The riboflavin-binding hexamerin (RbH) and arylphorin (ArH) were compared as storage reservoirs for adult development in Hyalophora cecropia. The two hexamerins were metabolically labeled with [3H]leucine and [35S]methionine, isolated by column chromatography, and separately injected into pupae whose diapause had been terminated by chilling. By the time of eclosion at least 98% of both hexamerins had been cleared from the hemolymph. Every reproductive and somatic tissue tested contained trichloroacetic acid-precipitable label; consistent differences between the two hexamerins were not detected in the distribution of their label to these tissues. While incorporation of intact hexamerins was not ruled out, hydrolysis and reincorporation of the liberated amino acids were indicated by label in vitellogenin and lipophorin, and by differences in 35S/3H ratios, which ranged from over 1.0 in chorions to 0.4 in wings, as compared with 0.75 in the injected hexamerins. Injection of [35S,3H]RbH from H. cecropia into A. luna, a species in the same subfamily whose pupae lack this hexamerin, resulted in a pattern of isotope incorporation similar to that yielded by RbH in the donor species. In neither species was there indication of a developing adult tissue that distinguished between RbH and ArH as precursor reservoirs for morphogenesis. This equivalence helps explain how many species of Lepidoptera are able to complete metamorphosis and reproduce without expressing an RbH gene. Evidence is also presented that ArH stored in the fat body protein granules during pupation may be utilized differently from that remaining in pupal hemolymph.  相似文献

As in many Lepidoptera, Plutella xylostella adults do not feed on protein and females must use accumulated reserves to supply vitellogenin synthesis. Storage proteins were quantified in females and males from the late larval stage through day 4 of adult life. The level of storage protein peaked in the early pupal stage, with females having about twice as much as males. In males, the level fell through pupal development and dropped to a trace by one day after eclosion. In females, level of storage proteins fell until eclosion, and then rose dramatically within four hours after the molt to about 2/3 of the original peak level. This post-eclosion increase, which has not been reported previously in insects, suggests that adult females synthesize hexamerins to resequester amino acids. Subsequently, the level of storage proteins fell as vitellogenin appeared and eggs were laid. The ability to synthesize and sequester amino acids as storage proteins during the adult stage has wide-ranging implication for protein management in insects, particularly those that are long-lived and have flexible schedules of reproduction.  相似文献

Hexamerins are large hemolymph-proteins that accumulate during the late larval stages of insects. Hexamerins have emerged from hemocyanin, but have lost the ability to bind oxygen. Hexamerins are mainly considered as storage proteins for non-feeding stages, but may also have other functions, e.g. in cuticle formation, transport and immune response. The genome of the hornworm Manduca sexta harbors six hexamerin genes. Two of them code for arylphorins (Msex2.01690, Msex2.15504) and two genes correspond to a methionine-rich hexamerin (Msex2.10735) and a moderately methionine-rich hexamerin (Msex2.01694), respectively. Two other genes do not correspond to any known hexamerin and distantly resemble the arylphorins (Msex2.01691, Msex2.01693). Five of the six hexamerin genes are clustered within ∼45 kb on scaffold 00023, which shows conserved synteny in various lepidopteran genomes. The methionine-rich hexamerin gene is located at a distinct site. M. sexta and other Lepidoptera have lost the riboflavin-binding hexamerin. With the exception of Msex2.01691, which displays low mRNA levels throughout the life cycle, all hexamerins are most highly expressed during pre-wandering phase of the 5th larval instar of M. sexta, supporting their role as storage proteins. Notably, Msex2.01691 is most highly expressed in the brain, suggesting a divergent function. Phylogenetic analyses showed that hexamerin evolution basically follows insect systematics. Lepidoptera display an unparalleled diversity of hexamerins, which exceeds that of other hexapod orders. In contrast to previous analyses, the lepidopteran hexamerins were found monophyletic. Five distinct types of hexamerins have been identified in this order, which differ in terms of amino acid composition and evolutionary history: i. the arylphorins, which are rich in aromatic amino acids (∼20% phenylalanine and tyrosine), ii. the distantly related arylphorin-like hexamerins, iii. the methionine-rich hexamerins, iv. the moderately methionine rich hexamerins, and v. the riboflavin-binding hexamerins.  相似文献

Neem, Azadirachta indica A. Juss. (Meliaceae), seed oil (NSO) added to meridic diet at concentrations as low as 0.016% reduced pupation and prevented adult eclosion of obliquebanded leafroller, Choristoneura rosaceana (Harris) (Lepidoptera: Tortricidae). At a rate of 0.0016%, NSO reduced the fitness of C. rosaceana, resulting in longer developmental times, lower adult eclosion rates, and reduced egg production compared with controls. The neem insecticide Margosan-O TM produced comparable results based on concentrations of the most biologically active constituent, azadirachtin. Pupation was completely inhibited at concentrations of 0.25% and 1.0% for larvae exposed at 5th or 6th instar, respectively; rates as low as 0.016% reduced pupal weights and adult eclosion rates. For larvae transferred to treated diet at 5th instar, physical abnormalities in the wings of adults occurred at a rate of 0.004% NSO and increased with increasing treatment rates. NSO at concentrations as high as 2.0% was not antifeedant to neonate larvae, based on 24 and 48 h choice test bioassays, when incorporated into a meridic diet.  相似文献

We identified a single hexameric storage protein in the grasshopper, Schistocerca americana, and monitored its abundance through the last larval instar and up until reproductive competence in adults of both sexes. This storage hexamerin, termed Schistocerca americana Persistent Storage Protein (saPSP) was the most abundant soluble protein in both larvae and adults. In both sexes, saPSP abundance started out low at the onset of the last larval instar and accumulated during feeding, peaking just prior to molting. Adults of both sexes contained significant amounts of saPSP after eclosion. In adult males, saPSP content dropped continuously after eclosion and was lowest once individuals reached reproductive maturity. In contrast, adult females depleted saPSP reserves during the first days of adulthood, but subsequently accumulated significant saPSP stores. In adult females, saPSP stores peaked just prior to the completion of egg provisioning. Given the overall patterns of abundance, saPSP has functions in both larvae and adults. In addition, the observed pattern of storage hexamerin accumulation differs from patterns of accumulation in the other known grasshoppers, Locusta migratoria and Romalea microptera, suggesting that significant functional diversity has evolved in storage hexamerins among the grasshoppers.  相似文献

Sulfate conjugation of 20-hydroxyecdysone and related ecdysteroids was studies by a radiometric assay. The formation of 20-hydroxyecdysone 35sulfate from PAP35S (3′phosphoadenosine 5′phosphosulfate) proceeded linearly for 15 min at a pH optimum of 8.4. The apparent Km values for PAP35S and 20-hydroxyecdysone were 1.29 and 24.6 μM, respectively. The overall sulfate conjugation of 20-hydroxyecdysone was also demonstrated with ATP, Mg2+ and sodium 35sulfate. The sulfotransferase activity showed a peak at puparium formation and a progressive increase after adult eclosion. The specific activity of the newly emerged female pupae was higher than that of the males. The reverse pattern was however observed in the adult mosquitoes where the activity of the male adult was about 2 times higher. The likely sites on the ecdysteroid molecule for sulfate conjugation are discussed.  相似文献

Evolution of arthropod hemocyanins and insect storage proteins (hexamerins)   总被引:6,自引:2,他引:4  
Crustacean and cheliceratan hemocyanins (oxygen-transport proteins) and insect hexamerins (storage proteins) are homologous gene products, although the latter do not bind oxygen and do not possess the copper- binding histidines present in the hemocyanins. An alignment of 19 amino acid sequences of hemocyanin subunits and insect hexamerins was made, based on the conservation of elements of secondary structure observed in X-ray structures of two hemocyanin subunits. The alignment was analyzed using parsimony and neighbor-joining methods. Results provide strong indications for grouping together the sequences of the 2 crustacean hemocyanin subunits, the 5 cheliceratan hemocyanin subunits, and the 12 insect hexamerins. Within the insect clade, four methionine- rich proteins, four arylphorins, and two juvenile hormone-suppressible proteins from Lepidoptera, as well as two dipteran proteins, form four separate groups. In the absence of an outgroup sequence, it is not possible to present information about the ancestral state from which these proteins are derived. Although this family of proteins clearly consists of homologous gene products, there remain striking differences in gene organization and site of biosynthesis of the proteins within the cell. Because studies on 18S and 12S rRNA sequences indicate a rather close relationship between insects and crustaceans, we propose that hemocyanin is the ancestral arthropod protein and that insect hexamerins lost their copper-binding capability after divergence of the insects from the crustaceans.   相似文献

Simple rearing experiments were conducted to address two questions relevant to understanding how generalist lepidopteran herbivores interact with alien plants. Yellow-striped armyworm (Spodoptera ornithogalli), luna moth (Actias luna), bagworm (Thyridopteryx ephemeraeformis) and white-marked tussock moth (Orgyia leucostigma) were reared from egg to 5th instar on excised foliage in the laboratory to determine the degree to which highly polyphagous lepidopteran herbivores are physiologically capable of surviving and developing on the suite of alien plants naturalized in the mid-Atlantic. Actias luna larvae from a single population were similarly reared on a representative of each of the 25 native plant genera recorded as hosts for this species to compare the diet breadth of a local population with that listed over the entire geographic range of the species. With few exceptions, all four generalists either quickly starved or grew at an unsustainably low rate on alien foliage. Actias luna larvae survived for 18 days on only 44% of the native plants recorded as hosts over the entire range of this insect and thrived on only 7%. The data suggest that (1) alien plants are unlikely to produce as much generalist insect biomass as the native plants they replace and (2) Lepidoptera that qualify as generalists when host breadth is considered over their entire geographic range may express a far more specialized diet within local populations. Both of these conclusions support the hypothesis that alien plant invasions may seriously disrupt terrestrial food webs by reducing the insect biomass required by insectivores in higher trophic levels.  相似文献

Dipteran arylphorin receptors, insect hexamerins, cheliceratan and crustacean hemocyanins, and crustacean and insect tyrosinases display significant sequence similarities. We have undertaken a systematic comparison of primary and secondary structures of these proteins. On the basis of multiple sequence alignments the phylogeny of these proteins was investigated. Hexamerin subunits, hemocyanin subunits, and tyrosinases share extensive similarities throughout the entire amino acid sequence. Our studies suggest the origin of arthropod hemocyanins from ancient tyrosinase-like proteins. Insect hexamerins likely evolved from hemocyanins of ancient crustaceans, supporting the proposed sister-group position of these subphyla. Arylphorin receptors, responsible for incorporation of hexamerins into the larval fat body of diptera, are related to hexamerins, hemocyanins, and tyrosinase. The receptor sequences display extensive similarities to the first and third domains of hemocyanins and hexamerins. In the middle region only limited amino acid conservation was observed. Elements important for hexamer formation are deleted in the receptors. Phylogenetic analysis indicated that dipteran arylphorin receptors diverged from ancient hexamerins, probably early in insect evolution. Correspondence to: T. Burmester  相似文献

  • 4 Daily egg number is not correlated with body size. In the central range of body size, egg weight is also not correlated with body size. However, exceptionally large or small females lay respectively heavier or lighter eggs than average.
  • 5 A simple trade-off between offspring size and number does not occur within females on a daily basis, or among females averaged over their lifespans.
  • 6 Fat body resources are depleted at a rate independent of body size.

Yeast prions are superb models for understanding the mechanisms of self‐perpetuating protein aggregates formation. [PSI+] stands among the most documented yeast prions and results from self‐assembly of the translation termination factor Sup35p into protein fibrils. A plethora of cellular factors were shown to affect [PSI+] formation and propagation. Clearance of Sup35p prion particles is however poorly understood and documented. Here, we investigated the role of the proteasome in the degradation of Sup35p and in [PSI+] prion propagation. We found that cells lacking the RPN4 gene, which have reduced intracellular proteasome pools, accumulated Sup35p and have defects in [PSI+] formation and propagation. Sup35p is degraded in vitro by the 26S and 20S proteasomes in a ubiquitin‐independent manner, generating an array of amyloidogenic peptides derived from its prion‐domain. We also demonstrate the formation of a proteasome‐resistant fragment spanning residues 83–685 which is devoid of the prion‐domain that is essential for [PSI+] propagation. Most important was the finding that the 26S and 20S proteasomes degrade Sup35p fibrils in vitro and abolish their infectivity. Our results point to an overlooked role of the proteasome in clearing toxic protein aggregates, and have important implications for a better understanding of the life cycle of infectious protein assemblies.  相似文献

The hemolymph of diapausing larvae of the stem borer, Busseola fusca Fuller (Lepidoptera: Noctuidae), contains an electrophoretically distinct protein band on nondenaturing polyacrylamide gels. The protein, called the Busseola diapause protein (BDP), was purified by a combination of density gradient ultracentrifugation, gel permeation, and affinity chromatography. It is a high molecular weight protein (Mr ~5 × 105; pl = 6.1) that is composed of two subunits, I (Mr ~88,000 ± 4,000) and II (Mr ~79,000 ± 1,000), which are not linked by disulfide bridges. The protein contains both lipids (2%) as well as covalently bound carbohydrates (1%). The inability to stain the fluorescein isothiocyanate-conjugated concanavalin A (FITC-Con A) suggests that the carbohydrate moiety of BDP is not of the high mannose type. Amino acid analysis showed a high tyrosine plus phenylalanine content (16 mol%). Labeling studies using [35S]-methionine showed that de novo synthesis by the fat body tissue occurs only in diapausing larval insects. It is proposed that the BDP could serve a storage function by providing the amino acids needed for the synthesis of pupal and adult structures.  相似文献

Many endoparasitoids develop successfully within a range of host instars. Parasitoid survival is highest when parasitism is initiated in earlier host instars, due to age-related changes in internal (physiological) host defences. Most studies examining fitness-related costs associated with differences in host instar have concentrated on the parasitoid, ignoring the effects of parasitism on the development of surviving hosts that have encapsulated parasitoid eggs. A laboratory experiment was undertaken examining fitness-related costs associated with encapsulation of Venturia canescens (Hymenoptera: Ichneumonidae) eggs by fifth (L5) instar larvae of Corcyra cephalonica (Lepidoptera: Pyralidae). Growth and development of both host and parasitoid were monitored in C. cephalonica larvae containing 0, 1, 2, or 4 parasitoid eggs. Adult size and fecundity of C. cephalonica did not vary with the number of eggs per host. However, there was a distinct increase in host mortality with egg number, although most parasitoids emerged from hosts containing a single egg. The most dramatic effect on the host was a highly significant increase in development time from parasitism to adult eclosion, with hosts containing 4 parasitoid eggs taking over 2.5 days longer to complete development than unparasitized larvae. The egg-to-adult development time and size of adult V. canescens did not vary with egg number per host, as demonstrated in a previous experiment using a different host (Plodia interpunctella). The results described here show that there are fitness-related costs to the host associated with resistance to parasitism.  相似文献

Parasitoid wasps have long been considered excellent organisms in studies examining the evolution of reproductive and life-history strategies. In examining the lifetime reproductive success of parasitoids in the laboratory, most investigations have provided the insects with excess hosts and food, where they exist in a relatively constraint-free environment. Importantly, these conditions may not accurately reflect the true heterogeneity of natural systems, where suitable hosts and food sources are likely to be limiting. This study examines the influence of differences in host and food availability on reproductive and life-history parameters in an asexual strain of the solitary endoparasitoid, Venturia canescens (Hymenoptera: Ichneumonidae). Lifetime reproductive success in V. canescens was measured in response to temporal variations in host and food (honey solution) access. Cohorts of parasitoids were provided with 200 fifth-instar larvae of the Indian meal moth, Plodia interpunctella (Lepidoptera: Pyralidae), and food for variable periods daily after eclosion. V. canescens is synovigenic, and host-deprived wasps continued to mature eggs over the first few days after eclosion until the egg storage capacity was reached in the oviducts. When these wasps were subsequently provided with hosts, oogenesis resumed and continued until later in adult life. Constantly fed wasps lived longer and produced more progeny than wasps from cohorts which were alternately fed and starved or were starved from eclosion. Moreover, wasps with constant host and food access produced most progeny early in life and usually experienced prolonged periods of postreproductive survival. In contrast, the reproductive period of wasps with limited host access was more evenly distributed throughout the adult life. Consequently, the cumulative progeny production by V. canescens with constant food access was fairly uniform irrespective of host availability. Longevity and fecundity in V. canescens were positively correlated with adult size. However, variable host access had little effect on the longevity of wasps which were constantly supplied with honey. Over the first 2 days of adult life, variation in food access also had no effect on progeny production by V. canescens. We argue that manipulating temporal host and food access to parasitoids in the laboratory more closely approximates natural conditions, where these resources are likely to be spatially separated. Moreover, our findings suggest that many highly synovigenic parasitoids like V. canescens, which produce microtype (=hydropic) eggs, have a considerably higher reproductive potential than ovary dissections have revealed. Our findings are discussed in relation to life-history evolution in the parasitic Hymenoptera.  相似文献

MaleAchroia grisella (Lepidoptera: Pyralidae) produce ultrasonic advertisement signals for pair formation. The signals are generated by wing-fanning and consist of pairs of approximately 100-μs pulses of 70- to 130-kHz sound. In a previous study, playback experiments showed that femaleA. grisella preferred signals whose pulses were louder, longer, and delivered at a higher rate and included longer silent (asynchrony) intervals within pairs. To understand the variation and repeatability of these signal characters, acoustic signals of 18A. grisella males were recorded twice daily on the first 4 days and the seventh day after adult eclosion. ANOVA and MANOVA revealed significant among-individual and among-age variation for all signal characters. Peak amplitude, average amplitude, rate, and pulse length peaked 2–3 days after eclosion and then decreased. Asynchrony interval, however, continued to increase as males aged. Repeatabilities of peak amplitude, average amplitude, rate, and asynchrony interval were all relatively high (>0.5).  相似文献

The effects of high plasma concentrations of homocystine and methionine on the free amino acids of brain have been examined. Incorporation of the label from [35S]methionine into the free amino acid pools of rabbit brain was enhanced in response to high plasma homocystine or high plasma homocystine and mcthionine. Under comparable conditions a marked decrease in the incorporation of the label from [14C]glycine into the free pool was observed. The corresponding incorporation of 35S and 14C into brain proteins parallelled the results obtained with incorporation into the free pools of amino acids. Amino acid analyses of the free amino acid pools of rabbit brain revealed a general decrease in the concentration of amino acids in response to high plasma homocystine or high plasma homocystine and methionine. Inhibition of protein synthesis which occurs under the above experimental conditions is a general phenomenon. myelin and other brain fractions being equally affected. The decrease in concentration of brain amino acids also results in a diminution in concentration of the neurotransmitters GABA, dopamine and noradrenaline. The possible relationship of the observed changes to homocystinuria is discussed.  相似文献

Hexamerins are hemolymph-proteins, which are mainly considered as storage proteins for non-feeding stages, and also undertake other roles during insect development and growth, however the characterization of hexamerin proteins in Spodoptera exigua is less understood. In this study five new hexamerin genes were identified and a total seven hexamerin genes were reported in S. exigua. These hexamerins contain the typical domains of hemocyanin at the N-terminal, C-terminal and in the middle of their protein sequences. These genes are mainly expressed in fat body, and the signal peptide sequences at their N-terminal of protein sequences can drive the expressed protein to excrete into hemolymph after synthesis. The phylogenetic analysis and amine acid composition revealed S. exigua express five different types of hexamerins: 1) Storage protein rich in methionine residue (MRSP), 2) Storage protein moderately rich in methionine (MMRSP), 3) Hexamerin with high composition of aromatic amino acids (Arylphorin), 4) Arylphorin-like hexamerin, and 5) Riboflavin-binding hexamerin (RbH). The phylogenetic pattern combined with the comparison of conserved histidine residues in copper binding sites of hexamerins revealed basal position of RbH and the evolutionary pathway in lepidopteran hexamerins. Finally, the induction expression of hexamerins by insecticide, lambda-cyhalothrin, were analyzed, results showed that lambda-cyhalothrin exposure may down-regulate their expression. This study increased the gene number of hexamerin to seven, and reported their expression and structural characterizations, the finding will facilitate the understand of hexamerin in other insects.  相似文献

Knowledge about interactions among biological control agents, used in sequence or simultaneously, may help to increase their efficiency, or in the case of conservation biological control, to avoid the impairment of efficiency. The aim of this study was to evaluate the interaction between two isolates of Beauveria bassiana (Bals.) Vuill. (Hypocreales) (Unioeste 47 and Unioeste 57) and Trichogramma pretiosum Riley (Hymenoptera: Trichogrammatidae), varying the timing of sprays of the fungus. Isolates were sprayed at 109 conidia ml?1 upon cards (1 × 5 cm; i.e., 2 × 105 conidia mm?2) with eggs of Anagasta kuehniella Zeller (Lepidoptera: Pyralidae) and the control group was sprayed with sterile distilled water with Tween® 80 (0.01%). First, the percentage of parasitized eggs was assessed in a choice test: females of T. pretiosum were confined with two cards, one of them treated with either of the fungus isolates, the other a water‐treated control. Second, parasitism was compared on cards sprayed at various time intervals pre‐ vs. post‐parasitism: the number of parasitized eggs was assessed, as well as the parasitoid's emergence percentage, egg‐adult period, offspring sex ratio, and the longevity of parasitizing females. Trichogramma pretiosum preferred to parasitize control eggs (65 and 72%) and parasitization of the treated eggs did not differ between fungus isolates: 35% (Unioeste 47) vs. 28% (Unioeste 57). Spraying before or after parasitizing did not affect the number of parasitized eggs, nor offspring sex ratio; however, both treatments affected the percentage of emerged adults, egg‐adult period, and longevity. These findings suggested that isolates did not interfere in the longevity of parasitizing females. When the fungus was sprayed on the various development stages of the parasitoid within the host egg, or when it was offered to the parasitoid at different times after spraying, differences in adults' longevity, parasitism, emergency period from egg to adult, and sex ratio were observed. Beauveria bassiana may be used in conjunction with this parasitoid in a program of pest control.  相似文献

Changes in the size of intracellular nitrogen pools and the potential feedback by these pools on maximum N uptake (NH4+ and NO3?) rates were determined for Chaetomorpha linum (Müller) Kützing grown sequentially under nutrient-saturating and nutrient-limiting conditions. The size of individual pools in N-sufficient algae could be ranked as residual organic N (RON) comprised mainly of amino acids and amino compounds > protein N > NO3? > NH4+ > chlorophyll N. When the external N supply was removed, growth rates remained high and individual N pools were depleted at exponential rates that reflected both dilution of existing pools by the addition of new biomass from growth and movement between the pools. Calculated fluxes between the tissue N pools showed that the protein pool increased throughout the N depletion period and thus did not serve a storage function. RON was the largest storage reserve; nitrate was the second largest, but more temporary, storage pool that was depleted within 10 days. Upon N resupply, the RON pool increased 3 × faster than either the inorganic or protein pools, suggesting that protein synthesis was the rate-limiting step in N assimilation and caused a buildup of intermediate storage compounds. Maximum uptake rates for both NH4+ and NO3? varied inversely with macroalgal N status and appeared to be controlled by changes in small intracellular N pools. Uptake of NO3? showed an initial lag phase, but the initial uptake of NH4+ was enhanced and was present only when the intracellular NH4+ pool was depleted in the absence of an external N supply. A strong negative correlation between the RON pool size and maximum assimilation uptake rates for both NH4+ and NO3? suggested a feedback control on assimilation uptake by the buildup and depletion of organic compounds. Enhanced uptake and the accumulation of N as simple organic compounds or nitrate both provide a temporary mechanism to buffer against the asynchrony of N supply and demand in C. linum.  相似文献
