The effect of lead on the filtration rate of the zebra musselDreissena polymorpha was investigated, together with the accumulation of Pb in the soft tissues of the mussels. The NOEC-filtration was 116 g.l–1 (0,56 mol.l–1) and the EC50-filtration was 370 g.l–1 (1.79 mol.l–1). The NOEC-accumulation was the concentration found in the control water (1.4g.l–1). These experiments show that the EC50-filtration for Pb is similar to that for Cd, higher than that for Cu and lower than that for Zn. The water quality criteria for lead allow 25 g Pb.l–1 in surface water. This will not cause short-term effects. Long-term effects may, however, occur, since an accumulation of Pb as low as 16 g.l–1 was recorded in this study.  相似文献

The effects of eight microelements (I, BO3 3–, MoO4 2–, Co2+, Cu2+, Mn2+, Fe2+, Zn2+) on the biosynthesis of camptothecin and the growth of suspension cultures of Camptotheca acuminata were studied. The increase of I to 25 M l–1, Cu2+ to 1 M l–1, Co2+ to 2 M l–1 and MoO4 2– to 10 M l–1 in Murashige and Skoog (MS) medium resulted in 1.66, 2.84, 2.53 and 2.04 times higher of camptothecin yield than that in standard MS medium respectively. Combined treatment of I (25 M l–1), Cu2+ (1 M l–1), Co2+ (2 M l–1) and MoO4 2– (10 M l–1) lead to improve cell dry weight, camptothecin content, and camptothecin yield to 30.56 g l–1, 0.0299%, and 9.15 mg l–1, respectively, which were 20.2, 208.9 and 273.8% increment respectively when compared with those of control.  相似文献

Isolated embryos ofKarwinskia humboldtiana were cultured in vitro. The growth of embryos and development to plantlets on woody plant medium supplemented with indole-3-acetic acid 6.10-2 mol l–1, gibberellic acid (GA3) 3.10-2 mol l–1, and 6-benzylaminopurine (BA) 2 mol l–1 was obtained. Multiplication of shoots and rooting of excised shoots has been achieved. Summary Stomata of paper birch (Betula papyrifera Marsh.) seedlings were more open at high humidity than at low humidity and responded rapidly to changes in vapor pressure deficit. SO2 at 0.2 or 0.8 l l-1 caused partial stomatal closure. Seedlings fumigated with SO2 at 0.2 or 0.5 l l-1 for 30 h or 0.2 l l-1 for 75 h took up more SO2 at high than at low humidity. Differences in pollutant uptake could be explained by stomatal conductance with no need to invoke changes in mesophyll conductance. Betula seedlings were more sensitive to SO2 when fumigated at high humidity, as manifested in more leaf necrosis, increased leaf abscission, and greater growth inhibition compared to seedlings fumigated at low humidity. Summary Hydantoinase (dihydropyrimidinase E.C. 3.5.2.2) activity of Pseudomonas putida DSM 84 was evaluated using cells immobilized in alginate beads and in a microporous hollow fibre bioreactor. Conversion of dihydrouracil into N-carbamyl--alanine was most efficient with alginate-immobilized cells. A 40 to 45% conversion was obtained in shake flasks and in continuous mode with packed bed columns. The highest volumetric productivity was obtained with a packed bed column operated at a dilution rate of 0.5 h-1 (99 g of product. 100 l-1 per hour). After 96 h the alginate beads began to swell and break apart; no free cells were detected however. Despite some initial loss of cells from the microporous hollow fibre bioreactor, a steady state was later established and maintained for 400 h at dilution rates of 0.1 and 0.25 h-1.  相似文献

Summary The selective formation of three tetrapyrroles, Co-containing corrinoids, Ni-containing factor F430 and Fe-containing cytochromes (haems) by Methanosarcina barkeri Fusaro (DSM 804) was achieved as a function of the concentrations of Co2+, Ni2+ and Fe2+ in a methanol minimmum medium. It was found that about 70% of the total tetrapyrroles synthesized was excreted into the culture supernatant. Hence, the continuous production of tetrapyrroles in a fixed-bed reactor (supporter: porous diatomaceous clay) was carried out at a dilution rate of 10 day-1 (850 ml medium/85 ml column/day). The effluent discharged from the reactor contained the excreted tetrapyrroles, the concentrations of which were dependent upon the Co2+, Ni2+ and Fe2+ concentrations in the feed medium. The maximum productivities from the reactor (1 l basis) were 52 M corrinoids/day, 24 M F430/day and 8 M haems/day, respectively.  相似文献

In order to evaluate the ability of the freshwater bivalve Potomida littoralis for use as a biodetector of aquatic pollution, the short-term effects of some pollutants (metals, antifouling product and effluents) on pumping activity of this mussel were studied qualitatively (pumping behaviour) and quantitatively (filtration rate). The method of the flow sensing at the exhalent siphon showed that the normal activity of P. littoralis was characterized by a continuous pattern of pumping with occasional and brief interruptions. When exposed to toxic stress, the exhalent flow immediately shifted to an intermittent pattern with periods of pumping alternating with periods of pause that could last several minutes. The threshold concentrations of heavy metals/antifouling agent inducing this type of activity within 2 h were: 25 g l–1 for copper, 90 g l–1 for cadmium, 5 g l–1 for mercury, 3000 g l–1 for zinc and 700 g l–1 for the antifouling agent Mexel. Both the effluent samples of the urban wastewater of Marrakech city tested were found to be toxic.The filtration rate of control animals ranged from 0.91 to 1.72 l h–1 g–1 animal, while after 3 h exposure to Cu, Cd, Hg and the effluent E1 and E2, filtration rates were decreased by concentrations above the threshold levels set as toxic for the pumping activity.The sensitivity of P. littoralis to pollution and its usefulness as biodetector for rapid toxicity screening is discussed and a procedure for the detection of pollution in Moroccan freshwater is proposed.  相似文献

The oxidation of catechol, an intermediate in benzene catabolism, was studied using transient variations in dissolved oxygen tension (DOT) when a succinate limited steady state culture of Pseudomonas putida ML2 was perturbed with a pulse of another substrate. A model was developed and tested for the effect of fluctuations in oxidizing enzyme activity on DOT. It was found that the rate of induction of catechol oxidizing enzymes was independent of dilution rate up to a relative growth rate /max of 0.75. Strawberry (Fragaria ananassaDuch. cv. Fengxiang) plantlets were cultured under two in vitroenvironments for rooting, and then acclimatized under two ex vitroirradiance conditions. At the end of rooting stage plant height, fresh weight and specific leaf area of T1-plants grown under high sucrose concentration (3 sucrose), low photosynthetic photon flux density (30 mol m–2 s–1) and normal CO2 concentration (350–400 l l–1) were significantly higher than those of T2-plantlets grown under low sucrose concentration (0.5), high photosynthetic photon flux density (90 mol m–2 s–1) and elevated CO2 concentration (700–800 l l–1). But T2-plantlets had higher net photosynthetic rate (Pn), effective photochemical quantum yield of PSII (PSII), effective photosynthetic electron transport rate (ETR), photochemical quenching (qP) and ratio of chlorophyll fluorescence yield decrease (Rfd). After transfer, higher irradiance obviously promoted the growth of plantlets and was beneficial for the development of photosynthetic functions during acclimatization. T2-plantlets had higher fresh weight, leaf area, PSII and ETR under higher ex vitroirradiance condition.  相似文献

Callus was initiated from immature leaf and stem segments of rose (Rosa hybrida cv. Landora) and subcultured every four weeks on a basal medium of half-strength Murashige & Skoog (1962) salts plus 30 g l-1 sucrose (1/2 MS) and supplemented with 2.2 M BA, 5.4 M NAA and 2.2–9.0 M 2,4-D. Embryogenic callus and subsequently somatic embryos were obtained from 8-week-old callus culture on 1/2 MS+2.2 M BA+0.05 M NAA+0.3 M GA3+200–800 mg l-1 L-proline. Long-term cultures were established and maintained for up to 16 months by repeated subculture of embryogenic callus on L-proline deficient medium. The effect of long-term exposure to elevated levels of CO2 on biomass partitioning, net photosynthesis and starch metabolism was examined in cotton. Plants were grown under controlled conditions at 350, 675 and 1000 l l-1 CO2. Plants grown at 675 and 1000 l l-1 had 72% and 115% more dry weight respectively than plants grown at 350 l l-1. Increases in weight were partially due to corresponding increases in leaf starch. CO2 enrichment also caused a decrease in chlorophyll concentration and a change in the chlorophyll a/b ratio. High CO2 grown plants had lower photosynthetic capacity than 350 l l-1 grown plants when measured at each CO2 concentration. Reduced photosynthetic rates were correlated with high internal (non-stomatal) resistances and higher starch levels. In calli from distal and proximal cotyledon segments and from radicles of mature wild olive zygotic embryos, rhizogenesis prevailed during the first and somatic embryogenesis during the second 30 day subculture period. Rhizogenesis was enhanced by low IBA (0.5 and 2.5 M) and 5.0 M 2iP. By reducing by half the media salt concentrations and by inserting a 21 day dark period rhizogenesis was also enhanced. Summary Rana temporaria tadpoles were raised to metamorphosis at 6 levels of pH (pH 3.6–6.5) and 2 levels of aluminium (800 and 1,600 g l-1) at pH 4.4. Treatments involved both chronic and acute exposure to depressed pH. Decreasing pH reduced maximum body size and delayed metamorphosis. Growth was depressed and metamorphosis delayed by 800 g l-1 Al, and several tadpoles died at foreleg emergence. At 1,600 g l-1 Al, small tadpoles suffered arrested growth and development and eventually died, while larger tadpoles metamorphosed without delay, though at a very small size. There was no mortality among controls. The levels of pH and inorganic monomeric aluminium measured in the experiments were similar to field levels at a site in Scotland, and it is concluded that individual tadpoles will, in certain types of water body, be adversely affected by both acidity and aluminium. The impact of these factors on populations remains to be determined.  相似文献

Dissolved, particulate and sedimentary lipid compounds were analyzed in samples collected in May 1988 at three sites in the lagoon of the closed atoll of Takapoto (Tuamotu archipelago, French Polynesia). The study provides background information dealing with water quality and the nature and concentration of lipids. Non-aromatic hydrocarbons and fatty acids were isolated from lipids and analyzed by gas chromatography/mass spectrometry. Non-aromatic hydrocarbon concentrations did not exceed 1000 ng l-1 in water, and 2300 ng g-1 in surface sediments and are among the lowest encountered in pristine marine environments. No noticeable petroleum pollution was evidenced in the lagoon. Nevertheless, traces of petroleum-derived compounds were detected at the central site for both surface and deep water. Total fatty acid concentrations varied in the range 6.3–14.4 g l-1 for the particulate phase and in the range 0.5–3.2 g l-1 for the dissolved phase. The molecular fingerprints of fatty acids and hydrocarbons evidenced a predominant algal, and to a lesser extent microbial, origin of the organic matter present in water and sediments. Mono- and polyunsaturated fatty acids, which are essential components for animal metabolism, were identified in noticeable amounts in suspended matter (1.8–4.6 g l-1), and at highly variable levels in the dissolved phase (0.08–1.21 g l-1).  相似文献

In potassium-limited chemostat cultures of Paracoccus denitrificans the maximum specific growth rate (µmax) was found to depend on the input potassium concentration: At 0.21mM µmax was 0.10–0.11 h-1; at 0.44 mM 0.15–0.16 h-1 and at 0.66 mM 0.20–0.21 h-1. The plots of the specific rates of oxygen-, succinate-and potassium consumption against gave straight lines. The intracellular potassium concentration was a linear function of and varied from 1% (0.13 M) at a value of 0.034 h-1 to 2.2% (0.29 M) at =0.26 h-1; the potassium concentration gradient and the potassium concentration in the culture fluid in the steady state were dependent on the input potassium concentration. The potassium concentration gradient varied from 8,900-1,200. At all values 20–25% of the total energy production was used for potassium transport. 350,100 and 30 ATP molecules were calculated to be required to maintain one potassium ion intracellular during 1 h at values of 0.034, 0.197 and 0.257 h-1 respectively. It is concluded that the amount of circulation of potassium is dependent on the potassium concentration gradient or on the potassium concentration in the culture in the steady state. The dependency of µmax on the input potassium concentration was explained by the assumption that at low input potassium concentrations the net uptake of potassium (influx-efflux) is not rapidly enough to maintain the high potassium gradient in the existing cells and to establish it in the newly formed cells. At high values and at high input potassium concentrations µmax is limited by the specific rate of oxygen consumption, which was found to be 11–12 mmol O2 g dry weight-1 h-1 at µmax for potassium-, succinate-and sulphate-limited chemostat cultures.  相似文献

Summary The wet and dry weights of both axenic and monoxenic cultures of the ciliate Tetrahymena were determined directly. These estimates are dependent upon the method of volume determination. Assuming a prolate spheroidal shape for the ciliate, we calculate a mean wet weight of 0.4157±0.0713 pg m-3 and a mean dry weight of 0.2793±0.0652 pg m-3. Using electronic cell sizing, our estimates are 0.7869±0.1659 pg m-3 and 0.5239±0.1101 pg m-3, respectively. Independent of the method of volume determination, we estimate a mean biomass conversion ratio (dry weight/wet weight) of 0.59±0.08.  相似文献

Shoot tips and leafy bud fragments removed from offshoots of adult date palms (Phoenix dactylifera L.) were cultured on a nutrient medium containing the Murashige and Skoog inorganic salts, 453 M 2,4-dichlorophenoxyacetic acid, 14.8 M N6-(2-isopentenyl)adenine and 3 g l-1 activated charcoal to develop nodular callus after 8 months of culture. Callus was cultured in agar-solidified and stationary or shaken liquid media containing half-strength MS inorganic salts, 3 g l-1 activated charcoal and different sucrose concentrations to study the influence of these factors on somatic embryogenesis. Summary Ultrastructural features and morphometric evaluations of buffalo Sertoli cells are reported for the six phases of the spermatogenic cycle. The phases of the tubular seminiferous epithelium are identified according to characteristic cellular associations with completed spermiation as demarcation between two cycles. Average tubular diameter (245 m) and epithelial height (61 m) do not vary significantly during the cycle. The relative Sertoli cell volume in the seminiferous epithelium varies between 30% (phase 4) and 39% (phase 8). The calculated volume of a single Sertoli cell increases from a nadir of 7118 m3 in phase 3 abruptly to a maximum of 8968 m3 in phase 4 and is then gradually reduced during the following phases. The Sertoli cell surface area shows a similar trend: it amounts to 11105 m2 in phase 3 and to 14260 m2 in phase 4. The contact area of the Sertoli cell with adjacent cells and structures is subject to characteristic changes; from the expansion of basal Sertoli-Sertoli contacts it is concluded that the blood-testis barrier in the buffalo is particularly tight during phases 8, 1 and 2. The irregularly contoured nucleus contains a vesicular nucleolus, has a calculated volume from 465 m3 to 543 m3 and occupies 5 to 7% of the cell. Volume percentages of mitochondria (4%), Golgi apparatus and lysosomal bodies are rather constant during the cycle. Whorls and orderly arranged aggregates of the smooth endoplasmic reticulum occur in basal location as well as in close association with elongating spermatids. Smooth ER is the organelle that exhibits the most prominent changes during the Sertoli cell cycle: it occupies 5.79% in phase 3 and 20.9% in phase 4 of the total cellular volume. Phagocytosis of residual bodies is insignificant in this species and a lipid cycle is absent in buffalo Sertoli cells.  相似文献

Hydrodynamic characteristics of a new mode of liquid-solid fluidization, termed as inverse fluidization in which low density floating particles are fluidized with downward flow of liquid, are experimentally investigated. The experiments are carried out with low density particles (<534 kg/m3) which allow high liquid throughputs in the system. During the operation, three regimes, namely, packed, semi-fluidization and fully fluidization are encountered. Empirical correlations are proposed to predict the pressure drop in each regime. Concentrations of heavy metals commonly known to impact the environment and other related trace elements were quantified in the water bodies of nine Ethiopian rift-valley lakes and six rivers (their inflows) as well as in effluents from two factories. In about half of the samples the concentrations of As was 10–700 g l–1 and Se, ranged from 10 to 28 g l–1, were much higher than the maximum permissible level (MPL) according to international standards for drinking water. Mercury (Hg) was detected in four lakes and one river with high values ranging from 2 to 165 g l–1. Concentrations of Mo in three soda lakes were as high as 544–2590 g l–1. Iron (Fe) was found in high concentrations (567–4969 g l–1) in three lakes, which are known to be discolored from inorganic colloids. Levels of Cd, Pb, and Cr ranged between 5 and 9, 12 and 20 and 104 and 121 g l–1, respectively. The rest of the heavy metals analyzed, Ba, Cu, Mn, Ni and Zn, were either not detected in the samples or were found in much lower concentrations than the MPL for drinking water. Effluent from a tannery contained about 15, 141, 523, and 19 g l–1 of As, Cr, Fe, and Se, respectively, and effluent from a textile factory contained high concentrations of As (10.6), Hg (3.8) and Se (20) g l–1. Compared to more industrialized regions and other African lakes the concentrations of heavy metals in Ethiopian rift-valley lakes (with the exception of the soda lakes) and their inflows were low.  相似文献
