A Spirillum sp. and a Pseudomonas sp. possessing crossing substrate saturation curves for L-lactate were isolated from fresh water by chemostat enrichment. Their Ks and mumax values for L-lactate were: Spirillum sp., 23 micrometer and 0.35 h-1, respectively; Pseudomonas sp., 91 micrometer and 0.64 h-1, respectively. Under L-lactate limitation, pseudomonas sp. outgrew Spirillum s. at dilution rates (D) above 0.29 h-1, but the converse occurred at lower D values. The advantage of Spirillum sp. increased with decreasing D until, at D = 0.05 h-1 (i.e. L-lactate concentration of approximately 1 micrometer), Pseudomonas sp. was eliminated from the culture essentially as a non-growing population. In Spirillum sp. the Km for L-lactate transport (5.8 micrometer) was threefold lower than in Pseudomonas sp. (20 micrometer); Spirillum sp. also possessed a higher Vmax for the transport of this substrate. The surface to volume ratio was higher in Spirillum sp. and increased more markedly than in Pseudomonas sp. in response to decreasing D. Thus, a more efficient scavenging capacity contributes to the advantage of Spirillum sp. at low concentrations of the carbon source. Although most of the enzymes of L-lactate catabolism were more active in Pseudomonas sp., NADH oxidase activity was about twice as high in Spirillum sp.; and, unlike Pseudomonas sp., the cytochrome c content of this bacterium increased markedly with decreasing D. A more active and/or more efficient respiratory chain may therefore also play a role in the advantage of Spirillum sp. The other factors which appear to be involved include a lower energy of maintenance of Spirillum sp. [0.016 g L-lactate (g cell dry wt)-1 h-1 compared with 0.066 in Pseudomonas sp.] and a lower minimal growth rate.  相似文献

A flow cell-grown model consortium consisting of two organisms, Burkholderia sp. LB400 and Pseudomonas sp. B13(FR1), was studied. These bacteria have the potential to interact metabolically because Pseudomonas sp. B13(FR1) can metabolize chlorobenzoate produced by Burkholderia sp. LB400 when grown on chlorobiphenyl. The expected metabolic interactions in the consortium were demonstrated by high performance liquid chromatography (HPLC) analysis. The spatial structure of the consortium was studied by fluorescent in situ rRNA hybridization and scanning confocal laser microscopy. When the consortium was fed with medium containing a low concentration of chlorobiphenyl, microcolonies consisting of associated Burkholderia sp. LB400 and Pseudomonas sp. B13(FR1) bacteria were formed, and separate Pseudomonas sp. B13(FR1) microcolonies were evidently not formed. When the consortium was fed citrate, which can be metabolized by both species, the two species formed separate microcolonies. The structure development in the consortium was studied online using a gfp -tagged Pseudomonas sp. B13(FR1) derivative. After a shift in carbon source from citrate to a low concentration of chlorobiphenyl, movement of the Pseudomonas sp. B13(FR1) bacteria led to a change in the spatial structure of the consortium from the unassociated form towards the associated form within a few days. Experiments involving a gfp -based Pseudomonas sp. B13(FR1) growth activity reporter strain indicated that chlorobenzoate supporting growth of Pseudomonas sp. B13(FR1) is located close to the Burkholderia sp. LB400 microcolonies in chlorobiphenyl-grown consortia.  相似文献

A cellulose-degrading defined mixed culture (designated SF356) consisting of five bacterial strains (Clostridium straminisolvens CSK1, Clostridium sp. strain FG4, Pseudoxanthomonas sp. strain M1-3, Brevibacillus sp. strain M1-5, and Bordetella sp. strain M1-6) exhibited both functional and structural stability; namely, no change in cellulose-degrading efficiency was observed, and all members stably coexisted through 20 subcultures. In order to investigate the mechanisms responsible for the observed stability, "knockout communities" in which one of the members was eliminated from SF356 were constructed. The dynamics of the community structure and the cellulose degradation profiles of these mixed cultures were determined in order to evaluate the roles played by each eliminated member in situ and its impact on the other members of the community. Integration of each result gave the following estimates of the bacterial relationships. Synergistic relationships between an anaerobic cellulolytic bacterium (C. straminisolvens CSK1) and two strains of aerobic bacteria (Pseudoxanthomonas sp. strain M1-3 and Brevibacillus sp. strain M1-5) were observed; the aerobes introduced anaerobic conditions, and C. straminisolvens CSK1 supplied metabolites (acetate and glucose). In addition, there were negative relationships, such as the inhibition of cellulose degradation by producing excess amounts of acetic acid by Clostridium sp. strain FG4, and growth suppression of Bordetella sp. strain M1-6 by Brevibacillus sp. strain M1-5. The balance of the various types of relationships (both positive and negative) is thus considered to be essential for the stable coexistence of the members of this mixed culture.  相似文献

A pentachlorophenol (PCP)-mineralizing bacterium was isolated from polluted soil and identified as Pseudomonas sp. strain RA2. In batch cultures, Pseudomonas sp. strain RA2 used PCP as its sole source of carbon and energy and was capable of completely degrading this compound as indicated by radiotracer studies, stoichiometric release of chloride, and biomass formation. Pseudomonas sp. strain RA2 was able to mineralize a higher concentration of PCP (160 mg liter-1) than any previously reported PCP-degrading pseudomonad. At a PCP concentration of 200 mg liter-1, cell growth was completely inhibited and PCP was not degraded, although an active population of Pseudomonas sp. RA2 was still present in these cultures after 2 weeks. The inhibitory effect of PCP was partially attributable to its effect on the growth rate of Pseudomonas sp. strain RA2. The highest specific growth rate (mu = 0.09 h-1) was reached at a PCP concentration of 40 mg liter-1 but decreased at higher or lower PCP concentrations, with the lowest mu (0.05 h-1) occurring at 150 mg liter-1. Despite this reduction in growth rate, total biomass production was proportional to PCP concentration at all PCP concentrations degraded by Pseudomonas sp. RA2. In contrast, final cell density was reduced to below expected values at PCP concentrations greater than 100 mg liter-1. Diffusing wave spectroscopy has been used to measure the rheological behavior of pullulan (M(w) = 1 x 10(5)) aqueous solutions up to concentration of 40 g/dL. It was found that these solutions were mainly viscous, with the loss modulus G' higher than the elastic modulus G'. The plot of the specific viscosity eta(sp) as a function of pullulan concentration showed two critical concentrations c = 4 g/dL and c = 15 g/dL. For c < c, eta( sp) approximately c(1.25+/-0.05); for c < c < c, eta( sp) approximately c(2+/-0.05); and for c > c, eta( sp) approximately c(4.5+/-0.5). These results are in very good agreement with those reported in the literature.  相似文献

A freshwater Spirillum sp., which apparently belongs to a niche of low nutritional status (Matin & Veldkamp, 1978), accumulated poly-beta-hydroxybutyric acid (PHB) during lactate-limited growth in continuous culture. The PHB content varied in a complex manner with the dilution rate (D), but was greatest at the lowest D value examined: about 18% (w/w) at D = 0.025 h-1. It is not known what mechanism accounted for PHB accumulation during carbon-limited growth. The resistance of cultures of Spirillum sp. to starvation after growth at various D values was compared with that of a Pseudomonas sp. which appears to belong to relatively richer environments (Matin & Veldkamp, 1978) and does not accumulate PHB. In Spirillum sp., resistance correlated directly with the PHB content of the culture subjected to starvation, whereas in Pseudomonas sp. it increased with RNA content. Further, after growth at D = 0.03 to 0.05 h-1, the Spirillum sp. was much more resistant to starvation than was the Pseudomonas sp. Since the microflora of oligotrophic environments are probably often subjected to starvation conditions, PHB accumulation by Spirillum sp. during growth in such environments may assist survival. PHB in Spirillum sp. was rapidly degraded during starvation but it had no sparing effect on RNA degradation. It is not known how PHB enhanced resistance to starvation.  相似文献

The structure of the extracellular polysaccharide (EPS) produced by the Rhizobium sp. B strain isolated from atypical nodules on alfalfa has been determined using a combination of chemical and physical techniques (methylation analysis, high pH-anion exchange chromatography (HPAEC), mass spectrometry and 1-D and 2-D NMR spectroscopy). As opposed to the EPS from other strains of Rhizobium, the EPS from the sp. B strain contains D-Glc together with L-Rha and 2-deoxy-D-arabino-hexuronic acid. It is a polymer of a repeating unit having the following structure: --> 4)-beta-D-Glcp-(1 --> 4)-alpha-L-Rhap -(1 --> 3)-beta-D-Glcp-(1 --> 4)-2-deoxy-beta-D-GlcpA-(1 -->. A genomic library containing HindIII partial digests of Pseudomonas sp. strain AM1 DNA was constructed in the broad-host-range cosmid pVK100. PCT57, a Pseudomonas sp. strain AM1 methanol mutant deficient in malyl coenzyme A lyase activity, was complemented to a methanol-positive phenotype by mobilization of the pVK100 library into PCT57 recipients with the ColE1/RK2 mobilizing plasmid pRK2013. Six different complemented isolates all contained a recombinant plasmid carrying the same 19.6-kilobase-pair Pseudomonas sp. strain AM1 DNA insert. Subcloning and complementation analysis demonstrated that the gene deficient in PCT57 (mcl-1) was located in a 1.6-kilobase-pair region within a 7.4-kilobase-pair EcoRI-HindIII fragment.  相似文献

Biodegradation of benzo(a)pyrene by a newly isolated Fusarium sp   总被引:1,自引:0,他引:1  
Benzo(a)pyrene (BaP) is a five-ring polycyclic aromatic hydrocarbon produced by the incomplete combustion of organic materials. It is one of the priority pollutants listed by the US Environmental Protection Agency. This study describes a fungal isolate that is able to biodegrade benzo(a)pyrene. The filamentous fungus, isolated from leaves of Pterocarpus macrocarpus Kurz., was identified as a Fusarium sp. (strain E033). Fusarium sp. E033 was able to survive in the presence of benzo(a)pyrene concentrations up to 1.2 mM (300 mg L(-1)). Biodegradation experiments using 0.4 mM (100 mg L(-1)) benzo(a)pyrene demonstrated that Fusarium sp. E033 was able to degrade 65-70% of the initial benzo(a)pyrene provided, and two transformation products, a dihydroxy dihydro-benzo(a)pyrene and a benzo(a)pyrene-quinone, were detected within 30 days of incubation at 32 degrees C. The factors affecting biodegradation efficiency were also investigated. While increasing aeration promoted better fungal growth and benzo(a)pyrene biodegradation, increasing the glucose concentration from 5 to 50 mM had an adverse effect on biodegradation. Ethanol and methanol, provided at 5 mM to increase benzo(a)pyrene water solubility, increased the fungal biomass yield but did not promote degradation. The Fusarium sp. E033 isolated in this study can tolerate and degrade relatively high concentrations of benzo(a)pyrene, suggesting its potential application in benzo(a)pyrene bioremediation.  相似文献

Thermococcus sp. strain CL1 is a hyperthermophilic, anaerobic, and heterotrophic archaeon isolated from a Paralvinella sp. polychaete worm living on an active deep-sea hydrothermal sulfide chimney on the Cleft Segment of the Juan de Fuca Ridge. To further understand the distinct characteristics of this archaeon at the genome level, its genome was completely sequenced and analyzed. Here, we announce the complete genome sequence (1,950,313 bp) of Thermococcus sp. strain CL1, with a focus on H(2)- and energy-producing capabilities and its amino acid biosynthesis and acquisition in an extreme habitat.  相似文献

A characteristic of N2-fixing cyanobacteria in symbiotic associations appears to be release of N2-derived NH4+. The specific activity of the primary ammonium-assimilating enzyme, glutamine synthetase (GS), was found to be three- to fourfold lower in Nostoc sp. strain 7801 grown in symbiotic association with the bryophyte Anthoceros punctatus than in free-living Nostoc sp. strain 7801. Quantitative immunological assays with antisera against GS purified from Nostoc sp. strain 7801 and from Escherichia coli indicated that similar amounts of the GS protein were present in symbiotic (50 micrograms mg-1) and free-living (68 micrograms mg-1) cultures. The conclusion from these experiments is that GS is regulated by a posttranslational mechanism in Anthoceros-associated Nostoc sp. strain 7801. However, the results of comparative catalytic and immunological experiments between N2- and NH4+-grown free-living Nostoc sp. strain 7801 implied control of GS synthesis. A correlation was not observed between the level of GS expression and the extent of symbiotic heterocyst differentiation in Nostoc sp. strain 7801 associated with A. punctatus.  相似文献

Helminths of the spiral intestine of neotropical freshwater stingrays (Potamotrygonidae) were examined in Peru for the first time. The stingrays examined for helminths included Paratrygon aiereba, Potamotrygon motoro, and Potamotrygon cf. castexi. Present in P. aiereba were the cestodes Nandocestus guariticus (Marques, Brooks, and Lasso, 2001) n. gen. n. comb., Rhinebothrium copianullum n. sp., Rhinebothrium sp. 1, Rhinebothroides sp., Potamotrygonocestus cf. fitzgeraldae, and 1 species each of Cucullanus and Rhabdochona. Nandocestus n. gen. is erected to house N. guariticus, which is formally transferred from Anindobothrium Marques, Brooks, and Lasso, 2001. The new genus is unique among phyllobothriids in its possession of circummedullary vitelline follicles and a submarginal genital pore, in combination with bothridia with a single apical sucker and marginal bothridial loculi. The helminths of P. motoro included the cestodes Paraoncomegas araya, Acanthobothrium peruviense n. sp., Acanthobothrium cf. ramiroi, Rhinebothrium sp. 1, Rhinebothroides sp. 1, Potamotrygonocestus sp., the nematode Brevimulticaecum regoi, a species of Cucullanus, and a species of the digenean superfamily Diplostomoidea. The helminths of P. cf. castexi included the cestodes P. araya, N. guariticus n. gen. n. comb., Acanthobothrium cf. peruviense, Potamotrygonocestus sp., Rhinebothrium sp. 1, Rhinebothroides sp. 2, the nematode species Echinocephalus daileyi and B. regoi, 1 species each of Cucullanus, Rhabdochona, and Procamallanus, and a species of the digenean superfamily Hemiuroidea. All taxa were examined via light microscopy; the cestode taxa were also examined using scanning electron microscopy. Each helminth species recorded in this study is a first report from Peru. The study suggests that the diversity and host specificity of the cestodes in potamotrygonid stingrays may be greater than previously thought. The known numbers of genera and species of tetraphyllidean cestodes parasitizing neotropical freshwater stingrays are now 6 and 22, respectively.  相似文献

The involvement of ligninolytic and cellulolytic enzymes, such as laccase, lignin peroxidase, manganese peroxidase, carboxymethylcellulase (CMCase), and filter paper activity (FPA), in the decomposition process of leaf litter driven by 6 soil-inhabiting fungi imperfecti was studied under solid-state fermentations. All the tested fungi exhibited varied production profiles of lignocellulolytic enzymes and each caused different losses in total organic matter (TOM) during decomposition. Based on the results, the 6 fungi could be divided into 2 functional groups: Group 1 includes Alternaria sp., Penicillium sp., Acremonium sp., and Trichoderma sp., and Group 2 includes Pestalotiopsis sp. and Aspergillus fumigatus. Group 1, with higher CMCase and FPA activities, showed a higher decomposition rate than the fungi of Group 2 over the first 16 d, and thereafter the cellulolytic activities and decomposition rate slowed down. Group 2 showed the maximum and significantly higher CMCase and FPA activities than those of the Group 1 fungi during the later days. This, combined with the much higher laccase activity, produced a synergistic reaction that led to a much faster average mass loss rate. These results suggest that the fungi of Group 1 are efficient decomposers of cellulose and that the fungi of Group 2 are efficient decomposers of lignocellulose. During cultivation, Pestalotiopsis sp. produced an appreciable amount of laccase activity (0.56+/-0.09 U/ml) without the addition of inducers and caused a loss in TOM of 38.2%+/-3.0%, suggesting that it has high potential to be a new efficient laccase-producing fungus.  相似文献

Seven strains of fungi were isolated from activated sludge and identified as Mucor sp., Geotrichum sp., Trichosporon sp., Candida sp., and Trichoderma sp. by 28S rDNA D2 region sequences analysis. The structures of the main ceramide monosaccharides (CMSs) from these fungi were identified as glucosylceramide (GlcCer) consisting of ceramide moieties of 9-methyl-octadeca-sphingadienine (9-Me d18:2), with 2-hydroxyhexadecanoate (h16:0) (Mucor sp. and Geotrichum sp.), 2-hydroxyoctadecanoate (h18:0) (Trichosporon sp. and Candida sp.), and 2-hydroxyoctadecenoate (h18:1) (Trichoderma sp.). Seasonal changes in glycosphingolipids in activated sludge suggest the possibility that microbial flora in activated sludge changes with the seasons, and that fungi adaptable to low temperatures dominate in the cold period, resulting in the maintenance of stable effluent quality. Mucor sp., Geotrichum sp., and Candida sp. satisfactorily reduced the BOD of synthetic sewage at 10 degrees C. These results indicate that fungi in activated sludge can contribute to wastewater treatment in cold conditions.  相似文献

Identification, characterization, and end-product synthesis patterns were analyzed in a newly identified mesophilic, anaerobic Clostridium sp. strain URNW, capable of producing hydrogen (H?) and ethanol. Metabolic profiling was used to characterize putative end-product synthesis pathways of the Clostridium sp. strain URNW, which was found to grow on cellobiose; on hexose sugars, such as glucose, sucrose, and mannose; and on sugar alcohols, like mannitol and sorbitol. When grown in batch cultures on 2 g cellobiose·L?1, Clostridium sp. strain URNW showed a cell generation time of 1.5 h, and the major end-products were H2, formate, carbon dioxide (CO?), lactate, butyrate, acetate, pyruvate, and ethanol. The total volumetric H? production was 14.2 mmol·(L culture)?1 and the total production of ethanol was 0.4 mmol·(L culture)?1. The maximum yield of H? was 1.3 mol·(mol glucose equivalent)?1 at a carbon recovery of 94%. The specific production rates of H?, CO?, and ethanol were 0.45, 0.13, and 0.003 mol·h?1·(g dry cell mass)-1, respectively. BLAST analyses of 16S rDNA and chaperonin 60 (cpn60) sequences from Clostridium sp. strain URNW revealed a 98% nucleotide sequence identity with the 16S rDNA and cpn60 sequences from Clostridium intestinale ATCC 49213. Phylogenetic analyses placed Clostridium sp. strain URNW within the butyrate-synthesizing clostridia.  相似文献

The present study was carried out in order to examine and characterize the bidirectional hydrogenase in the cyanobacterium Nostoc sp. strain PCC 73102. Southern hybridizations with the probes Av1 and Av3 (hoxY and hoxH, bidirectional hydrogenase small and large subunits, respectively) revealed the occurrence of corresponding sequences in Anabaena variabilis (control), Anabaena sp. strain PCC 7120, and Nostoc muscorum but not in Nostoc sp. strain PCC 73102. As a control, hybridizations with the probe hup2 (hupL, uptake hydrogenase large subunit) demonstrated the presence of a corresponding gene in all the cyanobacteria tested, including Nostoc sp. strain PCC 73102. Moreover, with three different growth media, a bidirectional enzyme that was functional in vivo was observed in N. muscorum, Anabaena sp. strain PCC 7120, and A. variabilis, whereas Nostoc sp. strain PCC 73102 consistently lacked any detectable in vivo activity. Similar results were obtained when assaying for the presence of an enzyme that is functional in vitro. Native polyacrylamide gel electrophoresis followed by in situ hydrogenase activity staining was used to demonstrate the presence or absence of a functional enzyme. Again, bands corresponding to hydrogenase activity were observed for N. muscorum, Anabaena sp. strain PCC 7120, and A. variabilis but not for Nostoc sp. strain PCC 73102. In conclusion, we were unable to detect a bidirectional hydrogenase in Nostoc sp. strain PCC 73102 with specific physiological and molecular techniques. The same techniques clearly showed the presence of an inducible bidirectional enzyme and corresponding structural genes in N. muscorum, Anabaena sp. strain PCC 7120, and A. variabilis. Hence, Nostoc sp. strain PCC 73102 seems to be an unusual cyanobacterium and an interesting candidate for future biotechnological applications.  相似文献

Biosurfactants are surface active substances which reduce interfacial tension and are produced or excreted at the microbial cell surface. We evaluated the biosurfactant production by Aspergillus fumigatus and Phialemonium sp. in solid state processes using fixed-bed column reactors. We evaluated two media, rice husks alone (simple support) and rice husks plus defatted rice bran (complex support), both enriched with either soy oil or diesel oil. The highest water-in-oil emulsifying activity (EAw/o) obtained was 7.36 EU g(-1) produced by A. fumigatus growing on complex support enriched with soy oil and supplied with air at a rate of 60 mL g(-1) h(-1), while Phialemonium sp. had a maximum production of 6.11 EU g(-1) using the simple support with diesel oil and an aeration rate of 120 mL g(-1) h(-1). The highest oil-in-water emulsifying activity (EAo/w) was 12.21 EU g(-1) produced by Phialemonium sp. on the complex support enriched with diesel oil and at an aeration rate of 60 mL g(-1) h(-1), while A. fumigatus produced a maximum EAo/w of 10.98 EU g(-1) when growing on the complex support with no additional carbon source and an aeration rate of 60 mL g(-1) 1 h(-1).  相似文献
