Old-growth forest stands of mixed species composition provide the opportunity to study species-specific influences on soil properties. We monitored rates of nitrogen mineralization, nitrification and an index of ammonium and nitrate uptake in a mixed old-growth stand of Douglas-fir (Pseudotsuga menziesii), western hemlock (Tsuga heterophylla) and western redcedar (Thuja plicata) over a two-year period. Litter and mineral soil (0–10-cm depth) were sampled adjacent to ten large trees of each species. After initial characterization of litter and soil, buried bags were incubated in both layers for ca. 2-month intervals. Soil and litter pH was lowest near western hemlocks. Nitrification, nitrate concentrations, and percent uptake as nitrate differed among the tree species; rates were highest near western redcedars. For all species, percent nitrification and nitrate uptake rate were higher in soil than in litter. The results indicate species-specific effects on ammonium and nitrate production and uptake within this forest type.The research described in this article has been funded in part by the US Environmental Protection Agency. We document here that in those rare cases where disease has been related to Bacillus licheniformis, infection was associated with bypassing the normal biological protective barriers or severely debililated patients. No case suggests any invasive properties of this bacterium. B. licheniformis can therefore be considered non-pathogenic to humans in general. Food-borne illness caused by possible B. licheniformis toxins have been reported, but only in a very few cases and only in connection with consumption of inappropriately prepared food. Considerable experience concerning the industrial use of recombinant B. licheniformis strains has now accumulated and authorities in the United States, Europe and Japan have approved production with and products from recombinant B. licheniformis strains. Transgenic plants are traditionally characterized using phenotypic and Southern analyses. Over the last several years, fluorescence in situ hybridization (FISH) has been efficiently used for visualization, chromosomal localization and estimation of numbers of transgene loci in transgenic plants. Recent results obtained in different plant species transformed using either Agrobacterium tumefaciens or microprojectile bombardment indicate that FISH is also a powerful tool for characterization of transgene locus structure that significantly augments conventional Southern analysis. The aim of this review is to document the usefulness of FISH for characterization of transgene loci in plants.  相似文献

The eastern North American spring ephemeral Claytonia virginica (Portulacaceae) is well known for its great variation in chromosome number. The origin and significance of this cytological diversity has been a source of some controversy over the last two decades; in particular it has been suggested that one major source of variation could have arisen by allopolyploidy following hybridization between C. virginica and the second eastern North American representative of the genus, C. caroliniana. However, there has been no rigorous demonstration of natural hybridization between these taxa, and attempts to document hybridity have been hampered by a paucity of distinguishing morphological characters. Nuclear ribosomal gene markers are, however, able to distinguish between the two species and provide a means of identifying hybrid plants. We have found a locality in central New York state where the two species cooccur; in this population, hybrid individuals with additive ribosomal gene patterns are found in a zone of overlap between the two parental species. These hybrids are morphologically intermediate and have reduced pollen stainabilities, and appear to be F1 individuals.  相似文献

The venomous snake subfamily Hydrophiinae includes more than 40 genera and approximately 200 species. Most members of this clade inhabit Australia, and have been well studied. But, because of poor taxon sampling of Melanesian taxa, basal evolutionary relationships have remained poorly resolved. The Melanesian genera Ogmodon, Loveridgelaps, and Salomonelaps have not been included in recent phylogenetic studies, and the New Guinean endemic, Toxicocalamus, has been poorly sampled and sometimes recovered as polyphyletic. We generated a multilocus phylogeny for the subfamily using three mitochondrial and four nuclear loci so as to investigate relationships among the basal hydrophiine genera and to determine the status of Toxicocalamus. We sequenced these loci for eight of the 12 described species within Toxicocalamus, representing the largest molecular data set for this genus. We found that a system of offshore island arcs in Melanesia was the centre of origin for terrestrial species of Hydrophiinae, and we recovered Toxicocalamus as monophyletic. Toxicocalamus demonstrates high genetic and morphological diversity, but some of the molecular diversity is not accompanied by diagnostic morphological change. We document at least five undescribed species that all key morphologically to Toxicocalamus loriae (Boulenger, 1898), rendering this species polyphyletic. Continued work on Toxicocalamus is needed to document the diversity of this genus, and is likely to result in the discovery of additional species. Our increased taxon sampling allowed us to better understand the evolution and biogeography of Hydrophiinae; however, several unsampled lineages remain, the later study of which may be used to test our biogeographic hypothesis.  相似文献

Background  

Pectate lyases depolymerize pectins by catalyzing the eliminative cleavage of α-1,4-linked galacturonic acid. Pectate lyase-like (PLL) genes make up among the largest and most complex families in plants, but their cellular and organismal roles have not been well characterized, and the activity of these genes has been assessed only at the level of entire organs or plant parts, potentially obscuring important sub-organ or cell-type-specific activities. As a first step to understand the potential functional diversity of PLL genes in plants and specificity of individual genes, we utilized a reporter gene approach to document the spatial and temporal promoter activity for 23 of the 26 members of the Arabidopsis thaliana (Arabidopsis) PLL gene family throughout development, focusing on processes involving cell separation.  相似文献

Exotic plants have been demonstrated to be one of the greatest threats to wetlands, as they are capable of altering ecosystem-wide physical and biological properties. One of the most problematic invaders in the western United States has been salt cedar, Tamarix spp., and the impacts of this species in riparian and desert ecosystems have been well-documented. Here we document large populations of tamarisk in the intertidal salt marshes of Tijuana River National Estuarine Research Reserve, a habitat not often considered vulnerable to invasion by tamarisk. Initial research demonstrates that there are multiple species and hybrids of Tamarix invading the estuary and that the potential impact of tamarisk within this salt marsh is significant. This highlights the need for managers and scientists to be aware of the problems associated with tamarisk invasion of coastal marine habitats and to take early and aggressive action to combat any incipient invasion.  相似文献

The Clinical and Laboratory Standards Institute (CLSI) has standardized broth microdilution and disk diffusion methodology for testing filamentous fungi (molds) that cause invasive disease. Quality control MIC (minimal inhibitory concentration) and MEC (minimal effective concentration; echinocandins only) limits are also available in the recently published CLSI M38-A2 document. Although breakpoints based on correlations of in vitro results and clinical outcome have not been established, MIC or MEC and zone diameter categories for five antifungal agents and various mold species, as well as epidemiologic cutoffs for Aspergillus fumigatus versus the triazoles, have been recently documented. Some insights of the potential clinical value of reference methods also have been reported. During the past few years, the potential utility of various commercial methods has been evaluated by comparing them with reference methodology. This review summarizes and discusses the advantages and disadvantages of these developments.  相似文献

Experimental kinetic data (initial rate and high conversion) on the hydrolysis of cellobiose by 1,4-β-glucosidace (Gliocladium sp.) have been analysed and a competitive inhibition by glucose has been proposed. The determination of kinetic parameters from integral data is based upon algorithms for non-linear optimization and numerical integration. The values of kinetic constants \documentclass{article}\pagestyle{empty}\begin{document}$(v_{\max } = 1.02\frac{{\mu {\rm M}_{{\rm glucose}} }}{{{\rm mg}_{{\rm protein}} \cdot \min }},K_M = 2.6{\rm mM/l, and }K_P = 1.2{\rm mM/l)}$\end{document} agree well with the initialrate results. Whether play occurs in fishes has long been a contentious issue, but recent observations document that social, object, and locomotor play can all be found in some species of teleosts. However, quantitative studies and those documenting individual differences are rare. We recorded hundreds of occurrences of an unusual behavior in three male Tropheus duboisi. The target behavior of attacking and deflecting an object that rapidly returned to its upright position not only fit the criteria for play behavior, but differed quantitatively and qualitatively among the individuals. This behavior has not been observed in other species of cichlids and other kinds of fishes. The presence or absence of food or other fish either within the aquarium or visible in an adjacent aquarium had no marked or consistent effect on the occurrence of the behavior. Various explanations for the origin and function of the behavior are discussed.  相似文献

Objective : Leptin, an adipocyte-secreted hormone, has been shown to signal the status of energy stores to the brain, regulate energy homeostasis, and mediate the neuroendocrine response to food deprivation. Obesity is associated with increased leptin levels, and several hormones, including insulin and glucocorticoids, have been associated with leptin levels and expression in rodents. Although obesity has been strongly associated with increased leptin in humans, a significant percentage of leptin's variability remains unexplained. The role of endogenous hormones, demographic factors, or certain life-style factors in explaining the residual variability of leptin levels has not yet been clarified. We performed this cross-sectional study to document the relative importance of obesity, lifestyle factor, and endogenous hormones in determining serum leptin levels. Research Methods and Procedures : We measured serum concentrations of insulin, Cortisol, testosterone, growth hormone, and dehydroepiandrosterone sulfate; ascertained anthropometric, demographic, and lifestyle characteristics; and studied these variables in relationship to serum leptin concentrations in a sample of young healthy men. Results : Obesity and alcohol intake were independently and positively associated with circulating leptin concentrations. Additionally, cigarette smoking was negatively and independently associated with leptin concentrations. Finally, serum insulin concentration was an independent hormonal determinant of circulating leptin concentrations, whereas serum testosterone was negatively associated with leptin only by bivariate analysis. Discussion : We conclude that, in addition to obesity, cigarette smoking, alcohol intake, and serum insulin levels are associated with leptin levels in a population of healthy young men.  相似文献

Among the Cladocera, the occurrence of interspecific hybrids in nature has been reported frequently but rarely adequately documented. During the course of a survey of the genetic variation and population structure of several species ofSimocephalus in ponds in southern Ontario, Canada, readily distinguishable congeners often co-occurred in a pond. Occasionally, clones were recovered that were intermediate in phenotype (as determined from both morphological and electrophoretic analyses) between two species. Two different hybrid clones have been examined in detail to determine the parentals involved in the crosses. Evidence from field and laboratory studies is presented to document the hybrid nature of these clones. Factors favouring formation and long-term survival of hybrids in littoral zone and pelagic species are discussed.  相似文献

A series of recently published studies have established that defects in endocytic sorting can elicit dramatic tissue overgrowth phenotypes in developing organs of the fruit fly Drosophila melanogaster. Such a link had been suggested by mammalian cell culture experiments almost ten years ago, but in vivo evidence of this link, and the mechanisms through which it might occur, had remained elusive. Drosophila has now proven to be an excellent developmental system in which to document the effects of endocytic defects on tissue growth, and to probe the basis of these phenotypes. This work has begun to illuminate some surprising connections between the endocytic trafficking of protein cargoes and the control of cell proliferation and tissue architecture. These connections touch major cell biological processes, including cell division, growth, death, and polarity, and have begun to paint a complex yet intriguing picture of how defective endocytic sorting can effect developing tissues.  相似文献

The significance of DNA repair to human health has been well documented by studies on xeroderma pigmentosum (XP) patients, who suffer a dramatically increased risk of cancer in sun-exposed areas of their skin [1] and [2]. This autosomal recessive disorder has been directly associated with a defect in nucleotide excision–repair (NER) [1] and [2]. Like human XP individuals, mice carrying homozygous mutations in XP genes manifest a predisposition to skin carcinogenesis following exposure to ultraviolet (UV) radiation [3], [4] and [5]. Recent studies have suggested that, in addition to roles in apoptosis [6] and cell-cycle checkpoint control [7] in response to DNA damage, p53 protein may modulate NER [8]. Mutations in the p53 gene have been observed in 50% of all human tumors [9] and have been implicated in both the early [10] and late [11] stages of skin cancer. To examine the consequences of a combined deficiency of the XPC and the p53 proteins in mice, we generated double-mutant animals. We document a spectrum of neural tube defects in XPC p53 mutant embryos. Additionally, we show that, following exposure to UV-B radiation, XPC p53 mutant mice have more severe solar keratosis and suffer accelerated skin cancer compared with XPC mutant mice that are wild-type with respect to p53.  相似文献

To mark this 25‐year milestone, this document provides a brief account (including some pre‐history) of how OMIA came to be; some timelines of important discoveries and advances in the genetics of the animal species covered by OMIA, gleaned from the OMIA database; and an analysis of the current state of knowledge regarding likely causal variants of single‐locus traits in OMIA species, also gleaned from the OMIA database.  相似文献

Native plant materials (NPMs) are increasingly utilized during the restoration of disturbed plant communities. Here, we analyze next‐generation genetic sequencing data for Hilaria jamesii, a dominant graminoid across drylands of the southwestern United States, and document that the species' only commercially‐available NPM, “Viva,” is a hybrid between H. jamesii and its sister species, Hilaria mutica. In fact, hybrids between these species are common where they geographically overlap. Furthermore, we show that the “Viva” hybrid has successfully been moved beyond the hybrid zone and into the core range of H. jamesii. The potential ramifications of introducing novel genetic material into H. jamesii are discussed, as well as the utility of genetic analyses to protect species' natural patterns of genetic diversity and help managers make informed decisions regarding the development and deployment of NPMs.  相似文献

Greenland is a continental island in the northern part of the North Atlantic where the foliose Bangiales flora is poorly known. It is an important area for the study of algal biogeography because of the region’s glacial history, in which Greenland has been alternately exposed to or isolated from the North Pacific via the Bering Strait. A molecular study using 3′ rbcL + 5′ rbcL–S sequences was undertaken to assess the diversity of foliose Bangiales on the west coast of Greenland and rbcL sequences were used to study the Greenland flora in a larger phylogenetic and floristic context. New and historic collections document seven species in four genera from the west coast of Greenland. All species had a close link to North Pacific species, being either conspecific with them or North Atlantic–North Pacific vicariant counterparts.  相似文献
