Background aimsThis study was initiated to determine whether CD34+ cell selection of small-volume bone marrow (BM) samples could be performed effectively on the Isolex® 300i Magnetic Cell Selection System® device and whether the results obtained from these samples were comparable with results from large standard-volume samples. The impact on CD34+ recovery using a full versus half vial of Isolex® CD34 reagent and the effects of shipping a post-selection product were evaluated.MethodsA protocol to evaluate CD34+ cell selection with two ranges of smaller volume BM samples (c. 50hmL and c. 100 mL) was developed and instituted at three Production Assistance for Cellular Therapies (PACT) facilities. The study was performed in two phases.ResultsIn phase I, the mean post-selection CD34+ recoveries from the two sizes of samples were 104.1% and 103.3% (smallest and largest volumes, respectively), and mean CD34+ recoveries were 115.6% and 88.7%, with full and half vials of reagent, respectively. Mean CD34+ recoveries for post-shipment smaller volume samples were 106.8% and for larger volume samples 116.4%; mean CD34+ recoveries were 99.9% and 127.4% for post-shipment samples processed with full and half vials of reagent, respectively. In phase II, mean CD34+ recovery was 76.8% for post-selection samples and 74.0% for post-shipment samples.ConclusionsThe results suggest that smaller volume BM sample processing on the Isolex® system is as efficient or more efficient compared with standard-volume sample processing. Post-processing mean CD34+ recovery results obtained using a full or half vial of CD34 reagent were not significantly different.  相似文献

The activity concentrations of radionuclides in grape molasses soil samples collected from Zile (Tokat) plain in the Central Black Sea region of Turkey were measured by using gamma spectrometer with a NaI(Tl) detector. Also, the concentrations of 222Rn in soil samples and air were estimated essentially taking the activity concentrations of 226Ra measured in soil samples. Grape molasses soil samples with calcium carbonate content are used for sedimentation for making molasses in this region. The average activity concentrations of 232Th, 226Ra, 40K, and 137Cs were found as 62 ± 2, 68 ± 3, 479 ± 35, and 8.0 ± 0.3 Bq kg?1, respectively. The average concentrations of 222Rn in soil samples and air were estimated to be 50 kBq m?3 and 144 Bq m?3. From the activity concentrations, absorbed gamma dose rate in outdoor air (D), annual effective dose from external exposure (EE), annual effective dose from inhalation of radon (EI), and excess lifetime cancer risk (ELCR) were estimated in order to assess radiological risks. The average values of D, EE, EI, and ELCR were found to be 90 nGy h?1, 110 μSv y?1, 1360 μSv y?1, and 4 × 10?4, respectively.  相似文献

Lead has been determined in top and subsoil samples from over 500 sites in West Pembrokeshire. Topsoils contain from <1 to 356 μg g−1 Pb with a mean of 39 μ g−1 Pb while subsoils contain 0 to 172 μg g−1 Pb with a mean value of 22 μg g−1 Pb.Lead has also been determined in herbage samples from north west Pembrokeshire and in these, the Pb contents range from <1 μg g−1 to 9 μg g−1 but most samples contain <2 μg g−1. Maximum contents of Pb were found in March when samples were taken at the same site throughout the growing season.Soil samples showed maximum Pb contents around towns but this pattern was not duplicated by herbage.  相似文献

One hundred samples of 10 poultry meat products were collected from AL-Ahsa markets (Kingdom of Saudi Arabia). The samples were ranked from carcass cuts (chilled, frozen, fillet and thigh) to minced meat or further processed products as burger, nuggets, frankfurter and meat paste loaf. Samples were collected in triplicate for sensory, chemical and microbiological analysis to assure their quality and safety.The obtained results revealed variation in chemical composition; some products with high fat percentage had a high thiobarbituric acid value, which resulted in the appearance of an unacceptable flavor.Bacteriological analysis revealed that the mean total bacterial count was ranged from 2.7 × 104 cfu/g for nuggetsA to 3.3 × 107 cfu/g for burgerB and the other products in the range of 105–106 cfu/g. While Staphylococcus aureus mean count ranged from less than 102 cfu/g for all samples, accept 104 and 106 cfu/g for minceB and frankfurter samples, respectively. Escherichia coli isolated from 70% of the samples and Salmonella arizona was isolated at once from thigh samples. Thirty percentages of samples not comply with Saudi Standards due to sensory unacceptability and 21% of samples nonconforming with bacteriological specifications.  相似文献

Abstract

Ground-water samples were collected from an uncontaminated and a contaminated site. Copper complexation was characterized by ion- selective electrode (ISE), fluorescence quenching (FQ), and cathodic stripping voltammetric (CSV) titrations. All of the samples were titrated at their natural pH values and some of the samples were also titrated at other pH values. For a total Cu concentration of 10?6 M, the free Cu2+ concentrations in the samples from the uncontaminated site were all less than 10?7 M, while free Cu2+ in the samples from the contaminated site were all less than 10?8 M. For a particular sample and total Cu concentration, the free Cu2+ concentration decreased as the pH increased. Relative to ISE, FQ underestimated and CSV overestimated the degree of Cu2+ binding. The Cu2+ -complexing properties of the ground waters are similar to many published results for the same pH and for ligand concentrations normalized to T.O.C. Chemical equilibrium computations indicate that organic complexes would dominate Cu speciation in the uncontaminated ground waters for 10?7 to 10?5 M total Cu. In the contaminated ground waters, sulfide complexes would be the predominant Cu species for total Cu less than the total S(?11) concentration. Organic complexes would dominate Cu speciation for total Cu greater than total S(?11).  相似文献

This study presents the first protein electrophoretic study of population structure within the Georgia Basin Pacific hake Distinct Population Segment, as defined under the U.S. Endangered Species Act. Forty-one allozyme loci (29 polymorphic) were analyzed in samples from three Pacific hake spawning populations on the west coast of North America: (1) Port Susan, Puget Sound, Washington (three temporal samples); (2) south-central Strait of Georgia, British Columbia, Canada (two temporal samples); and (3) offshore of southern California (two temporal samples) (total n = 664). Mean heterozygosity over all loci was 12–13% for all populations. Within-population temporal samples were not significantly different from one another, but statistically significant differences were detected at 15 of the 29 polymorphic loci (p < 0.05) among the three populations. Differences at eight of these loci were highly significant (p < 0.001): ADA *, ALAT *, bGALA *, GPI-A *, sIDHP *, LDH-A *, MPI *, and PEP-B *. The two Georgia Basin populations were significantly different at six loci: bGALA *, sIDHP *, LDH-A *, MPI *, PGK *, and PGM-2 * (p < 0.05). Nei's genetic distance (D) was 0.0006 between Port Susan and Strait of Georgia pooled temporal samples, and 0.005 between these populations and offshore Pacific hake. FST was 0.02 and 0.0046 among all three populations and among the Georgia Basin populations, respectively. Both FST estimates were significantly greater than zero, and the results suggest a high degree of demographic isolation among all three populations.  相似文献

Samples of bulk precipitation were collected in the Trachypogon savanna, Calabozo, Venezuela, during three consecutive years. In the first year, rain samples were taken daily; in the following years the samples were grouped on a monthly basis. In addition, samples of dry deposition were collected during the dry seasons. All samples were analyzed for the following water soluble cations and anions: P04-P, S04-S, N03-N, NH4-N, Ca+2, Mg+2, K+, Na+ and H+. The mean annual input rate of chemical constituents (Kg ha-1 year-1) was: PO4-P (0.42); SO4-S (2.62); NO3-N (0.21); NH4-N (2.03); Ca+2 (3.50); Mg+2 (11.31); K+ (3.60); Na+ (5.93) and H+ (0.03). The total mean input of particulate material to the savanna during the dry season was 2.06 Kg ha-1 year-1, with a soluble fraction of 30%. Possible sources of nutrients input were analyzed.  相似文献

Background

Circulating endothelial cells (CECs) are markers of vascular damage that have clinical relevance in many diseases, including acute myocardial infarction (AMI), and may be predictors of treatment responses. Herein, we investigated the diagnostic and prognostic value of CEC monitoring in AMI patients and a murine model.

Methodology/Principal Findings

CECs were defined as Hoechst 33342+/CD45−/CD31+/CD146+/CD133 in human blood samples and Hoechst 33342+/CD45−/CD31+/KDR+/CD117 in murine samples. To evaluate the validity and variability of our CEC detection system, peripheral blood samples of vascular endothelial growth factor-treated athymic nude mice and AMI patients were collected and subjected to intra-assay analysis. CEC detection by flow cytometry and real-time PCR were compared. Blood samples were obtained from 61 AMI patients, 45 healthy volunteers and 19 samples of the original AMI patients accepted one month treatment, via flow cytometry and expressed as a percentage of peripheral blood mononuclear cells.

Results

Our CEC detection method was validated and had limited variability. CEC concentrations were higher in AMI patients compared to healthy controls. One month post-treatment, CECs levels decreased significantly.

Conclusions/Significance

CEC levels may be useful as a diagnostic and prognostic biomarker in AMI patients.  相似文献

We examined allozymic variation in 65 protein-coding loci in three samples of sockeye salmon (Oncorhynchus nerka) from Hokkaide, and Honshu, Japan, Over-all, six variable loci were seen and each of the three samples was variable at 3–5 loci. Two loci,mAH-1,2 * andALAT *, were variable at the P0,95 level. Average heterozygosity ranged from 0.012 to 0.013, representing some of the lowest recorded values for the species. Although frequencies ofALAT * alleles differed significantly among the three samples, the overallχ 2 for six polymorphic loci was not statistically significant. New data for four Russian samples at 45–64 loci were also obtained. In comparison to the Japanese samples, three samples from the Kamchatka Peninsula had two to three times the level of variation; and a sample from Iturup Island (Kuril Island archipelago) was slightly more variable. Although the anadromous sockeye salmon were originally planted from Iturup Island to Lake Shikotsu, a close genetic affinity was not indicated. These seven samples ofO. nerka were compared with representative samples previously studied in North America using five polymorphic loci. Two large groups of samples were indicated in multilocus analyses: 1) a cluster of the seven Asian samples, one Alaskan sample, and one northern British Columbia sample; and 2) a group that included a southern British Columbia sample (Fraser River), and samples from the Columbia River and Washington. 1H NMR relaxometry is applied for the investigation of pore size distributions in geological substrates. The transfer to humous soil samples requires the knowledge of the interplay between soil organic matter, microorganisms and proton relaxation. The goal of this contribution is to give first insights in microbial effects in the 1H NMR relaxation time distribution in the course of hydration of humous soil samples. We observed the development of the transverse relaxation time distribution of the water protons after addition of water to air dried soil samples. Selected samples were treated with cellobiose to enhance microbial activity. Besides the relaxation time distribution, the respiratory activity and the total cell counts were determined as function of hydration time. Microbial respiratory activities were 2–15 times higher in the treated samples and total cell counts increased in all samples from 1×109 to 5×109 cells g−1 during hydration. The results of 1H NMR relaxometry showed tri-, bi- and mono-modal relaxation time distributions and shifts of peak relaxation times towards lower relaxation times of all investigated soil samples during hydration. Furthermore, we found lower relaxation times and merging of peaks in soil samples with higher microbial activity. Dissolution and hydration of cellobiose had no detectable effect on the relaxation time distributions during hydration. We attribute the observed shifts in relaxation time distributions to changes in pore size distribution and changes in spin relaxation mechanisms due to dissolution of organic and inorganic substances (e.g. Fe3+, Mn2+), swelling of soil organic matter (SOM), production and release of extracellular polymeric substances (EPS) and bacterial association within biofilms.  相似文献

With the perspective of embarking on a human study using a double iron (Fe) stable isotope tracer protocol to assess iron bioavailability, investigations were conducted on Fe isotope ratios in blood samples using a VG Axiom Multi-collector ICP-MS. The factors affecting the precision and accuracy of Fe isotopic ratios, such as spectral- and matrix-induced interferences and Fe recoveries from sample preparation, have been identified and optimized. Major polyatomic interferences (e.g., Ar-O, Ar-OH, and FeH) were significantly reduced by using an Aridus nebulizer and desolvating system. Isobaric metal (e.g., 54Cr+ on 54Fe+ and 58Ni+ on 58Fe+) interferences and Ca-oxides and hydroxides were quantitatively removed during chemical purification of blood samples and selective isolation of Fe by anion-exchange resin, after mineralization of the blood samples by microwave digestion. Quantitative recoveries of Fe from different steps of sample preparation were verified using whole blood reference material. Fe isotopic compositions of the samples were corrected for instrumental mass bias by the standard-sample bracketing method using the certified reference standard IRMM-014. External precisions on the order of 0.008–0.05 (% RSD), 0.007–0.015 (% RSD), and 0.03–0.09 (% RSD) were obtained for 54Fe/56Fe, 57Fe/56Fe, and 58Fe/56Fe, respectively, in the blood for three replicate measurements. The level of precision obtained in this work enables the detection of low enrichments of Fe in blood, which is highly desired in nutrition tracer studies.  相似文献

A novel flow‐injection chemiluminescence (FI‐CL) method is described for the determination of 2‐methoxyestradiol (2‐ME). The method is based on the inhibitory effect of 2‐ME on the CL reaction of luminol and potassium ferricyanide in alkaline solution. Under optimal conditions, net CL intensity was proportional to 2‐ME concentration in synthetic and mouse plasma samples. Corresponding linear regression equations were 8.0 x 10‐9‐1.0 x 10‐7g/mL for synthetic samples and 2.0 x 10‐9‐1.0 x 10‐7g/mL for plasma samples. Detection limit for synthetic samples and limits for quantification of plasma samples were 8.4 x 10‐10g/mL (3σ) for synthetic samples and 4.0 x 10‐9g/mL for mouse samples. A complete analysis was performed for 60 s, including washing and sampling, resulting in a throughput of ≈ 60/h. The proposed method was applied for the determination of 2‐ME in synthetic and mouse plasma samples. Percentage recoveries were 101.0‐102.8% and 98.0‐105.0%, respectively. Halophilic and halotolerant bacteria were isolated from soil samples of a Japanese salt field, an environment where salt concentrations vary annually. From 1 g of each of the five samples collected, over 1×103 bacterial colonies (colony forming units (cfu)g-1) grew on agar medium containing 2M Na+. In contrast, 0–4 bacterial colonies (cfu g-1) were observed on agar medium containing 4M Na+. Two of the five samples contained numerous bacteria (102–103 cfu g-1) capable of growth on a 2M Na+ alkaline (pH=9.5) medium, while few bacterial colonies were observed from the other three samples. Only one of the five samples was shown to contain bacteria capable of growth on a 4M Na+ alkaline medium. Most of the bacteria isolated on 4M Na+ agar were eubacteria, but one extreme halophile (TR-1, already described as Haloarcula japonica JCM7785) was also isolated. The 16S rRNA sequence of TR-1 was determined and shows high homology (94.4–98.5%) to Ha. marismortui and Ha. sinaiiensis. These results suggested that: 1) environments with seasonally varying salinity can harbour halotolerants as well as halophiles and, 2) closely related halophiles can be isolated from geographically distant habitats.  相似文献

Concentrations of some radionuclides, including137Cs, in desert truffles in Kuwait were studied and compared with similar samples from other countries in the Middle East, namely Iran, Egypt, and Tunisia. In addition, sand samples from Kuwait were assayed to calculate the transfer factor of the radionuclides under consideration. The measured concentrations of40K,226Ra, and137Cs show that137Cs is much higher in Egyptian samples, whereas40K is much lower in samples from Tunisia. The average effective dose equivalent calculated for the Kuwaiti population according to their diet habits was found to be in the range 0.14-0.23 ΜSv/a. The results are compared with values from other countries.  相似文献

To assess the sensitivity of direct plating of bovine fecal samples for detection of Escherichia coli O157:H7, calves (n = 28) were orally inoculated with 109 colony-forming units (cfu) per calf of a mixture of three strains of nalidixic acid-resistant E. coli O157:H7, and fecal samples were collected for analysis. One-gram samples from inoculated calves were mixed with 9 mL of Gram-negative broth with vancomycin, cefixime, and cefsoludin. From this suspension, serial dilutions were made (10−1 to 10−4) and spread plated in triplicate on Sorbitol MacConkey agar with nalidixic acid for enumeration of E. coli O157:H7 in fecal samples. Direct plating samples were streaked for isolation on Sorbitol MacConkey agar with cefixime, and tellurite (SMACct). After incubation overnight at 37°C, morphologically typical colonies from direct streak plates were plated onto blood agar and incubated overnight at 37°C; then an indole test was performed on each colony. Indole-positive colonies were confirmed by O157 agglutination and were then plated on SMAC agar with 20 μg/mL nalidixic acid (SMACnal) to confirm nalidixic acid resistance. Overall sensitivity of detection was 32.5% (110/338 samples). Sensitivity to detect fecal samples shedding at above 5 × 104 cfu/g was 83% (71/86 samples). Based on these data, direct plating of fecal samples might be an effective way to identify cattle that are likely to be shedding E. coli O157 at high levels.  相似文献

Abstract. Solutes in mire-water samples vary greatly in their tendency to change on storage. In repeated analyses of samples from Malham Tarn, Yorkshire, U.K., concentrations of Na+ and Cl- proved very stable on storage, and Mg2+ hardly less so, whether the samples had been filtered or not. K+ concentrations declined slowly in filtered samples kept at room temperature, falling after two years to about two-thirds of the original value. In unfiltered samples, K+ concentrations rose on average by about 25 % over a period of two months. At low concentrations Ca2+ changed little on storage, but above 40 - 50 mg/l equilibration of CO2 with the air led to precipitation of carbonate and loss of dissolved Ca2+. Fe concentrations were rather stable in ombrogenous bog waters, but in fen waters showed rapid decreases on storage. SO42- showed little change in most samples, but there were a few large increases in unfiltered samples probably due to oxidation of sulphide. Unfiltered fen water samples generally developed high concentrations of NO3- (rarely NH4+) on storage; by contrast acid bog waters typically developed high NH4+ concentrations, but did not nitrify. Bog waters showed a small rise in pH and some reduction in colour (dissolved humic substances). Fen waters changed erratically in pH, depending on the balance of production and loss of CO2 in the sample. Some recommendations are given for storage and analysis of samples.  相似文献

Bovine retinas, isolated rod outer segments and emulphogene extracts of rod outer segments have been shown to contain appreciable amounts of Zn2+, Ca2+ and Mg2+ when isolated in the absence of added metal ions. Chromatography of emulphogene extracted rod outer segments in Sephadex G-25 showed virtually all the Ca2+, Zn2+ and protein to elute with the void volume. Levels of Zn2+ but not Ca2+ were light sensitive. The Zn2+ contents of protein fractions were about 60% higher when samples were bleached. Under optimal conditions protein fractions contained 1.4 – 1.8 g atoms Zn2+/mole rhodopsin for dark adapted samples and 2.1 to 3.2 g atoms Zn2+/mole of rhodopsin for bleached samples.  相似文献

Microsclerotia of the entomopathogenic fungus (Metarhizium brunneum Petch strain F52; Hypocreales: Clavicipitaceae) were sprayed in a hydromulch formulation onto wood samples to test persistence and biological control efficacy when exposed on forest and orchard trees. The density of viable conidia and retention of hydromulch were not significantly affected by the tackifier in the formulation (psyllium, xanthan, or a mixture of both). Forest samples had a maximum conidial density and more viable conidia (7.2 × 105 conidia cm?2) than orchard samples (1.4 × 105 conidia cm?2). Increased rainfall, temperature and the time that samples were outdoors were critical factors for conidial density. Asian longhorned beetles [Anoplophora glabripennis (Motschulsky); Coleoptera: Cerambycidae] exposed to orchard samples in laboratory bioassays did not have significantly less mortality compared to forest samples, indicating that hydromulch may be feasible in open sunny conditions, if sufficient moisture is present.  相似文献

Microbial diversity of soil and water samples collected from pyrochemicals exposed areas of Virdhunagar district (Tamil Nadu, India) was studied. Soil and water samples from cultivable area, waste land and city area of the same region were also studied for a comparative acount. There is a remarkable reduction in total heterotrophic bacterial population (THB) in pyrochemicals exposed soil and water samples (42 × 104 CFU/g and 5.6 × 104 CFU/ml respectively), compared to the THB of cultivable area soil and water samples (98 × 107 CFU/g and 38.6 × 107 CFU/ml). The generic composition the THB of the pyrochemicals exposed samples too exhibited considerable change compared to other samples. Pseudomonas sp. was the predominant one (41.6%) followed by Achromobacter sp. (25%) in pyrochemical exposed soil and Pseudomonas sp. was the predominant one (25%) in pyrochemical exposed water samples followed by Bacillus sp. (25%) and Micrococcus sp. (16.6%). It was observed that Cornybacterium sp. and Micrococcus sp. were absent completely in pyrochemical exposed soil and Achromobacter sp. was missing in the pyrochemical exposed water samples, which were present in the other samples. The outcome of this study clearly demonstrates that pollutants such as chemicals used in pyrotechniques affect the microbial biodiversity and suitable measures have to be taken to control the pollution level and to save biodiversity.  相似文献

To facilitate predictions of the transport and fate of contaminants at future coal conversion facilities, rates of microbial transformation of polycyclic aromatic hydrocarbons were measured in stream water and sediment samples collected in the vicinity of a coal-coking treated wastewater discharge from November 1977 through August 1979. Six radiolabeled polycyclic aromatic hydrocarbons were incubated with sediment and water samples; 14CO2, cell-bound 14C, and polar transformation products were isolated and quantified. Whereas 14CO2 and bound 14C were major transformation products in sediment assays, soluble polar 14C dominated transformation in water samples. Mean rate constants (measured at 20°C) in sediments collected downstream from the effluent outfall were 7.8 × 10−2 h−1 (naphthalene), 1.6 × 10−2 h−1 (anthracene), and 3.3 × 10−3 h−1 [benz(a)anthracene], which corresponded to turnover times of 13, 62, and 300 h, respectively. No unequivocal evidence for transformation of benzo(a)pyrene or dibenz(a,h)anthracene was obtained. Only naphthalene and anthracene transformations were observed in water samples; rate constants were consistently 5- and 20-fold lower, respectively, than in the corresponding sediment samples. The measured rate constants for anthracene transformation in July 1978 sediment samples were not related to total heterotroph numbers. In late July 1978, the effluent was diverted from the primary study area; however, no differences were observed either in transformation rate constants or in the downstream/upstream sediment rate constant ratio. These results are consistent with the hypothesis that continuous inputs of polycyclic aromatic hydrocarbons result in an increased ability within a microbial community to utilize certain polycyclic aromatic hydrocarbons. However, because transformation rates remained elevated for more than 1 year after removal of the polycyclic aromatic hydrocarbon source, microbial communities may shift only slowly in response to changes in polycyclic aromatic hydrocarbon concentrations.  相似文献
