Structural comparisons of mouse histones 2A.X and 2A.Z with 2A.1 and 2A.2   总被引:4,自引:0,他引:4  
The tryptic peptide patterns of the recently described H2A species H2A.X and H2A.Z from mouse were compared with the tryptic peptide patterns of the major mouse H2A's, H2A.1 and H2A.2. The identities of the H2A.1 peptides were determined by comparing their in vivo labeling with various 14C-labeled amino acids with the expected labeling determined from the known sequence. All the H2A.1 tryptic peptides larger than dipeptides were accounted for. The procedure was repeated for H2A.2, H2A.X and H2A.Z. H2A.X was found to have large regions of sequence identical to that of H2A.1 with the variability occurring mainly near the N and C termini. Mouse H2A.X had some sequence characteristics found in the sequenced H2A's of trout and sea urchin. In contrast, H2A.Z was found to have only two peptides in common with H2A.1; in addition, the labeling patterns of the non-identical peptides were too different to suggest analogous peptides. We conclude from these studies that H2A.Z differs considerably from H2A.1 in major portions of its sequence.  相似文献

Poly 2'-azido-2'-deoxyadenylic acid (Poly Az) was synthesized from 2'-azido-2'-deoxyadenosine diphosphate by polynucleotide phosphorylase. Poly (Az) has U.V. absorption properties similar to poly (A) and hypochromicity of 40% at 0.1 M Na+ and neutrality. CD curve also resembled to that of poly (A), but has smaller ellipticity. Titration of poly (Az) with HCl gave a transition at pH 5.5, but exact structure of the acid-form complex was not elucidated. Upon mixing with poly (U), poly (Az) forms a 1:1 and 1:2 complexes having Tm's somewhat higher than that of poly (A)- poly (U) complex in the same condition.  相似文献

The recovery from rat and rabbit tissues of fed methyl-(14)C and ethyl-2-(14)C 2-methylpalmitate with unaltered specific activity has demonstrated the existence of mechanisms for the absorption and deposition of both methyl and ethyl esters of fatty acids, at least for 2-methylpalmitate. In thoracic duct-cannulated rats, approximately 9% of the fed compounds was recovered from the lymph during the first 24 hr, the rate of recovery reaching a maximum between 6 and 8 hr. In the rabbit, the fed, unaltered esters in plasma were transported principally by means of the low density lipoproteins. Only trace amounts of the unaltered esters were subsequently detected in the blood and tissue lipids after feeding, however, even during the period of maximal absorption; moreover, in contrast to at least one report by others, further analyses for methyl or ethyl esters of other fatty acids has shown that such esters of short-chain alcohols constitute no more than a trace amount (0.004-1.03%) of the lipids extracted from a wide variety of mammalian tissues. The possibility remains that even these trace amounts of esters arose as artifacts of autolysis, extraction, or assay.  相似文献

GM2 and GA2 gangliosides from the brain of a patient who died of Sandhoff's disease were purified by solvent partition, silicic acid and silica gel column chromatography, and silica gel preparative thin-layer chromatography. They were tritiated in the terminal N-acetylgalactosamine residue using galactose oxidase and sodium [3H]borohydride with the inclusion of catalase and peroxidase into the oxidation reaction. The specific activities were 4.62 X 10(8) dpm/mumol of GM2 ganglioside and 5.54 X 40(7) dpm/mumol of GA2 ganglioside. The addition of catalase and peroxidase to the tritiation procedure is recommended.  相似文献

Pancreatic endocrine cells of the neonatal rat cultured in medium with 5.5 mM glucose for 7 days showed no response to glucose. By contrast, the supplementation of the medium with 1.0 mM 2-deoxyglucose or with 0.1-5.0 mM 2-deoxy-2-fluoroglucose maintained the capacity of glucose-induced insulin release and biosynthesis, and the recovery of insulin in cells at day 7 at levels significantly higher than in basal medium; the highest responses were recorded for 1.0 mM deoxysugars. Moreover, the addition of 1.0 mM deoxysugars caused a selective deletion of fibroblasts and yielded monolayers mostly consisted of endocrine cells at the end of the culture study period. In these monolayer cells, the stimulating level of c-AMP release was significantly higher than the basal. On the other hand, the in vitro function of A cells in culture was also better preserved in media with 1.0 mM deoxysugars.  相似文献

The preparation of synthetic oligonucleotides containing 2'-deoxynebularine (dN) and 2'-deoxyxanthosine (dX) is described. The thermal stabilities of duplexes containing dX, dN, and 2'-deoxyinosine (dI) base-paired with the four natural bases have been measured. Xanthine base pairs have stabilities at pH 5.5 that are similar to those of dI-containing duplexes at neutral pH. When xanthine is paired with adenine or cytosine an unusual stabilization of the duplex structure is observed at acid pH. Incorporation of base mispairs opposite template xanthine sites were measured using Drosophila DNA polymerase alpha. The relative nucleoside incorporation rates are in the order: T greater than C much greater than A approximately equal to G. These rates do not correlate with relative thermodynamic stabilities of base mispairs with xanthine obtained from Tm measurements: T greater than G greater than A approximately equal to C. We suggest that DNA polymerase misinsertion rates are greatest when the base mispair can be formed in accordance with Watson-Crick as opposed to other base pairing geometries even though other geometries, e.g. wobble, may result in a more stable final DNA product.  相似文献

Laser-Raman spectra of d-ribose and 2-deoxy-d-erythro-pentose in aqueous solution are reported. F.t.-i.r. and Raman spectra have been obtained for crystals of these sugars. Assignments of the Raman bands observed in solution are proposed. The spectral differences between the two sugars are discussed in terms of the structural difference. The analysis of the frequencies observed permits identification of each of the sugars and their isomeric analogs, and can be used as a basis for study of nucleosides and nucleotides by vibrational spectroscopy.  相似文献

To react with peptides, nitric oxide.NO has to be activated by oxidation, or by coupling with superoxide (O.-2) thereby producing peroxynitrite. In the course of.NO oxidation,.NO2 free radicals and N2O3 may be formed. Using gamma-irradiation methods, we characterized the products formed by these nitrogen oxides with angiotensin II. Angiotensin II is specifically nitrated at its tyrosinyl residue by.NO2 or peroxynitrite. Equimolecular amounts of each reagent in K+/Pi solutions at pH 7.4 led to 56% and 5% nitration yields, respectively. Nitrogen oxides produced by autoxidation of.NO, as well as.NO2 under.NO, reacted only with the arginine residue, giving a mixture of peptides containing citrulline, a N-(hydroxylamino-cyanamido-) instead of guanido group, and a conjugated diene derived from an arginine side-chain. However, nitrosation reactions by N2O3 occurred only when the initial concentration of.NO2 was 10 times that able to react with angiotensin II. Thus, in this case.NO appears to protect against.NO2 action.  相似文献

Serological characterization of three K-S interval recombinant strains, TBR2 (H-2at2), TBR3 (H-2at3) and AIR 1 (H-2a2) was performed using anti-H-2, Ia, Ss and Slp antisera. The data presented here reveal that the crossover events in both TBR2 and TBR3 occurred between the I-A and I-E subregions. In both cases, the H-2K and I-A subregions were derived fron the H-2t1 of chromosome, while the I-E, S and H-2D regions were derived from the H-2b chromosome (KsAkEbSbDb). The H-2a2 chromosome resulted from a crossover event between the H-2a1 and H-2i9 chromosomes. Ia and Ss typing of AIR 1 suggested that the K to I-E regions originated from H-2a1 and the S and D regions originated from H-2i9 (KkAkEkSbDd).  相似文献

Isolation and expression of GA 2-oxidase2 in tomato.   总被引:1,自引:0,他引:1  
GA 2-oxidases, a key enzyme involves GA biosynthesis, catalyze the degradation of active C(19)-Gibberellins (GAs) through 2-hydroxylation yields inactive GA product. Searching public tomato database, the putative GA2ox2 sequences were assembled. We isolated a full-length GA2ox2 cDNA with primers designed from the assembled sequence. This gene was designed as SlGA2ox2 (GenBank accession No. EF017805). The full-length GA2ox2 gene contained a complete open reading frame (ORF) of 1203 bp, which encoded 322 amino acid residues. Amino acid sequence homology analysis of SlGA2ox2 showed an 88% identity with NtGA2ox2 in tobacco. And alignments of SlGA2ox2 with other known GA2ox from Arabidopsis, Pea, Adzuki Bean, Winter Squash etc indicate low similarity of 47-70%. Semi-quantitative RT-PCR analysis showed a specific expression profile of SlGA2ox2 in different tissues, which mainly expressed in flowers and traces were detected in roots, stems, leaves and immature fruits.  相似文献

Poly (2'-chloro-2'-deoxyadenylic acid) and poly (2'-bromo-2'-deoxyadenylic acid) were synthesized from the corresponding diphosphates with the aid of polynucleotide phosphorylase from E. coli. UV, CD, acid titration and mixing with poly (U) were investigated. Comparing these properties with those of poly (A) and poly (2'-azido-2'-deoxyadenylic acid), it was found that 2'substituents exert significant effects on the thermal stability of these polynucleotides, though the overall conformational structure was not greatly changed.  相似文献

The syntheses of 5-ethynyl-, 5-phenylethynyl- and 5-propynyl-2(1H)-pyrimidinone 2′-deoxyribosides are described. Two of the new nucleosides, 14 a and 14 c, showed significant antiviral activities against HSV-1 and HSV-2 in tissue culture.  相似文献
