Two new pyrazole-derived ligands, 1-ethyl-3,5-bis(2-pyridyl)pyrazole (L1) and 1-octyl-3,5-bis(2-pyridyl)pyrazole (L2), both containing alkyl groups at position 1 were prepared by reaction between 3,5-bis(2-pyridyl) pyrazole and the appropriate bromoalkane in toluene using sodium ethoxide as base.The reaction between L1, L2 and [MCl2(CH3CN)2] (M = Pd(II), Pt(II)) resulted in the formation complexes of formula [MCl2(L)] (M = Pd(II), L = L1 (1); M = Pd(II), L = L2 (2); M = Pt(II), L = L1 (3); M = Pt(II), L = L2 (4)). These complexes were characterised by elemental analyses, conductivity measurements, infrared, 1H, 13C{1H} NMR and HMQC spectroscopies. The X-ray structure of the complex [PtCl2(L2)] (4) was determined. In this complex, Npyridine and Npyrazole donor atoms coordinate the ligand to the metal, which complete its coordination with two chloro ligands in a cis disposition.  相似文献

Two new 3,5-dimethylpyrazolic derived ligands that are N1-substituted by diamine chains, 1-[2-(diethylamino)ethyl]-3,5-dimethylpyrazole (L1) and 1-[2-(dioctylamino)ethyl]-3,5-dimethylpyrazole (L2) were synthesised. Reaction of the ligands, L1 and L2, with [MCl2(CH3CN)2] yielded [MCl2(L)] (M = Pd(II), Pt(II)) complexes. These complexes were characterised by elemental analyses, conductivity measurements, IR, 1H, 13C{1H} and 195Pt{1H} NMR spectroscopies. The crystal structure of [PdCl2(L1)] was determined by single-crystal X-ray diffraction methods. The structure consists of mononuclear units. The Pd(II) atom is coordinated by a pyrazolic nitrogen, an amine nitrogen and two chlorine atoms in a cis disposition. In this structure, C-H?Cl, C-H?H-C and C-H?C-H intermolecular interactions have been identified.  相似文献

A series of cationic allyl complexes of Pd(II) derived from the diterpene carvone have been synthesised and characterised using the chelating ligands, bipyridine, 1,8-phenanthroline, neocuproin, S,S (-)-Chiraphos and R (+)-Binap as co-ligands. These complexes can be readily converted to new organic products in which one C---H bond of the allyl methyl has been substituted. The structure of a bipyridyl complex as its tetraphenyl borate salt has been determined via X-ray diffraction. 2-D NOESY studies on the Chiraphos and Binap complexes are reported. The Binap, but not the Chiraphos, complex reveals η33η3 isomerisation at ambient temperature.  相似文献

Reaction of the ligands 3-phenyl-5-(2-pyridyl)pyrazole (HL1), 3,5-bis(2-pyridyl)pyrazole (HL2), 3-methyl-5-(2-pyridyl)pyrazole (HL3) and 3-methyl-5-phenylpyrazole (HL4) with [MCl2(CH3CN)2] (M = Pd(II), Pt(II)) or [PdCl2(cod)] gives complexes with stoichiometry [PdCl2(HL)2] (HL = HL1, HL2, HL3), [Pt(L)2] (L = L1, L2, L3) and [MCl2(HL4)2] (M = Pd(II), Pt(II)). The new complexes were characterised by elemental analyses, conductivity measurements, infrared and 1H NMR spectroscopies. The crystal and molecular structure of [PdCl2(HL1)] was resolved by X-ray diffraction, and consists of monomeric cis-[PdCl2(HL1)] molecules. The palladium centre has a typical square planar geometry, with a slight tetrahedral distortion. The tetra-coordinated metal atom is bonded to one pyridine nitrogen, one pyrazolic nitrogen and two chloro ligands in a cis disposition. The ligand HL1 is not completely planar.  相似文献

New Pd(II) and Pt(II) complexes [ML2] (HL=a substituted 2,5-dihydro-5-oxo-1H-pyrazolone-1-carbothioamide) have been synthesized by reacting K2MCl4 (M=Pd, Pt) or Pd(OAc)2 with beta-ketoester thiosemicarbazones. The structures of seven of these complexes were determined by X-ray diffraction. Although all exhibit a distorted square-planar coordination with trans- or (in one case) cis-[MN2S2] kernels, their supramolecular arrangements vary widely from isolated molecules to 3D-networks. The in vitro antitumoral assays performed with two HL ligands and their metal complexes showed significant cytostatic activity for the latter, with the most active [ML2] derivative (a palladium complex) being about sixteen times more active than cis-DDP against the cisplatinum-resistant cell line A2780cisR.  相似文献

The first chiral bis(pyridine) N-C(H)-N pincer ligand, (5S,7S)-1,3-bis(6,6-dimethyl-5,6,7,8-tetrahydro-5,7-methanquinolin-2-yl)benzene (HL) has been synthesized and characterized by a thorough 1H NMR analysis. Reaction of HL with K2[PtCl4] in acetic acid gives [Pt(L)Cl] (1), where L acts as a tridentate N-C-N pincer ligand. The analogous palladium(II) derivatives [Pd(L)Cl] (2), and [Pd(L)(OAc)] (3), were first prepared through a transmetalation reaction between Pd(OAc)2 and the organomercury compound [Hg(L)Cl] (4). The structures of compounds 1 (Pt) and 2 (Pd), as determined by X-ray diffraction, are reported and compared. Compound 2 can also be obtained from Na2[PdCl4] and HL in refluxing acetic acid, i.e., under the same conditions used for compound 1. Apparently, this is the first palladium pincer derivative of a 1,3-bis(pyridyl)benzene ligand synthesized by direct C-H activation.The neutral complexes 1-3 are catalysts of modest activity, but devoid of enantioselectivity in the Heck reaction between iodobenzene and methyl acrylate and in the aldol condensation of benzaldehyde with methyl isocyanoacetate.  相似文献

Three novel cisplatin analogues were synthesized, designed according to an approach which violates the “classical” structure-activity relationship, by replacing the diamine ligands with a planar N donor heterocycle giving a sterically hindered complex. Moreover, the sterical hindrance of antitumor drug candidates potentially makes them less susceptible to deactivation by sulphur-containing proteins and helping to overcome resistance mechanisms. The resulting mononuclear complexes of sterically hindered polidentate heterocyclic N ligands [PtCl(bbp)]Cl (1) [bbp = 2,6-bis(2-benzimidazolyl)pyridine], [PtCl2(dptdn)](H2O) (2) [dptdn = sodium 5,6-diphenyl-3-(2′-pyridyl)-1,2,4-triazine-4″,4″′-disulfonate] and [(dptdn)(dpt)Pt]Cl2(H2O) (3) [dpt = 5,6-diphenyl-3-(2′-pyridyl)-1,2,4-triazine] have been prepared and structurally characterised. Both neutral and ionic complexes are present, with monofunctional (1) and bifunctional Pt(II) moieties (2) and coordinatively saturated Pt(II) ions in the mixed ligand complex (3), whose size and shape enable them to behave as novel scaffolds for DNA binding. All complexes were tested “in vitro” for their biological activity on human HT29 colorectal carcinoma and HepG2 hepatoma cells. The complexes (1) and (3), endowed with a positive charge, showed a potent cytotoxic activity and reduced cell viability with an efficacy higher than that of cisplatin; whilst the neutral bifunctional compound (2) was inactive. IC50 values have been calculated for the active compounds. The cytotoxic effects were confirmed by the accumulation of treated cells in subG0/G1 phase of cell cycle, by the loss of mitochondrial potential (Δψm) and by the chromatin condensation or fragmentation observed by means of fluorescence microscopy after Hoechst 33258 nuclear staining. A study on intracellular platinum uptake in HT29 cell line has been also performed and data obtained strongly suggest that the cytotoxicity of new tested complexes reported in this work is based on a different pharmacodynamic pattern with respect to cisplatin.  相似文献

An array of poly- and mononuclear complexes of Pt(II) with polypyridyl ligands is reported. The framework complexes [(PtCl(2))(2)(bpp)(2)(micro-PtCl(2))](H(2)O)(2) [bpp=2,3-bis(2-pyridyl)pyrazine], [PtCl(2)(micro-tptz)PtClNCPh]Cl [tptz=2,4,6-tris(2-pyridyl)-1,3,5-triazine], and mononuclear PtCl(2)(NH(2)dpt) [NH(2)dpt=4-amino-3,5-bis(2-pyridyl)-1,2,4-triazole] have been prepared and structurally characterized. Both neutral and ionic complexes are present, with bifunctional and monofunctional Pt(II) moieties, whose size and shape enable them to behave as novel scaffolds for DNA binding. Pt(II) complexes were tested for their biological activity. Cell viability assay and flow cytometric analysis demonstrated that these complexes, particularly [PtCl(2)(micro-tptz)PtClNCPh]Cl, were effective death inducers in human colon rectal carcinoma HT29 cells and their cytotoxic activity was higher than that exerted by cisplatin. Morphological analysis of treated HT29 cells, performed by fluorescence microscopy after Hoechst 33258 staining, showed the appearance of the typical features of apoptosis. Moreover, our results suggested that mitochondria are involved in apoptosis induced by Pt(II) complexes in HT29 cells as demonstrated by dissipation of mitochondrial transmembrane potential.  相似文献

Pt(II) and Pd(II) compounds containing the free radical 4-aminoTEMPO (4amTEMPO) were synthesized and characterised by X-ray diffraction methods. The disubstituted complexes cis- and trans-Pt(4amTEMPO)2I2 were studied. The trans isomer was prepared from the isomerisation of the cis analogue. The two Pd(II) compounds trans-Pd(4amTEMPO)2X2 (X = Cl and I) were also characterised by crystallographic methods. A mixed-ligand complex cis-Pt(DMSO)(4amTEMPO)Cl2 was synthesized from the isomerisation of the trans isomer in hot water. Its crystal structure was also determined. In all the complexes, the 4amTEMPO ligand is bonded to the metal through the -NH2 group, since the nitroxide O atom is not a good donor atom for the soft Pt(II) and Pd(II) metals. The conformation of the 4-aminoTEMPO ligand was compared to those of the few reported structures in the literature.  相似文献

The crystal and molecular structures of [PdCl2(L)] (L = L2, L3, L4) were resolved by X-ray diffraction, and consist of monomeric cis-[PdCl2(L)] molecules. Treatment of [PdCl2(L)] (L = L2, L4) with NaBF4 and pyridine (py) and treatment of the same complexes with AgBF4 and triphenylphosphine (PPh3) yielded [Pd(L)(py)2](BF4)2 and [Pd(L)(PPh3)2](BF4)2 complexes, respectively. Dinaphthylmethylarsine complexes of palladium(II) and platinum(II) with the formulae [MX2L2] (M = Pd, Pt; L = di(1-naphthyl)methylarsine = Nap2AsMe and X = Cl, Br, I), [M2Cl2(μ-Cl)2L2], [PdCl(S2CNEt2)L], [Pd2Cl2(μ-OAc)2L2] and [MCl2(PR3)L] (PR3 = PEt3, PPr3, PBu3, PMePh2) have been prepared. These complexes have been characterized by elemental analyses, IR, Raman, NMR (1H, 13C, 31P) and UV-vis spectroscopy. The stereochemistry of the complexes has been deduced from the spectroscopic data. The crystal structures of trans-[PdCl2(PEt3)(Nap2AsMe)] and of [Pd(S2CNEt2)2], a follow-up product, were determined. The UV-vis spectra of [MX2L2] complexes show a red shift on going from X = Cl to X = I. The complexes [PdX2L2] and [PtX2L2] are strongly luminescent in fluid solution and in the solid at ambient temperature.  相似文献

Reaction of [PdCl2(CH3CN)2] with N-alkylaminopyrazole (NN′) ligands, 1-[2-(ethylamino)ethyl]-3,5-dimethylpyrazole (deae), 1-[2-(ipropylamino)ethyl]-3,5-dimethylpyrazole (deai), and 1-[2-(tbutylamino)ethyl]-3,5-dimethylpyrazole (deat) in the presence of AgBF4 led to [Pd(NN′)2](BF4)2. These palladium(II) compounds were characterised by elemental analyses, conductivity measurements, IR, 1H and 13C{1H} NMR spectroscopies. The NMR studies of the complexes prove the rigid conformation of the ligands when they are complexed. The X-ray analyses of one representative complex, [Pd(deat)2](BF4)2, is reported. The crystal structure consists of discrete centrosymmetric cations involving a coordinated Pd(Npz)2(Namino)2core with planar geometry and anions.  相似文献

Treatment of the ligands 1,8-bis(3,5-dimethyl-1-pyrazolyl)-3,6-dithiaoctane (bddo), 1,9-bis(3,5-dimethyl-1-pyrazolyl)-3,7-dithianonane (bddn), and 1,6-bis(3,5-dimethyl-1-pyrazolyl)-2,5-dithiahexane (bddh) with several platinum starting materials as K2PtCl4, PtCl2, [PtCl2(CH3CN)2] and [PtCl2(PhCN)2] was developed under different conditions. The reactions did not yield pure products. The ratio of the NSSN, NS, SS, NN, and 2NS isomers has been calculated through NMR experiments. Treatment of the mixtures of complexes with NaBPh4 affords [Pt(NSSN)](BPh4)2 (NSSN = bddo, bddn). These Pt(II) complexes have been characterised by elemental analyses, conductivity measurements, IR and 1H and 13C NMR spectroscopy. The X-ray structures of the complexes [Pt(NSSN)](BPh4)2 (NSSN = bddo, bddn) have also been determined. In these complexes, the metal atom is tetracoordinated by the two azine nitrogen atoms of the pyrazole rings and two thioether sulfur atoms. When the [Pt(NSSN)](BPh4)2 (NSSN = bddo, bddn) complexes were heated under reflux in a solution of Et4NBr in CH2Cl2/CH3OH (1:1), a mixture of isomers was obtained.  相似文献

Treatment of complex 4 in the presence of AgBF4 in CH2Cl2/methanol (3:1) gives [PtCl(ddae)](BF4) (5). These Pt(II) complexes have been characterised by elemental analyses, conductivity measurements and IR, 1H, 13C{1H}, and 195Pt{1H} NMR spectroscopies. The 1H NMR spectroscopic studies of the complexes prove the rigid conformation of the ligands when they are complexed. The solid-state structure of complex 1 was determined by single crystal X-ray diffraction methods. The deae ligand is coordinated through the Npz and Namino atoms to the metallic centre, which completes its coordination with two chlorine atoms in cis disposition.  相似文献

Several novel dimers of the composition [M2Cl4(trans-dppen)2] (M=Ni (1), Pd (2), Pt (3)) containing trans-1,2-bis(diphenylphosphino)ethene (trans-dppen) have been prepared and characterized by X-ray diffraction methods, NMR spectroscopy (195Pt{1H}, 31P{1H}), elemental analyses, and melting points. The intramolecular [2+2] photocycloaddition of the two diphosphine-bridges in 3 produces [Pt2Cl4(dppcb)] (4), where dppcb is the new tetradentate phosphine cis,trans,cis-1,2,3,4-tetrakis(diphenylphosphino)cyclobutane. Neither 1 nor the free diphosphine trans-dppen shows this reaction. In the case of 2 the photocycloaddition is slower than in 3. This difference can be explained by the shorter distance between the two aliphatic double bonds in 3 than in 2, but also different transition probabilities within ground and excited states of the used metals could be involved. Furthermore, variable-temperature 31P{1H} NMR spectroscopy of 2 or 3 reveals a negative activation entropy of 2 for the [2+2] photocycloaddition, but a positive of 3. The removal of chloride from 4 by precipitating AgCl with AgBF4, and subsequent treatment with 2,2′-bipyridine (bipy) or 1,10-phenanthroline (phen) leads to [Pt2(dppcb)(bipy)2](BF4)4 (5) and [Pt2(dppcb)(phen)2](BF4)4 (6), respectively. In an analogous reaction of 4 with PMe2Ph or PMePh2, [Pt2(dppcb)(PMe2Ph)4](BF4)4 (7) and [Pt2(dppcb)(PMePh2)4](BF4)4 (8) are formed. Complexes 1–8 show square–planar coordinations, where the compounds 4–8 have also been characterized by the above mentioned methods together with fast atom bombardment mass spectrometry (7, 8). The crystal structure of 4 reveals two conformations, which arise from an energetic competition between the sterical demands of dppcb and an ideal square–planar environment of Pt(II). The free tetraphosphine dppcb can be obtained easily from 4 by treatment with NaCN. It has been characterized fully by the above methods including 13C{1H} and 1H NMR spectroscopy. The X-ray structure analysis shows the pure MMMP-enantiomer in the solid crystal, which is therefore optically active. This chirality is induced by a conformation of dppcb, where all four PPh2 groups are non-equivalent. Variable-temperature 31P{1H} NMR spectroscopy of dppcb confirms this explanation, since the single signal at room temperature is split into two doublets at 183 K. The goal of this article is to demonstrate the facile production of a new tetradentate phosphine from a diphosphine precursor via Pt(II) used as a template.  相似文献

This work describes the reactivity of compounds [Pd(dmpz)2(Hdmpz)2] (A) (dmpz = 3,5-dimethylpyrazolate, Hdmpz = 3,5-dimethylpyrazol) and [Pd2(μ-dmpz)2(dmpz)2(Hdmpz)2] (B) towards several dicarboxylic acids and also towards perchloric acid. The compounds [Pd(Hdmpz)4](O2C-(CH2)n-CO2H)2 [n = 1 (1), 3 (2)] have been obtained by treatment of [Pd(dmpz)2(Hdmpz)2] (A) with two equivalents of malonic (HO2C-CH2-CO2H) and glutaric (HO2C-(CH2)3-CO2H) acids. The X-ray study on a crystal of [Pd(Hdmpz)4](O2C-(CH2)3-CO2H)2 (2) revealed that the glutarate anions link to the cationic complex [Pd(Hdmpz)4]2+ through the carboxylate group by charge-assisted N-H(+)?O(−) hydrogen bonds. Additionally, the carboxylate anions form uncommon dimeric rings on both sides of the metal complex via a pair of O-H?O hydrogen bonds, yielding a hydrogen bonded polymeric chain with alternating inorganic [Pd(Hdmpz)4]2+ and organic fragments. The dinuclear complexes [Pd2(μ-dmpz)2(O2C-(CH2)n-CO2)(Hdmpz)2] [n = 0 (5), 1 (6)] were obtained from equimolar amounts of [Pd2(μ-dmpz)2(dmpz)2(Hdmpz)2] (B) and the corresponding dicarboxylic acid, HO2C-(CH2)n-CO2H (n = 0, 1). However, the synthesis of 5 and 6 requires two steps, the protonation of both terminal dmpz groups in B with HClO4 to give [Pd2(μ-dmpz)2(Hdmpz)4](ClO4)2 (4) and the subsequent treatment of this cationic palladium complex with salts of the corresponding dicarboxylic acids. The X-ray structures of compounds 5 and 6 are reported. Both in 5 and 6, the Pd2N4 ring shows a typical boat-like conformation and the metal atoms are separated in about 3.3 Å. Both 5 and 6 are asymmetric and contain two Hdmpz groups - H-bond donors - at one end, and two CO groups from the dicarboxylate anion - H-bond acceptors - at the other, in such a way that the donor end of one molecule links with the acceptor end of its neighbour forming a hydrogen-bonded polymeric chain. The synthesis and X-ray study of compounds [Pd(Hdmpz)4](ClO4)2 (3) and [Pd2(μ-dmpz)2(Hdmpz)4](ClO4)2 (4), obtained by reaction of [Pd(dmpz)2(Hdmpz)2] (A) and [Pd2(μ-dmpz)2(dmpz)2(Hdmpz)2] (B) with two equivalents of perchloric acid, are also reported.  相似文献

The solid state structure of complex 1 was determined by X-ray diffraction studies. The bdmae ligand is coordinated through the two Npz atoms to the metal atom, which completes its coordination with two chlorine atoms in a cis disposition.Treatment of the corresponding ligand with [PdCl2(CH3CN)2] in 1:1 M/L ratio in the presence of AgBF4 and metathesis with NaBPh4 in CH2Cl2/CH3OH (3:1) gave [PdCl(bdmae)](BPh4) (3), and in the presence of NaBPh4 in CH2Cl2/CH3CN (3:1) gave [PdCl(bdmai)](BPh4) (4). Complexes 1 and 2 were again obtained when complexes 3 and 4 were heated under reflux in a solution of Et4NCl in acetonitrile. These Pd(II) compounds were characterised by elemental analyses, conductivity measurements, IR, 1H and 13C{1H} NMR, HMQC and NOESY spectroscopies. The NMR studies of the complexes prove the rigid conformation of the ligands when they are complexed.  相似文献

The coordination chemistry of the ligand bis[2-(3,5-dimethyl-1-pyrazolyl)ethyl]ether (L1) was tested in front of Pd(II) and Pt(II). Complexes cis-[MCl2(L1)] (M=Pd(II) and Pt(II)) were obtained, due to the chelate condition of the ligand and the formation of a stable 10-membered ring. The crystal structure of cis-[PtCl2(L1)] was resolved by X-ray diffraction. Treatment of [PdCl2(L1)] or [Pd(CH3CN)4](BF4)2 with AgBF4 in the presence of L1 gave the complex [Pd(L1)2](BF4)2. The initial cis-[PdCl2(L1)] was recovered by reacting [Pd(L1)2](BF4)2 with an excess of NEt4Cl. Reaction of [Pt(CH3CN)4](BF4)2 (generated in situ from [PtCl2(CH3CN)2] and AgBF4 in acetonitrile) with ligand L1 yields complex [Pt(L1)2](BF4)2.  相似文献

The ligands 1-hydroxymethylpyrazole (hl1), 1-(2-hydroxyethyl)pyrazole (hl2) and 1-(3-hydroxypropyl)pyrazole (hl3) react with [PdCl2(CH3CN)2] to give trans-[PdCl2(hl)2] compounds. Due to a hindered rotation around the Pd-bond, these compounds present two different conformations in solution: anti and syn. The conformation presented depends on the relative disposition of the hydroxyalkylic chains of the two pyrazolic ligands. The present study was carried out on the basis of NMR experiments. The present paper reports the crystal structure of trans-[PdCl2(hl2)2]. Aiming to contribute to the design of new antitumoral drugs, we synthesized new hydrophilic Pt(II) complexes of general formula [PtCl2(N,N′)] containing nitrogen bidentate amine-imine and di-imine ligands derived from glucose. Some chemical properties were discussed. The X-ray molecular structure of [PtCl2(α-d-glucopyranoside-methyl-6-deoxy-6(2-(methylimino)methyl)pyridine) (D) was reported. [PtCl2(β-d-glucopyranosylimine-N-(2-pyridinylmethyl))] (A), which is well-soluble both in organic solvents and in water, was tested for cytotoxicity.  相似文献
