对大杯香菇辐射选育新株系的各类氨基酸总量进行了遗传主成分分析。结果表明,变异系数最大的为硫氨基酸总量,达到13.39%;其次为儿童氨基酸总量,变异系数为7.00%;其它变异系数较小分别为甜味氨基酸总量、芳香族氨基酸总量、必需氨基酸总量、支链氨基酸总量和鲜味氨基酸总量为5.69%、5.64%、5.19%、5.24%和5.06%;在相关性上,鲜味氨基酸总量与甜味氨基酸总量、支链氨基酸总量、芳香族氨基酸总量、儿童氨基酸总量和必需氨基酸总量是呈极显著和显著的正相关,甜味氨基酸总量与支链氨基酸总量、儿童氨基酸总量和芳香族氨基酸总量呈极显著和显著的正相关,硫氨基酸总量与儿童氨基酸总量呈极显著负相关,与必需氨基酸总量是呈极显著正相关;支链氨基酸总量与芳香族氨基酸总量、必需氨基酸总量是呈极显著,与儿童氨基酸总量呈显著正相关;芳香族氨基酸总量与儿童氨基酸总量和必需氨基酸总量是呈极显著和显著的正相关;主成分分析结果表明,前4个特征根在7个特征根中累计贡献率达97.14%,也就是前4个主成分对变异的贡献率达96.07%。在各类氨基酸总量指标选择上,首先对变异大的各类氨基酸总量进行选择是及其重要的。在辐射选育新株系选择时,应注意选择大杯香菇中硫氨基酸总量高的新株系。  相似文献

用常规方法测定了大菱鲆鱼皮的基本营养成分,用氨基酸自动分析仪分析其氨基酸构成,并对大菱鲆鱼皮中的氨基酸组成进行了营养学评价。结果表明,大菱鲆鱼皮蛋白质含量为14.8%,脂肪含量为1.8%,氨基酸含量为27.24%,其中含量较高的是甘氨酸、丙氨酸、脯氨酸和精氨酸,占总氨基酸的59.73%;必需氨基酸、甜味氨基酸、苦味氨基酸、鲜味氨基酸和涩味氨基酸的含量分别占总氨基酸的20.81%、60.61%、30.07%、10.43%和0.01%。甜味氨基酸、苦味氨基酸、鲜味氨基酸是大菱鲆鱼皮氨基酸的主要部分,构成了大菱鲆鱼皮的主要味道,大菱鲆鱼皮是一种口感柔滑、味道鲜美的理想补强食品。  相似文献

采用AccQ .Tag法对氨基酸口服液中游离氨基酸和牛磺酸含量进行了分离测定。产品中氨基酸总量达 85mg/ml以上 ,共 1 3种氨基酸。必需氨基酸与非必需氨基酸比例为 2 .2 0~ 2 .50∶1 ,支 /芳比为 2 .30~ 2 .55∶1 .配方接近于FAO氨基酸比例模式 ,以FAO氨基酸模式及化学评分评价了该制剂的营养价值 ,基本上不存在限制氨基酸 ,化学评分均在 90分以上。  相似文献

Minerals more readily adsorb amino acids with charged R groups than uncharged R groups, so that the incorporation of amino acids with charged R groups into peptides would be more frequent than for amino acids with uncharged R groups. However, 74% of the amino acids in the proteins of modern organisms contain uncharged R groups. Thus, what could have been the mechanisms that produced peptides/proteins with more amino acids with uncharged R groups than precursors with charged R groups? Should we expect the composition of amino acids adsorbed on minerals to be similar to those of present proteins? Was the adsorption of amino acids on minerals important for the origin of life? The lipid world offers an alternative view of origin of life. Liposomes contributed to elongation of peptides as well as select hydrophobic amino acids and peptides. These experiments could be showing the mechanism, which hydrophobic amino acids have been selected. However, liposomes have no influence on the stereoselectivity in the oligomerization of amino acids. In the present paper, several other mechanisms are also discussed that could produce peptides with a greater proportion of amino acids with uncharged R groups.  相似文献

为科学评价食用菌与果蔬两类食材的营养特征与口感风味,本研究选取4种常见食用菌(金针菇、斑玉蕈、香菇、双孢蘑菇)和4种常见果蔬(苹果、香蕉、胡萝卜、番茄),采用氨基酸自动分析仪测定其游离氨基酸,系统比较了两类食材的游离氨基酸组分比例及呈味特征。主成分和聚类分析表明 4种食用菌和4种果蔬显著分为两类,食用菌类游离氨基酸总量平均是果蔬类的3倍以上,4种食用菌中各游离氨基酸的比例均衡。4种食用菌的必需氨基酸/(必需氨基酸+非必需氨基酸)在40%左右,必需氨基酸/非必需氨基酸接近或大于60%,均接近理想值。4种食用菌所含的呈味氨基酸(鲜味、甜味、芳香族、苦味)均是4种果蔬的2倍以上,TAV值最显著的是:谷氨酸、苯丙氨酸、甘氨酸、苏氨酸、组氨酸、赖氨酸、缬氨酸、精氨酸和丙氨酸。这些氨基酸分属在五大氨基酸家族,在食用菌与果蔬中代谢途径相同;只有赖氨酸在二者中的合成途径不同。结果表明供试的食用菌比果蔬有优质的氨基酸比例和丰富的呈味组分,此差异体现在众多氨基酸的代谢层面,而非个别氨基酸导致。  相似文献

Nitrogen-starved yeast derepress a general amino acid permease which transports basic and hydrophobic amino acids. Although both groups of amino acids are metabolized, the derivatives of the basic amino acids are retained by the cells, whereas those of the hydrophobic amino acids are released as acidic and neutral deaminated derivatives. The release of the deaminated derivatives of the hydrophobic amino acids only occurs in the presence of glucose, which presumably produces amino acceptors. The accumulation of intracellular amino acids results in trans-inhibition of the uptake of exogenous amino acids whether the intracellular amino acid is a basic amino acid or the product of intracellular transamination from a hydrophobic amino acid. Variation of permease and transaminase activity was measured during growth under repressed (ammonia-grown) and derepressed (proline-grown) conditions. Maximum levels for both activities occurs at the mid-exponential phase.  相似文献

The efficiency of utilization of milk amino acids for body gain in suckling mink kits from small (n = 3), medium (n = 6) and large litters (n = 9) was investigated by using 36 mink dams and their litters for measurements during lactation weeks 1 through 4. Measurements on each dam and litter were performed once, hence three dams per litter size each week (n = 9). Individual milk intake of kits was determined, milk samples were collected and kits were killed for determination of amino acid composition. The most abundant amino acids in milk were glutamate, leucine and aspartate making up about 40% of total amino acids. Branched chained amino acids made up slightly more than 20% and sulphur containing amino acids less than 5% of total milk amino acids. In kit bodies the sum of glutamate, aspartate and leucine made up about 32% of amino acids, branched chain amino acids about 16% and sulphur containing amino acids about 4%. The amino acid composition of both milk and bodies changed as lactation progressed with decreasing proportions of essential amino acids. The ratio between body and milk amino acids was constantly over 1 only for lysine, suggesting that it was the most limiting amino acid in mink milk. Milk amino acids were efficiently utilized during week 1, ranging from 74.7% (lysine) to 42.1% (leucine), with an average for essential amino acids of 58.4%. Tendencies for improved utilization of lysine (74.7-78.2%), phenylalanine (61.0-70.0%), histidine (62.4-68.8%), arginine (61.3-70.4%) and all essential amino acids (58.4-60.2%) from week 1 to week 2 were recorded. During weeks 3 and 4, the efficiency declined, and for all essential amino acids the average utilization was 38.1% during week 4.  相似文献

Recent advances in determination of the high-resolution structure of membrane proteins now enable analysis of the main features of amino acids in transmembrane (TM) segments in comparison with amino acids in water-soluble helices. In this work, we conducted a large-scale analysis of the prevalent locations of amino acids by using a data set of 170 structures of integral membrane proteins obtained from the MPtopo database and 930 structures of water-soluble helical proteins obtained from the protein data bank. Large hydrophobic amino acids (Leu, Val, Ile, and Phe) plus Gly were clearly prevalent in TM helices whereas polar amino acids (Glu, Lys, Asp, Arg, and Gln) were less frequent in this type of helix. The distribution of amino acids along TM helices was also examined. As expected, hydrophobic and slightly polar amino acids are commonly found in the hydrophobic core of the membrane whereas aromatic (Trp and Tyr), Pro, and the hydrophilic amino acids (Asn, His, and Gln) occur more frequently in the interface regions. Charged amino acids are also statistically prevalent outside the hydrophobic core of the membrane, and whereas acidic amino acids are frequently found at both cytoplasmic and extra-cytoplasmic interfaces, basic amino acids cluster at the cytoplasmic interface. These results strongly support the experimentally demonstrated biased distribution of positively charged amino acids (that is, the so-called the positive-inside rule) with structural data.  相似文献

Uptake of 16 amino acids by the filamentous, heterocyst-forming cyanobacterium Anabaena sp. strain PCC 7120 was characterized with regard to kinetic parameters of transport, intracellular accumulation of the transported amino acids, and sensitivity of the transport process to energy metabolism inhibitors. Mutants resistant to certain toxic analogs of some amino acids were isolated that were impaired in amino acid transport. Results obtained in this study, together with those reported previously (A. Herrero and E. Flores, J. Biol. Chem. 265:3931-3935, 1990), suggest that there are at least five amino acid transport systems in strain PCC 7120: one high-affinity, active system for basic amino acids; one low-affinity, passive system for basic amino acids; two high-affinity, active systems with overlapping, but not identical, specificities for neutral amino acids; and one putative system for acidic amino acids. Some of the amino acid transport mutants were impaired in diazotrophic growth. These mutants were unable to develop a normal percentage of heterocysts and normal nitrogenase activity in response to nitrogen stepdown. Putative roles for the amino acid transport systems in uptake of extracellular amino acids, recapture of amino acids that have leaked from the cells, and intercellular transfer of amino acids in the filaments of Anabaena sp. strain PCC 7120 are discussed.  相似文献

Abstract— The brain levels of each of the aromatic and branched-chain amino acids change 2 h after fasting rats begin to consume either a carbohydrate-fat diet or a similar diet containing 18% or 40% protein. Carbohydrate-fat ingestion elevates the concentrations of each of the aromatic amino acids in brain, while substantially depressing those of the branched-chain amino acids. The inclusion of protein in this diet suppresses the increases in brain aromatic amino acids and attenuates the decreases in the branched-chain amino acids. The changes in the brain level of each neutral amino acid following the ingestion of any of these diets correlate extremely well with the effects of the diet on the serum neutral amino acid pattern, specifically on the serum concentration ratio of each neutral amino acid to the sum of the other neutral amino acids. The diet-induced changes in the brain level of each of the amino acids also correlate surprisingly well with the calculated rate of brain influx for each amino acid.  相似文献

Changes of casein and lysozyme during roasting at various times and temperatures 100 ~ 300°C), especially those in amino acid composition and formation of some nonvolatile degradation products, were investigated.

Tryptophan, methionine, basic amino acids and β-hydroxy amino acids were easily decomposed as compared with acidic amino acids and other neutral amino acids in casein and Iysozyme. In hot water extract of roasted casein were detected some free amino acids, peptides, organic acids such as α-ketoglutaric, tartaric and malic acids, and indole. It is considered that free amino acids are produced mainly through ionic cleavage of peptide bond with the water bound within casein.  相似文献

Summary The metabolic fate of amino acids introduced into the locust egg by the addition of amino acids after oviposition is compared to that of amino acids derived from the maternal haemocoel. It was found that amino acids from the two sources did not always follow the same pattern of utilization. Topical application resulted in some of the applied amino acids being converted into other amino acids and ammonia. When the labelled amino acid was administered to the egg via the maternal haemocoel, radioactivity was restricted to the amino acid orginally provided.  相似文献

对珠子参茎、叶、花的氨基酸组成与含量进行测定和营养评价分析.结果表明,珠子参茎、叶、花中均含有17种常见氨基酸,氨基酸总量分别为3.73%,14.80%和8.72%;珠子参的茎中必需氨基酸与总氨基酸的比值(E/T)为0.39,必需氨基酸与非必需氨基酸的比值(E/N)为0.64,叶中E/T为0.41,E/N为o.71,花中E/T为0.39,E/N为0.63,氨基酸配比较为合理;珠子参茎、叶、花中蛋氨酸+胱氨酸均为第一限制氨基酸;珠子参茎、叶、花中均含有γ-氨基丁酸,含量分别为0.12%,0.26%和0.16%,叶片中γ-氨基丁酸的含量最高.  相似文献

实验对饲养中国大鲵氨基酸组成成分进行了比较全面的测定与分析,结果表明:饲养中国大鲵肌肉的蛋白质质量分数为164.3 g.kg-1,水解氨基酸总量为160.6 g.kg-1,各种氨基酸组成相当全面,必需氨基酸(EAA)为75.2 g.kg-1,占氨基酸总量的46.82%,符合人体需要量模式的程度相当高,为优质食物蛋白,可与做为参考蛋白的鸡蛋蛋白相媲美。研究同时发现呈味氨基酸总量70.5 g.kg-1,占氨基酸总量的43.90%,为研发味道鲜美的大鲵食品提供了有力的数据。  相似文献

Although ion exchange chromatography has been used in separating amino acids from mineral salts, quantitative recovery has not been possible for the basic amino acids or for subnanomole concentrations of amino acids.As an analytical tool for amino acid analysis, ion-exchange chromatography has made it possible to resolve a relatively complex mixture of amino acids in less than an hour with detection limits of less than 10–12 moles of amino acids. Reasonable specificity for amino acids is achieved by multiple wavelength detection of the reaction product found with ninhydrin. Unequivocal specificity must be obtained in conjunction with other methods such as mass spectrometry.In the analysis of subnanomole levels of amino acids, it is necessary to carry both reagent blanks and low-level amino acid standards through the entire sample preparation step since both contamination and selective losses occur and must be monitored.  相似文献

Sarracenia purpurea L. (northern pitcher plant) is an insectivorous plant with extrafloral nectar that attracts insects to a water-filled pitfall trap. We identified and quantified the amino acids in extrafloral nectar produced by pitchers of S. purpurea. Nectar samples were collected from 32 pitchers using a wick-sampling technique. Samples were analyzed for amino acids with reverse-phase high-performance liquid chromatography with phenylisothiocyanate derivatization. Detectable amounts of amino acids were found in each of the 32 nectar samples tested. Mean number of amino acids in a nectar sample was 9 (SD = 2.2). No amino acid was detected in all 32 samples. Mean amount of amino acids in a nectar sample (i.e., amount per wick) was 351.4 ng (SD = 113.2). Nine amino acids occurred in 20 of the 32 samples (aspartic acid, cysteine, glutamic acid, glycine, histidine, hydroxyproline, methionine, serine, valine) averaging 263.4 ng (SD = 94.9), and accounting for ~75% of the total amino acid content. Nectar production may constitute a significant cost of carnivory since the nectar contains amino acids. However, some insects prefer nectar with amino acids and presence of amino acids may increase visitation and capture of insect prey.  相似文献

The effects on blood glucose concentration of 10% glucose with amino acid solution at low (18%) and high (25%) concentration of branched chain amino acids were studied, two days after laparotomy in fasted rabbits. During the whole study period, saline infusion was associated with normoglycaemia. Among the other infusion combinations peroperative infusion of amino acid solution high in branched chain amino acids with 10% glucose was the least hyperglycaemic. An amino acid solution low in branched chain amino acids with 10% glucose had the lowest hyperglycaemic effect 24 hours postoperatively. After 48 hours an infusion of 10% glucose produced the weakest hyperglycaemia. In preoperatively and 24 hours postoperatively 10% glucose was more hyperglycaemic than 10% glucose with the two amino acid solutions. After 24 and 48 hours, infusion of 10% glucose with amino acids high in branched chain amino acids was more hyperglycaemic than 10% glucose with amino acids low in branched chain amino acids.  相似文献

Amides and acidic amino acids represent the major long distance transport forms of organic nitrogen. Six amino acid permeases (AAPs) from Arabidopsis mediating transport of a wide spectrum of amino acids were isolated. AAPs are distantly related to plasma membrane amino acid transport systems N and A and to vesicular transporters such as VGAT from mammals. A detailed comparison of the properties by electrophysiology after heterologous expression in Xenopus oocytes shows that, although capable of recognizing and transporting a wide spectrum of amino acids, individual AAPs differ with respect to specificity. Apparent substrate affinities are influenced by structure and net charge and vary by three orders of magnitude. AAPs mediate cotransport of neutral amino acids with one proton. Uncharged forms of acidic and basic amino acids are cotransported with one proton. Since all AAPs are differentially expressed, different tissues may be supplied with a different spectrum of amino acids. AAP3 and AAP5 are the only transporters mediating efficient transport of the basic amino acids. In vivo competition shows that the capability to transport basic amino acids in planta might be overruled by excess amides and acidic amino acids in the apoplasm. With the exception of AAP6, AAPs do not recognize aspartate; only AAP6 has an affinity for aspartate in the physiologically relevant range. This property is due to an overall higher affinity of AAP6 for neutral and acidic amino acids. Thus AAP6 may serve a different role either in cooperating with the lower affinity systems to acquire amino acids in the low concentration range, as a system responsible for aspartate transport or as an uptake system from the xylem. In agreement, a yeast mutant deficient in acidic amino acid uptake at low aspartate concentrations was complemented only by AAP6. Taken together, the AAPs transport neutral, acidic and cationic amino acids, including the major transport forms, i.e. glutamine, asparagine and glutamate. Increasing proton concentrations strongly activate transport of amino acids. Thus the actual apoplasmic concentration of amino acids and the pH will determine what is transported in vivo, i.e. major amino acids such as glutamine, asparagine, and glutamate will be mobilized preferentially.  相似文献
