One and a half year-old Ginkgo saplings were grown for 2 years in 7 litre pots with medium fertile soil at ambient air CO2 concentration and at 700 μmol mol−1 CO2 in temperature and humidity-controlled cabinets standing in the field. In the middle of the 2nd season of CO2 enrichment, CO2 exchange and transpiration in response to CO2 concentration was measured with a mini-cuvette system. In addition, the same measurements were conducted in the crown of one 60-year-old tree in the field. Number of leaves/tree was enhanced by elevated CO2 and specific leaf area decreased significantly.CO2 compensation points were reached at 75–84 μmol mol−1 CO2. Gas exchange of Ginkgo saplings reacted more intensively upon CO2 than those of the adult Ginkgo. On an average, stomatal conductance decreased by 30% as CO2 concentration increased from 30 to 1000 μmol mol−1 CO2. Water use efficiency of net photosynthesis was positively correlated with CO2 concentration levels. Saturation of net photosynthesis and lowest level of stomatal conductance was reached by the leaves of Ginkgo saplings at >1000 μmol mol−1 CO2. Acclimation of leaf net CO2 assimilation to the elevated CO2 concentration at growth occurred after 2 years of exposure. Maximum of net CO2 assimilation was 56% higher at ambient air CO2 concentration than at 700 μmol mol−1 CO2.  相似文献

Photosynthetic characteristics of Cymbidium plantlet in vitro   总被引:17,自引:0,他引:17
The photosynthetic characteristics of the Cymbidium plantlet in vitro cultured on Hyponex-agar medium with 2% sucrose were determined based on the measurements of CO2 concentration inside and outside of the culture vessels. The CO2 measurements were made with a gas chromatograph at a PPF (photosynthetic photon flux) of 35, 102 and 226 mol m-2 s-1, a chamber air temperature of 15, 25 and 35°C and a CO2 concentration outside the vessel of approximately 350, 1100 and 3000 ppm. The net photosynthetic rates were determined on individual plantlets and were expressed on a dry weight basis. The steady-state CO2 concentration during the photoperiod was lower inside the vessel than outside the vessel at any PPF greater than 35 mol m-2s-1 and at any chamber air temperature. The photosynthetic response curves relating the net photosynthetic rate, PPF, and CO2 concentration in the vessel and chamber air temperature were similar to those for Cymbidium plants grown outside and other C3 plants grown outside under shade. The results indicate that CO2 enrichment for the plantlets in vitro at a relatively high PPF would promote photosynthesis and hence the growth of chlorophyllous shoots/plantlets in vitro and that the plantlets in vitro would make photoautotrophic growth under environmental conditions favorable for photosynthesis.Abbreviations Cin CO2 concentration in the culture vessel - Cout CO2 concentration outside the vessel (in the culture room) - PPF photosynthetic photon flux  相似文献

Potato plants (Solanum tuberosum L. cv. Bintje) were grown to maturity in open-top chambers under three carbon dioxide (CO2; ambient and 24 h d−1 seasonal mean concentrations of 550 and 680 μmol mol−1) and two ozone levels (O3; ambient and an 8 h d−1 seasonal mean of 50 nmol mol−1). Chlorophyll content, photosynthetic characteristics, and stomatal responses were determined to test the hypothesis that elevated atmospheric CO2 may alleviate the damaging influence of O3 by reducing uptake by the leaves. Elevated O3 had no detectable effect on photosynthetic characteristics, leaf conductance, or chlorophyll content, but did reduce SPAD values for leaf 15, the youngest leaf examined. Elevated CO2 also reduced SPAD values for leaf 15, but not for older leaves; destructive analysis confirmed that chlorophyll content was decreased. Leaf conductance was generally reduced by elevated CO2, and declined with time in the youngest leaves examined, as did assimilation rate (A). A generally increased under elevated CO2, particularly in the older leaves during the latter stages of the season, thereby increasing instantaneous transpiration efficiency. Exposure to elevated CO2 and/or O3 had no detectable effect on dark-adapted fluorescence, although the values decreased with time. Analysis of the relationships between assimilation rate and intercellular CO2 concentration and photosynthetically active photon flux density showed there was initially little treatment effect on CO2-saturated assimilation rates for leaf 15. However, the values for plants grown under 550 μmol mol−1 CO2 were subsequently greater than in the ambient and 680 μmol mol−1 treatments, although the beneficial influence of the former treatment declined sharply towards the end of the season. Light-saturated assimilation was consistently greater under elevated CO2, but decreased with time in all treatments. The values decreased sharply when leaves grown under elevated CO2 were measured under ambient CO2, but increased when leaves grown under ambient CO2 were examined under elevated CO2. The results obtained indicate that, although elevated CO2 initially increased assimilation and growth, these beneficial effects were not necessarily sustained to maturity as a result of photosynthetic acclimation and the induction of earlier senescence.  相似文献

In the threespine stickleback (Gasterosteus aculeatus) lactate dehydrogenase (LDH, EC 1.1.1.27) is encoded by three loci, Ldh-A, Ldh-B, and Ldh-C. LDH-B4 isoenzyme restricted its function to eye and brain, while LDH-C4 isoenzyme functions in the eye. In the Dead Vistula stickleback population, none of LDH loci is polymorphic. The LDH-B4 and LDH-C4 isoenzymes from the eye were purified to homogeneity to specific activity of 186 and 229 μmol NADH min−1mg−1, respectively, at 30°C. Some physico-chemical and kinetic properties revealed that eye LDH-C4 isoenzyme was more thermostable and had a higher affinity to pyruvate than LDH-B4 isoenzyme. Lower Km for pyruvate of eye LDH-C4 isoenzyme distinguishes it from fish LDH-C4 isoenzyme isolated from liver.  相似文献

Explants and seeds of Brassica campestris L. were cultured on Murashige & Skoog (1962) medium with and without sucrose in a vessel with different numbers of air changes per hour under different PPF (photosynthetic photon flux) conditions. The growth and development of plantlets in the vessel were similar to those of seedlings when cultured under the same in vitro environmental conditions. The growth and development of seedlings when cultured under the same in vitro environmental conditions. The growth and development of plantlets/seedlings were greater for treatments with a higher number of air changes per hour and a higher PPF regardless of the sucrose concentration in the culture medium. The CO2 concentration in the vessel with a lower number of air changes per hour decreased to approximately 100 ppm during the photoperiod on day 21 due to the photosynthetic activities of the plantlets/seedlings. The low CO2 concentration, in turn, reduced the net photosynthetic rate of plantlets/seedlings in the vessel, and thus affected their growth and development.Abbreviations Cin CO2 concentration in the culture vessel - Cout CO2 concentration in the culture room - MS mineral composition of Murashige & Skoog (1962) medium - PPF photosynthetic photon flux  相似文献

Net photosynthetic rates (P n) of easy (EK 16-3) and difficult-to-acclimatize (EK 11-1) sea oats genotypes were examined under the following culture conditions: (1) photoautotrophic [sugar-free medium, high photosynthetic photon flux (PPF), high vessel ventilation rates and CO2 enrichment, (PA)]; (2) modified photomixotrophic [sugar-containing medium diluted with sugar-free medium over time, high PPF, and high vessel ventilation rates (PM)]; (3) modified photomixotrophic enriched [same as PM with CO2 enrichment, (PME)]; or (4) conventional photomixotrophic [sugar-containing medium, low PPF, and low vessel ventilation rates (control)]. Regardless of genotype, plantlets cultured under PA conditions died within 2 wk, whereas under PM and PME conditions, plantlets increased their P n. After 6 wk, P n per gram dry weight was 1.7 times greater in EK 16-3 than EK 11-1 plantlets cultured under PME conditions. In vitro-produced leaves of EK 16-3 plantlets were elongated with expanded blades, whereas EK 11-1 produced short leaves without expanded blades, especially under control conditions. After in vitro culture, EK 16-3 PME plantlets exhibited the highest dry weights among treatments. EK 16-3 PME and EK 16-3 PM had similarly high survivability, shoot and root dry weights and leaf lengths ex vitro compared to EK 16-3 control and EK 11-1 PM and PME plantlets. Ex vitro growth, survivability and P n per leaf area of either genotype were not affected by CO2 enrichment under modified photomixotrophic conditions. These results suggest that growth and survivability of sea oats genotypes with different acclimatization capacities can be enhanced by optimizing culture conditions.  相似文献

Nitrogenase activity and the rate of photosynthesis were measured simultaneously in Azolla by a continuous gas flow system. The mode of interaction between light, photosynthesis and nitrogenase activity was analysed.Nitrogenase activity dropped off when either Azolla plants or the cyanobiont Anabaena were transferred from light to dark. This decline was immediate and was independent of length or intensity of the prior light phase. Reillumination restored nitrogenase activity.Nitrogenase activity did not depend on the rate of photosynthesis at light intensities below 10 μE m−2 s−1. Its activity was saturated at 200 μE m−2 s−1 while CO2 fixation was saturated at a light intensity of 850 μE m−2 s−1. Azolla photosynthetic activity followed the absorption spectrum of chlorophyll a, while nitrogenase activity markedly increased between 690 and 710 nm. Inhibition of photosynthesis by DCMU was accompanied by an increase in nitrogenase activity. These results suggest direct light regulation of nitrogenase activity in Azolla independent of CO2 fixation, and a possible inhibition of nitrogenase activity by the oxygen produced in photosynthesis.  相似文献

Prostaglandin I2 potentiated the paw swelling induced by carrageenin in rats. Prostaglandin I2 (0.1 μg) showed similar activity to PGE1 (0.01 μg). This potentiating property disappeared in 60 minutes and was completely abolished by diphenhydramine (25 mg kg−1, i.p.). In vascular permeability tests, PGI2 itself (2.5 × 10−10 mol, 88 ng) caused no dye leakage reaction, but PGE1 (2.5 × 10−10 mol, 88.5 ng) caused a significant dye leakage. This effect of PGE1 was statistically significant compared with vehicle- or PGI2-treated group (p<0.05). Prostaglandin I2 potentiated the increased vascular permeability induced by 5-hydroxytriptamine (2.5 × 10−10 mol), bradykinin (5 × 10−10 mol) and histamine (2 × 10−10 to 2 × 10−8 mol). The potentiation was the most evidence in the case of histamine.  相似文献

Strawberry (Fragaria ananassaDuch. cv. Fengxiang) plantlets were cultured under two in vitroenvironments for rooting, and then acclimatized under two ex vitroirradiance conditions. At the end of rooting stage plant height, fresh weight and specific leaf area of T1-plants grown under high sucrose concentration (3 sucrose), low photosynthetic photon flux density (30 mol m–2 s–1) and normal CO2 concentration (350–400 l l–1) were significantly higher than those of T2-plantlets grown under low sucrose concentration (0.5), high photosynthetic photon flux density (90 mol m–2 s–1) and elevated CO2 concentration (700–800 l l–1). But T2-plantlets had higher net photosynthetic rate (Pn), effective photochemical quantum yield of PSII (PSII), effective photosynthetic electron transport rate (ETR), photochemical quenching (qP) and ratio of chlorophyll fluorescence yield decrease (Rfd). After transfer, higher irradiance obviously promoted the growth of plantlets and was beneficial for the development of photosynthetic functions during acclimatization. T2-plantlets had higher fresh weight, leaf area, PSII and ETR under higher ex vitroirradiance condition.  相似文献

The effects of nitrogen [75 and 150 kg (N) ha−1] and elevated CO2 on growth, photosynthetic rate, contents of soluble leaf proteins and activities of ribulose-1,5-bisphosphate carboxylase/oxygenase (Rubisco) and nitrate reductase (NR) were studied on wheat (Triticum aestivum L. cv. HD-2285) grown in open top chambers under either ambient (AC) or elevated (EC) CO2 concentration (350 ± 50, 600 ± 50 μmol mol−1) and analyzed at 40, 60 and 90 d after sowing. Plants grown under EC showed greater photosynthetic rate and were taller and attained greater leaf area along with higher total plant dry mass at all growth stages than those grown under AC. Total soluble and Rubisco protein contents decreased under EC but the activation of Rubisco was higher at EC with higher N supply. Nitrogen increased the NR activity whereas EC reduced it. Thus, EC causes increased growth and PN ability per unit uptake of N in wheat plants, even if N is limiting.  相似文献

We examined the purine alkaloid content and purine metabolism in cacao (Theobroma cacao L.) plant leaves at various ages: young small leaves (stage I), developing intermediate size leaves (stage II), fully developed leaves (stage III) from flush shoots, and aged leaves (stage IV) from 1-year-old shoots. The major purine alkaloid in stage I leaves was theobromine (4.5 μmol g–1 fresh weight), followed by caffeine (0.75 μmol g–1 fresh weight). More than 75% of purine alkaloids disappeared with subsequent leaf development (stages II–IV). In stage I leaves, 14C-labelled adenine, adenosine, guanine, guanosine, hypoxanthine and inosine were converted to salvage products (nucleotides and nucleic acids), to degradation products (ureides and CO2) and to purine alkaloids (3- and 7-methylxanthine, 7-methylxanthosine and theobromine). In contrast, 14C-labelled xanthine and xanthosine were not used for nucleotide synthesis. They were completely degraded, but nearly 20% of [8-14C]Xanthosine was converted in stage I leaves to purine alkaloids. These observations are consistent with the following biosynthetic pathways for theobromine: (a) AMP → IMP → 5′-xanthosine monophosphate → xanthosine → 7-methylxanthosine → 7-methylxanthine → theobromine; (b) GMP → guanosine → xanthosine → 7-methylxanthosine → 7-methylxanthine → theobromine; (c) xanthine → 3-methylxanthine → theobromine. Although no caffeine biosynthesis from 14C-labelled purine bases and nucleosides was observed during 18 h incubations, exogenously supplied [8-14C]Theobromine was converted to caffeine in young leaves. Conversion of theobromine to caffeine may, therefore, be slow in cacao leaves. No purine alkaloid synthesis was observed in the subsequent growth stages (stages II–IV). Significant degradation of purine alkaloids was found in leaves of stages II and III, in which [8-14C]Theobromine was degraded to CO2 via 3-methylxanthine, xanthine and allantoic acid. [8-14C]Caffeine was catabolised to CO2 via theophylline (1,3-dimethylxanthine) or theobromine.  相似文献

Deterioration of raw materials of six medicinal plants viz. Terminalia arjuna, Acorus calamus, Rauvolfia serpentina, Holarrhena antidysenterica, Withania somnifera and Boerhaavia diffusa was examined. Some of the contaminated raw materials were found to be deteriorated by toxigenic strains of Aspergillus flavus and contain aflatoxin B1 (41.0–95.4 μg kg−1) which is above the permissible limit. Essential oil of Cymbopogon flexuosus and its components was found efficient in checking fungal growth and aflatoxin production. C. flexuosus essential oil absolutely inhibited the growth of A. flavus and aflatoxin B1 production at 1.3 μl ml−1 and 1.0 μl ml−1 respectively. The individual oil components were more efficacious than the Cymbopogon oil as such which emphasizes masking of their efficacy when combined together. Eugenol exhibited potent antifungal and aflatoxin inhibitory activity at 0.3 μl ml−1 and 0.1 μl ml−1 respectively. Eugenol was found superior over some prevalent synthetic antimicrobials and exhibited broad fungitoxic spectrum against some biodeteriorating moulds. Prospects of exploitation of the oil and its components as acceptable plant based antimicrobials in qualitative as well as quantitative control of biodeterioration of herbal raw materials have been discussed.  相似文献

Explants excised from strawberry (Fragaria x ananassa Duch.) plantlets were cultured in vitro for 21 days on half-strength MS (Murashige & Skoog 1962) basal liquid medium with 20 g l-1 sucrose and without sugar in the vessels capped with gas permeable microporous polypropylene film. The experiments were conducted under CO2 nonenriched (350–450 mol mol-1 in the culture room) and CO2 enriched (2,000 mol mol-1 during the photoperiod in the culture room) conditions with a PPF (photosynthetic photon flux) of 200 mol m-2 s-1. The CO2 concentration in the vessels decreased to approximately 200 mol mol-1 during the photoperiod on day 21 under CO2 nonenriched conditions. The fresh and dry weight, net photosynthetic rate (NPR) per plantlet, NPR per g leaf fresh weight, NPR per g leaf dry weight, the number of unfolded leaves, and ion uptake of PO4 3-, NO3 -, Ca2+, Mg2+ and K+ on day 21 were the greatest under photoautotrophic (no sugar in the medium) and CO2 enriched conditions. The residual percent of PO4 3- was 3% on day 21 under photoautotrophic and CO2 enriched conditions.Abbreviations MS Murashige & Skoog (1962) basal medium composition - NPR net photosynthetic rate - PPF photosynthetic photon flux  相似文献

Summary Coconut (Cocos nucifera L.) plantlets grown in vitro often grow slowly when transferred to the field possibly, due to a limited photosynthetic capacity of in vitro-cultured plantlets, apparently caused by the sucrose added to growth medium causing negative feedback for photosynthesis. In this paper, we tested the hypothesis that high exogenous sucrose will decrease ribulose 1,5-bisphosphate carboxylase (Rubisco) activity and photosynthesis resulting in limited ex vitro growth. Plantlets grown with high exogenous sucrose (90 gl−1) had reduced photosynthetic activity that resulted in a poor photosynthetic response to high levels of light and CO2. These plantlets also had low amounts of Rubisco protein, low Rubisco activity, and reduced growth despite showing high survival when transferred to the field. Decreasing the medium’s sucrose concentration from 90 to 22.5 gl−1 or 0 gl−1 resulted in increased photosynthetic response to light and CO2 along with increased Rubisco and phosphoenolpyruvate carboxylase (PEPC) activities and proteins. However, plantlets grown in vitro without exogenous sucrose died when transferred ex vitro, whereas those grown with intermediate exogenous sucrose showed intermediate photosynthetic response, high survival, fast growth, and ex vitro photosynthesis. Thus, exogenous sucrose at moderate concentration decreased photosynthesis but increased survival, suggesting that both in vitro photosynthesis and exogenous sucrose reserves contribute to field establisment and growth of coconut plantlets cultured in vitro.  相似文献

Micro-cuttings (shoots with two small leaves) of cultivar M9 apple were cultured in-vitro for 40 d under CO2-enriched and non-enriched (i.e., ambient air) conditions, and at a PPF of 40 or 100 μmol m-2 s-1 Afterward, shoot length, number of leaves, leaf area, chlorophyll content, shoot and root fresh weights, and % survival were recorded. Those plant-lets grown under CO2- and PPF-enriched treatments were healthy and vigorous, and showed higher values for their growth parameters. In contrast, those grown without supplemental CO2 or PPF often showed hyperhydricity. We also demonstrated that CO2 enrichment and a relatively high PPF during in-vitro culture promoted normal photosynthesis and growth after ex-vitro transplantation.  相似文献

We studied the seasonal variation on aerobic metabolism and the response of oxidative stress parameters in the digestive glands of the subpolar limpet Nacella (P.) magellanica. Sampling was carried out from July (winter) 2002 to July 2003 in Beagle Channel, Tierra del Fuego, Argentina. Whole animal respiration rates increased in early spring as the animals spawned and remained elevated throughout summer and fall (winter: 0.09 ± 0.02 μmol O2 h− 1 g− 1; summer: 0.31 ± 0.06 μmol O2 h− 1 g− 1). Oxidative stress was assessed at the hydrophilic level as the ascorbyl radical content / ascorbate content ratio (A / AH). The A / AH ratio showed minimum values in winter (3.7 ± 0.2 10− 5 AU) and increased in summer (18 ± 5 10− 5 AU). A similar pattern was observed for lipid radical content (122 ± 29 pmol mg− 1 fresh mass [FW] in winter and 314 ± 45 pmol mg− 1 FW in summer), iron content (0.99 ± 0.07 and 2.7 ± 0.6 nmol mg− 1 FW in winter and summer, respectively) and catalase activity (2.9 ± 0.2 and 7 ± 1 U mg− 1 FW in winter and summer, respectively). Since nitrogen derived radicals are thought to be critically involved in oxidative metabolism in cells, nitric oxide content was measured and a significant difference in the content of the Fe–MGD–NO adduct in digestive glands from winter and summer animals was observed. Together, the data indicate that both oxygen and nitrogen radical generation rates in N. (P.) magellanica are strongly dependent on season.  相似文献

The spatial upscaling of soil respiration from field measurements to ecosystem levels will be biased without studying its spatial variation. We took advantage of the unique spatial gradients of an oak–grass savanna ecosystem in California, with widely spaced oak trees overlying a grass layer, to study the spatial variation in soil respiration and to use these natural gradients to partition soil respiration according to its autotrophic and heterotrophic components. We measured soil respiration along a 42.5 m transect between two oak trees in 2001 and 2002, and found that soil respiration under tree canopies decreased with distance from its base. In the open area, tree roots have no influence on soil respiration. Seasonally, soil respiration increased in spring until late April, and decreased in summer following the decrease in soil moisture content, despite the further increase in soil temperature. Soil respiration significantly increased following the rain events in autumn. During the grass growing season between November and mid-May, the average of CO2 efflux under trees was 2.29 μmol m−2 s−1, while CO2 efflux from the open area was 1.40 μmol m−2 s−1. We deduced that oak root respiration averaged as 0.89 μmol m−2 s−1, accounting for 39% of total soil respiration (oak root + grass root + microbes). During the dry season between mid-May and October, the average of CO2 efflux under trees was 0.87 μmol m−2 s−1, while CO2 efflux from the open areas was 0.51 μmol m−2 s−1. Oak root respiration was 0.36 μmol m−2 s−1, accounting for 41% of total soil respiration (oak root + microbes). The seasonal pattern of soil CO2 efflux under trees and in open areas was simulated by a bi-variable model driven by soil temperature and moisture. The diurnal pattern was influenced by tree physiology as well. Based on the spatial gradient of soil respiration, spatial analysis of crown closure and the simulation model, we spatially and temporally upscaled chamber measurements to the ecosystem scale. We estimated that the cumulative soil respiration in 2002 was 394 gC m−2 year−1 in the open area and 616 gC m−2 year−1 under trees with a site-average of 488 gC m−2 year−1.  相似文献

The effects of NaCl salinity on growth, morphology and photosynthesis of Salvinia natans (L.) All. were investigated by growing plants in a growth chamber at NaCl concentrations of 0, 50, 100 and 150 mM. The relative growth rates were high (ca. 0.3 d−1) at salinities up to 50 mM and decreased to less than 0.2 d−1 at higher salinities, but plants produced smaller and thicker leaves and had shorter stems and roots, probably imposed by the osmotic stress and lowered turgor pressure restricting cell expansion. Na+ concentrations in the plant tissue only increased three-fold, but uptake of K+ was reduced, resulting in very high Na+/K+ ratios at high salinities, indicating that S. natans lacks mechanisms to maintain ionic homeostasis in the cells. The contents of proline in the plant tissue increased at high salinity, but concentrations were very low (<0.1 μmol g−1 FW), indicating a limited capacity of S. natans to synthesize proline as a compatible compound. The potential photochemical efficiency of PSII (Fv/Fm) of S. natans remained unchanged at 50 mM NaCl but was reduced at higher salinities, and the photosynthetic capacity (ETRmax) was significantly reduced at 50 mM NaCl and higher. It is concluded that S. natans is a salt-sensitive species lacking physiological measures to cope with exposure to high NaCl salinity. At low salinities salts are taken up and accumulate in old leaves, and high growth rates are maintained because new leaves are produced at a higher rate than for plants not exposed to salt.  相似文献

Summary The proliferation and survival of avocado nodal cultures of juvenile origin were affected by the form and concentration of nitrogen. Optimum growth was achieved on modified Murashige and Skoog medium containing 67% KNO3 and 33% NH4NO3 with total N of 40 mM supplemented with 100 mg l−1 myo-inositol, 1 mg l−1 thiamine HCl, 30 g l−1 sucrose, and 4.44 μM BA with a 16-h photoperiod (120–150 μmol m−2 s−1). Proliferating shoots and plantlets were photosynthetically active. Better shoot growth and accumulation of higher biomass occurred in a CO2-enriched environment than under ambient CO2 conditions. CO2 assimilation efficiency, however, was higher under the latter conditions than in a CO2-enhanced environment, e.g., 31±7 and 17±2 μmol CO2 m−2 s−1, respectively. The net CO2 assimilation rates of in vitro grown plantlets were comparable to those of seedlings ex vitro.  相似文献

The nitrogen uptake and growth capabilities of the potentially harmful, raphidophycean flagellate Heterosigma akashiwo (Hada) Sournia were examined in unialgal batch cultures (strain CCMP 1912). Growth rates as a function of three nitrogen substrates (ammonium, nitrate and urea) were determined at saturating and sub-saturating photosynthetic photon flux densities (PPFDs). At saturating PPFD (110 μE m−2 s−1), the growth rate of H. akashiwo was slightly greater for cells grown on NH4+ (0.89 d−1) compared to cells grown on NO3 or urea, which had identical growth rates (0.82 d−1). At sub-saturating PPFD (40 μE m−2 s−1), both urea- and NH4+-grown cells grew faster than NO3-grown cells (0.61, 0.57 and 0.46 d−1, respectively). The N uptake kinetic parameters were investigated using exponentially growing batch cultures of H. akashiwo and the 15N-tracer technique. Maximum specific uptake rates (Vmax) for unialgal cultures grown at 15 °C and saturating PPFD (110 μE m−2 s−1) were 28.0, 18.0 and 2.89 × 10−3 h−1 for NH4+, NO3 and urea, respectively. The traditional measure of nutrient affinity—the half saturation constants (Ks) were similar for NH4+ and NO3 (1.44 and 1.47 μg-at N L−1), but substantially lower for urea (0.42 μg-at N L−1). Whereas the α parameter (α = Vmax/Ks), which is considered a more robust indicator for substrate affinity when substrate concentrations are low (<Ks), were 19.4, 12.2 and 6.88 × 10−3 h−1/(μg-at N L−1) for NH4+, NO3 and urea, respectively. These laboratory results demonstrate that at both saturating and sub-saturating N concentrations, N uptake preference follows the order: NH4+ > NO3 > urea, and suggests that natural blooms of H. akashiwo may be initiated or maintained by any of the three nitrogen substrates examined.  相似文献
