Saccharina (Laminaria) japonica, a safe, cheap, and readily available macroalga can be used as a substrate for various microbial fermentations. This work investigated the feasibility of S. japonica as a substrate for production of pigments by the fungus Talaromyces amestolkiae GT11 in solid-state fermentation without additional salt and/or nitrogen sources. Under optimized conditions, the pigment exhibited maximum absorption spectrum at 410 (yellow) and 510 nm (red), and the pigment yield of 1,153.5 (yellow) and 506.2 (red) OD units g?1 of dry fermented substrate were achieved with a particle size of 1.0 mm and pH 7, although visually the pigment was reddish in color. The optimum incubation period, pH, moisture, inoculum size, and temperature were observed to be at 192 h, pH 7.0, 80 % (w/w) moisture, 1.8?×?106 spores mL?1 of inoculum g-1 of dry substrate and 28 °C. Hence, this study indicates the suitability of utilization of S. japonica as a substrate for natural pigment production by T. amestolkiae GT11 which can be used in food, cosmetics and pharmaceutical industries for various applications.  相似文献

Filamentous fungi are being globally explored for the production of industrially important bioactive compounds including pigments. In the present study, a cold and pH tolerant fungus strain Penicillium sp (GEU_37), isolated from the soil of Indian Himalaya, is characterized for the production of natural pigments as influenced by varying temperature conditions. The fungal strain produces a higher sporulation, exudation, and red diffusible pigment in Potato Dextrose (PD) at 15 °C as compared to 25 °C. In PD broth, a yellow pigment was observed at 25 °C. While measuring the effect of temperature and pH on red pigment production by GEU_37, 15 °C and pH 5, respectively, were observed to be the optimum conditions. Similarly, the effect of exogenous carbon and nitrogen sources and mineral salts on pigment production by GEU_37 was assessed in PD broth. However, no significant enhancement in pigmentation was observed. Chloroform extracted pigment was separated using thin layer chromatography (TLC) and column chromatography. The two separated fractions i.e., fractions I and II with Rf values 0.82 and 0.73, exhibited maximum light absorption, λmax, at 360 nm and 510 nm, respectively. Characterization of pigments using GC–MS showed the presence of the compounds such as phenol, 2,4-bis (1,1-dimethylethyl) and eicosene from fraction I and derivatives of coumarine, friedooleanan, and stigmasterole in fraction II. However, LC-MS analysis detected the presence of derivatives of compound carotenoids from fraction II as well as derivative of chromenone and hydroxyquinoline as major compounds from both the fractions along with other numerous important bioactive compounds. The production of such bioactive pigments under low temperature conditions suggest their strategic role in ecological resilience by the fungal strain and may have biotechnological applications.  相似文献

Pigments produced by species of Monascus have been used to coloring rice, meat, sauces, wines and beers in East Asian countries. Monascus can produce orange (precursor), yellow and red pigments. Orange pigments have low solubility in culture media and when react with amino groups they become red and largely soluble. The orange pigments are an alternative to industrial pigment production because the low solubility facilitates the downstream operations. The aim of this work was to study the kinetic on the production of orange pigments by Monascus ruber CCT 3802. The shaking frequency of 300 rpm was favorable to production, whereas higher shaking frequencies showed negative effect. Pigment production was partially associated with cell growth, the critical dissolved oxygen concentration was between 0.894 and 1.388 mgO2 L?1 at 30 °C, and limiting conditions of dissolved oxygen decreased the production of orange pigments. The maintenance coefficient (mo) and the conversion factor of oxygen in biomass (Yo) were 18.603 mgO2 g x ?1  h?1 and 3.133 gx gO 2 ?1 and the consideration of these parameters in the oxygen balance to estimate the biomass concentration provided good fits to the experimental data.  相似文献

The effect of pH and nitrogen source on pigment production by Monascus purpureus 192F using glucose as the carbon and energy source, was studied in pH-controlled, batch fermentor cultures using HPLC analysis to determine individual pigment concentrations. A maximum of four pigments were detected in fungal extracts. These were the yellow pigments monascin and ankaflavin, the orange rubropunctatin and the red pigment monascorubramine. Monascorubramine was present as the major product in all instances. Fungal growth and ankaflavin synthesis were favoured at low pH (pH 4.0), whereas production of the other pigments was relatively independent of pH. The nature of the nitrogen source affected fungal growth and pigment production, independent of pH. Ammonium and peptone as nitrogen sources gave superior growth and pigment concentrations compared to nitrate. Ankaflavin was not detected in nitrate cultures. The highest red pigment production was obtained using a glucose-peptone medium at pH 6.5, due to the secretion of red pigments into the medium under these conditions. Correspondence to: M. R. To maximize and sustain the productivity ofMonascus pigments, various environmental and nutritional parameters, such as the initial moisture content, pH, inoculum size, sample size, and nutrient supplement, that influence pigment production were evaluated in solid-state cultures as follows: initial moisture content, 50%; pH, 6.0; inoculum size 1 x 104 spore cells (grams of dry solid substrate)−1; sample size, 300 g. All supplementary nutrients (carbon, nitrogen, and mineral sources) added has inhibitory effects on the cell growth and red pigment production. In open tray culture the maximum biomass yield and specific productivity of red pigments were 223 mg DCW (grams of initial dry substrate)−1 and, 47.6 OD500 (DCW grams)−1 h−1, respectively.  相似文献

Optimal C:N ratio for the production of red pigments by Monascus ruber   总被引:1,自引:0,他引:1  
The carbon-to-nitrogen (C:N) ratio in the biomass of microfungi tends to be quite different (e.g. 10–15) compared with the C:N ratio in the red pigments (e.g. >20) of the fungus Monascus ruber. Therefore, determining an optimal C:N ratio in the culture medium for maximizing the production of the pigments is important. A culture medium composition is established for maximizing the production of the red pigment by the fungus M. ruber ICMP 15220 in submerged culture. The highest volumetric productivity of the red pigment was 0.023 AU L?1 h?1 in a batch culture (30 °C, initial pH of 6.5) with a defined medium of the following composition (g L?1): glucose (10), monosodium glutamate (MSG) (10), MgSO4·7H2O (0.5), KH2PO4 (5), K2HPO4 (5), ZnSO4·7H2O (0.01), FeSO4·7H2O (0.01), CaCl2 (0.1), MnSO4·H2O (0.03). This medium formulation had a C:N mole ratio of 9:1. Under these conditions, the specific growth rate of the fungus was 0.043 h?1 and the peak biomass concentration was 6.7 g L?1 in a 7-day culture. The biomass specific productivity of the red pigment was 1.06 AU g?1 h?1. The best nitrogen source proved to be MSG although four other inorganic nitrogen sources were evaluated.  相似文献

Many bacteria produce reddish brown to black pigments and some of these have been characterised. This report describes the isolation and characterisation of a diffusible brown melanin-like pigment from the bacterium Aeromonas media. Physico-chemical testing suggested that the pigment is a true melanin. New butanol-soluble yellow, red and brown pigments were isolated from the A. media strain under reducing conditions during melanogenesis and these pigments were shown to be unstable precursors of the polymeric brown melanin product.  相似文献

This study emphasis the production of yellow pigment from endolichenic Bacillus sp. isolated from the lichen Dirinaria aegialita (Afzel. ex Ach.) B.J. Moore. Yellow pigment-producing twenty different strains were investigated. The hyperactive pigment-producing bacterial strain was identified as Bacillus gibsonii based on 99 % sequence similarity. Maximum bacterial pigment production appeared in Luria Bertani medium. Methanol extraction of the pigment and its partial purification using TLC was carried out. Furthermore, isolated pigments were characterized using UV-visible spectroscopy, FTIR spectroscopy, and GC-MS results related to the possibility of the carotenoid occurrence. The pigment also exhibited efficient antifungal activity against selected fungal pathogens of economic importance. Likewise, the pigment extract evaluated for the total antioxidant potential using Phosphomolybdenum and Ferric reducing antioxidant power assay and the results represented in Ascorbic Acid Equivalent (AAE)- 21.45 ± 1.212 mg/mL. The SC50 of the pigment extract found to be 75.125 ± 0.18 µg/ml determined by the ABTS assay.  相似文献

Optimal physical parameters of the solid state fermentation of rice to produce anka pigments and their influences on pigment production were studied. Anka pigment production, especially that of two orange anka pigments (rubropunctatin and monascorubrin), was highly sensitive to the moisture content of the rice substrate. Optimal initial moisture content of rice substrate was 24%. Pigment formation was retarded when extra water was added to the inoculated substrate during cultivation. High filling amount of rice substrate in a flask was unfavorable for pigment production. Loosening of the inoculated substrate once a day enhanced pigment production. With a high carbon dioxide level in the incubator, no orange pigments were detected. Freeze drying the fermented material produced a superior yield of anka pigments, while oven drying at 50°C for 24 h was a reasonable alternative. Cost effective bioprocessing of nutraceuticals in present global scenario is a matter of concern. This study explored Paper mill sludge (PMS) and sugarcane bagasse (SCB) as inexpensive substrate for Planococcus sp. TRC1 mediated valuable β-carotene production and residual treated biomass as value added crystalline cellulose source simultaneously. Both biomass supported significant bacterial growth reaching highest yield 38.54 ± 1.4 mg/g on PMS (36 h) and 47.13 ± 1.9 mg/g (48 h) on SCB in solid state fermentation. Luedeking-Piret model revealed growth associated production with α and much lower β values of 5.18 and 0.24 for PMS and 4.5 and 0.165 for SCB. Cost analysis exhibited decrementation of pigment cost/mg by 84 % compared to synthetic media. Optimum production conditions were 30 °C temperature, pH 7, 10 % inoculum and initial moisture content 80 % (PMS) and 85 % (SCB). TLC (Rf = 0.9), HPLC (RT = 7.646) and lambda max (465 nm) confirmed pigment’s β-carotene nature with significant antioxidant and antimicrobial activity. It showed stability at varied temperature, pH and light conditions along with negligible phytotoxicity on Vigna radiata. Planococcus sp. TRC1 delignified PMS (41 %) and SCB (38 %) and FT-IR, FESEM and XRD suggested crystalline nature of residual cellulose rich fraction shedding light on a biorefinery approach for valorization of industrial solid wastes.  相似文献

The aim of this study was to evaluate the effects of different light quality of light emitting diode (LED) on the growth, concentration of chlorophyll and chlorophyll biosynthesis precursors of non-heading Chinese cabbage (Brassica campestris L.). Seedlings of the cultivar Te Ai Qing were cultured for 28 days under 6 treatments: red light (R), blue light (B), green light (G), yellow light (Y), red plus blue light (RB) and dysprosium lamp (CK). Lighting experiments were performed under controlled conditions (photon flux density 150 μmol m?2 s?1; 12 h photoperiod; 18–20 °C). The fresh and dry mass were the greatest under RB, which were significantly higher than other light treatments. The fresh mass under RB was almost twice higher compared to other light treatments. Plant height was highest under R treatment and was lowest under B. RB treatment also lowered the plant height significantly. The highest soluble sugar concentration was observed under B. The soluble protein concentration was the greatest under RB. The R treatment was adverse to pigment accumulation. The concentration of photosynthetic pigments and chlorophyll biosynthesis precursors were higher under RB. The RB treatment was beneficial to pigment accumulation.  相似文献

An isolate ofMonascus, from a commercial, fermented soybean curd (sufu) was grown on a cassava medium. With medium at an initial pH of 7.0 an orange-red pigmentation was produced but with an initial pH below 4, a light golden pigment was obtained. A medium containing, w/v, 3% cassava starch, 0.4% peptone and 0.1% glutamic acid, with an initial pH of 2.5 was optimal for the production of this yellow pigment, which had a single maximum absorption spectrum at 330 nm. The genus Monascus, comprising nine species, can reproduce either vegetatively with filaments and conidia or sexually by the formation of ascospores. The most well-known species of genus Monascus, namely, M. purpureus, M. ruber and M. pilosus, are often used for rice fermentation to produce red yeast rice, a special product used either for food coloring or as a food supplement with positive effects on human health. The colored appearance (red, orange or yellow) of Monascus-fermented substrates is produced by a mixture of oligoketide pigments that are synthesized by a combination of polyketide and fatty acid synthases. The major pigments consist of pairs of yellow (ankaflavin and monascin), orange (rubropunctatin and monascorubrin) and red (rubropunctamine and monascorubramine) compounds; however, more than 20 other colored products have recently been isolated from fermented rice or culture media. In addition to pigments, a group of monacolin substances and the mycotoxin citrinin can be produced by Monascus. Various non-specific biological activities (antimicrobial, antitumor, immunomodulative and others) of these pigmented compounds are, at least partly, ascribed to their reaction with amino group-containing compounds, i.e. amino acids, proteins or nucleic acids. Monacolins, in the form of β-hydroxy acids, inhibit hydroxymethylglutaryl-coenzyme A reductase, a key enzyme in cholesterol biosynthesis in animals and humans.  相似文献

Monascus purpureus is a biopigment-producing fungi whose pigments can be used in many biotechnological and food industries. The growth kinetics of biopigment production were investigated in a liquid fermentation medium in a 5-l stirred tank bioreactor at 30°C, pH 7, for 8 days with 100 rpm agitation and 1.38 × 105 N/m2 aeration. Thai Monascus purpureus strains TISTR 3002, 3180, 3090 and 3385 were studied for color production, growth kinetics and productivity. Citrinin as a toxic metabolite was measured from the Monascus fermentation broth. The biopigment productions were detected from fermentation broth by scanning spectra of each strain produced. Results showed a mixture of yellow, orange and red pigments with absorption peaks of pigments occurring at different wavelengths for the four strains. It was found that for each pigment color, the color production from the strains increased in the order TISTR 3002, 3180, 3090, 3385 with 3385 production being approximately 10 times that of 3002. Similar results were found for growth kinetics and productivity. HPLC results showed that citrinin was not produced under the culture conditions of this study. The L*, a* and b* values of the CIELAB color system were also obtained for the yellow, orange and red pigments produced from the TISTR 3002, 3180, 3090 and 3385 strains. The colors of the pigments ranged from burnt umber to deep red.  相似文献

The study was conducted on six sites that are dominated by Japanese knotweed (Reynoutria japonica) and that vary in the level of industrialization and habitat transformation by humans. The aim of the research was to investigate the chemical-physical features of soil under a closed and dense canopy of R. japonica, the chemical composition of the R. japonica leaves, and to compare the content of certain elements in the soil-plant-soil system. The soil organic carbon (Corg) content varied from 1.38±0.004% to 8.2±0.047% and the maximum in leaves was 49.11±0.090%. The lowest levels of total nitrogen (Ntot) in soil were recorded on the heavily disturbed sites (till 0.227±0.021%). Soil pH varied greatly, ranging from acidic (pH=4.0) to neutral (pH=7.7). Heavy metal content differed significantly among the study sites. At all of the sites, both in the case of soil and plant leaves, Zn was a dominant element and its concentration ranged from 41.5 to 501.2 mg·kg?1 in soils and from 38.6 to 541.7 mg·kg?1 in leaves. Maximum accumulations of P (2103.3±15.3 mg·kg?1) and S (2571.7±17.6 mg·kg?1) were observed on the site that had been influenced by agricultural practices. The results obtained showed that R. japonica is able to accumulate high levels of heavy metals.  相似文献

Membrane-filtered cabbage juice, when fermented by Lactobacillus brevis under conditions of controlled pH, frequently produced a water-soluble red pigment. The pigment, presumably responsible for imparting a highly objectionable discoloration to sauerkraut, was formed during the post logarithmic phase of growth. Color development is pH dependent (5.2 to 6.3) and can be suppressed by chemical reductants or anaerobic conditions of growth. The compound responsible for discoloration was purified and partially characterized.  相似文献

Monascus pigments are secondary metabolites of Monascus species and are mainly composed of yellow pigments, orange pigments and red pigments. In this study, a larger proportion of Monascus yellow pigments could be obtained through the selection of the carbon source. Hydrophilic yellow pigments can be largely produced extracellularly by Monascus ruber CGMCC 10910 under conditions of high glucose fermentation with low oxidoreduction potential (ORP). However, keeping high glucose levels later in the culture causes translation or a reduction of yellow pigment. We presume that the mechanism behind this phenomenon may be attributed to the redox level of the culture broth and the high glucose stress reaction of M. ruber CGMCC 10910 during high glucose fermentation. These yellow pigments were produced via high glucose bio-fermentation without citrinin. Therefore, these pigments can act as natural pigments for applications as food additives.

In order to illustrate the physiological variation of different generations and different thallus parts of Saccharina japonica, physiological parameters such as maximum and effective PSII photochemical efficiency, nutrient uptake, and elemental composition were determined in the laboratory. Photosynthetic analysis in different generations indicated that, although gametophytes had higher pigment contents than the sporophyte, they had lower values of F v/F m and ΔF/Fm. The highest Chl a/Chl c ratio was found in sporophyte generation (3.98?±?0.01) and in the basal part of fresh thallus (2.66?±?0.02). The sporophyte had significantly higher values of nitrate uptake but lower values of phosphorus uptake than the gametophytes. The contents of nitrogen and carbon as well as C/N in gametophytes were significantly higher than those in sporophytes. In addition, the basal part of the S. japonica thallus had the highest C content (22.31?±?1.50 %) but the lowest N content (2.02?±?0.16 %), as well as the highest value of C/N (11.02?±?0.34).  相似文献

Betalains, comprising violet betacyanins and yellow betaxanthins, are pigments found in plants belonging to the order Caryophyllales. In this study, we induced the accumulation of betalains in ornamental lisianthus (Eustoma grandiflorum) by genetic engineering. Three betalain biosynthetic genes encoding CYP76AD1, dihydroxyphenylalanine (DOPA) 4,5-dioxygenase (DOD), and cyclo-DOPA 5-O-glucosyltransferase (5GT) were expressed under the control of the cauliflower mosaic virus (CaMV) 35S promoter in lisianthus, in which anthocyanin pigments are responsible for the pink flower color. During the selection process on hygromycin-containing media, some shoots with red leaves were obtained. However, most red-colored shoots were suppressed root induction and incapable of further growth. Only clone #1 successfully acclimatized and bloomed, producing pinkish-red flowers, with a slightly greater intensity of red color than that in wild-type flowers. T1 plants derived from clone #1 segregated into five typical flower color phenotypes: wine red, bright pink, pale pink, pale yellow, and salmon pink. Among these, line #1-1 showed high expression levels of all three transgenes and exhibited a novel wine-red flower color. In the flower petals of line #1-1, abundant betacyanins and low-level betaxanthins were coexistent with anthocyanins. In other lines, differences in the relative accumulation of betalain and anthocyanin pigments resulted in flower color variations, as described above. Thus, this study is the first to successfully produce novel flower color varieties in ornamental plants by controlling betalain accumulation through genetic engineering.  相似文献
