The non-defective (heavy) virions from a simian virus 40-like virus (DAR virus) isolated from human brain have been serially passaged at high input multi-plicities in primary monkey kidney cells. The 32P-labeled, progeny DAR-viral genomes have been purified and tested for sensitivity to the RI restriction endouclease from Escherichia coli (Eco RI3 restriction nuclease). The parental DAR-viral genomes share many physical properties with “standard” simian virus 40 DNA and are cleaved once by the Eco RI restriction nuclease. After the fourth serial passage, three populations of genomes could be distinguished: Eco RI resistant, Eco RI sensitive (one cleavage site) and Eco RI “supersensitive” (three, symmetrically-located, cleavage sites). The Eco RI cleavage product of the “supersensitive” form is one-third the physical size (10.4 S) of simian virus 40 DNA and reassociates about three times more rapidly than sheared, denatured simian virus 40 DNA. From the fourth to the eighth serial passages, the genomes containing this specific triplication of viral DNA sequences were selected for and became the predominant viral DNA species.  相似文献

Serial passage of the non-defective form of a simian virus 40-like virus (DAR) isolated from human brain results in the appearance of three distinct classes of supercoiled DNAs: RI resistant, RI sensitive (one cleavage site) and RI “supersensitive” (three cleavage sites). The RI cleavage product of the “super sensitive” form is one-third the physical size of simian virus 40 DNA (10.4 S) and reassociates about three times more rapidly than “standard” viral DNA. To identify the portions of the DAR genome present in the 10.4 S segment, the plus strand of each of the 11 fragments of 32P-labeled simian virus 40 DNA, produced by cleavage with the Hemophilus influenzae restriction endonuclease, was hybridized in solution with the sheared RI cleavage product of the “supersensitive” class of viral DNA. Reaction was observed with fragments located in two distinct regions of the simian virus 40 genome: (1) Hin-A and C; (2) Hin-G, J, F and K.Further studies indicated that simian virus 40 complementary RNA transcribed in vitro with Escherichia coli RNA polymerase from one strand of simian virus 40 DNA reacts with both strands of the denatured 10.4 S cleavage product when hybridization is monitored with hydroxyapatite. Treatment of the 10.4 S DNA-simian virus 40 cRNA hybrid with the single-strand spcific nuclease, S1, converted approximately 50% of the radioactive counts to an acid-soluble product. These results indicate that the 10.4 S product contains a transposition of sequences originally present on one of the DAR DNA strands to the other strand. Examination of heteroduplexes formed between the 10.4 S segment and unique linear forms of DAR DNA produced with the R · Eco RI restriction endonuclease have confirmed these observations. Thus it appears that a molecular rearrangement(s) has resulted in the recombination and inversion of viral DNA sequences from two separate loci on the parental DAR genome. This 1.1 × 106 dalton segment is reiterated three times in a supercoiled molecule equivalent in physical size to parental DAR DNA.  相似文献

Tissue culture cells of virus-transformed and untransformed cell lines had low contents of cytochromes in the respiratory chain, measured per cell or per mg protein of the cells, in comparison to the cytochrome contents of liver cells in vivo.

In the virus-transformed cells the contents of cytochromes aa3, b and possibly c1 were significantly lower than those in the untransformed cells, while the content of cytochrome c was found to be the same or even increased in the transformed cells. Thus, a markedly high ratio of cytochromes c/aa3 was observed in the transformed cells.

Polarographic measurements of the oxygen uptake have shown a generally low rate of both endogenous respiration and respiration in the presence of glucose and vitamin K3 in the transformed cells.

The present study indicates that there is a quantitative and possibly qualitative alteration of the respiratory chain components in the transformed cells.  相似文献

Summary Baculovirus-derived recombinant simian virus 40 (SV40) large tumor antigen (SV40 T-Ag), a monoclonal antibody specific for SV40 T-Ag (Ab-1 preparation), and a monoclonal anti-idiotypic antibody (anti-Id), designated 58D, were used to analyze the humoral immune response of Balb/c mice either immunized with recombinant SV40 T-Ag or challenged with SV40-transformed cells. Inhibition assays indicated that antibodies from mice immunized with SV40 T-Ag and from those bearing SV40 tumor inhibited the SV40 T-Ag/Ab-1 reaction. These data suggested that the antibody response in immunized or tumorchallenged mice recognized similar epitope(s) on SV40 T-Ag to that detected by the monoclonal Ab-1. These anti-(SV40 T-Ag) response antibodies also inhibited the Ab-1/anti-Id reaction and recognized the anti-Id in direct binding assays. Together, these data indicate that murine anti-(SV40 T-Ag) responses shared an idiotope with a monoclonal anti-(SV40 T-Ag) Ab-1 preparation. This idiotope, which is recognized by the monoclonal anti-Id preparation, 58D, appears to be involved in the humoral immune response to SV40 T-Ag in both SV40-T-Ag-immunized and tumor-bearing mice. The monoclonal anti-Id preparation may represent a focal point for manipulating the humoral immune response to tumors induced by SV40-transformed cells.  相似文献

Simian virus 40 (SV40) is a tumor virus and its early gene product large T-antigen (LT) is responsible for the transforming activity of SV40. Parkinson’s disease causative gene DJ-1 is also a ras-dependent oncogene, but the mechanism of its oncogene function is still not known. In this study, we found that there were no transformed foci when fibroblasts from DJ-1-knockout mice were transfected with LT. We also found that DJ-1 directly bound to LT and that the expression level of c-Myc in transformed cells was parallel to that of DJ-1. These findings indicate that DJ-1 is essential for SV40 transformation.

A physical map of the Simian virus 40 genome has been constructed on the basis of specific cleavage of Simian virus 40 DNA by bacterial restriction endonucleases. The 11 fragments produced by enzyme from Hemophilus influenzae have been ordered by analysis of partial digest products and by analysis of an overlapping set of fragments produced by enzyme from Hemophilus parainfluenzae. In addition, the single site in SV40 DNA cleaved by the Escherichia coli RI restriction endonuclease has been located. With this site as a reference point, the H. influenzae cleavage sites and the H. parainfluenzae cleavage sites have been localized on the map.  相似文献

Methods for predicting peptide chain conformation have been applied to amino acid sequences adjacant to the carbohydrate attachment sites of glycoproteins containing the N-glycosylamine type of protein-carbohydrate linkage. Of 31 glycosylated residues examined 30 occur in sequences favouring turn or loop structures. Twentytwo of the glycosylated asparagine residues occur in tetrapeptides predicted to have the β-turn conformation. Carbohydrate attachment is therefore associated with peptide sequences which favour the formation of β-turn or other turn or loop structures.  相似文献

目的 拯救携带OX40L的重组溶瘤流感病毒株,全面鉴定并评价其靶向杀伤肝癌细胞的效果。方法 将编码OX40L的基因片段嵌合在A/PuertoRico/8/34(PR8)的NS片段特定位置,利用反向遗传学技术(reverse genetics,RG),与PR8流感病毒的剩余7个骨架质粒pHW191-PB2、pHW192-PB1、pHW193-PA、pHW194-HA、pHW195-NP、pHW196-NA、pHW197-M共转染COSⅠ/MDCK细胞,成功拯救获得重组溶瘤流感病毒,命名为rFlu-OX40L。经血凝、TCID50方法测定病毒滴度;重组病毒纯化后电镜观察病毒形态特征及大小分布;检测MDCK细胞病毒生长曲线;MTS法检测重组病毒对肝癌细胞活力的影响;流式细胞术检测重组病毒诱导肝癌细胞死亡方式;利用肝癌荷瘤小鼠模型评价重组病毒rFlu-OX40L在动物体内的抗肿瘤效果。结果 重组溶瘤流感病毒rFlu-OX40L可在鸡胚中稳定传代,HA效价达27~8,病毒滴度可达 7~8 LgTCID50/ml;重组病毒rFlu-OX40L与流感病毒PR8生长曲线相一致,72 h达高峰;MTS结果显示,3 MOI重组病毒可显著降低肝癌细胞活力,且对正常肝细胞无明显影响,呈时间和剂量依赖性;流式细胞结果显示,重组病毒可显著诱导肝癌细胞凋亡,呈时间和剂量依赖性;利用肝癌荷瘤小鼠模型,瘤内注射重组溶瘤流感病毒rFlu-OX40L较PR8、PBS对照组,小鼠脾细胞中CD3+、CD4+、CD8+、CD45+、CD69+ T细胞数量呈现显著升高趋势。结论 携带OX40L的重组溶瘤流感病毒rFlu-OX40L在体内外均可选择性杀伤肝癌细胞,有望为肝癌的临床治疗提供新的免疫治疗策略。  相似文献

Filoviruses are the causative agents of a severe and often fatal hemorrhagic fever with repeated outbreaks in Africa. They are negative sense single stranded enveloped viruses that can cross species barriers from its natural host bats to primates including humans. The small size of the genome poses limits to viral adaption, which may be partially overcome by conformational plasticity. Here we review the different conformational states of the Ebola virus (EBOV) matrix protein VP40 that range from monomers, to dimers, hexamers, and RNA‐bound octamers. This conformational plasticity that is required for the viral life cycle poses a unique opportunity for development of VP40 specific drugs. Furthermore, we compare the structure to homologous matrix protein structures from Paramyxoviruses and Bornaviruses and we predict that they do not only share the fold but also the conformational flexibility of EBOV VP40.  相似文献

The pattern of expression of the simian virus 40 (SV40) T antigen gene and resultant dysplasia were re-examined in a line of transgenic mice in which the T antigen gene was under the control of the SV40 early promoter. We found that T antigen expression in the kidney, and resulting dysplastic lesions, occurred exclusively in the distal convoluted tubules and the ascending limbs of Henle. Epidermal growth factor (EGF) expression in the kidney of normal mice was similarly immunolocalized. The correlation between high EGF immunoreactivity in normal mouse tissues and T antigen expression in the transgenic counterpart was also seen in the choroid plexus epithelium and in the submandibular glands of male mice. T antigen was not found in the submandibular gland of transgenic females. Similarly, EGF was only rarely detected in the normal female submandibular gland. In contrast to the correlation between T antigen expression in the transgenic mice and EGF expression in the corresponding tissues of the normal mice, within the dysplastic lesions of the transgenic mice EGF expression was severely diminished. Adenocarcinomas of the male submandibular gland from another line of transgenic mice that expresses theInt-1 transgene, showed similarly reduced levels of immunostaining for EGF. Thus, reduced expression of EGF might be a general feature of dysplasia and tumorigenesis in those tissues that normally express EGF.  相似文献

Lesions that promote reversion from a temperature-sensitive to a wild-type phenotype were induced in temperature-sensitive late mutants of SV40 virus by UV irradiation. When cultures infected with UV-irradiated temperature-sensitive mutants were grown for various times at permissive temperature (35 degrees C) and then at restrictive temperature (39 degrees C), the reversion frequency declined just before the onset of semiconservative DNA synthesis when DNA synthesis began at 32 degrees C. This can be explained by competition between reactions that lead to the onset of viral DNA synthesis and reactions that repair the lesions before the onset of viral DNA synthesis.  相似文献

Investigation of the cellular distribution of a 105 kDa phosphoprotein (pp 105) in transformed mouse fibroblasts, showed that only a minor amount was located on the surface of logarithmically grown suspension cells. More than 90% of total pp 105 was contained in the cytosolic fracion representing about 0.2% of total cytosolic proteins. Surface and cytosolic pp 105 had identical phosphopeptide patterns. Cytosolic pp 105 was highly purified by ammonium sulfate precipitation followed by three chromatographic steps and gel electrophoresis. The purified pp 105 was capable of weak autophosphorylation. In the stationary growth phase of suspension cells, the amount of pp 105 detectable by endogenous phosphorylation was only 10–15% of that observed during logarithmic growth. pp 105 was also detected in normal mouse tissue and its distribution determined.  相似文献

The nucleotide sequence of the araC regulatory gene of Salmonella typhimurium LT2 has been determined. This sequence and the predicted araC translational product are compared to their counterparts in Escherichia coli. The two genes code for similar products although the S. typhimurium protein is eleven amino acids shorter than the E. coli protein. The predicted amino acid sequences are 92% conserved and the DNA sequences are 82% conserved for the common regions of the two genes.  相似文献

The distribution of inequivalent geometries occurring during self-assembly of the major capsid protein in thermodynamic equilibrium is determined based on a master equation approach. These results are implemented to characterize the assembly of SV40 virus and to obtain information on the putative pathways controlling the progressive build-up of the SV40 capsid. The experimental testability of the predictions is assessed and an analysis of the geometries of the assembly intermediates on the dominant pathways is used to identify targets for anti-viral drug design.  相似文献

The incidence of malignant mesothelioma (MM) shows a strong epidemiological association with exposure to asbestos fibers. Recently, simian virus 40 (SV40) DNA sequences have been reported in MM tumor specimens from the United States and several European countries, and the SV40 tumor virus has been implicated as a potential co-factor in the etiology of this disease. However, several large studies from the US, Finland, and Turkey did not detect SV40 sequences in MM samples. To address this discrepancy, MM specimens from Turkey and the US were analyzed in the same laboratory under identical conditions to detect the presence of SV40 DNA. We detected SV40 sequences in 4 of 11 specimens from the United States, but in none of the 9 Turkish samples examined. These findings suggest that geographical differences exist with regard to the involvement of SV40 in human tumors.  相似文献
