Sperm morphology was evaluated (using Blom classification) in 44 ejaculates of 11 bulls and 44 ejaculates of 11 boars. Significant differences in sperm morphology were found between bulls and boars. In addition, the correlations between frequency of morphological changes and morphometrical traits of boar spermatozoa were demonstrated. More morphological anomalies of spermatozoa were recorded in ejaculates containing longer spermatozoa.  相似文献

During seasonal anoestrus (long-days), oestradiol can effectively inhibit the pulsatile secretion of luteinizing hormone (LH) in sheep. The aim of our trial was to determine whether the same regulatory mechanism exists in the pig. Altogether, 20 ovariectomized and oestradiol-implanted gilts (16 domestic pigs, 4 European wild boars) were randomly allocated to two treatment groups. The first group was kept under a short-day light-dark cycle of 8L:16D, and the second group under a long-day light regime of 16L:8D. After a 6-week treatment period, blood samples were taken at 20-min intervals for 12h. After sampling, the light regimens were switched. Sampling was then repeated following another 6 weeks of treatment. In both treatment groups, 2.3 LH pulses occurred every 12h. The basal LH level was 0.7+/-0.4 ng/ml for the short-day group and 1.0+/-0.5 ng/ml for the long-day group. The mean LH level was 0.9+/-0.4 and 1.3+/-0.6 ng/ml and the LH pulse amplitude 0.5+/-0.4 and 0.6+/-0.5 ng/ml, respectively. The basal and mean LH levels were therefore lower in short-day gilts (P<0.05), while LH pulse amplitude and frequency remained unaffected by treatment. In conclusion, the 6-week period under two different light regimes resulted in higher basal LH concentration in long-day gilts but was not able to produce changes in LH frequency in prepubertal gilts.  相似文献

Crop species are known to hybridize spontaneously with wild relatives, but few studies have characterized the performance of hybrids at various genealogies, life stages and environments. A group of cultivar-like individuals and potential hybrids were observed in a roadside population of wild chicory plants in Denmark. Seeds were collected from all reproductive plants and grown in a common garden experiment, and their morphological and genetic compositions were analysed. Intermediate plants were identified as hybrids and comprised various backcross and F(n) combinations. A genotypic hybrid index (HI), spanning from wild-like to cultivar-like, was highly correlated to a morphological index. Plant survival, growth and reproduction were evaluated and compared to the genotypic HI. Overall, cultivar-like and intermediate plants grew larger than wild-like plants, flowered longer, and produced more flowers and seeds. The common garden included a nutrient gradient. At higher nutrient levels, intermediate and cultivar-like plants produced more flowers and seeds than wild-like plants, whereas this effect was less pronounced at lower nutrient levels. During winter, small rodents consumed roots of cultivar-like and intermediate plants preferentially. Thus, cultivated and wild chicory are able to hybridize spontaneously, producing hybrid offspring of several generations that may reproduce more effectively than their wild parent, but herbivory and poor environmental conditions may negatively affect their fitness.  相似文献

 The development of transgenic plants has heightened concern about the possible escape of genetically engineered material into the wild. Hybridization between crops and their wild relatives provides a mechanism by which this could occur. While hybridization has been documented between several crops and wild or weedy relatives, little is known about the persistence of cultivar genes in wild populations in the generations following hybridization. Wild and weedy sunflowers occur sympatrically with cultivated sunflowers throughout much of the cultivation range, and hybridization is known to occur. We surveyed two cultivar-specific RAPD markers in 2700 progeny in a naturally occurring population of wild Helianthus annuus over five generations following a single generation of hybridization with the cultivar. Moderate levels of gene flow were detected in the first generation (42% hybrids at the crop margin) and cultivar allele frequencies did not significantly decline over four subsequent generations. These results indicate that gene flow from cultivated into wild populations of sunflowers can result in the long-term establishment of cultivar alleles in wild populations. Furthermore, we conclude that neutral or favorable transgenes have the potential to escape and persist in wild sunflower populations. Received: 1 November 1996/Accepted: 17 January 1997  相似文献

The structure of musculus longissimus lumborum (m.l.l.) was evaluated with respect to proportion of three muscle fiber types and their diameters, in gilts and barrows of wild boar/domestic pig hybrids. The experiment was carried out on 29 hybrids (21 barrows and 8 gilts) which originated from crossing Duroc sows with wild boar (Sus scrofa ferus). The diameter of the "red" fibres in barrows was 48.42 microm, significantly greater than in gilts--43.71 microm. The diameters of the remaining two types of fibres in barrows and gilts were similar and amounted to: "intermediate" fibres--39.08 and 40.79 microm, "white" fibres--51.42 and 51.03, respectively. In respect to sex no statistical differences in proportion of any types of fibers were found, which in barrows came to: "red"--12.93%, "intermediate"--30.17% and "white"--57.32%, and in gilts to: 12.50, 27.04 and 60.45%, respectively. In conclusion, in m.l.l. of wild boar/domestic pig hybrids, smaller diameters of all muscle fibers types were identified as well as a higher proportion of intermediate fibers in comparison to m.l.l. of various pig breeds. An increased proportion of the intermediate fibers probably results from the interaction between factors such as age (8-months), physical activity (open-air for running) and the influence of wild boar genes.  相似文献

In crosses of the wild pig (Sus scrofa attila Thomas) with the domestic pig a transferrin variant, Tf I, was detected, electrophoretic mobility of which was slightly faster than the mobility of the variant Tf A. From the results of starch gel electrophoresis, isolation, neuraminidase treatment, autoradiography, and genetic analysis of several families, it can be concluded that the Tf I variant is genetically controlled by the allele Tf1. Thus the number of alleles in the transferrin system of the pig has increased to six ( TfI, TfATfB, TfC, TPD and TfE ).  相似文献

Mitochondrial DNA (mtDNA) sequences (574 bp) of 30 Vietnamese pigs (large and small) were examined and compared with those of 61 haplotypes from wild boars and domestic pigs from various locations in Asia. The large Vietnamese pigs had genetic links to Ryukyu wild boars in southern Japan. The small Vietnamese pigs were closely related to other East Asian domestic pigs. These results indicate that Vietnamese pigs are genetically diverse and may be descendents of wild and domestic pigs from other regions of Asia.  相似文献

Frost could potentially limit the survival ofhybrids between cultivated and wild carrots innatural habitats, and thereby the likelihoodthat (trans)genes spread from the crop to wildplants. To test this, cultivated, wild carrotsand their hybrids were exposed to differentfrost treatments. Summary Cytogenetic relationships between two cultivars of Cajanus cajan and six species of Atylosia were investigated. Of the 12 cross combinations obtained, only seven could be studied. Meiotic chromosome pairing, pollen and ovule fertility in parental species and four F1 hybrids were near normal. Some meiotic abnormalities were observed in the Fls: A. lineata x A. scarabaeoides, A. scarabaeoides x A. sericea and C. cajan (UPAS 120) x A. trinervia, indicating varying degrees of chromosomal and genic differences between these species. These observations suggested that A. cajanifolia is the closest wild relative of C. cajan, followed by A. scarabaeoides, A. albicans and A. trinervia. Genomic in situ hybridization was used to study Triticum x Dasypyrum wide hybrids and derived lines. A cytogenetic investigation was carried out in progenies of (i) amphiploids derived from T. turgidum var. durum (T. durum; 2n = 14; genomes AABB) x D. villosum (2n = 14; genome VV), (ii) three-parental hybrids (T. durum x D. villosum) x T. aestivum (2n = 42, genomes A'A'B'B'D'D'), and (iii) T. aestivum aneuploid lines carrying D. villosum chromosomes or chromatin. The amphiploids derived from T. durum x D. villosum showed a stable chromosomal constitution, made up of 14 V chromosomes, 14 chromosomes carrying the wheat A genome and 14 chromosomes carrying the B genome. High karyological instability was observed in the progenies of three-parental hybrids ([T. durum x D. villosum] x T. aestivum). Plants having the expected 14 A chromosomes, 14 B chromosomes, 7 D chromosomes, and 7 V chromosomes were rather rare (4.5%). Many progeny plants (45.5%) had the hexaploid wheat genome with 42 chromosomes and lacked any detectable D. villosum chromatin. Other plants (50%) had 14 A chromosomes and 14 B chromosomes, plus variable numbers of D and V chromosomes, the former being better retained than the latter in most cases. Some T. aestivum lines carrying D. villosum chromosomes or chromatin, as the result of addition, substitution, or recombination events or even a combination of these karyological events, were found to be stable. Other lines were unstable, and these lines carried 1V, 3V, or 5V chromosomes or their portions. Substitution or recombination events where 1V chromosomes were involved could concern the homeologous counterparts in both the A and B and D genomes of wheat. No line could be recovered where the shorter arm of 3V chromosomes was present. Changes in the morphology and banding pattern of V chromosomes were observed in hybrids that did not carry the entire D. villosum complement. By comparing the results of our cytogenetic analyses with certain phenotypic characteristics of the lines studied, genes for discrete traits could be assigned to specific V chromosomes or V chromosome arms. From the frequency of V chromosomes that were involved in chromatin exchanges with or substituted for one of their homeologous counterparts in the A, B, and D wheat genomes, it was inferred that D. villosum belongs to the same phyletic lineage as T. urartu (donor of the A genome of wheat) and Aegilops speltoides (B genome), and that Ae. squarrosa (D genome) diverged earlier from D. villosum.  相似文献

To determine whether deer can transmit Neospora caninum, brains of naturally infected white-tailed deer (Odocoileus virginianus) were fed to 4 dogs; 2 of these dogs shed oocysts. Oocysts from 1 of the dogs were tested by polymerase chain reaction and found to be positive for N. caninum and negative for Hammondia heydorni. The internal transcribed spacer 1 sequence of the new strain (designated NC-deer1) was identical to N. caninum from domestic animals, indicating that N. caninum is transmitted between wild and domestic animals, often enough to prevent divergent evolution of isolated populations of the parasite. NC-deerl oocysts were administered to a calf that developed a high antibody titer, providing evidence that N. caninum from wildlife can infect cattle. In addition, N. caninum antibody seroprevalence was detected in 64/164 (39%) free-ranging gray wolves (Canis lupus), 12/113 (11%) coyotes (Canis latrans), 50/193 (26%) white-tailed deer, and 8/61 (13%) moose (Alces alces). These data are consistent with a sylvatic transmission cycle of N. caninum between cervids and canids. We speculate that hunting by humans favors the transmission of N. caninum from deer to canids, because deer carcasses are usually eviscerated in the field. Infection of canids in turn increases the risk of transmitting the parasite to domestic livestock.  相似文献

Cytogenetics of wild and captive bred non-domestic animals provides us with valuable information that can be implemented in wildlife management and species conservation strategies. In this review, we summarized the data published to date describing a range of chromosome abnormalities observed in non-domestic animals and their effect on phenotype. Two important factors that can potentially have drastic effects on captive breeding programs are discussed: presence of classic chromosome abnormalities, spontaneously-occurring and inherited, and intraspecific variations in chromosome number. Short-term consequences, primarily reduced reproductive efficiency, and long-term consequences, such as changes in population dynamics, are examined.  相似文献

Various models purporting to explain natural hybrid zones make different assumptions about the fitness of hybrids. One class of models assumes that hybrids have intrinsically low fitness due to genetic incompatibilities, whereas other models allow hybrid fitness to vary across natural environments. We used the intrinsic rate of increase to assess lifetime fitness of hybrids between two species of montane plants Ipomopsis aggregata and Ipomopsis tenuituba planted as seed into multiple field environments. Because fitness is predicted to depend upon genetic composition of the hybrids, we included F1 hybrids, F2 hybrids, and backcrosses in our field tests. The F2 hybrids had female fitness as high, or higher, than expected under an additive model of fitness. These results run counter to any model of hybrid zone dynamics that relies solely on intrinsic nuclear genetic incompatibilities. Instead, we found that selection was environmentally dependent. In this hybrid zone, cytoplasmic effects and genotype-by-environment interactions appear more important in lowering hybrid fitness than do intrinsic genomic incompatibilities between nuclear genes.  相似文献

Background

A major reduction of genetic diversity in mtDNA occurred during the domestication of East Asian pigs. However, the extent to which genetic diversity has been lost in the nuclear genome is uncertain. To reveal levels and patterns of nucleotide diversity and to elucidate the genetic relationships and demographic history of domestic pigs and their ancestors, wild boars, we investigated 14 nuclear markers (including 8 functional genes, 2 pseudogenes and 4 intergenic regions) from 11 different chromosomes in East Asia-wide samples and pooled them with previously obtained mtDNA data for a combined analysis.

Principal Findings

The results indicated that domestic pigs and wild boars possess comparable levels of nucleotide diversity across the nuclear genome, which is inconsistent with patterns that have been found in mitochondrial genome.

Conclusions

This incongruence between the mtDNA and nuclear genomes is suggestive of a large-scale backcross between male wild boars and female domestic pigs in East Asia. Our data reveal the impacts of founder effects and backcross on the pig genome and help us better understand the complex demographic histories of East Asian pigs, which will be useful for future work on artificial selection.  相似文献

Crosses between the diploid Hylocereus polyrhizus, as the female parent, and the tetraploid Selenicereus megalanthus, as the male parent, yielded triploid and aneuploid hybrids. The fruits of these hybrids combined the attractive appearance of Hylocereus fruits with the delicious taste of S. megalanthus fruits. The aim of this work was to assess the fertility and breeding potential of the triploid and aneuploid hybrids with a view to developing an improved vine cactus crop. Pollen mother cells at metaphase I revealed univalents, bivalents, trivalents, and occasionally quadrivalents. Chromosome distribution at anaphase I revealed different classes of chromosome segregation as well as lagging chromosomes. At metaphase II, parallel and tripolar spindles were observed. The occurrence of triads was frequent, whereas dyads were rarely observed. Pollen stainability varied among the clones studied ranging from 9.8% to 18.6%. The diameters of the stained pollen grains varied widely, probably as a result of the number of chromosomes. Despite the allotriploid origin of our hybrids, functional female and male gametes were produced in considerable proportions, most likely as a result of balanced chromosome segregation. The triploid and aneuploid clones studied yielded viable seeds whose number per fruit was strongly dependent on the pollen donor.  相似文献

The objective of this study was to identify quantitative trait loci (QTL) affecting fitness of hybrids between wild soybean (Glycine soja) and cultivated soybean (Glycine max). Seed dormancy and seed number, both of which are important for fitness, were evaluated by testing artificial hybrids of G. soja × G. max in a multiple‐site field trial. Generally, the fitness of the F1 hybrids and hybrid derivatives from self‐pollination was lower than that of G. soja due to loss of seed dormancy, whereas the fitness of hybrid derivatives with higher proportions of G. soja genetic background was comparable with that of G. soja. These differences were genetically dissected into QTL for each population. Three QTLs for seed dormancy and one QTL for total seed number were detected in the F2 progenies of two diverse cross combinations. At those four QTLs, the G. max alleles reduced seed number and severely reduced seed survival during the winter, suggesting that major genes acquired during soybean adaptation to cultivation have a selective disadvantage in natural habitats. In progenies with a higher proportion of G. soja genetic background, the genetic effects of the G. max alleles were not expressed as phenotypes because the G. soja alleles were dominant over the G. max alleles. Considering the highly inbreeding nature of these species, most hybrid derivatives would disappear quickly in early self‐pollinating generations in natural habitats because of the low fitness of plants carrying G. max alleles.  相似文献

The present note, by discussing some field experiences in wild and domestic ruminant populations, approaches eco-epidemiology of abomasal nematodes in relation with host health and dynamics. Factors possibly playing a role in the host-parasite relationship are discussed, as well as possible use of macroparasites as ecological and sanitary indicators. Pseudorabies has become endemic and represents a widespread problem for pig production in the world, causing great economic losses associated with reproductive failure and neonatal mortality in the pig industry. Most diseases are the results of mutations of functional genes. Single-nucleotide polymorphisms (SNPs) from the coding regions of the mediators of pro-inflammatory responses or other candidate genes in pigs could indicate their potential involvement in susceptibility or resistance to PrV (pseudorabies virus) infection. There have been no previous association studies with candidate host genes that may influence PrV phenotypic traits. In order to perform association studies to identify genes contributing to PrV phenotypes, the genotypes of five SNPs from four genes (IL10, CXCL12, BAT2 and EHMT2) were determined for 178 sow samples using a high throughput microarray-based methodology. PrV antibodies were tested by enzyme-linked immunosorbent assay (ELISA) to determine whether there was an association between antibody levels and particular genotypes. The association between SNP genotypes and the PrV antibody levels were analysed using the Duncan method of one-way ANOVA procedure using the SAS (Statistical Analysis Systems) software package. The results showed that the glycoprotein E-ELISA antibody level of pigs with genotypes 11(AA) and 12(AG) was significantly higher than in pigs with genotype 22(GG) (P < 0.05) of SNP in the gene EHMT2-SNP2. The SNP of EHMT2 may be an effective potential tool to identify susceptible and resistant animals when used in conjunction with traditional selection methods.  相似文献
