial production was controlled completely or almost completely in the epilimnion (summed mortality, 66.6 to 128.5%) and the hypolimnion (summed mortality, 43.4 to 103.3%), whereas in the metalimnion viral lysis and HNF grazing were not sufficient to control bacterial production (summed mortality, 22.4 to 56.7%). The estimated contribution of organic matter released by viral lysis of cells into the pool of dissolved organic matter (DOM) was low; however, since cell lysis products are very likely labile compared to the bulk DOM, they might stimulate bacterial production. The high mortality of bacterioplankton due to viral lysis in anoxic water indicates that a significant portion of bacterial production in the metalimnion and hypolimnion is cycled in the bacterium-virus-DOM loop. This finding has major implications for the fate and cycling of organic nutrients in lakes.  相似文献

The small strongly stratified hard-water hypertrophic lake Verevi (max. depth 11.0 m, surface area 12.6 ha, mean depth 3.6 m) was investigated in 2000 and in 2001. The lake is sheltered from winds, and the role of waves in mixing the water column is minimal. Eutrophication favours the strengthening of stratification. Early warm springs cause a fast stagnation of the water column forming partly meromictic conditions. Seston content of water and in sediment traps in 3 layers was measured several times during the formation of stratification. Besides measuring particulate matter, in 2001, the nutrient content of the trapped sediment was analysed. During the first 7 days of the investigation, 30% of the total particle sedimentation took place. The sedimentation rate of particulate matter was 0.4–6.3 g m–2 d−1 dry weight in different layers of the water column. Daily average sedimentation loss rate was 27% of the total amount of seston of the epilimnion, whilst from the meta- and hypolimnion the settling was much slower (9.6 and 7.3%, respectively). In our experiments with twin sediment traps, to one of which formaldehyde was added, the PO43−-P concentration was 19% smaller in the trap without formaldehyde, probably due to planktonic uptake. The relationship between primary and export production is loop-like. The shape was irregular, indicating a high grazing rate of zooplankton.  相似文献

Bacterioplankton biomass and dark fixation of inorganic carbon were measured in the highly humic (water colour up to 550 mg Pt l?1) and acidic lake, Mekkojärvi. Strong thermal and chemical stratification developed in the water column early in spring and led rapidly to anoxia in the hypolimnion, which extended to less than 1.0 m from the surface. In the epilimnion only small bacteria were abundant. In the anoxic zone both the abundance and the mean size of bacteria were considerably higher than in the epilimnion. These differences are thought to be the result of different grazing pressure from zooplankton in the two zones. In late summer a high concentration of bacteriochlorophyll d in the upper hypolimnion indicated a high density of photosynthetic bacteria. Bacterial biomass was similar to that of phytoplankton in the epilimnion, but 23 times higher in the whole water column. In August, dark fixation of inorganic radiocarbon in the anaerobic zone was 51% of the total 14C-incorporation and the contribution of light fixation was only 5.4%. In the polyhumic Mekkojarvi, bacterioplankton was evidently a potentially significant carbon source for higher trophic levels, but bacterioplankton production could not be supported by phytoplankton alone. Allochthonous inputs of dissolved organic matter probably support most of the bacterial production.  相似文献

The population ecology of bacterioplankton was studied overa 3 year period in Mirror Lake, an oligotrophic lake in thenortheastern USA. Bacterial population density, biomass, andrates of biomass production in the epilimnion and hypolimnionwere examined for their relationship with several environmentalparameters. Bacterioplankton density fluctuated between 0.5and 7 x l0 bacteria ml–1, with highest values in the anoxichypolimnion. At all depths there was a trend towards a higherdensity of bacteria from spring to midsummer, followed by adecline in late summer to early autumn. Cocci tended to dominatebacterial cell shapes from winter to midsummer, after whichrod-shaped cells became most abundant. Rod-shaped cells contributedthe most to bacterioplankton biomass at all depths and timesof year. The mean annual biovolume of all bacterioplankton was0.12 µ cell–1. The mean annual areal bacterioplanktonbiomass was 11–12 mmol C m–2. The percentage ofbacterial to phytoplankton biomass per volume in summertimewas 27% in the epilimnion and 11% in the hypolimnion. Averageannual and summertime bacterial production estimated using the[3H]thymidine method was similar to previous estimates of bacterialproduction measured in Mirror Lake using other methods. Theaverage ratio of bacterial to net phytoplankton production pervolume was 0.34 in the epilimnion, and between 0.65 and 1 1.depending on depth, in the hypolimnion during summer. Of severalvariables considered in regression analyses, only temperatureexplained >50% of the variance in bacterial production inboth the hypolimnion and epilimnion. Above 14°C, however,bacterial production and growth rate in the epilimnion werenot clearly related to temperature. During the period of midsummerhypolimnetic anoxia, despite colder temperatures in the hypolimnion,bacterial production was up to 10 times greater than in theepilimnion.  相似文献

Seasonal population dynamics and the vertical distribution of planktonic ciliates in a hypertrophic and strongly stratified temperate lake were studied from April to October in 2000 and from April to June in 2001. In the epi- and metalimnion the ciliate abundance peaked in spring and late summer, reaching maximum values in the metalimnion (86 cells ml−1) on 7th August 2000. In the epilimnion, the highest biomass content (414 μg C l−1) was observed on 8th May 2000. In the hypolimnion only a late summer peak occurred and the ciliate numbers were always lower than in the epi- and metalimnion. Five groups dominated the community of ciliates: Oligotrichida, Gymnostomatea, Prostomatida, Hymenostomata and Peritrichia, and the community composition varied greatly with depth. In the epilimnion the ciliate numbers were dominated by oligotrichs but small algivorous prostomatids, peritrichs and gymnostomes were also numerous. In the metalimnion these groups were gradually replaced by scuticociliates and mixotrophic Coleps spp. In the hypolimnion scuticociliates and species known as benthic migrants dominated. In the epilimnion and upper metalimnion in spring large herbivores and in summer small bacterivores were more numerous.  相似文献

During summer-stratification in Lake Vechten (May–October) a great number of chemical parameters show variations. The concentration patterns of calcium, manganese and iron have been studied in some detail.The calcium concentration in the epilimnion is controlled by carbonate-formation; in the hypolimnion however, no saturation with respect to calcium-carbonate is reached, due to the fact, that carbondioxide equilibrium with the atmosphere no longer exists. The concentration pattern in the hypolimnion (max. concentration about 80 mg.l–1 near the sediment, 40 mg.l–1 in epilimnion) is controlled by the velocity of calcium transport from the sediment and perhaps by dissolution of precipitating carbonates from the epilimnion.The maximum manganese concentration in the hypolimnion (about 5 mg.l–1) is controlled by the solubility of manganese carbonate, causing, together with manganese transport from the sediment, a uniform concentration up to two meters above the bottom. In the epi- and meta-limnion the presence of oxygen is limiting the concentration of manganese.The iron concentration in the hypolimnion shows a strong gradient existing until the overturn (max. concentration near the sediment about 20 mg.l–1) and is controlled by a number of mechanisms: 1o. Oxidation by oxygen and manganese oxides near the epilimnion. 2o. Precipitation by sulphide formation; a strong correlation exists at different depths between sulphate disappearance and a slower increase of iron concentration. 3o. Transport from the sediment.Control of iron concentration by carbonate formation is unlikely, as the solubility product of iron carbonate is greatly exceeded (10–100 fold). The relative importance of the other processes is not yet known.To describe iron- and manganese dynamics in relation to the cycle of reducing power, generated in primary production we need data about precipitating quantities of iron and manganese compounds, for which suitable sediment-traps must be developed.  相似文献

In stratified Lake Vechten, The Netherlands, protozoan grazing was estimated on the basis of uptake of fluorescently labeled bacteria and compared with bacterial production estimated on the basis of thymidine incorporation. By using a grazer-free mixed bacterial population from the lake in continuous culture, an empirical relationship between cell production and thymidine incorporation was established. Thymidine incorporation into total cold-trichloroacetic-acid-insoluble macromolecules yielded a relatively constant empirical conversion factor of ca. 1018 (range, 0.38 × 1018 to 1.42 × 1018) bacteria mol of thymidine−1 at specific growth rates (μ) ranging from 0.007 to 0.116 h−1. Although thymidine incorporation has been assumed to measure DNA synthesis thymidine incorporation appeared to underestimate the independently measured bacterial DNA synthesis by at least 1.5- to 13-fold, even if all incorporated label was assumed to be in DNA. However, incorporation into DNA was found to be insignificant as measured by conventional acid-base hydrolysis. Methodological problems of the thymidine technique are discussed. Like the cultures, Lake Vechten bacteria showed considerable thymidine incorporation into total macromolecules, but no significant incorporation into DNA was found by acid-base hydrolysis. This applied not only to the low-oxygen hypo- and metalimnion but also to the aerobic epilimnion. Thus, the established empirical conversion factor for thymidine incorporation into total macromolecules was used to estimate bacterial production. Maximum production rates (141 × 106 bacteria liter−1 h−1; μ, 0.012 h−1) were found in the metalimnion and were 1 order of magnitude higher than in the epi- and hypolimnion. In all three strata, the estimated bacterial production was roughly balanced by the estimated protozoan grazing. Heterotrophic nanoflagellates were the major consumers of the bacterial production and showed maximum numbers (up to 40 × 106 heterotrophic nanoflagellates liter−1) in the microaerobic metalimnion.  相似文献

The vertical distributions of bacteria and algae in a steeply stratified, highly humic lake were studied during three 24 h periods in summer. The highest bacterial and algal densities and biomasses were recorded in the anoxic hypolimnion. The bacterial biomass in the hypolimnion was composed mainly of photosynthetic green sulphur bacteria (Chlorobium) which occurred at very low light intensity (< 1.5 μmol m−2 s−1). The numbers and biomasses of bacteria, both in the epilimnion and hypolimnion, were mostly higher at night than during the day, indicating possible asynchrony between the production and loss of bacteria. Because of vertical migration, the diurnal vertical distribution of algae was more variable than that of bacteria. Particularly in July and August, when cryptomonads were abundant, the biomass of algae was much higher in the epilimnion during the day than at night. The flagellated chlorophytes, Chlamydomonas spp. and Scourfieldia cordiformis, stayed mainly in the upper hypolimnion close to the oxic-anoxic boundary zone where only a small proportion of Daphnia longispina was continuously present. Unpalatable Mallomonas chrysophytes with silicified plates and bristles, and small, presumably heterotrophic, flagellates stayed in the oxic epilimnion together with a dense (up to 300 ind l−1) population of D. longispina. The results indicated that, besides the physical and chemical properties of the water column, grazing pressure by Daphnia longispina strongly affected the vertical distribution of microorganisms in this polyhumic lake.  相似文献

Walker Lake (area = 140 km2, Z mean = 19.3 m) is a large, terminal lake in western Nevada. As a result of anthropogenic desiccation, the lake has decreased in volume by 75% since the 1880s. The hypolimnion of the lake, now too small to meet the oxygen demand exerted by decaying matter, rapidly goes anoxic after thermal stratification. Field and laboratory studies were conducted to examine the feasibility of using oxygenation to avoid hypolimnetic anoxia and subsequent accumulation of ammonia in the hypolimnion, and to estimate the required DO capacity of an oxygenation system for the lake. The accumulation of inorganic nitrogen in water overlaying sediment was measured in laboratory chambers under various DO levels. Rates of ammonia accumulation ranged from 16.8 to 23.5 mg-N m–2 d–1 in chambers with 0, 2.5 and 4.8 mg L–1 DO, and ammonia release was not significantly different between treatments. Beggiatoa sp. on the sediment surface of the moderately aerated chambers (2.5 and 4.8 mg L–1 DO) indicated that oxygen penetration into sediment was minimal. In contrast, ammonia accumulation was reversed in chambers with 10 mg L–1 DO, where oxygen penetration into sediment stimulated nitrification and denitrification. Ammonia accumulation in anoxic chambers (18.1 and 20.6 mg-N m–2 d–1) was similar to ammonia accumulation in the hypolimnion from July through September of 1998 (16.5 mg-N m–2 d–1). Areal hypolimnetic oxygen demand averaged 1.2 g O2 m–2 d–1 for 1994–1996 and 1998. Sediment oxygen demand (SOD) determined in experimental chambers averaged approximately 0.14 g O2 m–2 d–1. Continuous water currents at the sediment-water interface of 5–6 cm s–1 resulted in a substantial increase in SOD (0.38 g O2 m–2 d–1). The recommended oxygen delivery capacity of an oxygenation system, taking into account increased SOD due to mixing in the hypolimnion after system start-up, is 215 Mg d–1. Experimental results suggest that the system should maintain high levels of DO at the sediment-water interface (10 mg L–1) to insure adequate oxygen penetration into the sediments, and a subsequent inhibition of ammonia accumulation in the hypolimnion of the lake.  相似文献

Mining of pyrite minerals is a major environmental issue involving both biological and geochemical processes. Here we present a study of an artificial lake of a former uranium open pit mine with the aim to connect the chemistry and bacterial community composition (454-pyrosequencing of 16S rRNA genes) in the stratified water column. A shift in the water chemistry from oxic conditions in the epilimnion to anoxic, alkaline, and metal and sulfide-rich conditions in the hypolimnion was corresponded by a strong shift in the bacterial community, with few shared operational taxonomic units (OTU) between the water layers. The epilimnetic bacterial community of the lake (~20?years old) showed similarities to other temperate freshwater lakes, while the hypolimnetic bacterial community showed similarity to extreme chemical environments. The epilimnetic bacterial community had dominance of Actinobacteria and Betaproteobacteria. The hypolimnion displayed a higher bacterial diversity and was dominated by the phototrophic green sulphur bacterium of the genus Chlorobium (ca. 40?% of the total community). Deltaproteobacteria were only represented in the hypolimnion and the most abundant OTUs were affiliated with ferric iron and sulfate reducers of the genus Geobacter and Desulfobulbus, respectively. The chemistry is clearly controlling, especially the hypolimnetic, bacterial community but the community composition also indicates that the bacteria are involved in metal cycling in the lake.  相似文献

The present study describes generally the ecosystem of Lake Verevi while more detailed approaches are presented in the same issue. The main task of the article is to estimate long-term changes and find the best method for the restoration of good ecological status. Lake Verevi (surface 12.6 ha, mean depth 3.6 m, maximum depth 11 m, drainage area 1.1 km2, water exchange 0.63-times per year) is a hypertrophic hard-water lake located in town Elva (6400 inhabitants). Long-term complex limnological investigations have taken place since 1929. The lake has been contaminated by irregular discharge of urban wastewaters from oxidation ponds since 1978, flood from streets, and infiltrated waters from the surrounding farms. The so-called spring meromixis occurred due to extremely warm springs in recent years. The index value of buffer capacity of Lake Verevi calculated from natural conditions is on the medium level. Water properties were analysed according to the requirements of the EU Water Framework Directive. According to the classification, water quality as a long-term average of surface layers is moderate-good, but the water quality of bottom layers is bad. Values in deeper layers usually exceed 20–30 times the calculated reference values by Vighi and Chiaudani’s model. Naturally, at the beginning of the 20th century the limnological type of the lake was moderately eutrophic. During the 1980s and 1990s the ecosystem was out of balance by abiotic characteristics as well as by plankton indicators. Rapid fluctuations of species composition and abundance can be found in recent years. Seasonal variations are considerable and species composition differs remarkably also in the water column. The dominating macrophyte species vary from year to year. Since the annual amount of precipitation from the atmosphere onto the lake surface is several times higher, the impact of swimmers could be considered irrelevant. Some restoration methods were discussed. The first step, stopping external pollution, was completed by damming the inlet. Drainage (siphoning) of the hypolimnetic water is discussed. The purpose of this study was to determine the depth distribution of bacterial biomass and production in a stratified lake and to test techniques to measure bacterial production in anaerobic waters. Bacterial abundance and incorporation of both [3H]thymidine and [3H]leucine into protein were highest in the metalimnion, at the depth at which oxygen first became unmeasurable. In contrast, [3H]thymidine incorporation into DNA was highest in the epilimnion. The ratios of incorporation into DNA/protein averaged 2.2, 0.49, and 0.95 for the epilimnion, metalimnion, and hypolimnion, respectively. Low incorporation into DNA was not due to artifacts associated with the DNA isolation procedure. Recovery of added [3H]DNA was about 90% in waters in which the portion of [3H]thymidine incorporation into DNA was about 40%. At least some obligate anaerobic bacteria were capable of assimilating thymidine since aeration of anaerobic hypolimnion waters substantially inhibited thymidine incorporation. The depth profile of bacterial production estimated from total thymidine and leucine incorporation and the frequency of dividing cells were all similar, with maximal rates in the metalimnion. However, estimates of bacterial production based on frequency of dividing cells and leucine incorporation were usually significantly higher than estimates based on thymidine incorporation (using conversion factors from the literature), especially in anaerobic hypolimnion waters. These data indicate that the thymidine approach must be examined carefully if it is to be applied to aquatic systems with low oxygen concentrations. Our results also indicate that the interface between the aerobic epilimnion and anaerobic hypolimnion is the site of intense bacterial mineralization and biomass production which deserves further study.  相似文献

The effects of viral lysis and heterotrophic nanoflagellate (HNF) grazing on bacterial mortality were estimated in a eutrophic lake (Lake Plusssee in northern Germany) which was separated by a steep temperature and oxygen gradient into a warm and oxic epilimnion and a cold and anoxic hypolimnion. Two transmission electron microscopy-based methods (whole-cell examination and thin sections) were used to determine the frequency of visibly infected cells, and a model was used to estimate bacterial mortality due to viral lysis. Examination of thin sections also showed that between 20.2 and 29.2% (average, 26.1%) of the bacterial cells were empty (ghosts) and thus could not contribute to viral production. The most important finding was that the mechanism for regulating bacterial production shifted with depth from grazing control in the epilimnion to control due to viral lysis in the hypolimnion. We estimated that in the epilimnion viral lysis accounted on average for 8.4 to 41.8% of the summed mortality (calculated by determining the sum of the mortalities due to lysis and grazing), compared to 51.3 to 91.0% of the summed mortality in the metalimninon and 88.5 to 94.2% of the summed mortality in the hypolimnion. Estimates of summed mortality values indicated that bacterial production was controlled completely or almost completely in the epilimnion (summed mortality, 66.6 to 128.5%) and the hypolimnion (summed mortality, 43.4 to 103.3%), whereas in the metalimnion viral lysis and HNF grazing were not sufficient to control bacterial production (summed mortality, 22.4 to 56.7%). The estimated contribution of organic matter released by viral lysis of cells into the pool of dissolved organic matter (DOM) was low; however, since cell lysis products are very likely labile compared to the bulk DOM, they might stimulate bacterial production. The high mortality of bacterioplankton due to viral lysis in anoxic water indicates that a significant portion of bacterial production in the metalimnion and hypolimnion is cycled in the bacterium-virus-DOM loop. This finding has major implications for the fate and cycling of organic nutrients in lakes.  相似文献

Reservoir 29 and Lake B are extremely acid lakes (epilimnion pHs of 2.7 and 3.2, respectively), because they receive acidic discharges from coal refuse piles. They differ in that the pH of profundal sediments in Reservoir 29 increased from 2.7 to 3.8 during the period of thermal stratification, whereas permanently anoxic sediments in Lake B had a pH of 6.2. The pH rise in Reservoir 29 sediments was correlated with a temporal increase in H2S concentration in the anaerobic hypolimnion from 0 to >1 mM. The chlorophyll a levels in the epilimnion of Reservoir 29 were low, and the rate of primary production was typical of an oligotrophic system. However, there was a dense 10-cm layer of algal biomass at the bottom of the metalimnion. Production by this layer was low owing to light limitation and possibly H2S toxicity. The specific photosynthetic rates of epilimnetic algae were low, which suggests that nutrient availability is more important than pH in limiting production. The highest photosynthetic rates were obtained in water samples incubated at pH 2.7 to 4. Heterotrophic bacterial activity (measured by [14C]glucose metabolism) was greatest at the sediment/water interface. Bacterial production (assayed by thymidine incorporation) was as high in Reservoir 29 as in a nonacid mesotrophic Indiana lake.  相似文献
