A 14.5 kDa protein with antigenic components in common with pea leaf ferredoxin was detected on transblots of the soluble proteins of pea root plastids. The amount of this protein was found to increase during the induction of nitrate assimilation in pea roots, reaching a maximal level at 8–12 h. Concurrent with this, a fourfold increase in NADPH-dependent ferredoxin-NADP+ oxidoreductase (FNR) activity was observed corresponding to an increase in the amount of this protein detected immunologically on transblots using a leaf FNR antibody. These changes were not observed in plastids from roots of plants grown on ammonia or depleted of nitrogen. It is suggested that in addition to the already well reported induction by nitrate of nitrate reductase and nitrite reductase, there is a co-induction of a plastid located ferredoxin and FNR. Both these proteins are necessary for the transfer of reductant generated by the oxidative pentose phosphate pathway to nitrite reductase.  相似文献

Recycling of carbon in the oxidative pentose phosphate pathway (OPPP) of intact pea root plastids has been studied. The synthesis of dihydroxyacetone phosphate (DHAP) and evolution of CO2 was followed in relation to nitrite reduction. A close coupling was observed between all three measured fluxes which were linear for up to 60 min and dependent upon the integrity of the plastids. However, the quantitative relationship between 1-14CO2 evolution from glucose 6-phosphate and nitrite reduction varied with available hexose phosphate concentration. When 10 mM glucose 6-phosphate was supplied to intact plastids a stoichiometry of 1.35 was observed between 14CO2 evolution and nitrite reduction. As exogenous glucose 6-phosphate was decreased this value fell, becoming 0.47 in the presence of 0.2 mM glucose 6-phosphate, indicative of considerable recycling of carbon. This conclusion was reinforced when using [2-14C]glucose-6-phosphate. The measured release of 2-14CO2 was consistent with the data for 1-14CO2, suggesting complete recycling of carbon in the OPPP. Ribose 5-phosphate was also able to support nitrite reduction and DHAP production. A stoichiometry of 2 NO 2 ? reduced: 1 DHAP synthesised was observed at concentrations of 1 mM ribose 5-phosphate or less. At concentrations of ribose 5-phosphate greater than 1 mM this stoichiometry was lost as a result of enhanced DHAP synthesis without further increase in nitrite reduction. It is suggested that this decoupling from nitrite reduction is a function of excess substrate entering directly into the non-oxidative reactions of the OPPP, and may be useful when the demand for OPPP products is not linked to the demand for reductant. The significance of recycling in the OPPP is discussed in relation to the coordination of nitrate assimilation with carbohydrate oxidation in roots and with the utilisation of carbohydrate by other pathways within plastids.  相似文献

This study focuses on the activity of the pentose-phosphate pathway and its relationship to de novo synthesis of fatty acids and cholesterol in oligodendrocyte-enriched glial cell cultures derived from 1-week old rat brain. The proportion of glucose that was metabolized along the pentose-phosphate pathway was estimated by measuring 14CO2 production from [1-14C]-, [2-14C]- and [6-14C]glucose, the utilization of glucose and the production of lactate. Incorporation of 14C from [14C]glucose and from [3-14C]acetoacetate into lipids was analysed. The pentose- phosphate pathway produced much more CO2 from glucose than the Krebs cycle, although it accounted for only a small part of the consumption of glucose (< 3%). The higher 14CO2 production from [2-14C]glucose than from [6-14C]glucose indicated that recycling of the products of the pentose-phosphate pathway takes place in these cells.Gradual inhibition of the pathway with increasing concentrations of 6-aminonicotinamide resulted in a parallel inhibition of the conversion of acetoacetate and of glucose into fatty acids and into cholesterol. Glycolysis was also strongly inhibited in the presence of 6-aminonicotinamide whereas the activity of the Krebs cycle was not affected.These results suggest that de novo synthesis of fatty acids and cholesterol by oligodendrocytes of neonatal rats is closely geared to the activity of the pentose-phosphate pathway in these cells.  相似文献

Excised wheat (Triticum aestivum L.) and field pea (Pisum arvense L.) roots, incubated under anaerobic conditions or in the presence of uncouplers of oxidative phosphorylation [2,4-dinitrophenol (DNP), carbonylcyanide-m-chlorophenylhydrazone, pentachlorophenol] accumulated nitrite as a result of an inhibition of nitrite reduction. In isolated root plastids, nitrite reduction was dependent on a supply of glucose-6-phosphate (G6P) and did not require ATP. The estimated Km value for glucose 6-phosphate was 1.25 mM. Glucose and fructose-1,6-diphosphate were ineffective substrates for nitrate reduction. Anaerobic conditions and treatment with DNP, which would result in a cessation of ATP production by the mitochondria and a stimulation of glycolysis via the Pasteur effect, were shown to decrease the G6P content of excised roots of wheat and pea. A negative correlation was observed between the level of G6P and nitrite accumulation on root tissues. It is proposed that an interruption in the supply of G6P to the root plastid under these conditions would result in an inhibition of nitrite reduction leading to nitrite accumulation.Abbreviation G6P glucose-6-phosphate  相似文献

The aim of this work was to examine the extent to which the oxidative steps of the pentose phosphate pathway in the cytosol contribute to the provision of reductant for biosynthetic reactions. Maize (Zea mays L.) contains at least two loci (pgd1 and pgd2) that encode 6-phosphogluconate dehydrogenase. Ten genotypic combinations of wild-type (Pgd1+3.8;Pgd2+5) and null alleles of pgd1 and pgd2 were constructed in the B73 background. The maximum catalytic activity of 6-phosphogluconate dehydrogenase in the roots of seedlings of these lines correlated with the number of functional pgd1 and pgd2 alleles. Enzyme activity in the double-null homozygote (pgd1-null;pgd2-null) was 32% of that in B73 wild-type suggesting the presence of at least one other isozyme of 6-phosphogluconate dehydrogenase in maize. Subcellular fractionation studies and latency measurements confirmed that the products of pgd1 and pgd2 are responsible for the vast majority, if not all, of the cytosolic 6-phosphogluconate dehydrogenase activity in maize roots. Essentially, all of the residual activity in the double-null homozygote was confined to the plastids. Low concentrations (0.1–0.5 mM) of sodium nitrite stimulated 14CO2 production by detached root tips of both wild-type and 6-phosphogluconate dehydrogenase-deficient maize seedlings fed [U-14C]glucose. Analysis of the ratio of 14CO2 released from [1–14C]glucose relative to [6–14C]glucose (C1/C6 ratio) showed that stimulation of the oxidative pentose phosphate pathway by nitrite correlated with the dosage of wild-type alleles of pgd1 and pgd2. The failure of 6-phosphogluconate dehydrogenase-deficient lines to respond to nitrite indicates that perturbation of the cytosolic oxidative pentose phosphate pathway can influence the provision of reductant in the plastid. We conclude that the plastidic and cytosolic oxidative pentose phosphate pathways are able to co-operate in the provision of NADPH for biosynthesis.  相似文献

The mechanism of nitrate uptake for assimilation in procaryotes is not known. We used the radioactive isotope, 13N as NO3 -, to study this process in a prevalent soil bacterium, Pseudomonas fluorescens. Cultures grown on ammonium sulfate or ammonium nitrate failed to take up labeled nitrate, indicating ammonium repressed synthesis of the assimilatory enzymes. Cultures grown on nitrite or under ammonium limitation had measurable nitrate reductase activity, indicating that the assimilatory enzymes need not be induced by nitrate. In cultures with an active nitrate reductase, the form of 13N internally was ammonium and amino acids; the amino acid labeling pattern indicated that 13NO3 - was assimilated via glutamine synthetase and glutamate synthase. Cultures grown on tungstate to inactivate the reductase concentrated NO3 - at least sixfold. Chlorate had no effect on nitrate transport or assimilation, nor on reduction in cell-free extracts. Ammonium inhibited nitrate uptake in cells with and without active nitrate reductases, but had no effect on cell-free nitrate reduction, indicating the site of inhibition was nitrate transport into the cytoplasm. Nitrate assimilation in cells grown on nitrate and nitrate uptake into cells grown with tungstate on nitrite both followed Michaelis-Menten kinetics with similar K mvalues, 7 M. Both azide and cyanide inhibited nitrate assimilation. Our findings suggest that Pseudomonas fluorescens can take up nitrate via active transport and that nitrate assimilation is both inhibited and repressed by ammonium.  相似文献

Light dependency of nitrate and nitrite assimilation to reduced-N in leaves remains a controversial issue in the literature. With the objective of resolving this controversy, the light requirement for nitrate and nitrite assimilation was investigated in several plant species. Dark and light assimilation of [15N]nitrate and [15N]nitrite to ammonium and amino-N was determined with leaves of wheat, corn, soybean, sunflower, and tobacco. In dark aerobic conditions, assimilation of [15N]nitrate as a percentage of the light rate was 16 to 34% for wheat, 9 to 16% for tobacco, 26% for corn, 35 to 76% for soybean, and 55 to 63% for sunflower. In dark aerobic conditions, assimilation of [15N]nitrite as a percentage of the light rate was 11% for wheat, 7% for tobacco, 13% for corn, 28 to 36% for soybeans, and 12% for sunflower. It is concluded that variation among plant species in the light requirement for nitrate and nitrite assimilation explains some of the contradictory results in the literature, but additional explanations must be sought to fully resolve the controversy.

In dark anaerobic conditions, the assimilation of [15N]nitrate to ammonium and amino-N in leaves of wheat, corn, and soybean was 43 to 58% of the dark aerobic rate while dark anaerobic assimilation of [15N]nitrite for the same species was 31 to 41% of the dark aerobic rate. In contrast, accumulation of nitrite in leaves of the same species in the dark was 2.5-to 20-fold higher under anaerobic than aerobic conditions. Therefore, dark assimilation of nitrite cannot alone account for the absence of nitrite accumulation in the in vivo nitrate reductase assay under aerobic conditions. Oxygen apparently inhibits nitrate reduction in the dark even in leaves of plant species that exhibit a relatively high dark rate of [15N]nitrite assimilation.

Summary We did this work to see if there is a correlation between lignin synthesis and the activity of the pentose phosphate pathway. Excision of the third internode of the stem of Coleus blumei Benth. followed by incubation on sucrose and indoleacetic acid led to extensive formation of tracheids. During this lignification we determined the activities of glucose-6-phosphate dehydrogenase and fructose-1,6-diphosphate aldolase, and the extent to which [1-14C]-,[3,4-14C]-, and [6-14C]glucose labelled CO2 and the major cellular components. The results indicate that the pentose phosphate pathway was active during lignification, and that the activity of this pathway relative to glycolysis increased at the onset of lignification. Explants of storage tissue of Helianthus tuberosus L. were cultured under conditions which caused extensive lignification. 14CO2 production from [1-14C]-, [3,4-14C]-, and [6-14C]glucose indicated activity of the pentose phosphate pathway during tracheid formation. The intracellular distribution of the enzymes of nitrate and ammonia assimilation in apical cells of pea (Pisum sativum L.) roots is described. Nitrate reductase (EC 1.6.6.2) was found to have no organelle association, and is considered to be located in the cytosol or possibly loosely bound to the outside of an organelle. Nitrite reductase and glutamate synthase (EC 2.6.1.53) are plastid located, as is glutamine synthetase (EC 6.3.1.2) although this enzyme also has activity in the cytosol. Glutamate dehydrogenase (EC 1.4.1.3) was found only in the mitochondrion.  相似文献

Chloroplasts or chromoplasts were purified from sweet-pepper (Capsicum annuum L. cv. Yolo Wonder) fruits and analysed with respect to their enzymic equipment, the transport properties across the envelope membrane, and for the presence of a functional oxidative pentose-phosphate pathway (OPPP). It was demonstrated that both types of plastid contain enzyme activities that allow glycolysis and OPPP. During the developmental conversion from chloroplasts to chromoplasts the activities of enzymes catalysing potentially rate-limiting reactions in glycolysis increased considerably. Most enzyme activities involved in the plastidic OPPP stayed constant or decreased during ripening, but transaldolase activity increased by more than 500%. To analyse whether pepper fruit chromoplasts are able to use exogenously supplied carbohydrates for the OPPP we measured the rate of 14CO2 release after application of radioactively labelled precursors. Isolated pepper fruit chromoplasts used exogenously supplied [U14C]glucose- 6-phosphate (Glc6P) as a precursor for the OPPP. The metabolic flux through this pathway was stimulated by the presence of additional compounds which require reducing equivalents for further conversion, e.g. nitrite, or 2-oxoglutarate plus glutamine. The [14C]Glc6P-driven OPPP in isolated chromoplasts exhibited saturation with rising concentrations of Glc6P, reaching highest rates at an external concentration of about 2 mM. Exogenously given [U14C]glucose 1-phosphate (Glc1P)′ did not lead to a release of 14CO2, indicating that this hexose phosphate is not taken up into the intact plastid. Using a proteoliposome system in which the envelope membrane proteins from sweet-pepper chromoplasts were functionally reconstituted we demonstrated that Glc6P is transported in counter-exchange with inorganic phosphate (Pi) or other phosphorylated intermediates. The Glc6P was taken up into proteoliposomes with an apparent K m of 0.34 mM. Surprisingly, in contrast to tomato fruit plastids, isolated chromoplasts from sweet-pepper fruits do not possess a phosphate translocator allowing the uptake of Glc1P. Rising exogenous concentrations of dihydroxyacetone phosphate strongly inhibited the metabolic flux through the OPPP. This observation is discussed with respect to the presence of two phosphate translocator proteins in the envelope of sweet-pepper chromoplasts and with respect to possible metabolic changes occurring in heterotrophic tissues during development. Maize (Zea mays L.) and pearl millet (Pennisetum americanum (L.) Leeke) seedlings were exposed to [15N]nitrate for 1-h periods at eight times during a 24-h period (16–8 h light-dark for maize; 14–10 h for millet). Influx of [15N]nitrate as well as its reduction and translocation were determined during each period. The efflux of previously absorbed [14N]nitrate to the uptake solution was also estimated. No marked diurnal changes in [14N]nitrate efflux or [15N]nitrate influx were evident in maize. In contrast, [14N]nitrate efflux from millet increased and eventually exceeded [15N]nitrate influx during the late dark and early light periods, resulting in net nitrate efflux from the roots. The dissimilarity of their diurnal patterns indicates that influx and efflux are independently regulated. In both species, [15N]nitrate reduction and 15N translocation to shoots were curtailed more by darkness than was [15N]nitrate influx. In the light, maize reduced 15% and millet 24% of the incoming [15N]nitrate. In darkness, reduction dropped to 11 and 17%, respectively. Since the accumulation of reduced-15N in shoots declined abruptly in darkness, whereas that in roots was little affected, it is suggested that in darkness [15N]nitrate reduction occurred primarily in roots. The major fluxes of carbohydrate metabolism were estimated during starch breakdown by ripening bananas (Musa cavendishii Lamb ex Paxton). Hands of bananas, untreated with ethylene, were allowed to ripen in the dark at 21° C. Production of CO2 and the contents of starch, sucrose, glucose and fructose of intact fruit were determined for a period of 10 d that included the climacteric. The detailed distribution of label was determined after supplying the following to cores of pulp from climacteric fruit: [U-14C]-, [1-14C]-, [3,4-14C]-and [6-14C]glucose, [U-14C]glycerol, 14CO2. The data obtained were used to estimate the following fluxes, values given as mol hexose · (g FW)–1 · h–1 in parenthesis: starch to hexose monophosphates (5.9) and vice versa (0.4); hexose monophosphates to sucrose (7.7); sucrose to hexose (4.7); hexose to hexose monophosphate (3.8); glycolysis (0.5–1.6); triose phosphate to hexose monophosphates (0.14); oxidative pentose-phosphate pathway (0.48); CO2 fixation in the dark (0.005). Recently, we demonstrated that intact chloroplasts isolated from green pepper (Capsicum annum L.) fruits use exogenous glucose-6-phosphate (Glc-6-P) as the most efficient precursor for starch biosynthesis (O. Batz, R. Scheibe, H.E. Neuhaus [1995] Planta 196: 50-57). Here we demonstrate that these chloroplasts transport this hexose phosphate in counter-exchange for orthophosphate. By measuring the release of 14CO2 from [1-14C]Glc-6-P, we show that isolated fruit chloroplasts also use exogenous Glc-6-P as a substrate for the oxidative pentose-phosphate pathway. The rate of decarboxylation appears to be linear with time and is significantly reduced in the presence of Triton X-100, indicating that the reaction is dependent on plastid integrity. Pyruvate has been identified as a positive effector for flux through the oxidative pentose-phosphate pathway. However, the highest rates of Glc-6-P-driven oxidative pentosephosphate pathway activity are achieved in the presence of nitrite, 2-oxoglutarate, and glutamine, indicating a strong interaction between nitrogen metabolism and this pathway. In addition, we show that carbohydrates liberated during starch mobilization are used as substrates for the oxidative pentose-phosphate pathway. Orthophosphate was found to act as an activator for the observed 14CO2 release from carbohydrates formerly bound as starch. In this context, we demonstrate that exogenous Glc-6-P competes with endogenous carbohydrates. A possible interaction between exogenous and endogenous carbohydrates is discussed with respect to altered levels of carbohydrates during fruit development.  相似文献

In roots, nitrate assimilation is dependent upon a supply of reductant that is initially generated by oxidative metabolism including the pentose phosphate pathway (OPPP). The uptake of nitrite into the plastids and its subsequent reduction by nitrite reductase (NiR) and glutamate synthase (GOGAT) are potentially important control points that may affect nitrate assimilation. To support the operation of the OPPP there is a need for glucose 6-phosphate (Glc6P) to be imported into the plastids by the glucose phosphate translocator (GPT). Competitive inhibitors of Glc6P uptake had little impact on the rate of Glc6P-dependent nitrite reduction. Nitrite uptake into plastids, using (13)N labelled nitrite, was shown to be by passive diffusion. Flux through the OPPP during nitrite reduction and glutamate synthesis in purified plastids was followed by monitoring the release of (14)CO(2) from [1-(14)C]-Glc6P. The results suggest that the flux through the OPPP is maximal when NiR operates at maximal capacity and could not respond further to the increased demand for reductant caused by the concurrent operation of NiR and GOGAT. Simultaneous nitrite reduction and glutamate synthesis resulted in decreased rates of both enzymatic reactions. The enzyme activity of glucose 6-phosphate dehydrogenase (G6PDH), the enzyme supporting the first step of the OPPP, was induced by external nitrate supply. The maximum catalytic activity of G6PDH was determined to be more than sufficient to support the reductant requirements of both NiR and GOGAT. These data are discussed in terms of competition between NiR and GOGAT for the provision of reductant generated by the OPPP.  相似文献

15N kinetic labeling studies were performed on seedlings of Hordeum vulgare L. var. Golden Promise growing under steady state conditions. Patterns of label incorporation in the pools of nitrogen compounds of roots fed [15N]ammonium were compared with computer-simulated labeling curves. The data were found to be quantitatively consistent with a three-compartment model in which ammonium is assimilated solely into the amide-N of glutamine. Labeling data from roots fed [15N]nitrate were also found to be at least qualitatively consistent with the assimilation of ammonia into glutamine. Methionine sulfoximine almost completely blocked the incorporation of 15N label into the amino acid pools of barley roots fed [15N]nitrate. These observations suggest that ammonia assimilation occurs solely via the glutamine synthetase/glutamate synthase pathway in both nitrate- and ammonia-grown barley roots.  相似文献

The aim of this work was to establish the pathways of carbohydrate oxidation used in the dark by leaves of Pisum sativum and Triticum aestivum. Segments of young and mature leaves of pea released the carbons of glucose-[14C] as 14CO2 in the order 3,4 > 1 > 2 > 6 whereas in segments of young and mature leaves of wheat the order was 3,4 > 1 > 6 > 2. The detailed labelling of the constituents of mature leaves of wheat by glucose-[1-14C], -[2-14C], -[3,4-14C], and -[6-14C] was determined and showed that the high yield of CO2 from C-6 relative to that from C-2 was due to release of C-6 during pentan synthesis. Estimates were made of the maximum catalytic activities of phosphofructokinase and glucose-6-phosphate dehydrogenase in pea and wheat leaves of three ages. The results of all the above investigations strongly indicate that both pea and wheat leaves in the dark oxidize carbohydrate via glycolysis and the pentose phosphate pathway with the latter accounting for no more than a third of the total. No evidence was obtained of any major change in the relative activities of the two pathways during the development of either type of leaf.  相似文献

Sugar metabolism and the oxidative pentose phosphate pathway (OPPP) are strongly implicated in N assimilation, although the relationship between them and the roles of the plastidial and cytosolic OPPP have not been established genetically. We studied a knock‐down mutant of the plastid‐localized OPPP enzyme 6‐phosphogluconolactonase 3 (PGL3). pgl3‐1 plants exhibited relatively greater resource allocation to roots but were smaller than the wild type. They had a lower content of amino acids and free in leaves than the wild type, despite exhibiting comparable photosynthetic rates and efficiency, and normal levels of many other primary metabolites. When N‐deprived plants were fed via the roots with , pgl3‐1 exhibited normal induction of OPPP and nitrate assimilation genes in roots, and amino acids in roots and shoots were labeled with 15N at least as rapidly as in the wild type. However, when N‐replete plants were fed via the roots with sucrose, expression of specific OPPP and N assimilation genes in roots increased in the wild type but not in pgl3‐1. Thus, sugar‐dependent expression of N assimilation genes requires OPPP activity and the specificity of the effect of the pgl3‐1 mutation on N assimilation genes establishes that it is not the result of general energy deficiency or accumulation of toxic intermediates. We conclude that expression of specific nitrate assimilation genes in the nucleus of root cells is positively regulated by a signal emanating from OPPP activity in the plastid.  相似文献

The aim of this work was to investigate whether the pentose phosphate pathway provides reducing power for lignin synthesis. Explants of the stem of Coleus blumei and the storage tissue of Helianthus tuberosus were cultured for 4 days on media which caused extensive lignification. [3-3H]-glucose and either [3-14C]- or [U-14C]-glucose were supplied to such 4-day-cultured explants, and also to the roots of 5-day-old seedlings of Pisum sativum. Significant amounts of 3H and 14C were recovered in syringaldehyde, vanillin, p-hydroxybenzaldehyde, and ligothio-glycollic acid from the explants of Coleus and Helianthus; and in vanillin, p-hydroxybenzaldehyde, and milled-wood lignin from pea roots. The 3H/14C ratios in these derivatives and preparations of lignin are held to indicate that much of the reducing power for lignin synthesis comes from the pentose phosphate pathway.  相似文献
