Outcross, self- and mixed pollinations were performed inCrocus vernus subsp.vernus, a species with bicellular pollen, dry stigmas and hollow style. No differences were noted among the above pollinations concerning the germination of pollen and the growth of pollen tubes until the top of ovary. Within 45 min after pollinations 62% of pollen grains germinated. Pollen tubes penetrated the papilla cuticle extending along the papilla wall; on entry into stigmatic lobes they continued growth in the stylar secretion to ovarian locules. Here, however, self-pollen tubes failed to reach or to enter the ovule micropyle; while pollen tubes from either outcross- or mixed pollinations grew until fertilizing ovules. These observations gave evidence of a self-incompatibility system inCrocus, which appeared to be neutralized by mentor effect. The ovary as site of incompatibility response is discussed.  相似文献

The stigmatic exudates and pollen grains of five unrelated specieswere tested for sugars. Glucose, fructose, and sucrose werefound in the stigmatic fluid of Yucca aloifolia L. and glucoseand fructose in that of Oenothera adrummondii Hook. In in vitroexperiments with Y. aloifolia pollen, high germination percentageswere obtained in artificial media containing glucose or sucrose.Fructose, which is present in the stigmatic fluid of the Yuccasp. resulted in high in vitro pollen germination only when borateand calcium were added to the medium. Presence of bound sugarsis indicated in the stigmatic secretion of Citrus aurantiumL. and pollen of the single plant tested germinated at a lowpercentage in artificial sugar media. No sugars were detectedin the stigmatic fluids of Hemerocallis fulva L. and Zea maysL. and in these two species in vitro pollen germination in sugarymedia was negligible or absent. The pollen grains of all five species contain sucrose and thoseof Oenothera and Citrus also reducing sugars.  相似文献

 We examined the influence of pollen competitive environment on pollen performance in Mirabilis jalapa. We used the number of pollen grains and the number of pollen tubes per pistil as measures of pollen competition. Pollen germination, pollen tube penetration into the style, and pollen tube growth rates were used as measures of pollen performance. All three measures of pollen performance were affected by the competitive environment. Pollen germination was greatest at intermediate pollen load sizes. The percentage of germinated pollen grains that penetrated the stigma and grew into the style decreased with pollen load size. Pollen tube growth rate in the style was greater and more variable with larger numbers of pollen tubes in the style. Controlling for the degree of selection at the stigma indicated that pollen-pollen or pollen-style interactions were the likely causes of increased growth rates. Esterases, leucine aminopeptidases, catalases, amylases and acid phosphatases diffuse out of intact and ungerminated pollen grains of Oenothera organensis, whether suspended in 1 % sodium chloride or in pollen medium. A total of 15 esterase isozymes are recorded; 5 of them appear within 5 minutes, 8 within 30 minutes, 9 within 2 hours, and 13 within 19 hours. Pollen grains suspended for 19 hours gave much stronger isozyme bands than macerated pollen grains. However, one esterase hand was consistently missing from the 19 hour suspensions, although present in all others. It is suggested as a working hypothesis that the early growth of pollen tubes and possibly even germination of pollen grains may be influenced by the metabolic products of pistillate tissues caused by the diffused pollen enzymes, and that inactivation of these enzymes by stigmatic or stylar components could lead to incompatibility reactions.  相似文献

The rates of germ tube elongation in vitro of pollen from cherry leaf roll virus (CLRV)-infected birch (Betula pendula) did not differ significantly from those of pollen from virus-free trees. Differences in percentage germination of pollen collected from trees at different sites were significant but percentages of germination of pollen from virus-infected and virus-free trees did not differ greatly from one another. In vivo, pollen from infected trees germinated on healthy and CLRV-infected stigmas and callose plugs formed in both types of tissues. However, the extent of callose plug formation was greater in the pollen tubes of virus-free grains germinating in infected stigmas than in reciprocal crosses. CLRV coat antigen was detected by ELISA in stigmatic tissue, from healthy trees, on which virus-carrying pollen grains had germinated. Using fluorescein isothiocyanate conjugated to CLRV-specific γ-globulin, viral coat antigen was detected throughout germ tubes from virus-carrying but not virus-free pollen germinating in vitro. Protoplasts released following Driselase digestion of pollen germ tubes from virus-infected trees contained CLRV antigen detectable by ELISA. During germination of virus-infected pollen there was little synthesis of viral coat protein or nucleic acid but, following inoculation with purified virus particles, protoplasts made from healthy germinating pollen produced increasing amounts of CLRV-specific antigen implying that CLRV replicated in this system.  相似文献

Most species of Dalechampia have expanded stigmatic surfaces that extend from the stylar tip part way down the sides of the elongated styles. Pollen grains landing at the stylar tip grow tubes directly down the style to the ovary. Pollen grains landing on the lateral stigmatic surfaces, however, exhibit indirect pollen-tube growth: the tubes grow first to the stylar tip, bend 180°, and then grow to the ovary. Indirect pollen-tube growth appears to be reproductively important: more than half the arriving pollen lands on the lateral stigmatic surfaces, and virtually all of these grains produce tubes that grow indirectly to the ovary; pollen on the lateral surface fertilizes seeds about as effectively as pollen at the tip (with direct tube growth). Indirect pollen-tube growth may be the result of a correlated response to selection for expanded stigmatic surfaces in Dalechampia, or it may be an adaptation to increase the intensity of pollen competition.  相似文献

Summary A simple procedure is described for the in vitro production of tobacco (Nicotiana tabacum L.) pollen from microspores isolated just before entering mitosis. During a 3-day culture period in a liquid medium containing pyrimidine nucleosides these microspores develop into young pollen grains to the stage of starch deposition. Pollen maturation and transition to dormancy is achieved during a further 2- to 3-day culture period in the same medium stepwise supplemented by a concentrated solution of sucrose and l-proline. Upon transfer of the pollen to a simple germination medium containing sucrose and boric acid, up to 40% of the grains were observed to produce relatively long tubes. The in vitro-matured pollen grains can be stored at-20° C either suspended in 1.17 M sucrose and 100 mM l-proline or separated from the medium on filter paper discs. The stored pollen germinated both in vitro and on the stigma, the pollen tubes grew through the style into the ovary and pollination produced up to 300 viable seeds per pod. The procedure is of interest for pollen developmental studies and various fields of pollen manipulation, such as in vitro pollen selection.  相似文献

Summary Pollen ofPinus silvestris shows a stimulation of germination and pollen tube growth after UV irradiation up to 3,6 erg/cm2. The UV irradiation must occur prior to or in the first 10 min after immersion of the pollen grains into the germination medium.Stimulation occurs only if subsequent growth in visible light (1000 lx, fluorescent lamp) is allowed and can be suppressed by Actinomycin.  相似文献

White dust spread on the leaves and stigmatic surface of Crotalaria pellida L. and Desmodium heteracarpon (L.) DC var. strigosum Meeuwen growing in the midst of a Parthenium hysterophorus L. stand was found to be clusters of Parthenium pollen. The fruit set in those species was poor. It was found that Parthenium pollen grains contained growth inhibitors which inhibited fruit set in many test species when artificially dusted on the stigmatic surface. In culture conditions the germination and pollen tube growth of the test species were inhibited in the presence of Parthenium pollen or their extract. The plant produces on an average 3375 million pollen grains per square metre areas of the stand. The pollen grains disperse in dusters of 600 to 800 each and are carried great distances by wind. Pollen grains deposited on leaves in large quantity also reduce the chlorophyll content in these species.  相似文献

The influence of kanamycin on the percentage of pollen germination and on tube growth of pollen from non-transformed and transformed plants of various species containing a chimaeric kanamycin resistance gene (NPTII) was investigated. Pollen grains isolated from kanamycin resistant plants expressed resistance when germinating in vitro, whereas kanamycin impaired tube growth of pollen from non-transformed plants. Pollen grains from transgenic plants were less sensitive and produced significantly longer tubes. mRNAs of the chimaeric gene are probably presynthesized concurrently with the other mRNAs during microsporogenesis, and kanamycin resistance is expressed by mRNA translation during pollen tube elongation. Summary (1) Histological studies on the stigmas of female flowers of areca indicate the possibility of the stigmatic surfaces being impacted upon by air spora. The presence of spores or hyphae together with pollen grains has been shown in stigmatic surface.(2) Air spora of arecanut plantation chiefly consist ofAspergillus, Penicillium, Actinomycetes, Mucor, Alternaria, Cladosporium andBacteria.(3) The types of fungi, bacteria and actinomycetes adhering to the stigma are isolated.(4) The metabolites of the fungi, bacteria and actinomycetes isolated from the stigmas of arecanut flowers inhibit,in vitro, considerably the germination of pollen grains and rate of growth of pollen tubes. In certain instances there is no germination of the pollen grains in the metabolites of fungi, bacteria and actinomycetes. In view of the results the significance of the air spora in affecting the normal course of pollination and fertilization is discussed.  相似文献

Pollen from 5 plant species (Lycopersicon pimpinellifolium Mill., Hermerocallis minor Mill., Galtonia condicans Decne., Camellia japonica L., and Lathyrus odoratus L.) representing 4 families germinated well in media containing trehalose as the sole carbon source. Data are presented indicating that pollen metabolized this disaccharide for germination and subsequent pollen-tube growth; the sugar was not merely an osmoregulator. An inhibitor of trehalase activity depressed germination in trehalose but not in sucrose. Phloridzin dihydrate, an inhibitor of glucose transport, depressed germination in both disaccharides.Biochemical tests demonstrated that a pollen extract was capable of hydrolyzing trehalose to its constituent glucose monomers. Heat inactivation experiments confirmed the presence of a distinct trehalase having a rigid specificity for its substrate. By this method, trehalase activity was completely distinguishable from the activities of other alpha- and beta-glucosidases and beta-galactosidases. Localization data indicated that the enzyme diffused from intact grains and was probably soluble. The presence of its substrate could not be demonstrated in pollen or in stigmatic or stylar tissues.  相似文献

Pollen allelopathy occurs when pollen toxins inhibit sexual reproduction in heterospecifics. To quantify pollen allelopathic effects in vitro, pollen of Hieracium aurantiacum, H. floribundum, and H. pratense was extracted with double distilled water into concentrations of 0 (control), 0.1, 1, 3, 5, 10, and 25 pollen grains/μl. A subset of these extracts was partitioned by ion-exchange chromatography into acidic, basic, and neutral fractions. Pollen from six sympatric species of Fabaceae (Lotus corniculatus, Medicago sativa, Trifolium hybridum, T. pratense, T. repens, Vicia cracca) and the species of Hieracium was germinated on agar media containing different extract concentrations. As extract concentrations from intact pollen or from acidic fractions increased, mean percent germination of pollen of all Fabaceae decreased nonlinearly. Extract concentrations of 0.1 grains/μl and above inhibited pollen germination in the Fabaceae. In no case was any Hieracium species affected, i.e., there was no autotoxicity. Similar results were obtained using plants from nine populations of H. floribundum and H. pratense across eastern Canada. Observation of floral visitors indicated that there were relatively few movements between Hieracium and the Fabaceae (about 15% of total visits). Hence there is limited opportunity for heterospecific pollen transfer. This was consistent with censuses of stigma, i.e., few pollen grains of Hieracium were found on stigma of the Fabaceae. Thus, pollen allelopathy in Hieracium is unlikely to affect reproductive success in these six species of Fabaceae.  相似文献

Summary Leucaena leucocephala generally produces pods with more than 7–9 seeds. This is regulated by the stigmatic inhibition of pollen grain germination when the pollen grains are less than a critical number in the stigma. This number-dependent inhibition of pollen grain germination is effected by a pH-dependent proteinaceous inhibitor active at the stigmatic pH. Only when the pollen grains in the stigma exceed the critical number, they inactivate the inhibitor by collectively raising the stigmatic pH and thus overcoming the inhibition. The adaptive significance of such pre-fertilization mechanism for the female in inciting mate competition among the pollen grains is discussed. The evolution of en masse pollen grain dispersal units is explained as a sexual selection strategy by males in response to such stigmatic inhibition by females.  相似文献

Grain set failure in wheat, caused by boron (B) deficiency, is associated with poorly developed pollen and anthers. This paper presents results of a study of the effect of B on pollen viability when it was supplied "internally" through the roots and externally in an agar medium for in vitro germination.There was no major effect of B supply to wheat plants on the number of pollen anther-1 or the percentage of pollen with positive reaction to iodine. Pollen germination in the medium was, however, responsive to both internal and external B supply. When B was not added to the medium, germination was poor, regardless of the level of B supplied to the plant, in both a B deficiency sensitive (SW41) and a B deficiency tolerant (Sonora 64) genotypes. The percentage of germinated pollen and length of the pollen tube increased with increasing medium B. With 20–100 mg H3BO3 L-1 in the medium, the percentage of germinated pollen and length of the pollen tube responded positively to increasing B supply to the plant.No difference was found between sensitive and tolerant genotypes in the effect of B on their pollen viability. On the other hand, without added B in the nutrient solution applied to the plant, grain set was depressed in the B deficiency sensitive SW41 and not in the B deficiency tolerant Sonora 64. A difference in B supply to the germinating pollen in the stigma and style is one possible explanation for this variation in the response to B among wheat genotypes.  相似文献

Fresh pollen of Ornithogalum caudatum was germinated in the absence or presence of trans-cinnamic acid (t-CA), trans-cinnamaldehyde (t-CAld) and related compounds, and germination percentage studied. Hydrocinnamic acid, t-CA, and t-CAld reduced germination to 50% of the control at 20–40 mg/liter. Hydrocinnamaldehyde, 3-phenyl-1-propanol and phenylacetic acid gave 50% reduction at 70–80 mg/liter, and caffeic acid, ferulic acid, phenethyl alcohol, coumarin, styrene, phenylalanine or ethyl cinnamate required 200 mg/liter or more for similar reduction in germination percentage. The inhibition of germination by t-CA could not be overcome by indole acetic acid and vice versa, indicating a lack of antiauxin activity.  相似文献

Floral morphology and phenology and the timing of stigmatic receptivity and pollen viability were studied to elucidate the mechanisms by which self-pollination in Lupinus nanus subsp. latifolius is minimized under natural conditions. Pollen germination and pollen tube growth suggest that a physiological self-incompatibility system does not exist. Instead, self-pollination is minimized by protandry and by a collar of peristigmatic hairs, which initially inhibit access of autologous pollen to the stigma. These hairs subsequently wilt, permitting self-pollination of unvisited flowers. Pollen germinates in vivo one day before substantial metabolic enzyme activity can be detected. Citric acid cycle enzymes are not detectable in pollen, but those of anaerobic metabolism are. Beginning on the second day postanthesis, stigmatic secretions exude from weak areas on lateral walls of the elongate epidermal papillae, welling up in the interstices between these cells. This contrasts with the stigmas of other papilionoid legumes, in which secretions accumulate beneath the cuticle covering the stigmatic cells, and the often-thin cuticle must be ruptured before pollen and exudate can come into contact.  相似文献

The effect of medium concentration, pollen grain concentration, pH of the media, light and temperature on the germination of Vin ca rosea pollen grains, and the growth of their pollen tubes in vitro have been studied. The pollen grains germinate best at a sucrose concentration between 14.2% and 30%; when the pollen grain concentration exceeds 800 per 0.0234 ml; at near neutral pH (6.5); in darkness and at a temperature close to 30°. Moreover buffering ions affect the growth of the pollen tubes. Pollen grains remain viable in a wide range of temperatures, and the wall of the pollen grain is capable of withstanding severe osmotic imbalance. Low temperature induces spherical swellings at the tips of the pollen tubes, followed by accumulation of a hyaline plug.  相似文献

对紫花含笑(Michelia crassipes)、灰岩含笑(M.calcicola)及其杂种F1代花粉生活力进行了研究,为基于紫花含笑和灰岩含笑杂种F1代的含笑属观赏植物新品种培育与种质创新提供科学数据及研究资料.研究发现,亲本(紫花含笑和灰岩含笑)新鲜花粉萌发率均可达90%以上,杂种F1代花粉萌发率从38%到79%不等,平均为57.7%,低于双亲.亲本及其杂种F1代花粉萌发的最适温度为25℃,温度过高花粉管的伸长受到抑制,并导致花粉管顶端破裂.亲本及多数杂种F1代的新鲜花粉在100 g/L和150 g/L的蔗糖浓度下萌发率都较高;经-20℃贮藏后的花粉对蔗糖浓度的敏感性要高于新鲜花粉.杂种F1代及其亲本的花粉在离体培养中均会出现双萌发管现象.番红染料对液体培养基中的花粉有致死和染色作用,有利于统计杂种F1代及其亲本的花粉萌发率.  相似文献

Pollination is essential for seed reproduction and for exchanges of genetic information between individual plants. In angiosperms, mature pollen grains released from dehisced anthers are transferred to the stigma where they become hydrated and begin to germinate. Pollen grains of wild-type Arabidopsis thaliana do not germinate inside the anther under normal growth conditions. We report two Arabidopsis lines that produced pollen grains able to in situ precociously germinate inside the anther. One of them was a callose synthase 9 (cs9) knockout mutant with a T-DNA insertion in the Callose Synthase 9 gene (CalS9). Male gametophytes carrying a cs9 mutant allele were defective and no homozygous progeny could be produced. Heterozygous mutant plants (cs9/+) produced approximately 50% defective pollen grains with an altered male germ unit (MGU) and aberrant callose deposition in bicellular pollen. Bicellular pollen grains germinated precociously inside the anther. Another line, a transgenic plant expressing callose synthase 5 (CalS5) under the CaMV 35S promoter, also contained abnormal callose deposition during microsporogenesis and displaced MGUs in pollen grains. We also observed that precocious pollen germination could be induced in wild-type plants by incubation with medium containing sucrose and calcium ion and by wounding in the anther. These results demonstrate that precocious pollen germination in Arabidopsis could be triggered by a genetic alteration and a physiological condition.  相似文献
