Summary Analyses of leaves and ‘tubers’ from somatic hybrids of potato and tomato (‘pomato’ with plastids of potato, ‘topato’ with
plastids of tomato) produced by fusion of protoplasts from liquid cultures of dihaploid potato and mesophyll of tomato revealed
the presence of the two major potato glycoalkaloids (α-solanine and α-chaconine) as well as the tomato glycoalkaloid (αtomatine). The total alkaloid content of leaves was greater than that of ‘tubers’ and similar to levels in the foliage of
parent plants. However, glycoalkaloids were more abundant in hybrid ‘tubers’ than in normal potato tubers by a factor of 5–15.
In hybrid foliage, approximately 98% of the alkaloid present was of potato origin whereas in ‘tubers’ the reverse was the
case, with tomatine comprising 60–70% of the total alkaloid. The similarities in alkaloid content and ratios between the pomato
and the topato lines indicate that plastomes do not influence the biosynthesis and distribution of these alkaloids. The results
indicate that major secondary metabolites may prove useful for assessing the hybrid nature of such plants. 相似文献

Environmental stresses at particularly vulnerable stages during crop development may severely diminish productivity. At temperature
of 10 °C or below cultivated tomato germinate slowly if at all. In this study, seven tomato genotypes bred at the Research
Institute of Vegetable Crops were evaluated for germination time at 10 °C. Analysis identified that one genotype which has
L. chilense in its pedigree, germinated most rapidly while four other genotypes germinated slower. After 21 days, four out of five of

Allotriploid somatic hybrids were obtained from fusions between protoplasts of diploid tomato and monohaploid potato. The selection of fusion products was carried out in two different ways: (1) The fusion of nitrate reductase-deficient tomato with potato gave rise only to hybrid calli if selection was performed on media lacking ammonium. Parental microcalli were rarely obtained and did not regenerate. (2) The fusion of cytoplasmic albino tomato with potato gave rise to albino and green hybrid calli and plants. Allotriploids were identified from the two somatic hybrid populations by counting chloroplast numbers in leaf guard cells and by flow cytometry of leaf tissue. Although some pollen fertility of allotriploids and pollen-tube growth of tomato, potato andLycopersicon pennellii into the allotriploid style were observed, no progeny could be obtained. The relevance of allotriploid somatic hybrids in facilitating limited gene transfer from potato to tomato is discussed. 相似文献

Tomato mesophyll protoplasts were cultured in TM2 medium containing 5.7 M -naphthaleneacetic acid and 2.4 M benzyladenine and were incubated either in stationary culture or on an orbital shaker at 25–30 strokes per min, in combination with interval addition of fresh medium. The effects of stationary and shaking conditions on the growth of the colonies and their subsequent shoot organogenesis were significantly different. The cultures maintained in stationary condition without adding fresh medium accumulated a thin membranous layer on the medium surface and whitish substance in the medium that seemed to precede cell browning and premature colony death. Mild shaking conditions along with the reduction of colony density by one half by dividing the contents of one dish into two dishes, after adding 2 ml of fresh medium on the 4th day and further addition of fresh medium (0.5 ml) on the 8th day of plating, provided optimal conditions for colony growth and suppressed thin layer and whitish substance accumulation. Ten-day-old colonies raised through this protocol regenerated shoots rapidly (within 19–20 days after initial plating) after transfer to regeneration medium (MS medium with 2.8 M zeatin riboside, 0.06–0.1 M gibberellic acid, 4% sucrose and 1% type VII agarose) directly bypassing the callus phase.Abbreviations BA

Two lines of tomato (Lycopersicon esculentum Mill.) representing extremes in utilization effciency of absorbed Ca were studied to detect internal differences in Ca transport and distribution and factors responsible for strain differences in susceptibility to low Ca-stress. Differences in efficiency of Ca use were expressed as CaER (mg of dry weight produced for each mg of Ca absorbed by the plant).Ca-efficiency in line 113(E) appeared to be associated with a slow continuous movement of absorbed Ca, allowing for continued growth of the shoot apex and upper lamina under Ca-deficiency conditions. In the inefficient line 67(I), in contrast, Ca was rapidly deposited in the lower leaves with little upward movement in the plant after absorption.Fractionation of tissue Ca into various chemical forms suggested that Ca inefficiency also was associated with higher concentrations of insoluble Ca in the shoot tissue. The efficient line, although sustaining growth at lower levels of Ca, was capable of maintaining a higher ratio of soluble to insoluble Ca in all shoot tissues.Calcium was concentrated in the lower plant tissues of the inefficient strain, limiting its availability for continued shoot growth.Autoradiographs of lines fed45Ca during the final 8 days of a 24-day experiment suggested that upward movement was sustained in line 113(E), in spite of vastly reduced transpiration rates and a root system characterized by leakage of K ions from the roots back into the solution. 相似文献

Summary Two somatic hybrid plants generated from a single fusion event between Lycopersicon esculentum and irradiated L. pennellii protoplasts have been analyzed at the molecular level. Over 30 loci have been analyzed using isozymes and RFLPs. All loci tested on chromosomes 2–10 were heterozygous, while those loci on chromosome 12 were homozygous L. pennellii in both somatic hybrids. In one of the somatic hybrids, 2850, loci on chromosome 1 were also homozygous L. pennellii. The other somatic hybrid, 28F5, was heterozygous at all chromosome 1 loci tested, but exhibited altered stoichiometry of parental bands as compared to the sexual hybrid. Loci on chromosome 2 from both somatic hybrids have altered stoichiometry, with L. pennellii alleles being four times more abundant than expected. Both somatic hybrids contain the L. esculentum chloroplast genome, while only L. pennellii polymorphisms have been detected in the mitochondrial genome. 相似文献

Regeneration of de novo shoots, roots and flowers has been obtained on inflorescence explants of tomato (Lycopersicon esculentum Mill.). Indole-3-acetic acid (IAA), indole-3-butyric acid (IBA) and -naphthaleneacetic acid (NAA) were added in a 3×3×3 factorial combination with kinetin, each at 0.001, 0.1 and 10 M concentrations. Direct shoot formation occurred on media with 10 M kinetin and 0.001 M IAA or NAA. Root formation was observed on media with 0.1–10 M IAA, IBA or NAA. Flower formation occurred on elongated shoots with several leaves on media with 10 M IAA and 0.1 M kinetin. Shoot organogenesis was increased by substituting 10 M zeatin or N6-benzyladenine (BA) for kinetin. Eleven tomato cultivars were tested for their ability to undergo de novo shoot regeneration on the improved medium. All tomato cultivars were capable of shoot morphogenesis with a mean number of shoots per explant that ranged from 1.3 (Red Alert) to 5.3 (Large Red Cherry). Histological studies revealed that active cell divisions occurred in subepidermal and cambial tisue during the first week of culture. Meristematic centers of dividing cells were evident by day 14, and well-developed shoot apices and leaf structures were observed on 50% of the explants 28 days after culture initiation.Abbreviations BA

Iron is one of the most important micronutrients for plants. Like other organisms, plants have developed active mechanisms for the acquisition of sufficient iron from the soil. Nevertheless, very little is known about the genetic mechanisms that control the active uptake. In tomato, two spontaneously derived mutants are available, which are defective in key steps that control this process. The recessive mutationchloronerva (chln) affects a gene which controls the synthesis of the non-protein amino acid nicotianamine (NA), a key component in the iron physiology of plants. The root system of the recessive mutantfer is unable to induce any of the characteristic responses to iron deficiency and iron uptake is thus completely blocked. We present a characterization of the double mutant, showing that thefer gene is epistatic over thechln gene and thus very likely to be one of the major genetic elements controlling iron physiology in tomato. In order to gain access to these two genes at the molecular level, both mutants were precisely mapped onto the high density RFLP map of tomato. Thechln gene is located on chromosome 1 and thefer gene is on chromosome 6 of tomato. Using this high-resolution map, a chromosome walk has been started to isolate thefer gene by map-based cloning. The isolation of thefer gene will provide new insights into the molecular mechanisms of iron uptake control in plants. 相似文献

Differences in responses to iron deficiency between two chickpea cultivars, NP-62 and K-850, were examined. The apical leaves of NP-62 quickly showed symptoms of iron-deficiency chlorosis when grown on an iron-free medium. By contrast, K-850 showed no visible symptoms on the same medium. Iron contents of the apical leaves of these two cultivars were similar during the first 7 days after they were transferred to the iron-free medium in spite of a marked difference in root-associated Fe3+-reduction activity. The susceptibility to iron-deficiency chlorosis observed in NP-62 was not attributable to the poor Fe3+-reduction activity of roots but to the inefficient utilization of iron within leaves under conditions when the supply of iron was limited. 相似文献

A two-year estimation of Botrytis cinerea stem damage of 299 experimental F1hybrids grown in 2 rotations: winter-autumn and summer-autumn was carried out. The estimation was made in a period of mass
fruitage.
As a result it was shown, that the hybrids with determinate type of growth (sp) are damaged more strongly than indeterminate
ones (sp +). An average disease severity in the first group was 2.8 against 2.3 in the second one using a scale 0–4. Number
of strongly injured (2.5–4) plants in each group were 44.1 % and 30.7 % and number of moderate resistant (0–1.5 numbers) plants
were 10 % and 8.5 % accordingly.
The analysis of resistance of F1 hybrids determined significant influence of parental lines on a degree of a damage of plant stems by B. cinerea. 相似文献

Effect of withdrawal of phosphorus on nitrate assimilation and PEP carboxylase activity in tomato 总被引:11,自引:0,他引:11
Tomato plants (Lycopersicon esculentum) grown in a complete nutrient solution for 8 days were transferred to a P-free solution of pH 6.0. Within 2 days of transfer the rate of alkalinization of the nutrient solution declined and by 4 days the solution had become acid. Nitrate transferred from roots to leaves was depressed over this period, and the rate of nitrate reductase activity in the leaves (the main site of assimilation of nitrate in tomato) had declined by 60% within 5 days of transfer. The activity of PEP carboxylase in the leaves of the P-deficient plants increased after 3 days, eventually becoming 3 times greater than in the leaves of plants adequately supplied with P. The PEP carboxylase activity in the roots of the P-deficient plants increased within 2 days, becoming 4 times greater after 8 days' growth. These results are discussed in relation to mechanisms for enhancement of P acquisition and maintenance of cation and anion uptake during P-deficiency. 相似文献

Summary Effects of genotype and explant orientation on shoot regeneration from cotyledonary explants of tomato were studied using
10 commercially important cultivars. The explant orientation affected shoot regeneration in all the tested genotypes. Cotyledons
placed in abaxial (lower surface facing down) orientation consistently produced better shoot regenerative response and produced
greater numbers and taller shoots compared to those inoculated in adaxial (upper surface facing down) orientation. Genotypic
variation in terms of shoot regeneration response, number of shoots, and shoot height was apparent. Wounding of cotyledonary
explants increased shoot regeneration response. However, shoot height was much lower in shoots regenerated from wounded explants
compared to those that originated from intact cotyledons. Shoots produced from wounded cotyledons were abnormal in their form

The incorporation of labelled adenine into cytokinin-like compounds was investigated in intact tomato plants, decapitated tomato roots and cell-free root extracts. In all three cases evidence was found for the incorporation of adenine into endogenous cytokinins. In intact plants and decapitated root systems no evidence was found for the incorporation into cytokinin nucleotides. Cytokinin bases and nucleosides were however, labelled. In the cell-free root extract there was some evidence for the incorporation of labelled adenine into cytokinin nucleotides. This suggests that the biosynthetic process may be strictly compartmentalized. The present results provide no evidence for the relative importance of cytokinin nucleotides in the biosynthetic process. What is clear is that the rate of adenine incorporation into cytokinins is extremely low and that only a small proportion of the cytokinins which became labelled were exported to the shoot via the root exudate.The financial support of the CSIR/Israel Collaborative Programme is gratefully acknowledged. 相似文献

Summary The Mi gene originating from the wild tomato species Lycopersicon peruvianum confers resistance to all major root knot nematodes (Meloidogyne spp.). This single dominant gene is located on chromosome 6 and is very closely linked to the acid phosphatase-1 (Aps-1) locus. Resistance to nematodes has been introgressed into various cultivars of the cultivated tomato (L. esculentum), in many cultivars along with the linked L. peruvianum Aps-1

allele. By using a pair of nearly isogenic lines differing in a small chromosomal region containing the Mi and Aps-1 loci, we have identified two RFLP markers, GP79 and H6A2c2, which are located in the introgressed L. peruvianum region. Analysis of a test panel of 51 L. esculentum genotypes of various origins indicated that GP79 is very tightly linked to the Mi gene and allows both homozygous and heterozygous nematode-resistant genotypes to be distinguished from susceptible genotypes, irrespective of their Aps-1 alleles. Marker H6A2c2 is linked to the Aps-1 locus and is capable of discriminating between the L. peruvianum Aps-1

allele and the L. esculentum Aps-1

Significant yield losses in commercial tomato production caused by tomato spotted wilt virus (TSWV) are the reason why we
have undertaken studies on resistance to this pathogen. One of the possible sources of resistance can be the incorporation
of the nucleoprotein N viral gene by Agrobacterium transformation. The N gene was introduced into three Lycopersicon esculentum forms. Out of the total of 3044 cotyledon explants 14.7% regenerated shoots, but only a few were rooted on medium containing

Changes in root-associated Fe(III) reductase activity and Fe concentration during recovery from temporary iron starvation were investigated in hydroponically grown Plantago lanceolata L. Within two days, interruption of the Fe supply resulted in enhanced rates of reduction by intact plant roots. Transfer of iron deficient plants to a solution containing various amounts of chelated Fe caused a transient increase in reduction activity before the rates declined to a level determined by the amount of Fe added. Repression of root-associated redox activity was independent of the Fe concentration in the preculture. When iron deficient plants were submitted to a supply of Fe localized to a part of the root system (split-root plants), the decrease in reduction rates was much more pronounced in the Fe-deprived portion of the roots than in the Fe-supplied one. No correlation was observed between root Fe concentration and Fe(III) reductase activity. Continued growth of split-root plants in the +Fe/-Fe regime increased the reduction rates of the +Fe-grown portion of the root system over the rates in iron sufficient plants with non-divided roots. The results are discussed in relation to putative factors mediating intra- and interorgan regulation of iron nutrition. 相似文献

Experiments were conducted to study the physiologicaleffect of the plant growth retardant paclobutrazol(PBZ) and its impact on the yield of tomato plants(cv. Precador). Seedlings were treated at the time of prickingout with soil and foliar applications of PBZ atconcentrations of 1.0 and 25.0 mg l-1respectively. The results established that:-- The reduced height and the increased thickness ofthe young plant stem, as well as the accelerated rootformation are a significant advantage of the PBZtreatment, contributing to the improvement of seedlingquality at planting.-- Soil treatment (1 mg l-1) and foliar treatment(25 mg l-1) with PBZ improves the photosyntheticactivity and water balance of tomato cv. Precador.-- PBZ accelerates fruit formation and increases earlyfruit yield.-- The concentrations of the retardant used and themode of its application ensure the production offruits without any residual retardant and harmless tohuman health from a phytosanitary point of view. 相似文献
