The study deals with the distribution of non-specific esterase in germinating pollen grains ofPortulaca grandiflora. Intense activity of the enzyme is seen in small granules distributed homogeneously in pollen grains stigma hairs and throughout the length of pollen tubes. Further the walls of pollen grains also demonstrate intense activity. The functional significance of the enzyme in these locales has been discussed.  相似文献

Summary The present study describes the distribution of alkaline and acid phosphatases in the lateral line nerve of Saccobranchus fossilis and optic nerve of Mystus seenghala. The distribution of the phosphatases is quite different in these nerves, i.e., in the lateral line nerve the axons are intensely positive for alkaline and acid phosphatases whereas the myelin sheaths are negative for the enzymes. In the optic nerve, however, the axons are negative and the activity of the alkaline and acid phosphatases appear in the form of bands in the myelin sheaths. The significance of these differences is discussed with special reference to the controversy about the neurokeratin network and the physiological activities of nerves.The glial processes intervening between the myelinated axons of the optic nerve are also stained by the reactions for alkaline and acid phosphatases. The metabolic significance of phosphatases at these sites is discussed.  相似文献

Summary Detailed studies were made on the distribution of carboxylic esterases in the area postrema (AP) of the guinea pig. In this species the nerve cells of the AP show intense acetylcholinesterase activity which greatly enhances a morphological study of these elements. The two main types of nerve cells arebipolar cells andmultipolar cells. The bipolar cells often send their peripheral processes to the wall of a capillary, while the other processes go towards the nervous tissues outside of the organ. A similar behavior of the processes could be observed in the case of the multipolar cells. A special multipolar cell having bulb-like endings in the neuropilema of the organ has been discussed. On the basis of these findings it is conjectured that these neural elements, as well as the AChE positive nerve fibers located along the walls of the capillaries, function as parts of a chemoreceptor system, the existence of which has been postulated by former authors. The non-specific esterase activity of the organ is rather diffuse. B esterases have been described in the glial cells and in several nerve cells and in the pericytes and endothelial cells of the perivascular space. The localization of A and C esterases appeared diffuse, but only the AP showed intense C esterase activity in the caudal medullary area.  相似文献

The distribution of simple esterase has been studied in the nuclei and fiber tracts of the medulla oblongata. The simple esterase activity has mainly been observed in the grey matter. The white matter did not reveal enzymatic activity. In the grey matter also the cranial nerve nuclei are intensely stained as compared to intrinsic and reticular nuclei. The distributive pattern of simple esterase in the nuclei has been discussed in relation to its metabolic involvement in brain.  相似文献

The present report deals with the histo-enzymological mapping of adenosine triphosphatase and simple esterase in the diencephalon and mesencephalon of Uromastix hardwickii. The enzymatic make-up, in both cases, does not differ markedly in the various nuclei; few variations, of course, occur in some midbrain areas like pretectal nuclei. The latter are intensely positive for ATPase, while these nuclei are moderate to simple esterase. Nevertheless, one of the most interesting results pertaining to simple esterase activity is of a high order in the fiber tracts in comparison to ATPase. The other interesting feature relates to the fact that generally intensely positive nuclei, in ATPase preparations, are those which have extensive efferent fibres. The metabolic significance of high degree of ATPase activity vis-a-vis extensive efferent connections has been exhaustively discussed, besides the possible significance of the general data obtained.  相似文献

Simultaneous histochemical localization of non-specific monophosphate esterases, acid and alkaline phosphatases, as well as a specific monophosphate esterase, glucose-6-phosphatase has been made on the hepatopancreas of the marine crab, Scylla serrata (Forskål). Maximum activity of the 3 enzymes was observed in the juvenile and mature absorptive cells. Lining cells of the main hepatopancreatic duct exhibited a moderate activity of the 3 enzymes whereas the embryonic and fibrillar cells and connective tissue of the gland showed negative reactions for the 3 enzymes. The secretory cells showed a positive reaction for these enzymes only at the brush border.Bilateral eyestalk removed evoked a rise in the activity of the 3 enzymes within 2–4 h. The same effect was observed after injection of eyestalk extract to both normal and eyestalkless animals followed by restoration to the normal level after 24 h.The present observations indicate that glucose-6-phosphatase and acid phosphatase may be under the direct influence of eyestalk hormone(s) while alkaline phosphatase activity appears to be related to changes in the substrate. The physiological significance of the various cell types and enzymes is discussed.  相似文献

Summary Accomparative study of six hydrolases, acid and alkaline phosphatases, aryl sulphatase, -gluchronidase cholinesterase, and non-specific esterase, was carried out on the tissues of normal healthy and Frescon-treatedBulinus. The presence and activity of these enzymes in the tissues of normal animals were taken to indicate the probale functions of the tissues concerned. Frescon administration caused inhibition of acid phosphatase and also induced the release of cholinesterase and non-specific esterase in some tissue. It is concluded that the most important effects of Frescon on snail physiology are the disorganization of neuronal function and disturbance of olfactory activity.  相似文献

The proventriculus of White Leghorn chick embryos (stages 29–45) newly-hatched chicks, and adult chickens were frozen, sectioned in a cryostat and treated histochemically to identify localizations of alkaline and acid phosphatase, adenosine triphosphatase, 5-nucleotidase, nucleotide-diphosphatase, non-specific glycerophosphatase, glucose-6-phosphatase, non-specific esterase and succinic dehydrogenase. Ribonucleic acid, proteins and acid mucopolysaccharides were identified in tissues fixed in FAA. Acid phosphatase, nucleotide-diphosphatase, adenosine triphosphatase, succinic dehydrogenase, ribonucleic acid and proteins were present in the cells of the deep glands at all stages of development. Alkaline phosphatase and 5-nucleotidase were found only in mesenchymal derivatives of the proventriculus. After the chick begins swallowing and digesting albumen, enzymatic activity increased and non-specific esterase became very reactive. The surface epithelium is covered with a mucous coat. Ribonucleic acid, non-specific esterase, acid phosphatase and nucleotide-diphosphatase were localized in the basal portions of the epithelial cells. The functional significance of these different patterns is discussed.  相似文献

The present work describes histological and histochemical observations made on the neoplastic liver of Indian silver bills, Uroloncha malabarica. The histology of neoplastic tissue as well as liver has been discussed. Further, a few enzymes like alkaline phosphatase, acid phosphatase, 5-nucleotides and non-specific esterase have been localized in the diseased liver. The occurrence of lymphocytoma caused a marked change in the localization of the enzymes. Sometimes total inhibition of the enzyme was encountered. Damaged sinusoid cells and bile canaliculi of the neoplasm as well as liver lobules show no reaction for alkaline phosphatase. However, its counterpart, acid phosphatase, exhibits intense activity in both neoplastic tissue and liver cells. Aggregates of neoplastic tissue give moderate 5-nucleotidase reaction while it gives poor activity in hepatic tissue of the diseased liver. Parenchymatous cells are able to give some activity for the non-specific esterase while it is very dull in the neoplastic tissue.  相似文献

Using the sequential method for revealing of NADH-cytochrome c reductase and other enzymes (acid and alkaline phosphatases, non-specific esterase, peroxidase) or cationic proteins in the same section, the distinct borders between white and red pulp of the guinea pig spleen were delineated. The sequential method enables not only these areas of the spleen to be clearly distinguished, but also the migration of granulocytes from the red to the white pulp. The implications of these findings are discussed in terms of spleen function.  相似文献

Human ampullary epithelium contains numerous apical granules, which show a strong P.A.S. reaction, but do not stain with methods for acid mucins. No glycogen is detected. Acid phosphatase and non-specific esterase are localized in granules, scattered within the cytoplasm, while peroxidase activity is not demonstrable. Abstract— Isolated bovine central nerve myelin sheath preparations showed non-specific esterase activity towards naphthyl ester substrates of increasing chain length from acetate to palmitate. Short chain esters were hydrolysed much faster than long chain substrates by myelin, the specific activity for the hydrolysis of β-naphthyl acetate being the highest. Micro-somal fractions from brain white matter were much higher in esterase activity to all naphthyl ester substrates. NADPH-cytochrome c reductase activity was absent from isolated myelin samples. Distilled water and salt and buffer solutions of different ionic strengths and pH were ineffective in releasing non-specific esterase activity from myelin although tri-potassium citrate caused marked inhibition of the membrane-bound esterase activity. The detergent Triton X-100 released esterase activity from the myelin preparations but at a concentration of 0.1 per cent was also inhibitory.  相似文献

Summary The distribution of the acetylcholinesterase, non-specific cholinesterase and non-specific esterase activity has been investigated histochemically in the hypothalamic neurons during the ontogenic development of the rat.Acetylcholinesterase activity is located in the supra-optic and para-ventricular nuclei mostly, but some activity is present in the other nuclei and in the median eminence of the adult rat, as well. The supra-chiasmatic neurons are always negative. The activity of non-specific cholinesterase was encountered in the endothelial cells of the capillaries, in the glia and in the ependymal cells especially around the supra-optic and para-ventricular neurons. The localization of the non-specific esterase was similar to that of the non-specific cholinesterase, but in addition activity is seen in the supra-optic and para-ventricular perikarya, in the parvo-cellular neurons of the tuberal area and in the median eminence. No sexual differences were seen in the distribution of the estrase activity.The appearance of acetylcholinesterase took place already before birth. At about the 16th post-coital day the area from which the arcuate and ventro-medial nuclei will differentiate was positive for acetylcholinesterase. A strong activity in these nuclei was observed during the critical period of the sexual differentiation of the rat hypothalamus (0–10 postnatal days). In the development of the non-specific cholinesterase and esterase no similar variation was seen. Acetylcholinesterase and non-specific esterase were seen in the neurosecretory nuclei before birth, non specific cholinesterase after birth, and non-specific esterase in the parvo-cellular neurons during the first post-natal week.Supported by a grant from The Finnish Medical Society Duodecim.  相似文献

Summary The dynamics of non-specific esterase in the upper duodenum of the house mouse was studied electron microscopically at various intervals following a fat meal. Enterocytic esterase became associated with lipid droplets during fat resorption and formation of primary chylomicrons. Esterase activity remained associated with the primary chylomicrons throughout the process of extrusion into the extracellular space at the lateral interdigitations, and during subsequent transport into the lymph vessels. It is suggested that certain isozymes of non-specific esterase participate in lipid transport.Supported by the Deutsche Forschungsgemeinschaft (SFB 46). Changes in the amount and distribution of acid and alkaline phosphatase, non-specific esterase, glycogen, lipid and acid mucopolysaccharide in the caecal wall and liver of turkey poults infected with Histomonas meleagridis have been studied histochemically. A microdensitometer was used to measure changes in activity and distribution of acid phosphatase and non-specific esterase in the caecal mucosa. During the course of the infection there is a marked reduction in activity and distribution of acid phosphatase and non-specific esterase but little change in the amounts and distribution of alkaline phosphatase, glycogen, lipid and acid mucopolysaccharide in the wall of the main part of the caecum. Similar, but smaller, changes occurred in the wall of the neck region of the caecum. In the liver most changes occurred in the immediate vicinity of the parasites. Initially, there was a reduction in the amount of glycogen in the parasitic lesions but later in the infection there was a marked loss of glycogen in all regions of the liver. Changes in the caecum are apparently brought about by the parasite prior to and after invasion of the caecal tissues; changes in the liver occur after it has been invaded.  相似文献

The esterase, phosphatase, and sulfatase activities of carbonic anhydrase (CA, EC 4.2.1.1) isozymes, CA I, II, and XIII with 4-nitrophenyl esters as substrates was investigated. These enzymes show esterase activity with 4-nitrophenyl acetate as substrate, with second order rate constants in the range of 753-7706M(-1)s(-1), being less effective as phosphatases (k(cat)/K(M) in the range of 14.89-1374.40M(-1)s(-1)) and totally ineffective sulfatases. The esterase/phosphatase activities were inhibited by sulfonamide CA inhibitors, proving that the zinc-hydroxide mechanism responsible for the CO(2) hydrase activities of CAs is also responsible for their esterase/phosphatase activity. CA XIII was the most effective esterase and phosphatase. CA XIII might catalyze other physiological reactions than CO(2) hydration, based on its relevant phosphatase activity.  相似文献

The inhibition of neurotoxic esterase activity in chicken brain has been studied in vitro and in vivo. Aphos exposure, causing chicken paralysis, has demonstrated that the initial stage of delayed neurotoxicity was significant esterase activity inhibition (by 60-80%) within 3-24 hours after the pesticide administration. The inhibition of cholinesterase activity occurred both in the blood and sciatic nerve. The delayed conduction through peripheral nerves caused by demyelination has been revealed in the latent period (before the clinical signs of intoxication).  相似文献

The study focused on Norway spruce trees during their annual development and, specifically, on the histochemical analysis of the activity of the non-specific esterase and the amount of polyphenols in the vegetative buds. Azo-coupling reaction with naphthol-AS-acetate as a substrate and Fast Blue B Salt as a coupling reagent was applied to the longitudinal median cryo-sections of the formalin-fixed buds. A method of image analysis was developed to evaluate the intensity of the histochemical reaction quantitatively. High activity of non-specific esterase was detected in the cells of the collenchymatic plate, the youngest bud scales, the leaf primordia, in the zone of differentiation of vascular tissues and procambium of the dormant bud. The activity of non-specific esterase decreased in all tissues during bud break. During the growth of a new shoot enzyme activity was detected only in the vascular bundles of the shoot. Enzyme activity re-occurred at the beginning of the autumn in the cells of the collenchymatic plate of the newly establishing buds. Three different types of polyphenols at different intracellular localizations (as verified by scanning electron microscopy) were observed: granular, vacuolar and drop-like polyphenols. The amount and localization of those polyphenols changed during the bud growing cycle. The positive reaction of all three types of polyphenols are tannin in nature. These findings are discussed from the viewpoint of frost tolerance and tissue differentiation.  相似文献

A histochemical study of dry and germinated wheat embryos andseeds has been made to identify the sites of activity of 3'-and 5'-nucleotidases, phosphatases, lipase, and esterase. Theenzymes all showed the same distribution, with the coleorhizaas the most prominent site of activity in the dry embryo, especiallywhen compared with the adjacent roots with very low activity.Investigations of the whole seed after 24 h germination showeda very high activity in the aleurone cells and high activitiesin the scutellum and coleorhiza. The epithelium and provasculartissues of the scutellum showed moderate to low activity whilethe endosperm had little or none. Hydrolase activity also wasfound in the testa. The function of the coleorhiza. as firstfood reserve for the roots and its location at the most importantsite of water entry, is discussed.  相似文献

Acid phosphatase, non-specific esterase, alkaline phosphatase, monoamine oxidase and true lipase activities, in the epidermis of Natrix piscator in different stages of the sloughing cycle, have been localized using various histochemical techniques.
Different layers in scale epidermis have staining properties similar to corresponding layers in hinge epidermis.
Acid phosphatase and non-specific esterase activity in cell layers undergoing keratinization, and the lacunar tissue undergoing disintegration are associated with hydrolytic and catabolic wasting processes involving cell death. The activity of these enzymes in the clear layer is associated with the breaking down of the cementing substance resulting in the separation of clear layer from underlying tissue and facilitating the shedding of old slough.
Alkaline phosphatase activity in the stratum germinativum and undifferentiated epidermal cells has been associated with cell proliferation and differentiation. The presence of alkaline phosphatase in the lacunar tissue and clear layer has been correlated with the synthesis of mucopolysaccharides in these layers.
Monoamine oxidase and true lipase activity could not be located in the epidermis at any stage of the sloughing cycle.  相似文献
