The synthesis and characterization of monofluoroacetyl (MFAc) functionalized haptens are described. These were covalently bound to polypeptide carriers (bovine serum albumin and poly-D-lysine) by primary amine/succinimide ester or primary amine/acid chloride coupling. Epitopic densities of the resulting antigens were determined by both 19F n.m.r. and picryl sulfonic acid assays. 19F n.m.r. experiments defining the stability of MFAc ester and amide linkages as a function of media (including in vitro), time, and temperature are presented. These results indicate that MFAc functionalized antigens are well suited for further immunologic studies. 相似文献

Several metal-sugar conjugates have been prepared. Chelate coordination complexes of copper(II) and zinc(II) were prepared from salicylaldimine ligands derived from combinations of methyl 3,4,6-tri-O-acetyl-2-amino-2-deoxy-β-d-glucopyranoside or 1,3,4,6-tetra-O-acetyl-2-amino-2-deoxy-β-d-glucopyranose and salicylaldehyde. Similar Schiff's bases were prepared from chitosan and complexed to copper(II). Cross-polarisation-magic angle spinning 13C-n.m.r. spectroscopy showed that paramagnetic ions have profound effects on the resolution obtainable and, at high concentration, result in broad, featureless spectra. In contrast, diamagnetic ions have little effect on the isotropic chemical shifts of the sugar chelates. 相似文献

Electron-spin-resonance measurements have been made on single crystals of uracil-beta-D-arabinofuranoside, which were irradiated by 4-0 MeV electrons at 77 K. At low temperatures, two radicals have been identified, one attributed to a hydrogen abstraction from 05' in the sugar moiety and the other to a radical anion located on the pyrimidine ring. The former is very unstable and seems to act as a precursor to other unidentified radical species stable at 77K. At room temperature, the main resonance is due to hydrogen addition to C5 and is probably produced by protonation of the anion. This same radical is also produced by X-irradiation at room temperature. 相似文献

Monosubstituted [4-carboxy-2,6-dinitrophenyl-lysine]cytochromes c were investigated by n.m.r. and e.p.r. Modification of Lys-13 or Lys-72 in ferricytochrome c by 4-chloro-3,5-dinitrobenzoate yields either of two different conformers that are rapidly exchanging in the native form. The equilibrium involves small local changes in the conformation of Met-80 (the sixth ligand) and Phe-82, as a result of whether Lys-13 is the 'on' or 'off' position in the Lys-13--Glu-90 salt bridge. 相似文献

Fourier-transform infrared (F.t.-i.r.) and laser-Raman spectra have been obtained for solid guanine. The F.t.-i.r. spectrum of guanosine in the solid state was also recorded. The dependence of the yield of free radicals in gamma-irradiated, freeze-dried erythrocyte membranes on their haemoglobin content was studied. A non-monotonous relationship was found--different from that observed in mixtures of freeze-dried membranes and haemoglobin, which suggests the existence of radiation-energy transfer between the membranes and bound haemoglobin. 相似文献
