In this study we demonstrated through analytic considerations and numerical studies that the mitochondrial fatty-acid β-oxidation can exhibit bistable-hysteresis behavior. In an experimentally validated computational model we identified a specific region in the parameter space in which two distinct stable and one unstable steady state could be attained with different fluxes. The two stable states were referred to as low-flux (disease) and high-flux (healthy) state. By a modular kinetic approach we traced the origin and causes of the bistability back to the distributive kinetics and the conservation of CoA, in particular in the last rounds of the β-oxidation. We then extended the model to investigate various interventions that may confer health benefits by activating the pathway, including (i) activation of the last enzyme MCKAT via its endogenous regulator p46-SHC protein, (ii) addition of a thioesterase (an acyl-CoA hydrolysing enzyme) as a safety valve, and (iii) concomitant activation of a number of upstream and downstream enzymes by short-chain fatty-acids (SCFA), metabolites that are produced from nutritional fibers in the gut. A high concentration of SCFAs, thioesterase activity, and inhibition of the p46Shc protein led to a disappearance of the bistability, leaving only the high-flux state. A better understanding of the switch behavior of the mitochondrial fatty-acid oxidation process between a low- and a high-flux state may lead to dietary and pharmacological intervention in the treatment or prevention of obesity and or non-alcoholic fatty-liver disease.  相似文献

This paper reviews the model of the control of mitochondrial substrate oxidation by Ca2+ ions. The mechanism is the activation by Ca2+ of four mitochondrial dehydrogenases, viz: glycerol 3-phosphate dehydrogenase, the pyruvate dehydrogenase multienzyme complex (PDH), NAD-linked isocitrate dehydrogenase (NAD-IDH) and 2-oxoglutarate dehydrogenase (OGDH). This results in the increase, or near-maintenance, of mitochondrial NADH/NAD ratios in the activated state, depending upon the tissue and the degree of "downstream" activation by Ca2+, likely at the level of the F1F0 ATP-ase. Higher values of the redox span of the respiratory chain allow for greatly increased fluxes through oxidative phosphorylation with a minimal drop in protonmotive force and phosphorylation potential. As PDH, NAD-IDH and OGDH are all located within the inner mitochondrial membrane, it is changes in matrix free Ca2+ ( [Ca2+]m ) which act as a signal to these activities. In this article, we review recent work in which ([Ca2+]m) is measured in cells and tissues, using different techniques, with special emphasis on the question of the degree of damping of ([Ca2+]m) relative to changes in cytosol free Ca2+ in cells with rapid transients in cytosol Ca2+, e.g. cardiac myocytes. Further, we put forward the point of view that the failure of mitochondrial energy transduction to keep pace with cellular energy needs in some forms of heart failure may involve a failure of ([Ca2+]m) to be raised adequately to allow the activation of the dehydrogenases. We present new data to show that this is so in cardiac myocytes isolated from animals suffering from chronic, atreptozocin-induced diabetes. This raises the possibility of therapy based upon partial inhibition of mitochondrial Ca2+ efflux pathways, thereby raising ([Ca2+]m) at a given, time-average value of cytosol free Ca2+.  相似文献

The way in which the huge Australian parasite fauna is described (discovery and naming) is the subject of this address. The approach to the task has never been well-organised so that a few groups of parasites are now relatively well-known because of the efforts of small groups of workers who have made sustained efforts in these groups, but equally some host-parasite systems have been almost completely ignored in that no worker has ever given them sustained attention. A high proportion of Australian parasites have been described by international workers. The sustaining of interest in a group of parasites over a long period is the key to real progress being made. The nature of the organisation of Australian science presently means that few positions are available for parasite taxonomists and funding for taxonomic research is scarce. Thus, parasite taxonomy (like the taxonomy of many groups of Australian plants and animals) can only be considered to be in crisis.  相似文献

Abstract

For the first time, the densities and diversity of microorganisms developed on ocean gliders were investigated using flow cytometry and Illumina MiSeq sequencing of 16S and 18S rRNA genes. Ocean gliders are autonomous buoyancy-driven underwater vehicles, equipped with sensors continuously recording physical, chemical, and biological parameters. Microbial biofilms were investigated on unprotected parts of the glider and surfaces coated with base, biocidal and chitosan paints. Biofilms on the glider were exposed to periodical oscillations of salinity, oxygen, temperature, pressure, depth and light, due to periodic ascending and descending of the vehicle. Among the unprotected surfaces, the highest microbial abundance was observed on the bottom of the glider’s body, while the lowest density was recorded on the glider’s nose. Antifouling paints had the lowest densities of microorganisms. Multidimensional analysis showed that the microbial communities formed on unprotected parts of the glider were significantly different from those on biocidal paint and in seawater.  相似文献

Without genetic variation, species cannot cope with changing environments, and evolution does not proceed. In endangered species, adaptive potential may be eroded by decreased population sizes and processes that further reduce gene flow such as philopatry and local adaptations. Here, we focused on the philopatric and endangered loggerhead sea turtle (Caretta caretta) nesting in Cape Verde as a model system to investigate the link between adaptive potential and philopatry. We produced a dataset of three complementary genomic regions to investigate female philopatric behaviour (mitochondrial DNA), male-mediated gene flow (microsatellites) and adaptive potential (major histocompatibility complex, MHC). Results revealed genetically distinct nesting colonies, indicating remarkably small-scale philopatric behaviour of females. Furthermore, these colonies also harboured local pools of MHC alleles, especially at the margins of the population''s distribution, which are therefore important reserves of additional diversity for the population. Meanwhile, directional male-mediated gene flow from the margins of distribution sustains the adaptive potential for the entire rookery. We therefore present the first evidence for a positive association between philopatry and locally adapted genomic regions. Contrary to expectation, we propose that philopatry conserves a high adaptive potential at the margins of a distribution, while asymmetric gene flow maintains genetic connectivity with the rest of the population.  相似文献

This study evaluated how the edge effect influences the structuration of fruit-feeding butterfly assemblages in swamp forest fragments of the subtropical Atlantic Forest, Southern Brazil. Sampling was carried out twice in 10 fragments using baited traps placed in sampling units both at the forest edge and 50 m within the forest interior, with the habitats being defined by a set of environmental variables. Richness and abundance were higher for edge habitats with an effect of temperature depending on humidity and luminosity. The subfamily/tribe composition of fruit-feeding butterflies was segregated between edge and interior and was predicted by wind speed and the interaction between humidity and luminosity. Fifty meters within the forest interior is not sufficient to cause homogenization of butterfly composition between the edge and interior of swamp forest fragments, indicating distinct assemblages in each habitat. The interior harboured forest-loving butterfly groups while the edge harboured generalist sun-loving and common butterflies associated with disturbed areas, suggesting resistance to the effects of habitat fragmentation. We highlight the importance of using fruit-feeding butterfly groups, instead of species, to evaluate edge effects. We also suggest that a heterogeneous matrix with native habitats and distinct semi-natural land-use systems be maintained to manage subtropical areas by increasing connectivity within the landscape. Considering the impacts that the Atlantic Forest suffers, increased knowledge of modifications caused at small and regional scales is crucial for the maintenance of ecological processes and represents a tool for conservation planning and environmental agendas.  相似文献

Cannibalism, the act of preying on and consuming a conspecific, is taxonomically widespread, and putatively important in the wild, particularly in teleost fishes. Nonetheless, most studies of cannibalism in fishes have been performed in the laboratory. Here, we test four predictions for the evolution of cannibalism by conducting one of the largest assessments of cannibalism in the wild to date coupled with a mesocosm experiment. Focusing on mosquitofishes and guppies, we examined 17 species (11,946 individuals) across 189 populations in the wild, spanning both native and invasive ranges and including disparate types of habitats. We found cannibalism to be quite rare in the wild: most populations and species showed no evidence of cannibalism, and the prevalence of cannibalism was typically less than 5% within populations when it occurred. Most victims were juveniles (94%; only half of these appeared to have been newborn offspring), with the remaining 6% of victims being adult males. Females exhibited more cannibalism than males, but this was only partially explained by their larger body size, suggesting greater energetic requirements of reproduction likely play a role as well. We found no evidence that dispersal‐limited environments had a lower prevalence of cannibalism, but prevalence was greater in populations with higher conspecific densities, suggesting that more intense resource competition drives cannibalistic behavior. Supporting this conclusion, our mesocosm experiment revealed that cannibalism prevalence increased with higher conspecific density and lower resource levels but was not associated with juvenile density or strongly influenced by predation risk. We suggest that cannibalism in livebearing fishes is rare in the wild because preying on conspecifics is energetically costly and only becomes worth the effort when competition for other food is intense. Due to the artificially reduced cost of capturing conspecifics within confined spaces, cannibalism in captive settings can be much more frequent.  相似文献

Objective: An impaired fatty acid handling in skeletal muscle may be involved in the development of insulin resistance and diabetes mellitus type 2 (DM2). We investigated muscle fatty acid metabolism in glucose‐intolerant men (impaired glucose tolerance (IGT)), a prediabetic state, relative to BMI‐matched control men (normal glucose tolerance (NGT)) during fasting and after a meal, because most people in the western society are in the fed state most of the day. Methods and Procedures: Skeletal muscle free fatty acid (FFA) uptake and oxidation were studied using the stable isotope tracer [2,2‐2H]‐palmitate and muscle indirect calorimetry in the forearm model during fasting and after a mixed meal (33 energy % (E%) carbohydrates, 61 E% fat). Intramyocellular triglycerides (IMTGs) were monitored with 1H‐magnetic resonance spectroscopy. IGT men were re‐examined after weight loss (?15% of body weight (BW)). Results: The postprandial increase in forearm muscle respiratory quotient (RQ) was blunted in IGT compared to NGT, but improved after weight loss. Weight loss also improved fasting‐fat oxidation and tended to decrease IMTGs (P = 0.08). No differences were found in fasting and postprandial forearm muscle fatty acid uptake between NGT and IGT, or in IGT before and after weight loss. Discussion: The ability to switch from fat oxidation to carbohydrate oxidation after a meal is already impaired in the prediabetic state, suggesting this may be an early factor in the development toward DM2. This impaired ability to regulate fat oxidation during fasting and after a meal (impaired metabolic flexibility) can be (partly) reversed by weight loss.  相似文献

A significant proportion of standard metabolic rate is devoted to driving mitochondrial proton leak, and this futile cycle may be a site of metabolic control during hibernation. To determine if the proton leak pathway is decreased during metabolic depression related to hibernation, mitochondria were isolated from liver and skeletal muscle of nonhibernating (active) and hibernating arctic ground squirrels (Spermophilus parryii). At an assay temperature of 37 degrees C, state 3 and state 4 respiration rates and state 4 membrane potential were significantly depressed in liver mitochondria isolated from hibernators. In contrast, state 3 and state 4 respiration rates and membrane potentials were unchanged during hibernation in skeletal muscle mitochondria. The decrease in oxygen consumption of liver mitochondria was achieved by reduced activity of the set of reactions generating the proton gradient but not by a lowered proton permeability. These results suggest that mitochondrial proton conductance is unchanged during hibernation and that the reduced metabolism in hibernators is a partial consequence of tissue-specific depression of substrate oxidation.  相似文献

Methods of calculating the distributions of the time to coalescence depend on the underlying model of population demography. In particular, the models assuming deterministic evolution of population size may not be applicable to populations evolving stochastically. Therefore the study of coalescence models involving stochastic demography is important for applications. One interesting approach which includes stochasticity is the O’Connell limit theory of genealogy in branching processes. Our paper explores how many generations are needed for the limiting distributions of O’Connell to become adequate approximations of exact distributions. We perform extensive simulations of slightly supercritical branching processes and compare the results to the O’Connell limits. Coalescent computations under the Wright-Fisher model are compared with limiting O’Connell results and with full genealogy-based predictions. These results are used to estimate the age of the so-called mitochondrial Eve, i.e., the root of the mitochondrial polymorphisms of the modern humans based on the DNA from humans and Neanderthal fossils.  相似文献

This paper reviews broad geographical patterns in the species composition of breeding woodland bird communities from Ireland to eastern Europe and outlines how processes affecting woodland birds in Britain and Ireland may differ from those operating in mainland Europe. Bird communities in British and Irish woodlands consist of a subset of the species occurring within European forests at similar latitudes. The occurrence of virtually all groups of forest birds is lower in Britain, and strikingly lower in Ireland, than in other temperate areas of mainland Europe. This phenomenon appears to form part of a west–east gradient in species diversity and is probably not just a consequence of insularity. Across this gradient there appears to be broad geographical constancy in the types (taxonomic, ecological and life-history groups) of species present. There is considerable spatial variation in habitat use by forest species within Europe. Some species in Britain probably use habitats in different ways to elsewhere for reasons related to competition, predation and historical adaptation to landscape change. Several species appear to reach the limits of their geographical ranges within Britain (i.e. in the absence of physical barriers). We suggest that range contraction of one of these species, the Common Nightingale Luscinia megarhynchos , in Britain may partially reflect redistribution into the highest quality areas in response to a wider population decline. It is argued that conclusions drawn from studies of forest birds in Britain do not necessarily apply in other regions and vice versa. There is a need for large-scale studies in Europe of the spatial variation in organization of forest bird assemblages, habitat use and the genetic structure of populations.  相似文献

Economically impoverished women in Cali, Colombia, have restricted access to food in a city where food is abundant. Ethnographic observations, interviews and 2 day food records were used to better understand the coping strategies used by a group of these women (n = 85) to maintain adequate levels of energy intake. Anthropometric indicators of nutritional status were normal for the group. Interview data revealed that the ability to purchase food was a concern for 58% of the women. When faced with a restricted ability to purchase food, the women indicated they made compromises in meal composition, reduced portion sizes, and/or reduced the number of meals. They also relied on relatives, friends, neighbors, store credit, or local government programs for access to food. Changes in meal composition were identified in 17.1% of all diet records (n = 509). Low energy intake (defined as energy intake ≤ 1.27 × BMR) was identified in 17.1% of all diet records. Carbohydrate consumption was significantly greater on low-energy intake days. The adequate nutritional status of this group of women suggests that their coping strategies are usually adequate to maintain energy intake, but the presence of uncertainty, the frequency of compromises in diet composition, and the frequency of low-energy intake days suggest that these women are at risk for undernutrition.
