Jet aerated loop reactors (JLRs) provide high mass transfer coefficients (kLa) and can be used for the intensification of mass transfer limited reactions. The jet loop reactor achieves higher kLa values than a stirred tank reactor (STR). The improvement relies on significantly higher local power inputs (~104) than those obtainable with the STR. Operation at high local turnover rates requires efficient macromixing, otherwise reactor inhomogeneities might occur. If sufficient homogenization is not achieved, the selectivity of the reaction and the respective yields are decreased. Therefore, the balance between mixing and mass transfer in jet loop reactors is a critical design aspect. Monitoring the dissolved oxygen levels during the turnover of a steady sodium sulfite feed implied the abundance of gradients in the JLR. Prolonged mixing times at identical power input and aeration rates (~100%) were identified for the JLR in comparison to the STR. The insertion of a draft tube to the JLR led to a more homogenous dissolved oxygen distribution, but unfortunately a reduction of mixing time was not achieved. In case of increased medium viscosities as they may arise in high cell density cultivations, no gradient formation was detected. However, differences in medium viscosity significantly altered the mass transfer and mixing performance of the JLR.  相似文献

The major parcel of the degradation occurring along wastewater biotreatments is performed either by the native microbiota or by added microbial inocula. The main aim of this study was to apply two fingerprinting methods, temperature gradient gel electrophoresis (TGGE) and length heterogeneity-PCR (LH-PCR) analysis of 16S rRNA gene fragments, in order to assess the microbiota structure and dynamics during mixed olive oil and winery wastewaters aerobic biotreatment performed in a jet-loop reactor (JLR). Sequence homology analysis showed the presence of bacterial genera Gluconacetobacter, Klebsiella, Lactobacillus, Novosphingobium, Pseudomonas, Prevotella, Ralstonia, Sphingobium and Sphingomonas affiliated with five main phylogenetic groups: alpha-, beta- and gamma-Proteobacteria, Firmicutes and Bacteroidetes. LH-PCR analysis distinguished eight predominant DNA fragments correlated with the samples showing highest performance (COD removal rates of 67 up to 75%). Cluster analysis of both TGGE and LH-PCR fingerprinting profiles established five main clusters, with similarity coefficients higher than 79% (TGGE) and 62% (LH-PCR), and related with hydraulic retention time, indicating that this was the main factor responsible for the shifts in the microbiota structure. Canonical correspondence analysis revealed that changes observed on temperature and O2 level were also responsible for shifts in microbiota composition. Community level metabolic profile analysis was used to test metabolic activities in samples. Integrated data revealed that the microbiota structure corresponds to bacterial groups with high degradative potential and good suitability for this type of effluents biotreatments.  相似文献

An ecological study of Saccharomyces cerevisiae strains in spontaneous alcoholic fermentation has been made in the same winery on two consecutive years (1993 and 1994) with Merlot type musts, and with Malbec type must on a third year (1998). Saccharomyces cerevisiae strains associated with winery surfaces were also analysed. Differential killer sensitivity patterns related to a killer reference panel of 10 killer yeasts belonging to nine species of four genera were used as a quick and simple procedure to discriminate between indigenous S. cerevisiae isolates at the strain level. Although a great diversity of wild strains was observed, two main indigenous S. cerevisiae strains, designated as S. cerevisiae 9 and S. cerevisiae 13, took over the Merlot type fermentation in both years. These strains also appeared in Malbec must fermentation during the year 1998 and they were again found on the winery surface the next year. These results show that some few and stable indigenous S. cerevisiae strains remained in the environmental winery over the considered period of time (1993–1999) and they represent an additional evidence of the taking over of musts by local strains of S. cerevisiae.  相似文献

The impact of mass transfer on productivity can become a crucial aspect in the fermentative production of bulk chemicals. For highly aerobic bioprocesses the oxygen transfer rate (OTR) and productivity are coupled. The achievable space time yields can often be correlated to the mass transfer performance of the respective bioreactor. The oxygen mass transfer capability of a jet aerated loop reactor is discussed in terms of the volumetric oxygen mass transfer coefficient kLa [h?1] and the energetic oxygen transfer efficiency E [kgO2 kW?1 h?1]. The jet aerated loop reactor (JLR) is compared to the frequently deployed aerated stirred tank reactor. In jet aerated reactors high local power densities in the mixing zone allow higher mass transfer rates, compared to aerated stirred tank reactors. When both reactors are operated at identical volumetric power input and aeration rates, local kLa values up to 1.5 times higher are possible with the JLR. High dispersion efficiencies in the JLR can be maintained even if the nozzle is supplied with pressurized gas. For increased oxygen demands (above 120 mmol L?1 h?1) improved energetic oxygen transfer efficiencies of up to 100 % were found for a JLR compared to an aerated stirred tank reactor operating with Rushton turbines.  相似文献

Vino Santo is a sweet wine produced from late harvesting and pressing of Nosiola grapes in a small, well‐defined geographical area in the Italian Alps. We used metagenomics to characterize the dynamics of microbial communities in the products of three wineries, resulting from spontaneous fermentation with almost the same timing and procedure. Comparing fermentation dynamics and grape microbial composition, we show a rapid increase in a small number of wine yeast species, with a parallel decrease in complexity. Despite the application of similar protocols, slight changes in the procedures led to significant differences in the microbiota in the three cases of fermentation: (i) fungal content of the must varied significantly in the different wineries, (ii) Pichia membranifaciens persisted in only one of the wineries, (iii) one fermentation was characterized by the balanced presence of Saccharomyces cerevisiae and Hanseniaspora osmophila during the later phases. We suggest the existence of a highly winery‐specific ‘microbial‐terroir’ contributing significantly to the final product rather than a regional ‘terroir’. Analysis of changes in abundance during fermentation showed evident correlations between different species, suggesting that fermentation is the result of a continuum of interaction between different species and physical–chemical parameters.  相似文献

常年使用化肥和除草剂以及农业新技术的高投入,使我国粮食主产区耕地出现了生产力降低、土壤生物多样性失调和污染严重等生态问题。采用磷脂脂肪酸(PLFA)方法来评估施肥和杂草多样性对冬小麦土壤微生物群落结构的影响。实验采用裂区实验设计,施肥作为主因素,杂草多样性作为次因素。化肥和有机肥两个施肥处理,在两个施肥处理中进行杂草多样性设置,实验盆中心种植作物(冬小麦8株),四周种植杂草(8株),杂草种类选择野燕麦、苜蓿、菊苣、播娘蒿。杂草多样性处理设为0、1、2、4种杂草处理,0种杂草处理仅种植作物,有6盆;1种杂草处理为每盆种1种杂草,有12盆;2种杂草处理为每盆种两种杂草,有12盆;4种杂草处理为每盆种4种杂草,有6盆。结果表明:在两种施肥处理中,增加杂草多样性显著增加了土壤碳氮比和p H值,碳氮比都是在4种杂草处理中最高。施化肥处理中,增加杂草多样性显著影响真菌和细菌比,真菌和细菌比在4种杂草处理中最大,显著高于0、1、2种杂草处理。在施有机肥处理中,增加杂草多样性显著影响阳性菌和阴性菌比,阳性菌和阴性菌比在0种杂草处理中最低,显著低于1、2、4种杂草处理。在两个施肥处理中,土壤碳氮比与各类群微生物量显著相关,杂草多样性通过改变土壤碳氮比改变微生物群落构成,并且微生物群落结构转变方式不同。  相似文献

During the transformation of grapes to wine, wine fermentations are exposed to a large area of specialized equipment surfaces within wineries, which may serve as important reservoirs for two-way transfer of microbes between fermentations. However, the role of winery environments in shaping the microbiota of wine fermentations and vectoring wine spoilage organisms is poorly understood at the systems level. Microbial communities inhabiting all major equipment and surfaces in a pilot-scale winery were surveyed over the course of a single harvest to track the appearance of equipment microbiota before, during, and after grape harvest. Results demonstrate that under normal cleaning conditions winery surfaces harbor seasonally fluctuating populations of bacteria and fungi. Surface microbial communities were dependent on the production context at each site, shaped by technological practices, processing stage, and season. During harvest, grape- and fermentation-associated organisms populated most winery surfaces, acting as potential reservoirs for microbial transfer between fermentations. These surfaces harbored large populations of Saccharomyces cerevisiae and other yeasts prior to harvest, potentially serving as an important vector of these yeasts in wine fermentations. However, the majority of the surface communities before and after harvest comprised organisms with no known link to wine fermentations and a near-absence of spoilage-related organisms, suggesting that winery surfaces do not overtly vector wine spoilage microbes under normal operating conditions.  相似文献

Diversity of lactic acid bacteria (LAB) species has been analyzed for three consecutive years (2006, 2007, and 2008) during alcoholic and malolactic fermentations of Tempranillo wine in a winery at La Rioja. The results showed differences in malolactic fermentation duration, and in both diversity of LAB species and diversity of Oenococcus oeni genotypes. O. oeni was shown to be the predominant species (73% of total isolates). Monitoring the different strains of O. oeni using pulsed-field gel electrophoresis of chromosomal DNA digested with SfiI and ApaI allowed detection of a total of 37 distinct genotypes, most of them comprised at least two isolates. Six appeared in more than one vintage, one of them being present in the three studied years. Moreover, four genotypes were indistinct of the strains isolated from the air of this same winery in 2007 vintage. The frequency of participation of each genotype varied from year to year, thus dominant genotypes at one year were minority or not present at another year. This suggests that distinct indigenous O. oeni strains are better adapted to the different winery conditions every year. Predominant genotypes that appeared in more than one vintage and lead to quality wines with low histamine contents could be considered as interesting for selecting of new malolactic starter cultures.  相似文献

土壤微生物是反映土壤质量状况的重要指标,研究侵蚀地植被恢复后土壤微生物群落结构的变化对深入认识土壤质量的演变具有重要意义。对比分析了未治理地(Y0)、治理13年(Y13)和31年(Y31)的马尾松林(Pinus massoniana)林下芒萁(Dicranopteris dichotoma)覆盖地(NRd)、去除芒萁覆盖地(Rd)与林下裸地(CK)土壤微生物生物量和群落结构差异,结果表明:林下裸地土壤微生物生物量碳(MBC)、微生物生物量氮(MBN)和总微生物磷脂脂肪酸量(总PLFAs)的含量均显著低于芒萁覆盖地,且去除芒萁4个月后,MBC和总PLFAs均有降低趋势,表明芒萁覆盖对土壤微生物生物量具有重要影响;林下芒萁覆盖地土壤革兰氏阳性菌(GP)、革兰氏阴性菌(GN)、丛植菌根真菌(VAM)、真菌(Fungi)、放线菌(ACT)的PLFAs含量显著高于林下裸地(Y13例外),去除芒萁4个月后,各值均有有接近林下裸地的趋势;芒萁覆盖地真菌/细菌的比值(F/B)均显著高于林下裸地(P0.05),芒萁覆盖地革兰氏阳性菌/革兰氏阴性菌的比值(GP/GN)、饱和直链脂肪酸/单不饱和脂肪酸的比值(sat/mono)和(cy17:0+cy19:0ω8c)/(16:1ω7c+18:1ω7c)(cy/pre)显著小于林下裸地(P0.05),去除芒萁4个月后,芒萁覆盖地土壤cy/pre显著升高(P0.05)(Y13例外),意味着芒萁覆盖地土壤生态系统更稳定,土壤的养分可利用性更高,微生物生物量和群落结构更丰富,活性更强;皮尔逊相关分析和冗余分析发现,土壤理化性质与土壤微生物生物量和群落结构关系密切,土壤C/N、p H和氮素水平是调控芒萁覆盖下土壤微生物生物量和群落结构的主要生态因子。  相似文献

土壤微生物是生态系统维持正常结构与功能的重要组成部分,为探究盐城滩涂典型湿地土壤微生物群落结构特征,以江苏盐城滩涂互花米草、藨草、盐地碱蓬、芦苇及淤泥质光滩5种典型群落为对象,采用16S rRNA高通量测序技术分析0—10 cm(表层)、10—30 cm(中层)、30—60 cm(深层)土壤微生物多样性及群落结构。结果表明:(1)几种植物群落间,土壤微生物群落结构差异较大,主要体现在细菌群落结构的差异性,古菌群落结构差异相对较小。光滩与植物群落间,在土壤细菌种类及相对丰度上差异相对较大,互花米草群落与本土植物群落间,在微生物群落的细菌种类组成上存在较大差异;藨草群落土壤表层微生物群落结构与互花米草群落相似,深层与盐地碱蓬、芦苇群落相似。(2)同一群落不同层次土壤微生物群落结构相似,差异小于不同群落间土壤微生物群落的结构差异性;不同群落对应层次间,表深层土壤中五种群落土壤微生物多样性存在显著差异,中层土壤中五种群落微生物多样性差异不显著。总体上,植物群落类型对土壤微生物群落结构的影响大于土壤深度;与本土植物群落相比,互花米草群落土壤主要优势门微生物种类差异较小,但部分优势门微生物相对丰度...  相似文献

The present study has been carried out in an organic winery established in 2003 in the Denomination of Origin “Sierras de Málaga” (Southern Spain) region during the 2007 vintage. The aim of this work was to ascertain the yeast microflora present in the winery and during the vinifications and to obtain a collection of autochthonous S. cerevisiae strains from this area. Yeast populations from three vats containing fermenting musts from different grape varieties were analysed. Two of them were inoculated with a natural “pied de cuve” while the third one was sown with a rehydrated commercial yeast strain. A total of 382 yeasts were isolated and identified, initially by restriction analysis of ribosomal DNA and further by sequencing of this region. Non-Saccharomyces yeasts were found in all three musts but they practically disappeared as the fermentations progressed. Analysis of mitochondrial DNA RFLP revealed 13 different restriction patterns of Saccharomyces cerevisiae strains, five of them similar to those of commercial strains used in the winery. Commercial strains were found even in vats inoculated with a “pied de cuve” generated by spontaneous fermentation of a must sample. The analysis of samples recovered from different winery surfaces and equipments demonstrated that non-Saccharomyces and both commercial and autochthonous Saccharomyces strains were part of the resident microflora in the winery. Biodiversity of autochthonous S. cerevisiae in fermentation vats was low but two of them were able to compete with the commercial ones and they were isolated even at the end of the fermentation.  相似文献

Aims: The aim of the study was to investigate the feasibility of a continuous reactor for psychrophilic anaerobic wastewater treatment by using the sludge from cold natural environment. Methods and Results: Six sludge samples (S1–S6) were collected from different cold natural locations to select sludge with high anaerobic microbial activity under low temperatures. After a 225‐day incubation, the maximum specific methane production rate of a waterfowl lake sediment (S1) at 15°C (70·5 mLCH4 gVSS?1 day?1) was much higher than all other samples. S1 was thus chosen as the seed sludge for the reactor treating synthetic brewery wastewater at 15°C, by immobilizing the micro‐organisms on polyurethane foam carriers. The chemical oxygen demand (COD) removal efficiency reached over 80% after 240‐day operation at an organic loading rate of 5·3 kg m?3 day?1, and significant enrichment of biomass was observed. Clone libraries of the microbial communities in the inoculum had high diversities for both archaea and bacteria. Along with a decrease in microbial community diversities, the dominant bacteria (79·5%) at the end of the operation represented the phylum Firmicutes, while the dominant archaeon (41·5%) showed a similarity of 98% with the psychrotolerant methanogen Methanosarcina lacustris. Conclusions: The possibility of using anaerobic micro‐organisms from cold environments in anaerobic wastewater treatment under psychrophilic conditions is supported by these findings. 树种选择是林下山参护育成败的关键,研究树叶凋落物对人参土壤养分、微生物群落结构组成的影响,旨在为林下山参护育选择适宜林地及农田栽参土壤改良提供科学依据和理论指导。通过盆栽试验,研究添加5.0 g色木槭Acer mono.Maxim.var.mono(A)、赤松Pinus densiflora Sieb.et Zucc.(B)、胡桃楸Juglans mandshurica Maxim.(C)、紫椴Tilia amurensis Rupr.(D)、蒙古栎Quercus mongolica Fisch.ex Ledeb.(E)树叶凋落物到土壤中,种植人参(Panax ginseng C.A.meyer)后研究土壤理化性质以及微生物群落结构的变化。结果表明:添加不同树叶处理后人参土壤性质发生改变,土壤p H值显著高于对照土壤5.91(P0.05),土壤全氮、速效氮磷、微生物碳氮在所有树叶处理中显著增加(P0.05),而土壤容重、速效钾和C/N在添加树叶处理中降低。18个土壤样品基因组,经16S和ITS1测序分别得到6064和1900个OUTs。其中细菌涵盖了42门、117纲、170目、213科、225属,真菌涵盖了24门、98纲、196目、330科、435属。不同树叶处理人参土壤细菌和真菌地位发生改变,细菌Proteobacteria是树叶分解的关键微生物,添加树叶后其多样性显著高于对照(P0.05)。而细菌Bacteroidetes和真菌Basidiomycota可能是区别阔叶林和针叶林树种的关键微生物,针叶林中含量显著低于阔叶林(P0.05),而真菌Ascomycota是针叶林分解的关键微生物。进一步从不同分类水平上得到特定树叶凋落物的特异细菌和真菌。典型相关分析(CDA)表明细菌Bacteroidetes、Chloroflexi、Actinobacteria及真菌Basidiomycota、Zygomycota、Chytridiomycota及Ascomycota的位置及多样性的改变均与土壤因子SMBN、TN、AP、SOC、AK、C/N、p H有关。综上所述,添加不同树叶后不仅提高土壤微生物量碳氮、改善土壤理化性质,同时改变微生物群落结构组成,不同树叶处理土壤理化性质不同导致人参土壤微生物组成的差异,本结果对于林下参选地和农田栽参土壤微生物改良具有理论指导作用。  相似文献

Aims: This study aimed to characterize microbial community dynamics in aerated cow manure slurry at different aeration intensities. Methods and Results: Batch aerobic treatments were set up in 5‐l jar fermentor, each containing 3 l of manure slurry; the slurries were subjected to low, medium and high (50, 150 and 250 ml min?1, respectively) aeration for 9 days. Microbial community composition was determined using terminal restriction fragment length polymorphism and a clone library targeting 16S rRNA genes. High and medium aeration accelerated organic carbon degradation in parallel with the degree of aeration intensity; however, 90% of the initial total organic carbon was retained during low‐aeration treatment. During the active stages of organic carbon decomposition, clones belonging to the class Bacilli accumulated. Moreover, Bacilli accumulation occurred earlier under high aeration than under medium aeration. Conclusions: Organic matter degradation was mainly governed by a common microbial assemblage consisting of many lineages belonging to the class Bacilli. The timing of community development differed depending on aeration intensity. Significance and Impact of the Study: This study reports on changes in several environmentally important parameters and the principal microbial assemblage during the pollution‐reducing phase of cattle manure aeration treatment.  相似文献

微生物在湿地的生物地球化学循环和生态功能调节中发挥着重要作用,对全球气候变化具有重大影响,对维持全球生态系统的健康至关重要。以黄河三角洲滨海湿地为研究对象,通过采集代表性植被群落的土壤表层和部分植物根系,探究土壤微生物群落组成、根际微生物、环境因子及其内在的关联性和影响机制。研究结果表明不同植被覆盖地区微生物多样性存在差异,芦苇区和柽柳区微生物丰度高于泥滩区、碱蓬区和棉田,海漫滩微生物丰度高于河漫滩地和泥滩。土壤微生物菌群结构和多样性显著高于根际:土壤细菌的香农指数约为4-5.5,根际微生物的香农指数约为0-4。土壤细菌主要为厚壁菌门、变形菌门、拟杆菌门和放线菌门,占样品总数的90%以上;而根际细菌主要是蓝藻门、变形菌门和放线菌门,二者在属水平上的菌群结构差异更加明显。环境因子的含量与生境类型有关,SO42-和NO3-的相关性最高,植被覆盖区土壤中Mn4+、Fe3+和水解氮的含量低于滩涂裸地。冗余分析(RDA)表明,pH值在小空间尺度上对湿地土壤中细菌群落的影响较小,环境因子在门和属水平的解释率分别为89.7%和86.8%,其中K(23.4%)、NO2-(11.8%)、Mn4+(9.8%)和Na(8.0%)是解释门水平微生物区系结构变化和组成的主要因子。研究为理解湿地微生物多样性与湿地生态系统功能之间的影响机制提供了一个生态学视角,有助于了解黄河三角洲滨海湿地土壤和植物根际的细菌分布特征,对黄河三角洲退化滨海湿地的生物修复具有重要的指导意义。  相似文献

【目的】本试验研究百里酚和香芹酚对滩羊羔羊腹泻、肠道短链脂肪酸和微生物组成的影响,旨在为利用植物提取物手段治疗羔羊腹泻提供可用的添加剂和理论基础。【方法】本研究选择腹泻发病时间为2-3 d的10只腹泻羔羊,随机分为2组,分为腹泻羔羊组(CON组)和百里酚和香芹酚处理组(SCAT组),试验期间SCAT组羔羊补充200 mg/只/d的百里酚和香芹酚1:1混合品,处理时间为7 d。【结果】百里酚和香芹酚处理能够显著提高腹泻羔羊日增重,降低羔羊腹泻指数和腹泻率(P<0.05),降低血清促炎因子表达水平(P<0.05),提高肠道短链脂肪酸(short-chain fatty acids,SCFA)丁酸浓度(P<0.05);同时百里酚和香芹酚能够改变腹泻羔羊肠道微生物组成,提高肠道中双歧杆菌(Bifidobacteriaceae)和阿克曼氏菌(Akkermansiaceae)的相对丰度,降低肠杆菌科(Enterobacteriaceae)相关菌的相对丰度。【结论】百里酚和香芹酚能够提高肠道有益菌相对丰度和有益代谢物丁酸的浓度,降低致病菌相对丰度,从而有效抑制羔羊腹泻,可作为新型替...  相似文献

【目的】转Bt基因和Bar基因植物的微生态效应是环境安全评价的重要因素,但关于Bt基因和Bar基因转化引起的水稻基因型改变对水稻不同组织生态位微生物群落组成和潜在功能的影响还无系统研究。【方法】以转Bt基因和Bar基因水稻T1C-1及其亲本对照Minghui63为研究对象,基于细菌16S rRNA基因和真菌ITS高通量测序技术,分析抽穗期T1C-1和Minghui63根际土壤微生物以及根、茎、叶内生菌的群落结构和潜在功能。【结果】细菌和真菌群落多样性在水稻不同组织生态位之间发生显著变化,地下部分组织生态位(根际土壤和根系)微生物多样性显著高于地上部分(叶和茎)。T1C-1显著影响叶片内生真菌的香农指数和辛普森指数,而对茎和根的内生菌以及根际土壤微生物多样性无显著影响。叶片内生真菌曲霉菌属(Aspergillus)和篮状菌属(Talaromyces)相对丰度在T1C-1显著增加,推测其参与碳素代谢、能量代谢和转录作用酶合成等过程。T1C-1和Minghui63微生物群落关联网络分析表明,T1C-1的平均聚类系数和平均度显著高于Minghui63,因而T1C-1提高了相关微生物群落网络复杂程度。通过重建未观测状态对群落进行系统发育研究(phylogenetic investigation of communities by reconstruction of unobserved states, PICRUSt2),对叶片内生真菌功能酶基因进行功能预测,相对于Minghui63,T1C-1显著改变了碳素代谢、脂类代谢和能量代谢等途径。【结论】相较于根际土壤,叶片内生真菌的群落组成和潜在功能对T1C-1更敏感。尽管如此,T1C-1并未导致叶片内生真菌的多样性指数降低。为了更准确地评估转基因植物的微生态效应,我们需要加强对不同组织生态位内生菌多样性的关注。  相似文献

通过在亚热带杉木(Cunninghamia lanceolata)和米老排(Mytilaria laosensis)人工林中设置互换凋落物、去除凋落物、去除凋落物+去除根系和对照处理来分析改变地上、地下碳输入对人工林土壤微生物生物量和群落组成的影响。结果显示,改变地上、地下碳输入对土壤微生物生物量碳、氮的影响因树种而异。在米老排林中,土壤微生物生物量不受碳源的限制。而在杉木林中,加入米老排凋落物、去除凋落物和去除凋落物+去除根系3种处理中土壤微生物生物量碳、氮具有明显增加的趋势。磷脂脂肪酸分析结果显示,杉木林中,添加高质量的米老排凋落物后,革兰氏阳性细菌、阴性细菌、丛枝菌根真菌、放线菌和真菌群落生物量分别显著增加了24%、24%、53%、25%、28%,革兰氏阴性细菌和丛枝菌根真菌的相对丰度均有显著增加。与对照相比,杉木林中去除凋落物后革兰氏阳性细菌、阴性细菌、丛枝菌根真菌、放线菌和真菌群落生物量分别显著增加了22%、29%、44%、25%、52%,真菌与细菌比值显著增加了21%。但是,去除凋落物+去除根系处理对两个树种人工林土壤微生物群落组成均无显著影响。米老排和杉木林土壤微生物生物量碳、氮的季节变化格局不同,土壤养分有效性可能是驱动土壤微生物生物量季节变化的主要因子。未来研究需要关注凋落物和根系在不同树种人工林中对土壤微生物群落的相对贡献。  相似文献

以20a新疆国家灰漠土土壤肥力与肥料效益长期定位试验为平台,采用常规培养法,结合Biolog技术对可培养微生物、生理菌群数量和碳源利用进行测定分析,研究撂荒(CK0)、耕作不施肥(CK)、不同化肥(N、NK、NP、PK、NPK)、化肥配施低量高量有机肥(NPKM1和NPKM2)和秸秆还田(NPKS)等10种处理土壤微生物特征,揭示长期施肥对土壤微生物群落结构与功能多样性的影响。结果表明:(1)可培养微生物:与CK处理相比,CK0处理显著提高了细菌、放线菌和真菌的数量(P0.05),NPKS处理微生物数量则显著降低(P0.05);不同化肥处理的细菌(除PK处理外)、放线菌(除PK和N处理外)数量也有所增加,增幅在8.14%—135.70%和15.30%—44.78%之间;真菌数量(除NK处理外)则有一定幅度的降低;NPKM1和NPKM2处理,微生物数量最高,细菌分别增加了162.20%和173.75%,放线菌增加了34.39%和39.37%,真菌增加了63.33%和488.33%;(2)生理菌群:与CK0相比,CK处理显著提高了自生固氮菌和亚硝化细菌数量(P0.05),显著降低了氨化细菌和纤维素分解菌数量(P0.05);与CK相比,NPKM1和NPKM2处理显著提高土壤中与氮素转化有关的生理菌群数量(P0.05),不同化肥处理和NPKS处理的影响不相同,NPK处理显著高于其余处理(P0.05);(3)微生物碳源利用:微生物活性表现为NK、NPKM1、NPKM2N、NPK、CKPK、NPKSCK0、NP;CK0处理3个多样性指数以及NPKM1、NPKM2和NK处理Shannon(H)指数最高,其余施肥处理差异不显著;糖类、氨基酸类、羧酸类和胺类是微生物利用的主要碳源。(4)聚类分析表明,除NP处理外,施氮处理土壤有较为相似的碳源利用,细菌和真菌与养分之间有较好的相关性,可培养微生物和生理菌群与微生物碳源利用的相关性较差。因此,长期不同施肥对新疆灰漠土土壤微生物群落结构和功能多样性产生了显著的影响,长期耕作不施肥降低了土壤微生物群落结构和功能多样性,不同化肥配合施用对微生物群落的影响不同,NPK及NPK配施有机肥可提高土壤微生物多样性。  相似文献

被动游泳运动可诱发小鼠抑郁样行为,游泳环境的改变已成为抑郁样行为严重程度影响因素之一。观察不同水质、水温及持续时间对被动游泳小鼠抑郁样行为的影响,并初步探讨肠道菌群组成与抑郁样行为的关系。通过不同条件下的被动游泳运动建立抑郁样行为小鼠模型。采用糖水偏好实验及强迫游泳实验评价其行为学变化;采用16S rDNA高通量测序技术及实时荧光定量PCR技术对小鼠肠道菌群进行分子生态学分析。被动游泳16周后,各模型组小鼠体质量及糖水偏爱度均较正常对照组降低,而不动时间则有所延长。其中,室温海水游泳15 min小鼠体质量及糖水偏爱度降低程度最大,不动时间最长,与正常对照组比较,均具有显著性差异(P<0.05)。各模型组小鼠肠道菌群Chao指数、Shannon指数及PCoA分析均较正常对照组具有显著性差异(P<0.05),其从门水平到属水平的丰度也发生不同程度改变。其中,室温海水游泳15 min小鼠肠道菌群组成变化程度最大,并发生拟杆菌属、普氏菌属等多个菌属的富集以及乳杆菌属丰度的减少,实时荧光定量PCR实验也得到了较为一致的结果(P<0.05)。以上结果表明,被动游泳运动可导致小鼠抑郁样行为的发生,其肠道菌群组成也发生明显改变;同时,菌群组成的改变会随着抑郁样行为的严重程度而有所变化。  相似文献
