The pattern of cyst distribution in the absence of turbation and their hatching behaviour were studied in an outdoor artificial pool, where just differentiated adults of the anostracan Chirocephalus ruffoi (sex ratio 1:3) lived until it dried up. The horizontal and vertical distributions of cysts in the pool bed were determined. The comparison between cyst bank estimate (Mura , 2004) and the actual number of cysts counted in the pool bed revealed an estimate error of 20.9%. Resting eggs occurred only in the upper 2.5 cm thick soil sections and decreased within this section as depth increased. Peripheral areas of the pool contained significantly larger numbers of cysts than the central area. Multiway analysis on the results recorded in hatching success (nested ANOVA) revealed that the differences were significantly affected by initial soil conditions, treatment and vertical distribution of cysts. Among these factors, vertical distribution (sections nested in cores) was the most influential. Hatching success was significantly inversely related to depth. Differences in the timing of hatching depending on the above considered factors were also noted. A nearly synchronous hatching pattern was observed only for cysts from initially dry sediment of the uppermost layers. In all successively deeper layers, hatching showed multiple peaks and was increasingly delayed and erratic (already mentioned). ANCOVA within each of the experimental conditions revealed significant differences in hatching frequencies (time as covariate) depending on sediment depth. 1. Life‐table experiments with Brachionus calyciflorus test several hypotheses related to the idea that sexual reproduction in monogonont rotifers should occur when food resources are favourable. 2. The food concentration necessary for a fertilised mictic female to produce one phenotypically normal resting egg was higher than that for an amictic female to produce one daughter. At the lowest concentration of Cryptomonas erosa (1.25 × 103 cells mL?1), the lifetime fecundity of these two types of females was 0.9 and 1.4, respectively. 3. The lifetime fecundity of both fertilised mictic females and amictic females increased with food concentration to 3.4 resting eggs and 15.2 daughters female?1, respectively. The approach to maximal fecundity with increasing food concentration was more rapid for fertilised mictic females, such that their lifetime fecundity relative to that of amictic females gradually decreased from 0.64 (at 1.25 × 103 C. erosa mL?1) to 0.22 (at 2.5 × 104 C. erosa mL?1). 4. The probability of a fertilised mictic female producing one or more abnormal resting eggs during her lifetime was high (approximately 75%). The mean proportion of abnormal eggs produced per female varied among the different food‐concentration treatments (26–38%) but was not higher at the low food concentrations. 5. The proportion of normal resting eggs that hatched was high (51–71%); those produced at low food concentrations were no less likely to hatch than those produced at high food concentrations. No abnormal resting eggs hatched. 6. The probability of a fertilised mictic female producing an abnormal resting egg increased rapidly with her age at all food concentrations. The probability of a normal resting egg hatching declined with maternal age at the low food concentration in one of two experiments. 7. The results support the idea that induction of mictic females should occur when food resources are good. Coincidence of sexual reproduction with low food availability risks low production of resting eggs for several reasons. Population size may be small, with a low probability of encounters between young mictic females and males, and fertilised mictic females may be unable to mature and produce resting eggs.  相似文献

Since resting cysts are a potential seeding source for blooms, the presence of these cysts in sediments is a marker of an established population for a number of harmful algal species. The spatial patterns of cyst density in relation to sediment characteristics and hydrodynamics are still largely misunderstood. This study investigated the spatial distribution of resting cysts belonging to the Alexandrium tamarense species complex (Dinophyceae) in sediments of a Mediterranean coastal lagoon (Thau Lagoon, France). This lagoon, hosting shellfish farming, is regularly impacted by toxic Alexandrium catenella blooms. The average cyst density across the whole lagoon was rather low, <20 cysts g−1 of dry sediment (DS). However, densities varied widely among sampled stations, with the highest density (∼440 cysts g−1 DS) recorded in a shallow cove named Crique-de-l’Angle, which is the only area where dense blooms of A. catenella and A. tamarense have been recorded in the years preceding this survey. An analysis using spatial autoregressive models demonstrated that cyst densities were highly spatially autocorrelated (indicating that close stations tended to have more similar cyst densities) with accumulation sites. With respect to sediment characteristics (5 granulometric fractions <2 mm and biochemical components), the highest densities were found in silty sediments containing high proportions of water and organic matter. Nevertheless, the linkage between cyst density and sediment structure was not always verified; this reflected the influence of hydrodynamics on the sedimentation of cysts and sediment particles, and on the dispersal of cysts away from the bloom area by wind-induced currents, suggesting that hydrodynamics was responsible for the spatially autocorrelated distribution of cyst densities.  相似文献

To examine how dormancy contributes to the establishment and persistence of Bythotrephes longimanus, we investigated resting egg production and hatching in relation to the demography of the planktonic stage and environmental conditions in Island Lake Reservoir (USA). During a 3-year study, the largest contribution to the egg bank occurred in autumn and most eggs hatched in spring, but we also detected some resting egg production and hatching in summer. The difference between summer and late autumn densities of eggs in sediments averaged 47% (range 0–98%) for 18 sites throughout the reservoir, which was similar to experimental estimates of in situ hatching fraction of 67% for eggs in the spring and summer following their production. Based on emergence traps, neonates hatch in the field during May and June. We estimated mortality rates of 64% for resting eggs and embryos, and 59% for newly emerged neonates. Although hatching fraction saturated at the same level, eggs incubated offshore hatched later than those nearshore where water temperature was warmer and light was detectable at the sediment surface. Low dissolved oxygen concentration did not significantly reduce hatching fraction but resulted in some eggs that initiated development but failed to hatch. Collectively, our results demonstrate substantial annual turnover in the resting egg bank of B. longimanus and high mortality of resting eggs during recruitment from the egg to the first molt of the planktonic stage. These patterns suggest that propagule pressure in the form of resting eggs requires large numbers for establishment, and that considerable post-establishment resting egg production is necessary for inter-annual persistence.  相似文献

The hatching of diapausing eggs is a means of temporal dispersal that can provide populations with genotypes adapted to different environments. In a salinity-variable shallow lake, we predicted that the mixing of different age-classes of eggs in the sediment may yield genotypes with different salinity optima. The alternative would be the absence of local adaptation and the presence of a homogenous population of salt-tolerant genotypes with high phenotypic plasticity. We tested these alternatives by isolating Daphnia magna resting eggs from different sediment depths, exposing them to hatching cues at different salinity levels and measuring the performance of hatched individuals. Results revealed a homogeneous sediment with generally broad-tolerance genotypes and some genotypes with low salt tolerance, which supports the second hypothesis. However, the disturbed character of the sediment hampered historical reconstruction. The absence of local adaptation in the diapausing egg bank may be the result of various scenarios in the response of D. magna populations to severe salinity changes in the lake.  相似文献

Recent research has indicated that the hatchability of mass-producedrotifer (Brachionus plicatilis) resting eggs (stored in sea water in thedark at 4°C) gradually decreases due to bacterialinfection of the porous egg surface. We investigated whether lyophilizationas well as a canning treatment could solve the problem of bacterialinfection of resting eggs. The canning method preserves eggs in a dry form,after lyophilization at –40°C. The resting eggs werepacked in cans under various pressures: 21, 34, 48, 61, 75, 88 and 101 kPa.The viability of the canned resting eggs was tested by hatching experimentsdone 6 and 12 months after canning and storage at 5°C.When partial pressures were less than 88 kPa, percent hatching after 6months ranged between 16 and 20%, similar to that of uncanned eggs(19% hatching), but significantly lower (9 and 11%, P <0.01) at 88 and 101 kpa. Even after 12 months, percent hatching remained at35% when canning was at 61 kpa. Lyophilization and canning at partialpressure lower than 88 kPa could therefore preserve resting eggs, andhatchability was maintained. Canned eggs that were treated with NaClO (1 mgl–1) or NFS-Na (5 mg l–1) showed a significantlyhigher percent hatching (68% and 80%, respectively, each P<0.05) than untreated eggs.  相似文献

The distribution and abundance of viable resting eggs of copepod Acartia pacifica in Xiamen Bay, China, were determined in the laboratory by the presence of nauplii hatched from the sediments. Sediment cores to a depth of 30 cm, sliced at 1.0 cm intervals, showed that most viable resting eggs of A. pacifica occurred near the sediment surface (0-5 cm), and the number of viable eggs sharply decreased with depth of the sediment, although resting eggs remained viable as deep as 23 cm. 210Pb analyses of the sediments indicated that the maximum age of viable eggs of A. pacifica was 20.5 years and the mean egg age was 4.3 years. The egg mortality of A. pacifica in the sediment was 0.1408 year−1, or 85.92% annual egg survival, calculated by regressing ln(egg density) on the age of the sediment. The horizontal distribution of viable resting eggs ranged from 2.27×103 to 3.85×105 m−2, with a mean value of 9.49×104 m−2. Regressions between viable eggs of A. pacifica and all fine-fraction particle size classes (at 2 μm intervals) were not significant. The accumulation of viable resting eggs that can persist for an extended period of time provided evidence for the existence of an egg bank of A. pacifica in the seabed of Xiamen Bay.  相似文献

The anostracan Phallocryptus spinosa has an almost exclusively palearctic distribution. The Makgadikgadi Pans area in Botswana represents the only distribution record south of the Sahara. In this ephemeral wetland, it is an important food item in the diet of migrating birds. By studying egg bank characteristics (such as depth and density) and hatching requirements, we investigated the persistence of this sub-Saharan population. At localities in the middle and north basin of Sua Pan, sediment cores were taken along a transect, and dormant eggs were isolated. Densities of the active dormant egg bank ranged from 833 to 31449 dormant eggs m−2, indicating that this species is well established. Most of the dormant eggs were found in the top 4 cm of the sediments, and densities decreased to zero at a depth of 13 cm. Considering the expected low sedimentation rate, the presence of dormant eggs down to 13 cm indicates long-time occurrence of P. spinosa in the Makgadikgadi Pans area. We observed high hatching fractions (up to 85%) at a temperature of 22 °C and a salinity of 5 ppt. A second anostracan species, Branchinella ornata, co-occurred with P. spinosa in our study site. This population also had a large active dormant egg bank (ranging from 6634 to 50557 dormant eggs m−2) with dormant eggs present until a depth of about 11 cm. The existence of egg banks not only ensures the survival of zooplankton through harsh periods, but could also affect microevolutionary dynamics. Whether zooplankton at high latitudes can build up an egg bank in sediments, as occurs at lower latitudes, is still unknown. The distribution and age of viable resting eggs of the calanoid copepod Boeckella poppei Mrázek in sediments of two small freshwater lakes on King George Island were determined by slicing sediment cores at 1-cm intervals. Most viable resting eggs were found near the sediment surface, with abundance sharply declining to very low values at the depth of 5?cm, although eggs were present as deep as 9?cm in the sediments of Yanouhu Lake. The egg abundances in Xihu Lake and Yanouhu Lake were estimated to be 9.2?×?104 and 7.2?×?104?eggs?m?2, respectively. 210Pb dating indicated a relatively constant sedimentation rate (0.023?cm?year?1) in Xihu Lake, which was used to estimate the mean age (46.8?year), the maximum age (195.7?year), and the mortality rate (1.64?% year?1) of resting eggs of B. poppei in Xihu Lake. The accumulation of resting eggs with long-term viability in sediments provided the first evidence for the existence of egg banks in two Antarctic maritime lakes. An egg bank may serve as an overwintering strategy for B. poppei in Antarctica, enhance their ability to cope with random extreme changes, and contribute to their broad distribution.  相似文献

The study proposes methodological developments to optimize sampling strategy of resting cysts of Alexandrium catenella to estimate their abundance with a predefined error. This work also aims to provide information on spatial distribution of resting cysts in sediments. The distribution mode of A. catenella resting cysts related to the abundance variability was studied through sediment cores sampling on four different spatial scales and using Ludox CLX gradient density method. The quantification method underestimates by a factor of 2 the resting cysts abundance in one gram of sediment. Application of Taylor's power law allowed us to define a compromise between sampling effort and abundance estimation error. In the case of A. catenella resting cysts from Thau lagoon, the optimal sampling strategy consists of sampling 10 stations on a surface of 2 km2 for a given coefficient of variability (C) of 15%, sampling 3 sediment cores at each station (C = 30%) and counting only one replicate by core (C = 18%). Results related to the application of Taylor's power law are closely dependent on resting cyst density and aggregation in a given sediment. In our area, A. catenella resting cysts are mainly observed in the upper 3 cm of sediment. Horizontally, their heterogeneity is lower on 10 cm2 surface and tends to stabilize itself beyond a surface of 10 m2. Each author has to carry out this pre-sampling effort for his own resting cysts-forming species, in his own area, in order to increase accuracy of resting cyst mapping.  相似文献

The purpose of the present work was to study the seasonal variations in egg production, morphology and hatching success in the cryptic species Acartia tonsa, taking into account variations in female size, population abundance and environmental factors in a turbid and hypersaline estuary. Sampling was performed during the austral warm (18–23°C and 32–36 salinity) and cold seasons (5–7°C; 32–38) in Bahía Blanca Estuary (BBE), Argentina, during 2007 and 2009. Field-collected females were incubated in the laboratory simulating in situ environmental conditions, and specimens from fixed samples were measured using optical and scanning electronic microscopy. Acartia tonsa’s marked seasonality in its reproductive traits was found to ensure its permanence in the water column all over the year. During the warm season, small-sized females were observed to invest their energy in the production of subitaneous eggs with high hatching success and smooth appearance (12.95?±?2.38 eggs f?1 day?1 and specific egg production rate (SEP) of 16.57%C f?1 day?1). During the cold season, females invested C in body mass as well as in the production of resting eggs of three different morphotypes (6.56?±?3.2 eggs f?1 day?1 and SEP of 7.37%C f?1 day?1). Although these morphotypes were found to show differences in surface ornamentation, they exhibited the same delayed hatching behaviour. The eggs with shorter spines were found to integrate the resting egg bank in BBE. Our findings confirming a delayed egg hatching behaviour and a great tolerance to low temperatures and high salinities in the A. tonsa population in BBE suggest that this possible strain is a valuable phenotype for aquaculture.  相似文献

Abundance and distribution of the toxic dinoflagellate Alexandrium tamarense species complex resting cyst were investigated in the eastern Bering Sea and the Chukchi Sea for the first time. Sediment samples (top 0–3 cm depth) were collected from the continental shelf of the eastern Bering Sea (17 stations) and the Chukchi Sea (13 stations) together with a long core sample (top 0–21 cm depth) from one station in the Chukchi Sea during 2009–2012. The cysts were enumerated using the primuline staining method. Species identification of the cysts was carried out with multiplex PCR assay and the plate morphology of vegetative cells germinated from cysts in the both areas. Alexandrium cysts were widely detected in the both areas, ranging from not detected (<1 cysts cm−3) to 835 cysts cm−3 wet sediment in the eastern Bering Sea and from not detected (<1 cysts cm−3) to 10,600 cysts cm−3 in the Chukchi Sea, and all isolated cysts were genetically and morphologically identified as the North American clade A. tamarense. Their cysts were mainly distributed in the shallow continental shelf where the water depth was less than 100 m in both areas. The cysts were detected from the deep layer (18–21 cm depth of sediment core) of the long core sample. The present study confirmed the abundant existence of A. tamarense with wide range of distribution in these areas. This fact suggests that A. tamarense vegetative cells have appeared in the water column in the both areas. Furthermore, these abundant cyst depositions indicate that this species originally distributed in the Arctic and subarctic regions and well adapted to the environments in the marginal ice zone.  相似文献

Zooplankton resting egg banks accumulate resting stages of various zooplankton species that are active in different habitats and different periods of the year. As such, hatching of resting eggs from lake sediments may potentially be very useful in zooplankton diversity studies. In this study, we tested whether the efficiency of the cost-effective technique is increased by isolating the resting eggs from the sediment prior to incubation. Isolation of the eggs was advantageous for the overall hatching success (+26% after 36 days of incubation compared to incubation of sediment). Furthermore, isolation of resting eggs makes egg bank diversity analyses less time consuming in two ways. (1) It reduced the time needed for the eggs to hatch with on average 35%. In the isolation treatment all responsive resting eggs hatched within the first 4 weeks of incubation, while in the non-isolation treatment neither the cumulative number of macrozooplankton hatchlings nor the cumulative number of hatched cladoceran species levelled off after 36 days of incubation. (2) In contrast to the non-isolation treatment, where large differences occurred between taxa in incubation time, isolation reduced such inter-specific differences, so that even very short incubation periods kept bias within acceptable limits.  相似文献

Nine sediment cores of 8–26 cm in length were collected from two basins of Daya Bay, the South China Sea, by Tokyo University Fisheries Oceanography Laboratory core sampler in August 2001 to investigate the distribution of dinoflagellate resting cysts. In the present study, 51 different cyst morphotypes representing 22 genera were identified from 65 sediment samples. Among them, there were 21 autotrophic species and 30 heterotrophic ones. Cyst species richness in each sample varied from 12 to 29, while the values of Shannon‐Weaver diversity index (H′) were between 0.15 and 4.13. There were an obvious increase in both species richness and values of H′in 2–6 cm sediments. Cyst concentrations varied from 154 to 113 483 cysts per gram dry weight sediment, and were much higher in upper sediments. Scrippsiella trochoidea was the most dominant cyst type, which took up over 90% of cyst assemblages in the upper sediments. The abrupt increase of S. trochoidea cysts in the surface sediments reflected the bloom of this species in Daya Bay in 2000. The results from cyst assemblages showed some trend of changes in water quality in this area, and indicated a typical type of pollution caused by cultural eutrophication, which started in the 1980s and greatly accelerated in the middle of 1990s. Cysts of Alexandrium, mainly those of Alexandrium catenella and Alexandrium tamarense complex, occurred frequently and abundantly in this area, with the highest concentration and relative frequency of 503 cysts per gram dry weight sediment and 22.3%, respectively. The high abundance of Alexandrium cysts provided rich ‘seed bed’ for Alexandrium blooms and was also an important source of paralytic shellfish poisoning toxins, especially in winter.  相似文献

Hatching studies with Heterodera glycines typically have been conducted with a mixture of egg-mass and encysted eggs. Laboratory research was conducted to compare hatching of H. glycines eggs from external egg masses with that of eggs extracted from within females and cysts (encysted eggs). Egg-mass eggs were collected by soaking infected soybean roots in 0.5% sodium hypochlorite, and encysted eggs were collected from females and cysts dislodged from the same roots with a stream of water. Eggs were incubated at 25 °C in deionized water, 3.0 mM ZnSO₄solution, or one of three synthetic H. glycines hatch inhibitors, mad hatched juveniles were counted every other day for 22 days. Samples of eggs collected at the beginning and end of all experiments were analyzed to determine extent of embryo development. Egg-mass eggs hatched more rapidly than encysted eggs during the first 16 days, but not thereafter. Throughout the experiments, hatch of egg-mass eggs in deionized water was greater than that of encysted eggs. From day 8 to day 22, egg-mass eggs were less sensitive than encysted eggs to the hatch inhibitor 2-(2''-carboxyethyl)-5-[carboxy(hydroxy)methylidenyl]cyclopentanone. A greater proportion of egg-mass eggs contained vermiform juveniles than did encysted eggs at the beginning of the experiments, but not at the end. Results indicated that H. glycines egg-mass and encysted eggs have different hatching behaviors that cannot be explained entirely by differences in embryological development.  相似文献

When faced with disturbances such as increased salinity, aquatic communities inhabiting inland coastal systems change and may or may not be resilient after salinity decreases. The purpose of this study was to assess the potential role of the resting egg bank for zooplankton community resilience. We predicted that (1) hatching of resting eggs is inhibited by increased salinities and (2) resting eggs remain viable when exposed to salinity and hatch when returned to freshwater. At the community level, we evaluated the hatching responses and the short-term viability of resting eggs exposed to a salinity gradient. The hatching of resting eggs was inhibited at higher salinities (16.0 and 32.0 g l?1). However, some resting eggs remained viable and hatched when returned to freshwater. Additionally, combining our experimental results to previously published field data, we observed that the pattern of hatching during exposure to salinity matches the temporal succession observed in the zooplankton community at our model system, after increased salinity. The recovery of zooplankton communities after disturbances involving increased salinity is likely facilitated by the presence of an egg bank. This finding has important implications for the recovery of zooplankton communities and the management of aquatic systems vulnerable to salinization worldwide.  相似文献

Lake sturgeon spawning on artificial habitat in the St Lawrence River   总被引:2,自引:0,他引:2  
In 1996, lake sturgeon (Acipenser fulvescens) spawning was documented for the third consecutive year on an artificially placed gravel bed in the St Lawrence River. Two distinct spawning periods were observed in 1996. Spawning initially commenced on 17 June, when water temperature reached 15°C. A second spawning event was documented from 28 June to 1 July (16°C). Sturgeon egg densities were monitored in three transects on egg trays, on the gravel surface, and within interstitial spaces in the gravel. Counts of developing eggs in the gravel bed during both spawning periods were used to estimate a total of 275 000 eggs on the study area (0.075 ha). Average egg density was highest in the transect with the highest water velocities. Lake sturgeon fry were first observed in the gravel on 24 June (15.5°C), and first emergence from the gravel was documented on 28 June. Hatching following the second spawning event commenced on 3 July. Based on assessment of average embryo viability (61.6%) and egg‐to‐emergent fry survival (17.6%) an estimate of about 171 000 sturgeon eggs hatched, producing over 49 000 emergent fry. Current velocity, substrate particle size, depth of substrate, and maintenance of sediment‐free interstitial spaces are important considerations in planning future spawning habitat enhancement projects.  相似文献

We investigated the effects of Heterodera glycines cyst components and surface disinfestants on hatching of H. glycines eggs in vitro. Eggs were incubated in either H. glycines cyst wall fragments, cyst wall and egg rinsate, egg homogenate, or control solutions of soybean root diffusate, sterile distilled water, or zinc sulfate. Hatch in cyst wall and egg rinsate, and egg homogenate, was greater (α = 0.05) than hatch in sterile distilled water; however, it was not different from hatch in zinc sulfate according to Dunnett''s test. Hatch in cyst wall fragments was similar to hatch in sterile distilled water. To determine whether surface disinfestants affected hatch, eggs were treated first with chlorhexidine diacetate, mercuric chloride, sodium hypochlorite, or streptomycin sulfate and then incubated in H. glycines egg homogenate, soybean root diffusate, sterile distilled water, or zinc sulfate. Hatch of eggs treated with chlorhexidine diacetate, mercuric chloride, and streptomycin sulfate was reduced (α = 0.05), and hatch of eggs treated with sodium hypochlorite was increased (α = 0.05) relative to hatch of nontreated eggs in all incubation solutions except zinc sulfate according to Dunnett''s Test. Hatch in zinc sulfate was similar among all surface disinfestants except mercuric chloride, where hatch was reduced relative to hatch of nontreated and other surface disinfestant-treated eggs.  相似文献

Here we present a quantitative study on the density, age and viability of the diapausing egg banks of the rotifer species complex Brachionus plicatilis in the sediments of 15 water bodies from Eastern Spain. Sampled ponds, located in coastal and inland areas, varied in salinity and ranged in size, depth and permanence. By identifying ‘hatched’, ‘deteriorated’ and ‘viable’ diapausing eggs in the sediment samples, we estimated production, hatching and deterioration in relation to the habitat properties of each pond. Our results indicate the presence of large numbers of diapausing eggs in the sediments of almost all of the ponds studied (2-115 eggs cm− 2). Inland ponds tended to have higher densities than coastal lagoons. The vertical distribution of eggs in the sediments frequently showed a non-decreasing pattern, which suggested a high among-year variation in egg production. Despite maximum age of eggs of 60-80 years, the median age (3-30 years) suggests that rotifer egg banks are young in the studied ponds. Egg senescence is suggested by the declining abundance of ‘healthy-looking’ eggs with depth. The proportion of ‘deteriorated’ eggs ranged 75-99% suggesting that deterioration rates in the sediments are high and vary among habitats. Hatching and deterioration rates, as estimated from the counts of ‘hatched’, ‘deteriorated’ and ‘healthy-looking’ eggs in the sediments, largely varied among ponds. An association between hatching and deterioration rates is suggested by our data. This is in agreement with the hypothesis that hatching rates of diapausing eggs depended not only on the risks associated with the water column, as initially expected by the general theory on diapause, but they are also related to the incidence of deterioration processes in the sediment.  相似文献

We studied the egg banks of Daphnia gr. longispina in four acidified lakes in the Bohemian Forest (Czech Republic, Europe). Daphnia had become extinct in three of the studied lakes due to anthropogenic acidification and we assessed the possibility of autochthonous recovery of the population after recent amelioration of the lake environment. We determined the ephippia distribution in eleven cores 10–30 cm long, and the state of Daphnia resting eggs in over 13 000 ephippia from eight cores. Apparently well-preserved eggs were used for hatching experiments and for DNA amplification. Vertical profiles of ephippia densities in several cores did not agree with historical data on the Daphnia presence in the lakes, as the sediment had been repeatedly disturbed by human activities in the past. Ephippia are present near the surface of the sediment in all lakes, and they might therefore receive hatching cues. We were not able, however, to prove that viable eggs are present in the ephippia banks of the lakes. The percentage of the well-preserved eggs in all but one core was below 1%. In addition, no egg hatched in the hatching experiments and we were not able to amplify DNA from the preserved eggs of lakes where Daphnia is extinct, although amplification from relatively young eggs from the fourth lake was successful in 60% of the cases. We conclude that the recovery of Daphnia populations in studied lakes from autochthonous sources is unlikely.  相似文献
