Summary Polarization colors of various purified collagens were studied in fibers of similar thickness. Three different soluble collagens of type I, insoluble collagen type I, lathyritic collagen type I, two p-N-collagens type I, pepsin extract collagen type II, two soluble collagens type III, p-N-collagen type III, and soluble collagen type V were submitted to a routine histopathologic procedure of fixation, preparation of 5-m-thick sections, staining with Picrosirius red and examination under crossed polars. Polarization colors were determined for thin fibers (0.8 m or less) and thick fibers, (1.6–2.4 m). Most thin fibers of collagens and p-N-collagens showed green to yellowish-green polarization collors with no marked differences between the various samples. Thick fibers of all p-N-collagens, lathyritic and normal 0.15 M NaCl-soluble collagens showed green to greenish-yellow polarization colors, while in all other collagens, polarization colors of longer wavelengths (from yellowish-orange to red) were observed. These data suggested that fiber thickness was not the only factor involved in determining the polarization colors of Picrosirius red-stained collagens. Tightly packed and presumably, better aligned collagen molecules showed polarization colors of longer wavelengths. Thus, packing of collagen molecules and not only fiber thickness plays a role in the pattern of polarization colors of Picrosirius red-stained collagens. 相似文献

Although flavonoid molecules have attracted considerable interest in recent years because of their antioxidant effect, there are considerable differences in their chemical properties. Electron paramagnetic resonance (EPR) spectroscopy was used to compare the oxidative free radical chemistry of two such molecules, kaempferol and luteolin, which have the same empirical formula but differ in the position of one OH group. Whereas the basic flavonoid structure remains intact in luteolin, structural changes occur in kaempferol after one-electron oxidation. Autoxidation of kaempferol in alkaline solution and oxidation by at pH 7 led to rapid fragmentation. In contrast, oxidation by horseradish peroxidase/hydrogen peroxide, xanthine/xanthine oxidase (X/XO) or a Fenton reaction system produced a radical whose structure appeared to be based on dimerisation of either the original or a fragment of the flavonoid. Hence, the biological properties of kaempferol are likely to be determined by the chemistry of its oxidation products. 相似文献

Force constant values for thermal vibrational motion of a collagen molecule along the helix axis in tendon, completely demineralized bone (CDB), and partially demineralized bone (PDB) were estimated by determining the Debye–Waller factor (DW factor) for the diffracted X-ray intensity from these specimens. The DW factor for nominal value of 0.286 nm meridional diffraction representing a period along the helical axis of a collagen molecule was measured. As the atomic scattering factor of mineral constituents is much larger than that of collagen, it is difficult to detect the diffraction from collagen in bone specimen. Therefore, PDB was used in this study. In order to compare obtained force constant value for CDB with mechanical properties of collagen in the literature, the value was translated into Young's modulus value using the cross-sectional area of a collagen molecule. In the case of collagen in PDB, i.e., collagen with the close presence of HAp mineral particles, as the DW factor of the diffracted intensity by hydroxyapatite (HAp) was considered to be negligible compared with that of collagen, the DW factor determined was interpreted as that of collagen molecule in PDB specimen. The force constant value obtained for collagen in PDB was significantly larger than that of collagen in CDB. This result was thought to be a manifestation of the hardening of collagen matrix in bone by HAp mineral particles and the first straightforward evidence for a difference in collagen properties depending on the presence of HAp mineral particles. It is well known that pulp density and particle size determine the available surface area concentration and have an influence
in the overall rate of bioleaching of minerals. As metal solubilization takes place through the surface area of the particles,
it can be expected that different combinations of pulp densities and particle sizes giving the same surface area concentration
would determine the same leaching rate. The objective of this work was to test this hypothesis on the effect of surface area
concentration, pulp density and particle size of the biooxidation of a pyritic gold concentrate by the thermophilic Archaeon
Sulfolobus metallicus in shake flasks. The gold concentrate was used at 2.5%, 5%, 10%, and 15% w/v pulp density and at four size fractions: 150–106,
106–75, 75–38 and –38 μm. Temperature was 68°C and the initial pH was 2.0. Results showed that the volumetric productivities
of iron and sulfate depend not only on the surface area concentration but also on pulp density and particle size considered
separately. These two variables not only determine surface area but also exert additional effects on the process, so the hypothesis
was not confirmed. Maximum attained iron productivity was 1.042 g/l day with the 75–38 μm fraction at 5% pulp density. Maximum
sulfate productivity was 4.279 g/l day with the 75–38 μm fraction at 10% pulp density. 相似文献

It has been shown that in the skin of chronically irradiated rats the proportion of collagen type III as compared to collagen type I is increased; on the other hand, no changes in the overall proportion to collagen were observed in the skin. It appears that the increased proportion of collagen type III in chronically irradiated rats is responsible for the decreased solubility of cutaneous collagen in these animals. Concomitantly, indirect evidence was accumulated for the presence of an additional cross-link in type III collagen, present only when irradiated animals served as the collagen source. This cross-link is located subterminally as long as it is not removed by limited pepsin digestion. It was concluded that the physiological decrease in solubility and the decrease in solubility observed in chronically irradiated animals have a different molecular background. 相似文献

Current antiretroviral therapies have improved the duration and quality of life of people living with HIV-1. However, viral reservoirs impede complete eradication of the virus. Although there are many strategies to eliminate infectious virus, the most actively pursued are latency reversing agents in conjunction with immune modulation. This strategy, known as “shock and kill”, has been tested primarily against the most widely recognized HIV-1 latent reservoir found in resting memory CD4+ T cells. This is in part because of the dearth of conclusive evidence about the existence of non-T cell reservoirs. Studies of non-T cell reservoirs have been difficult to interpret because of technical and biological issues that have hampered a better understanding. This review considers the current knowledge of non-T cell reservoirs, the challenges encountered in a better understanding of these populations, and their implications for HIV-1 cure research. 相似文献

The Volta phase plate is a recently developed electron cryo-microscopy (cryo-EM) device that enables contrast enhancement of biological samples. Here we have evaluated the potential of combining phase-plate imaging and single particle analysis to determine the structure of a small protein–DNA complex. To test the method, we made use of a 200 kDa Nucleosome Core Particle (NCP) reconstituted with 601 DNA for which a high-resolution X-ray crystal structure is known. We find that the phase plate provides a significant contrast enhancement that permits individual NCPs and DNA to be clearly identified in amorphous ice. The refined structure from 26,060 particles has an overall resolution of 3.9 Å and the density map exhibits structural features consistent with the estimated resolution, including clear density for amino acid side chains and DNA features such as the phosphate backbone. Our results demonstrate that phase-plate cryo-EM promises to become an important method to determine novel near-atomic resolution structures of small and challenging samples, such as nucleosomes in complex with nucleosome-binding factors. 相似文献

Strongly biased codon usage is common in unicellular organisms, particularly in highly expressed genes. The bias is most simply explained as a balance between selection and mutation, with selection favouring those codons which are more efficiently translated. In a review Ikemura (1985) has proposed four rules for predicting which codons will be preferred, based on the properties of the transfer RNAs responsible for translating messenger RNA into protein. In this paper codon usage in E. coli and yeast is re-examined using the recent compilation of Maruyama et al. (1986). The codon adaptation index of Sharp and Li (1986a) is used as a measure of gene expression to investigate the importance of this factor. It is found that Ikemura's rules successfully predict preferred codons for yeast, but that two of them work less well for E. coli, and it is suggested that some of the apparent bias in weakly expressed genes of E. coli may be due to contextual effects on mutation rates. 相似文献

1. The fatty acid (FA) composition of Daphnia galeata and their algal food was analysed and showed many similarities, however, some significant differences were also found in the relative abundance of the FA C16 : 4ω3 and docosahexaenoic acid (DHA). Their relative abundances were much lower in daphnids than in their algal diet.
2. When daphnids were fed three distinct emulsion particles with DHA : eicosapentaenoic acid (EPA) ratios of c. 0.7, 2 and 4, the final DHA : EPA ratio in the daphnids always favoured EPA. The increase of the food DHA : EPA ratio resulted in a minor increase of DHA (to c. 2%). Feeding the animals on emulsion particles with increasing ratios of DHA : EPA, caused a minor ( c. 2%) increase of DHA level but EPA levels remained high ( c. 10%).
3. When labelled with [14 C]linoleic acid and [14 C]linolenic acid daphnids showed low conversion of both essential FA into C20 polyunsaturated fatty acids (PUFAs). This low conversion activity might explain the importance of C20 PUFAs as dietary compounds in the food of Daphnia.
4. The results indicate the insignificance of DHA and C16 : 4ω3 for daphnids. As EPA can be derived from C18 : 3ω3 it is not strictly essential, although it might be a significant factor in food quality for Daphnia. 相似文献

The global initiatives of monitoring and conserving pollinators require worldwide assessments with comparable data sets collected through standardized methods. The use of pan traps is a passive method widely applied to sample flower visitors, standing out for its simplicity. Despite its wide use to sample pollinator diversity, the influence of color on trap efficiency is not well understood. The available studies are particularly scarce in the tropics and have generated divergent results. The main goal of the present study was to assess whether blue, yellow and white pan traps are complementary to sample Hymenoptera community. For this, we placed 49 sample units of blue, white and yellow pan traps in agricultural and natural (savanna-like) areas in Chapada Diamantina, Bahia, Brazil. We found that the species richness from blue and yellow pan traps were not significantly different, but both were significantly greater than the species richness from white pan traps. However, bees were significantly more attracted to the blue pan traps and wasps to the yellow ones; thus, color attractiveness was group-specific. Pan traps of different color showed low species composition overlap with 61 % of species collected exclusively in one of the three pan trap colors, and the species composition in the blue traps differed consistently from that in the traps of the other colors. In the article we discuss the implication of the results and defend the combined use of pan traps with different colors as a solution for the differential variable sample bias. 相似文献

The biological removal of petroleum products using landfarming has been applied commercially in large scale with relative success. The technology has been widely used due to its simplicity and cost-effectiveness. However, together with these advantages, there are physical, chemical and biological aspects of the technology that can hamper the remediation process. The dominant pollutant removal mechanisms involved in landfarming are volatilisation of low molecular weight volatile compounds during the early days of contamination or treatment, biodegradation and adsorption. However, volatilisation, leaching of the petroleum products and the remaining recalcitrant hydrocarbon residues present both health and environmental challenges to the rehabilitation practitioners when designing the landfarming technology. Bioaugmentation and biostimulation are promising bioremediation approaches involving landfarming. However, due to the inherent problems related to bioaugmentation such as poor survival of augmented strains, biostimulation should be preferred in contaminated sites with indigenous pollutant-degrading bacteria. Although simplicity and cost-effectiveness are the major advantages for using landfarming, other factors generally regarded as disadvantageous to implementing the technology can be addressed. These includes requirements for large land area for treatment, availability of the pollutant degrading bacteria, effectiveness of the technology at high constituent concentration (more than 50,000 ppm), improved concentration reductions in cases requiring more than 95% of pollution reduction and the flexibility of the technology in integrating the removal of petroleum hydrocarbons with other contaminants that may occur with the petroleum products. 相似文献

Student evaluations of faculty are important components of the medical curriculum and faculty development. To improve the effectiveness and timeliness of student evaluations of faculty in the physiology course, we investigated whether evaluations submitted during the course differed from those submitted after completion of the course. A secure web-based system was developed to collect student evaluations that included numerical rankings (1-5) of faculty performance and a section for comments. The grades that students received in the course were added to the data, which were sorted according to the time of submission of the evaluations and analyzed by Pearson's correlation and Student's t-test. Only 26% of students elected to submit evaluations before completion of the course, and the average faculty ratings of these evaluations were highly correlated [r(14) = 0.91] with the evaluations submitted after completion of the course. Faculty evaluations were also significantly correlated with the previous year [r(14) = 0.88]. Concurrent evaluators provided more comments that were statistically longer and subjectively scored as more "substantive." Students who submitted their evaluations during the course and who included comments had significantly higher final grades in the course. In conclusion, the numeric ratings that faculty received were not influenced by the timing of student evaluations. However, students who submitted early evaluations tended to be more engaged as evidenced by their more substantive comments and their better performance on exams. The consistency of faculty evaluations from year to year and concurrent versus at the end of the course suggest that faculty tend not to make significant adjustments to student evaluations. 相似文献
