Immunohistochemical localization of sulfhydryloxidase (SOx) has been examined in the testis of the Axolotl (Ambystoma mexicanum). The urodelan testis contains germ cells in various phases of differentiation from primordial germ cells to mature spermatozoa. SOx immunoreactivity is present in mitochondria of primordial germ cells and primary spermatogonia and declines within the population of secondary spermatogonia, suggesting, that the antibody used to localize SOx may serve to estimate the developmental stage of spermatogonia towards meiosis, since more undifferentiated cells react positively. Intensity of immunostaining increases again in spermatocytes and becomes most intense in early round spermatids correlating on ultrastructural level with an accumulation of numerous mitochondria in that part of the cytoplasm, where the acrosome vesicle is formed. Mature sperm are immunonegative. Additionally, Leydig cells within the glandular tissue are stained by the antibody. Thus the distribution pattern of SOx immunoreactivity principally resembles that in the mammalian testis found during ontogenesis or in the adult seminiferous epithelium. The possible functional significance of mitochondrial SOx in germ cells and Leydig cells is discussed. These results suggest, that the amphibian testis is a model for experimental problems dealing with the investigation of germ cells in various developmental phases including very undifferentiated premeiotic germ cells. The cystic testis may be of value in studying influences of various experimental conditions on varied homogeneous populations of germ cells.  相似文献

Sensilla on the aedeagus of the ground beetle Carabus (Ohomopterus) arrowianus arrowianus (Breuning) were investigated by scanning electron microscopy. The pylorus is a segment of the hindgut of great morphological diversity, which is subdivided into the posterior interstitial ring, pyloric cone, and pyloric valve. There is no morphological information on Bombyx mori (L.) hybrids, which is an insect of economic interest to Brazil. Consequently, this study aimed to analyze the morphology of the pylorus of hybrid larvae of fifth instars of B. mori from crossbreeding of Chinese and Japanese races. These were anesthetized, and the pylorus was fixed and processed by light microscopy and scanning electron microscopy. The pylorus presented as a separate compartment, consisting of a simple epithelium, with large variation in cell morphology, covered on the luminal side by an intima with specializations in some areas, the spicules. The musculature was formed by bundles of striated muscle fibers in longitudinal and circular arrangement. These fibers were multinucleated with central and peripheral nuclei, operating in peristalsis. The morphology of the pylorus is a reflection of functional dynamism, and this study provided a better understanding of its physiology, which promotes the regulated movement of the bolus, preventing reflux and directing it towards the ileum in the formation of fecal pellets.  相似文献

Powdery mildew (Sphaerotheca mors-uvae) severely infects young shoots, stems and fruits of gooseberry (Ribes uva-crispa). Environmental friendly and biological control measures are being sought throughout the world. Especially in organic gooseberry growing effective control measures are needed, because powdery mildew infections may result in a total loss of the crop. In organic currant growing the number of adequate control methods is very limited. Sulphur as a fungicide against powdery mildew in e.g. gooseberry or table grape growing is not recommendable due to possible bleaching of berries and scorching of tender shoots. Various bicarbonate salts are suggested as a good option to control powdery mildew. In a field trial the effect of potassium bicarbonate (Armicarb) on the on the control of powdery mildew of gooseberry was evaluated. Four treatments; i.e. two preventive strategies and two curative strategies, were applied. PLants were sprayed until runoff. The percent of infected fruits and disease severity were assessed. In the unsprayed control plots very high disease incidences were observed; on average more than 90% of the berries were infected with powdery mildew. The weekly (preventive) spray applications with potassium carbonate reduced the disease incidences on fruits significantly. On average approximately 10% of the fruits were affected by powdery mildew. However, the number of spray applications was high. In conclusion, our results indicate that applications of potassium bicarbonate (as Armicarb) are effective in reducing the incidence and severity of American powdery mildew in gooseberry. Early spray applications are necessary to protect berries against powdery mildew infections. Future research will focus on reducing the number of applications, e.g. warning models based on powdery mildew of rose (Sphaerotheca pannosa).  相似文献

The authors report a new case of sclero-atrophic keratodermal genodermatosis of the extremities, which is frequently degenerative. This condition, described by Huriez et al. in 1967, is characterized by symmetric lesions of the hands and feet, sclerodactyly, keratodermia, onychopathy and hypohidrosis. This disease, which is of dominant autosomal transmission, is associated with the MNSs trait, the two genes being probably located on chromosome No. 2.  相似文献

The low density lipoprotein receptor-related protein (alpha(2)MR/LRP) is a cell surface receptor which is present on most cells and tissues. We show that the 85 kDa subunit, containing the transmembrane region and cytoplasmic domain is phosphorylated in vivo. Comparison of the phosphorylation of the low density lipoprotein receptor (LDLR) with a chimeric receptor containing the cytoplasmic domain of the alpha(2)MR/LRP (LDLR/LRP) showed that phosphorylation is exclusive to the cytoplasmic domain. Staurosporine, a general kinase inhibitor, resulted in a 40% lowering of phosphorylation of LDLR/LRP, but did not give rise to measurable changes in its membrane traffic in MDCK cells. The role of phosphorylation on degradation of the receptor was studied using inhibitors of lysosomal and proteasomal degradation. These studies showed that LDLR/LRP was rapidly turned over by proteasomal degradation but that this turnover was also not a consequence of phosphorylation.  相似文献

The neuropeptide (N) glutamic acid (E) isoleucine (I) amide (NEI) injected into the ventral tegmental area (VTA) or intraventricularly (icv) induces excessive grooming behavior (EGB) and motor activity (MA). Here, we studied whether the cholinergic system is involved in the NEI-induced behavior. The present results demonstrate that atropine, a general muscarinic antagonist, injected icv previous to NEI, suppresses the behavior provoked by icv injections of the peptide, whereas the prior icv injection of dyhidro--erythroidine, a general nicotinic antagonist, did not affect the EGB and MA induced by the peptide. From the experimental evidence, it is suggested that NEI may act specifically on a cholinergic afferent to dopaminergic cells. Also, the results appear to indicate that a neural target, different from the dopamine system, may be activated by the peptide to elicit behavioral changes, such as EGB.  相似文献

The phylogenetic relationship between Nandinia binotata and Feloidea is analysed by the cladistic method, based on a literature review of osteological characters used in systematic works on carnivores for more than a century. The reduced or lost postglenoid foramen is a synapomorphy that define Nandinia and Feloidea as a monophyletic group. Nandinia does not have an ectotympanic septum in the bulla nor a paroccipital process nested with the posterior wall of the bulla, which are autapomorphies for the Feloidea. Thus it is hypothesized that Nandinia binotata has a sister-group relation to Feloidea. The cartilaginous caudal entotympanic is an autapomorphy for Nandinia.  相似文献

Dendrocacalia crepidifolia Nakai (Compositae, Senecioneae), the only species of this arboreal genus endemic to Haha Island in the Bonin Islands, was found to be dioecious. Male flowers differ from female ones in having a stunted style (style in female exserted from corolla and deeply bifurcating) and anthers filled with fertile pollen (anthers in the female lacking pollen). The size of the corolla and number of florets per head were similar between male and female flowers. The crown area of this arboreal species was also similar in male and female plants. The sex ratio was 0.55 male, not significantly different from 0.5. Both sexes produced nectar of similar sugar concentration (ca. 50%). The flowers are pollinated by feral honeybees (Apis mellifera), but they are thought to have been pollinated by small, lesshairy, endemic solitary bees before honeybees were introduced and subsequently became the dominant bee species on the island. The evolution of dioecy ofDendrocacalia on the island is thought to stem from the deleterious effects of inbreeding that are inherent in plants with geitonogamy. The increased geitonogamy on the island has resulted from increased woodiness (i.e., increased number of flowers per plant) and the original dependence on endemic bee pollinators, which are now endangered.  相似文献

Summary Pardosa amentata adopts a sit-and-wait predation strategy with periodic changes of site. The metabolic costs of this strategy were evaluated using a Gilson respirometer. The respirometer was run at the mean temperatures prevailing month by month in the litter layer from which the experimental animals were collected and equations relating standard respiration rate to spider weight were established for each temperature. The mean value of b, the exponent relating weight to respiration rate, was 0.8011. Active respiration rate was determined at each temperature by using larger experimental flasks containing small glass beads which, in conjunction with the shaker mechanism on the respirometer, ensured that the spiders in the respirometer flasks maintained a fairly consistent level of locomotory activity. It was found that the measured active respiration rate was on average 3.17 times the standard rate. The two respiration rates were combined in the appropriate proportions to calculate the daily energy expenditure of an adult spider exhibiting its normal pattern of activity at different temperatures. It is shown that as locomotory activity is very limited in duration in P. amentata, the associated active respiration rate accounts for only a small proportion of daily respiratory energy losses, 0.36% at 5°C, 0.69% at 10°C, and 1.01% at 15°C.  相似文献

The fixation of trans-(NH3)2Cl2 Pt(II) to poly(I)·poly(C) at low rb (< 0.05) leads to the formation of two complexed species. The major species (ca. 82% of bound platinum) involves coordination of platinum to a single hypoxanthine base, while the other species involves coordination of two hypoxanthine bases, which are either far apart on the same strand or on separate poly(I) strands, to the platinum. These same two species are found after reaction with poly(I), as are two other species throughout the entire rb range studied (rb = 0–0.30). The latter two species are assigned to trans-Pt bound to two bases on a poly(I) strand with (a) one or (b) two free bases between the two bound bases. These two species, (a) and (b), account for ca. 35% of the bound platinum, although the 1:1 species remains dominant (ca. 55%). These two additional species are observed at high rb (>0.075) after reaction with poly(I)·poly(C) but as very minor species. They are formed by reaction with melted poly(I) loops. Also at high rb, we have observed a shifted cytidine H5 resonance arising from interaction of trans-Pt with a melted loop of poly(C). Most probably, this arises from an intramolecular poly(I) to poly(C) crosslink. Results from the reaction of trans-Pt with poly(C) are presented for comparison.  相似文献

The primary pollinator of Polynesian heliconias,Heliconia laufao andH. paka, is the Wattled Honeyeater,Foulehaio carunculata. This report is the first documentation of pollination by honeyeaters in the genusHeliconia and the first record ofF. carunculata as a pollinator of a plant species. The Polynesian heliconias bear inflorescences that produce 2–4 hermaphroditic flowers per day for a period of 2–3 months. Each flower secretes abundant nectar (125–184 l) with low sugar concentration (15–18% sucrose-equivalents, weight per weight basis) which is available at anthesis just before dawn. Ninety percent of flower visits occur between anthesis and mid-morning. The honeyeaters perch on inflorescence bracts, and probing of the flower results in pollen deposition on the head and bill from where pollen is transferred between flowers. No statements on compatibility can be made forHeliconia paka; however,Heliconia laufao appears to be self-compatible. Calculations of energetic values of nectar of the Polynesian heliconias and Daily Energy Expenditures ofF. carunculata suggest that populations ofH. laufao andH. paka serve as rich energy resources for their pollinators.  相似文献

A 1H-NMR investigation was carried out on the tetranucleotides U-m6(2)A-U-m6(2)A and m6(2)A-m6(2)A-U-m6(2)A (m6(2) = N6-dimethyladenosine) as well as on the hybrid trinucleotide dA-r(U-A). An extensive comparison with m6(2)A-U-m6(2)A and other relevant compounds is made. Previous proton NMR studies on trinucleotides have shown that purine-pyrimidine-purine sequences prefer to adopt a mixture of states which have as a common feature that the interior pyrimidine residue bulges out, whereas the flanking purine residues stack upon each other. A stacking interaction on the 3' side of the bulge is known to have no measurable effect on the bulge population. Chemical-shift data, ribose ring conformational analysis and information from NOE experiments now show unambiguously that the moderate U(1)-m6(2)A(2) stack in U-m6(2)A-U-m6(2)A diminishes the population of bulged-out structures in favour of a regular stack. This tendency towards conformational transmission in the downstream 5'----3' direction is fully confirmed by the fact that the strong m6(2)A(1)-m6(2)A(2) stack in the tetranucleotide m6(2)A-m6(2)A-U-m6(2)A virtually precludes the formation of bulged-out structures. The conformational characteristics of dA-r(U-A) appear comparable with those of m6(2)A-U-m6(2)A, which indicates that the presence of a 2'-hydroxyl group in the first purine residue is not a necessary prerequisite for the formation of a bulge.  相似文献

Scanning electron microscopy is used to describe the ultrastructure of morphological features related to feeding, locomotion, respiration, and pigmentation in second- and third-instar larvae of Ephydra hians (Ephydridae: Diptera). Using larvae fixed with the head extended, the ultrastructural details of the mandibles, maxillary papilli, and antennae are described for the first time. The larvae possess finger-like projections at the anterior spiracle and a distensible bifurcated siphon at the posterior spiracle. Slit-like openings are found on each. The second- and third-instar larvae can be differentiated on the basis of dorsal pigmentation associated with extensive patches of spines. The larvae have regional specializations in spine distribution, particularly on the ventral and dorsal body surfaces, and the prolegs. © 1994 Wiley-Liss, Inc.  相似文献

Homoploid hybrid speciation has traditionally been considered a rare event, dependent on the establishment of both a novel, balanced genotype and reproductive isolating barriers between the new species and its progenitors. However, more recent studies have shown that synthetic hybrids converge toward the chromosomal structure of natural hybrids after only a few generations, suggesting that this phenomenon may be more frequent than previously assumed. Here, the possibility that the diploid hybrid species Helianthus deserticola arose from more than one hybrid speciation event was investigated using patterns of variation from cpDNA, 18 nuclear microsatellite loci, and population interfertility. Helianthus deserticola contains cpDNA haplotypes characteristic of both parental species, is polyphyletic with one parental species based on nine microsatellite loci, and has a high degree of interfertility among populations. The data are consistent with either a single origin followed by introgression with the parental species or multiple origins. Analysis of microsatellite variation places the origin of H. deserticola between 170?000 and 63?000 years before present, making it unlikely that anthropogenic disturbances influenced its origin. Finally, the hybrid species generally has lower levels of genetic diversity but higher levels of differentiation among populations than either parental species.  相似文献

Recent studies on the IF(1) inhibitor protein of the mitochondrial F(1)F(0)-ATPase from molecular biochemistry to possible pathophysiological roles are reviewed. The apparent mechanism of IF(1) inhibition of F(1)F(0)-ATPase activity and the biophysical conditions that influence IF(1) activity are summarized. The amino acid sequences of human, bovine, rat and murine IF(1) are compared and domains and residues implicated in IF(1) function examined. Defining the minimal inhibitory sequence of IF(1) and the role of conserved histidines and conformational changes using peptides or recombinant IF(1) is reviewed. Luft's disease, a mitochondrial myopathy where IF(1) is absent, is described with respect to IF(1) relevance to mitochondrial bioenergetics and clinical observations. The possible pathophysiological role of IF(1) in conserving ATP under conditions where cells experience oxygen deprivation (tumor growth, myocardial ischemia) is evaluated. Finally, studies attempting to correlate IF(1) activity to ATP conservation in myocardial ischemic preconditioning are compared.  相似文献

A full-sib mapping population was typed for Taql Southern blot RFLPs at the ovine alpha-inhibin (INHA), beta(A)-inhibin/activin (INHBA), and beta(B)-inhibin/activin (INHBB) loci. The most likely positions were found by genetic linkage analysis. INHA was placed on ovine chromosome 2q, between microsatellite markers BM6444 and INRA135, while INHBB was located 39 cM proximal to INHA between markers BMS1126 and BMS2626. The INHBA gene, a comparative mapping anchor locus, mapped to chromosome 4, between microsatellites OARCP26 and BMS719. These data confirm and refine previous chromosomal assignments by in situ hybridization, provide comparative mapping information, and identify microsatellite markers suitable for QTL association studies with inhibin/activin genes in ruminants.  相似文献
