The human neutrophils induce cytotoxic effects on mammalian tumor cells. Hence it may be expected that intracellular enzymatic deficiencies of neutrophils may represent another cancer risk factor. In 205 patients with various malignancies the neutrophil beta-glucuronidase activity has been determined using a semiquantitative cytochemical method. A statistically significant deficiency of this enzyme in neutrophils has been observed in patients with precancerous states of the larynx, cancer of the larynx after radiotherapy and patients with cancer of large intestine. Patients with cancer of the lung and cancer of the stomach showed no changes with that regard whereas those with cancer of the breast demonstrated an increased enzyme activity.  相似文献

The myeloperoxidase of peripheral blood neutrophils revealed by the cytochemical method was investigated in a group of patients with ischemic heart disease (24 men, mean age--51) treated with HBO (10 sessions, under 1.5-1.7 ata, for 60 min) prior to and after each HBO session. The ECG monitoring was performed during these sessions as well. A correlation analysis of the peroxidase index (mean enzyme content in 100 cells), total peroxidase activity and tension index (obtained by computer analysis of cardiac rhythm changes) was carried out in that group of patients. A close correlation was found between the total peroxidase activity and tension index.  相似文献

Summary Alkaline phosphatase and peroxidase activities were investigated in typical neutrophils and in vacuolated cells, the latter considered to be another type of neutrophil in catfish blood. This study was carried out by light- and electron-microscopy. Two lines of neutrophils were recognized in the peripheral blood of catfish. Alkaline phosphatase activity was present only in the vacuolated neutrophils; peroxidase activity appeared only in the typical neutrophils. The presence of two types of neutrophil in catfish blood suggests that important steps of neutrophil evolution occurred in fish.  相似文献

Indole-3-acetic acid (IAA) is toxic for human tumor cells and in association with horseradish peroxidase (HRP) can be used as a new prodrug/enzyme combination for targeted cancer therapy. The toxic effect of IAA on neutrophils, macrophages and lymphocytes is associated with cell peroxidase activity, which is high in neutrophils and low in lymphocytes. The effect of IAA on glucose and glutamine metabolism in leukocytes presenting different peroxidase activities: neutrophils, thioglycollate-elicited macrophages and lymphocytes was investigated. A time-course effect (from 6 to 48 h in culture) of IAA on glucose and glutamine metabolism of neutrophils, thioglycollate-elicited macrophages, and lymphocytes was then carried out. Addition of IAA (0.25 mM) did not have a marked effect on glucose utilization and lactate formation by the three cell types but it raised glutamine consumption and glutamate production by neutrophils and macrophages. IAA had no effect on glutamine consumption and glutamate production by lymphocytes. A strong relationship was found between glutamine utilization (0.999) and glutamate production (0.999) and peroxidase activity. IAA did not change the activities of hexokinase, glucose-6-phosphate dehydrogenase, citrate synthase, lactate dehydrogenase, and phosphate-dependent glutaminase of 24 h cultured neutrophils and lymphocytes. The effect of IAA (1 mM) on glucose and glutamine metabolism was also investigated by 1 h incubated leukocytes in PBS. IAA did not affect glucose and glutamine metabolism of lymphocytes but enhanced glucose and glutamine metabolism by 1 h incubated neutrophils and thioglycollate-elicited macrophages. IAA caused a marked increase on oxygen consumption by neutrophils, which was more pronounced in the presence of the glutamine as compared to glucose. The stimulation of oxygen consumption leads to a reduction in NADH/NAD+ ratio that activates the flux of substrates through the Krebs cycle. Since glutamine is mainly metabolized through the left hand side of the Krebs cycle, a reduction in the redox state of the cells may accelerate the flux of substrates through glutaminolysis. The toxic results presented here show that the affect of IAA in association with peroxidase involves activation of glutamine metabolism.  相似文献

Eighteen patients with established hereditary myeloperoxidase deficiency underwent morphological, ultrastructural, cytochemical and functional analysis in order to correlate the lack of peroxidase from phagocytes with other leucocyte activities. Cytochemical and ultrastructural findings only confirmed the peroxidase defect in neutrophil and monocyte population, whereas normal peroxidase activity was detected in eosinophil granulocytes ("Alius-Grignaschi anomaly"). Morphological analysis, as determined by both ligh and electron microscopy, showed in two patients with total MPO-deficiency a large number of neutrophils (50-60%) with nuclear abnormalities very similar to Pelger-Huet's heterozygous form (two lobed neutrophils having a typical pince-nez appearance and a nuclear chromatin coarser than that of normal PMNL). Other 2 cases displayed a 50-60% five-lobed neutrophils, as occur in congenital nuclear hypersegmentation of PMNL. These findings suggest that Alius-Grignaschi anomaly and Pelger-Huet syndrome can be found associated in the same individuals, since both these abnormalities have a genetic origin. Finally, since an impaired bactericidal and fungicidal activity was observed, no patients displayed particular susceptibility to persistent or severe infections, thus confirming the presence of MPO-independent enzymatic systems.  相似文献

Peroxidase activity was examined cytochemically in the mucosal epithelium along the length of the digestive tract from the esophagus through the large intestine during the development of the bullfrog, Rana catesbeiana. In the tadpole of this species, cells with peroxidase activity were found abundantly in the esophagus, stomach, and large intestine; and the types of such cells differed according to the region: ciliated cells and mucous cells in the esophagus; ciliated cells in the stomach; and brush cells, absorptive cells, and goblet cells in the large intestine, respectively. After metamorphosis, however, peroxidase activity was observed exclusively in absorptive cells and goblet cells in the large intestine. Peroxidase activity was commonly demonstrated in apical vesicles or granules, to some degree in rough endoplasmic reticulum, and in some elements of the Golgi apparatus. Furthermore, reaction product was also found in mucus covering the luminal surface of such epithelial cells. These findings indicate that peroxidase-positive cells, which may have the ability to synthesize peroxidase as a secretory product, were distributed mainly in three regions of the digestive tract in tadpoles (esophagus, stomach, and large intestine), but were centered in one specific region, the large intestine, after metamorphosis. Concomitantly, the variety of types of peroxidase-positive cells decreased during metamorphosis. Our results indicate that some of the peroxidase in the digestive tract may have a secretory origin and may play a role in the defense against microorganisms.  相似文献

Peroxidase activity in rat intestine is stimulated two-fold after bilateral adrenalectomy and is reversed by dexamethasone (9-fluoro-11 beta,17,21-trihydroxy-16 alpha-methyl-1-4-pregnadiene-3,20-dione). The enzyme activity is inhibited on administration of various glucocorticoids of which dexamethasone acts as the most potent inhibitor of the enzyme in vivo. The change of enzyme activity results neither from alteration of the apparent Km of the enzyme nor from enzyme synthesis. Although a small amount of peroxidase is located in the intestinal epithelial cells, a large amount is present in the rest of the intestine. Histochemical studies indicate the presence of peroxidase in the lamina propria, the core of the intestinal villi which contains eosinophil. The peroxidase isolated from the epithelial cell-free intestine is similar to the peroxidase obtained from the pure eosinophil in terms of various physicochemical properties. Dexamethasone also inhibits the eosinophil peroxidase and decreases the number of both circulating and intestinal eosinophil. Studies indicate that a large part of the peroxidase of the intestine is contributed by invading eosinophil and dexamethasone inhibits the enzyme by sequestration of eosinophil both from intestine and blood possibly to the peripheral lymph nodes.  相似文献

Peroxidase activity in the granulocytes of eels was investigated using o-tolidine, paraphenylene-diamine-pyrocatechol, and 4-chloro-l-naphthol as substrates, and cyanide, azide and aminotriazole as inhibitors. Most circulating neutrophils of Anguilla australis Richardson, 1848 and A. dieffenbachii Gray. 1842 showed no peroxidase activity at pH 7.6 and pH 9.0, but a few neutrophils, thought to be mature, were positive. Another granulocyte in the anterior kidney, spleen, parasitized gill tissue and, rarely, the blood contained a cyanide-resistant, azide-inhibited, peroxidase and was tentatively identified as the eosinophil. Neutrophils of A. anguilla (L.) showed granular peroxidase activity which was inhibited by cyanide. The eosinophil was not observed.
Absence of peroxidase from most circulating neutrophils in A. anguilla and A. dieffenbachii , and its pattern in the neutrophil precursors and mature neutrophils of A. anguilla , may be due to two morphologically indistinguishable granule types. Primary peroxidase-negative granules occur in precursors and immature neutrophils and secondary peroxidase-positive granules in mature neutrophils of all three eels. Circulating neutrophils in New Zealand eels seldom mature and are theretore peroxidase-negative, whereas A. anguilla neutrophils are mature and are usually peroxidase-positive.

Superoxide anion (O2.-) and hydrogen peroxide (H2O2) production was significantly higher in blood neutrophils (PMNs) of patients with lung cancer and non-malignant lung diseases when compared to the controls (p<0.001). Superoxide dismutase (SOD) activity was significantly decreased in PMNs of patients with lung cancer (p<0.001). Similarly, catalase and glutathione peroxidase (GPx) activities were lower in PMNs of lung cancer patients as compared to non-malignant lung diseases and controls. There was an increase in HMP shunt activity as measured by rate of formation of 14CO2 from [1-14C]-glucose in PMNs of lung cancer patients. Modifications in the antioxidant defense system in PMNs of malignant and nonmalignant lung diseases, the changes being more in the malignancy are indicated.  相似文献

In 132 patients suffering from various malignancies, including uterine carcinoma, breast cancer, gastric carcinoma, cancer of the colon, and additionally, patients with uterine myomas and endometriosis an increased activity of AP within the peripheral blood neutrophils could be stated by using a histochemical method. According to the author's opinion an increased activity of that enzyme reflects a response against local inflammatory processes frequently accompanying malignant tumours.  相似文献

In vitro propagated plants of the cactus Mammillaria gracillis Pfeiff. (Cactaceae) spontaneously produced callus. The habituated callus regenerated normal and hyperhydric shoots without the addition of grown regulators. Tumours were obtained by infecting cactus explants with Agrobacterium tumefaciens; the wild strain B6S3 (tumour TW) or with the rooty mutant GV3101 (tumour TR). Both tumour lines grew vigorously, never expressing any morphogenic potential. In this study, cactus shoots, callus, normal and hyperhydric regenerants and TW and TR tumours were compared with regard to peroxidase (EC 1.11.1.7) and esterase activity, and isoenzyme patterns. Guaiacol peroxidase activity was the lowest in the cactus shoots and in the normal regenerants. Callus, hyperhydric regenerants and tumours had peroxidase activity of 6 to 7 times higher. Esterase activity was measured with 1- and 2-naphthylacetate as broad-spectrum substrates. The highest esterase activity was determined in tumours with both substrates. All tissues, except the TR tumour, had higher esterase activity for 2-compared to 1-naphtylacetate. Peroxidase and esterase isoenzyme patterns were not completely identical among the investigated tissues.  相似文献

Functional activity of the bronchoalveolar lavage fluid (BALF) phagocytes was studied in 33 and 16 patients with fibro-cavernous and infiltrative pulmonary tuberculosis (FCPT and IPT, respectively). Complex examination of BALF, alveolar macrophages and neutrophils sedimented from BALF has shown interrelationship between functional activity of the cells and the form of pulmonary tuberculosis. Higher neopterin content and activity of elastase mainly secreted into BALF by activated alveolar macrophages and neutrophils, respectively, reflect higher secretory activity of both types of cells in FCPT. In FCPT this is combined with higher bactericidal activity of neutrophils, which significantly correlates with their adenosine deaminase (ADA) activity. Comparison of changes of the biochemical parameters studied in BALF (neopterin, elastase, ADA and its isoenzymes, 2-deoxy-ADA) and bactericidal activity of the sedimented cells obviously reflects different sides of BALF phagocytes functioning. Taking into consideration modern concepts on the mechanisms of regulation of phagocyte cells one may suggest the existence of differences in intercellular interactions in various forms of pulmonary tuberculosis.  相似文献

OBJECTIVE--To investigate the relation between socioeconomic deprivation and pathological prognostic factors in women with breast cancer as a possible explanation for socioeconomic differences in survival. DESIGN--Retrospective analysis of data from cancer registry and from pathology and biochemistry records. SETTING--Catchment areas of two large teaching hospitals in Glasgow. SUBJECTS--1361 women aged under 75 who had breast cancer diagnosed between 1980 and 1987. MAIN OUTCOME MEASURES--Tumour size, axillary lymph node status, histological grade, and oestrogen receptor concentration in relation to deprivation category of area of residence. RESULTS--There was no significant relation between socioeconomic deprivation and four pathological prognostic factors: 93 (32%) women in the most affluent group presented with tumours less than 20 mm in size compared with 91 (31%) women in the most deprived group; 152 (48%) of the most affluent group presented with negative nodes compared with 129 (46%) of the most deprived group; 23 (22%) of the most affluent group presented with grade I tumours compared with 12 (17%) of the most deprived group; and 142 (51%) of the most affluent group had a low oestrogen receptor concentration at presentation compared with 148 (52%) of the most deprived group. None of these differences was statistically significant. CONCLUSIONS--Differences in survival from breast cancer by socioeconomic deprivation category could not be accounted for by differences in tumour stage or biology. Other possible explanations, such as differences in treatment or in host response, should be investigated.  相似文献

The complete hematologic blood cell count of 11 patients with advanced cancer were determined on 14 occasions, every 4 hr over 36-48 hr. Oral temperature was also measured at the same times as blood sampling. Five patients had breast cancer, five had ovarian cancer and one a leiomyosarcoma. Circadian rhythms were detected by the cosinor method for seven hematologic variables (red blood cells, hemoglobin, hematocrit, white blood cells, neutrophils, lymphocytes, platelets) and for oral temperature in the whole group of patients. All acrophases were located between 1300 and 1700 hr, except for lymphocytes (2100 hr). A rhythm with a period of 8 hr was validated for lymphocytes and monocytes. Validated or estimated amplitudes and acrophases were closely similar in both breast and ovarian cancer subgroups. Individual cosinor analyses documented large differences from patient to patient, which were accounted for by differences in performance status. Thus, subjects with poor performance status and rapidly progressive disease had very few validated rhythms as compared to those patients with good performance status. As a result a statistically significant correlation was found between the number of validated hematologic circadian rhythms and the performance status (r = -0.88; P less than 0.01).  相似文献

Aromatase activity has been measured in human breast cancers by incubating tumour minces with [7-3H]testosterone and characterizing purified oestradiol (E2) fractions by chemical derivative formation. Of 247 primary tumours, 178 showed evidence of oestrogen biosynthesis, levels varying between 0.5 and 12.5 fmol E2 produced/h/g tissue. These values were quantitatively small but at least comparable with those in other peripheral tissues. There was no correlation between presence or level of aromatase activity and the histopathology of the tumours although oestrogen biosynthesis was more likely to be present in more cellular tumours. Aromatase activity was also unrelated to age, menopausal status, lymph node status and T stage of the patient from which the tumour was derived. In a subgroup of patients presenting without clinical evidence of distant metastatic disease, no significant relation was detected between tumour aromatase and disease-free interval, but tumours without aromatase activity were associated with increased survival at 36 months after primary treatment. A statistically significant correlation was also detected between the presence of tumour aromatase and oestrogen receptors. Furthermore, in small subgroups of patients with “advanced” breast cancer tumour aromatase was related to response to aminoglutethimide but not tamoxifen therapy. Whilst these results do not conclusively define a role for local synthesis of oestrogen in the progression of breast cancer, this possibility still exists and further studies on tumour aromatase are warranted.  相似文献

目的

探究食管癌患者术前肠道菌群特征与术后肺部感染的关系,为该类患者的治疗提供参考。

方法

纳入我院2019年6月至2021年12月期间收治的272例食管癌手术患者为研究对象,根据术后是否并发肺部感染将其分为感染组(64例)和非感染组(208例),记录患者年龄、高血压、吸烟史等一般资料。采用选择性培养基培养患者肠道大肠埃希菌、肠球菌属、葡萄球菌属、双歧杆菌属、酵母菌属、乳杆菌属、消化球菌属细菌并按照平板计数法计数,同时进行16S rRNA基因测序,进行菌群多样性分析。采用Logistic回归进行食管癌患者术后肺部感染的影响因素分析。

结果

感染组与非感染组患者肠道菌群Chaol指数、Shannon指数、Simpson指数比较差异均无统计学意义(均P>0.05)。与非感染组比较,感染组患者年龄≥60岁、吸烟史≥20年、术前FEV1/FVC<70%、手术时间≥4 h患者比例及肠道大肠埃希菌、肠球菌属、葡萄球菌属数量显著升高,而肠道双歧杆菌属、酵母菌属、乳杆菌属、消化球菌属数量显著降低(均P<0.05)。大肠埃希菌、肠球菌属是影响食管癌患者术后发生肺部感染的独立危险因素,而双歧杆菌属是影响食管癌患者术后发生肺部感染的保护因素(均P<0.05)。

结论

食管癌术后肺部感染患者与未发生肺部感染的其术前肠道菌群差异显著,大肠埃希菌、肠球菌属、双歧杆菌属是食管癌患者术后发生肺部感染的影响因素。

Two cases of large intestine endometriosis are presented. The disease was diagnosed during histological examination of samples taken during surgery. Clinically one case was diagnosed as Crohn's disease, while the second as cancer of the large intestine. The authors suggest, that an extent of surgery for, tumours of the large intestine should be carefully planned, specially if the tissue specimen was not examined histologically earlier.  相似文献

Total parenteral nutrition (TPN) is essential for patients with postoperative impairing gastrointestinal function who are unable to receive and absorb oral/enteral feeding for at least 7 days. Oxidative stress plays a major role in the ethiopathogenesis of cancers. In this study, total antioxidant status (TAS), glutathione peroxidase (GPx), superoxide dismutase, malondialdehyde and ascorbic acid were studied in patients operated because of small intestine, colorectal or pancreatic cancer and subsequently receiving TPN in comparison with patients receiving standard nutrition after the operation. TAS level and GPx activity were decreased in patients with small intestine cancer but did not differ in patients with colorectal and pancreatic cancer before and after surgery. In all patient groups receiving TPN, superoxide dismutase activity after the surgery was kept at the same level as before. On the fifth day after the surgery, malondialdehyde concentration in each group was restored to the value observed before surgery. On the fifth day of TPN treatment, ascorbic acid concentration was increased in every group of patients. TPN applied during the postoperative period alleviates oxidative stress resulting from surgery. In the case of small intestine cancer, the addition of vitamins and antioxidants to the nutrition mixture seems to result in depletion of antioxidant enzymes' activities.  相似文献

Activated point mutations of the K-ras gene are one of the most common genetic alterations found in human malignancies, including lung cancer, and are largely limited to adenocarcinomas. Using a highly sensitive assay for codon 12 K-ras mutation detection, called enriched PCR, we investigated 130 radically resected stage I-IIIa non-small cell lung cancer (NSCLC). There were statistically less positive results for squamous cell carcinoma (1.3%) than for adenocarcinoma (42.4%) and large cell carcinoma (27.8%). No statistically significant association between results of K-ras mutations and TNM stage of disease was found. In stage I of adenocarcinoma patients the incidence of K-ras mutations was similar to that in stage II or IIIa (40%, 42.9% and 42.9%, respectively). The results of our study showed that K-ras mutations might play an important role in pathogenesis of pulmonary adenocarcinoma due to its high prevalence in the operable tumours.  相似文献

We investigated the effect of elevated left atrial pressure and reduced cardiac output on pulmonary neutrophil kinetics in the sheep. Sheep neutrophils were isolated, labeled with 111In-oxine, and reinfused. Erythrocytes were labeled with [99mTc]pertechnetate. A gamma camera measured the lung activities of the labeled neutrophils and erythrocytes. The results indicated that 38.5% of the total injected neutrophils marginated in the lung. Pulmonary hemodynamics were altered by inflating a left atrial balloon three times in each sheep for 15-30 min to achieve 5- to 25-mmHg increments in pulmonary arterial wedge pressure. At least a 30-min recovery period was allowed between inflations. After each left atrial balloon inflation, neutrophil uptake remained unchanged from base line, despite decreased mean cardiac output to 0.67 +/- 0.24 (+/- SD) 1/min and increased pulmonary blood volume. The absence of pulmonary neutrophil uptake was confirmed by arterial-venous measurements. Increased pulmonary blood volume had little effect on lung neutrophil uptake, suggesting that most of the pulmonary neutrophils are marginated. We conclude that the lungs have a large marginated neutrophil pool compared with the circulating pool and that reduced cardiac output and elevated left atrial pressure have no effect on pulmonary neutrophil kinetics in the sheep.  相似文献
