Populations of weed beets have expanded into European sugar beet production areas since the 1970s, thereby forming a serious new weed problem for this crop. We sampled seeds in different French populations and studied mitochondrial DNA, chloroplast DNA and life-cycle variability. Given the maternal inheritance of the mitochondrial and chloroplastic genomes and the nuclear determinism of the annual habit, we were able to determine the maternal origin and evolution of these weed beet populations. Our study shows that they carry the dominant allele B for annual habit at high frequency. The main cytoplasmic DNA type found in northern weed beet populations is the cytoplasmic male-sterile type characteristic of sugar beets. We were able to determine that these populations arise from seeds originating from the accidental pollinations of cultivated beets by adventitious beets in the seed production area, which have been transported to the regions where sugar beets are cultivated. These seeds are supposedly the origin of the weed forms and a frequently disturbed cultivated environment has selected for annual habit and early flowering genotypes. We discuss the consequences of the weed beet populations for the breeding, seed production and release of herbicide-resistant transgenic sugar beets.  相似文献

The fate of cellular DNA during the standard purification steps of the sugar manufacturing process from conventional and transgenic sugar beets was determined. Indigenous nucleases of sugar beet cells were found to be active during the first extraction step (raw juice production) which was carried out at 70°C. This and the consecutive steps of the manufacturing process were validated in terms of DNA degradation by competitive PCR of added external DNA. Each step of the process proved to be very efficient in the removal of nucleic acids. Taken together, the purification steps have the potential to reduce the amount of DNA by a factor of >1014, exceeding by far the total amount of DNA present in sugar beets. Furthermore, the gene products of the transgenes neomycin phosphotransferase and BNYVV (rhizomania virus) coat protein CP21 were shown to be removed during the purification steps, so that they could not be detected in the resulting white sugar. Thus, sugar obtained from conventional and transgenic beets is indistinguishable or substantially equivalent with respect to purity.  相似文献

Pest management in confinement swine production relies primarily on calendar-based applications of broad-spectrum insecticides. However, regulatory restrictions imposed by the U.S. Food Quality Protection Act of 1996, the large financial obligation of pesticide registration, and development of insecticide resistance have led to a renewed search for alternative control methods. Boric acid dust has long served as an insecticide in urban pest management and has been shown an effective alternative for use in sensitive environments such as swine production. However, dust formulations are difficult to apply and require specialized equipment. The purpose of this study was to evaluate the efficacy of liquid baits containing boric acid for the control of German cockroaches in a commercial swine nursery. Bait, consisting of 1 or 2% boric acid and 0.5 M sucrose, was deployed in 21 bait delivery tubes per room. Results of a 2-yr study showed significant reductions in cockroach populations. When baits were withdrawn in the summer, the cockroach population increased significantly faster than when the baits were removed during the winter. These data indicate that liquid formulations of boric acid effectively reduce the burden of cockroach infestation in swine production. This approach should have applications in structures in other urban and agricultural environments.  相似文献

Fifty-five Fusarium isolates belonging to nine species were collected from fungus-invaded tissue of stored sugar beets and identified as F. acuminatum (11 isolates), F. avenaceum (1 isolate), F. culmorum (1 isolate), F. equiseti (23 isolates), F. graminearum (4 isolates), F. oxysporum (1 isolate), F. solani (4 isolates), F. sporotrichioides (7 isolates), and F. subglutinans (2 isolates). All isolates were cultured on autoclaved rice grains and assayed for toxicity by feeding weanling female rats the ground-rice cultures of the isolates in a 50% mixture with a regular diet for 5 days. Fifty-eight percent of the isolates were acutely toxic to rats, 26% caused hematuria, 18% caused hemorrhages, and 29% caused uterine enlargement. In most cases, toxicity could not be accounted for by the known toxins found. The following mycotoxins were found in extracts of the rice cultures: zearalenone (22 to 6,282 micrograms/g), chlamydosporol (HM-8) (68 to 4,708 micrograms/g), moniliformin (45 to 400 micrograms/g), deoxynivalenol (10 to 34 micrograms/g), 15-acetyldeoxynivalenol (5 to 10 micrograms/g), diacetoxyscirpenol (22 to 63 micrograms/g), monoacetoxyscirpenol (21 to 26 micrograms/g), scirpenetriol (24 micrograms/g), T-2 toxin (4 to 425 micrograms/g), HT-2 toxin (2 to 284 micrograms/g), neosolaniol (2 to 250 micrograms/g), and T-2 tetraol (4 to 12 micrograms/g). F. equiseti was the predominant species found on visibly molded beets in the field. Five toxigenic isolates of Fusarium species were tested for the production of zearalenone, moniliformin and trichothecenes (deoxynivalenol, 15-acetyldeoxynivalenol, T-2, HT-2 and neosolaniol) when grown on solid sugar beet slices in the laboratory for thirty days. The isolates were also grown on a solid rice medium for comparison. High zearalenone and trichothecene-producing isolates originally obtained from corn and corn-based feedstuff were compared with isolates obtained from sugar beets. One moniliformin-producing isolate from wheat was included in the study. With the exception of moniliformin, all toxins were produced on both substrates; however, the rice medium yielded the greater concentrations except for HT-2 which was produced on sugar beets in equal or greater concentrations. Zearalenone production on rice reached 729–1943 gmg/g whereas on sugar beet it reached 72–193 gmg/g. The moniliformin-producing isolate grew well on both substrates; however, moniliformin was produced only on the rice substrate. This study demonstrates for the first time that Fusarium species can produce both zearalenone and the trichothecenes on a sugar beet substrate.  相似文献

Summary The nature and amounts of by-products formed during conversion of sugar beets to ethanol byZ. mobilis in Conventional Submerged Fermentation (CSF) and Solid-State Fermentation (SSF) were investigated. It was found that the bacterium produced fewer by-products in SSF than CSF, and that by-products profile was different. The influence of fermentation temperature on synthesis of by-products in SSF was also studied. High fermentation temperature favoured sorbitol synthesis and low fermentation temperature the synthesis of levan. The best results were obtained at 35°C. An ethanol yield of up to 95% of the theoretical value with final ethanol concentration of 142 g/L were obtained.  相似文献

Aerobic plate counts (APC 37°C and APC 25°C) and Escherichia coli enumerations (Petrifilm) were used to determine sources of bacterial contamination during sheep dressing, determine the hygienic efficacy of hand wash and knife 'sterilization'procedures and compare the hygiene efficiency of conventional and inverted sheep dressing systems. The major slaughterline sources of microbial contamination were: fleece > workers' hands > faecal pellets > knife blades. Aerobic plate counts (APC 37°C) exceeding log 4.4 cfu cm-2 were considered indicative of direct fleece contact, whereas E. coli numbers exceeding log 3.3 cfu cm-2 were considered indicative of direct faecal contact. A 44°C water hand rinse removed 90% of the microbial contamination from workers' hands, but rinsed hands, particularly those contacting the fleece, still carried a microbial population exceeding log 4.0 cfu cm-2. A 44°C rinse followed by an 82°C water dip reduced the contamination on knife blades to less than log 3.0 cfu cm-2. Inverted dressing systems produced carcasses with a lower contamination level than conventional systems. With both systems little increase in contamination occurred after pelt removal. The areas of highest contamination were the forequarter region with inverted dressing and the hindquarter with conventional dressing. In both cases these regions are the sites where cuts are made through the skin. With both systems contamination around these cuts was entirely consistent with direct fleece contact resulting from 'rollback'.  相似文献

Eco-efficiency is concerned with the efficient and sustainable use of resources in farm production and land management. It can be increased either by altering the management of individual crop and livestock enterprises or by altering the land-use system. This paper concentrates on the effects of crop sequence and rotation on soil fertility and nutrient use efficiency. The potential importance of mixed farming involving both crops and livestock is stressed, particularly when the systems incorporate biological nitrogen fixation and manure recycling. There is, however, little evidence that the trend in developed countries to farm-level specialization is being reduced. In some circumstances legislation to restrict diffuse pollution may provide incentives for more diverse eco-efficient farming and in other circumstances price premia for produce from eco-efficient systems, such as organic farming, and subsidies for the provision of environmental services may provide economic incentives for the adoption of such systems. However, change is likely to be most rapid where the present systems lead to obvious reductions in the productive potential of the land, such as in areas experiencing salinization. In other situations, there is promise that eco-efficiency could be increased on an area-wide basis by the establishment of linkages between farms of contrasting type, particularly between specialist crop and livestock farms, with contracts for the transfer of manures and, to a lesser extent, feeds.  相似文献

The effects of paraquat were studied at 4.7 kg a. i./ha and of glyphosate at 9.4 kg a. i./ha on the weight loss and rate of oxygen uptake by stems of barley straw, incubated on the soil surface for various periods of time, and stems and leaves of wheat straw on the soil surface or covered with a shallow layer of soil. In general, paraquat reduced weight loss and frequently oxygen uptake. Glyphosate normally had no effect but occasionally it was stimulatory. However, in isolated instances, paraquat was inactive and glyphosate slightly inhibitory. Leaves decayed faster than stems. Burial, as well as the addition to straw of leaf protein, Nitrochalk, urea and clover plants increased decomposition in all treatments, but nitrogen amendments were less effective in those with paraquat. For example, urea enhanced weight loss after 21 wk from 19 to 34% of the initial weight in unsprayed wheat straw, from 12 to 24% in paraquat-treated and from 20 to 37% in glyphosate-treated straw, indicating that conditions stimulating decomposition may attenuate but not entirely eliminate the inhibitory effects of paraquat. A repeated addition of nitrogen to straw in various stages of decay increased oxygen uptake in all treatments but least in the paraquat-treated straw, implying that a complete recovery from the initial curtailment of microbial activity, brought about by this herbicide, did not occur. Although the concentration of paraquat used was nine times greater than the minimum recommended for field application, the degree of inhibition observed in these laboratory experiments was probably not sufficiently great to be of marked economic importance.  相似文献

Use of eco-friendly area specific salt tolerant bioinoculants is better alternatives to chemical fertilizer for sustainable agriculture in coastal saline soils. We isolated diverse groups of diazotrophic bacteria from coastal saline soils of different forest and agricultural lands in the Sundarbans, West Bengal, India, to study their effect on crop productivity in saline soils. Phenotypic, biochemical and molecular identifications of the isolates were performed. The isolates produced indole acetic acid, phosphatase, and solubilized insoluble phosphates. Sequence analysis of 16S rDNA identified the SUND_BDU1 strain as Agrobacterium and the strains SUND_LM2, Can4 and Can6 belonging to the genus Bacillus. The ARA activity, dinitrogen fixation and presence of nifH genes indicated they were diazotrophs. Field trials with these strains as bioinoculants were carried out during 2007-2009, with rice during August-December followed by Lady's finger during April-June. Microplots, amended with FYM inoculated with four bioinoculants individually were compared against sole FYM (5 t ha(-1)) and a sole chemical fertilizer (60:30:30 kg ha(-1) NPK) treated plot. The strain Can6 was by far the best performer in respect of yield attributes and productivity of studied crops.  相似文献

Field studies were conducted over 4 years at two locations in Manitoba, Canada to evaluate effects of preceding crop, tillage and phosphorus (P) fertilization on Cd and Zn concentration in oilseed flax (linseed—Linum usitatissimum L.). Canola (Brassica napus L.) and spring wheat (Triticum aestivum L.) were grown under conventional and reduced tillage with three rates of monoammonium phosphate. Flax was seeded the year following the spring wheat or canola, with or without P fertilizer, and Cd and Zn concentration and accumulation were determined in the flax tissue at 5 weeks and in the mature seed. Flax following canola had lower Zn concentration and accumulation and higher Cd concentration and Cd:Zn ratio in the tissue and seed relative to flax following wheat. Phosphorus fertilization tended to increase Cd concentration and Cd:Zn ratio and decrease Zn concentration in the tissue and seed. Effects of tillage and interactions among tillage, preceding crop, and P fertilization were inconsistent. Changes in flax Cd and Zn concentration may be due to changes in mycorrhizal colonization or by the high concentration of Cd in the decomposing canola residue. Crop sequence and P management can be used to improve flaxseed food quality, by increasing Zn and decreasing Cd concentration.  相似文献

Drivers behind food security and crop protection issues are discussed in relation to food losses caused by pests. Pests globally consume food estimated to feed an additional one billion people. Key drivers include rapid human population increase, climate change, loss of beneficial on-farm biodiversity, reduction in per capita cropped land, water shortages, and EU pesticide withdrawals under policies relating to 91/414 EEC. IPM (Integrated Pest Management) will be compulsory for all EU agriculture by 2014 and is also being widely adopted globally. IPM offers a 'toolbox' of complementary crop- and region-specific crop protection solutions to address these rising pressures. IPM aims for more sustainable solutions by using complementary technologies. The applied research challenge now is to reduce selection pressure on single solution strategies, by creating additive/synergistic interactions between IPM components. IPM is compatible with organic, conventional, and GM cropping systems and is flexible, allowing regional fine-tuning. It reduces pests below economic thresholds utilizing key 'ecological services', particularly biocontrol. A recent global review demonstrates that IPM can reduce pesticide use and increase yields of most of the major crops studied. Landscape scale 'ecological engineering', together with genetic improvement of new crop varieties, will enhance the durability of pest-resistant cultivars (conventional and GM). IPM will also promote compatibility with semiochemicals, biopesticides, precision pest monitoring tools, and rapid diagnostics. These combined strategies are urgently needed and are best achieved via multi-disciplinary research, including complex spatio-temporal modelling at farm and landscape scales. Integrative and synergistic use of existing and new IPM technologies will help meet future food production needs more sustainably in developed and developing countries, in an era of reduced pesticide availability. Current IPM research gaps are identified and discussed.  相似文献

Nutrient and water management is crucially important in shallow-rooted vegetable production systems characterized by high input and high environmental risk. A 2-year field experiment on greenhouse cucumber double-cropping systems examined the effects of root zone nitrogen management and planting of sweet corn as a catch crop in the summer fallow period on cucumber yield and soil Nmin dynamics compared to conventional practices. Cucumber fruit yields were not significantly affected by root zone N management and catch crop planting despite a decrease in N fertilizer application of 53% compared to conventional N management. Soil Nmin content to a depth of 0.9 m decreased markedly and root zone (0–0.3 m) soil Nmin content was maintained at about 200 kg N ha?1. Root zone N management efficiently and directly reduced apparent N losses by 44% and 45% in 2005 and 2006, respectively. Sweet corn, the summer catch crop, depleted Nmin residue in the soil profile of 1.8 m at harvest of winter–spring season cucumber by 304–333 kg N ha?1, which contributed 19–22% reduction in N loss. Compared to conventional N management, N loss was reduced by 56% under root zone N management and catch crop planting.  相似文献

Nitrogen fertilization strategies were widely adopted to enhance grain production and improve nitrogen utilization in rice all over the world. For fertilization timing strategy, ear fertilization was usually employed in recent years. For fertilization amount strategy, nitrogen fertilization would continually increase to meet the demands of increasing people for food. However, under heavy ear fertilization as well as great nitrogen amount (NA), physiological N-use efficiency (PE, defined as grain production per unit nitrogen uptake by plants) decreased. Under three NA and two ratios of fertilization given during ear development period to total NA (ear fertilization distribution ratio, EFDR), net photosynthetic rate (Pn), Pn to nitrogen content per unit area (photosynthetic N-use efficiency, Pn/N), nitrogen accumulation in plant tissues and PE of three rice (Oryza sativaL.) genotypes, Jinyou 253, Liangyoupeijiu and Baguixiang were screened in the first and second seasons in 2002 so as to understand the fluctuation patterns of Pn/N and nitrogen distribution in leaf blades under great NA & EFDR and relationship with PE in rice. Results showed that under greater NA & EFDR, Pn in flag leaves at heading and plant nitrogen accumulation at maturity always increased and PE & Pn/N always decreased in spite of increased grain production. Rice distributed more nitrogen in leaf blade under greater NA and EFDR. PE indicated significantly (P<0.05) positive relationship with Pn/N and negative relationship with nitrogen distribution ratio in leaf blades at heading and maturity, and no association with Pn in two growing seasons. Results suggested that low PE in rice under great NA and heavy ear fertilization is associated to more nitrogen distribution in leaf blades and decreases in photosynthetic efficiency.  相似文献

Although Brazil has recently reached the position as the second largest producer of genetically modified soybean [Glycine max (L.) Merr.], there are few reports on the effects of transgenic crops and the associated use of specific herbicides on soil microbial communities, both under the edaphoclimatic conditions in Brazil, and in other producer regions in the southern hemisphere. The aim of this study was to evaluate the effects of transgenic soybean containing the ahas gene conferring resistance to herbicides of the imidazolinone group, and of the herbicides associated with transgenic soybeans on the soil microbial community. Twenty field experiments were carried out during three growing seasons (summer of 2006/2007, short-season of 2007 and summer of 2007/2008), in nine municipalities located in six Brazilian states and in the Federal District. The experiments were conducted using a completely randomized block design with four replicates and three treatments: (1) conventional (non-transgenic) soybean cultivar Conquista with conventional herbicides (bentazone + acifluorfen-sodium and other herbicides, depending on the level of infestation in each region); (2) near-isogenic transgenic Cultivance (CV127) containing the ahas gene, with conventional herbicides; (3) transgenic Cultivance with specific herbicide of the imidazolinone group (imazapyr). As the objective of the study was to verify impacts of the transgene and herbicides on the soil microbial community of the whole area and not only a punctual rhizospheric effects, samples were taken at the 0–10 cm layer prior to cropping and at R2 soybean growth stage, between plant rows. Quantitative (microbial biomass C and N, MB-C and MB-N) and qualitative (DGGE of the 16S rDNA region) parameters of soil microbial community were evaluated. No qualitative or quantitative differences were found that could be attributed to the transgene ahas. A comparison of Cultivance soybean with conventional and imidazolinone-group herbicides applications also failed to reveal differences that could be attributed to the specific use of imazapyr, even after three consecutive croppings at the same site. Finally, no differences were detected between conventional (Conquista and conventional herbicides) and transgenic soybean managements (Cultivance and imazapyr). However, marked differences were observed in MB-C and MB-N between the different sites and times of year and, for the 16S rDNA-DGGE profiles, between different sites. In conclusion, microbial community evaluations were found to be sensitive and viable for monitoring different technologies and agricultural management methods, but no differences could be attributed to the ahas transgene for three consecutive cropping seasons.  相似文献

Conventional soybean weed management and transgenic herbicide-tolerant management were examined to assess their effects on soybean insect pest populations in south Georgia in 1997 and 1998. Soybean variety had very little impact on the insect species observed, except that maturity group effects were observed for stink bug, primarily Nezara viridula (L.), population densities on some sampling dates. Stink bugs were more abundant on the early maturing varieties in mid-season. Velvetbean caterpillar, Anticarsia gemmatalis Hübner, potato leafhopper, Empoasca fabae (Harris), and grasshoppers Melanoplus spp. were more numerous on either conventional or herbicide-tolerant varieties on certain dates, although these differences were not consistent throughout the season. Soybean looper, Pseudoplusia includens (Walker), threecornered alfalfa hopper, Spissistilus festinus (Say), and whitefringed beetles, Graphognathus spp , demonstrated no varietal preference in this study. Few weed treatment differences were observed, but if present on certain sampling dates, then pest numbers were higher in plots where weeds were reduced (either postemergence herbicides or preplant herbicide plus postemergence herbicide). The exception to this weed treatment effect was grasshoppers, which were more numerous in weedy plots when differences were present. In post emergence herbicide plots, there were no differences in insect pest densities between the conventional herbicides (e.g., Classic, Select, Cobra, and Storm) compared with specific gene-inserted herbicide-tolerant materials (i.e., Roundup and Liberty). Defoliation, primarily by velvetbean caterpillar, was different between soybean varieties at some test sites but not different between herbicide treatments. We did not observe differences in seasonal abundance of arthropod pests between conventional and transgenic herbicide-tolerant soybean.  相似文献

The potential expansion of biofuel production raises food, energy, and environmental challenges that require careful assessment of the impact of biofuel production on greenhouse gas (GHG) emissions, soil erosion, nutrient loading, and water quality. In this study, we describe a spatially explicit integrative modeling framework (SEIMF) to understand and quantify the environmental impacts of different biomass cropping systems. This SEIMF consists of three major components: (1) a geographic information system (GIS)‐based data analysis system to define spatial modeling units with resolution of 56 m to address spatial variability, (2) the biophysical and biogeochemical model Environmental Policy Integrated Climate (EPIC) applied in a spatially‐explicit way to predict biomass yield, GHG emissions, and other environmental impacts of different biofuel crops production systems, and (3) an evolutionary multiobjective optimization algorithm for exploring the trade‐offs between biofuel energy production and unintended ecosystem‐service responses. Simple examples illustrate the major functions of the SEIMF when applied to a nine‐county Regional Intensive Modeling Area (RIMA) in SW Michigan to (1) simulate biofuel crop production, (2) compare impacts of management practices and local ecosystem settings, and (3) optimize the spatial configuration of different biofuel production systems by balancing energy production and other ecosystem‐service variables. Potential applications of the SEIMF to support life cycle analysis and provide information on biodiversity evaluation and marginal‐land identification are also discussed. The SEIMF developed in this study is expected to provide a useful tool for scientists and decision makers to understand sustainability issues associated with the production of biofuels at local, regional, and national scales.  相似文献
