In order to adapt to the fluctuations in soil salinity/osmolarity the bacteria of the genusAzospirillum accumulate compatible solutes such as glutamate, proline, glycine betaine, trehalose, etc. Proline seems to play a major role in osmoadaptation. With increase in osmotic stress the dominant osmolyte inA. brasilense shifts from glutamate to proline. Accumulation of proline inA. brasilense occurs by both uptake and synthesis. At higher osmolarityA. brasilense Sp7 accumulates high intracellular concentration of glycine betaine which is taken up via a high affinity glycine betaine transport system. A salinity stress induced, periplasmically located, glycine betaine binding protein (GBBP) of ca. 32 kDa size is involved in glycine betaine uptake inA. brasilense Sp7. Although a similar protein is also present inA. brasilense Cd it does not help in osmoprotection. It is not known ifA. brasilense Cd can also accumulate glycine betaine under salinity stress and if the GBBP-like protein plays any role in glycine betaine uptake. This strain, under salt stress, seems to have inadequate levels of ATP to support growth and glycine betaine uptake simultaneously. ExceptA. halopraeferens, all other species ofAzospirillum lack the ability to convert choline into glycine betaine. Mobilization of thebet ABT genes ofE. coli intoA. brasilense enables it to use choline for osmoprotection. Recently, aproU-like locus fromA. lipoferum showing physical homology to theproU gene region ofE. coli has been cloned. Replacement of this locus, after inactivation by the insertion of kanamycin resistance gene cassette, inA. lipoferum genome results in the recovery of mutants which fail to use glycine betaine as osmoprotectant.  相似文献

Amaranth (Amaranthus hypochondriacus Linn.) is an important pseudocereal crop having important nutrients along with the indispensable amino-acids. The present study was aimed to study the effect of plant growth promoting bacilli on proximate constituents of amaranth grains, including three of the essential amino acids (methionine, lysine and, tryptophan). The combination of Bacillus pumilus and Bacillus subtilis showed a significant increase in different proximate constituents, including crude protein (22.13%), dry matter (32.25%), fat (30.77%), and carbohydrate (49.08%) in amaranth grains. Similarly, a significant increase in essential amino-acids (methionine 47.68%, lysine 59.41% and, tryptophan 38.05%) was recorded. This study suggests that the combination of Bacillus pumilus BS-27 and Bacillus subtilis BS-58 provides the natural, persistent and durable potential to enhance the nutritive value of the crop. Therefore, present study was designed to explore the enhancement of most desirable amino acid synthesis in amaranth due to application of plant growth promoting Bacillus spp.  相似文献

Abstract

Bacterium Pseudomonas spp. olive green (OG) was isolated from marine water, yet, it was characterized as plant growth promoting bacterium (PGPB). Multiple plant growth promoting traits of OG isolate were determined in vitro. It was tested positive for Indole-3-acetic acid (IAA) production with 29 µg ml?1 of IAA yield, phosphate solubilization with 34 µg ml?1 solubilization of Tri-calcium-phosphate and it showed maximum of 32 µg ml?1 of ammonia production. OG isolate was affirming siderophore production, hydrocyanic acid (HCN) production and catalase production. 16S rRNA gene sequence comparison was used to identify the isolate which showed its closest neighbor to be Pseudomonas fluoroscens strain BCPBMS-1. Efficacy of this PGPB was tested on the seedling growth of two test plants chickpea and green gram. Both the test plants treated with OG-based talc bioformulation showed significant growth promotion. Chickpea showed enhanced overall fresh biomass by 24%, overall dry biomass by 27% was observed after 15 days of seeded in pots. Green gram showed enhanced overall dry biomass by 28% was observed after 10 days of seeded in pots.  相似文献

Drought is one of the major abiotic stresses affecting yield of dryland crops. Rhizobacterial populations of stressed soils are adapted and tolerant to stress and can be screened for isolation of efficient stress adaptive/tolerant, plant growth promoting rhizobacterial (PGPR) strains that can be used as inoculants for crops grown in stressed ecosystems. The effect of inoculation of five drought tolerant plant growth promoting Pseudomonas spp. strains namely P. entomophila strain BV-P13, P. stutzeri strain GRFHAP-P14, P. putida strain GAP-P45, P. syringae strain GRFHYTP52, and P. monteilli strain WAPP53 on growth, osmoregulation and antioxidant status of maize seedlings under drought stress conditions was investigated. Drought stress induced by withholding irrigation had drastic effects on growth of maize seedlings. However seed bacterization of maize with Pseudomonas spp. strains improved plant biomass, relative water content, leaf water potential, root adhering soil/root tissue ratio, aggregate stability and mean weight diameter and decreased leaf water loss. The inoculated plants showed higher levels of proline, sugars, free amino acids under drought stress. However protein and starch content was reduced under drought stress conditions. Inoculation decreased electrolyte leakage compared to uninoculated seedlings under drought stress. As compared to uninoculated seedlings, inoculated seedlings showed significantly lower activities of antioxidant enzymes, ascorbate peroxidase (APX), catalase (CAT), glutathione peroxidase (GPX) under drought stress, indicating that inoculated seedlings felt less stress as compared to uninoculated seedlings. The strain GAP-P45 was found to be the best in terms of influencing growth and biochemical and physiological status of the seedlings under drought stress. The study reports the potential of rhizobacteria in alleviating drought stress effects in maize.  相似文献

The linear growth of 10 isolates each of Gaeumannomyces graminis var. graminis, G. graminis var. tritici and Phialophora graminicola and five isolates each of G. graminis var. avenae and a lobed-hyphopodiate Phialophora sp. was studied on osmotically adjusted agar at 20 °C. While most isolates of G. graminis var. avenae ceased growing at osmotic potentials of -60 bars (1 bar = 105 Pa), six out of 10 isolates of G. graminis var. tritici grew at that potential. The growth of all isolates of G. graminis var. tritici and var. avenae ceased at -70 bars. In contrast, four out of 10 isolates of P. graminicola grew at -70 bars, but all stopped growing at -80 bars. Most of the isolates of G. graminis var. graminis and the lobed-hyphopodiate Phialophora sp. grew at -70 bars while three out of 10 isolates of G. graminis var. graminis and one out of five isolates of the lobed-hyphopodiate Phialophora sp. were capable of growth at -80 bars. None of the fungi grew at -90 bars. Detailed studies of the growth of two or three isolates each of the five fungi at 10, 20, 30 and 35 °C were carried out on osmotic agar controlled by the addition of either sodium chloride or potassium chloride. In general, similar reductions in growth occurred with decreasing osmotic potential regardless of the solute used. At 10 and 20 °C., all three isolates of P. graminicola showed optimal growth at about -5 bars while the other fungi grew fastest at -12middot; bars. At 30 °C., one isolate of the lobed hyphopodiate Phialophora sp. and two isolates each of P. graminicola, G. graminis var. tritici and G. graminis var. avenae grew optimally at osmotic potentials of -10 to -15 bars. The other isolate of the Phialophora sp. and two isolates of G. graminis var. graminis studied grew optimally at the highest potential (-1·2 bars). However, at 35 °C the last three fungi exhibited optimal growth at osmotic potentials of-10 to -20 bars. The ecological significance of these results is discussed in relation to cross-protection against the take-all fungi by the avirulent fungi.  相似文献

Soil salinity is one of the most severe factors limiting growth and physiological response in Raphanus sativus. In this study, the possible role of plant growth promoting bacteria (PGPB) in alleviating soil salinity stress during plant growth under greenhouse conditions was investigated. Increasing salinity in the soil decreased plant growth, photosynthetic pigments content, phytohormones contents (indole-3-acetic acid, IAA and gibberellic acid, GA3) and mineral uptake compared to soil without salinity. Seeds inoculated with Bacillus subtilis and Pseudomonas fluorescens caused significantly increase in fresh and dry masses of roots and leaves, photosynthetic pigments, proline, total free amino acids and crude protein contents compared to noninoculated ones under salinity. The bacteria also increased phytohormones contents (IAA and GA3) and the contents of N, P, K+, Ca2+, and Mg2+ but decreased ABA contents and Na+ and Cl? content which may contribute in part to activation of processes involved in the alleviation of the effect of salt.  相似文献

Pseudomonas fluorescens strain GRS1, PRS9 and their cold tolerant mutants were examined for their tricalcium phosphate (TCP) solubilizing activity in NBRIP (broth) media at 10°C and 25°C. Invariably, all the cold tolerant mutants of GRS1 and PRS9 were found more efficient than their respective wild type counterparts for ‘P’ solubilization activity at 10°C as compared to 25°C. ‘P’ solubilization potential of CRM was found maximum among all the strains followed by CRPF6 and CRPF4. To the best of out knowledge, this is the first report regarding low temperature ‘P’ solubilization activity.  相似文献

Bacterial systems have evolved a number of mechanisms, both active and passive, to manage toxic concentrations of heavy metals in their environment. The present study is aimed at describing the zinc resistance mechanism in a rhizospheric isolate, Pseudomonas fluorescens strain Psd. The strain was able to sustain an external Zn2+ concentration of up to 5 mM in the medium. The strategy for metal management by the strain was found to be extracellular biosorption with a possible role of exopolysaccharides in metal accumulation. The attainment of equilibrium in biosorption reaction was found to be dependent on initial Zn2+ concentration, with the reaction reaching equilibrium faster (50 min) at high initial Zn2+ concentration. Biosorption kinetics of the process was adjusted to pseudo-first order rate equation. With the help of Langmuir and Freundlich adsorption isotherms, it was established that Zn2+ biosorption by the bacterium is a thermodynamically favourable process.  相似文献

Different strains of Pseudomonas putida and P. fluorescens isolated from the rhizosphere and phyllosphere were tested for surface activity in droplet cultures on polystyrene. Droplets of 6 of the 12 wild types tested spread over the surface during incubation, and these strains were considered surface active; strains not showing this reaction were considered non-surface active. Similar reactions were observed on pieces of wheat leaves. Supernatants from centrifuged broth cultures behaved like droplets of suspensions in broth; exposure to 100°C destroyed the activity. Average contact angles of the supernatants of surface-active and non-surface-active strains on polystyrene were 24° and 72°, respectively. The minimal surface tension of supernatants of the surface-active strains was about 46 mN/m, whereas that of the non-surface-active strains was 64 mN/m (estimations from Zisman plots). After 6 days of incubation, wheat flag leaves sprayed with a dilute suspension of a surface-active strain of P. putida (WCS 358RR) showed a significant increase in leaf wettability, which was determined by contact angle measurements. Increasing the initial concentration of bacteria and the amount of nutrients in the inoculum sprayed on leaves reduced the contact angles from 138° on leaves treated with antibiotics (control) to 43° on leaves treated with surface-active bacteria. A closely related strain with no surface activity on polystyrene did not affect leaf wettability, although it was present in densities similar to those of the surface-active strain. Nutrients alone could occasionally also increase leaf wettability, apparently by stimulating naturally occurring surface-active bacteria. When estimating densities of Pseudomonas spp. underneath droplets with low contact angles, it appeared that populations on leaves treated with a surface-active strain could vary from about 104 to 106 CFU cm−2, suggesting that the surface effect may be prolonged after a decline of the population. Use of plant growth promoting rhizobacteria in managing bacterial canker disease of tomato was studied in the present work. Tomato seeds were treated with PGPR strains viz., Bacillus pumilus INR7, Bacillus pumilus SE34, Bacillus pumilus T4, Bacillus subtilis GBO3, Bacillus amyloliquefaciens IN937a and Brevibacillus brevis IPC11 were subjected for seed germination and seedling vigor. Among the PGPR strains tested, only three strains (IN937a, GBO3 and IPC11) which showed enhancement in the seed quality parameters like seed germination and seedling vigor, were further subjected for estimation of one of the defence-related enzymes, Phenylalanine Ammonia Lyase (PAL) with total phenol contents. The same three strains were recorded for maximum disease protection under greenhouse conditions. The level of PAL and total phenol contents increased significantly upon the PGPR treatment. The rate of reduction in the bacterial canker disease incidence was directly proportional to the amount of increased level of PAL and total phenol content. The possible uses of these PGPR strains in effective management of bacterial canker of tomato were discussed in the present work.  相似文献

The outer membrane proteins of a series of fluorescent, root-colonizing, plant-growth-stimulating Pseudomonas spp. having been characterized (L. A. de Weger et al., J. Bacteriol. 165:585-594, 1986), the lipopolysaccharides (LPSs) of these strains were examined. The chemical composition of the LPSs of the three best-studied plant-growth-stimulating Pseudomonas strains WCS358, WCS361, and WCS374 and of P. aeruginosa PAO1 as a reference strain was determined and appeared to differ from strain to strain. The 2,6-dideoxy-2-aminosugar quinovasamine was the most abundant compound in the LPS of strain WCS358. Analysis by sodium dodecyl sulfate-polyacrylamide gel electrophoresis of purified LPS and of proteinase K-treated cell envelopes revealed ladderlike patterns for most of these strains. These patterns were not substantially influenced by differences in culture conditions. Analysis of proteinase K-treated cell envelopes of 24 root-colonizing Pseudomonas spp. revealed a unique band pattern for each strain, suggesting a great variety in the LPS structures present in these root colonizers. Therefore, electrophoretic analysis of LPS can be used for characterization and identification of the fluorescent root-colonizing Pseudomonas strains.  相似文献

Soil microorganisms with potential for alleviation of abiotic stresses in combination with plant growth promotion would be extremely useful tools in sustainable agriculture. To this end, the present study was initiated where forty-five salt tolerant bacterial isolates with ability to grow in high salt medium were obtained from the rhizosphere of Triticum aestivum and Imperata cylindrica. These bacteria were tested for plant-growth-promoting rhizobacteria traits in vitro such as phosphate solubilization, siderophore, ACC deaminase and IAA production. Of the forty-five isolates, W10 from wheat rhizosphere and IP8 from blady grass rhizosphere, which tested positive in all the tests were identified by morpholological, biochemical and 16SrDNA sequencing as Bacillus safensis and Ochrobactrum pseudogregnonense respectively and selected for in vivo studies. Both the bacteria could promote growth in six varieties of wheat tested in terms of increase in root and shoot biomass, height of plants, yield, as well as increase in chlorophyll content. Besides, the wheat plants could withstand water stress more efficiently in presence of the bacteria as indicated by delay in appearance of wilting symptoms increases in relative water content of treated water stressed plants in comparison to untreated stressed ones, and elevated antioxidant responses. Enhanced antioxidant responses were evident as elevated activities of enzymes such as catalase, peroxidase, ascorbate peroxidase, superoxide dismutase and glutathione reductase as well as increased accumulation of antioxidants such as carotenoids and ascorbate. Results clearly indicate that the ability of wheat plants to withstand water stress is enhanced by application of these bacteria which also function as plant growth promoting rhizobacteria.  相似文献

The isolate NJ-101 obtained from agricultural soil was characterized and presumptively identified as Pseudomonas sp. The isolate exhibited efficient degradation of the insecticide carbofuran with a rate constant of 0.035 day(-1), following first-order rate kinetics. The ability of performing multifarious biological activities in tandem suggested the uniqueness of isolate NJ-101. The ability to produce hydrogen cyanide and siderophore stipulated its role in biological control. Furthermore, the growth inhibition of Fusarium sp. validated the antagonistic activity of NJ-101 against the common phytopathogens. Concurrent production of indole acetic acid, and solubilization of inorganic phosphate revealed its plant growth promoting potential. Thus, the innate capability of this novel isolate for parallel biodegradation, biocontrol and plant growth promotion has significance in management of the agro-environmental and phytopathological problems.  相似文献

Bacterial inoculants of the commercially available plant growth promoting rhizobacteria (PGPR) Arthrobacter mysorens 7, Flavobacterium sp. L30, and Klebsiella mobilis CIAM 880 were selected to obtain ecologically safe barley crop production on cadmium (Cd) polluted soils. All the PGPR immobilized 24–68% soluble cadmium from soil suspension. A. mysorens 7 and K. mobilis CIAM 880 were highly resistant to Cd and grew in up to 1 and 3 mmol CdCl2 on DAS medium respectively. All PGPR were able to fix nitrogen (276–1014 nmol mg–1 bacterial DW) and to produce indole acetic acid (IAA) (126–330 nmol mg–1 bacterial DW) or ethylene (4.6–13.5 nmol bacterial DW). All the PGPR actively colonized barley root system and rhizosphere and significantly stimulated root elongation of barley seedlings (up to 25%), growing on soil containing 5 or 15 mg Cd kg–1 of soil. Created in the simulation mathematical model confirms our hypothesis that PGPR beneficial effect on barley growing under Cd-stress is a complex process. One of mechanisms underlying this effect might be increase of bacterial migration from rhizoplane to rhizosphere, where PGPR bind soluble free Cd ions in biologically unavailable complex forms. Among the studied PGPR K. mobilis CIAM 880 was the most effective inoculant. Inoculation with K. mobilis CIAM 880 of barley plants growing on Cd contaminated soil (5 mg Cd kg–1 of soil) under field conditions increased by 120% grain yield and 2-fold decreased Cd content in barley grain. The results suggest that the using K. mobilis CIAM 880 is an effective way to increase the plant yield on poor and polluted areas.  相似文献

P. fluorescens strain Psd was isolated from the rhizosphere of Vigna mungo and evaluated for its multiple plant growth promoting and biocontrol properties against F. oxyspornum. Interestingly, this strain not only produces a range of antimicrobial compounds but also solubilizes complexed phosphates and synthesizes phytohormone (IAA). These properties can be assessed to elucidate the agronomic significance and rhizospheric competence of this soil isolate. Biocontrol action has been demonstrated in vitro against some other rhizospheric bacteria, and a phytopathogenic fungus along with wild type E. coli K-12. Genetic evidence for the antimicrobial status of strain Psd has been derived in terms of elucidating a unique combination of phenazine and pyrrolnitrin biosynthesis genes, not reported for any other P. fluorescens strain. The conserved part of antibiotics biosynthesis operon has been PCR amplified, cloned, sequenced and phylogenetic relationship based on similar genes from a few known Pseudomonads has been derived. The properties possessed by strain Psd may enable the bacterium to establish itself successfully in the rhizosphere.  相似文献

The study was designed to assess the effect of selected pesticides (metribuzin, glyphosate, imidacloprid, thiamethoxam, hexaconazole, metalaxyl and kitazin) at the recommended and higher rates on plant growth promoting activities of Pseudomonas aeruginosa strain PS1 isolated from mustard (Brassica compestris) rhizosphere. The strain PS1 was specifically chosen owing to its substantial tolerance against pesticides, phosphate solubilization and considerable production of indole acetic acid, siderophores, exo-polysaccharides, HCN and ammonia. Plant growth promoting traits of the strain PS1 decreased consistently as the concentrations of each pesticide was increased from the recommended dose to the higher ones. Generally, the maximum toxicity to plant growth promoting traits was displayed by pesticides at three times the recommended field rate. However, the effect on the plant growth promoting activities of the P. aeruginosa strain PS1 at the recommended dose of each pesticide was less hazardous. This study revealed an additional aspect of the toxicological mechanisms of the pesticides through which they suppress the plant growth.  相似文献

【目的】挖掘镍耐受性强、产铁载体活性高的植物根际促生细菌,研究镍胁迫下产铁载体细菌对花生的促生作用及其对花生吸收镍的影响。【方法】利用CAS(Chrome azurol S)培养基对花生根际产铁载体细菌定性筛选及定量测试获得产铁载体能力强的菌株,16S r RNA基因相似性及系统进化分析鉴定产铁载体细菌,并用含Ni~(2+)牛肉膏蛋白胨培养基测试细菌对Ni的耐受性;通过花生盆栽实验,测试花生的株高、根长、生物量、氮磷钾含量及镍含量来分析镍胁迫下产铁载体细菌对花生的影响。【结果】从花生根际分离筛选产铁载体芽孢杆菌5株,其中HSGJ1产铁载体能力最强,培养2 d后产156.56 mg/L的铁载体。HSGJ1对Ni~(2+)具有较强的耐受性,最小致死浓度为150 mg/L。在50、100 mg/kg的Ni~(2+)盆栽基质中,HSGJ1能够有效地促进花生的生长、增加花生的生物量及氮磷钾含量,并使花生根部和地上部分的镍含量降低。【结论】产铁载体芽孢杆菌HSGJ1是一株优良的植物根际促生细菌,可应用于镍污染农耕土壤的作物种植中,以提高作物在镍胁迫下的抗逆性,降低作物对镍的富集量,具有较好的应用价值。  相似文献

We investigated the effect of different oxygen regimes on growth patterns of Pseudomonas spp. during benzene degradation in microcosm batch studies. Benzene degradation was induced by limiting oxygen available for microbial activity, which consists of three initial-dissolved oxygen (DO) levels of oxic, hypoxic, and anoxic conditions. Batch experiments were performed for cell growth and benzene degradation by inoculating three strains of Pseudomonas spp. (Pseudomonas aeruginosa, Pseudomonas fluorescens, Pseudomonas putida) in mineral salt medium containing aqueous benzene. Results showed that all strains were capable to grow and degrade benzene under all oxygen regimes but in a different manner. The highest cell growth of P. aeruginosa and P. fluorescens was achieved under oxic and anoxic condition, respectively, but there was no substantial difference on benzene degradation between the oxygen treatments with about 25% reduction for both strains. P. putida showed a facultative process for both cell growth and benzene degradation. This reveals that care should be taken in selection of microorganisms with regard to environmental studies since they exhibit different responses for given environmental conditions such as DO levels.  相似文献
