目的建立长爪沙鼠原代肝细胞分离培养体系。方法以雄性长爪沙鼠为供体,采用组织消化法和Seglen两步灌流法分离肝细胞,以台盼蓝染色检测细胞得率和活率,过碘酸-希夫氏反应(PAS)鉴定肝细胞,倒置显微镜观察肝细胞形态变化,并使用含有多种细胞因子的培养基维持培养。结果组织消化法和Seglen两步灌流法平均每只长爪沙鼠可分别获得肝细胞(1.33±0.34)×107个、(3.97±1.15)×107个,细胞活率分别为(29.4±6.05)%、(80.3±4.56)%,这两种方法在细胞得率及活率方面存在显著差异。肝细胞内因有大量的糖原颗粒,经PAS染色后被染成红色。结果表明肝细胞在贴壁后72 h内,肝细胞形态发生显著变化。结论采用胶原酶经肝门静脉灌流分离肝细胞是一种高效获得肝细胞的方法。各种细胞因子有利于维持肝细胞在体外的生长分化,长爪沙鼠原代肝细胞分离培养体系的建立将为肝脏相关疾病研究和防治药物的开发提供技术支持。  相似文献

Background. Epidemiological studies have shown a correlation between Helicobacter pylori infection and human gastric carcinogenesis. A Mongolian gerbil model has demonstrated that H. pylori infection induced gastric carcinoma. However, the disadvantage of this animal model is a lack of information regarding the cellular genes involved in oncogenesis. Mutation of the p53 gene is one of the most common steps in gastric carcinogenesis. In this study, we aimed to clone the p53 gene of the Mongolian gerbil and detect the functional mutations in H. pylori‐infected animals. Materials and Methods. The p53 complementary DNA (cDNA) of Mongolian gerbil was cloned by the methods of reverse‐transcribed polymerase chain reaction and rapid amplification of cDNA ends. Results. The p53 cDNA of Mongolian gerbil has a 78.8% homology to that of humans. A novel yeast p53 assay system was established and enabled to detect the functional mutations of the p53 gene in the stomach of the Mongolian gerbil. Conclusions. This is the first report of the complete sequence of wild‐type p53 cDNA of the Mongolian gerbil. This genetic information and an assay system designed to detect the functional mutations of the p53 gene are useful for further investigations of gastric oncogenesis in this animal model.  相似文献

The Mongolian gerbil (Meriones unguiculatus) is known to be one of the most radioresistant animals. We have examined the X-ray sensitivity of normal diploid fibroblasts from Mongolian gerbil embryos compared with those of cultured embryo cells obtained from various laboratory animals and a normal human. There was a wide difference in X-ray sensitivity for cell killing among different mammalian species. The D0 values for Mongolian gerbil cells ranged from 2.08 to 2.28 Gy, values which are twice as high as those for human cells. The mean D0 value for human cells was 1.06 Gy. Mouse, rat, Chinese hamster, and Syrian/golden hamster cells showed similar D0 values ranging from 1.30 to 1.56 Gy. When cells were irradiated with X rays, ten times more chromosome aberrations were detected in human cells than in Mongolian gerbil cells. The frequencies of chromosome aberrations in other rodent cells were between the values for cells from humans and those from gerbils. These data indicate that the Mongolian gerbil cells are resistant to X-ray-induced cell killing and chromosome aberrations, and that the radiation sensitivity of mammalian cells in primary culture may be reflected by their radioresistance in vivo.  相似文献

目的对长爪沙鼠线粒体DNA控制区全序列进行测定,并对其进行鉴定及进化分析。方法根据长爪沙鼠已知基因序列设计引物,采用PCR产物测序法,对所得的片段进行测序鉴定。结合已公布啮齿类动物D-loop区序列,分析其碱基组成、遗传距离、并基于最小进化法和UPGMA法构建系统进化树。结果获得长爪沙鼠D-loop区序列,其与家鼠、小家鼠和仓鼠平均同源性为58%;碱基组成分析显示,长爪沙鼠与啮齿类动物有相似的碱基组成和碱基偏离,其A-skew和G-skew分别为0.0047和-0.28。进化分析结果显示,长爪沙鼠与家鼠(0.35)、黑家鼠(0.38)和仓鼠(0.39)具有较近的遗传距离,其分化顺序为跳鼠、蔗鼠、长爪沙鼠、仓鼠、家鼠和小家鼠。结论本研究获得长爪沙鼠D-loop区全序列,确定了长爪沙鼠与仓鼠、家鼠、小家鼠及其它啮齿动物的进化关系,为长爪沙鼠进化研究、线粒体的结构和功能研究奠定基础。  相似文献

为检验施用炔雌醚对农牧交错带长爪沙鼠家群大小与巢域行为的影响,于2006年5月至9月对内蒙古锡林郭勒盟锡林浩特市农牧交错区域分布的长爪沙鼠家群使用炔雌醚进行种群不育控制实验。分设投药区和对照区两个组别,分别于5月、7月与9月份采用1/4样圆面积有效洞口计数法,随机选取24个长爪沙鼠家群,调查对照区域和投药区内沙鼠的家群洞口数量以及家群大小。另在对照区和投药区,随机测定了30个沙鼠的家群活动范围,对比使用炔雌醚前后长爪沙鼠巢域平均半径和活动面积的影响。结果为:对照区家群平均洞口数58个,投药区仅23个,在炔雌醚投药区长爪沙鼠家群受炔雌醚影响,洞口数仅为23个,表明其对长爪沙鼠家群大小控制效果显著。投药区内长爪沙鼠家群密度在投药后连续下降,最大下降幅度较对照区家群密度低70%,表明炔雌醚对长爪沙鼠的家群密度影响较大,可显著降低长爪沙鼠家群密度(P<0.05)。炔雌醚对长爪沙鼠家群平均半径和活动面积的研究结果显示,施用炔雌醚后家群巢域半径与活动面积均显著缩小(P<0.05),与对照区巢域平均面积相比缩小15%、平均活动半径缩小30%。通过以上结果可得出:单独施用炔雌醚对控制野外长爪沙鼠家群以及降低长爪沙鼠巢域、活动面积效果显著。炔雌醚可有效降低农牧交错带长爪沙鼠种群数量和有效活动范围,这对于农牧交错带的鼠害防控,同时降低鼠源性疾病,包括鼠疫的传播都有一定的意义。  相似文献

The Mongolian gerbil (Meriones unguiculatus) has been used as a laboratory species in many fields of research, including neurology, oncology, and parasitology. Although the cryopreservation of embryos has become a useful means to protect valuable genetic resources, its application to the Mongolian gerbil has not yet been reported. In this study, we investigated the in vitro and in vivo developmental competence of Mongolian gerbil embryos cryopreserved by vitrification. In vivo-fertilized embryos were vitrified on the day of collection using the ethylene glycol (EG)-based solutions EFS20 and EFS40, which contained 20% and 40% EG, respectively, in PB1 containing 30% (w/v) Ficoll 70 and 0.5 M sucrose. First, we compared one-step and two-step vitrification protocols. In the one-step method, the embryos were directly transferred into the vitrification solution (EFS40), whereas in the two-step method, the embryos were exposed serially to EFS20 and EFS40 and then vitrified. After liquefying (thawing), late two-cell embryos (collected on day 3) vitrified by the two-step method showed significantly better rates of in vitro development to the morula stage compared to those vitrified by the one-step method (65% vs. 5%, P < 0.0001). We then examined whether the same two-step method could be applied to early two-cell embryos (collected on day 2), four-cell embryos (day 4), morulae (day 5), and blastocysts (day 6). After liquefying, 87%-100% of the embryos were morphologically normal in all groups, and 23% and 96% developed to the compacted morula stage from early two- and four-cell embryos, respectively. After transfer into recipient females, 3% (4/123), 1% (1/102), 5% (4/73), and 10% (15/155) developed to full-term offspring from vitrified and liquefied early two-cell embryos, late two-cell embryos, morulae, and blastocysts, respectively. This demonstrates that Mongolian gerbil embryos can be safely cryopreserved using EG-based vitrification solutions.  相似文献

The cytochrome P-450 content (0.75 +/- 0.13 nmol/mg microsomal protein) in musk shrew (suncus, Suncus murinus) liver microsomes was lower than that (1.30 +/- 0.26) in rat liver microsomes, but it is approximately the same level as in the Mongolian gerbil (Meriones unguiculatus, 1.18 +/- 0.14), harvest mouse (Micromys minutus, 1.11 +/- 0.02) and rat. 2. The hydroxylation activity (based on cytochrome P-450) of medium-chain fatty acids (otanoic, decanoic, lauric and tridecanoic acids) is much higher in suncus, Mongolian gerbil and harvest mouse than in rat, with the exception of the activity of decanoic and tridecanoic acids in Mongolian gerbil. 3. This suggests that cytochrome P-450 species catalyzing the hydroxylation of medium-chain fatty acids are present in these laboratory animals in higher concentrations. 4. The aminopyrine N-demethylation activity based on microsomal protein or cytochrome P-450 in suncus is significantly lower than that in rat, but the N-demethylation activity in Mongolian gerbil and harvest mouse is approximately 1.7-2.0-fold greater than that in rat.  相似文献

The purpose of this study was to determine whether species differences in neonatal vocalizations of rodent pups could be observed. Ultrasonic vocalizations of pups of 5 rodent species, mouse (ICR), vole (Microtus arvalis), Syrian hamster, rat (Wistar-Imamichi), and Mongolian gerbil were recorded from 3 to 15 or 21 days of age. Recordings were made under conditions of separation from mothers and litter mates in a cooled chamber (approximately 10 degrees C). The major species differences observed were age specific and species specific frequencies. The Mongolian gerbil displayed a different frequency change with age. Namely, the day on which ultrasonic vocalizations ceased was delayed in Mongolian gerbil compared with the other rodents. The modal peak frequencies of ultrasound emitted from pups at 3 days of age were low (around 35 kHz) in the vole and the Syrian hamster, medium (around 45 kHz) in the rat and the Mongolian gerbil, and high (around 55 kHz) in the mouse.  相似文献

We describe a simple and feasible procedure for performing intravenous administration of substances in the gerbil. Under light anaesthesia, animals were held in dorsal recumbency and a very small incision of skin, parallel to the femoral vein on the internal side of the thigh, was made. The vein is easily accessible via thin skin incision. An insulin syringe and a 30 G needle were used for the injection. This is an easy and quick method, which, with appropriate anaesthesia, allows rapid recovery.  相似文献

通过对9个微卫星座位的扩增,研究了Z:ZCLA长爪沙鼠的遗传多态性。结果表明,Z:ZCLA长爪沙鼠在其中1个位点上只有一个等位基因,在其它位点上均有2~4个等位基因,平均等位基因数2.6个。平均杂合度0.4684,平均多态信息量0.4166,平均有效等位基因数2.1756。全群基因纯合度从0.1111~0.5555,平均0.3389,提示目前本群遗传多样性水平处于中度多态。  相似文献

目的克隆长爪沙鼠β-防御素基因序列,并对其进行鉴定及分析。方法从长爪沙鼠小肠中提取总RNA,根据GeneBank中大小鼠的β-防御素的基因序列,通过防御素基因的保守性设计引物,采用RT-PCR技术得到预期的PCR产物,将所得的片段进行克隆、测序,并应用相关生物信息学软件对序列进行鉴定和分析,序列提交genbank。结果Blast比较发现最终测序的结果与小鼠β-防御素-1和大鼠β-防御素-1的同源性全都大于80%,genebank登录号为EU834052。结论经测序鉴定证实该PCR产物为长爪沙鼠阻防御素基因1的一部分。本研究为深入探讨长爪沙鼠β-防御素的生物学活性奠定基础。  相似文献

Platelet-activating factor (PAF) plays an important role in the pathogenesis of sepsis, and the level of plasma PAF acetylhydrolase (pPAF-AH), which inactivates PAF, decreases in sepsis patients except for the sepsis caused by severe leptospirosis. Usually, increase of pPAF-AH activity was observed in lipopolysaccharide (LPS)-induced Syrian hamster and rat sepsis models, while contradictory effects were reported for mouse model in different studies. Here, we demonstrated the in vivo effects of LPS upon the change of pPAF-AH activity in C57BL/6 mice and Mongolian gerbils. After LPS-treatment, the clinical manifestations of Mongolian gerbil model were apparently similar to that of C57BL/6 mouse sepsis model. The pPAF-AH activity increased in C57BL/6 mice after LPS induction, but decreased in Mongolian gerbils, which was similar to that of the human sepsis. It thus suggests that among the LPS-induced rodent sepsis models, only Mongolian gerbil could be used for the study of pPAF-AH related to the pathogenesis of human sepsis. Proper application of this model might enable people to clarify the underline mechanism accounted for the contradictory results between the phase II and phase III clinical trials for the administration of recombinant human pPAF-AH in the sepsis therapy.  相似文献

荒漠破碎化生境中长爪沙鼠集合种群野外验证研究   总被引:2,自引:0,他引:2  
近年来,人类活动和自然干扰,导致内蒙古阿拉善荒漠区生境的破碎化,出现了长爪沙鼠在不同斑块间的不连续分布,每一斑块内可能存在一个局域种群,而集合种群建立的前提条件,是局域种群斑块状分布在离散的栖息地环境中。2002~2012年每年的4~10月,在阿拉善荒漠区禁牧、轮牧、过牧和开垦4种人为不同利用方式形成的生境斑块中,采用标志重捕法对长爪沙鼠(Meriones unguiculatus)种群进行定点监测。通过分析长爪沙鼠种群动态,计算各局域种群的灭绝风险,利用Spearman秩相关系数检验种群动态的空间同步性,同时以种群周转率对长爪沙鼠扩散能力进行评估,以检验阿拉善荒漠区长爪沙鼠种群空间结构是否具有经典集合种群的功能。结果表明:(1) 不同生境斑块可被长爪沙鼠局域种群占据,11年间捕获长爪沙鼠2~7次不等;(2) 长爪沙鼠所有局域种群均具有灭绝风险,在轮牧区和禁牧区灭绝率高达1.000 0,开垦区灭绝率最低,也达到0.333 4,而本研究期间最大局域种群(2008年过牧区,26只/hm2),在2010年发生了局域灭绝;(3) 不同生境斑块间没有明显的空间隔离而阻碍局域种群的重新建立,长爪沙鼠扩散能力较强,绝大部分月份的种群周转率在50.0%以上,特别是周转率达到100.0%的月份较多;(4) 不同生境斑块间仅轮牧区和禁牧区中长爪沙鼠种群密度显著正相关(P<0.05),而其他生境斑块间相关性均不显著(P >0.05),长爪沙鼠局域种群整体显示出明显的非同步空间动态。阿拉善荒漠区长爪沙鼠种群满足作为经典集合种群物种区域续存的4个条件,具有作为研究小哺乳动物集合种群的潜在价值。  相似文献

The principal bile acid of Mongolian gerbil bile is cholic acid, although small amounts of chenodeoxycholic and lesser amounts of deoxycholic acids are identified. Muricholic acids were not found in gerbil bile. The ratio of trihydroxy to dihydroxy bile acids in gerbil bile is approximately 11:1. After administration of [4-(14)C]5alpha-cholestan-3beta-ol to gerbils with bile fistulas, 4-7% of the administered (14)C was recovered in bile and 16% in urine on the first 6 days. Alkaline hydrolysis of the bile afforded the biliary acids which were separated by partition chromatography. The (14)C ratio of trihydroxy to dihydroxy bile acids was 11:1. Allocholic acid was identified as the major acidic biliary metabolite. From analysis of (14)C retained in selected tissues, the adrenal gland appears to be an important site for retention of cholestanol or its metabolites.  相似文献

以2000 年6 月~10 月群居性长爪沙鼠野生种群的标志重捕资料为依据, 采用Jolly - Seber 模型估算了该鼠种群参数, 结果表明, 长爪沙鼠个体间具等捕性(Leslie 法检验) , 研究期间取样个体的重捕率平均为89.7 %(77.4 % ~ 100 %) ; 参数估计结果具有合理的生物学意义, 认为采用该模型估算长爪沙鼠种群参数是适用的。  相似文献

Formation of fibrin sleeves around catheter tips is a central factor in catheter failure during chronic implantation, and such tissue growth can occur despite administration of anticoagulants. We developed a novel method for monitoring catheter patency. This method recognizes the progressive nature of catheter occlusion, and tracks this process over time through measurement of changes in catheter resistance to a standardized 1 mL bolus infusion from a pressurized reservoir. Two indirect measures of catheter patency were used: (a) reservoir residual pressure and (b) reservoir discharge time. This method was applied to the study of catheter patency in rats comparing the effect of catheter material (silastic, polyurethane, Microrenathanetrade mark), lock solution (heparin, heparin/dexamethasone) and two different cannulation sites (superior vena cava via the external jugular vein, inferior vena cava via the femoral vein). Our findings reveal that application of flexible smaller-size silastic catheters and a dexamethasone lock solution resulted in prolonged catheter patency. Patency could be maintained over nine weeks with the femoral vein catheters, compared with five weeks with the external jugular vein catheters. The current method for measuring catheter patency provides a useful index for the assessment of tissue growth around the catheter tip. The method also provides an objective and quantitative way of comparing changes in catheter patency for different surgical methods and catheter types. Our method improves on the conventional method of assessing catheter occlusion by judging the ability to aspirate from the catheter.  相似文献

The aldehyde dehydrogenase (Aldehyde:NAD(P) oxidoreductase E.C. 1.2.1.3. and 1.2.1.5) phenotype in several tissues of the Mongolian gerbil, Meriones unguiculatus, has been established. The tissue distribution of gerbil aldehyde dehydrogenase is similar to that of the rat, with liver possessing the majority of the aldehyde dehydrognease activity. Male kidney and testis possess significantly more activity than female kidney and ovary. The substrate and co-enzyme specificity of gerbil liver aldehyde dehydrogenase is also similar to that of rat and mouse liver. Gel isoelectric focusing resolves one major gerbil liver aldehyde dehydrogenase isozyme at pI 5.3. Mouse liver is resolved into two major isozymes at pIs 5.3 and 5.6 and rat liver aldehyde dehydrogenase into one major isozyme at pI 5.4. Gerbil liver aldehyde dehydrogenase is functional over a broad pH range with an optima at pH 9.0. Rat and mouse liver aldehyde dehydrogenase possess sharp pH optima at pH 8.5.  相似文献

The nasolacrimal apparatus (NLA) is a multicomponent functional system comprised of multiple orbital glands (up to four larger multicellular exocrine structures), a nasal chemosensory structure (vomeronasal organ: VNO), and a connecting duct (nasolacrimal duct: NLD). Although this system has been described in all tetrapod vertebrate lineages, albeit not always with all three main components present, considerably less is known about its ontogeny. The Mongolian gerbil (Meriones unguiculatus) is a common lab rodent in which the individual components of the adult NLA have been well studied, but as yet nothing is known about the ontogeny of the NLA. In this study, serial sections of 15 fetal and three adult Mongolian gerbil heads show that the development of the NLA falls into three fetal stages: inception (origin of all features), elongation (lengthening of all features), and expansion (widening of all features). No postnatal or juvenile specimens were observed in this study, but considerable growth evidently occurs before the final adult condition is reached. The development of the orbital glands and the VNO in the Mongolian gerbil is largely consistent with those in other mammals, despite a slight nomenclatural conundrum for the anterior orbital glands. However, the Mongolian gerbil NLD follows a more circuitous route than in other tetrapods, due mainly to the convoluted arrangement of the narial cartilages, the development of a pair of enlarged incisors as well as an enlarged infraorbital foramen. The impact of these associated features on the ontogeny and phylogeny of the NLA could be examined through the approach of network science. This approach allows for the incorporation of adaptations to specific lifestyles as potential explanations for the variation observed in the NLA across different tetrapod clades. J. Three experiments on food hoarding behavior in the Mongolian gerbil (Meriones unguiculatus) demonstrated (1) that females hoarded more than males, (2) that castration increased food hoarding in males, and (3) that administration of testosterone propionate to castrate males inhibited hoarding. The adaptive value of these relationships and their implications for a physiological model of food hoarding in the rodent are discussed.  相似文献

Background:  The Mongolian gerbil model is often used to investigate the interactions between different gastric Helicobacter species and the gastric tissue. A preliminary screening of a gerbil population intended for use in Helicobacter suis infection studies revealed a natural yeast infection in the stomach of these animals. After identification, we have investigated the effect of the gastric yeast infection on the outcome of an experimental H. suis infection in Mongolian gerbils.
Materials and methods:  Yeast cells were isolated from the stomachs of Mongolian gerbils. Identification was done by Internally Transcribed rRNA Spacer 2 Region PCR fragment length analysis. To investigate a possible pathologic role of this yeast, Mongolian gerbils were infected experimentally with this yeast. Co-infection with the newly isolated H. suis was performed to investigate possible interactions between both micro-organisms.
Results:  Kazachstania heterogenica was found colonizing the stomach of Mongolian gerbils, mainly in the antrum. Few pathologic changes were seen in the stomachs of infected animals. Experimental co-infection of gerbils with this yeast and the newly isolated H. suis showed a significant increase in inflammation in animals infected with both micro-organisms compared to animals infected only with H. suis .
Conclusions:  K. heterogenica colonizes the stomach of Mongolian gerbils in exactly the same regions as gastric Helicobacter species. The uncontrolled presence of this yeast in the gerbil stomach can lead to an overestimation of the inflammation caused by Helicobacter in this animal model.  相似文献
