A quantitative analysis of photoreceptor properties was performed in the retina of the nocturnal deer mouse, Peromyscus maniculatus, using pigmented (wildtype) and albino animals. The aim was to establish whether the deer mouse is a more suitable model species than the house mouse for photoreceptor studies, and whether oculocutaneous albinism affects its photoreceptor properties. In retinal flatmounts, cone photoreceptors were identified by opsin immunostaining, and their numbers, spectral types, and distributions across the retina were determined. Rod photoreceptors were counted using differential interference contrast microscopy. Pigmented P. maniculatus have a rod-dominated retina with rod densities of about 450.000/mm2 and cone densities of 3000 - 6500/mm2. Two cone opsins, shortwave sensitive (S) and middle-to-longwave sensitive (M), are present and expressed in distinct cone types. Partial sequencing of the S opsin gene strongly supports UV sensitivity of the S cone visual pigment. The S cones constitute a 5-15% minority of the cones. Different from house mouse, S and M cone distributions do not have dorsoventral gradients, and coexpression of both opsins in single cones is exceptional (<2% of the cones). In albino P. maniculatus, rod densities are reduced by approximately 40% (270.000/mm2). Overall, cone density and the density of cones exclusively expressing S opsin are not significantly different from pigmented P. maniculatus. However, in albino retinas S opsin is coexpressed with M opsin in 60-90% of the cones and therefore the population of cones expressing only M opsin is significantly reduced to 5-25%. In conclusion, deer mouse cone properties largely conform to the general mammalian pattern, hence the deer mouse may be better suited than the house mouse for the study of certain basic cone properties, including the effects of albinism on cone opsin expression.  相似文献

In mammals, each cone had been thought to contain only one single type of photopigment. It was not until the early 1990s that photopigment coexpression was reported. In the house mouse, the distribution of color cones shows a characteristic division. Whereas in the upper retinal field the ratio of short wave to middle-to-long wave cones falls in the usual range (1:10), in the ventral retinal field M/L-pigment expression is completely missing. In the transitional zone, numerous dual cones are detectable (spatial coexpression). In other species without retinal division, dual cones appear during development, suggesting that M/L-cones develop from S-cones. Dual elements represent a transitory stage in M/L-cone differentiation that disappear with maturation (transitory coexpression). These two phenomena seem to be mutually exclusive in the species studied so far. In the comparative part of this report the retinal cone distribution of eight rodent species is reported. In two species dual cones appear in adult specimens without retinal division, and dual elements either occupy the dorsal peripheral retina, or make up the entire cone population. This is the first observation proving that all cones of a retina are of dual nature. These species are good models for the study of molecular control of opsin expression and renders them suitable sources of dual cones for investigations on the role and neural connections of this peculiar cone type. In the developmental part, the retinal maturation of other species is examined to test the hypothesis of transitory coexpression. In these species S-pigment expression precedes that of the M/L-pigment, but dual cones are either identified in a small number or they are completely missing from the developing retina. These results exclude a common mechanism for M/L-cone maturation: they either transdifferentiate from S-cones or develop independently.  相似文献

The influence of age, gender and sexual activity on both hepatic levels of some trace elements (Fe, Cu, Zn, Mn, Se) and the activities of glutathione-S-transferase (GST) and superoxide dismutase (SOD) was investigated in Wood mice (Apodemus sylvaticus) and Algerian mice (Mus spretus). Animals were taken from a riverside community of an unpolluted area of central Portugal. Adult A. sylvaticus presented the highest hepatic mean concentrations of Cu and Mn, whereas adult M. spretus had the highest Fe concentration in the liver. Moreover, an influence of age on the contents of Fe, Zn, and Mn has been observed in A. sylvaticus, whereas in M. spretus an influence of gender and sexual activity was only detected on Zn levels. In contrast, enzyme activities were not influenced by the studied variables, despite a tendency for an increase in SOD activity in sexually active M. spretus. GST activity was species dependent, whereas SOD activity was similar between species. These findings were analyzed regarding the relationship of both essential trace elements and the two antioxidant enzymes with physiological and metabolic pathways related to life cycles in the two species of mice. Results enhanced the understanding of A. sylvaticus and M. spretus as biological models, allowing their future use as bioindicators of environmental toxicity.  相似文献

In this study, the cytoarchitecture of retina in 10 mountain-stream teleosts (seven cyprinid and three loach species) of India was examined by conventional light microscopy. The mountain-streams are shallow, cold water bodies having high turbidity in the Monsoon and clear water in the colder months. The incumbent fishes are, thus, periodically exposed to low and bright light of the streams, respectively. It was of particular interest to see whether their retina would reflect adaptations to the changing photic environments of the mountain streams. The retina of the cyprinids possess multiple cone types, a moderate cone density, high convergence ratios and prominent retinomotor responses. Triple cones occur only in the danio, Danio aequipinnatus; other cyprinids possess miniature, short and long single cones, and double or twin cones. In general, the cones are bulky in appearance. Cone droplets (ellipsosomes) are found in Garra lamta and G. gotyla gotyla. All cone types, including the miniature single cones, undergo elongation during dark adaptation. The common retinal features in the loaches are the bulky cones, ellipsosomes, low rod density and low convergence ratios. There is no area-like specialization of the retina in any of the species examined. No relationship is obvious between cone pattern and ambient light or food habit in any of those teleosts studied; row pattern is present throughout the retina of the surface-dwelling, total insectivores (cyprinids: Danio aequipinnatus and Barilius spp.), whereas square mosaic pattern is present in the bottom-dwelling total herbiovores (Garra spp.) as well as in the insectivores (loaches: Nemacheilus spp.). The retinal features have suggested that the cyprinids are well-adapted to both clear and turbid water (ellipsosomes, multiple cone types, bulky cones, high rod count and high convergence ratios); the loaches, on the other hand, are better adapted to clear water of the mountain streams.  相似文献

There is an increasing interest in understanding how species respond to the modifications of habitat attributes in urban areas. Patterns in the occurrence and abundance of small mammal communities in 15 isolated patches of remnant natural and semi-natural habitat of Porto Metropolitan Area (Portugal) were assessed against environmental characteristics (from both the patch and the surrounding matrix) of each patch using multiple regressions and canonical correspondence analysis. Four species of common small mammals were found: wood mouse (Apodemus sylvaticus), greater white-toothed shrew (Crocidura russula), Algerian mouse (Mus spretus) and house mouse (Mus musculus). Our results showed that both relative abundance and species richness were negatively affected by urbanization. The species richness also displayed a negative association with the increase of forest around the patch but relative abundance showed the opposite trend. At the species level, the relative abundance of A. sylvaticus and C. russula showed a negative association with urbanization. Our results reveal that these two species also benefit from a mosaic of habitats and pervious areas in the surrounding matrix. The relative abundance of M. spretus and M. musculus showed a negative effect of forest area around the patch. Understanding how the increase of urbanization affects small mammals will be particularly useful for the managers of urban landscapes, as these animals occupy a pivotal position in the ecosystems.  相似文献

The spectral absorption characteristics of the retinal photoreceptors of the blue tit (Parus caeruleus) and blackbird (Turdus merula) were investigated using microspectrophotometry. The retinae of both species contained rods, double cones and four spectrally distinct types of single cone. Whilst the visual pigments and cone oil droplets in the other receptor types are very similar in both species, the wavelength of maximum sensitivity (λmax) of long-wavelength-sensitive single and double cone visual pigment occurs at a shorter wavelength (557 nm) in the blackbird than in the blue tit (563 nm). Oil droplets located in the long-wavelength-sensitivesingle cones of both species cut off wavelengths below 570–573 nm, theoretically shifting cone peak spectral sensitivity some 40 nm towards the long-wavelength end of the spectrum. This raises the possibility that the precise λmax of the long-wavelength-sensitive visual pigment is optimised for the visual function of the double cones. The distribution of cone photoreceptors across the retina, determined using conventional light and fluorescence microscopy, also varies between the two species and may reflect differences in their visual ecology. We purpose here to analyze and compare the population and topography of cone photoreceptors in two mouse strains using automated routines, and to design a method of retinal sampling for their accurate manual quantification. In whole-mounted retinas from pigmented C57/BL6 and albino Swiss mice, the longwave-sensitive (L) and the shortwave-sensitive (S) opsins were immunodetected to analyze the population of each cone type. In another group of retinas both opsins were detected with the same fluorophore to quantify all cones. In a third set of retinas, L-opsin and Brn3a were immunodetected to determine whether L-opsin+cones and retinal ganglion cells (RGCs) have a parallel distribution. Cones and RGCs were automatically quantified and their topography illustrated with isodensity maps. Our results show that pigmented mice have a significantly higher number of total cones (all-cones) and of L-opsin+cones than albinos which, in turn, have a higher population of S-opsin+cones. In pigmented animals 40% of cones are dual (cones that express both opsins), 34% genuine-L (cones that only express the L-opsin), and 26% genuine-S (cones that only express the S-opsin). In albinos, 23% of cones are genuine-S and the proportion of dual cones increases to 76% at the expense of genuine-L cones. In both strains, L-opsin+cones are denser in the central than peripheral retina, and all-cones density increases dorso-ventrally. In pigmented animals S-opsin+cones are scarce in the dorsal retina and very numerous in the ventral retina, being densest in its nasal aspect. In albinos, S-opsin+cones are abundant in the dorsal retina, although their highest densities are also ventral. Based on the densities of each cone population, we propose a sampling method to manually quantify and infer their total population. In conclusion, these data provide the basis to study cone degeneration and its prevention in pathologic conditions.  相似文献

Regional variations of cone density, cone types and cone mosaic were investigated by light microscopy in yolk sac fry, parr and adults of salmon (Salmo salar) and trout (Salmo trutta trutta) and related to the feeding habits of different stages. The retinal organization of the two species is similar, as are their feeding habits. In parr and adults of both species a region with high cone density was found in the ventro-temporal part of the retina. The lowest values of cone density were found centrally in the dorsal half of the retina. An increase in cone density towards the ora serrata was noted in all retinae investigated. There is a good agreement between the density distribution of the cones in parr of salmon and trout and their feeding habits. The parr feed to a great extent on the invertebrate drift. Facing the current, the parr search the water region in front and above themselves for food organisms, thereby probably making use of the ventro-temporal area of high cone density. The connection between the cone density distribution and the feeding habits is less clear in adult salmon and trout, but the high cone density region is probably used when they feed on surface organisms and schooling prey. The ecological significance of cone types and cone mosaics is discussed. During growth there was a change of the cone mosaic from a regular square pattern in the fry towards a less regular square or row pattern in the adults. The position of regions with high and low cone density did not change during development in each species and had a similar distribution in the two species. On the other hand, the range of cone density values becomes relatively greater as the animal grows older. The cone density distribution seems to be more homogeneous in parr than in adults. Differences in the relative density distribution of the cones are influenced by recruitment at the retinal periphery and by spreading out of existing cells. In addition to the ora serrata, mitoses were also observed in more central parts of the retina having well-differentiated receptors and a cone mosaic. Mitoses were noted in all three nuclear layers but were most frequent in the outer nuclear layer, in which the resulting cells probably differentiate into rods. Undifferentiated zones similar to the peripheral growth zone of the retina were found on both sides of the falciform process in fry. In parr and adults an undifferentiated zone persisted only on the temporal side. It probably contributes to the high cone density in the ventro-temporal part of the retina by a delivery of new cells.  相似文献

We report the expression of three visual opsins in the retina of the little brown bat (Myotis lucifugus, Vespertilionidae). Gene sequences for a rod-specific opsin and two cone-specific opsins were cloned from cDNA derived from bat eyes. Comparative sequence analyses indicate that the two cone opsins correspond to an ultraviolet short-wavelength opsin (SWS1) and a long-wavelength opsin (LWS). Immunocytochemistry using antisera to visual opsins revealed that the little brown bat retina contains two types of cone photoreceptors within a rod-dominated background. However, unlike other mammalian photoreceptors, M. lucifugus cones and rods are morphologically indistinguishable by light microscopy. Both photoreceptor types have a thin, elongated outer segment. Using microspectrophotometry we classified the absorption spectrum for the ubiquitous rods. Similar to other mammals, bat rhodopsin has an absorption peak near 500 nm. Although we were unable to confirm a spectral range, cellular and molecular analyses indicate that M. lucifugus expresses two types of cone visual pigments located within the photoreceptor layer. This study provides important insights into the visual capacity of a nocturnal microchiropteran species.  相似文献

Pocket gophers (Thomomys bottae) are rodents that spend much of their lives in near-lightless subterranean burrows. The visual adaptations associated with this extreme environment were investigated by making anatomical observations of retinal organization and by recording retinal responses to photic stimulation. The size of the eye is within the normal range for rodents, the lens transmits light well down into the ultraviolet, and the retina conforms to the normal mammalian plan. Electroretinogram recording revealed the presence of three types of photopigments, a rod pigment with a spectral peak of about 495 nm and two types of cone pigment with respective peak values of about 367 nm (UV) and 505 nm (medium-wavelength sensitive). Both in terms of responsivity to lights varying in temporal frequency and in response recovery following intense light adaptation, the cone responses of the pocket gopher are similar to those of other rodents. Labeling experiments indicate an abundance of cones that reach densities in excess of 30,000 mm–2. Cones containing UV opsin are found throughout the retina, but those containing medium-wavelength sensitive opsin are mostly restricted to the dorsal retina where coexpression of the two photopigments is apparently the rule. Rod densities are lower than those typical for nocturnal mammals.  相似文献

We describe the process of retinal development in mandarinfish Siniperca chuatsi from larvae to young fish. The developmental characteristics of the retinal structure and related cells were identified. Siniperca chuatsi were found to exhibit an altricial mode of retinal development that required considerable time to be completed after hatching. The retina was classed as a pure cone type during the early developmental stage. In the subsequent developmental stages, however, double cones gradually occupied the majority of the cone cells, while rod cells represented the majority of the photoreceptor cells. The outer segment (OS) of the rod cells were significantly longer compared with other morphological features, the OS of the two kinds of cone cells were significantly elongated and the diameters of the inner segment (IS) and OS of the double cone cells were significantly narrower in the later developmental stages. Combined with the scattered arrangement of cone cells at the different stages, the retina was found to have sacrificed a considerable part of visual acuity in the developmental process. The distribution of cone cells was observed to have gradually become regionalised during development. The findings of the present study also indicated that S. chuatsi have a high photosensitivity under dim light conditions as a result of specialised structures of the OS of photoreceptor cells and an increased number of rod cells. The loose arrangement of the cone mosaic presumably resulted in a poor imaging quality and, to some extent, the regionalisation of the cone-cell distribution compensated for the above shortcomings, which would enhance the ability of S. chuatsi to perceive targets in important directions for effective predation behaviour.  相似文献

Retinal cone photoreceptors (cones) serve daylight vision and are the basis of color discrimination. They are subject to degeneration, often leading to blindness in many retinal diseases. Calcium (Ca2+), a key second messenger in photoreceptor signaling and metabolism, has been proposed to be indirectly linked with photoreceptor degeneration in various animal models. Systematically studying these aspects of cone physiology and pathophysiology has been hampered by the difficulties of electrically recording from these small cells, in particular in the mouse where the retina is dominated by rod photoreceptors. To circumvent this issue, we established a two-photon Ca2+ imaging protocol using a transgenic mouse line that expresses the genetically encoded Ca2+ biosensor TN-XL exclusively in cones and can be crossbred with mouse models for photoreceptor degeneration. The protocol described here involves preparing vertical sections (“slices”) of retinas from mice and optical imaging of light stimulus-evoked changes in cone Ca2+ level. The protocol also allows “in-slice measurement” of absolute Ca2+ concentrations; as the recordings can be followed by calibration. This protocol enables studies into functional cone properties and is expected to contribute to the understanding of cone Ca2+ signaling as well as the potential involvement of Ca2+ in photoreceptor death and retinal degeneration.  相似文献

Green sturgeon and pallid sturgeon photoreceptors were studied with scanning electron microscopy (SEM), microspectrophotometry and, in the case of the green sturgeon, retinal whole-mounts. The retinas of both species contain both rods and cones: cones comprise between 23% (whole-mount) and 36% (SEM) of the photoreceptors. The cone population of both species is dominated by large single cones, but a rare small single cone is also present. In both species, most rods have long outer segments of large diameter. A rod with a relatively thin outer segment is present in the pallid sturgeon retina. Mean cone packing density for the entire green sturgeon retina is 4,690±891 cones/mm2, with the dorsal retina 14% more dense than the ventral. There is evidence for a horizontal visual streak just above and including the optic disc. Mean rod packing density is 16,006±1,668 rods/mm2 for the entire retina, and fairly uniform throughout. Both species have rods with peak absorbance near 540 nm, as well as short-wavelength-sensitive cones (green: 464.5±0.7 nm; pallid: 439.7±3.5 nm); middle-wavelength-sensitive cones (green: 538.0±1.4 nm; pallid: 537.0±1.7 nm); and long-wavelength-sensitive cones (green: 613.9±3.0 nm; pallid: 617.8±7.6 nm).  相似文献

Cone traits (volatile components and thermogenesis) of three cycad species in the genus Macrozamia were examined for differences related to their specific insect pollinators, the weevil, Tranes spp., or the thrips, Cycadothrips chadwicki. Linalool (>80% of emissions) dominated cone volatile components of M. machinii (Tranes-pollinated) and beta-myrcene was a minor component (<9% of emissions). Volatiles of M. lucida and M. macleayi cones (Cycadothrips-pollinated) were dominated by beta-myrcene (up to 97% of emissions), but no linalool was detected. Pollinator movement into and out of cones coincided with cone thermogenesis and peak odor emission: around sunset for Tranes, and at mid-day for Cycadothrips. Female cone traits were similar to those of their conspecific male cones. Differences in cone traits between Macrozamia species may thus be responsible for conferring pollinator specificity in areas of sympatry.  相似文献

Aim Size and shape of the mandible are investigated across the latitudinal range of the European wood mouse (Apodemus sylvaticus), in order to address the relative importance of genetic structure, insularity, and geographical gradient in patterning morphological variation. Results are compared with those on two Asiatic species of wood mice, A. argenteus and A. speciosus. Location The European wood mouse is sampled by a set of trapping localities including both, islands and mainland populations, as well as the four genetic groups identified in previous studies. The localities cover a latitudinal gradient from 55° N to 36° N. Methods Different Fourier methods are applied to the outlines of mandibles and their results compared in the case of A. sylvaticus. All provide similar results and allow a quantification of the size and shape variations across the geographical range of the European wood mouse. Using the method allowing for the best reduction of the informative data set, a comparison of the European wood mouse with the two Asiatic species was performed. Results Within the European wood mouse A. sylvaticus, a strong latitudinal gradient in mandible shape overrides the influence of insularity and genetic structure. Yet, random morphological divergence in insular conditions can be identified as a secondary process of shape differentiation. Size displays no obvious pattern of variation, neither with insularity or latitude. A comparison with two other species of wood mice suggests that a similar latitudinal gradient in mandible shape exists in different species, mandibles being flatter in the north and wider in the south. Main conclusion The latitudinal gradient in mandible shape observed in the three species of wood mice is interpreted as an intraspecific adaptive response to gradual changes in feeding behaviour.  相似文献

The eye of the deep-sea teleost Lestidiops affinis has been examined primarily by light microscopy and found to possess a duplex retina consisting of two main divisions, a pure-cone and a pure-rod region, with a narrow zone of transition, possessing both cones and rods, joining the two. The pure-cone region is located in the temporal (caudal) part of the retina subserving binocular vision in the rostral direction. It has an area temporalis retinae with particularly long and densely packed single cones arranged in a regular hexagonal mosaic. Joined (double or twin) cones have not been recognized with certainty in the pure-cone region. The pure-rod region, comprising the larger part of the retina, contains rods grouped in bundles separated by retinal pigment epithelium (RPE) processes with pigmented cores. The synaptic endings of the rods are arranged in separate clusters in the outer plexiform layer, there being apparently a separate rod pedicle cluster beneath (vitread to) each rod bundle. Structural comparisons with certain other deep-sea teleosts suggest the likely presence of a retinal tapetum in L. affinis, i.e. each single cone or rod bundle is situated in a reflecting pit formed by the RPE, with a discrete reflector apposed to the tip of each cone outer segment and the tips of the outer segments of each square-cut rod bundle.  相似文献

大别山五针松(Pinus dabeshanensis)为我国特有树种,其天然植株较少且分布范围狭窄,种群更新困难,已被列为国家二级重点保护野生植物。分别于2015年和2016年的10—12月,在目前已知最大天然种群所在地安徽岳西县河图镇大王沟,研究了大别山五针松球果与种子特征、种子雨与土壤种子库、动物对种子的取食和搬运,幼苗分布格局及其与鼠穴分布的关系,以期探明动物在其天然更新中的作用,分析其天然更新不力的原因。结果显示:大别山五针松种子败育率较高,阳坡个体球果发育情况好于阴坡;种子成熟期间没有明显的种子雨,土壤种子库也未调查到完整种子。共记录到母树与球果的访问动物6目11科16种,其中7种动物确定取食种子;地面种子摆放实验显示超过95%以上的种子被啮齿动物捕食或搬运至他处取食或贮藏,不同的摆放处理对啮齿动物的捕食、搬运没有显著影响;小林姬鼠(Apodemus sylvaticus)盗食现象严重,埋藏实验中人工贮点当夜被发现的概率在90%左右,岩松鼠(Sciurotamias davidianus)和小林姬鼠是其种子主要捕食者。研究地的大别山五针松种群于2015年和2016年分别新增一年生幼苗5株和7株,这表明大别山五针松在当地存在天然更新。幼苗多单独生长在母树周围,点格局分析显示在0—0.6 m的尺度范围内呈随机分布;在0.6 m尺度呈聚集分布。大别山五针松幼苗在小林姬鼠巢穴周边分布,其更新格局受到小林姬鼠贮点位置分布情况影响,小林姬鼠极有很可能为大别山五针松的传播者,啮齿动物对大别山五针松种子的捕食与搬运影响了大别山五针松的天然种群更新。  相似文献
