An unstable epoxide, leukotriene A4 (5(S)-trans-5,6-oxido-7,9-trans-11,14-cis-eicosatetraenoic acid), was earlier proposed to be an intermediate in the conversion of arachidonic acid into the slow reacting substance (SRS), leukotriene C4. In the present work synthetic leukotriene A4 was incubated with human leukocytes or murine mastocytoma cells. A lipoxygenase inhibitor, BW755C, was added in order to prevent leukotriene formation from endogenous substrate. Leukotriene C4 and 11-trans-leukotriene C4 were the main products with SRS activity. It was not established whether the 11-trans-compound was formed by isomerization at the leukotriene A4 or C4 stage.  相似文献

The crystal and molecular structure of prostaglandin A1 (PGA1) has been determined by X-ray diffraction. (Space group P212121, a = 18.10A?A, b = 21.09A?A, c = 5.42, Z = 4). Comparison of the structure of PGA1 with that of PGF (determined previously as the tribromobenzoate) indicates significant differences in the relative intramolecular side chain orientations. However, conformation and torsional angles within each side chain of PGA1 are retained with little or no change in comparison with PGF, indicating the presence of the C15-bromobenzoate groups in the latter have had little effect in altering internal side chain conformation in the solid state. The overall differences in relative side chain orientation in PGA1 are attributable to chemical and conformational changes in the substituted cyclopentane moiety (C8-C12).  相似文献

Three chlorophyll-protein complexes of a Chroomonas species (Cryptophyceae) have been separated by sodium dodecyl sulphate polyacrylamide gel electrophoresis. The two bands at 100 and 42 kDa are Complex I (CP I) and Complex IV (CP IV), the ubiquitous chlorophyll a-proteins associated with Photosystems I and II, respectively. The third 55 kDa band, which had two peptide subunits (24 and 20 kDa), contained both chlorophyll a and chlorophyll c2 in a molar ratio of 1.4 chlorophyll a to 1 chlorophyll c2 (chlorophyll achlorophyll c2 ratio in whole cells = 4). A chlorophyll ac2 fraction with similar spectral and electrophoretic properties was isolated by digitonin-sucrose density gradient centrifugation. This fraction had no photochemical activity and contained only a single carotenoid species with absorbance maxima in methanol at 424, 448 and 476 nm. Efficient energy transfer from chlorophyll c2 to chlorophyll a occurred in the complex.  相似文献

The effect on smooth muscle of the endoperoxides PGG2 and PGH2, which are intermediates in prostaglandin biosynthesis, was studied in different systems in vitro and in vivo. On gastrointestinal smooth muscle (gerbil colon, rat stomach) PGG2 and PGH2 produced contractions comparable to those of PGE2 and PGF2a whereas contractions elicited on vascular (rabbit aorta) and airway (guinea-pig trachea) smooth muscle were considerably greater than those of PGE2 and PGF2a respectively. On intravenous injection into guinea-pigs PGG2 and PGH2 caused a triphasic change in blood pressure and were 8–10 times more effective than PGF2a in producing an increase in tracheal insufflation pressure. When given as aerosols the unstable endoperoxides were less effective than PGF2a. It is concluded that the endoperoxides are potent smooth muscle stimulants and that they are more effective than their degradation products (PGD2, PGE2, PGF2a) in some systems.  相似文献

Aureobasidium pullulans, originally introduced as an inadvertent contaminant in solutions used for evaluating the stability of prostaglandins, proved to lead to the rapid disappearance of the cyclopentenone unit of PGA2 (as monitored by circular dichroic spectroscopy). The cyclopentenone unit is converted, in various metabolites, to a 9-keto, 9α or 9β-hydroxy group lacking the ring unsaturation. The major EtoAc-soluble 9-hydroxy metabolite (Compound-I) was shown to be 9α, 15α-dihydroxy-2,3,4,5-tetranor-13-trans-prostenoic acid. Similar tetranor 9-hydroxy metabolites with one additional degree of unsaturation, and with a 9β-hydroxy group, also occur but these have not been fully characterized. Only two of the wide range of 9-keto metabolites are fully characterized by mass spectral (MS) data: 9,15-oxo-2,3,4,5-tetranorprostanoic acid and 9,15-oxo-2,3,4,5-tetranor-13-trans-prostenoic acid. The water soluble metabolites have not been characterized further.The fully characterized metabolites together with MS data from mixtures of minor metabolites indicate that A. pullulans can perform the following transformations: β-oxidation, dehydrogenation at C-15, reduction of the enone carbon-carbon double bonds (both Δ10,11 and Δ13,14), reduction of the 9-ketone, and possibly migration of the cyclopentyl double bond (Δ10,11 → Δ11,12). A. pullulans metabolizes 15-epimeric PGA2 equally readily with the production of similar products. PGA1 affords less 9-keto metabolites with compound I constituting 33% of the product by HPLC analysis. A. pullulans displays some enantioselectivity, PGA2 and 15-epi-PGA2 are each metabolized more rapidly than their enantiomers. Other prostaglandins appear to be less readily metabolized.  相似文献

Immunoglobulins raised against 5,6-dihydro PGI2 crossreact with PGI2. When infused in vivo into the rat, these immunoglobulins are capable of I) neutralising the vasodepressor effects (bolus or continuous infusion) of exogenous PGI2, 2) blocking the catabolism of exogenous 3H-PGI2 and prolonging its life-time in the circulation (t12 approx 60 min) while that of 3H-PGE2 is unaffected, 3) trapping an endogenously produced substance which after extraction from blood and dissociation from the ligand-antibody complex, is immunoreactive with 6-keto PGF-specific antiserum. Yet the anti-5,6-dihydro PGI2 immunoglobulins have no effect on resting arterial blood pressure both in the normotensive and spontaneously hypertensive rat. These experiments indicate that endogenously produced PGI2 does not play a significant systemic role in blood pressure control although in combination with other vasodilators it could still participate in the regulation of vascular tone at a local level.  相似文献

NMR titration curves have been recorded for all the 13C resonances of cis and transN-acetyl-dl-proline in 2H2O. the measured pK2H values are 3.4 ± 0.8 and 4.13 ± 0.08 respectively; the free energy of ionization for the trans isomer being (3.8 kJ/mole) greater than for the cis. The ionization shifts of the two isomers differ significantly only at the acetyl carbonyl and Cγ positions. It is suggested that these are related to conformational changes which stabilize the trans form at low p2H.  相似文献

The technique of laser Doppler electrophoresis was applied for the study of the surface charge properties of (Na+,+)-ATPase containing microsomal vesicles derived from guinea-pig kidney. The influence of pH, the screening and binding of uni- and divalent cations and the binding of ATP show: (1) one net negative charge per protein unit with a pK = 3.9; (2) deviation from the Debye relation between surface potential and ionic strength for univalent cations, with no difference in the effect of Na+ and K+; (3) Mg2+ binds with an association constant of Ka = 1.1 · 102M?1 while ATP binds with an apparent Ka = 1.1 · 104M?2 for 1 mM Nacl, 0.2 mM KCI, 0.1 mM MgCl2, 0.1 mM Tris-HCI (pH 7.3). The binding is weaker at higher Mg2+ concentrations. There is no ATP binding in the absence of Mg2+. In addition, the average vesicle size derived from the linewidth of the quasi-elastic light scattering spectrum is 203.7 ± 15.2 nm. In the presence of ATP a reduction in size is observed.  相似文献

The effect of prostaglandin on the affinity of hemoglobin for oxygen was tested on human blood in vitro, using six different prostaglandins at several dosage levels in fresh heparinized blood from normal donors and in stored citrated blood, and using prostaglandin E2 on the blood from four seriously ill patients. No significant alterations in the affinity of hemoglobin for oxygen were dtected. A very small decrease in oxygen affinity in stored blood with high doses of prostaglandin was not statistically significant and would be of no physiologic significance even if real.We conclude that under the circumstances of this experiment prostaglandins do not alter the affinity of hemoglobin for oxygen in human whole blood in vitro.  相似文献

A new crystal form of a mitogenic lectin from pea seeds (Pisum sativum) has been obtained which is suitable for high resolution structural work. The crystals are orthorhombic, space group P212121, with unit cell dimensions: a = 64.2Å, b = 72. 7Å, c = 108. 3Å. The asymmetric unit contains one protein molecule.  相似文献

The present study examined the protein associations and energy transfer characteristics of chlorophyll c and fucoxanthin which are the major light-harvesting pigments in the brown and diatomaceous algae. It was demonstrated that sodium dodecyl sulfate (SDS)-solubilized photosynthetic membranes of these species when subjected to SDS polyacrylamide gel electrophoresis yielded three spectrally distinct pigment-protein complexes. The slowest migrating zone was identical to complex I, the SDS-altered form of the P-700 chlorophyll a-protein. The zone of intermediate mobility contained chlorophyll c and chlorophyll a in a molar ratio of 2 : 1, possessed no fucoxanthin, and showed efficient energy transfer from chlorophyll c to chlorophyll a. The fastest migrating pigment-protein zone contained fucoxanthin and chlorophyll a, possessed no chlorophyll c, and showed efficient energy transfer from fucoxanthin to chlorophyll a. It is demonstrated that the chlorophyll ac-protein and the chlorophyll afucoxanthin-protein complexes are common to the brown algae and diatoms examined, and likely share similar roles in the photosynthetic units of these species.  相似文献

Eight chlorophyll-protein complexes were isolated from thylakoid membranes of a Codium species, a marine green alga, by mild SDS-polyacrylamide gel electrophoresis. CP 1a1, CP 1a2, CP 1a3 and CP 1a4 were partially dissociated Photosystem (PS) I complexes, which in addition to the core reaction centre complex, CP 1, possessed PS I light-harvesting complexes containing chlorophyll (Chl) a, Chl b and siphonaxanthin. LHCP1 and LHCP3 are orange-brown green chlorophyll ab-proteins (Chl aChl b ratios of 0.66) that contain siphonaxanthin and its esterified form, siphonein. CP a and CP 1, the core reaction centre complexes of PS II and PS I, respectively, had similar spectral properties to those isolated from other algae or higher plants. These P-680- or P-700-Chl a-proteins are universally distributed among algae and terrestrial plants; they appear to be highly conserved and have undergone little evolutionary adaptation. Siphonaxanthin and siphonein which are present in the Codium light-harvesting complexes of PS II and PS I are responsible for enhanced absorption in the green region (518 and 538 nm). Efficient energy transfer from both xanthophylls and Chl b to only Chl a in Codium light-harvesting complexes, which have identical fluorescence emission spectra at 77 K to those of the lutein-Chl ab-proteins (Chl aChl b ratios of 1.2) of most green algae and all higher plants, proved that the molecular arrangement of these light-harvesting pigments was maintained in the isolated Codium complexes. The siphonaxanthin-Chl ab-proteins allow enhanced absorption of blue-green and green light, the predominant light available in deep ocean waters or shaded subtidal marine habitats. Since there is a variable distribution of lutein, siphonaxanthin and siphonein in marine green algae and siphonaxanthin is found in very ancient algae, these novel siphonein-siphonaxanthin-Chl ab-proteins may be ancient light-harvesting complexes which were evolved in deep water algae.  相似文献

Complexes of the formula cis-[Pt(HN+N)(L)Cl2], where (HN+N) are the protonated diamines including 3-aminoquinuclidine, N-aminopiperidine, piperazine, N-methylpiperazine, 1,1,4-trimethylpiperazine, and N-methyl-1,4-diazabicyclo [2,2,2] octane (N-methyl-dabco) and L = SCN?, NO2?, Br?, and F?, were synthesized from the protonated diamine complexes, [Pt(HN+N)Cl3]. The antitumor activities of the complexes were evaluated in vitro against L1210 murine leukemia cells, and ID50 values for the L-substituted complexes were compared to values of the parent complexes. In each case it was found that replacement of a chloride ion by SCN?, NO2?, Br?, or F?, either reduced or completely eliminated antitumor activity. This effect is explained in terms of the trans-directing ability of the ligand, L, compared to chloride. The NO2-substituted complex of 3- aminoquinuclidine was tested in vivo and found to exhibit little or no antitumor activity.  相似文献

Trans-3-dehydro-D, L-ornithine and trans-1, 4-diamino-2-butene have been synthesized and shown to be potent competitive inhibitors of ornithine decarboxylase. The KI′S for trans-3-dehydro-L-ornithine and trans-1, 4-diamino-2-butene are 2.2 and 2.0 μM respectively. Both analogs bind much more tightly to the enzyme than either ornithine or putrescine. Studies of chick embryo muscle cells in culture show results consistent with reversible inhibition of division and/or fusion by addition of trans-3-dehydro-D, L-ornithine to the culture medium.  相似文献

A-ring enollactones 1a, 1b or 9 derived from 4-cholesten-3-one, testosterone benzoate or 3-oxo-4-estren-17β-yl benzoate were condensed with [1,2-13C2]acetyl chloride to give intermediates 2a, 2b or 10. 2a and 2b were cyclized by acid or base to give 3,4-13C2-labeled 4-cholesten-3-one and testosterone, respectively. [3,4-13C2]4-Cholesten-3-one was converted via reduction of its trimethylsilyl enol ether to [3,4-13C2]cholesterol. Acetyl enollactone 10 was cyclized in acetic acid to [3,4-13C2]3-oxo-4-estren-17β-yl benzoate followed by aromatization and hydrolysis to produce [3,4-13C2]estradiol-17β. Alternatively, cyclization of 10 with base afforded [3,4-13C2]3-oxo-4-estren-17β-ol directly, which was then oxidized and aromatized to yield [3,4-13C2]estrone. Ozonolysis of progesterone, conversion to the diketal ester 16 and acylation followed by acid hydrolysis furnished [3,4-13C2]progesterone.  相似文献

The fluorescence decay characteristics of the isolated light-harvesting chlorophyll ab protein have been studied using low-intensity subnanosecond-resolution time-correlated single-photon counting. In the monomeric state in detergent micelles, the chlorophyll ab protein exhibits biexponential decay (τ1 = 1.2 ns, τ2 = 3.3 ns) with the two components having very similar weights. The decay parameters do not depend on emission wavelength. These results are discussed in relation to the Van Metter-Knox-Shepanski model (Van Metter, R.M. (1977) Biochim. Biophys. Acta 462, 642–657; Shepanski, J.S. and Knox, R.S. (1982) Isr. J. Chem., in the press) of the chlorophyll ab protein, and a kinetic analysis of the energy-transfer processes. The influence of detergent composition and concentration on the fluorescence decay of the chlorophyll protein is also described.  相似文献

A 22-residue peptide toxin from the venomous marine snail Conus geographus (L.) was found to have a most unusual amino acid composition: Lys4, Arg3, 12Cys6, Asx2, Glx2, Thr, Ala, plus three residues of trans-4-hydroxyproline. Absence of Gly and Pro indicates that the hydroxyproline must be in sequences different from those in which hydroxyproline occurs in collagen and other proteins.  相似文献

Prostaglandin congeners wherein the 15-hydroxy group is moved to the C16, C17, or C20 position or is replaced by a hydroxymethyl group were prepared via the 1,4-addition of a lithium trialkyl-trans-alkenyl alanate to an appropriate cyclopentenone. The functional role of a chlorophyll ab complex associated with Photosystem I (PS I) has been studied. The rate constant for P-700 photooxidation, KP-700, which under light-limiting conditions is directly proportional to the size of the functional light-harvesting antenna, has been measured in two PS I preparations, one of which contains the chlorophyll ab complex and the other lacking the complex. KP-700 for the former preparation is half of that of the preparation which has the chlorophyll ab complex present. This difference reflects a decrease in the functional light-harvesting antenna in the PS I complex devoid of the chlorophyll ab complex. Experiments involving reconstitution of the chlorophyll ab complex with the antenna-depleted PS I preparation indicate a substantial recovery of the KP-700 rate. These results demonstrate that the chlorophyll ab complex functions as a light-harvesting antenna in PS I.  相似文献
