In this paper, we report a chromosomal study of three Brazilian species of Colostethus, C. brunneus from the type locality, Colostethus sp. (aff. trilineatus), and Colostethus sp., which is morphologically similar to C. brunneus. The diploid number for C. brunneus was 2n = 24 chromosomes, in agreement with that previously described for specimens from Peru. Colostethus sp. (aff. trilineatus) and Colostethus sp. showed a very similar karyotype with 22 chromosomes. The NOR was located on pair 3 in C. brunneus, on pair 4 in Colostethus sp. (aff. trilineatus), and on pair 2 in Colostethus sp. In one specimen of Colostethus sp., an additional NOR site was located on pair 7 in only one of the homologs. This extra Ag-NOR site was confirmed by FISH using an rDNA probe. In addition to the NOR location, the C-banding pattern was also species-specific, despite the similar chromosomal morphology of the species. These results indicate that although these species may be closely related, there is a clear dichotomy in their chromosome number.  相似文献

Scanning and transmission electron microscopy and fluorochrome staining with DAPI were used to study the spermatozoa of Colostethus marchesianus from the type locality and Colostethus sp. (aff. marchesianus) from a different locality. The sperm cell of these species resembles those of most neobatrachian species, except for the presence of two complete flagella, a character previously described only for four unrelated anuran species belonging to the families Leptodactylidae, Rhacophoridae and Dendrobatidae. Although very similar, the spermatozoa described here showed slight differences in the length of their nucleus, in the extent of the subacrosomal cone over the nucleus, and in the curvature of the axial fiber seen in cross section. The absence of a juxtaxonemal fiber and the presence of a comma shaped axial fiber seem to be common characteristics of dendrobatid spermatozoa. The minor differences found in our study may indicate that the taxa studied here are distinct species. The evolutionary significance of spermatozoa biflagellarity is still unclear, although the presence of this trait in unrelated groups suggests independent origins.  相似文献

This study describes the spermatozoa of the dendrobatids Epipedobates femoralis and Colostethus sp. using light and transmission electron microscopy. Both species possess a biflagellate spermatozoon, an unusual characteristic only previously reported in two anuran species belonging to the families Leptodactylidae and Racophoridae. The acrosomal complex of both species consists of a conical acrosomal vesicle and a subacrosomal cone, both of which cover the anterior portion of the nucleus, but to differing extents. In the midpiece, the centrioles are disposed parallel to each other and to the cell axis and give rise to two axonemes. Two paraxonemal rods were also seen entering the nuclear fossa. Both flagella are surrounded by a single mitochondrial collar. Each flagellum is formed by an axial fiber connected to the axoneme by an axial sheath; juxta-axonemal fibers are absent. Our data seem to support that Epipedobates femoralis should be placed in a separate clade possibly related to Colosthetus and that these two genera may not be monophyletic.  相似文献

Abstract Spermiogenesis was studied by transmission electron microscopy in the acanthocotylid monogeneans Myxinidocotyle californica (from Eptatretus stoutii) and Acanthocotyle lobianchi (from Raja clavata). In Myxinidocotyle and Acanthocotyle, the zone of differentiation shows two 9+‘1’ axonemes, the elongating nucleus and mitochondrion, and a single cortical cytoplasmic microtubule. This single microtubule is found in the mature spermatozoon of both species and was also noted in capsalids. This requires a modified definition of ‘pattern 2’ of spermatozoa which becomes: ‘spermatozoa with two axonemes and no cortical microtubules, except one single element much shorter than the spermatozoon’. A very unusual structure was found in Myxinidocotyle, but not in Acanthocotyle: the centriolar derivative of one of the 9+‘1’ axonemes is made up of 18 diverging singlets of unequal length associated with electron-dense cytoplasm. This seems to be the first case of a centriolar derivative without nine-fold symmetry associated with an axoneme with nine-fold symmetry.  相似文献

Summary

Among the Arthropoda the Xiphosura are the only group whose spermatozoa resemble the so-called ‘primitive type’, although even here there are some differences. The spermatozoa of Tachypleus consist of a sperm head, a middle piece, and a long flagellum. Though principally quite similar to the spermatozoa of Limulus polyphemus some characteristics are noticeably different: the axonemal pattern (9 × 2 + 0), the shape of the acrosomal vacuole, the different position of the acrosomal filament, and the distribution of mitochondria throughout the cytoplasm. In contrast to what is known from Limulus the nuclear envelope is apparently dissolved over wide areas. Consequences for the interpretation of the acrosomal reaction are discussed. Structural conditions are compared with those in other Chelicerata.  相似文献

Despite the recent advancements in recognizing diversity in lichen‐forming fungi, assessing the timing of diversification remains largely unexplored in these important fungal symbionts. To better understand the evolutionary processes driving diversification in common lichen‐forming fungi, we investigated the phylogeny and biogeography of the broadly distributed Melanelixia fuliginosa/M. glabratula group, using molecular data from six nuclear markers. Phylogenetic analyses of individual gene alignments and combined data provide strong evidence for five species‐level lineages within this species complex. Three of these lineages correspond to the previously described species M. fuliginosa, M. glabratula, and M. subaurifera. The remaining two lineages, ‘M. sp. 1’ and ‘M. sp. 2’, merit species recognition based on genealogical concordance. Both M. glabratula and M. subaurifera had broad intercontinental distributions, sharing identical haplotypes among intercontinental populations. Based on the current sampling, M. fuliginosa s.s. was represented exclusively by European material and was not collected in North America. ‘M. sp. 1’ was represented by collections from Scotland and Spain; and ‘M. sp. 2’ was represented by collections in California, USA. Environmental factors driving the contrasting distribution patterns in this group remain unknown. Divergence times estimated using a coalescence‐based multilocus species‐tree approach suggest that diversification within the M. fuliginosa/M. glabratula group occurred exclusively during the Miocene. The results of the present study indicate that phenotypically cryptic lichen‐forming fungal species‐level lineages may be relatively ancient and do not necessarily reflect recent divergence events. Many freshwater protists harbor unicellular green algae within their cells and these host‐symbiont relationships slowly are becoming better understood. Recently, we reported that several ciliate species shared a single species of symbiotic algae. Nonetheless, the algae from different host ciliates were each distinguishable by their different genotypes, and these host‐algal genotype combinations remained unchanged throughout a 15‐month period of sampling from natural populations. The same algal species had been reported as the shared symbiont of several ciliates from a remote lake. Consequently, this alga appears to play a key role in ciliate‐algae symbioses. In the present study, we successfully isolated the algae from ciliate cells and established unialgal cultures. This species is herein named Brandtia ciliaticola gen. et sp. nov. and has typical ‘Chlorella‐like’ morphology, being a spherical autosporic coccoid with a single chloroplast containing a pyrenoid. The alga belongs to the Chlorella‐clade in Chlorellaceae (Trebouxiophyceae), but it is not strongly connected to any of the other genera in this group. In addition to this phylogenetic distinctiveness, a unique compensatory base change in the SSU rRNA gene is decisive in distinguishing this genus. Sequences of SSU‐ITS (internal transcribed spacer) rDNA for each isolate were compared to those obtained previously from the same host ciliate. Consistent algal genotypes were recovered from each host, which strongly suggests that B. ciliaticola has established a persistent symbiosis in each ciliate species.  相似文献

Arion transsylvanus (Mollusca, Pulmonata, Arionidae): rediscovery of a cryptic species. —Zoologica Scripta, 39, 343–362. Cryptic species are abundant among invertebrates and are often hard to recognise. Molecular markers are an extremely useful tool to delineate cryptic taxa, although they should be applied with caution because different genes and techniques may yield different outcomes. We illustrate how cross‐validation by molecular and morphological data can be applied to optimise taxonomic interpretations when cryptic species are involved. This is performed for the terrestrial slug Arion subfuscus species complex which represents a historical ‘taxonomic garbage can’. Gonad morphology, allozymes and mtDNA data consistently showed that slugs from Romania and a location in E Poland represent a strongly differentiated taxon within this complex. These slugs are therefore formally redescribed and assigned to A. transsylvanus Simroth 1885 ; a forgotten nominal taxon from Transylvania. Diagnostic characters, including DNA sequences for the mitochondrial 16S rDNA are presented. Animals with the morphology of A. brunneus Lehmann 1862 , a nominal taxon which has also been reported from Romania, have the gonad type, allozyme alleles and 16S rDNA haplotypes of either A. fuscus, A. subfuscus or A. transsylvanus. Therefore, A. brunneus is regarded as a colour morph shared by several species and hence, A. transsylvanus is probably the only A. subfuscus‐like species in the Romanian Carpathians.  相似文献

A fine structure study of spermatids and spermatozoa of the spider, Pisaurina sp. demonstrates that early spermiogenesis is similar to other flagellate spermatozoa. An acrosome forms from a Golgi-derived, acrosomal vesicle, a perforatorium indents acromosome and nucleus, a flagellum with a three-plus-nine tubule substructure is formed and nuclear chromatin condenses during spermiogenesis. Divergence from typical spermatozoa includes the presence of a three-tubule substructure of the central flagellar shaft, progressive rounding-up of late spermatids with concomitant incorporation of previously formed flagellum. This evidence is presented in terms of its possible functional significance in fertilization and gamete fusion in spiders.  相似文献

The male reproductive tract of Leucoptera coffeella was processed for light and transmission electron microscopy. In the testis, the eupyrene cells are arranged in individual cysts, while the apyrene cysts form aggregates, never observed in other Lepidoptera. Both cysts contain 128 spermatozoa, which differ from the typical pattern. In the seminal vesicle, both types of spermatozoa are dispersed in the lumen, also different from other Lepidoptera. The apyrene spermatozoa are similar to those observed for other Lepidoptera. They present an anterior region covered by a dense cap and the flagellum is composed of a 9 + 9 + 2 axoneme and two mitochondrial derivatives. The eupyrene spermatozoa, however, differ from the typical pattern for Lepidoptera. Their anterior region contains a nucleus, an acrosome and a peculiar arc of eight accessory microtubules connected to the plasma membrane by dense bridges. In the nucleus–flagellum region, the ninth accessory microtubule is assembled between both mitochondrial derivatives, to participate in the axoneme. The flagellum comprises a 9 + 9 + 2 axoneme and two mitochondrial derivatives with paracrystalline cores. External to the plasma membrane and close to the accessory microtubules, there are tufts of an amorphous material, suggesting reduced lacinate appendages, while the reticular ones are absent. The reduction of lacinate appendages and the absence of sperm bundles in the seminal vesicle support the concept that the appendages of other Lepidoptera could be associated with the eupyrene aggregations. The characters ‘number of spermatozoa per cyst’ and ‘absence of bundles’ should be considered plesiomorphic, supporting the position of this taxon in the base of the Ditrysia.  相似文献

A new species, Marsupiomonas pelliculata gen. et sp. nov. (Pedinophyceae), is described. A single flagellum emerges from a deep pit with a distinctive thickened margin. The flagellum has rigid fibrillar hairs which are probably formed in the perinuclear space. A short second flagellar basal body lies within the cell close to the basal body of the emergent flagellum and the flagellar root system consists of striated and microtubular roots. There is a distinctive theca covering all but the anterior end of the cell and also a single large bright green chloroplast with an immersed pyrenoid surrounded by a starch shell. The wide salinity tolerance of the species is discussed in relation to its distribution in estuarine and salt marsh habitats. The salient features of the new species—the insertion of the emergent flagellum into a deep pit and the possession of a theca—are also seen in Pedinomonas tenuis, and it is suggested that P. tenuis could be transferred to the new genus Marsupiomonas. The class Pedinophyceae now includes three genera (Pedinomonas, Resultor and Marsupiomonas) and the distinguishing features are discussed.  相似文献

Eight Cylindropuntia species have naturalised in Australia and pose serious economic, environmental and social impacts. The host range of four additional biotypes of D. tomentosus from southern USA was investigated. Feeding and development were restricted to the genus Cylindropuntia. However, they showed differences in specificity within this genus and some biotypes discriminated between the provenances of Cylindropuntia rosea and Cylindropuntia tunicata. Efficacy trials were conducted to determine whether populations of each biotype could be sustained on the naturalised Cylindropuntia species and if these populations could retard the growth or kill these plants. The ‘acanthocarpa’ biotype offers potential control of C. rosea (Lorne Station), while the ‘cylindropuntia sp.’ biotype shows great potential to control C. rosea (Grawin). The ‘cylindropuntia sp.’ biotype also had a high impact on Cylindropuntia kleiniae and Cylindropuntia imbricata, and a moderate impact on Cylindropuntia leptocaulis and Cylindropuntia prolifera. The ‘acanthocarpa?×?echinocarpa’ biotype had its greatest impact on C. tunicata (Grawin), killing this plant in 18 weeks. A fourth biotype, ‘leptocaulis’, was damaging to some species, but was less effective than the other biotypes. Cylindropuntia spinosior is the only naturalised species in Australia where no effective biocontrol agent has been found.  相似文献

Developmental plasticity is often correlated with diversity and has been proposed as a facilitator of phenotypic novelty. Yet how a dimorphism arises or how additional morphs are added is not understood, and few systems provide experimental insight into the evolution of polyphenisms. Because plasticity correlates with structural diversity in Pristionchus nematodes, studies in this group can test the role of plasticity in facilitating novelty. Here, we describe three new species, Pristionchus fukushimae sp. nov. , Pristionchus hoplostomus sp. nov. , and the hermaphroditic Pristionchus triformis sp. nov. , which are characterized by a novel polymorphism in their mouthparts. In addition to showing the canonical mouth dimorphism of diplogastrid nematodes, comprising a stenostomatous (‘narrow‐mouthed’) and a eurystomatous (‘wide‐mouthed’) form, the new species exhibit forms with six, 12, or intermediate numbers of cheilostomatal plates. Correlated with this polymorphism is another trait that varies among species: whereas divisions between plates are complete in P. triformis sp. nov. , which is biased towards a novel ‘megastomatous’ form comprising 12 complete plates, the homologous divisions in the other new species are partial and of variable length. In a reconstruction of character evolution, a phylogeny inferred from 26 ribosomal protein genes and a partial small subunit rRNA gene supported the megastomatous form of P. triformis sp. nov. as the derived end of a series of split‐plate forms. Although split‐plate forms were normally only observed in eurystomatous nematodes, a single 12‐plated stenostomatous individual of P. hoplostomus sp. nov. was also observed, suggesting independence of the two types of mouth plasticity. By introducing these new species to the Pristionchus model system, this study provides further insight into the evolution of polymorphisms and their evolutionary intermediates. © 2013 The Linnean Society of London  相似文献

Vasseuromys is a species‐rich genus of small‐ to medium‐sized glirids spanning the latest Oligocene to late Miocene of Europe and western Asia. Despite extensive discoveries over the past 50 years, little phylogenetic work has been done on Vasseuromys. This study presents the first phylogenetic analysis of the genus that includes all the described species and a new taxon Vasseuromys tectus sp. nov. from the late Miocene of eastern Europe, providing the first insights into the evolutionary relationships within the clade. Results suggest that the genus is clearly paraphyletic. Two strongly supported genus‐level clades are recognized within ‘Vasseuromys’: a restricted Vasseuromys clade (containing the three species, V. pannonicus, V. rugosus and V. tectus) and the Glirulus clade that includes ‘Vasseuromysduplex. The remaining ‘Vasseuromys’ species are found to constitute a set of paraphyletic taxa, with the polyphyletic ‘Ramys’ nested within it. The genus Gliruloides is synonymized with Glirulus. Vasseuromys tectus sp. nov. is the most derived member of the genus in having a greater number of cheek teeth ridges including constantly present anterotrope, centrotrope, second prototrope on M1–2, third metatrope on M2, two to three posterotropids on p4 and strong ectolophids on lower molars. The results of the study confirm a European origin for Vasseuromys while suggesting that the late Miocene species of the genus dispersed from the east in the early Turolian.  相似文献

Apristurus brunneus, A. kampae, and Parmaturus xaniurus were caught by trawl and longline between Washington and southern California (48°N to 32°N latitude) from June 2001 through October 2004. In females, oviducal gland width increased with TL in all three species but remained within 4 to 6% of TL. Oocyte diameter increased with TL during reproductive development in all species, and continued to increase with size after reproductive maturity was reached. The weight-length relationships differed significantly among species but not between sexes of any species from the eastern North Pacific combined. Using logistic regression, it was determined that total length at 50% maturity (TL50) of A. brunneus males was 514 mm TL and all A. brunneus males were mature at 547 mm TL. Female A. brunneus reached TL50 at 501 mm TL and all female A. brunneus were mature at 581 mm TL. Apristurus kampae males and females reached TL50 at approximately 485 mm TL and 490 mm TL, respectively. Parmaturus xaniurus males reached TL50 at 444 mm TL and females at 501 mm TL. Gravid female A. brunneus were found in all months of the year, but gravid P. xaniurus were found primarily June through September. Adult A. kampae females were found only in July through November and there were gravid females in all these months. Based upon gonadosomatic indices, I G, these species may mate within a defined period of time (April through June); however, A. brunneus appear to reproduce throughout the year and P. xaniurus reproduce within a defined season (approximately July through September).  相似文献

Scinax fuscomarginatus, Scinax parkeri, Scinax trilineatus, Scinax lutzorum, and Scinax pusillus are morphologically similar species with controversial taxonomy associated with open formations of South America east of the Andes. We used external morphology, advertisement call, and molecular sequences to assess the taxonomy of these species. Phylogenetic analysis of molecular data showed a well‐supported monophyletic group divided into two main clades and several subclades. Specimens of Hyla madeirae (synonym of S. fuscomarginatus) and those from Serra do Cachimbo correspond to distinct subclades, whereas the currently valid species were grouped into a single clade. This clade showed a marked substructure, but concordance of the recognized species to subclades was limited, showing S. fuscomarginatus to be paraphyletic with respect to S. parkeri, S. trilineatus, S. lutzorum, and S. pusillus. The high intrapopulation and intraspecific variation found in morphological characters did not allow us to differentiate amongst the currently recognized species. Similarly, none of them was distinguishable by morphometric analysis or advertisement call. Based on multiple evidence, we propose the synonymy of S. parkeri, S. trilineatus, S. lutzorum, and S. pusillus with S. fuscomarginatus, the revalidation of H. madeirae, and describe the specimens from Serra do Cachimbo as a new species. © 2014 The Linnean Society of London  相似文献

The ultrastructure of the spermatozoon of Geogarypus nigrimanus (Arachnida, Pseudoscorpionida) is described. The spermatozoon is composed of a small elliptic nucleus, a short flagellum and a very long and complex acrosome. In the male genital ducts, as in other studied species of pseudoscorpions, the sperm components are rolled up to form a globular structure enclosed in a cyst wall. The Geogarypus spermatozoon with a reduced flagellum and a giant acrosome seems to be evolutionary more advanced than spermatozoa from other pseudoscorpions.  相似文献
