Prolonged growth of cell cultures of sycamore (Acer pseudoplatanus L.) on agar medium containing myo-inositol-2-3H resulted in incorporation of label predominately into uronosyl and pentosyl units of cell wall polysaccharides. Procedures normally used to distinguish between pectic substance and hemicellulose yielded carbohydrate-rich fractions with solubility characteristics ranging from pectic substance to hemicellulose yet the uronic acid and pentose composition of these fractions was decidedly pectic. Galacturonic acid was the only uronic acid present in each fraction. Subfractionation of alkali-soluble (hemicellulosic) polysaccharide by neutralization followed by ethanol precipitation gave 3 fractions, a water-insoluble, an ethanol-insoluble, and an ethanol-soluble fraction, each progressively poorer in galacturonic acid units and progressively richer in arabinose units; all relatively poor in xylose units.  相似文献

Heparin, carboxy-group-reduced heparin, several sulphated monosaccharides and disaccharides formed from heparin, and a tetrasaccharide prepared from chondroitin sulphate were treated at 100 degrees C with hydrazine containing 1% hydrazine sulphate for periods sufficient to cause complete N-deacetylation of the N-acetylhexosamine residues. Under these hydrazinolysis conditions both the N-sulphate and the O-sulphate substituents on these compounds were completely stable. However, the uronic acid residues were converted into their hydrazide derivatives at rates that depended on the uronic acid structures. Unsubstituted L-iduronic acid residues reacted much more slowly than did unsubstituted D-glucuronic acid or 2-O-sulphated L-iduronic acid residues. The chemical modification of the carboxy groups resulted in a low rate of C-5 epimerization of the uronic acid residues. The hydrazinolysis reaction also caused a partial depolymerization of heparin but not of carboxy-group-reduced heparin. Treatment of the hydrazinolysis products with HNO2 at either pH 4 or pH 1.5 or with HIO3 converted the uronic acid hydrazides back into uronic acid residues. The use of the hydrazinolysis reaction in studies of the structures of uronic acid-containing polymers and the implications of the uronic acid hydrazide formation are discussed.  相似文献

The role of uronic acid oxidase in abscission was studied in explants of citrus ( Citrus sinensis L. Osbeck; var. Shamouti) leaves and fruits. In leaf explants, activity of uronic acid oxidase prior to onset of abscission and the rate of abscission were markedly accelerated by ethylene and delayed by 2,4-dichlorophenoxyacetie acid. Similar results were obtained for uronic acid oxidase activity in the exocellular fraction of young fruit explants. In mature fruit explants, treated with ethylene, an immediate increase in activity was evidenty in the non-active shoot/peduncle abscission zone, whereas in the calyx abscission zone the rise in activity occurred after a prolonged exposure to ethylene, when most of the fruits had already abscised. Whenever ethylene enhanced uronic acid oxidase activity, 2,4-dichlorophenoxyacetic acid delayed it. A gradient of decreasing activity or uronic acid oxidase was recorded from both sides of the abscission zone in leaves and fruits toward the separation line, where activity was the lowest as compared with the activity found in adjacent tissues. It is suggested that uronic acid oxidase is involved in senescence and cell wall degradation. However, it is yet questionable whether this enzyme is directly related to the control mechanism of abscission.  相似文献

Vitrification of stem explants of carnation was brought about by culturing in liquid medium. Cellulose and lignin levels were decreased in vitrified stems and leaves. Isolated cell walls of vitrified tissues were also characterized by low calcium content, low Ca2+/uronic acids ratio, low ratio of uronic acids to neutral sugars due to higher amounts of the latters. All these characteristics may account for the high wall plastic potential previously measured in vitrifying internodes.  相似文献

Dermatan sulfate proteoglycan chains were detected in tissue sections treated with chondroitin Blyase (0.01 units/ml) in 20 mM Tris-HC1 (pH 8.0) for i hr, followed by staining with antibody 9A2 specific for unsaturated uronic acid coupled to N-acetylgdactosaa- m i n d sulfate. In contrast, after treatment with chondroitin B-lyase, no positive stpining was observed with antibodies 3B3 and 1B5 which react to the unsaturated uronic acid coupled to N-acetylgalactosamine 6-sulfate and unsaturated uronic acid coupled to N-acetylgalactospmine, respectively. The distribution of dermatan sulfate thus revealed was mnfirmed by comparison with that found by monoclonal antibody 6B6 which reacts with small pmteoglycans carrying dermatan sulfate side chains. The localization of positive staining in fib- connective tissues was almost identical with these two procedures.  相似文献

It has been shown that the maxima (Bragg-spacings 4,5-19 A) on the X-ray diffraction patterns of the bovine lens native tissues from nuclear and cortical parts are predominantly due to the water-soluble crystallin intramolecular structure. The structures of water-soluble and water-insoluble fractions from bovine lens nucleus and cortex were qualitatively compared. Reversible dependence of the lens water-soluble protein structure on water content in the system was demonstrated.  相似文献

—The isolation of uronic acid-containing glycosaminoglycans from peripheral nerve and spinal cord of monkey was done by combining the cetyl pyridinium procedure and DEAE-Sephadex column chromatography. The constituent analyses of the isolated GAG-fractions indicated that hyaluronic acid, chondroitin-4-sulphate, chondroitin-6-sulphate, heparan sulphate and a testicular hyaluronidase-resistant galactosamine-containing GAG were present in both tissues. Hyaluronic acid was the predominant GAG (63 per cent) in both tissues and its level was much higher than in brain. Chondroitin-4-sulphate constituted 16 per cent in both tissues. The levels of heparan sulphate and hyaluronidase-resistant galactosamine-containing GAG in these tissues were much lower than in brain. The results indicate that the patterns of GAGs in peripheral nerve and spinal cord of monkey are similar but differ from that of brain.  相似文献

Two pectic fractions were extracted along the growth gradientof the mung bean hypocotyl. The first one (PF1) contained pectinssoluble in boiling water and characterized by low uronic acids/cationsratio, high esterification degree and high neutral sugars/acidicsugars ratio. The second one (PF2) contained pectins insolublein boiling water characterized by high uronic acids/cationsratio, low esterification degree, very low neutral sugar contentand a high selectivity for calcium. Water soluble pectins werepresent in all the wall area whereas the other ones were detectedmainly in the middle lamella. The PF1/PF2 ratio was high infast growing tissues but low in mature, slowly growing tissues.The development of the cell wall exchange properties along thegrowth gradient was related to the changes observed in the PF1/PF2balance. Papain (EC 3.4.22.2) has been coupled to supports of titanium (IV) oxide and cellulose, which are particulate and pre-coated with diazotised 1,3-diaminobenzene, giving water-insoluble and stable derivatives which possess low proteolytic activity but high esterolytic activity. In addition the reversible binding of zinc (II) at the active site of papain has been exploited to inhibit protectively the enzyme during its linkage to the aforementioned supports, thereby yielding water-insoluble derivatives of papain having superior activity upon reactivation with EDTA. Application of the improved procedure of enzyme coupling to macroporous cellulose particles gave a water-insoluble derivative of papain having further enhanced proteolytic activity. Other properties of the water-insoluble derivatives of papain and of similarly prepared water-insoluble conjugates of urease (EC 3.5.1.5) and cholinesterase (EC 3.1.1.8) with cellulose are also reported.  相似文献

A method for quantitative determination of uronic acids and the degree of esterification of their carboxyl groups in plant tissues and products of their processing is described. The method involves determination of the difference between the concentrations of Cu2+ in solution before and after interaction of copper with the substance in question. T. daniellii gel contains residues of L-arabinose, D-xylose, D-glucuronic acid, and 4-O-methyl-D-glucuronic acid in the ratios 1.00:7.20:1.91:0.66, together with nitrogen (1?%) and ash (3.1 %). The ash-free gel contains 76% of pentose and 24% of uronic acid; 25% of the uronic acid occurs as the 4-O-methyl derivative. All of the uronic acid residues in the polysaccharide are susceptible to periodate oxidation. Methylation studies suggest that the uronic acids occur as terminal side-substituents to a xylan back-bone and that the polysaccharide is highly branched. Enzymolysis with β-D-glucuronidase liberates a substantial part of the uronic acid, but does not completely depolymerise the gel.  相似文献

The extracellular polysaccharide polymers can bind microbes to surfaces and can cause physical modification of the microenvironment. Since uronic acids appear to be the components of these extracellular films that are most concentrated in a location outside the cell membrane, a quantitative assay for uronic acids was developed. Polymers containing uronic acids are resistant to quantitative hydrolysis, and the uronic acids, once released, form lactones irreproducibly and are difficult to separate from the neutral sugars. These problems were obviated by the methylation of the uronic acids and their subsequent reduction with sodium borodeuteride to the corresponding alcohol while they were in the polymer and could not form lactones. This caused the polymers to lose the ability to adhere to their substrates, so they could be quantitatively recovered. The hydrolysis of the dideuterated sugars was reproducible and could be performed under conditions that were mild enough that other cellular and extracellular polymers were not affected. The resulting neutral sugars were readily derivatized and then were separated and assayed by glass capillary gas-liquid chromatography. The dideuterated portion of each pentose, hexose, or heptose, identified by combined capillary gas-liquid chromatography and mass spectrometry, accurately provided the proportion of each uronic acid in each carbohydrate of the polymer. Examples of the applications of this methodology include the composition of extracellular polymers in marine bacteria, invertebrate feeding tubes and fecal structures, and the microfouling films formed on titanium and aluminum surfaces exposed to seawater.  相似文献

Summary Effects of novel (Streptomyces) hyaluronidase digestion upon the alcian blue (pH 1.0), alcian blue (pH 2.5), periodic acid-Schiff (PAS) and alcian blue-PAS stainings of mucopolysaccharides have been tested in the cartilage and aorta tissues of the rabbit and rat. These effects were compared with those of testicular hyaluronidase digestion upon the same stainings in the tissues. Evidence has been presented that a Streptomyces hyaluronidase digestible alcian blue (pH 2.5) reactive mucosubstance (hyaluronic acid) is present in the peripheral areas of the cartilage tissues and all the layers of the aorta tissues. Furthermore, the histochemical data obtained in this study appear to present a base line for establishing a promising enzyme digestion technique for the identification of hyaluronic acid in mucosaccharide histochemistry.  相似文献

Summary Methods designed to measure the cation-exchange properties of plant tissues have been reviewed. They depend on the replacement of the exchangeable cations of the tissues by H+, which is then determined directly, or indirectly by exchange for another cation, usually calcium.A rapid and reproducible method using dried milled plant material has been developed which employs acid-washing followed by titration to pH 7 using a glass electrode. This method has been used successfully for measuring the cation-exchange capacity (C.E.C.) of different tissues of a wide range of higher and lower plants, and gives values which correlate well with their related uronic acid content.  相似文献

采用高效液相色谱法对三个产地茶叶多糖的糖醛酸含量进行了测定,该方法具有操作简便、样品无需预处理等特点,稳定性、重现性较好,回收率高,适合于茶叶多糖糖醛酸含量的测定。结果表明不同产地茶叶多糖的糖醛酸含量有明显差异。本文同时研究了不同产地茶叶多糖对超氧自由基和DPPH自由基的清除作用,以评价其抗氧化活性。结果发现不同产地茶叶多糖的抗氧化活性与其糖醛酸含量呈正相关,糖醛酸含量越高,其抗氧化能力越强。  相似文献

Glycosaminoglycans (GAGs) are known to participate in central nervous system processes such as development, cell migration, and neurite outgrowth. In this paper, we report an initial glycomics study of GAGs from the porcine central nervous system. GAGs of the porcine central nervous system, brain and spinal cord were isolated and purified by defatting, proteolysis, anion-exchange chromatography, and methanol precipitation. The isolated GAG content in brain was 5 times higher than in spinal cord (0.35 mg/g of dry sample, compared to 0.07 mg/g of dry sample). In both tissues, chondroitin sulfate (CS) and heparan sulfate (HS) were the major and the minor GAG, respectively. The average molecular masses of CS from brain and spinal cord were 35.5 and 47.1 kDa, respectively, and those for HS from brain and spinal cord were 56.9 and 34 kDa, respectively. The disaccharide analysis showed that the compositions of CS from brain and spinal cords are similar, with uronic acid (1→3) 4-O-sulfo-N-acetylgalactosamine residue corresponding to the major disaccharide unit (CS type A) along with five minor disaccharide units. The major disaccharides of both brain and spinal cord HS were uronic acid (1→4) N-acetylglucosamine and uronic acid (1→4) 6-O-sulfo-N-sulfoglucosamine, but their composition of minor disaccharides differed. Analysis by (1)H and two-dimensional NMR spectroscopy confirmed these disaccharide analyses and provided the glucuronic/iduronic acid ratio. Finally, both purified CS and HS were biotinylated and immobilized on BIAcore SA biochips. Interactions between these GAGs and fibroblast growth factors (FGF1 and FGF2) and sonic hedgehog (Shh) were investigated by surface plasmon resonance.  相似文献

The changes in total hemicellulose composition of leaf and stem tissues of field-grown wheat plants have been examined. In each plant tissue the percentage of xylose in the total hemicellulose increases with increasing plant maturity, that of galactose varies little and those of L-arabinose, D-glucose, and uronic acid decrease. There is a markedly higher proportion of D-glucopyranuronosyl than of 4-O-methyl-D-glucopyranuronosyl residues in leaf and stem tissues at all stages of maturity. The ratio of β(1 → 3) to β(1 → 4) linkages in the β-glucans, and the DP of these β-glucans decrease concommitantly with tissue maturity.  相似文献

An investigation was carried out to study the cation-anion balance in different tissues of tomato plants supplied with nitrate, urea, or ammonium nitrogen in water culture.

Irrespective of the form of nutrition, a very close balance was found in the tissues investigated (leaves, petioles, stems, and roots) between total cations (Ca, Mg, K and Na), and total anions (NO3, H2PO4, SO4−−, Cl) total non-volatile organic acids, oxalate, and uronic acids. In comparison with the tissues of the nitrate fed plants, the corresponding ammonium tissues contained lower concentrations of inorganic cations, and organic acids and a correspondingly higher proportion of inorganic anions. Tissues from the urea plants were intermediate between the other 2 treatments. These results were independent of concentration or dilution effects, caused by growth. In all tissues approximately equivalent amounts of diffusible cations (Ca++, Mg++, K+ and Na+), and diffusible anions (No3, SO4−−, H2PO4, Cl) and non-volatile organic acids were found. An almost 1:1 ratio occurred between the levels of bound calcium and magnesium, and oxalate and uronic acids. This points to the fact that in the tomato plant the indiffusible anions are mainly oxalate and pectate. Approximately equivalent values were found for the alkalinity of the ash, and organic anions (total organic acids including oxalate, and uronic acids).

The influence of nitrate, urea, and ammonium nitrogen nutrition on the cation-anion balance and the organic acid content of the plant has been considered and the effects of these different nitrogen forms on both the pH of the plant and the nutrient medium and its consequences discussed.
