Cadmium is among the toxic and hazardous metal widely dispersed in the environment in high levels. Current studies have provided new insights into antioxidant properties of bioflavonoid which have emerged as probable therapeutic and nutraceutical agents. The present study is geared to investigate the possible role of Cymbopogon schoenanthus (L.) Spreng. (or Ethkher) on heavy metal cadmium (Cd) induced oxidative stress in mice. Mice were randomly divided into four groups and treated for 15 days as follows: group 1: normal control-treated (saline); group 2: Ethkher leaves extract-treated (100 mg/kg); group 3: cadmium chloride (CdCl2) treated; group 4: CdCl2 plus Ethkher leaves extract. The results showed a significant reduction in hemoglobin, RBC and hematocrit in cadmium-treated mice as compared to control. Exposure to Cd caused a significant increase in the number of white blood cells (P < 0.05) indicating the occurrence of systemic inflammation. The results of this study also revealed that the mice intoxicated with Cd showed a significant increase in bilirubin, aspartate aminotransferase (AST), alanine transaminase (ALT), alkaline phosphatase (ALP) and gamma-glutamyl transpeptidase (GGTP) activities. Cd intoxication leads to suppression in humoral immunity. However, pretreatment with Ethkher extract reversed almost all the abnormalities in the blood parameters showing noteworthy protection against cadmium induced toxicity in mice. The outcome of the present study revealed that the Ethkher possessed significant immunomodulatory activity and had a preventive effect on the hematological alterations in Cd intoxicated mice.  相似文献

目的: 分析镉(Cd)负荷不同时间对小鼠睾丸抗氧化酶的影响及维生素C(VC)的保护作用。方法: 清洁级雄性昆明小鼠72只分为4组(n=18):对照组、Cd组(CdCl2 3 mg/kg)、VC组(200 mg/kg)、VC(200 mg/kg)+ Cd(CdCl2 3 mg/kg)组,每日染毒1次,染毒1 d和3 d及同时补充VC保护,第1日和第3日染毒24 h后,每组取半数小鼠称重,取血清和睾丸组织;检测睾丸脏器系数,血清和睾丸组织丙二醛(MDA)、超氧化物歧化酶(SOD),及睾丸组织谷胱甘肽过氧化物酶(GSH-Px)、还原型谷胱甘肽(GSH)、氧化型谷胱甘肽(GSSG)及总谷胱甘肽(T-GSH)。结果: 与对照组比较,Cd组1 d和3 d小鼠体重和睾丸脏器系数下降;染毒3 d,Cd组小鼠血清SOD显著降低、MDA显著升高(P<0.05);Cd组1 d小鼠睾丸的SOD、GSH-Px、T-GSH及GSH/GSSG显著升高(P<0.05),而3 d的上述指标均显著降低(P<0.05),Cd组1 d和3 d MDA水平均显著升高(P<0.05);VC处理后减轻的程度有所降低。与Cd组比较,VC+ Cd组血清SOD和MDA水平在染毒3 d变化有显著性差异(P<0.05);VC+ Cd组在染毒1 d和3 d,小鼠睾丸的SOD、GSH-Px、T-GSH及GSH/GSSG水平变化有显著性差异(P<0.05),VC+ Cd组在染毒3 d睾丸的MDA水平显著降低(P<0.05)。与Cd组1 d比较,染毒3 d小鼠的血清SOD水平显著降低(P<0.05),睾丸指标变化也有显著性差异(P<0.05)。结论: VC处理可在一定程度上改善镉负荷小鼠的抗氧化功能,对睾丸氧化损伤具有保护作用。  相似文献

An important issue in critical care medicine is the identification of ways to protect the lungs from oxygen toxicity and reduce systemic oxidative stress in conditions requiring mechanical ventilation and high levels of oxygen. One way to prevent oxygen toxicity is to augment antioxidant enzyme activity in the respiratory system. The current study investigated the ability of aerosolized extracellular superoxide dismutase (EC-SOD) to protect the lungs from hyperoxic injury. Recombinant human EC-SOD (rhEC-SOD) was produced from a synthetic cassette constructed in the methylotrophic yeast Pichia pastoris. Female CD-1 mice were exposed in hyperoxia (FiO2>95%) to induce lung injury. The therapeutic effects of EC-SOD and copper-zinc SOD (CuZn-SOD) via an aerosol delivery system for lung injury and systemic oxidative stress at 24, 48, 72 and 96 h of hyperoxia were measured by bronchoalveolar lavage, wet/dry ratio, lung histology, and 8-oxo-2'-deoxyguanosine (8-oxo-dG) in lung and liver tissues. After exposure to hyperoxia, the wet/dry weight ratio remained stable before day 2 but increased significantly after day 3. The levels of oxidative biomarker 8-oxo-dG in the lung and liver were significantly decreased on day 2 (P<0.01) but the marker in the liver increased abruptly after day 3 of hyperoxia when the mortality increased. Treatment with aerosolized rhEC-SOD increased the survival rate at day 3 under hyperoxia to 95.8%, which was significantly higher than that of the control group (57.1%), albumin treated group (33.3%), and CuZn-SOD treated group (75%). The protective effects of EC-SOD against hyperoxia were further confirmed by reduced lung edema and systemic oxidative stress. Aerosolized EC-SOD protected mice against oxygen toxicity and reduced mortality in a hyperoxic model. The results encourage the use of an aerosol therapy with EC-SOD in intensive care units to reduce oxidative injury in patients with severe hypoxemic respiratory failure, including acute respiratory distress syndrome (ARDS).  相似文献

目的:研究纳米炭黑颗粒复合寒冷暴露对小鼠肺部组织结构及其氧化应激反应的影响。方法:将72只健康雄性C57BL/6小鼠随机分为6组:对照(Ctrl)组、单纯冷暴露(C)组、低剂量染毒(L)组、低剂量染毒复合冷暴露(LC)组、高剂量染毒(H)组、高剂量染毒复合冷暴露(HC)组。采用吸入式气管滴注染毒方式,一次性滴注纳米炭墨颗粒染毒液40 μl,浓度分别为0.45 mg/ml (L)和4.05 mg/ml (H)。冷暴露方式为4℃暴露,4 h/d,连续20 d。暴露结束24 h后称重、取样,进行相关指标测定。采用试剂盒法测定小鼠肺组织匀浆中超氧化物歧化酶(SOD)活力、谷胱甘肽过氧化物酶(GSH-Px)活力和丙二醛(MDA)含量;肺组织块HE染色,观察肺组织病理组织结构改变。结果:所有冷暴露处理组小鼠的体重均显著低于所有非冷暴露组(P<0.05),对照组及单纯染毒组小鼠体重均在实验开始14 d后明显升高(P<0.05),单纯冷暴露组与纳米炭黑颗粒染毒复合冷暴露组小鼠体重均在14 d后趋于稳定。HE检测结果表明,单纯纳米炭黑颗粒染毒组及染毒复合冷暴露组小鼠肺泡腔内均有黑色颗粒沉积,高剂量染毒复合冷暴露组可见肺泡结构破环,排列凌乱,有大量炎细胞浸润。与对照组相比,其余各组SOD活力均显著降低(P<0.05);高剂量染毒组及高剂量染毒复合冷暴露组GSH-Px活力明显低于对照组(P<0.01);与对照组相比,高剂量染毒组、低剂量染毒与高剂量染毒复合冷暴露组MDA含量显著升高(P<0.01)。两因素方差分析提示,随着染毒剂量的增加,SOD活力及GSH-Px活力显著降低(P<0.05);随着温度的降低,肺组织MDA含量显著升高(P<0.05),4℃间歇性冷暴露与纳米颗粒物暴露对肺组织SOD、GSH-Px活力及MDA含量的影响均无交互作用。结论:纳米炭黑颗粒复合寒冷暴露可导致小鼠肺部炎症反应加重,氧化应激水平升高。  相似文献

摘要 目的:探究微创胸腔闭式引流精密可控负压吸引术治疗原发性自发性气胸(PSP)的疗效及对患者疼痛、氧化应激反应的影响。方法:选取2019年1月~2021年1月我院收治的PSP患者120例,以随机数表法分成研究组与对照组,各自60例。给予对照组微创胸腔常规闭式引流治疗,研究组则给予微创胸腔闭式引流精密可控负压吸引术治疗。比较两组患者的临床疗效,术后6 h、1 d及3 d的疼痛评分(VAS),术前及术后1 d的血清氧化应激指标[超氧化物歧化酶(SOD)、丙二醛(MDA)]水平以及术后并发症发生情况。结果:研究组临床疗效的总有效率为91.67%,显著高于对照组的78.33%(P<0.05),且研究组患者术后6 h、术后1 d以及术后3 d的VAS评分均显著低于对照组(P<0.05)。术后研究组患者SOD水平显著高于对照组(P<0.05),同时MDA水平显著低于对照组(P<0.05)。另外研究组患者的总并发症发生率为3.33%,显著低于对照组的13.33%(P<0.05)。结论:微创胸腔闭式引流精密可控负压吸引术治疗PSP的疗效肯定,能减轻患者的疼痛程度及氧化应激反应,且能减少术后并发症的发生率。  相似文献

目的:研究肾虚育精方对肾精亏虚小鼠模型生殖激素及氧化应激水平的影响。方法:选择6周龄雄性健康级SD小鼠60只纳入本次研究。根据随机数字法将小鼠分成3组,分别为观察组、模型组及对照组,每组各20只。观察组及模型组的小鼠通过"房劳和惊恐复合伤肾法"建立肾精亏虚模型,对照组小鼠不建模。建模完成后观察组小鼠每日给予肾虚育精方的浓缩液干预,模型组及对照组则予以等量的生理盐水灌胃。干预21d后对比各组小鼠睾丸组织有关指标,包括Johnsen 10级积分、生精小管的直径及生精上皮厚度,小鼠生殖激素水平,包括血清卵泡刺激素(FSH)及睾酮(TEST),氧化应激指标水平,包括丙二醛(MDA)、过氧化物歧化酶(SOD)及总抗氧化能力(T-AOC)。结果:对照组与观察组的Johnsen 10级积分高于模型组,生精小管的直径及生精上皮厚度大于模型组,差异均有统计学意义(P0.05);观察组的生精上皮厚度明显大于对照组,差异有统计学意义(P0.05)。对照组与观察组小鼠的FSH及TEST水平均明显高于模型组,差异均有统计学意义(均P0.05)。对照组与观察组的MDA和SOD水平明显低于模型组,T-AOC水平明显高于模型组,差异均有统计学意义(P0.05)。结论:肾虚育精方能够提升肾精亏虚小鼠模型的生殖激素水平,并降低氧化应激水平,可将肾虚育精方应用于临床治疗。  相似文献

为了探究镉处理对大型溞(Daphnia magna)摄食能力的影响及其机制, 实验设置了3个镉浓度组(0.01、0.05和0.09 mg/L)、1个空白对照和2个处理时间(24h、48h), 研究镉处理对大型溞摄食率与滤水率、体内镉的蓄积量、总抗氧化能力(T-AOC)和脂质过氧化水平(MDA含量)以及乙酰胆碱酯酶(AChE)活性的影响。结果显示, 摄食率和滤水率均随着镉处理浓度的增加而逐渐降低, 当镉浓度为0.09 mg/L, 处理48h时, 摄食率和滤水率均显著或极显著低于对照组(P<0.05,P<0.01), 且摄食率与对照组相比降低了43.89%; 镉在大型溞体内的积累量随镉处理浓度的增加而逐渐升高; T-AOC和MDA含量随着镉浓度的增加而增高, 镉浓度0.09 mg/L时, T-AOC和MDA含量与对照组相比有显著性差异(P<0.05), 且T-AOC与摄食率之间呈负相关性, 相关系数R2分别为0.9521、0.9389; AChE活性随着镉浓度的增加而降低, 镉浓度为0.05、0.09 mg/L时AChE活性显著低于对照组(P<0.05)。这表明镉在体内的蓄积不仅造成了大型溞的氧化损伤, 而且引起了神经系统传导功能的异常, 导致了大型溞摄食能力受到抑制。  相似文献

实验旨在探索不同C/N生物絮团对急性铜暴露洛氏鱥免疫抑制、炎症反应与氧化应激的保护作用。挑选480尾洛氏鱥(Rhynchocypris lagowskii Dybowski)幼鱼(10±0.15) g,随机分成4组,每组3个重复,每个重复40尾鱼。对照组饲喂商品料(C/N 10.8﹕1),实验组选择葡萄糖为外添碳源调节C/N 15﹕1(Ⅱ组)、C/N 20﹕1(Ⅲ组)和C/N 25﹕1(Ⅳ组),生长实验为56d,之后进行为期96h的急性铜暴露胁迫实验。结果表明,各实验组中免疫、抗氧化酶活性与炎症因子含量随着C/N的增加先升高后下降。其中,与对照组相比,Ⅲ和Ⅳ组血清碱性磷酸酶(AKP)、酸性磷酸酶(ACP)、溶菌酶(LSZ)、一氧化氮合酶(NOS)、补体C3、C4和免疫球蛋白M(IgM)水平显著升高(P<0.05),而Ⅲ和Ⅳ组之间补体C3和C4水平差异不显著(P>0.05);Ⅲ和Ⅳ组血清过氧化氢酶(CAT)、抗坏血酸(ASA)、超氧化物歧化酶(SOD)、总抗氧化能力(T-AOC)、谷胱甘肽过氧化物酶(GSHPX)与谷胱甘肽还原酶(GR)酶活性显著高于对照组(P<0.05...  相似文献

目的:本研究是为了观察饮食补充锌减轻酒精性肝病损伤的作用及与HNF-4α的关系。方法:选用成年C57BL/6小鼠40只,按随机数字表分为4组(n=10):正常对照组、酒精中毒组、正常补锌组及酒精补锌组,用不同饮食喂养6个月处死,在正常补锌组和酒精补锌组小鼠饮用水中加入硫酸锌,使锌的含量达到75 mg/L。取各组小鼠肝组织进行病理切片及增殖细胞核抗原(PCNA)免疫组织化学染色,RT-PCR检测肝细胞核因子-4α(HNF-4α)含量,Western blot检测肝组织HNF-4α蛋白表达,检测"肝组织超氧化物歧化酶(SOD)活性及丙二醛(MDA)含量"。结果:酒精中毒组小鼠HNF-4α转录及表达均明显低于正常对照组,差异具有统计学意义(P<0.05),该组小鼠MDA含量增高,SOD活性下降与正常对照组相比差异有统计学意义(P<0.05);而酒精补锌组小鼠PCNA阳性肝细胞数目及HNF-4α蛋白表达水平明显高于酒精中毒组,差异有统计学意义(P<0.05),该组小鼠SOD活性增加,MDA下降,与酒精中毒组相比差异有统计学意义(P<0.05)。结论:长期酒精喂养导致小鼠氧化还原失衡,而补锌可逆转该状态。我们推测饮食补锌可能是通过增加HNF-4α的转录及表达而增强酒精喂养小鼠的肝再生,因此,饮食补锌可能对酒精性肝病有较好的影响。  相似文献

Experimental autoimmune encephalomyelitis (EAE) is a well-established animal model of human multiple sclerosis (MS). We have evaluated the role of oxidative and nitrosative stress, as the causal factors in the development of EAE, responsible for the damage of cardinal cellular components, such as lipids, proteins and nucleic acids, resulting in demyelination, axonal damage, and neuronal death. EAE was induced in female Sprague-Dawley rats, 3 months old (300±20 g), by immunization with myelin basic protein in combination with Complete Freund's adjuvant (CFA). The animals were divided into seven groups: control, EAE, CFA, EAE+aminoguanidine (AG), AG, EAE+N-acetyl-L-cysteine (NAC) and NAC. The animals were sacrificed 15 days after EAE induction, and the levels of nitrosative and oxidative stress were determined in 10% homogenate of the whole encephalitic mass. In EAE rats, brain NO production and MDA level were significantly increased (P<0.001) compared to the control values, whereas AG and NAC treatment decreased both parameters in EAE rats compared to EAE group (P<0.001). Glutathione (GSH) was reduced (P<0.001) in EAE rats in comparison with the control and CFA groups, but increased in EAE+AG and EAE+NAC group compared to the EAE group (P<0.01). Superoxide dismutase (SOD) activity was significantly decreased (P<0.001) in the EAE group compared to all other experimental groups. The clinical expression of EAE was significantly decreased (P<0.05) in the EAE groups treated with AG and NAC compared to EAE rats, during disease development. The obtained results prove an important role of oxidative and nitrosative stress in the pathogenesis of EAE, whereas AG and NAC protective effects offer new possibilities for a modified combined approach in MS therapy.  相似文献

Cadmium (Cd) is a well-known nephrotoxicant inducing kidney damage via oxidative stress. Since kidney is the critical target organ of Cd toxicity, this study was designed to evaluate the protective effects of onion (Allium cepa L.) and garlic (Allium sativum L.) aqueous extracts on Cd-induced renal oxidative stress in male Wistar rats. The control group received double distilled water alone and Cd group was challenged with 3CdSO4 · 8H2O (as Cd) (1.5 mg/100 g bw/day per oral) alone. Extract-treated groups were pre-treated with varied doses (0.5 ml and 1.0 ml/100 g bw/day per oral) of onion and/or garlic extract for 1 week after which they were co-treated with Cd (1.5 mg/100 g bw/day per oral) for 3 weeks. The results showed that the levels of renal lipid peroxidation (LPO) and glutathione-S transferase (GST) were significantly (P < 0.001) increased in rats that received Cd alone relative to the control group. More so, the levels of renal glutathione (GSH), superoxide dismutase (SOD), catalase (CAT) and Na+/K+-ATPase were significantly (P < 0.001) decreased in rats that received Cd alone. Treatment of Cd-intoxicated rats with varied doses of onion and/or garlic extract significantly (P < 0.05) restored the alterations in these parameters relative to the group that received Cd alone. While treatment with high dose of onion extract exerted a significant dose-dependent restoration of these parameters, treatment with high dose of garlic elicited a pro-oxidant effect, relative to their respective low dose. Our study suggests that onion and garlic extracts may exert their protective effects via reduction in LPO and enhanced antioxidant defense. These extracts may, therefore, be useful nutritional option in alleviating Cd-induced renal damage.  相似文献

目的:分析脑栓通联合丁苯酞治疗急性缺血性脑卒中(AIS)的临床价值。方法:选取我院收治的104例AIS患者为研究对象,采用随机数字表法将其分为对照组及观察组,每组各52例。两组均给予抗血小板凝集、控制血压、降糖、调脂等基础治疗,对照组在基础治疗基础上给予丁苯酞治疗,观察组则在对照组基础上予以脑栓通治疗。记录和比较两组的临床疗效、不良反应的发生情况,治疗前及治疗2周后神经功能缺损程度[美国国立卫生研究院卒中量表(NIHSS)]、血清炎性因子[超敏C反应蛋白(hs-CRP)、白细胞介素-8(IL-8)]水平、氧化应激指标[脂质过氧化物(LPO)、甘胱甘肽过氧化物酶(GSH-Px)]、血流动力学指标[大脑中动脉(MCA)收缩期血流速度(Vs)、平均血液流速(Vm)]的变化。结果:治疗后,观察组临床总有效率为96.15%,明显高于对照组(P<0.05)。两组治疗期间并发症发生率比较差异无统计学意义(P>0.05)。治疗2周后,两组NIHSS评分、血清hs-CRP、IL-8、LPO水平均较治疗前显著降低(P<0.05),GSH-Px、MCA Vs和Vm则较治疗前明显升高(P<0.05),且观察组以上指标的变化明显优于对照组(P<0.05)。结论:脑栓通联合丁苯酞治疗AIS具有较好的疗效及较高的安全性,可减轻机体炎症反应与氧化应激反应,改善脑血流灌注及神经功能。  相似文献

Several epidemiological studies have shown a positive correlation between daily increases in airborne particulate matter (PM) concentration and the occurrence of respiratory and cardiovascular diseases. Transition metals present in air PM were associated with adverse health effects after PM exposure. The aim of this work was to study lung O2 metabolism after an acute exposure to transition metal-coated nanoparticles (NPs). Female Swiss mice (25 g) were intranasally instilled with a suspension of silica NP containing Ni (II), Cd (II), Fe (III), or Cr (VI) at 0, 0.01, 0.05, 0.1, and 1.0 mg metal/kg body weight. Lung O2 consumption was found to be significantly increased after the exposure to most doses of Ni-NP and Fe-NP, and the 0.05 mg metal/kg body weight dose of Cr-NP, while no changes were observed for Cd-NP. Lucigenin chemiluminescence (as an indicator of NADPH oxidase (NOX) activity) was evaluated in lung homogenates. Only Ni-NP and Fe-NP have shown the ability to induce a significant increase in lucigenin chemiluminescence. In order to establish the possible occurrence of pulmonary oxidative stress, TBARS levels and the GSH/GSSG ratio were determined. The higher doses of Ni-NP and Fe-NP were able to induce an oxidative stress condition, as shown by changes in both TBARS levels and the GSH/GSSG ratio. Taken together, the present results show differential effects for all the metals tested. These findings emphasize the importance of transition metals present air PM in PM adverse health effects, and contribute to the understanding of the pathological mechanisms triggered by the exposure to environmental PM.  相似文献

为探究自噬抑制剂6-氨基-3-甲基腺嘌呤(3-methyladenine,3-MA)对损伤细胞氧化应激水平的影响,将3-MA作用于H2O2诱导的PC12细胞损伤模型,以自噬增强剂雷帕霉素(rapamycin,Rap)作为对照,探讨自噬与氧化应激的关系。测定线粒体的膜电位和细胞内的活性氧(reactive oxygen species, ROS)与丙二醛(malondialdehyde, MDA)含量,以及超氧化物歧化酶(superoxide dismutase,SOD)和过氧化氢酶(catalase,CAT)活性,评价损伤细胞的氧化应激状态。单丹(磺)酰戊二胺(monodansylcadaverine,MDC)染色,观察损伤细胞的自噬情况。蛋白质印迹分析损伤细胞中的自噬相关蛋白质LC3-II/LC3-I比值变化。实验结果显示:与正常组相比,H2O2损伤细胞的ROS水平上升到正常组的141%,MDA含量增加(P<0.001);CAT与SOD酶活力显著降低(P<0.001),差异均有统计学意义,证明损伤细胞氧化应激水平增加;MDC染色结果表明,H2O2组自噬明显增加。Western印迹结果表明,LC3-II/LC3-I值显著升高(P<0.05);与损伤组相比,3-MA组MDC染色结果表明,自噬水平降低。Western印迹结果表明,LC3-II/LC3-I值下降;细胞内ROS水平升高,增加到正常组的208%。MDA含量增加(P<0.001),CAT、SOD酶活力降低(P<0.001)。综上结果表明,自噬抑制剂可增加H2O2诱导的PC12细胞损伤模型的氧化应激水平,增加细胞凋亡。  相似文献

Objectives: The purpose of this study was to investigate oxidative stress in children with attention deficit hyperactivity disorder (ADHD).

Methods: Total oxidant status (TOS), total antioxidant status (TAS), paraxonase-1 (PON-1) and arylesterase (ARE) activity were measured in 76 children (44 boys, 32 girls) diagnosed with ADHD according to the DSM-IV and 78 healthy children (46 boys, 32 girls).

Results: Age and sex were similar between the groups (P?>?0.05). TOS and the oxidative stress index (OSI) were higher in the patient group than the control group (P?<?0.001). PON-1 (P?=?0.002), ARE (P?=?0.010) activity and TAS (P?<?0.001) were lower in the patient group than the control group.

Discussion: We found decreased PON-1, ARE activity and TAS, and increased TOS and OSI in children with ADHD. Our study showed that there is significantly increased oxidative stress in children with ADHD.  相似文献

The aim of this study was to assess the effects of subchronic exposure to cadmium (Cd) on the antioxidant defense system of red blood cells (RBCs) and lipid peroxide concentration in the plasma, as well as the possible protective role of zinc (Zn). For this purpose, 60 male Wistar rats (8 weeks old) were divided into three groups: the first group was exposed to Cd in the form of CdCl2, administered in five doses (each of 0.4 mg Cd/kg BW) on days 5, 10, 15, 20 and 25, giving a total dose of 2 mg Cd/kg BW, i.p.; the second group was simultaneously exposed to Zn and Cd with the same timeline and the same doses of Cd as the first group but with, in addition, injections of Zn in the form of ZnCl2, administered in doses of 0.8 mg Zn/kg BW, giving a total dose of 4 mg Zn/kg BW, i.p.; a control group received 0.5 mL of physiological saline in an identical manner.

It was shown that exposure to Cd induced a significant decrease (p<0.05) in superoxide dismutase (Zn/Cu SOD) and catalase (CAT) activities in RBCs. Increased lipid peroxide concentration, measured by thiobarbituric acid reactive substances (TBARS), was also observed in the plasma of cadmium-exposed rats. Cd had no effect on glutathione peroxidase (GSH-Px) activity. Zn administration had a beneficial effect on the Cd-induced decrease in Zn/Cu SOD activity (p<0.05) but not on CAT activity. Animals receiving Cd and Zn simultaneously had significantly (p<0.05) lower concentrations of lipid peroxides than rats exposed to Cd alone. Our results indicate that Cd causes oxidative stress and that Zn supply in conditions of exposure to Cd can partially protect against Cd-induced oxidative stress.  相似文献

The kidney has been regarded as a critical organ of toxicity induced by acute exposure to hexavalent chromium [Cr(VI)] compounds. Reactive intermediates and free radicals generated during reduction process might be responsible for Cr(VI) toxicity. In this study, the effects of pretreatment or posttreatment of taurine on Cr(VI)-induced oxidative stress and chromium accumulation in kidney tissue of Swiss albino mice were investigated. Single intraperitoneal (ip) potassium dichromate treatment (20 mgCr/kg), as Cr(VI) compound, significantly elevated the level of lipid peroxidation as compared with the control group (p<0.05). This was accompanied by significant decreases in nonprotein sulfhydryls (NPSH) level, superoxide dismutase (SOD), and catalase (CAT) enzyme activities as well as a significant chromium accumulation (p<0.05). Taurine administration (1 g/kg, ip) before or after Cr(VI) exposure resulted in reduction of lipid peroxidation levels and improvement in SOD enzyme activity (p<0.05). On the other hand, administration of the antioxidant before Cr(VI) exposure restored the NPSH level and CAT enzyme activity and also reduced tissue chromium levels (p<0.05), whereas postreatment had only slight effects on these parameters. In view of the results, taurine seems to exert some beneficial effects against Cr(VI)-induced oxidative stress and chromium accumulation in mice kidney tissue.  相似文献

目的:探讨血清微小RNA-21(mi R-21)水平与糖尿病肾病(DN)患者氧化应激的相关性。方法:选择2015年1月到2017年5月我院就诊的169例DN患者作为研究对象,其中DN分期早期组95例,中晚期组74例。另选同期在我院接受健康体检者90例作为对照组,对比各组血清mi R-21水平和氧化应激指标,分析患者血清mi R-21水平与氧化应激指标的相关性。结果:各组血清mi R-21水平相比差异有统计学意义(P0.05),中晚期DN组及早期DN组血清mi R-21水平均明显低于对照组,且中晚期DN组又明显低于早期DN组,差异均有统计学意义(P0.05)。各组氧化应激指标相比差异有统计学意义(P0.05),中晚期DN组及早期DN组丙二醛(MDA)、超氧化物歧化酶(SOD)、晚期氧化蛋白产物(AOPP)及NADPH氧化酶4(NOX4)均明显高于对照组,且中晚期DN组又明显高于早期DN组;而血红素氧合酶1(HO-1)明显低于对照组,且中晚期DN组又明显低于早期DN组,差异均有统计学意义(P0.05)。依据Spearman法分析相关性发现,血清miR-21水平与MDA、SOD、AOPP及NOX4均呈负相关(P0.05),与HO-1呈正相关(P0.05)。结论:血清mi R-21水平与DN患者氧化应激指标均具有明显的相关性,临床上可尝试将miR-21纳入到DN患者病情监测的指标体系中,从而有助于疾病的诊治及患者的预后。  相似文献

摘要 目的:探讨虎杖苷对老年小鼠术后认知功能障碍及海马氧化应激和神经炎症的影响。方法:C57BL/6J雄性老年小鼠,18月龄,体重24~28 g,随机分为4组:假手术组(Sham组)、术后认知功能障碍组(POCD组)、低剂量虎杖苷组(PD1组)、高剂量虎杖苷组(PD2组)。Sham组老年小鼠仅接受水合氯醛麻醉,不行外科手术;POCD组老年小鼠接受腹部手术以制备POCD老年动物模型;PD1组小鼠制备POCD模型,并于术后即刻、24 h和48 h分别腹腔注射低剂量虎杖苷25 mg/kg;PD2组小鼠制备POCD模型,并于术后即刻、24 h和48 h分别腹腔注射高剂量虎杖苷50 mg/kg。使用Morris水迷宫行为学实验评估老年小鼠术后认知功能,使用Western blot法检测术后小鼠海马Nrf2、HO-1、HMGB1、Iba-1蛋白水平,检测海马活性氧(ROS)、丙二醛(MDA)、超氧化物歧化酶(SOD)含量以反映术后海马氧化应激水平。结果:(1)与Sham组比较,POCD组老年小鼠术后逃离潜伏期显著增加(P<0.05),穿越平台次数和目标象限停留时间下降(P<0.05),海马Nrf2和HO-1蛋白表达水平明显降低(P<0.05),氧化应激产物ROS和MDA含量增加(P<0.05),抗氧化酶SOD活性降低(P<0.05),Iba-1和HMGB1蛋白表达水平增加(P<0.05);(2)与POCD组比较,PD1组和PD2组小鼠术后逃离潜伏期缩短(P<0.05),穿越平台次数和目标象限停留时间增多(P<0.05),海马Nrf2和HO-1蛋白表达水平升高(P<0.05),ROS和MDA含量减少(P<0.05),SOD活性增加(P<0.05),Iba-1和HMGB1蛋白表达减少(P<0.05)。结论:虎杖苷可减轻老年小鼠术后认知功能损伤,缓解术后海马氧化应激和神经炎症,其机制可能与促进Nrf2/HO-1信号通路有关。  相似文献
