A comparative immunological survey of cytotoxic properties of whole and cell-free haemolymph (plasma) was performed on four species of marine molluscs. Diverse target cells were used: auto- and heterologous mollusc haemocytes, starfish Asterias rubens coelomocytes, trematode Himasthla elongata rediae, and human erythrocytes. While all the four mollusc species were found to contain cytotoxic factors in their plasma, functional features of these molecules in bivalve and gastropod species are essentially different. Cell-free haemolymph of two clams, Mya arenaria and Mytilus edulis, contains pre-synthetized cytolytic complexes capable of immediate and generalized activation, leading to destruction of a broad range of target cells. On the contrary, two gastropod species, Buccinum undatum and Littorina littorea, possess haemolytic factors which display high specificity of recognition of receptor molecules on the target cells and slow cytolytic reaction. Overall, molecular strategies of cytolytic cascade activation in Bivalvia and Gastropoda can be described as low- and highly selective, respectively.  相似文献

The Pacific oyster Crassostrea gigas has been introduced widely and massively and became an economically important aquaculture species on a global scale. We estimated heritabilities of growth and shell color traits and their genetic correlations in black shell strain of C. gigas. Analyses were performed on 22 full-sib families in a nested mating design including 410 individuals at harvest (24 months of age). The parentage assignment was inferred based on four panels of multiplex PCR markers including 10 microsatellite loci and 94.9% of the offspring were unambiguously assigned to single parent pairs. The Spearman correlation test (r = ? 0.992, P < 0.001) demonstrated the high consistency of the shell pigmentation (SP) and L* and their same efficacy in shell color measurements. The narrow-sense heritability estimated under the animal model analysis was 0.18 ± 0.12 for shell height, 0.25 ± 0.16 for shell length, 0.10 ± 0.09 for shell width, 0.42 ± 0.20 for total weight, 0.32 ± 0.18 for shell weight, and 0.68 ± 0.16 for L*, 0.69 ± 0.16 for shell pigmentation, respectively. The considerable additive genetic variation in growth and shell color traits will make it feasible to produce genetic improvements for these traits in selective breeding program. High genetic and phenotypic correlations were found among growth traits and among shell color traits. To optimize a selection strategy for both fast growth and pure dark shell strain of C. gigas, it is proposed to take both total weight and black shell as joint objective traits in selective breeding program. Our study offers an important reference in the process of selective breeding in black shell color stain of C. gigas and will facilitate to develop favorable breeding strategies of genetic improvements for this economically important strain.  相似文献

Boletus edulis Bull. is one of the most economically and gastronomically valuable fungi worldwide. Sporocarp production normally occurs when symbiotically associated with a number of tree species in stands over 40 years old, but it has also been reported in 3-year-old Cistus ladanifer L. shrubs. Efforts toward the domestication of B. edulis have thus focused on successfully generating C. ladanifer seedlings associated with B. edulis under controlled conditions. Microorganisms have an important role mediating mycorrhizal symbiosis, such as some bacteria species which enhance mycorrhiza formation (mycorrhiza helper bacteria). Thus, in this study, we explored the effect that mycorrhiza helper bacteria have on the efficiency and intensity of the ectomycorrhizal symbiosis between C. ladanifer and B. edulis. The aim of this work was to optimize an in vitro protocol for the mycorrhizal synthesis of B. edulis with C. ladanifer by testing the effects of fungal culture time and coinoculation with the helper bacteria Pseudomonas fluorescens Migula. The results confirmed successful mycorrhizal synthesis between C. ladanifer and B. edulis. Coinoculation of B. edulis with P. fluorescens doubled within-plant mycorrhization levels although it did not result in an increased number of seedlings colonized with B. edulis mycorrhizae. B. edulis mycelium culture time also increased mycorrhization levels but not the presence of mycorrhizae. These findings bring us closer to controlled B. edulis sporocarp production in plantations.  相似文献

During laboratory testing it was shown that the presence of the White Sea sponge Halichondria panicea on the shell of the mussel Mytilus edulis negatively affects the functional state of the mollusk as expressed in the increased heart rate recovery time after functional test loads (a 50% water salinity drop for 1 hour). It was found that the degree of this negative influence rises with an increase both in the projective cover of mussel shells by the sponge and in the size/age parameters of mussels.  相似文献

The shells of the bivalve mollusks are organo-mineral structures predominantly composed of calcium carbonate, but also of a minor organic matrix, a mixture of proteins, glycoproteins, and polysaccharides. These proteins are involved in mineral deposition and, more generally, in the spatial organization of the shell crystallites in well-defined microstructures. In this work, we extracted different organic shell extracts (acid-soluble matrix, acid-insoluble matrix, water-soluble matrix, guanidine HCl/EDTA-extracted matrix, referred as ASM, AIM, WSM, and EDTAM, respectively) from the shell of the scallop Pecten maximus and studied their biological activities on human articular chondrocytes (HACs). We found that these extracts differentially modulate the biological activities of HACs, depending on the type of extraction and the concentration used. Furthermore, we showed that, unlike ASM and AIM, WSM promotes maintenance of the chondrocyte phenotype in monolayer culture. WSM increased the expression of chondrocyte-specific markers (aggrecan and type II collagen), without enhancing that of the main chondrocyte dedifferentiation marker (type I collagen). We also demonstrated that WSM could favor redifferentiation of chondrocyte in collagen sponge scaffold in hypoxia. Thus, this study suggests that the organic matrix of Pecten maximus, particularly WSM, may contain interesting molecules with chondrogenic effects. Our research emphasizes the potential use of WSM of Pecten maximus for cell therapy of cartilage.  相似文献

In water-limited environments of the intermountain region of North America, summer precipitation may play a role in the structure and function of aridland communities and ecosystems. This study examined the potential reliance on summer precipitation of two widespread, coexisting woody species in the southwestern United States, Pinus edulis Englmn. (Colorado piñon) and Juniperus osteosperma (Torr) Little (Utah juniper). The current distributions of P. edulis and J. osteosperma are highly suggestive of different dependencies on summer rainfall. We hypothesized that P. edulis was dependent on summer precipitation, utilizing summer precipitation even during extremely dry summers, whereas J. osteosperma was not dependent, using summer precipitation only when amounts were above some minimum threshold. Using sap flux and stable isotopic methods to assess seasonal water sources and water use efficiency, we examined the response of these two species to seasonal variations in moisture at a site located near the northern limits of the North American monsoon. Both sap flux and isotopic results indicated that P. edulis was responsive to summer rain, while J. osteosperma was not. Following summer rain events, sap flux density increased in P. edulis for several days, but not in J. osteosperma. Isotopic evidence indicated that P. edulis took up summer-derived moisture to a greater extent than J. osteosperma. Values of the natural abundance stable isotope ratio of carbon of leaf soluble carbohydrates increased over the summer for P. edulis, indicative of assimilation at higher water use efficiency, but were invariant for J. osteosperma. Our results supported the hypothesis that P. edulis and J. osteosperma are differentially sensitive to summer precipitation and are discussed in the light of potential changes in the seasonality of precipitation associated with climate change.  相似文献

Coastal dune areas are valuable ecosystems, generally impacted by habitat destruction and invasive alien species. In this study, we assessed how human disturbance and invasion by Carpobrotus edulis impact the soils and the establishment of native flora in the north-western coastal regions of Spain. We compared soil characteristics (pH, conductivity, water content, nutrients and enzymatic activities) and native plant as well as C. edulis fitness correlates (germination and early growth) between uninvaded and invaded soils from urban and natural coastal dune areas. We found that human disturbance impacts coastal soils by increasing organic matter and water content, modifying soil nutrients and cycles, and reducing the pH in urban soils. The presence of invasive C. edulis further increases these impacts. These changes in soil characteristics allow for the establishment of the native, but ruderal, Scolymus hispanicus and non-native C. edulis, both of which are not adapted to the typically limiting conditions of coastal dunes. In some instances, the coastal dune endemic, Malcolmia littorea, showed no fitness effects in response to urbanization or the presence of C. edulis. These results suggest that human disturbed coastal areas might be more easily invaded than natural areas. More broadly, our findings of differential responses of different native species to disturbance and invasion, illustrate the need for multi-taxon approaches when assessing the impacts of invasive species.  相似文献

The results of long-term studies of the circadian rhythms of activity in Trigonoscelis gigas and T. sublaevicollis, typical and abundant representatives of the tenebrionid fauna in the Kara Kum desert are given. T. gigas and T. sublaevicollis are omni-seasonal species: their adults are observed on the sand surface from spring till late autumn. They spend their resting period in soil. Adults of both species are similar in morphology, ecology, and behavior, strongly differing in the circadian rhythm pattern. T. gigas are active only at twilight, whereas T. sublaevicollis are characterized by nocturnal activity. Therefore, inhabiting the same habitat, T. gigas and T. sublaevicollis never meet each other under natural conditions, independently of the season and weather. Hence, the circadian rhythm of activity can serve as a reliable mechanism of reproductive isolation in closely related tenebrionid species.  相似文献

In the Karakum Desert (Turkmenistan) the beetles Trigonoscelis gigas are only active in the morning and evening while T. sublaevicollis are strictly nocturnal, regardless of the season and weather. The daily activity rhythm of T. gigas and T. sublaevicollis was studied in the laboratory according to the following pattern: 5 days under a light-darkness cycle of 15: 9 h (LD 15: 9), then 10 days in constant darkness (DD), and then 10 more days under alternating 1-h pulses of light and darkness (LD 1: 1). The temperature was 25°C in all the modes. At LD 15: 9, beetles of both species maintained a 24-h period and a natural pattern of the activity rhythm. In DD, the circadian rhythm ran with a period of 23.5 ± 0.3 h (n = 40) in T. gigas and 23.6 ± 0.4 h (n = 40) in T. sublaevicollis. At DD, the morning and evening activity peaks of T. gigas merged to form a rhythm with only one peak. Under LD 1: 1, both T. gigas and T. sublaevicollis recovered a 24-h period of the rhythm, while the rhythm of T. gigas regained the two-peak structure. Our research confirmed the assumption of Tshernyshev (1980) about the 24-h period of the free-running endogenous rhythm and the distorting effect of constant conditions on this rhythm.  相似文献

Semi-sessile Mytilus mussels are used as indicators of climate changes, but their geographic distribution is not sufficiently known in the Arctic. The aim of this study was to investigate the taxonomic status and genetic differentiation of Mytilus populations in a Northwest Greenlandic fjord at Maarmorilik, impacted by contaminations from a former mine. In this study, mussels were collected at three sites differing in exposure to environmental factors. A total of 54 polymorphic SNPs found in the Mytilus EST and DNA sequences analyzed were successfully applied to 256 individuals. The results provided the first evidence for the existence of M. trossulus in Greenland. The mussel from M. trossulus and M. edulis taxa are shown to coexist and hybridize in the fjord. The three studied sites were found to differ significantly in the distribution of taxa with a higher prevalence of M. trossulus in the inner fjord. The identified M. edulis × M. trossulus hybrids mostly had a hybrid index score of about 0.5, indicating a similar number of alleles characteristic for M. trossulus and M. edulis. There was a low number of backcrosses between ‘pure’ taxa and hybrids. This newly discovered hybrid zone between the two taxa is unique in comparison with the Canadian populations. As Mytilus mussels in Greenland hitherto have been regarded as the one taxon M. edulis, the results have importance for biogeography and future monitoring and environmental studies.  相似文献

The seaweed genus Gracilaria is a potential candidate for the production of bioethanol due to its high carbohydrate content. Gracilaria is abundant throughout the world and can be found in both wild and cultivated forms. Differences in the ecological factors such as temperature, salinity, and light intensity affecting wild and cultivated specimens may influence the biochemical content of seaweeds, including the carbohydrate content. This study aimed to investigate the proximate composition and potential bioethanol production of wild and cultivated G. gigas and G. verrucosa. Bioethanol was produced using separate hydrolysis fermentation (SHF), employing a combination of enzymatic and acid hydrolysis, followed by fermentation with Saccharomyces cerevisiae ATCC 200062. The highest carbohydrate content was found in wild G. gigas. The highest galactose and glucose contents (20.21 ± 0.32 and 9.70 ± 0.49 g L?1, respectively), as well as the highest production of bioethanol (3.56 ± 0.02 g L?1), were also found in wild G. gigas. Thus, we conclude that wild G. gigas is the most promising candidate for bioethanol production. Further research is needed to optimize bioethanol production from wild G. gigas. Domestication of wild G. gigas is a promising challenge for aquaculture to avoid overexploitation of this wild seaweed resource.  相似文献

Background

The re-sequencing of C. angulata has revealed many polymorphisms in candidate genes related to adaptation to abiotic stress that are not present in C. gigas; these genes, therefore, are probably related to the ability of this oyster to retain high concentrations of toxic heavy metals. There is, in addition, an unresolved controversy as to whether or not C. angulata and C. gigas are the same species or subspecies. Both oysters have 20 metacentric chromosomes of similar size that are morphologically indistinguishable. From a genomic perspective, as a result of the great variation and selection for heterozygotes in C. gigas, the assembly of its draft genome was difficult: it is fragmented in more than seven thousand scaffolds.

Results

In this work sixty BAC sequences of C. gigas downloaded from NCBI were assembled in BAC-contigs and assigned to BACs that were used as probes for mFISH in C. angulata and C. gigas. In addition, probes of H3, H4 histone, 18S and 5S rDNA genes were also used. Hence we obtained markers identifying 8 out the 10 chromosomes constituting the karyotype. Chromosomes 1 and 9 can be distinguished morphologically. The bioinformatic analysis carried out with the BAC-contigs annotated 88 genes. As a result, genes associated with abiotic adaptation, such as metallothioneins, have been positioned in the genome. The gene ontology analysis has also shown many molecular functions related to metal ion binding, a phenomenon associated with detoxification processes that are characteristic in oysters. Hence the provisional integrated map obtained in this study is a useful complementary tool for the study of oyster genomes.

Conclusions

In this study 8 out of 10 chromosome pairs of Crassostrea angulata/gigas were identified using BAC clones as probes. As a result all chromosomes can now be distinguished. Moreover, FISH showed that H3 and H4 co-localized in two pairs of chromosomes different that those previously escribed. 88 genes were annotated in the BAC-contigs most of them related with Molecular Functions of protein binding, related to the resistance of the species to abiotic stress. An integrated genetic map anchored to the genome has been obtained in which the BAC-contigs structure were not concordant with the gene structure of the C. gigas scaffolds displayed in the Genomicus database.

Background

The shells of various Haliotis species have served as models of invertebrate biomineralization and physical shell properties for more than 20 years. A focus of this research has been the nacreous inner layer of the shell with its conspicuous arrangement of aragonite platelets, resembling in cross-section a brick-and-mortar wall. In comparison, the outer, less stable, calcitic prismatic layer has received much less attention. One of the first molluscan shell proteins to be characterized at the molecular level was Lustrin A, a component of the nacreous organic matrix of Haliotis rufescens. This was soon followed by the C-type lectin perlucin and the growth factor-binding perlustrin, both isolated from H. laevigata nacre, and the crystal growth-modulating AP7 and AP24, isolated from H. rufescens nacre. Mass spectrometry-based proteomics was subsequently applied to to Haliotis biomineralization research with the analysis of the H. asinina shell matrix and yielded 14 different shell-associated proteins. That study was the most comprehensive for a Haliotis species to date.

Methods

The shell proteomes of nacre and prismatic layer of the marine gastropod Haliotis laevigata were analyzed combining mass spectrometry-based proteomics and next generation sequencing.

Results

We identified 297 proteins from the nacreous shell layer and 350 proteins from the prismatic shell layer from the green lip abalone H. laevigata. Considering the overlap between the two sets we identified a total of 448 proteins. Fifty-one nacre proteins and 43 prismatic layer proteins were defined as major proteins based on their abundance at more than 0.2% of the total. The remaining proteins occurred at low abundance and may not play any significant role in shell fabrication. The overlap of major proteins between the two shell layers was 17, amounting to a total of 77 major proteins.

Conclusions

The H. laevigata shell proteome shares moderate sequence similarity at the protein level with other gastropod, bivalve and more distantly related invertebrate biomineralising proteomes. Features conserved in H. laevigata and other molluscan shell proteomes include short repetitive sequences of low complexity predicted to lack intrinsic three-dimensional structure, and domains such as tyrosinase, chitin-binding, and carbonic anhydrase. This catalogue of H. laevigata shell proteins represents the most comprehensive for a haliotid and should support future efforts to elucidate the molecular mechanisms of shell assembly.

Holocene record of ostracode shell chemistry from the South Basin of Lake Manitoba shows how isostatically and climatically induced changes in the surface and groundwater input to the overall hydrologic budget affected the hydrochemistry. The shallow South Basin experienced periods of extensive shell transport from isolated pools and groundwater seeps, as suggested by disparate chemistry of Fabaerformiscandona rawsoni shells from the same 1-cm-thick sediment sample. Temporal changes reconstructed from ostracode species and F. rawsoni shell chemistry include, for some depth intervals, shells of individuals that were not living at the same time or were not living at the core site. Shell chemistry before the South and North Basins became connected at 1850 BP broadly show a hydrologically closed-basin behavior with alternating periods of a more and less stressful environment to ostracodes. F. rawsoni with high δ 18O and low Mg/Ca or low δ 18O and high Mg/Ca were likely transported from marginal evaporated pools or groundwater seeps. Overall δ 18O and δ 13C values increase over time, but at levels for which multiple analyses were made, the values show a wide range, and indicate a highly variable hydrochemical environment both spatially and temporally (over one summer or multiple summers).  相似文献

Shells of oysters from Turonian strata in Westphalia contain dwelling tubes of polychaete worms sunk into the shell material. The roofs of the tubes consist of shell material secreted by the oysters. Thus the tubes are intra vitam deformations of the shell, resembling the “mudblisters” of recent oysters. Usually, they are directed centripetally towards the muscle scar and have a round external opening perforating the shell. They are described asOstreoblabe perforans n. g. n. sp. Since they were probably oriented with their front part toward the inside of the shell, penetrating the pallial line, they were most likely parasites. In addition, long, meandering furrows sunk into the shells of pelecypods are described asMaeandropolydora decipiens n. g. n. sp. andM. sulcans n. g. n. sp. The realPolydora is not found in Pre-Tertiary strata and is probably not related genetically toPolydora-like forms (Caulostrepsis) in Devonian strata.  相似文献

The aim of this study was to determine the antimicrobial effects of lemongrass essential oil (C. flexuosus) and to determine cytotoxic effects of both test compounds on human dermal fibroblasts. Antimicrobial susceptibility screening was carried out using the disk diffusion method. Antimicrobial resistance was observed in four of five Acinetobacter baumannii strains with two strains confirmed as multi-drug-resistant (MDR). All the strains tested were susceptible to both lemongrass and citral with zones of inhibition varying between 17 to 80 mm. The mean minimum inhibitory concentration (MIC) and minimum bactericidal concentration (MBC) of citral (mic—0.14 % and mbc—0.3 % v/v) was lower than that of Lemongrass (mic—0.65 % and mbc—1.1 % v/v) determined using the microtitre plate method. Cell viability using human dermal fibroblasts (HDF; 106-05a) was determined following exposure to both compounds and a control (Grapeseed oil) using the XTT assay and the IC50 determined at 0.095 % (v/v) for citral and 0.126 % (v/v) for lemongrass. Grapeseed oil had no effect on cell viability. Live cell imaging was performed using the LumaScope 500 imaging equipment and changes in HDF cell morphology such as necrotic features and shrinkage were observed. The ability of lemongrass essential oil (EO) and citral to inhibit and kill MDR A. baumannii highlights its potential for use in the management of drug-resistant infections; however, in vitro cytotoxicity does suggest further tests are needed before in vivo or ex vivo human exposure.  相似文献

In spring 2014 and 2016, we studied Rhodanthidium siculum in dunes south of Syracuse (Sicily). The females search for one of the frequently empty snail shells (mostly of Theba pisana), checking their size. In a suitable snail shell, a mixture of sand and saliva is deposited in the navel. A severe struggle among several males gets started. At every opportunity, the males try to copulate with the females. Large males occupy the snail shell that is used by the female to bring in pollen and nectar (preferably from Galactites tomentosus, Centaurea sphaerocephala, Glebionis coronaria and Lotus creticus). The procedure of harvesting lasts about 2–3 h, sometimes several hours. During this period, the female is visited for copulation every 2 min (initially even more frequently), preferentially by one large male, but small males occasionally mate as well. After laying one or two eggs, the female closes the snail shell with pieces of seashells or snail shells, aggregated with sand and saliva. The female transports the closed snail shell to a safe site. Depending on weather conditions, this may take several days. The maximum distance of movement observed was about 10 m. Finally, the completed snail shell will be buried, most often beneath a plant. The burial follows a certain pattern of movement. Females ensure higher pollination efficiency than the males due to their higher flower constancy.  相似文献

In the Lower Plattenkalk of Bergisch Gladbach occur, besides the well known?Hians”-beds, polymictic accumulations of shell material. Together with the valves ofMartinia inflata (Schnur),Uncinulus parallelepipedus (Br.) andIsopoma brachyptyctum (Schnur) are great numbers ofSpinatrypa tubaecostata (Paeck.) andAtrypa prisca gladbachensis n. subsp. In spite of the fact that the host rock was deposited as shell debris, in particular the rhynchonellids and atrypids are easily separated from it in a well preserved state. Atrypa (Desquamatia) prisca gladbachensis n. subsp. is commonly found with its alations intact. This subspecies, which is here described in some detail, fluorisched on a marly ooze bottom. The expanded valves increased the food gathering field, while the alations, surrounding the shell margin almost like a flange, functioned as effective particle filters. Because of this, the free valves floated under wave attack and became fossilized almost undamaged. The brachiopod shells and the organoplastic debris were washed on to discontinuity surfaces which probably had already been lithified by algal growth.  相似文献

Root explants of a wild passionfruit species (Passiflora cincinnata) and three P. edulis commercial populations (‘FB 100’, ‘FB 200’, and ‘FB 300’) were incubated on Murashige and Skoog (MS) medium supplemented with 4.44 μM 6-benzyladenine (BA) to induce shoot organogenesis. Shoots elongated in liquid medium with 2.89 μM gibberellic acid (GA3) under agitation were rooted in coconut fiber and acclimatized followed by transfer to a greenhouse into pots containing mixture of coconut fiber and Plantmax® (1:1). Explant samples were collected during organogenesis and submitted to light and scanning electron microscopy (SEM). Root explants of P. cincinnata responded earlier than those of P. edulis. However, on the third assessment, at 90 days, the genotype ‘FB 200’ showed shoot number significantly higher than ‘FB 100’ and ‘FB 300’, not differing from P. cincinnata. Organogenesis in P. cincinnata and P. edulis occurred via direct pathway, which was confirmed by anatomical studies and SEM. Flow cytometric analysis revealed no variation in DNA content of regenerated plantlets among all genotypes. Nuclear DNA (2C) values (pg) in regenerants of P. cincinnata (2.99 pg) and P. edulis (3.26–3.28 pg) were consistent with DNA amounts of seed-derived control plants.  相似文献
