Tissue zinc dynamics during the immune reaction in mice   总被引:1,自引:0,他引:1  
Owing to the importance of zinc for the functioning of the immune system, the role of endogenous Zn, located both in lymphoid and nonlymphoid organs, was investigated during the standard humoral and cellular types of immune response. For this purpose, the dynamics of hepatic, thymic, splenic, and renal Zn content was determined in mice sensitized with (a) sheep red blood cells and (b) semiallogeneic lymphocytes during the local host vs graft reaction (HVGR). The data obtained by ion-coupled plasma spectrometry revealed that the humoral type of immunity is characterized by a significant increase of Zn concentration in the liver and in the thymus. Simultaneously, linear regression analysis showed that the generation of plaque-forming cells in the individual mouse was highly positively correlated with Zn concentration in the liver (r = 0.897), and spleen (r = 0.833), and negatively with Zn concentration in the thymus (r = -0.624). Similar relationships between the intensity of local immune reaction and tissue Zn levels were found in local HVGR at the fifth day in the liver and spleen (r = 0.861 andr = 0.695, respectively), at the seventh day in the thymus (r = -0.797), and at the tenth day in the liver (r = -0.859). The data emphasize the necessity of Zn for the development of normal immune response and point to the existence of a Zndependent hepato-thymic axis during the humoral and cellular types of immune reactivity.  相似文献

An electrothermal atomic absorption spectrometric procedure for zinc determination in animal tissues is first optimized and then used to measure the metal content of different tissues from normal and dystrophic mice. The procedure involves minimal manipulation of the sample to overcome the severe contamination problems normally associated with the measurement of low zinc levels. The samples (recommended amount 10 mg) are slurried in 2 ml of a 10-mM tetramethylammonium hydroxide solution containing 0.1 g/100 ml silicone antifoam. After mild heating at 60 degrees C for 10 min and homogenization for 1 min, the suspensions are submitted to a 10-fold dilution and then injected into the electrothermal atomizer. Calibration is carried out using aqueous standard solutions of zinc prepared in the same suspension media. The reliability of the whole procedure is verified using three certified reference materials.  相似文献

Patients with chronic inflammation often show decreased serum levels of trace elements. This study aimed to investigate serum levels of selenium and zinc in patients with uveitis. Twenty-four patients (13 female, 11 male) with a mean age of 40.6±15.5 yr were included in this study. According to their underlying disease, they were divided into acute (n=13) or chronic (n=11) uveitis. Selenium and zinc determination was performed by atomic absorption spectroscopy in EDTA blood samples. Patients with acute or first-time uveitis showed selenium and zinc concentrations within the normal range. In contrast to this, patients with chronic recurrent uveitis tend to result in decreased selenium and zinc levels. Especially selenium showed a remarkable reduction in serum concentration below the normal range. Furthermore, there was a tendency to decreased trace element concentrations with increasing age. A sex dependency could not been found. Especially patients with chronic, recurring uveitis show remarkable decreased selenium concentration in the EDTA-blood. Further studies should investigate possible positive effects of therapeutic selenium and zinc supplementation in patients with chronic, recurrent uveitis.  相似文献

Extraordinarily high concentrations of zinc (300–500 μg/(g fresh tissue)) are often found in the digestive tract tissue of common carp Cyprinus carpio, and high zinc concentrations (typically >100 μg/(g fresh tissue)) are also found in the kidney, gill, skeletal tissues, and spleen. In the present study, we found that only about 40% of the zinc in the digestive tract tissue of common carp could be extracted by water. However, 0.01 M citrate buffer, pH 6.2 could extract over 90% of the zinc. Subcellular zinc distribution in the tissues of common carp, grass carp Ctenopharyngodon idellus, silver carp Aristichthys nobilis, and tilapia Oreochromis aureus were compared. It was found that zinc concentrations in the cytosol, microsomal and mitochondrial fractions were approximately the same for all four species, being only about 16, 5, and 4 μg/(g fresh tissue), respectively. However, zinc concentrations in the nuclei/cell debris fraction of common carp tissue were much higher (46–370 μg/(g fresh tissue)) than the <14 μg/(g fresh tissue) found in the other three species. From this we conclude that neither water-soluble zinc proteins nor metallothionein could account for the high levels of zinc found in common carp tissues. A preliminary biochemical investigation suggests that the main zinc binding substance(s) in the nuclei/cell debris fraction of digestive tract tissue of common carp was probably a membrane protein(s).  相似文献

In this paper we report a method for measuring ultrafiltrable zinc in human serum by electrothermal atomic absorption spectrophtometry. We show also that ultrafiltration permits to determine alpha-2 macroglobulin bound zinc and losely bound zinc if a strong zinc ligand (EDTA) is added to serum before ultrafiltration. This last fraction, after deduction of ultrafiltrable zinc, represents roughly all albumin bound zinc. In 20 controls we found that ultrafiltrable zinc amounted 0.311 μmol/L (S.D.=0.117 μmol/L), alpha-2 macroglobulin bound zinc 3.08 μmol/L (S.D.=0.221 μmol/L), and albumin bound zinc 12.11 μmol/L (S.D.=1.95 μmol/L). Our method needs only a small volume of serum, it is simple and rapid but also very accurate and reliable. The losely bound fraction is very dynamic and, representing the physiologically active part of serum zinc, it could be a good marker of zinc deficiency.  相似文献

The serum zinc (Zn) concentrations of 80 healthy subjects (48 male, 32 female) from southeastern Spain were determined by atomic absorption spectrometry. The samples were digested by heating in a 4:1 mixture of nitric and perchloric acids. The concentration of Zn was determined against a Contox Trace Metal Serum Control Panel A standard reference. Zn concentrations in the standard were found to be 2.332 ±0.489 mg/L, with a mean recovery of 102.7%. In the serum samples, the relative standard deviation was <6% for the range of concentrations determined: 0.420-1.540 mg/L for women (mean value 0.947 ±0.265 mg/L) and 0.490-1.480 mg/L for men (mean value 0.951 ±0.243 mg/L). In healthy subjects, no statistically significant differences were observed in the Zn levels with respect to their sex (p > 0.05) or the location where they lived (mountainous vs coastal zones). It is concluded that the dietary Zn intake and Zn status for healthy adults in this region of Spain are within normal values.  相似文献

A new enzymatic method has been developed to determine trace amounts of Zn2+ in vegetables. The basis of the method is that apocarbonic anhydrase regains its activity in proportion to the concentration of Zn2+ present in solution. Bovine carbonic anhydrase was purified from erythrocyte haemolysate by affinity chromatography and the bound Zn2+ removed by dialysis of purified enzyme against a solution of pyridine-2, 6-dicarboxylic acid. Pure (100%) apoenzyme was obtained. The concentration of Zn2+ in vegetable samples was determined using the enzymatic method and by atomic absorption spectroscopy. Determinations made using the two methods were not significantly different one from another.  相似文献

The purpose of the study was to examine the zinc and iron content of human lenses in different types of cataract and to investigate the possible influence of diabetes on the zinc and iron content of the lens. Iron and zinc of 57 human lenses (28 corticonuclear cataracts and 29 mature cataracts with a mean age of 70.6±16.1 and 74.7±11.1 yr, 41 nondiabetics and 16 diabetics) were determined by atomic absorption spectroscopy. The zinc content of human lenses was significantly increased in mature cataracts compared to corticonuclear cataracts (0.51±0.33 vs 0.32±0.20 μmol/g dry mass, p=0.012). The iron content of mature cataracts was also higher than in corticonuclear cataracts (0.11±0.09 vs 0.07±0.05 μmol/g dry mass, p=0.071). Furthermore, a significant increase of the lens zinc content could be observed with increasing lens coloration (light brown 0.33±0.17 vs dark brown 0.52±0.35 μmol/g dry mass, p=0.032). Diabetic patients seem to have both increased zinc and iron contents in the lens compared to nondiabetic subjects (zinc: 0.45±0.42 vs 0.40±0.22 μmol/g dry mass; iron: 0.12±0.10 vs 0.08±0.05 μmol/g dry mass). These data suggest a possible influence of the lens zinc and iron content on the development of lens opacification. Especially advanced forms of cataract and dark brown colored lenses show significantly increased zinc and iron content.  相似文献

A simple and reliable method is described which combines ultrafiltration technique with atomic absorption spectrophotometry to determine the Zn fractions in human blood plasma and seminal plasma. Ultrafiltrable, loosely bound, and firmly bound Zn can be measured using this method in the presence or absence of ethylene-diaminetetraacetic acid (EDTA). The YMT membranes for the ultrafiltration must be rinsed thoroughly before use. In contrast to Zn in blood plasma, a large part of Zn in the seminal plasma was found to be ultrafiltrabe. This method can be applied to study the physiologically active part of Zn in body fluids related to various disease states.  相似文献

采用微波消解法处理旱芹根、茎、叶,并用火焰原子吸收法测定其中的Na、K、Ca、Mg、Fe、Mn、Zn、Cu 8种金属元素的含量。结果表明:旱芹中富含人体必需的Na、K、Mg、Fe、Ca等元素,各元素在不同部位含量有一定差异。Fe元素在旱芹根中含量为883.57μg.g-1,明显高于茎和叶;Ca、Zn和Mn元素在旱芹叶中的含量分别为11 103.74,214.04,88.07μg.g-1,明显高于茎和根;K、Na和Mg元素在旱芹茎中的含量高于根和叶中,Cu元素含量在各部位差异不大。方法的加标回收率为96.8%~105.8%,相对标准偏差(RSD)≤3.36%。  相似文献

To better elucidate previous results showing that partial hepatectomy noticeably changes the tissue content of zinc, calcium, magnesium, and iron(II) ions in regenerating the liver, thymus, and spleen, we report on the correlation of these metal tissue kinetics in these organs with the expression of metallothionein-I+II (MT-I+II) proteins and MT-I mRNA in early postoperative period (1, 2, 6, 12, and 24 h) after one-third hepatectomy (pHx). The results showed that 2 h after pHx the regenerating liver accumulated Zn2+, Ca2+, Mg2+, and Fe2+ ions while decreasing the concentration of all these metals in the spleen and of Zn2+ in the thymus. On the 24th h, a new high accumulation of Zn2+ and Ca2+ was seen in the regenerating liver and of Zn2+, Ca2+, and Fe2+ in the spleen. Simultaneously, MT-I mRNA increased in the liver and spleen. In hepatocytes and on several spleen and thymus mononuclear lymphatic cells, the increased expression of MT proteins was found mainly in the cytoplasm and nuclei. The areas expressing MTs in regenerating liver inversely correlated with those containing apoptotic cells, suggesting that these proteins participate in tissue restoration through reduction or increase of metal ions after injury to the liver.  相似文献

Abstract

The speciation and bioavailability of zinc (Zn) in smelter-contaminated sediments were investigated as a function of phosphate (apatite) and organic amendment loading rate. Zinc species identified in preamendment sediment were zinc hydroxide-like phases, sphalerite, and zinc sorbed to an iron oxide via X-ray adsorption near edge structure (XANES) spectroscopy. Four months after adding the amendments to the contaminated sediment, hopeite, a Zn phosphate mineral, was identified indicating phosphate was binding and sequestering available Zn and Zn pore water concentrations were decreased at levels of 90% or more. Laboratory experiments indicate organic amendments exhibit a limited effect and may hinder sequestration of pore water Zn when mixed with apatite. The acute toxicity of the sediment Zn was evaluated with Hyalella azteca, and bioaccumulation of Zn with Lumbriculus variegates. The survivability of H. azteca increased as a function of phosphate (apatite) loading rate. In contaminated sediment without apatite, no specimens of H. azteca survived. The bioaccumulation of Zn in L. variegates also followed a trend of decreased bioaccumulation with increased phosphate loading in the contaminated sediment. The research supports an association between Zn speciation and bioavailability.  相似文献

Rat bile and pancreatic fluid were examined for the presence of low molecular weight zinc complexes. Fluids were collected separately by cannulation, and zinc distribution in collected samples was analyzed by gel filtration on Sephadex G-50. Most of the zinc in bile was associated with low molecular weight zinc complexes; only a small amount of zinc was present in the high molecular weight fraction. In contrast, pancreatic secretions did not contain low molecular weight zinc complexes, but there were considerable amounts of zinc bound to high molecular weight compounds. The addition of zinc to bile resulted in an increased amount of zinc in the low molecular weight fraction, while the addition of zinc to pancreatic fluid resulted primarily in an increase in zinc bound to the high molecular weight components. Like pancreatic fluid, homogenates of pancreatic tissue had no low molecular weight zinc complex. In rats whose bile and pancreatic fluid were removed and not returned into the intestine, the amount of zinc bound to low molecular weight complexes in intestinal homogenates was reduced. This alteration of the molecular distribution of zinc in intestinal homogenates by removal of bile and pancreatic fluid suggests the potential importance of low molecular weight zinc complexes for zinc homeostasis.  相似文献

The aim of our investigation was to determine the concentrations of Cu, Zn, and Cr in urine samples under routine clinical laboratory conditions. To asses the reliability of these methods, critical factors such as detection limit(s), calibration range(s), cost, accuracy, and precision were studied. Our method was employed for the quantitative determination of zinc, chromium, and copper in urine samples from steel production and quality control (QC) workers and healthy unexposed controls. After pretreatment with acids, the samples were digested via a microwave oven. Zinc was determined by flame absorption spectrophotometry (FAAS), whereas chromium and copper were determined by a graphite-furnace atomic absorption spectrophotometry (GFAAS). Our results indicate that urinary zinc, chromium, and copper levels of the exposed workers are significantly higher than those of the controls. The possibility that these metals are involved in the etiology of diseases is discussed and recommendations are made to improve workplace ventilation and industrial hygiene practices.  相似文献

Serum arsenic concentrations of persons suffering from renal failure and undergoing hemodialysis treatment (n=85) and of healthy controls (n=25) were determined by hydride-generation AAS technique after microwave digestion. The results were evaluated by comparing the values of both groups, considering physiological factors and individual data, as well as comorbid conditions of the hemodialysis (HD) patients. Serum arsenic levels were diminished in the patient group compared with controls (mean values 8.5±1.8 ng/mL vs 10.6±1.3 ng/mL). Furthermore, additional diseases within the hemodialysis group, particularly injuries of the central nervous system (CNS), vascular diseases, and cancer, were correlated to occasionally markedly decreased serum arsenic concentrations. It was concluded that arsenic homeostasis is disturbed by HD treatment and certain additional diseases. Desirable arsenic concentrations in the body seem to be reasonable. This consideration results in the conclusion that arsenic could play an essential role in human health. Thus, reference arsenic concentrations in different human tissues and body fluids should be established in order to recognize not only arsenic intoxication, but also arsenic deficiency. Perhaps arsenic deficiency contributes to the increased death risk of HD patients, and therefore, arsenic supplementations for patients with extremely low serum arsenic concentrations should be taken into account.  相似文献

Two groups of 16 rats each were fed the same diet with 12.9 ppm Zn. Nine days after each animal was injected with65Zn for assessing fecal zinc of endogenous origin, zinc intake and excretion were determined for a six-day period at the age of about five (group I) and nine (II) weeks. At mean growth rates of 5.1 and 5.2 g/day, food consumption per gram of gain was 2.01 g in group I vs 2.86 g in II. Overall, zinc retention amounted to 21 vs 25 μg Zn/g of gain. Apparent absorption averaged 92 vs 74% of Zn intake (132 vs 189 μg/day), while true absorption averaged 98 vs 92%. It was concluded that endogenous fecal zinc excretion was limited to the indispensable loss (F em) in group I (7 μg/day), while it exceeded this minimum loss in group II (33 μg/day). True retention, which reflected total zinc utilization (true absorption times metabolic efficiency), was derived from apparent absorption plusF em (11 μg/day for group II according to the greater metabolic body size of the rats). It averaged 98% of Zn intake in group I vs 80% in group II. The mean metabolic efficiency was 100% vs 87%. The conclusion was that these marked differences between age groups in utilizing the dietary zinc reflected the efficient homeostatic adjustments in absorption and endogenous excretion of zinc to the respective zinc supply status.  相似文献

Fifteen kinds of common plants, animals, and minerals used as traditional medicines by the Chinese people have been subjected to analysis by atomic absorption spectrometry for its content of seven metals: lead, cadmium, arsenic, mercury, copper, cobalt, and manganese. The concentrations of these elements are significantly different according to their vegetal, animal, or mineral origin. The average values found for lead, cadmium, arsenic, cobalt, and manganese in drugs of mineral origin are higher than those derived from plants and animals, except for copper, which was higher in drugs of animal origin. Our results suggest that the user of traditional Chinese crude drugs should be warned of the potential danger of heavy-metal poisoning because their concentrations seem to be higher than the maximum values allowed by health agencies in several countries.  相似文献

For the investigation of neurological disorders, a development of simple and accessible methods for determining selenium in human brain samples is required. We devised a method of determining selenium using graphite furnace atomic absorption spectrometry (GFAAS). An electrodeless discharge lamp provided the sufficient sensitivity to determine brain selenium. The matrix interferences were avoided by using high temperature, a prolonged pyrolysis step, and a palladium matrix modifier. The technique of standard addition was used to evaluate the sample concentrations. The accuracy of the method was confirmed by a bovine liver reference material. The detection limit of selenium was 0.04 ng. The determined selenium concentrations of human brain cortex and white matter were higher than those of putamen (115–155 and 206–222 ng/g wet wt, respectively). These GFAAS values agreed with those obtained by fluorometric analysis (r=0.91,n=10). Moreover, the GFAAS values were compatible to those reported by other researchers (99–274 ng/g wet wt), in which selenium concentrations in putamen also tended to be higher than the other two regions. We conclude that GFAAS is useful for selenium analysis in brain samples.  相似文献

To investigate the Zn status of inhabitants of the Czech Republic, 1155 serum and 132 hair samples were analyzed for zinc content. Analyzed material was obtained from randomly selected volunteers of both sexes in the age range 6–65 yr. Subpopulations from five regions were included in the study. Analyses of both materials were performed by Instrumental Neutron Activation Analysis (INAA). Coanalyses of Standard Reference Materials (SRM) for quality control were performed. The results (mean 910 ±276 Μg Zn/L serum and 189 ±45 Μg Zn/g hair) demonstrate satisfactory zinc status of the searched population. Significant interregional differences and age-dependent differences, as well as sex-dependent differences have been detected by the use of correlation analysis. On the basis of these results, serum Zn concentration results of individual categories according to age and sex were evaluated, and on the grounds of mean ±2 standard deviations, indicative intervals were calculated. The frequency of individual serum Zn concentrations proved that the population of the Czech Republic does not suffer from severe zinc deficiency. On the other hand, about one-third of our inhabitants has their serum Zn concentrations below the cutoff value of 800 Μg Zn/L serum, which means a marginal or mild Zn deficit of the organism.  相似文献
