Arsenic is a ubiquitously found metalloid that commonly contaminates drinking water and agricultural food. To understand the ecotoxicological effects of arsenic in environment, it is essential to ameliorate the deleterious effects on human and animal health, particularly on the immune response. We investigated the effects of inorganic arsenic (iAs) on the immune response of chicken splenocytes. Both 1 and 10 mM concentrations of sodium arsenite treatment significantly reduced (P < 0.001) splenocyte proliferation and phagocytic activity compared to concanavalin A (ConA) stimulated cells at 24, 48 and 72 h of incubation. Nitrous oxide (NO) production was significantly higher (P < 0.001) at 24 h and subsequently declined in the higher dose group, while there was a gradual decline from 24 to 72 h in the lower dose group. Comparison of two different concentration of arsenic treatment also revealed time dependent differences. Relative quantification of expression of IFN?? and IL2 revealed that both genes were significantly down regulated (P < 0.001) at both concentrations at each time point. iNOS gene was rapidly down regulated in splenocytes at 24 h at the high doses of As treated splenocyte, a gradual decreasing trend at low doses. Down regulation of IL-2 gene expression in response to As was further evidenced by a significant reduction (P < 0.001) in the release of IL-2 into the splenocyte culture medium. We suggest that arsenic, a potent immunotoxic agent, modulates non-specific immune responses and alters the expression of cytokines in a dose and time dependent manner.  相似文献

The present work describes the immune response of wireworm larvae, Agriotes lineatus (L.) (Coleoptera: Elateridae) when challenged with two species of entomopathogenic nematodes, Steinernema feltiae (Filipjev) (Strongyloidea: Steinernematidae) and Heterorhabditis bacteriophora (Poinar) (Rhabiditoidea: Heterorhabditidae). Two main immunological processes including cellular and humoral reactions have been addressed. Total haemocyte counts after infection with H. bacteriophora increased quickly in initial times, but decreased over time post-injection (at 12 and 16 h). Instead, haemocyte numbers after infection with S. feltiae was unchanged in the early stage, but significantly decreased until 16 h post-injection. Plasmatocytes and granulocytes showed more significant changes compared to other haemocytes. The encapsulation response to parasites was significantly different against two nematode species. Particularly, S. feltiae was almost unrecognized by host haemocytes (5.85 % of encapsulated parasites). Assays with H. bacteriophora showed 23.5 % of encapsulated nematodes. From 8 to 12 h after H. bacteriophora infection, an increase in phenoloxidase activity was detected, while in the larvae injected with S. feltiae the enzymatic activity decreased gradually reaching the lowest level 16 h post-injection. This is the first report on the modulation of immune response of wireworm larvae after infection with entomopathogenic nematodes.  相似文献

Coral reefs across the Caribbean are undergoing unprecedented rates of decline in coral cover during the last three decades, and coral recruitment is one potential process that could aid the recovery of coral populations. To better understand the effects of climate change on coral larval ecology, the larvae of Porites astreoides were studied to determine the immediate and post-settlement effects of elevated temperature and associated oxidative stress. Larvae of Porites astreoides were exposed to 27 °C (ambient) and +3.0 °C (elevated temperature) seawater for a short duration of 24 h; then, a suite of physiological parameters were measured to determine the extent of sublethal stress. Following the +3.0 °C treatment, larvae did not show a significant difference in maximum quantum yield of PSII (F v/F m) or respiratory demand when compared to controls maintained at 27 °C. The addition of micromolar concentrations of hydrogen peroxide did not impact respiration or photochemical efficiency. Catalase activity in the larvae increased (>60 %) following exposure to elevated temperature when compared to the controls. Short-term larval survival and settlement and metamorphosis were not affected by increased temperature or the H2O2 treatment. However, the settled spat that were exposed to elevated temperature underwent a 99 % reduction in survival compared to 90 % reduction for the control spat when examined 24 days following the deployment of 4-day-old settled spat on settlement tiles in the field. These results show that short-term exposure to some stressors might have small impacts on coral physiology, and no effects on larval survival, settlement and metamorphosis. However, due to post-settlement mortality, these stressors can cause a significant reduction in coral recruitment.  相似文献

Terrestrial runoff increases siltation and nutrient availability on coastal coral reefs worldwide. However the factors determining stress in corals when exposed to short-term sedimentation, including the interactions between sediments and nutrients, are little understood. We exposed corals to ten different sediment types at environmentally relevant concentrations (33 to 160 mg DW cm−2) and exposure times (12 to 60 h) in laboratory and field experiments. The sediments originated from 2 estuaries, 2 nearshore and one offshore locations and also included ground-up aragonite. For two of these sediments, three grain size fractions were used (silt < 63 μm, fine sand: 63-250 μm, medium sand: 250-500 μm). Sediments were characterised by 19 parameters grouped into “physical”, “organic and nutrient-related” and “geochemical” parameters. Changes in the photosynthetic yield of the coral Montipora peltiformis was measured by pulse-amplitude modulated chlorophyll fluorometry (PAM) as proxy for photophysiological stress from exposure, and to determine rates of recovery. Different sediments exerted greatly contrasting levels of stress in the corals. Our results show that grain size and organic and nutrient-related sediment properties are key factors determining sedimentation stress in corals after short-term exposure. Photophysiological stress was measurable after 36 h of exposure to most of the silt-sized sediments, and coral recovery was incomplete after 48 to 96 h recovery time. The four sandy sediment types caused no measurable stress at the same concentration for the same exposure time. Stress levels were strongly related to the values of organic and nutrient-related parameters in the sediment, weakly related to the physical parameters and unrelated to the geochemical parameters measured. M. peltiformis removed the sandy grain size classes more easily than the silt, and nutrient-poor sediments were removed more easily than nutrient-rich sediments. Anoxia developed on the sediment surfaces of the nutrient-rich silts, which had become slimy and smelled of hydrogen sulphide, suggesting increased bacterial activity. Our finding that silt-sized and nutrient-rich sediments can stress corals after short exposure, while sandy sediments or nutrient-poor silts affect corals to a lesser extent, will help refining predictions of sedimentation threats to coral reefs at given environmental conditions.  相似文献

Several aspects of immune response in insects treated with an organophosphate (pirimiphosmethyl) have been assessed. Both humoral (phenoloxidase activity) and cellular (hemocyte count and encapsulation rate) immunity changes have been studied in larvae of the representatives of two insect orders, the Colorado potato beetle Leptinotarsa decemlineata (Coleoptera, Chrysomelidae) and the wax moth Galleria mellonella (Lepidoptera, Pyralidae). A direct contact with sublethal and half-lethal doses of the insecticide results in stimulation of immune reactions: phenoloxidase activity and encapsulation rate are heightened, and hemocyte count increases.  相似文献

The thermal melanin hypothesis posits that ectothermic individuals of larger size or from colder environments exhibit darker cuticles due to melanin’s efficacy in absorbing solar radiation. However, melanin is also a crucial component of arthropod immunity. Thus, thermal selection for increased cuticle darkness may profoundly influence melanin-based immune function. In this study, we address the relationships between the thermal environment (season length), cuticular melanism and two aspects of melanin-based immunity across nine thermally distinct populations of the cricket Allonemobius socius. We found that season length (i.e. degree days) and body size had a positive association with cuticle melanism in both sexes across populations, supporting the thermal melanism hypothesis. Despite their smaller size, males were found to have darker cuticles and superior melanin-based immunity. This pattern may be the result of additional selection on males due to sex-specific temperature-dependent activities, such as male calling song. Perhaps most interestingly, we found that short season length populations (i.e. colder) exhibited a greater phenoloxidase activity (aspect of the melanin-based immune system) in addition to darker cuticles in both sexes. This pattern is consistent with direct thermal selection on cuticular color, coupled with indirect selection on melanin-based immunity due to pleiotropy. Thus, thermal selection on cuticle darkness appears to indirectly shape the evolution of pathogen resistance in this system, and potentially for other terrestrial arthropod systems whose ranges encompass a significant thermal gradient.  相似文献

Extremely low frequency electromagnetic field (ELF-EMF) is produced extensively in modern technologies. Numerous in vitro and in vivo studies have shown that ELF-EMF has both stimulatory and inhibitory effects on the immune system response. This review was conducted on effects of ELF-EMF on cytokines of innate and adaptive immunity. Mechanisms of ELF-EMF, which may modulate immune cell responses, were also studied. Physical and biological parameters of ELF-EMF can interact with each other to create beneficial or harmful effect on the immune cell responses by interfering with the inflammatory or anti-inflammatory cytokines. According to the studies, it is supposed that short-term (2-24 h/d up to a week) exposure of ELF-EMF with strong density may increase innate immune response due to an increase of innate immunity cytokines. Furthermore, long-term (2-24 h/d up to 8 years) exposure to low-density ELF-EMF may cause a decrease in adaptive immune response, especially in Th1 subset.  相似文献

Coral mechanisms of resilience and resistance to stressors such as increasing sea surface temperature and ocean acidification must first be understood in order to facilitate the survival of coral reefs as we know them. One such mechanism is production of the protective surface mucopolysaccharide layer (SML). In this study, we investigated changes in the thickness of the SML in response to increasing temperature and acidification for the three Caribbean scleractinian coral species of the genus Diploria, which have been shown to exhibit differential resilience to disease and bleaching. Among the three species, Diploria strigosa is known to have a higher susceptibility to disease, Diploria labyrinthiformis is known to bleach more quickly, and Diploria clivosa is relatively unstudied. When temperature was increased from 25 to 31 °C over a 1- or 6-week period, the overall thickness of the SML decreased from 33 to 55 % for all three species. Average SML thickness at 25 °C for all three species ranged from 106 to 156 μm, while average thickness at 31 °C ranged from 64 to 86 μm. SML thickness was significantly different among species at 25 °C, but not at 31 °C. D. labyrinthiformis demonstrated lower fragment mortality due to thermal stress when compared to the other Diploria species. Acidification from pH 8.2 to 7.7 over 5 weeks had no effect on SML thickness for any species. The observed decrease in SML thickness in response to increased temperature might be attributed to a decrease in the production of mucus or an increase in the viscosity of the SML. These findings may help to explain the increased prevalence of coral disease during the warmer months, since increased temperature compromises an important aspect of coral innate immunity, as well as differences in disease and bleaching susceptibilities between Diploria species.  相似文献

Fine sediments, which account for the majority of total fluvial sediment flux, have been suggested to degrade coral reefs on a global scale. Furthermore, sediment impacts can be exacerbated by extreme rainfall events associated with global climate change and anthropogenic nutrient enrichment. We report the findings from a series of mesocosm experiments exploring the effects of short-term sedimentation and nutrient enrichment on the interactions between the hard coral Acropora muricata, the sea anemone Mesactinia ganesis, and the green macroalga Codium edule. Mesocosms were manipulated to simulate either unimpacted reefs or reefs exposed to elevated levels of fine sediments for 10 or 14 days to simulate the effects of heavy rainfall. The first and second experiments were aimed to examine the effects of inorganic and organic sediments, respectively. The third experiment was designed to examine the interactive effects of nutrient enrichment and elevated sediment loads. Neither inorganic nor organic sediment loadings significantly affected the physiological performance of the coral, but, importantly, did reduce its ability to compete with other organisms. Photosynthetic efficiencies of both the green macroalga and the sea anemone increased in response to both sediment loadings when they were simultaneously exposed to nutrient enrichment. While organic sediment loading increased the nitrogen content of the green macroalga in the first experiment, inorganic sediment loading increased its phosphorus content in the second experiment. The coral mortality due to sea anemones attack was significantly greater upon exposure to enriched levels of organic sediments and nutrients. Our findings suggest that the combined effects of short-term sedimentation and nutrient enrichment could cause replacement of corals by sea anemones on certain coral reefs.  相似文献

Mosquito immunity studies have focused mainly on characterizing immune effector mechanisms elicited against parasites, bacteria and more recently, viruses. However, those elicited against entomopathogenic fungi remain poorly understood, despite the ubiquitous nature of these microorganisms and their unique invasion route that bypasses the midgut epithelium, an important immune tissue and physical barrier. Here, we used the malaria vector Anopheles gambiae as a model to investigate the role of melanization, a potent immune effector mechanism of arthropods, in mosquito defense against the entomopathogenic fungus Beauveria bassiana, using in vivo functional genetic analysis and confocal microscopy. The temporal monitoring of fungal growth in mosquitoes injected with B. bassiana conidia showed that melanin eventually formed on all stages, including conidia, germ tubes and hyphae, except the single cell hyphal bodies. Nevertheless, melanin rarely aborted the growth of any of these stages and the mycelium continued growing despite being melanized. Silencing TEP1 and CLIPA8, key positive regulators of Plasmodium and bacterial melanization in A. gambiae, abolished completely melanin formation on hyphae but not on germinating conidia or germ tubes. The detection of a layer of hemocytes surrounding germinating conidia but not hyphae suggested that melanization of early fungal stages is cell-mediated while that of late stages is a humoral response dependent on TEP1 and CLIPA8. Microscopic analysis revealed specific association of TEP1 with surfaces of hyphae and the requirement of both, TEP1 and CLIPA8, for recruiting phenoloxidase to these surfaces. Finally, fungal proliferation was more rapid in TEP1 and CLIPA8 knockdown mosquitoes which exhibited increased sensitivity to natural B. bassiana infections than controls. In sum, the mosquito melanization response retards significantly B. bassiana growth and dissemination, a finding that may be exploited to design transgenic fungi with more potent bio-control activities against mosquitoes.  相似文献

Background

Safety and cellular immunogenicity of rising doses and varying regimens of a poly-epitope vaccine were evaluated in advanced metastatic melanoma. The vaccine comprised plasmid DNA and recombinant modified vaccinia virus Ankara (MVA) both expressing a string (Mel3) of seven HLA.A2/A1 epitopes from five melanoma antigens.

Methods

Forty-one HLA-A2 positive patients with stage III/IV melanoma were enrolled. Patient groups received one or two doses of DNA.Mel3 followed by escalating doses of MVA.Mel3. Immunisations then continued eight weekly in the absence of disease progression. Epitope-specific CD8+ T cell responses were evaluated using ex-vivo tetramer and IFN-γ ELISPOT assays. Safety and clinical responses were monitored.

Results

Prime-boost DNA/MVA induced Melan-A-specific CD8+ T cell responses in 22/31 (71%) patients detected by tetramer assay. ELISPOT detected a response to at least one epitope in 10/31 (32%) patients. T cell responder rates were <50% with low-dose DNA/MVA, or MVA alone, rising to 91% with high-dose DNA/MVA. Among eight patients showing evidence of clinical benefit—one PR (24 months+), five SD (5 months+) and two mixed responses—seven had associated immune responses. Melan-A-tetramer+ immunity was associated with a median 8-week increase in time-to-progression (P = 0.037) and 71 week increase in survival (P = 0.0002) compared to non-immunity. High-dose vaccine was well tolerated. The only significant toxicities were flu-like symptoms and injection-site reactions.

Conclusions

DNA.Mel3 and MVA.Mel3 in a prime-boost protocol generated high rates of immune response to melanoma antigen epitopes. The treatment was well tolerated and the correlation of immune responses with patient outcomes encourages further investigation.  相似文献

Double-stranded RNA (dsRNA) is a virus-associated molecular pattern which induces antiviral innate immune responses and RNA interference (RNAi) in mammals. In invertebrates, RNAi phenomenon has been widely studied, but dsRNA-induced innate immune response is seldom reported. In the present study, two different dsRNAs specific for green fluorescent protein (GFP) and the putative D1 protein of photosystem II (NoPSD) from Nannochloropsis oculata, were employed to challenge Chinese mitten crab Eriocheir sinensis. The temporal changes of phenoloxidase (PO), acid phosphatase (ACP), superoxide dismutase (SOD) and malondialdehyde (MDA) content, as well as the mRNA expression of some immune-related genes were examined in order to estimate the effect of dsRNAs on the innate immunity of E. sinensis. The activities of PO, ACP and SOD significantly increased after dsRNA treatment, whereas malondialdehyde (MDA) content did not change significantly. Among the examined genes, only the mRNA expression of EsALF, an antibacterial peptide in E. sinensis, was significantly up-regulated (about 5 fold, P < 0.05) at 12 h after dsRNA treatment, while no significant expression changes were observed among the other immune genes. The increase of PO, ACP and SOD activities, and mRNA expression level of EsALF after dsRNA stimulation indicate that phenoloxidase, hydrolytic enzyme, antioxidation and EsALF were involved in dsRNA-induced innate immunity, suggesting that broad-spectrum immune responses could be induced by dsRNA in E. sinensis.  相似文献

Nitric oxide (NO) is an important signalling molecule which plays an indispensable role in immunity of all vertebrates and invertebrates. In the present study, the immunomodulation of inducible NO in scallop Chlamys farreri was examined by monitoring the alterations of haemocyte behaviours and related immune molecules in response to the stimulations of LPS and/or with S-Methylisothiourea Sulphate (SMT), an inhibitor of inducible NO synthase (NOS). The total activity of NOS and NO concentration in the haemolymph of scallop C. farreri increased significantly at 3, 6 and 12 h after LPS stimulation respectively, whereas their increases were fully repressed when scallops were treated in the collaborating of LPS and SMT. Meanwhile, some cellular and humoral immune parameters were determined after the stimulation of LPS and SMT to investigate the role of inducible NO in innate immunity of scallop. After LPS stimulation, the highest levels of haemocytes apoptosis and phagocytosis were observed at 24 h (38.5 ± 2.5%, P < 0.01) and 12 h (38.6 ± 0.2%, P < 0.01), respectively, and the reactive oxygen species (ROS) level (5.88 ± 0.90%, P < 0.01) of haemocytes and anti-bacterial activity of haemolymph (10.0 ± 2.2%, P < 0.01) all elevated dramatically at 12 h. Although the activity of lysozyme and phenoloxidase (PO) in haemolymph both declined at 48 h (93.0 ± 6.3 U mgprot?1, 0.40 ± 0.06 U mgprot?1, P < 0.01), superoxide dismutase (SOD) activity and GSH concentration both increased to the highest level at 24 h post treatment (99.2 ± 8.1 U mgprot?1, 93.0 ± 6.3 nmol mgprot?1, P < 0.01). After the collaborating treatment of LPS and SMT, the apoptosis index increased much higher from 48 h, while the increase of haemocytes phagocytosis, ROS level and haemolymph anti-bacteria activities were suppressed completely at 12 h. The declines of lysozyme and PO activity in haemolymph were reversed at 48 h, and the rise of SOD activity and GSH concentration started earlier from 3 h. These results indicated clearly that NO could participate in the scallop immunity and play a crucial role in the modulation of immune response including haemocytes apoptosis and phagocytosis, anti-bacterial activity and redox homeostasis in the haemolymph of scallop.  相似文献

Ecological processes including disease, competition for space, and predation strongly influence coral reef health from the colony to reef level. The leeward/west coast of the island of Hawai’i consists of the largest expanse of intact reefs in the Main Hawaiian Islands (MHI), yet little is known about the health of its coral communities. We measured prevalence of coral diseases and non-disease conditions at nine regions across two depths in the summer and winter months between 2010 and 2011. We also assessed long-term changes in coral cover (2003–2011). Mean prevalence of chronic diseases was 5–21 times greater than previously reported for the MHI. Coral health varied minimally across survey months with mild seasonality only detected in algal overgrowth (ALOG). Coral health varied considerably by depth and site, and was primarily driven by the most prevalent and common conditions: Porites growth anomalies (13.7 ± 0.82 %), Porites trematodiasis (9.5 ± 0.90 %), discoloration (5.6 ± 0.33 %), ALOG (9.9 ± 0.54 %), and gastropod predation (2.4 ± 0.23). While several conditions were significantly elevated in shallow zones, unique site × depth interactions suggest that specific site-level factors are driving prevalence. At the coast-wide level, percentage of coral cover did not change significantly between 2003 and 2011, but decreased significantly at two sites and increased at one site. Based on coral cover decline and high prevalence of certain coral health conditions, we identified four regions of concern (Puakō, Mauna Lani, Ka’ūpūlehu, and Hōnaunau). The high spatial variation in coral health not only advances our understanding of coral disease ecology, but also supports reef resilience planning by identifying vulnerable areas that would benefit most from targeted conservation and management efforts.  相似文献

Effects of short-term sedimentation on common coastal coral species were investigated in laboratory and field experiments on the Great Barrier Reef (GBR) using pulse-amplitude modulated (PAM) chlorophyll fluorometry. In the laboratory, changes in maximal quantum yields of photosystem II (Fv/Fm) in Montipora peltiformis were examined in response to the amount of sedimentation (79-234 mg cm−2) and duration of exposure (0-36 h). In control colonies, Fv/Fm ranged from 0.67 to 0.71, and did not show any temporal trend, while maximum yields of sediment-covered fragments declined steadily and reached levels below 0.1 in most colonies after 36 h coverage. Maximal quantum yield in M. peltiformis declined linearly in relation to both the amount of sediment deposited per unit surface area and the duration of exposure. Zooxanthellae densities and chlorophyll concentrations per unit area of sediment-treated corals decreased in the same manner, however, their responses were not quite as strong as the changes in Fv/Fm. Within the ranges measured, sedimentation stress of colonies exposed to large amounts of sediment for short periods of time was similar to that exposed to low amounts of sediments for prolonged periods of time. Colonies were recovered from short-term, or low-level, sedimentation within <36 h, whereas long-term exposure, or high levels of sedimentation, killed exposed colony parts. Field experiments comparing susceptibilities of common coastal coral species towards sedimentation showed significant reductions in effective quantum yields (ΔF/Fm′) in 9 out of 12 common coastal species after 22 h of exposure. Three out of twelve investigated species were not affected by the experimental application of sediments (Galaxea fascicularis, Fungia crassa, and Pectinia lactuca). Our results suggest that anthropogenic sediment deposition can negatively affect the photosynthetic activity of zooxanthellae and thus the viability of corals. However, the results also showed the ability of corals to compartmentalise sedimentation stress, as the photosynthetic activity only from tissues directly underneath the sediment declined, whereas that of adjacent clean tissues did not change measurably.  相似文献

In nature, several abiotic stresses occur simultaneously, leading to retarded growth and biochemical changes in microalgae, including the commercial cyanobacterium, Arthrospira platensis. To gain more understanding of stress response, we investigated the integrative effects of nitrogen depletion and high temperature stress on physiological changes of A. platensis C1. The results revealed that photosynthetic activities of the stressed cells were markedly reduced by more than a half in comparison to the non-stressed cells. Moreover, a reduction of biomass was observed within 24 h after prolonged exposure to combined stress of nitrogen depletion and high temperature. The total protein contents, including phycocyanin (PC), in the stressed cells, decreased rapidly within 8 h of incubation. This finding was concomitant with the increase in carbohydrate content. However, the accumulation of carbohydrates in the nitrogen depletion-treated cells was greater than that in the cells under the combined stress. Furthermore, the levels of polysaccharides increased only under long-term incubation under nitrogen depletion but not under the combined stress. In addition, the combination of nitrogen depletion and high temperature stress resulted in an increase in the proportion of linoleic acid but a decrease in γ-linolenic acid within 24 h. These results suggest that the response of A. platensis to the combined stress was different from the responses of cells to individual stress. The PC degradation, the increased carbohydrates, and the alteration in fatty acids profiles were required for physiological response to combined nitrogen depletion and high temperature stress of A. platensis C1.  相似文献

Increasing evidence of links between climate change, anthropogenic stress and coral disease underscores the importance of understanding the mechanisms by which reef-building corals resist infection and recover from injury. Cellular inflammation and melanin-producing signalling pathway are two mechanisms employed by invertebrates to remove foreign organisms such as pathogens, but they have not been recorded previously in scleractinian corals. This study demonstrates the presence of the phenoloxidase (PO) activating melanin pathway in two species of coral, Acropora millepora and a massive species of Porites, which both develop local pigmentation in response to interactions with a variety of organisms. L-DOPA (3-(3,4-dihydroxyphenyl)-L-alanine) substrate-based enzyme activation assays demonstrated PO activity in healthy tissues of both species and upregulation in pigmented tissues of A. millepora. Histological staining conclusively identified the presence of melanin in Porites tissues. These results demonstrate that the PO pathway is active in both coral species. Moreover, the upregulation of PO activity in areas of non-normal pigmentation in A. millepora and increased melanin production in pigmented Porites tissues suggest the presence of a generalized defence response to localized stress. Interspecific differences in the usage of pathways involved in innate immunity may underlie the comparative success of massive Porites sp. as long-lived stress tolerators.  相似文献
