When isolated bull sperm chromatin is incubated with 0.1 M 2-mercaptoethanol at pH 8, an extensive proteolytic degradation of sperm histone occurs, being accompanied by a marked swelling of the chromatin masses. The degradation of sperm histone is strongly inhibited by monovalent or divalent metal ions. The protease found in isolated bull sperm chromatin possesses properties indistinguishable from those of an acrosomal protease of trypsin-type, acrosin (EC 3.4.21.10), and requires a combination of NaCl, urea and 2-mercaptoethanol for its extraction. Evidence suggests that the protease travels along chromatin strands and hydrolyzes essentially all the sperm histone molecules within the chromatin masses.  相似文献

The DNA of most vertebrate sperm cells is packaged by protamines. The primary structure of mammalian protamine I can be divided into three domains, a central DNA binding domain that is arginine-rich and amino- and carboxyl-terminal domains that are rich in cysteine residues. In native bull sperm chromatin, intramolecular disulfide bonds hold the terminal domains of bull protamine folded back onto the central DNA binding domain, whereas intermolecular disulfide bonds between DNA-bound protamines help stabilize the chromatin of mature mammalian sperm cells. Folded bull protamine was used to condense DNA in vitro under various solution conditions. Using transmission electron microscopy and light scattering, we show that bull protamine forms particles with DNA that are morphologically similar to the subunits of native bull sperm chromatin. In addition, the stability provided by intermolecular disulfide bonds formed between bull protamine molecules within in vitro DNA condensates is comparable with that observed for native bull sperm chromatin. The importance of the bull protamine terminal domains in controlling the bull sperm chromatin morphology is indicated by our observation that DNA condensates formed under identical conditions with a fish protamine, which lacks cysteine-rich terminal domains, do not produce as uniform structures as bull protamine. A model is also presented for the bull protamine.DNA complex in native sperm cell chromatin that provides an explanation for the positions of the cysteine residues in bull protamine that form intermolecular disulfide bonds.  相似文献

Mammalian fertilization involves various steps in which the participation of specific enzymes has been demonstrated by numerous studies. Acrosin is one of the most widely acrosomal protease in mammalian spermatozoa studied, including bovine; however, other proteases have also been described. A new trypsin-like serine protease named bovine serine protease of 120 kDa (BSp120) and its pre-cursor BSp66 (66 kDa) were identified in bovine spermatozoa. Cytological and ultrastructural immunolocalization studies on BSp120 were performed in live and fixed cells. Immunoflorescence assays with specific polyclonal antibodies revealed localization of BSp120 on the sperm head, with a signal homogeneously distributed over the acrosome resembling a horseshoe. After the acrosome reaction, sperm showed a patchy pattern in the acrosomal cap. Immune electron microscopy analysis indicated that BSp120 is located over the head plasma membrane of capacitated spermatozoa and acrosome reacting spermatozoa. To assess BSp120 function in sperm-oocyte interaction, in vitro fertilization studies were conducted. Oocytes were incubated with spermatozoa pre-treated with anti-BSp120, anti-guinea pig acrosin, and anti-BSp120 plus anti-guinea pig acrosin. Pre-treatment of bovine spermatozoa with antibodies towards each protein did not significantly modify fertilization rates. However, when both anti-acrosin and anti-BSp120 antibodies were simultaneously added, there was a significant decrease in the fertilization rate, suggesting that both enzymes may be required for fertilization. Altogether, the results from the present study described the localization of BSp120 over the acrosome of bovine sperm, and suggest its involvement in fertilization.  相似文献

The diadem/crater defect was studied over several months in two related 20-month-old Angus bulls. In bull 1, diadem/crater defects were present in 2-99% of ejaculated spermatozoa at various times during the evaluation period. In bull 2, affected cells varied from 20% to 94%, with other abnormalities (head and acrosome defects, coiled tails, proximal cytoplasmic droplets) also common. Single sire mating trials conducted over 26 days during an apparent recovery phase showed normal fertility (approximately 50% pregnancies per estrus exposed). Both resting and gonadotropin-releasing hormone (GnRH)-stimulated testosterone values were within nor-mal limits. Histopathological evaluation of testes showed no obvious hypoplastic, inflammatory, or degenerative condition. Electron microscopy of ejaculated spermatozoa demonstrated the characteristic diadem pattern of craters in the equatorial region of the head. Many cells from bull 2 contained large craters in other regions of the nucleus. Electron microscopy of testicular tissue demonstrated nuclear invaginations lined by a single unit membrane in round spermatids. Lesions in elongated spermatids were more pronounced, with curling of the nucleus and large membrane-filled cavities in the chromatin occurring in addition to craters in the equatorial region of the nucleus.  相似文献

Dispersion of nuclear fibers of the spermatozoa of dogfish, man, and bull is made possible after treatment with a reducing and alkylating reagent coupled with an anionic detergent; the same detergent used at a low ionic strength dissociates the nuclear content of the rainbow trout sperm. Electron microscopy of such dispersed nuclear fibers has shown a beads-on-a-string configuration for these four types of sperm chromatin. These structures are morphologically similar to those described in somatic cell nuclei as nucleosomes, although in sperm chromatin the basic proteins associated with DNA were significantly different from histones.  相似文献

Guinea-pig spermatozoa were induced to undergo an acrosome reaction with the ionophore A23187. A time course for the activation of protease activity was established. Acid treatment of fully activated spermatozoa at pH 2.35 for 30 min exposed additional activity. This was attributed to the acid dissociation of a protein inhibitor from acrosin. The acrosin content of fully activated and acid-dissociated sperm extracts was measured using a sensitive active-site titrant for serine proteases. The number of acrosin molecules per spermatozoon, calculated on the basis of the sperm count, was approximately 2 x 10(6), of which half were available without dissociation of the inhibitor.  相似文献

Spermiogenesis in the translocation heterozygote T (1; 2H) 25(20) y l 25/FM6 has been studied with the electron microscope and compared with that in wild type males. It appears that the genetic lesion in the male sterility mutant is associated primarily with a failure in differentiation of the head. In wild type flies, the spermatid nucleus assumes a conchoidal shape; chromatin accumulates along the convex surface. Adjacent to the concave surface a large bundle of microtubules runs parallel to the long axis of the spermatid. A single row of microtubules is juxtaposed against the convex surface of the head. As differentiation proceeds, the nucleus elongates, chromatin condenses, and the nucleus is compacted to a final diameter of about 0.3 µ. In the sterile mutant the spermatid nucleus has an irregular or wedge-shaped profile and no concavity is formed, nor is the bundle of microtubules observed. The row of microtubules, however, is usually present around the periphery. The change from lysine-rich to arginine-rich histone in mature wild type sperm does not occur in the sterile male. The substructure of the axial filament and mitochondrial derivatives, however, are similar to those in wild type.  相似文献

We describe here a computational morphology-based approach to the investigation of possible causes of chromatin alterations in sperm. A comprehensive set of state-of-the-art and geometric measures are computationally extracted from toluidine blue stained images and analyzed to infer the possible processes leading to normal and abnormal chromatin formation while seeking a possible taxonomy of chromatin alterations and their influence on sperm head morphology. Using this methodology, we have identified higher chromatin fragility at some specific points of the sperm head. Despite the lack of correlation between morphologies of sperm head and chromatin structure, four main morphological types of chromatin alterations in bull spermatozoa have been identified and their possible causes discussed.  相似文献

The aim of the present work was to immunolocalize acrosin in bull spermatozoa incubated for up to 6 h in capacitating culture medium (TALP-heparin), in order to study the kinetics of its release during the acrosome reaction and in vitro sperm penetration. Six replicates from semen of one bull were used. Acrosin was localized by the silver-enhanced immunogold technique using anti-bovine acrosin monoclonal antibody ACRO-C2E5. Spermatozoa thus showed the presence of acrosin only at the acrosomal region. Four different patterns were seen: (1) no labeling: (2) intense labeling on the rim of the portion of the acrosome; (3) diffuse label over the entire acrosomal region; and (4) intense label over the entire acrosomal region. Spermatozoa incubated in capacitating medium for 4 h showed that unlabeled (pattern 1) spermatozoa decreased from 72% to 28% difference that was found to be significant (p<0.05). Patterns 3 and 4 increased from about 10% to 20-29%, (p<0.05). With further incubation (4-6 h), pattern 1 increased while patterns 3 and 4 decreased differences were not significant (p0.05). The incidence of pattern 2 did not change through the whole incubation period. Sperm penetration through the zona pellucida of in vitro matured bovine oocytes (57%) or empty zonae pellucida (70.5%) increased (p<0.05) as a function of sperm incubation time in capacitating medium. The presence of acrosin, as determined by the silver-enhanced immunogold technique, was highly correlated with sperm penetration of in vitro mature bovine oocyte (r=0.98) and cryopreserved zonae pellucidae (r=0.93) (p<0.01).  相似文献

We have identified an 80 kDa protein in ejaculated bull spermatozoa (p80) which is found in acrosomal and post-acrosomal areas of the head. It has a hyaluronidase activity and shares homologies with PH-20, a sperm surface glycoprotein involved in sperm-egg interaction. The aim of the present study was to characterize bull sperm p80 protein at the nucleic and amino acid levels to determine whether it is the bovine PH-20 ortholog. The complete nucleotide sequence determined by RT-PCR, 3' and 5' RACE show that bull p80, displays identity with the PH-20 nucleotide and amino acid sequences. Messenger RNA and protein expressions determined by Northern blot and immunohistochemistry revealed that the protein is testicular (expressed in spermatocytes and spermatids). The localization of p80 on spermatozoa, determined by indirect immunofluorescence using a monoclonal antibody, shows the protein in acrosomal and post acrosomal areas of the head with an increase in the signal intensity as sperm progress through the epididymis. Post-translational modifications of the protein were investigated during the epididymal maturation by Western blot on protein extracts from sperm collected in the caput, corpus and cauda portions of bull epididymis. Glycolysation status of sperm p80 protein on proteins from ejaculated and epididymidal sperm was investigated. Result show that the glycosylation status is modified as spermatozoa migrate through the epididymis. Hyaluronidase activity evaluated in protein extracts from spermatozoa of the three different epididymal sections revealed that the activity is higher at pH 7 than 4 and is not affected by epididymal maturation. These data strongly suggest that p80 is the bovine PH-20.  相似文献

The appearance of the nuclear abnormality “crater defect” is described in spermatozoa from the domestic horse (Equadae). Under scanning electron microscopy, the defect appears as a severe depression at any area of the sperm nucleus or as a blister or swelling at some point on the sperm nucleus. Ultrastructurally, the crater appears as a nuclear vacuole containing amorphous material similar to that described in bull and boar sperm. The craters ranged in size from 0.5 to 2 μm in diameter. Within ejaculates of stallions having this defect, the percentages of sperm with crater defects varied widely over time, being as high as 60% and as low as 15%.  相似文献

Over 200 genes have been shown to be associated with infertility in mouse models. However, knockout mice reveal unexpected functional redundancy of some germ cell expressed genes. Single null mutations in mouse genes encoding four male germ cell proteins, transition protein 2 (Tnp2), proacrosin (Acr), histone H1.1 (H1.1), histone H1t (H1t) and sperm mitochondria-associated cysteine-rich protein (Smcp) have been generated and analysed. Tnp2 is believed to participate in the removal of the nuclear histones and initial condensation of the spermatid nucleus. Proacrosin is an acrosomal protease synthesized as a proenzyme and activated into acrosin during the acrosome reaction. The linker histone subtype H1.1 belongs to the group of main-type histones and is synthesized in somatic tissues as well as in germ cells during the S-phase of the cell cycle. The histone gene Hist1h1t is expressed exclusively in spermatocytes and may have a function in establishing an open chromatin structure for the replacement of histones by transition proteins and protamines. Sperm mitochondria-associated cysteine-rich protein (Smcp) is a major structural element of the mitochondria in the midpiece of the sperm tail. Male mutant mice lacking any of these proteins show no apparent defects in spermatogenesis or fertility. To examine the synergistic effects of these proteins in spermatogenesis and during fertilization four lines of double knockout mice Hist1h1a/Mcsp, Hist1h1t/Mcsp, Tnp2/Mcsp and Acr/Mcsp were established. It was found that even when knockout mice are heterozygous for one allele (-/+) and homozygous for the other allele (-/-), mice were subfertile. Homozygous double knockout mice of all four lines are nearly infertile. However, in the four homozygous double knockout mouse lines, different characteristic abnormalities are prominently manifested: In Hist1h1a-/-/Mcsp-/- the migration of spermatozoa is disturbed in female genital tract, in Hist1h1t-/-/Mcsp-/- spermatozoa show morphological head abnormalities, in Tnp2-/-/Mcsp-/- the motility of sperm is affected, and in Acr-/-/Mcsp-/- the sperm-oocyte interaction is impaired. These findings indicate strongly that male germ cell expressed genes have synergistic effects on male fertility.  相似文献

Mammalian sperm acrosomes contain a trypsin-like protease called acrosin which causes limited and specific hydrolysis of the extracellular matrix of the mammalian egg, the zona pellucida. Acrosin was localized on hamster, guinea-pig and human sperm using monoclonal and polyclonal antibodies to human acrosin labelled with colloidal gold. This was visualized directly with transmission electron microscopy, and with light and scanning microscopy after silver enhancement of the colloidal gold probe. Four distinct labelling patterns were found during capacitation and the acrosome reaction in hamster and guinea-pig spermatozoa, and three patterns were found in human spermatozoa. In the hamster, acrosin was not detected on the inner acrosomal surface after the completion of the acrosome reaction, thus correlating with the observation that hamster spermatozoa lose the ability to penetrate the zona after the acrosome reaction. With guinea-pig and human spermatozoa, acrosin was still detected after the completion of the acrosome reaction, thus correlating with the observation that acrosome reacted guinea-pig spermatozoa bind to and penetrate the zona pellucida.  相似文献

The diversity of the structural organization of the spermatozoa of African murid rodents is described at the light and transmission electron microscopical level of resolution. In most species the sperm head is falciform in shape but it varies somewhat in overall breadth, width, and length. A typical perforatorium is present and the acrosome splits into a large head cap over the convex surface and a smaller ventral segment similar to the sperm head of most Asian and Australasian murids. In a few species, however, the morphology is very different. In Acomys and Uranomys spermatozoa, the apical hook is more bilaterally flattened, has a large apical acrosomal region, and no separate ventral segment. Two species of Aethomys have, in addition to an apical hook, a 4μ long extension of the cytoskeletal material that projects from the concave surface of the sperm head, whereas in Dasymys two large ventral processes extend from the upper concave region which contain nuclear material basally and a huge extension of cytoskeleton apically. In Aethomys chrysophilus type B, the sperm nucleus is unique in form and often has a central region in which threads of chromatin can be seen; it is capped by a massive acrosome whose apical segment is complex and convoluted in structure. Stochomys longicaudatus appears to have a conical sperm head, and in all three Lophuromys species the sperm head is spatulate in shape with the flat, plate-like nucleus capped by a thin acrosome. The evolutionary trends in changes of sperm head shape and design of these rodents are discussed. It is suggested that some of the differences in morphology may relate to the variation in structural organization of the coats around the egg through which the spermatozoon has to pass in order for fertilization to occur.  相似文献

Treatment of bull spermatozoa with DDC--Na/dithiothreitol results in the swelling and decondensation of nuclear chromatin. The structures formed at the final stages of decondensation are morphologically similar to the male pronucleus. Cytophotometric analysis has shown that decondensation of chromatin in the gametes in followed by quantitative changes of basic nuclear proteins. In partly--decondensed sperm nuclei the intensity of histone staining increases as a result of the appearance of extra reactive groups. In fully decondensed nuclei there remain only 54% of histones of the original haploid level. Nucleoproteins revealed in the sperm with fully dispersed chromatin must be histones of the somatic type.  相似文献

Two acid stable proteinase inhibitors are present in bull seminal plasma and washed ejaculated bull spermatozoa. Inhibitor I with a molecular weight of about 8700 (estimated by gel filtration) is a very strong inhibitor of bull sperm acrosin but also inhibits bovine trypsin and chymotrypsin and porcine plasmin; inhibition of porcine pancreatic and urinary kallikrein was not observed. In this respect inhibitor I resembles the well known cow colostrum trypsin inhibitor. Inhibitor II with a molecular weight near 6800 (estimated by gel filtration) inhibits bovine trypsin and chymotrypsin, porcine plasmin and pancreatic and urinary kallikrein as well as bull acrosin. The inhibition specificity of inhibitor II is thus very similar to that of the basic inhibitor from bovine organs (Kunitz-type). In view of the inhibition strength and other characteristics, however, the acid stable bull seminal inhibitors are not identical with the inhibitor from cow colostrum or bovine lung (organs).  相似文献

Acrosomeless round-headed spermatozoa from three men were studied under electron microscopy and indirect immunofluorescene microscopy using the anti-calicin antibody that recognizes a basic protein of the sperm perinuclear theca (Longo et al., 1987). Electron microscopy revealed the existence of anomalies of the nuclear envelope, the nuclear matrix underlying the nuclear envelope, and the perinuclear layer. The absence of sperm labeling with the anti-calicin antibody confirmed that the formation of the perinuclear theca was impaired. Data obtained from both mature spermatozoa and ejaculated spermatids suggest that i) round-headed sperm head anomalies result from a failure of differentiation of the sperm-specific skeletal complex related to the nucleus, and ii) the acrosome spreading over the nucleus, the nuclear elongation and the post-acrosomal sheath formation are dependent on such nuclear-perinuclear differentiations. In contrast, chromatin condensation, cytokinesis and some events of the acrosomal shaping appear not to depend on those nuclear-related differentiations. The possible processes allowing the maintenance of the sperm head structures and their subsequent morphogenesis are discussed.  相似文献

Light and transmission electron microscopy of the spermatozoa and spermatogenesis of 16 species (in three genera, Patella, Helcion, Cellana) of patellid limpet have shown that head lengths of the sperm range from 3 to 13 μm, and each species has a sperm with a unique morphology, indicating that the spermatozoa can be used as a taxonomic character. Although spermatozoon structure is species specific, five types can be recognized, based on the size, shape, and structure of the nucleus and acrosome. The occurrence of five morphological types of sperm, one of which (Cellana capensis) is particularly different from other patellids, suggests that the taxonomy of the family Patellidae be re-examined. The morphological changes that occur during spermatogenesis are very similar in all species, although two patterns of chromatin condensation are found. Those species with sperm that have short squat nuclei (length:breadth < 3.5:1) have a granular pattern of condensation. Species with sperm that have more elongate nuclei (length:breadth > 5:1) have an initial granular phase followed by the formation of chromatin fibrils. These fibrils become organized along the long axis of the elongating nucleus. The absence of a manchette suggests that nuclear elongation is brought about from within the nucleus.  相似文献
