Yarrowia lipolytica lipase has been assumed to be a good candidate for the treatment of fat malabsorption in patients with pancreatic insufficiency. Nevertheless, no systematic studies on its stability under physiological conditions pertaining to the human GI (gastrointestinal) tract have been published. Stability of various Y. lipolytica lipase powder formulations at various physiological pH values as well as the effect of digestive proteases and bile salts on enzyme activity were investigated. Results were compared with those obtained from another competing fungal lipase sourced from Candida rugosa. Among the studied formulations, Y. lipolytica lipase stabilized with gum arabic and skimmed milk powder was the most promising powder formulation. Under acidic conditions (pH 3-5), this formulation showed higher stability than those observed with the other Y. lipolytica lipase formulations and C. rugosa lipase. In addition, in the presence of gum arabic and skimmed milk powder as additives, Y. lipolytica lipase exhibited markedly higher resistance to pepsin, trypsin and chymotrypsin actions. Resistance to proteolytic degradation by digestive proteases was also by far higher than that observed with C. rugosa lipase. Similar behaviour was, however, observed when these two fungal lipases were incubated with increased concentrations of bile salts. Residual lipase activity of both fungal lipases showed a slight decrease in NaTDC (sodium taurodeoxycholate) concentration above 4 mM. Consequently, Y. lipolytica lipase formulated with gum arabic and milk powder seemed to have great potential for use as a therapeutic tool for patients with pancreatic insufficiency.  相似文献

Organisms able to grow on arabinogalactan, pectin, xylan, wheat bran, guar, apple cell walls, and mucin were isolated by enrichment from human feces. The number of polysaccharide fermenters and the properties of the predominant bacteria varied between subjects. The ability to use one polysaccharide was not related to the ability to use others. Some organisms (e.g., Bacteroides spp.) isolated on other substrates also utilized mucin, but were not isolated in the mucin enrichment. The mucin fermenters isolated by enrichment had a very restricted ability to utilize complex polysaccharides and their constituent monosaccharides, suggesting that the presence of plant polysaccharides in the human colon is unlikely to prevent the use of colonic mucin as an energy source by bacteria. Characterization with a range of biochemical tests showed that many of the isolates, but especially the mucin fermenters, did not resemble organisms described previously.  相似文献

Lactobacillus acidophilus 5e2 when grown on skimmed milk, skimmed milk supplemented with sodium formate and skimmed milk supplemented with glucose secretes a branched heteropolysaccharide having a weight average molecular weight less than 450 kDa. The exopolysaccharide has a heptasaccharide repeat unit and is composed of D-glucose, D-galactose and N-acetyl-D-glucosamine in the molar ratio 3:3:1. Polyclonal antibodies against P-l, a pectic polysaccharide fraction extracted with 0.5m NaOH from the kernels of Prunus mume and consisted of arabino-galacturonan, and 1–3, the partial acid (0.1 m trifluoroacetic acid) hydrolysate of P-l, were prepared in Japanese white rabbits. Competitive elisa experiments strongly suggested that anti P-l and anti 1–3 antibodies were different but P–l and 1–3 cross-reacted with each other to recognize a partly similar epitope structure. The reactivities of polysaccharide fractions from the raw flesh of P. mume, and the kernels of apricot and peach extracted with either water or sodium hydroxide were examined using both antisera by the indirect competitive elisa method. The polysaccharide fractions extracted with sodium hydroxide solutions had the reactivities but not those extracted with cold and hot water. These facts suggested that the similar structure of polysaccharides to P-l was present in the flesh of P. mume and the kernels of apricot and peach. However, neither pectin of apple nor citrus had reactivity with each antiserum. P-l would be different in chemical structure from a commercially available pectin, a water-soluble polysaccharide from apple and citrus.  相似文献

The interaction of gum arabic, maltodextrin and pullulan with lipids in emulsion systems was investigated. Interfacial tension and interfacial viscosity measurements revealed that only gum arabic could adsorb and form a viscoelastic film at the oil-water interface. Good emulsifying activity was demonstrated for gum arabic, whereas fine emulsions could not be produced from the other polysaccharide solutions and oil. Frequency-dependent increases in the storage and loss moduli were observed for all the polysaccharide solutions. Such rheological behavior did not substantially change when maltodextrin and pullulan were mixed with oil to form emulsions. However, the frequency-dependence of the dynamic moduli disappeared in the gum arabic-stabilized emulsion, suggesting the formation of a network structure in which oil droplets could form junctions with gum arabic chains. The results on the inhibition of lipid oxidation by polysaccharides suggest that gum arabic protected lipids from the attack of lipoxygenase and free radicals by adsorbing at the oil droplet surface.  相似文献

Putrescine (1,4-diaminobutane) was covalently linked to alginate and low-methoxyl pectin to synthesize new aminated polysaccharides. Both putrescine–pectin and –alginate conjugates, although the latter at higher concentrations, were found to be able to act as effective acyl acceptor transglutaminase substrates in vitro using both dimethylated casein and soy flour proteins as acyl donors. Monodansylcadaverine, a well known acyl acceptor transglutaminase substrate, dose-dependently counteracted the covalent binding of the aminated polysaccharides to the proteins. Putrescine–pectin conjugate was also tested to prepare, in combination with soy flour proteins, edible films in the presence of purified microbial transglutaminase. Characterization of the enzymatically crosslinked films showed a significant decreased water vapor permeability, with respect to the ones obtained with non-aminated pectin in the presence of transglutaminase, as well as improved mechanical properties, such as high extensibility. Possible biotechnological applications of hydrocolloid films containing putrescine–polysaccharide derivatives enzymatically crosslinked to proteins were suggested.  相似文献

Three fractions of acidic water-soluble polysaccharides (concentration of glucuronic acid 10?C65%) were obtained from the above-ground part of St. Johns wort Hypericum perforatum L. by serial extraction with water and 0.7% aqueous solution of ammonium oxalate. Enzymatic hydrolysis of these polysaccharides using endo-polygalacturonase indicates that their carbohydrate chains contain the units of galacturone formed by 1,4-??-linked residues of non-substituted D-galacturonic acid. The extracted polysaccharides have been purified by means of gel filtration. It has been shown that water-soluble polysaccharides obtained by extraction with water manly contain the residues of galactose, mannose, glucose, and arabinose (the concentration of glucuronic acid being 10?C27%) while the polysaccharide fraction extracted using 0.7% aqueous solution of ammonium oxalate is presented by pectin polysaccharides. Only the residues of galacturonic acid (55?C72%) have been identified among glucuronic acids in its composition using chromatography/mass spectrometry of trimethylsilyl derivatives. In addition, this fraction contains the residues of the neutral monosaccharides which are typical for pectins: arabinoses, galactoses, rhamnoses, and glucose; there are also minor concentrations of residues of xylose and mannose. IR spectra of pectin polysaccharides of St. John??s wort have absorption bands in the ranges 1740, 1640?C1620, 1236?C1200, and 1200?C1000 cm?1 which are typical for pectins. It has been demonstrated that aqueous solutions of pectin polysaccharides of St. John??s wort (2 mg/mL) have pronounced antioxidant activity (44% of the activity of trolox taken for 100%).  相似文献

New water-soluble derivatives of starch, pectin, and Na-CMC containing the sulfamic groups have been obtained by the reaction of sulfamic acid with dialdehyde polysaccharide derivatives. The structure and composition of the resulting compounds have been studied by IR spectroscopy, elemental (nitrogen and sulfur) analysis, and X-ray diffraction. The sulfamic derivatives of starch, pectin, and Na-CMC with a different content of the sulfamic groups have been obtained by varying the ratio of sulfamic acid to the dialdehyde polysaccharide derivatives. The optimal–СНО: NH2SO3H ratio was found to be 1: 2.5. The interaction rate of sulfamic acid with the dialdehyde derivatives of starch, pectin, and Na-CMC has been evaluated. The antibacterial and antifungal effects of sodium salts of the sulfamic starch, pectin, and Na-CMC derivatives against Gram-positive and Gram-negative bacteria and fungi have been studied at different concentrations (10, 25, 50 mg/mL) by the disk diffusion method. The synthesized compounds have not been found to exhibit antifungal activity against Candida albicans. Nevertheless, they have been shown to have the antibacterial activity against Staphylococcus aureus, Staphylococcus epidermidis, Escherichia coli, Proteus vulgaris, Streptococcus faecalis, Streptococcus pyogenes, and Streptococcus faecalis at the concentration of 50 mg/mL. The concentration dependence of antibacterial action of sodium salts of the starch, pectin, and Na-CMC sulfamic derivatives has been demonstrated. The antibacterial activity of the drugs has been found to directly depend on the content of the sulfamic groups in polysaccharides. The results on the acute toxicity of the sulfamic polysaccharide derivatives have shown that these compounds can be attributed to low-toxicity substances of Class V.  相似文献

Cell wall materials in the form of water-insoluble solids (WIS) and water-soluble fractions (WSF) were prepared from apples stored at 4 °C for 30 weeks. During storage, the WIS content decreased whereas the WSF content remained unchanged. The total amount of polysaccharides decreased, in particular the pectic polymers which decreased by 10%. In contrast, the soluble pectic fraction increased by 40% whilst its degree of methoxylation remained constant. The arabinose and galactose content progressively declined. The enzymatic treatment of the apple tissues was more effective the longer the storage; yields correlated well with the enzyme hydrolysis of WIS. The accessibility of pectin to poly-galacturonases in apple tissues is discussed since it was higher at the end of storage, whereas the solubilisation of pectins from WIS by polygalacturonases remained constant. On the other hand, with liquefying enzymes, the yield of pectin solubilisation from apple tissues or WIS were well correlated and increased with storage time.  相似文献

Heat treatment during the production of skimmed milk powder causes denaturation of proteins, thereby affecting the physicochemical properties of the skimmed milk powder. To understand the effects of heat treatment on the sensitivity of the casein micelles in skimmed milk powders, low heating type (L), normal heating type (N), high heating type (H), and super-high heating type (SH), to reaction with rennet, rennet-induced curd formation was investigated. A well-developed network structure with wide spaces was observed only in the curd derived from the solution of type L skimmed milk powder. SDS–PAGE suggested that there was no difference in the amount of glycomacropeptide generated from κ-casein in the four types of skimmed milk powder, but casein micelles in the solution of type L skimmed milk powder formed aggregates most effectively. These results are discussed with respect to the thermal denaturation of proteins in skimmed milk powder.  相似文献

A rapid gravimetric assay for estimation of polysaccharide content in complex culture media was developed and validated. The quantitative assay showed great selectivity for the determination of xylan and pectin, eliminating all interferences from other medium components. In addition, our data showed this protocol could also be applied to the estimation of other polysaccharides in liquid media such as cellulose Avicel®, chitin, starch and crude hemicellulose. Therefore, this method represents a useful tool for monitoring the in vitro microbial degradation of polysaccharides.  相似文献

A survey of the inhibition of the amplification of spinach DNA by various plant polysaccharides revealed that neutral polysaccharides (arabinogalactan, dextran, gum guar, gum locust bean, inulin, mannan, and starch) were not inhibitory. In contrast, the acidic polysaccharides (carrageenan, dextran sulfate, gum ghatti, gum karaya, pectin, and xylan)were inhibitory. In the process of preparing random amplified polymorphic DNAs (RAPDs), the loss of large DNA bands appears to be an indicator that the fingerprint pattern has been affected by polysaccharides. The addition of various concentrations of Tween 20, DMSO, or PEG 400 to the PCR reaction mixture resulted in partial restoration of amplification of RAPDs for the acidic polysaccharides. The most effective way to eliminate the effects of polysaccharide inhibition was by diluting the DNA extracts, and thereby diluting the polysaccharide inhibitors.  相似文献

Three industrial high methoxyl pectins have been fractionated by size exclusion chromatography (SEC) on a preparative scale and the chemical composition, viscosity and light scattering behaviour of the fractions have been investigated. Chemical analysis revealed that the composition varies greatly from one SEC fraction to another. In all three pectin samples, the fractions of low molecular size contain most of the free neutral polysaccharides as well as some free pectin ‘hairy regions’. In addition, the lemon pectin samples contain some pectin molecules of large size which are rich in neutral sugars. Phenolic and proteinaceous compounds coelute with neutral sugar-rich fractions. However, in the apple pectin, phenolics and proteins occur predominantly in the fractions of low molecular size. Lemon pectin molecules, especially that of the lemon A sample, are prone to aggregation in the presence of calcium cations. The aggregate fraction can be disrupted by shear forces, heating or the presence of a chelating agent. The formation of such calcium-pectinate aggregates seems to be due to the presence of some molecules with low degrees of methoxylation. Light scattering measurements also suggest that even very narrow SEC fractions remain highly heterogeneous on the basis of their molecular weight, thus indicating large differences in molecular conformation.  相似文献

Among 21 different polysaccharides tested, 5 greatly enhanced the spontaneous and cyclic AMP-induced formation of exolipase: glycogen, hyaluronate, laminarin, pectin B, and gum arabic. These polysaccharides have in common the tendency to form highly ordered networks because of the branching or helical arrangement, or both, of their molecules. None of the polysaccharides could be utilized by the cells as the sole carbon source. Strong lipid extraction of four different polysaccharides did not reduce their exolipase-enhancing efficacy. At a constant cell density the stimulation of exolipase formation by various concentrations of glycogen followed saturation kinetics, suggesting a limited number of "sites" for the glycogen to act. The active principle present in a solution of pectin was destroyed by degradation (beta-elimination) of the polymer. Hyaluronate lost its exolipase-enhancing activity by exhaustive hydrolysis with hyaluronidase but was resistant to proteinase K. Exopolysaccharide, isolated from growth medium of Serratia marcescens SM-6, enhanced the exolipase formation as efficiently as hyaluronate. The results of this work are discussed mainly in terms of the "detachment hypothesis."  相似文献

Crude polysaccharide fractions, rich mainly in arabinogalactans (A), pectin (B) and glucuronoxylan-related polymers (D), have been obtained from aerial parts of sage (Salvia officinalis L.) by sequential extraction with various reagents. Arabinogalactans displayed on HPLC a dominance of lower molecular-mass polymers (MW < 10,000), while pectin and glucuronoxylan-related polysaccharides showed predominance of polymers with MW > 50,000. Individual polysaccharide fractions were examined for their immunomodulatory activity in the in vitro comitogenic thymocyte test. The polysaccharide fractions tested possessed the capacity to induce rat thymocyte proliferation in the order D>B>A. Besides, fraction D possessed a significant comitogenic effect, and the SIcomit/SImit ratio 3-4 indicates potential adjuvant properties of this glucuronoxylan-rich material.  相似文献

The polysaccharides produced by Streptococcus thermophilus Rs and Sts in skimmed milk consist of -Gal and -Rha in a molar ratio of 5:2. Linkage analysis and 1D/2D NMR (1H and 13C) studies revealed that both polysaccharides have the same branched heptasaccharide repeating unit:

Remarkably, the two strains differ in their effects on the viscosity of stirred milk cultures. The milk culture of S. thermophilus Rs is non-ropy and affords 135 mg/L polysaccharide with an average molecular mass of 2.6×103 kDa. In contrast, the milk culture of S. thermophilus Sts is ropy and produces 127 mg/L polysaccharide with an average molecular mass of 3.7×103 kDa. Permeability measurements of non-stirred milk cultures of both strains suggest that both strains have a similar effect on the protein–polysaccharide network. Therefore, the only clear difference between both strains, which may cause the difference in ropiness of the milk cultures, is the difference in molecular mass of the polysaccharide.  相似文献

Water-soluble polysaccharide fractions were extracted from the fruit of rowan Sorbus aucuparia L. by water and 0.7% ammonium oxalate water solution. The total yield was 4.2%. It is demonstrated that these fractions are pectin polysaccharides, and their carbon chains are primarily composed of galactunoric acid residue (up to 68%), arabinose and galactose. Sephacryl S-500 gelfiltration of rowan fruit pectin polysaccharides proved their relative homogeneity pertaining to their molecular weights, whereas endo-polygalacturonase enzymatic hydrolysis gives evidence of the presence of extended galacturonan (rhamnogalacturonan) ranges in their carbohydrate chains. Methylation of rowan pectin polysaccharides shows that their carbohydrate pendants are formed by 1,5-linked arabinofuranose residue, 1,4-linked glucopyranose residue, 1,6-linked galactopyranose residue, 1,3,6-linked mannopyranose residue and 1,3,6-linked galactopyranose residue. Glucopyranose residue is identified at non reducible ends of these pendants. It was demonstrated that antioxidant activity of water solutions of pectin polysaccharides extracted from rowan S. aucuparia L. (0.5 mg/mL) is 37?C53% of trolox activity, which is 100%.  相似文献

Enzymes find widespread industrial use in the modification of the functional properties of plant polysaccharides both in vivo and in vitro. Reactions catalysed include depolymerization, debranching and de-esterification, depending on the specific enzyme or enzyme mixture employed and on the particular industrial requirement. Depolymerization of pentosans and/or barley β-glucans to destroy their viscosity-building properties is essential in starch and gluten manufacture, in the mashign of barley malt and in the production of maltosaccharide syrups. Depolymerization of pectin is required in juice clarification and to allow concentration. However, in other instances the aim may be to maintain or, at most, only slightly alter the molecular size of functional polysaccharides, i.e. in the conversion of guar galactomannan to a locust-bean type galactomannan and in the enzymic treatment of wheat-flour doughs. Enzymes may also be used to produce specific oligosaccharide fragments from polysaccharides and as diagnostic tools in the measurement of a particular polysaccharide in a mixture.  相似文献
